The annual shoots of apple and pear trees which accumulated a high concentration of arginine during the dormant stage also contained Nα-acylarginine derivatives. Nα-(2-Hydroxysuccinyl)arginine, Nα-(3-hydroxysuccinyl)arginine and Nα-oxalylarginine were found in apple trees, and Nα-succinylarginine and Nα-(2-carboxymethyl-2-hydroxysuccinyl)arginine, besides the former three, were found in pear trees. Nα-(3-Hydroxysuccinyl)arginine, Nα-oxalylarginine and Nα-succinylarginine are new arginine derivatives.  相似文献

Summary The influence of partial substitution of agar by galactomannans (GMs) in culture media was studied in pear (Pyrus communis L. cv. ‘Durondeau’) micropropagation. GMs. extracted from seeds of Cassia fastuosa (cassia) or Cyamopsis tetragonolobus (guar gum, a commercial GM), were mixed in equal proportions with agar to a final concentration of 0.3% (w/v) for each type of gelling agent. The production of multiple shoots and the formation of roots from shoots were compared with the control solidified with agar alone at a concentration of 0.6% (w/v). In the media solidified with the mixtures of agar/guar and agar/cassia GMs, an, increase of 32 and 17%, respectively, was obtained in the number of regenerated shoots. The modified media promoted a higher number of roots and increased the rooting percentage. A maximum of 91% rooting was obtained in the medium solidified with the agar/cassia GM and containing 9.80 μM indole-3-butyric acid. Less callus formation at the base of the shoot was also observed on this medium. The improved in vitro performance of shoot formation and rooting, combined with a significantly lower cost, suggests a potential use of agar/GM gels in plant tissue culture.  相似文献

Pear (Pyrus L.) is one of the most important fruits in temperate regions. Even in the mountainous border region of Tibet, Sichuan, and Yunnan Provinces (China), a variety of highly adapted pear landraces were discovered in Tibetan Autonomous Prefecture and were named “Zangli pears.” Here, we analyzed genetic diversity of 29 genotypes of Zangli distributed in Tibetan Region, together with 30 genotypes of sand pear (P. pyrifolia) in border regions, and 8 genotypes of P. ussuriensis as out groups, using simple sequence repeats (SSR) markers. Twenty-eight SSR markers displayed high polymorphism with a total of 202 alleles. Cluster analysis revealed a close genetic relationship between Zangli and sand pears, while population structure analysis revealed that gene flow between the western and eastern side of the Jin-sha River was restricted by geographic isolation. Diversity statistics for five geographic groups implied a migration event from Sichuan and Yunnan Provinces to the border region of western Sichuan, eastern Tibet, and northwestern Yunnan Provinces. Six specific genetic alleles in Zangli pear landraces might be related to their adaption to severe environmental conditions.  相似文献

Mature fruit of ‘Dangshansuli’ pear has yellow-green skin, while its mutant ‘Xiusu’ has russet fruit skin, which is a genetic variation. To explore the mechanism underlying the russet formation, the fruit spot and epidermal structure were observed, the color, texture, and wax and suberin components were evaluated, and the gene expression levels were confirmed. In the present study, the color, texture and fruit spot of the epidermis differed significantly between ‘Dangshansuli’ and ‘Xiusu’ at 25 days after full bloom (DAFB). The cuticular wax components were alkanes, olefins, alkanoic acids, alcohols and terpenes, and the suberin was composed of fatty acid, α,ω-diacids, ω-hydroxy fatty acids, mainly ferulic acid and primary alcohols in the epidermis of ‘Dangshansuli’ and ‘Xiusu’, which exhibited significant differences at most stages of the development of pear fruits. Moreover, the expression levels of genes involved in wax and suberin were consistent with morphological and biochemical analyses. The results indicated that the suberization of epidermal cells occurred when pear fruit was young and that wax and suberin might contribute to the russet formation on the epidermis of ‘Xiusu’, leading to the significant differences in color, texture, fruit spot, and exocarp structure between ‘Dangshansuli’ and ‘Xiusu’ pears.  相似文献

