Growth of Botryococcus braunii (race 'A') and production of its constituents viz, hydrocarbon, carbohydrate, fatty acid, and carotenoids were influenced by different levels of salinity. Under salinity at 34 mM and 85 mM, 1.7-2.25-fold increase in the relative proportion of palmitic acid and two fold increase in oleic acid were observed. A twofold increase in carotenoid content was noticed at 85 mM salinity with lutein (75% of total carotenoid) as the major carotenoid followed by beta-carotene. The increase in biomass yields and changes in other constituents indicated the influence of salinity and the organism's adaptability to the tested levels of salinity (17 mM to 85 mM).  相似文献

A laboratory study was conducted on the removal of nitrogen and phosphorus from piggery wastewater during growth of Botryococcus braunii UTEX 572, together with measurements of hydrocarbon formation by the alga. The influence was tested of the initial nitrogen and phosphorus concentration on the optimum concentration range for a culture in secondarily treated piggery wastewater. A high cell density (> 7 g L–1 d. wt) was obtained with 510 mg L–1 NO3-N. Growth increased with nitrogen concentration at the basal phosphorus concentration (14 mg P L–1). The growth rate was nearly independent ( = 0.027 0.030 h–1) of the initial phosphate concentration, except under conditions of phosphate deficiency ( = 0.019 h–1). B. braunii grew well in piggery wastewater pretreated by a membrane bioreactor (MBR) with acidogenic fermentation. A dry cell weight of 8.5 mgL–1 and hydrocarbon level of 0.95 gL–1 were obtained, and nitrate was removed at a rate of 620 mg NL–1. These results indicate that pretreated piggery wastewater provides a good culture medium for the growth and hydrocarbon production by B. braunii.  相似文献

Of various methods for lipid recovery in Botryococcus braunii UTEX 572, the most effective method was disruption of the cells with a bead-beater followed by extraction with chloroform/methanol (2:1, v/v). This gave a lipid content of 28.6% of dry wt. There was a significant relationship between in vivo fluorescence of cells stained with Nile Red and lipid content in B. braunii determined gravimetrically (r2 = 0.997). This suggested that the Nile Red staining as a rapid method was as good as the gravimetric method commonly used for lipid determination which requires toxic solvents and considerable time-consuming manipulations. © Rapid Science Ltd. 1998  相似文献

Flocculating activity was determined to evaluate the effective harvesting method and an optimal growth stage for the recovery of the green alga Botryococcus braunii growing in batch cultures. Flocculating activity was highest at 2 weeks of incubation, regardless of harvesting methods. The degree of flocculation was different with harvesting methods and growth stages. A high pH value (pH 11) was more effective for flocculation than treatment with aluminium sulphate or a microbial flocculant, Pestan, until the third week of incubation. The lipid content of the algae on a dry weight basis was unaffected by harvesting methods. These results indicate that the most effective harvesting method is to adjust pH to 11 after 2 weeks of incubation.  相似文献

The various strains of the hydrocarbon-rich alga Botryococcus braunii (A race) examined up to now produce the Z-isomers of the following dienes: tricosa-1,14-diene, pentacosa-1,16-diene, heptacosa-1,18-diene, nonacosa-1,20-diene and hentriaconta-1,22-diene. A C29 triene was also detected in these strains, but its structure was not fully elucidated. In the present work, spectroscopic data and oxidative degradations, established the structure of the C29 trienic hydrocarbon. These techniques were also used to examine the dienic series of a new A strain, collected from nature and laboratory grown. In addition to the usual Z-dienes, E-isomers of tricosa-1,14-diene, pentacosa-1,16-diene, heptacosa-1,18-diene and nonacosa-1,20-diene, were characterized.  相似文献

Contamination by other algal species is a major problem in the culture of the oil-producing green alga Botryococcus braunii (Bb). In this study, the population interactions between Bb and two small green algae, Chlorella vulgaris (Cv) and Chlamydomonas reinhardtii (Cr), were separately investigated in two controlled experiments. Filtrates of Cv or Cr significantly stimulated the growth of Bb in the first 9–11 days when the filtrates were derived from a low- or medium-density algal culture. However, filtrates of Cv and Cr from the high-cell-density cultures significantly inhibited Bb growth from day 11 and 7, respectively. In Bb–Cv competition, the growth of Bb was significantly inhibited by Cv at the biomass ratio of Bb/Cv?=?1:1. However, when the Cv component reduced to 0.5 (Bb/Cv?=?1:0.5) or 0.25 (Bb/Cv?=?1:0.25), the growth of Bb was faster than that in the control in the first 13 days. Similarly, in Bb–Cr competition, the biomass ratios of 1:0.5 and 1:0.25 (Bb/Cr) favoured the growth of B. braunii in the first 15 days, but the growth of Bb decreased in the Bb/Cr ratio of 1:1 from day 15. By contrast, the growth of Cv and Cr were not affected by the presence of Bb in all biomass ratios. These results suggest that Cv and Cr could impact the growth of Bb by metabolites in the culture medium, and when the biomass of Bb is two to four times that of other algae, this oily alga becomes competitive over other algal species. This study reveals the mechanism of population dynamics of this oily alga competing with small and fast-growing algae. The results are applicable in reducing algal competition and regulating species succession between slow and fast-growing algae species.  相似文献

