Six chlorophyll (Chl)-protein complexes associated with photosystemI (CPla), and the PS I reaction center complex (CPl) were isolatedfrom the thylakoid membranes of the green alga, Bryopsis maxima,by SDS-polyacrylamide gel electrophoresis. CPla had four polypeptides(22, 24, 25, 26 kDa) in addition to the 67 kDa polypeptide ofCPl. These complexes may thus possibly be a combination of CPland antenna complexes for PS I. Six CPla showed almost the sameoptical properties, with absorption maxima at 650 and 677 nmand contained carotene and a small amount of xanthophylls. TheChl a/b ratios of these CPla were about 2, while that of CPlwas 14. CPla showed a fluorescence emission maximum at 695 nm;its excitation spectrum had peaks at 438, 470 and 540 nm, correspondingto the absorption maxima of Chl a, Chl b, xanthophylls, respectively.An antenna complex free of CPl has been detected in some plantsbut was not found in the present alga. 1Present address: Department of Botany, The University of Adelaide,Adelaide, S.A. 5001, Australia (Received April 17, 1986; Accepted June 26, 1986)  相似文献

Previous cell cycle studies have been based on cell-nuclearproliferation only. Eukaryotic cells, however, have double membranes-boundorganelles, such as the cell nucleus, mitochondrion, plastidsand single-membrane-bound organelles such as ER, the Golgi body,vacuoles (lysosomes) and microbodies. Organelle proliferations,which are very important for cell functions, are poorly understood.To clarify this, we performed a microarray analysis during thecell cycle of Cyanidioschyzon merolae. C. merolae cells containa minimum set of organelles that divide synchronously. The nuclear,mitochondrial and plastid genomes were completely sequenced.The results showed that, of 158 genes induced during the S orG2-M phase, 93 were known and contained genes related to mitochondrialdivision, ftsZ1-1, ftsz1-2 and mda1, and plastid division, ftsZ2-1,ftsZ2-2 and cmdnm2. Moreover, three genes, involved in vesicletrafficking between the single-membrane organelles such as vps29and the Rab family protein, were identified and might be relatedto partitioning of single-membrane-bound organelles. In othergenes, 46 were hypothetical and 19 were hypothetical conserved.The possibility of finding novel organelle division genes fromhypothetical and hypothetical conserved genes in the S and G2-Mexpression groups is discussed.  相似文献

Domozych, D. S. 1987. An experimental analysis of the dictyosomein the green alga, Tetraselmis convolutae.—J. exp. Bot.38: 1399–1411. Tetraselmis convolutae possesses two well-defined, polar-orienteddictyosomes. These endo-membrane components have various functionsthat change during the cell cycle phases. The dictyosomes arealso very sensitive to various subcellular poisons. Disruptionof the cellular ionic balance with monovalent ionophores yieldsmaturing face curling, swelling and the production of unusualmultivesicular elements. Divalent ionophore application alsoimpairs maturing face vesiculation and vesicle transport inthe anterior cytoplasm. Caffeine restricts maturing face vesicleproduction, subsequently leading to gross enlargement of cisternallength. Application of the microtubule-inhibitor, colchicine,and the MTOC-affecting agent, IPC, leads to polar axis disruptionof the dictyosome and the formation of multiple, cisternal stacks.A discussion of dictyosome-microtubule-MTOC associations ispresented. The division of chloroplasts in a green alga, Pediastrum duplex,was studied by electron microscopy. Cells were treated for observationwith the freeze-substitution method. Fibrils, or fibrous belts,which we had observed previously at the dividing constrictionsof chloroplasts in Trebouxia potteri were not visible in Pediastrum,even though the method of preparation was the same for bothsets of samples. Microtubules (MTs) and the septum seem notto participate directly in the division of the single chloroplastin Pediastrum cells. Many thin fibrils, 7–20 nm in diameter,attached to, or protruding from, the surface of the dividingconstriction were seen. These fibres were less densely distributedat the constrictions of non-dividing chloroplasts. It is suggestedthat these fibrils are involved in the divison of chloroplastsin Pediastrum duplex. SYNOPSIS. Agelena consociata Denis, which Chauvin discoveredin Gabon, when put in a laboratory, weaves nests which are 2m2 and consist of three different parts: shelters, a web, andvertical threads used as a snare. The snare stops insects intheir flight and causes them to fall into the web which thespiders use as their hunting field. These spiders have a nocturnalactivity which can be divided into two successive periods: afirst stage during which the spiders weave vertical threads,and a second stage of general activity. Hunting and eating are done in common. These activities andthe relations between the individuals which they involve, showin a definite way the social characteristics of these spiders. Eating favors an exchange of chemical substance between theindividuals, but we have not been able to determine the exactpart played by such an exchange. As study proceeds, we shall try to make clear the relationshipsbetween the individuals inside the society. We shall analyzegroup effects on physiology and behavior, comparing them tothose found for Agelena labyrinthica.  相似文献