Cacopsylla pyricola (Förster) (Hemiptera: Psyllidae) is an important pest of commercial pear in all pear-growing regions of Iran. In the scope of an integrated pest management, a research was carried out on the impact of treatment with biorational compounds in comparison with conventional chemical insecticides for controlling the pear psyllid. The experiments were done with five treatments consisted of diflubenzuron and lufenuron as biorational insecticides and thiacloprid and diazinon as conventional chemical insecticides and untreated check. The trials were set up in a randomised complete block design. The treatments were replicated four times. Samplings were carried out one day before spraying and 3, 7, 15, 30 and 45 days after spraying through clipping 15 leaves in each replicate and counting the number of pear psyllid live nymphs. Mortality percentage was calculated using Henderson–Tilton formula. The data were subjected to analysis of variance (ANOVA) and the means comparison was performed using Duncan’s multiple range test. The results indicated that the highest mortality in diflubenzuron and lufenuron treatments occurred after 15 days, with 82.09% and 71.01% mortality, respectively. In comparison with conventional chemical insecticides, the efficacy of biorational compounds was higher or not significantly different. The results of the trials are discussed in terms of improving management of the populations of pear psylla.  相似文献

Auxin‐deprived, senescent, suspension‐cultured pear ( Pyrus communis cv. Passe Crassane) fruit cells that can be stimulated to produce ethylene were employed in the search for a possible interdependence between DNA methylation levels and ethylene production. Neither short‐term stimulation of ethylene production by CuCl2, nor longer‐term stimulation by auxin, nor the inhibition of ethylene biosynthesis had a significant effect on the cellular level of DNA methylation. However, short‐term exposure to S‐adenosylhomocysteine enhanced cellular ethylene production and long‐term exposure to azacytidine resulted in the reduction of both DNA methylation levels and stress‐induced ethylene production. These and other correlative findings, or lack thereof, are discussed in the context of ethylene physiology, DNA methylation/demethylation, and epigenetic control.  相似文献

Summary Large numbers of highly viable mesophyll protoplasts were isolated from shoot cultures of the scion cv Passe Crassane and the rootstock genotype Old Home of common pear (Pyrus communis L.). Protoplasts were cultured for both genotypes either as liquid layers or as liquid-over-agar cultures, in ammonium-free MS medium with 0.5 M mannitol, 50 mg/l casein enzymatic hydrolysate (CEH), 2.0 mg/l NAA and 1.0 mg/l BAP, plus either 0.5 mg/l IAA (for Old Home) or 2.0 mg/l IAA (for Passe Crassane). Protoplast microcalli, obtained by day 60 (Passe Crassane) or day 80 (Old Home), were transferred for further growth to ammonium-free MS medium with 2.0 mg/l NAA and 1.0 mg/l BAP. Shoot bud regeneration from the protoplastderived callus was first attempted between 100 (Passe Crassane) and 120 (Old Home) days after protoplast isolation. For Passe Crassane, shoot buds were regenerated (day 130) on a half-strength MS medium with 0.1 mg/l IBA, 0.5 mg/l BAP, 50 mg/l CEH and 20 mg/l Ca-panthotenate. For Old Home, shoot but regeneration only occurred 30 days later and on the same medium as above, which was additionally supplemented with double the concentration of the group B vitamins found in the original MS formulation and 0.05 mg/l GA3. Following micropropagation and in vitro rooting of shoots, the plants were transferred to soil following standard procedures. Trueness-to-type of the regenerated plants was assessed by analysing their leaf isozyme banding profiles (for EST, AP, PRX, SOD, ENP, LAP, PGI, AAT, ADH, MDH and PGM) and comparing them to those corresponding to the original shoots that provided the protoplasts. No differences between the mother shoots and the protoclones were observed for any one of the 11 isozyme systems studied.  相似文献

Women have more body fat than men, but in contrast to the deleterious metabolic consequences of the central obesity typical of men, the pear-shaped body fat distribution of many women is associated with lower cardiometabolic risk. To understand the mechanisms regulating adiposity and adipose tissue distribution in men and women, significant research attention has focused on comparing adipocyte morphological and metabolic properties, as well as the capacity of preadipocytes derived from different depots for proliferation and differentiation. Available evidence points to possible intrinsic, cell autonomous differences in preadipocytes and adipocytes, as well as modulatory roles for sex steroids, the microenvironment within each adipose tissue, and developmental factors. Gluteal-femoral adipose tissues of women may simply provide a safe lipid reservoir for excess energy, or they may directly regulate systemic metabolism via release of metabolic products or adipokines. We provide a brief overview of the relationship of fat distribution to metabolic health in men and women, and then focus on mechanisms underlying sex differences in adipose tissue biology.  相似文献