The outer walls of the green alga Botryococcus braunii (main sites of hydrocarbon production and accumulation) show a complex constitution. They comprise a biopolymer highly resistant to non-oxidative degradation. The resistant polymer accounts for ca 9% of the cell dry wt and appears, along with hydrocarbons, as one of the major constituents of the alga. In addition to chemical resistance, B. braunii polymer exhibits other properties: mode of deposition and fluorescence, often used to identify sporopollenins. (Class of wall components generally regarded as originating from polymerization of carotenoid derivatives.) Nevertheless further studies, using IR spectroscopy and high resolution 13C NMR of solids, along with determination of elemental composition and unsaturation levels, indicate that the bulk of the resistant polymer from B. braunii outer walls does not derive from carotenoids; accordingly it cannot be considered, in this respect, as a sporopollenin. In fact the information obtained on the structure of this important constituent of the alga is consistent with its formation via oxidative polymerization of B. braunii dienic hydrocarbons.  相似文献

We tested for chemical reagents that would be useful in preparing a large number of vital single cells from colonial Botryococcus braunii B-race, variety Showa. Among the 18 reagents assayed, glycerol and erythritol showed the highest potency for releasing single cells. Incubation in medium containing these reagents released 40–50 % single cells in 15 min. Fluorescent staining with Nile red revealed that except for the cap-like structures the released single cells were free of hydrocarbon oils that accumulated in the extracellular matrix where the single cells were embedded. However, to maintain the prepared single cells in vital condition, they must be maintained at a high concentration (>2?×?107 cells/ml); at low concentrations, they rapidly lost chlorophyll and get disrupted. In contrast to the above results obtained using B-race, Showa, single cells prepared from A-race varieties survived even at low cell concentrations.  相似文献

Four new strains of Botryococcus braunii were isolated from Japanese waters and cultured under defined conditions. Their hydrocarbon content and composition were analyzed and compared with those of the Darwin and Berkeley strains. The Yamanaka strain produced only alkadienes characteristic of the A race, whereas the others, the Yayoi, Kawaguchi-1 and -2 strains as well as the Darwin and Berkeley strains, produced botryococcenes peculiar to the B race. The hydrocarbon content of the Yamanaka strain was 16.1 % dry weight and that of the B race strains ranged from 9.7 to 37.9%. Botryococcene composition of the Japanese strains differed from each other as well as from the Darwin and Berkeley strains. More than 50% of the hydrocarbons in the Yayoi, Darwin, and Berkeley strains were composed of C34H58, but the main components were different from one another as isomers. The Kawaguchi-1 and -2 strains did not have a high level of C34 botryococcenes, C32 ones being the main components. In these strains significant amounts of squalene-related compounds were detected.  相似文献

通过对布朗葡萄藻分别在Chu13、Chu13×2和BG-11培养基中培养结果的比较,发现在气升式光照生物反应器中Chu13培养基最有利于布朗葡萄藻的生长和烃的合成,培养15d后,其生物量和粗烃质量分数分别为1.82g/L和58.7%;棕榈酸、油酸和亚麻酸是布朗葡萄藻的主要脂肪酸组成,Chu13培养获得的藻体不饱和脂肪酸比例最高。Chu13培养基中布朗葡萄藻代谢规律的研究表明:粗烃含量随着生物量的增加而逐渐增大,15d后粗烃产量达到最大值1.07g/L,不同生长周期烃的组成保持一致,布朗葡萄藻的烃主要由C33H56和C34H58组成;在布朗葡萄藻生长周期中,不饱和脂肪酸的比例显著上升,培养15d达到64%以上。  相似文献