The light-harvesting system of photosynthesis was studied infour strains of Porphyra yezoensis differing in their phycoerythrin(PE) content; the red strain, richer in PE than the wild strain,and the green and the yellow strains, poorer in PE. Specialattention was given to possible alteration of pigment systemin response to PE content, especially in the green and the yellowstrains. The relative quantum yields of Chi a fluorescence at–196°C and O2 evolution were compared. Four strains commonly showed a low yield in pigment system II(PS II) fluorescence on Chl a excitation. The yield was as lowas in those algae in which PS II has only a small portion ofChl a as the light harvester. Measurement of O2 evolution gavethe same results. Results indicate that the functional compositionof Chl a system remains unaltered in four strains with differentPE content. PS II in the green and the yellow strains reflectsa reduction in the size of light-harvesting components, suggestingthat pigmentation in these strains is fixed genetically as asun-type. 4 On leave from University of Washington, Seatle, Washington,U.S.A. The intracellular concentration of pyruvic and -ketoglutaricacids in Chlorella when grown under various conditions has beeninvestigated. The keto-acids were assayed after quantitativeelution of their 2,4: dinitrophenylhydrazones, chromatographicallyseparated. Very small amounts of keto-acids are found in Chlorella whengrown auto-trophically. The level was much higher under heterotrophicconditions and in nitrogen starvation. Further, with nitratethe level was considerably higher than if ammonia was supplied.In the presence of glucose this differential effect occurredwith pyruvic acid, but the level of -ketoglutaric acid appearedto be stabilized. An outbreak of a dryberry disease caused by Peronospora sparsa (syn. P. rubi) occurred in plantations of arctic bramble (Rubus arcticus subsp. arcticus) in Finland in the middle of 1990s. The disease persists and is most severe in cool and rainy summers. The disease has not been encountered in northern Sweden where cultivars (R. arcticus nothosubsp. stellarcticus) different from those in Finland are used. The occurrence of P. sparsa in wild Rubus spp. is virtually unknown in both areas and it is not known whether they constitute a potential infection source. Therefore, the aim of this study was to investigate the occurrence of P. sparsa on wild Rubus spp. growing in the vicinity of cultivations of arctic bramble. Symptomatic plants were sampled in 1997–1999. P. sparsa was detected using a light microscope, preceded by incubation of the sample in vitro if necessary, and by a polymerase chain reaction (PCR) based method. Plants of cultivated R. arcticus subsp. arcticus were commonly infected by P. sparsa in Finland. P. sparsa was also found on the cultivated R. arcticus nothosubsp. stellarcticus in Finland and Sweden. However, the infected plants of the cultivars of nothosubsp. stellarcticus seemed to be much less damaged than the cultivars of subsp. arcticus. Plants infected with P. sparsa were found in the populations of wild R. arcticus subsp. arcticus in both countries, and in cloudberry (R. chamaemorus) in natural habitats in Finland. In addition, P. sparsa was detected on specimens of R. arcticus subsp. arcticus (collected in 1966–1985) and R. chamaemorus (collected in 1899–1981) in Finnish herbaria. The samples of R. idaeus and R. saxatilis collected from the field in this study or investigated in the herbaria were not infected with P. sparsa. These data show that P. sparsa has not recently invaded Finland but has become an economically significant pathogen during the rapid expansion of cultivation of the apparently sensitive clones of arctic bramble.  相似文献