Because Y-tube olfactometer experiments in the laboratory showed a response of anthocorid bugs to odour fromPsylla-infested leaves, it was of interest to assess its relevance under field circumstances. This was done by measuring the density of predatory bugs on pear trees adjacent toPsylla-infested or control trees that were covered with fine mesh gauze-screens. In this way odours from these caged trees could spread through the screen, while contact with thePsylla prey in the cage was prevented. The density of anthocorid predators around cages with heavily infested trees was significantly higher than around uncaged control trees and around cages containing uninfested or little infested trees. Covering a cage withPsylla-infested trees by an airtight plastic sheet led to an immediate drop in the density of anthocorid predators, whereas removal of the sheet led to predator aggregation again. The results of these field experiments strongly support the hypothesis that anthocorid predators respond to volatile chemicals emanating fromPsylla-infested pear trees.  相似文献

Genomic sequences of the self-incompatibility genes, the S-RNase genes, from two rosaceous species, Japanese pear and apple, were characterized. Genomic Southern blot and sequencing of a 4.5-kb genomic clone showed that the S4-RNase gene of Japanese pear is surrounded by repetitive sequences as in the case of the S-RNase genes of solanaceous species. The flanking regions of the S2- and Sf-RNase genes of apple were also cloned and sequenced. The 5′ flanking regions of the three alleles bore no similarity with those of the solanaceous S-RNase genes, although the position and sequence of the putative TATA box were conserved. The putative promoter regions of the Japanese pear S4- and apple Sf-RNase genes shared a stretch of about 200 bp with 80% sequence identity. However, this sequence was not present in the S2-RNase gene of apple, and thus it may reflect a close relationship between the S4- and Sf-RNase genes rather than a cis-element important in regulating gene expression. Despite the uniform pattern of expression of the rosaceous S-RNase genes, sequence motifs conserved in the 5′ flanking regions of the three alleles were not found, implying that the cis-element controlling pistil specific gene expression also locates at the intragenic region or upstream of the analyzed promoter region.  相似文献

Fruit ripening can be considered as a complex set of biochemical and physiological changes occurring at the end of the developmental stage. Ripe fruit texture notably affects overall quality and consumer appreciation. Excessive softening limits shelf-life and storability, thereby increasing disease susceptibility and economic loss. Fruit softening is a process due to the depolymerisation of different polysaccharide classes, an event controlled by a synergic and coordinated action of several enzymes among which expansins play a fundamental role. To date, six expansin genes are known to be expressed during apple fruit ontogeny, from full bloom up to fruit ripening. We identified a novel expansin apple homolog (Md-Exp7) sharing high sequence similarity with specific-ripening expansin genes of other crops. A functional marker (Md-Exp7SSR) based on an SSR motif located within the untranslated region of the gene was developed and mapped on Linkage Group 1 of the apple and pear genomes in a region where one major apple QTL for fruit firmness had been previously identified. The allelic composition of 31 apple varieties for the SSR marker was associated with differences in fruit softening.  相似文献

Background

Rootstocks play an essential role to determining orchard performance of fruit trees. Pyrus communis and Cydonia oblonga are widely used rootstocks for European pear cultivars. The lack of rootstocks adapted to different soil conditions and different grafted cultivars is widely acknowledged in pear culture. Cydonia rootstocks (clonal) and Pyrus rootstocks (seedling or clonal) have their advantages and disadvantages. In each case, site-specific environmental characteristics, specific cultivar response and production objectives must be considered before choosing the best rootstock. In this study, the influence of three Quince (BA 29, Quince A = MA, Quince C = MC) and a local European pear seedling rootstocks on the scion yield, some fruit quality characteristics and leaf macro (N, P, K, Ca and Mg) and micro element (Fe, Zn, Cu, Mn and B) content of ‘Santa Maria’ pear (Pyrus communis L.) were investigated.

Results

Trees on seedling rootstock had the highest annual yield, highest cumulative yield (kg tree−1), largest trunk cross-sectional area (TCSA), lowest yield efficiency and lowest cumulative yield (ton ha−1) in the 10th year after planting. The rootstocks had no significant effect on average fruit weight and fruit volume. Significantly higher fruit firmness was obtained on BA 29 and Quince A. The effect of rootstocks on the mineral element accumulation (N, K, Ca, Mg, Fe, Zn, Cu, Mn and B) was significant. Leaf analysis showed that rootstocks used had different mineral uptake efficiencies throughout the early season.