The B race of the green alga Botryococcus braunii is characterized by the production of large amounts of botryococcenes, i.e. triterpenoid hydrocarbons of general formula CπH2π-109 n= 30–37. The axenic strain used in this work produces botryococcenes ranging from C30 to C34 when fast growth is promoted by air-lift. Sequential extraction of hydrocarbons with solvents showed that botryococcenes accumulate in two distinct sites: externally in the successive outer walls forming a dense matrix and internally, probably in cyctoplasmic inclusions. Moreover, chase experiments after feeding the algae with sodium [1,2-14C]acetate, and feeding experiments with L-[Me-14C]methionine established the existence of an excretory process from the cells towards the matrix. The results of the radio GC analyses of the botryococcenes synthesized during the feeding experiments provided good evidence to show that the C30 botryococcene is the precursor of all the higher hydrocarbons, and that each intermediate botryococcene C31-C33 is the precursor of its next highest homologue. L-Methionine acts as the methyl donor in the methylation process, leading from the C30 precursor to the botryococcene family. The 13C NMR spectra of the botryococcenes produced when the algae were fed with L-[Me-13C]methionine indicate that the methylation takes place on the C30 backbone in positions 37, 16 and 20.  相似文献

Botryococcus braunii is a colonial green microalga with recognized potential to synthesize lipids and hydrocarbons for biofuel production. Besides this ability, this microalga also produces exopolysaccharides (EPS). Nevertheless, there are few reports about their biotechnological aspects and industrial applications. In this study, the effect of the nutritional conditions was examined by using two different culture media (BG11 and D medium). To our knowledge, the latter has not been reported before for culturing B. braunii. After 49 days of incubation, the final production of EPS was found to be statistically higher (P < 0.05) in the D medium (0.549?±?0.044 g L?1) than in BG11 (0.336?±?0.009 g L?1). On the contrary, the biomass production was found to be higher in BG11 (1.019?±?0.051 g L?1) than in the D medium (0.953?±?0.056 g L?1). However, this difference was not statistically significant. The difference in salinity and nitrogen concentration between both media is suggested as the main factor involved in the EPS and biomass results. FTIR spectra of B. braunii EPS from both media revealed presence of uronic acids and absence of amino and sulfate groups. Despite the similarity between both spectra, there were some different signals (at 1,921.52 and 720.60 cm?1) which may mean a difference in glycosyl composition.  相似文献

Glutamine-synthetase (GS; EC 6.3.1.2) activity and protein levels were measured in crude extracts from Monoraphidium braunii Näegeli, strain 202-7d, cultures grown under different nitrogen sources. Only ammonium and l-glutamine promoted a partial enzyme inactivation, which, in the case of l-glutamine, was accompanied by a significant repression of GS. Methionine sulfoximine (MSX), a strong inhibitor of GS, produced a drastic inactivation of GS which was concomitant with a marked increase in GS protein as measured by rocket immunoelectrophoresis. Such an increase was prevented in the presence of cycloheximide. The effect of the l-glutamine analog on GS activity and protein was partially inhibited if l-glutamine was also added to cell cultures, possibly indicating competition in the transport of these two substances. In addition, the effects of MSX were reversed after longer times when cultures were treated with smaller concentrations of inhibitor. Treatment of cell cultures with azaserine, a specific inhibitor of glutamate synthase, the second enzyme acting in the ammonium assimilation pathway, promoted a strong GS inactivation and a partial repression of this enzyme, which paralleled a specific increase in the intracellular pools of glutamine High-performance liquid chromatography measurements of intracellular amino-acid concentrations showed that glutamine levels correlated negatively with GS concentration. Six novel tetraterpenoid ethers, lycopanerols B-G, were isolated from lipidic extracts of the green microalga Botryococcus braunii (L race), along with a series of phytyl esters and alpha- and beta-tocopherols. The structures of the compounds were determined by means of spectral analyses including 2D NMR techniques. A biogenetic relationship is proposed between lycopanerols and lycopadiene, the acyclic diunsaturated tetraterpenoid hydrocarbon synthesized by the alga.  相似文献

对烟道气培养产烃葡萄藻的可行性进行了研究,重点考察了SO2和NOx的水溶形态对葡萄藻的影响。结果表明,亚硫酸氢盐浓度低于0.8mmol/L时,对葡萄藻生长没有明显抑制作用,可以提供葡萄藻生长的硫源,但高浓度(大于2mmol/L)时抑制细胞生长;当亚硝酸盐浓度小于8mmol/L时,可以作为葡萄藻生长的唯一氮源,亚硝酸盐的去除主要是由微藻利用所致。当起始浓度为2mmol/L和4mmol/L时,亚硝酸盐的去除率分别为100%和99.7%。  相似文献