Carbonic anhydrase (CA) activity was detected in homogenatesfrom Anabaena variabilis ATCC 29413, M-2 and M-3, but not inthe suspension of the intact cells. Activity was higher in cellsgrown in ordinary air (low-CO2 cells) than in those grown inair enriched with 2–4% CO2 (high-CO2 cells). Fractionationby centrifugation indicated that the CA from A. variabilis ATCC29413 is soluble, whereas both soluble and insoluble forms existin A. variabilis M-2 and M-3. The addition of dithiothreitoland Mg2 $ greatly decreased the CA activity of A. variabilisATCC 29413. The specific activity of the CA from A. variabilis ATCC 29413was increased ca. 200 times by purification with ammonium sulfate,DEAE-Sephadex A-50 and Sephadex G-100. Major and minor CA peaksin Sephadex G-100 chromatography showed respective molecularweights of 48,000 and 25,000. The molecular weight of the CAdetermined by polyacrylamide disc gel electrophoresis was 42,000?5,000.The activity of CA was inhibited by ethoxyzolamide (I50=2.8?10-9M), acetazolamide (I50=2.5?10-7 M) and sulfanilamide (I50=2.9?10-6M). A rapid induction of sulfate transport was observed in the greenalga Chlorella ellipsoidea during sulfur-limited growth. Bothaffinity and Vmax increased about five-fold within 6 h of transferringcells from Bold's basal medium with 350 µM MgSO4 to sulfur-deficientBold's medium. High affinity sulfate transport was induced within15 min and reached maximum rate within 3 h of transferring cellsto sulfur-deficient condition, indicating that a new, high-affinity-sulfatetransport system is induced by sulfur starvation in C. ellipsoidea.Eadie-Hofstee plots of initial rates of sulfate uptake indicatedthat the K of sulfur-starved cells was about 17 µM. Bothsulfur-starved and unstarved cells grown in air had a Vmax of1.5 times higher than that of high-CO2 grown cells. Sulfatetransport was completely inhibited by 30 µM CCCP or 800µMKCN both in the light and the dark but transport in the lightwas not inhibited by 20 µM DCMU. Treatment with 50 µMor 500 µM vanadate caused 50% inhibition of uptake. Therate of sulfate uptake in the dark was twice that in the lightand was stimulated by low pH. These results suggest that thesulfate transport system in C. ellipsoidea is operated by protonsymport across the plasmamembrane which is partially mediatedby P-type ATPase and that these systems depend exclusively onenergy derived from oxidative phosphorylation in the mitochondria. Cylindrical 52.5-nm-long phycobilisomes were observed in Anabaenavariabilis, differing from the generally accepted hemidiscoidalmorphology. The central part of such a phycobilisome has a network-likefine structure of slightly greater diameter (16 nm) than theconnected end parts of stacked-disc structure (12 nm in diameter).On the basis of this morphology, the molecular mass of thisphycobilisome was calculated to be 3.27?106, about 60% of whichis accounted for by phycocyanin with the rest being due to allophycocyanin.Separately prepared 23 S allophycocyanin particles with a molecularmass of 1.13?106 have the dimensions (16?23 nm) and network-likefine structure similar to the central part of phycobilisomes,while an aggregate form of phycocyanin (18 S) has a fine structureof stacked discs similar to the connecting end part of phycobilisomes,suggesting that the central part constitutes the core at whichthese phycobilisomes attach to the thylakoid membranes. The positional distribution of fatty acids in glycerolipidsfrom thalli of Porphyra yezoensis was studied by enzymatic hydrolysis.In monogalactosyl diacylglycerol, icosapentaenoic acid was amajor fatty acid at both the sn-1 and sn-2 positions of theglycerol moiety, whereas palmitic acid was a minor componentat both positions. In digalactosyl diacylglycerol and sulfoquinovosyldiacylglycerol, icosapentaenoic and palmitic acids were almostexclusively distributed at the sn-1 and sn-2 positions, respectively.In phosphatidylglycerol, palmitic and trans--13-hexadecenoicacid were exclusively located at the sn-2 position. In phosphatidylcholine,icosapentaenoic acid occurred in both the sn-l and sn-2 positions,whereas palmitic acid was confined to the sn-1 position. Itis suggested that monogalactosyl diacylglycerol in P. yezoensissynthesized in both the cytoplasmic and chloroplastic pathways,while the diacylglycerol moieties of the other chloroplast lipidsare virtually all derived from the chloroplastic pathway. Chaenothecopsis consociata regularly is lichenized withDictyochloropsis symbiontica, nova spec., a member of theChlorococcales. During early developmentChaenothecopsis consociata possibly parasitizes onChaenotheca chrysocephala.Dictyochloropsis symbiontica is characterized by its hollow spherical chloroplast which consists of a complex threedimensional network, the absence of a pyrenoid, and the possession of one central nucleus. Acriflavine and acridine orange were highly effective in producingmutants defective in spore differentiation in the blue-greenalga, Anabaena doliolum. Acridine orange produced both oligosporogeneous(Osp) and asporogeneous (Sp) mutants in high yield withthe frequency ranging from 2.4 to 21 per 104 survivors, whereaswith acriflavine no oligosporogeneous mutants were detected,although it produced non-sporulating (Sp) mutants withthe frequency of 1.4 per 104 survivors. Mutants isolated throughthese dyes were stable and showed no tendency towards spontaneousor induced reversion. Behaviour of various mutants indicatedtheir being blocked at different stages of sporulation withdifferent mutagenic sites. Induction of high numbers of Ospand Sp mutants after acridine dye treatment indicatesthe involvement of an extrachromosomal determinant in sporulationin Anabaena doliolum. The rove beetle genus Drusilla includes some myrmecophilous species. The Japanese species Drusilla sparsa (Sharp, 1874) has been regarded as a non‐myrmecophilous beetle. In Kagawa Prefecture, Shikoku Island, western Japan, however, we often observed that D. sparsa adults were walking in the vicinity of foraging workers of the myrmicine ant Crematogaster osakensis Forel, 1990. The body color of the beetle is similar to C. osakensis as in other myrmecophilous beetles found near the trails of the host ants. To examine whether D. sparsa is myrmecophilous, we investigated the distribution of D. sparsa and C. osakensis in the field, as well as their behavior including prey preference of the beetle in the laboratory. Drusilla sparsa beetles were collected only in sites where C. osakensis ants occurred. When the beetles encountered the ant workers, they bent the abdominal tip toward the ants. The ants licked the abdominal tip, and then the beetles usually walked away. Such behavioral reaction of the ants was not observed when the beetles encountered workers of the formicine ant Nylanderia flavipes (Smith, 1874) that continuously attacked the beetles. Drusilla sparsa preferred to feed on dead workers of C. osakensis even when other ants were available as food, indicating that D. sparsa is a myrmecophilous species associated with C. osakensis. Crematogaster osakensis was frequently found in the stomach in the ant predator, the Japanese treefrog Hyla japonica Günther, 1859. Thus, the significance of body color similarity between the host ants and beetles is not a case of Batesian mimicry.  相似文献