Conclusion

The results showed that the rootstocks strongly affected fruit yield, fruit quality and leaf mineral element uptake of ‘Santa Maria’ pear cultivar. Pear seedling and BA 29 rootstock found to be more prominent in terms of several characteristics for ‘Santa Maria’ pear cultivar that is grown in highly calcareous soil in semi-arid climate conditions. We determined the highest N, P (although insignificant), K, Ca, Mg, Fe and Cu mineral element concentrations on the pear seedling and BA 29 rootstocks. According to the results, we recommend the seedling rootstock for normal density plantings (400 trees ha−1) and BA 29 rootstock for high-density plantings (800 trees ha−1) for ‘Santa Maria’ pear cultivar in semi-arid conditions.  相似文献

In pear, fruit ripening is not homogeneous, which leads to problems in harvest and storage management. To identify factors affecting the ripening homogeneity, structural, biochemical and molecular parameters were investigated. Fruits were sampled from trees trained with three different systems, spindle, V-shaped and bi-axis, and were further grouped on the basis of canopy position (top or bottom) and branch type (short-old-spurs, 3-year-old or older branches, 2-year-old branches and twigs). This study confirmed that the position in the canopy plays a crucial role in fruit ripening in V-shaped and bi-axis training systems. Fruit on the spindle training system was unaffected by the position of the canopy, albeit showing a large variability within the fruit of each part of the canopy. Such variability is much lower in the bi-axis and V-shaped systems. The spindle training system had no similarity in homogeneity indices between the molecular and physiological measurements. Partial least square discriminate analysis (PLS-DA) showed differences in training system, branch type and canopy position separated by the different physiological and molecular parameters. Short-old-spurs and twigs had the highest homogeneity and least variability in all training systems and branch types. In addition, partial least square path modelling (PLS-PM) was able to identify a high correlation between physiological measurements and ripening compared to that of molecular data.  相似文献

Scab, caused by the ascomycete fungus Venturia pirina, leads to severe damage on European pear varieties resulting in a loss of commercial value and requiring frequent use of fungicides. Identifying scab resistance genes, developing molecular markers linked to these genes and establishing marker-assisted selection would be an effective way to improve European pear breeding for scab resistance. Most of the European pear cultivars (Pyrus communis) are currently reported to be sensitive. The pear cultivar ‘Navara’ was shown to carry a major scab resistance gene whose phenotypic expression in seedling progenies was a typical stellate necrosis symptom. The resistance gene was called Rvp1, for resistance to V. pirina, and was mapped on linkage group 2 of the pear genome close to microsatellite marker CH02b10. This genomic region is known to carry a cluster of scab resistance genes in apple indicating a first functional synteny for scab resistance between apple and pear.  相似文献

The genomic clone encoding the pistil-specific thaumatin/PR5-like protein (PsTL1) was isolated from Japanese pear (Pyrus serotina). Sequence analysis showed that the genomic clone contained the 5-flanking sequence of 2.4 kb, the 3-flanking sequence of 648 bp and the coding region interrupted by a intron of 351 bp. A sequence motif conserved in some pistil self-incompatibility gene promoters of solanaceous and brassicaceous species was located in the 5-flanking region of the PsTL1 gene. The 2.4 kb 5-flanking region was fused to the GUS coding sequence and transferred to tobacco. Transgenic tobacco showed GUS activity in pistil and, at low level, in anther, but not in other floral organs and leaf. Histochemical analysis localized GUS activity to stigma, transmitting tissue, anther and pollen of transgenic tobacco.  相似文献

The effect of various basal salts media, containing different nitrogen levels on in vitro adventitious shoot regeneration from leaf explants of Louise Bonne Panachee and Seckel pear (Pyrus communis L.) were investigated. Among the different basal salt formulae tested, Nitsch (1969) gave significantly better regeneration in most of the experiments. Shoot regeneration was altered with different NH4 +-N/NO3 -N ratios. The best regeneration was obtained when NH4 +:NO3 was either 1:2 or 1:3 regardless of overall N concentration. In addition, these data show that NH4 + was essential for adventitious shoot regeneration from pear leaf explants on White's (1943) medium.  相似文献