The spores of Anabaena doliolum formed in light (light spores)and after transfer to darkness (dark spores) are biochemicallydifferent in that the light spores contain chlorophyll a andphycocyanin, while dark spores seem to lack them. The apparentbiosyntheses accompanying dark-spore germination seem to proceedin the following order: RNA, chlorophyll a, phycocyanin andDNA. Results of chloramphenicol treatment indicate that proteinsynthesis precedes RNA synthesis. The biosynthetic events followingRNA synthesis show a requirement for light.  相似文献

The carbohydrate content of various cellular fractions of theblue-green alga Nostoc was studied as a function of age of theculture. The production of extracellular and intracellular polysaccharideswas higher in actively growing cultures. Mannose and glucosewere the main components of cell wall polysaccharides. Glucosamineand diaminopimelic acid were also detected in the cell walls.The kinetics of incorporation of radioactivity from sodium [14C]bicarbonate showed that the extracellular polysaccharides werelabelled within 10 min of incubation suggesting the active exudationof polysaccharides by this alga. The selective excretion ofpolysaccharides by the alga Nostoc is also suggested.  相似文献

SDS-solubilized thylakoid membranes of Bryopsis maxima showeda similar pattern to those of higher plants in SDS-poIyacrylamidegel electrophoresis. Absorption spectra and pigment compositionof both CP1 and CPa bands were similar to those of higher plantsand other algae. Five bands containing chlorophyll (Chl) b weredivided into three categories; a group of major light-harvestingChl a/b-protein complexes (LHCP 1, LHCP 2 and LHCP 3), a minorLHCP (LHCP 3') and a photosystem I complex (CP1a). LHCP 1, thehigh molecular form, showed the lowest Chl a/b ratio among theLHCPs, and contained only xanthophylls as carotenoids. LHCP2, LHCP 3 and LHCP 3' bands contained xanthophylls and carotene.Carotenoid composition of LHCP 3' was different from that ofthe major LHCPs. It has been shown by experimentation that two morphologicallydistinct clones of Fragilaria crotonensis Kitton, isolated fromthe plankton of Windermere at approximately the same time, wereaffected quite differently by the chytrid Rhizophydium fragilariaeCanter. In one case the diatom filaments became heavily parasitizedand could be used for the maintenance of the fungus. In theother case infection was negligible. Additional clones, representative of these two morphologicalforms, from other bodies of water in the Windermere drainagebasin were tested. The same pattern of infection or relativenon-infection again ensued in direct accordance with the particulartype of filament used. Some preliminary experiments undertaken with a second, as yetlittle known chytrid parasite of F. crotonensis are reported.In this instance the fungus appeared to find its natural hoston cells belonging to the morphological form which failed tosupport the continued increase of R. fragilariae. The parasitism of morphological variants of F. crotonensis bythese fungi in the wild from Britain and else where is discussed. The presence of nucleus, chloroplasts, mitochondria, microbodiesand a vacuole are confirmed in Cyanidium caldarium strain CCAP1355/1. Chloroplasts usually contain parallel rows of unstackedthylakoids surrounded by a peripheral thylakoid, although theconcentric arrangement has also been observed. The chloroplastenvelope consists of two closely appressed membranes which canonly be resolved after gluteraldehyde—osmium fixation.The chloroplast of Rhodosorus marinus also contains parallel,unstacked thylakoids surrounded by a peripheral thylakoid but,in addition, a prominent, stalked pyrenoid projects into thecytoplasm and is bounded by both the peripheral thylakoid andthe chloroplast envelope. This structure is covered by a capof starch grains and contains membranous vesicles in the matrix.Phycobilisomes are absent from the chloroplasts of both algae.The recognition of C. caldarium as a rhodophyte is supportedby these observations.
