Effects of methyl jasmonate (MeJA) on floret opening were investigated in male-sterile and male-fertile rice ( Oryza sativa subsp. indica ). Excised or intact panicles with several top florets flowered one day before the experiments were soaked in MeJA, salicylic acid (SA), 24-epibrassinolide (24-epiBL) or control solution for 2 min. MeJA at the concentration of 4 mmol/L significantly promoted rice floret opening; the response of floret opening of male-sterile rice to MeJA was more sensitive than that of male-fertile rice (such as maintenance line and conventional variety). The percentage of finally opened-floret per panicle of Zhenshan 97A (cytoplasmic male sterile line) or Pei'ai 64S (thermo-sensitive genic male sterile line) treated with MeJA was over 40% in the day of experiment, that of Zhenshan 97B (maintenance) and Pei'ai 64 (conventional variety) was (21±1.5)% and (25±2.1)%, respectively; floret opening of intact Pei'ai 64S panicles in the field could also be induced by 2-mmol/L MeJA. The percentage of opened floret in all control panicles was 0% or less than 5% in the day of experiment. The percentage of fertilized-grain of Zhenshan 97A and Longtepu A could be increased over 24% by spraying panicles with 2-mmol/L MeJA solution (circa 6.0 μmol per panicle). The lag time (time from the finishing of treatment to first floret opening) of male sterile line responsing to MeJA was 10 min shorter than that of male fertile line. MeJA induction on the rice floret opening could markedly be inhibited by SA solution at the concentration of 1 mmol/L, and the inhibition of SA on MeJA induction on floret opening of male-sterile rice could be nullified by MeJA treatment again. MeJA effect on floret opening of male-sterile rice could be clearly synergized by pretreatment with 24-epibrassinolide (24-epiBL).  相似文献

The effects of benzyladenine (BAP), kinetin (KIN), zeatin (ZEA), isopentenyladenine (2iP), and thidiazuron (TDZ) were studied on in vitro growth of rudimentary embryos of Ilex paraguariensis St. Hil. Heart stage zygotic embryos were removed from seeds of immature, light green fruits and cultured aseptically on quarter-strength Murashige and Skoog medium containing 3% sucrose, 0.65% agar, and supplemented with or without three concentrations of BAP, KIN, ZEA, 2iP, or TDZ. Cultures were incubated in darkness at 27 ± 2°C. Media containing 4.4 × 10−6 m BAP, 4.6 × 10−6 m KIN, or 4.9 × 10−6 m 2iP were totally ineffective in inducing embryo growth after culture for 28 days. However, lower concentrations of these compounds (4.4 × 10−8 m BAP, 4.6 × 10−8 m KIN, 4.5 × 10−8 m ZEA, or 4.9 × 10−8 m 2iP) promoted embryo growth. TDZ at 9.9 × 10−9 m, 9.9 × 10−8 m, or 9.9 × 10−7 m induced embryo growth at similar rates. Brush border membrane vesicles, BBMV, from eel intestinal cells or kidney proximal tubule cells were prepared in a low osmolarity cellobiose buffer. The osmotic water permeability coefficient P f for eel vesicles was not affected by pCMBS and was measured at 1.6 × 10−3 cm sec−1 at 23°C, a value lower than 3.6 × 10−3 cm sec−1 exhibited by the kidney vesicles and similar to published values for lipid bilayers. An activation energy E a of 14.7 Kcal mol−1 for water transport was obtained for eel intestine, contrasting with 4.8 Kcal mol−1 determined for rabbit kidney proximal tubule vesicles using the same method of analysis. The high value of E a , as well as the low P f for the eel intestine is compatible with the absence of water channels in these membrane vesicles and is consistent with the view that water permeates by dissolution and diffusion in the membrane. Further, the initial transient observed in the osmotic response of kidney vesicles, which is presumed to reflect the inhibition of water channels by membrane stress, could not be observed in the eel intestinal vesicles. The P f dependence on the tonicity of the osmotic shock, described for kidney vesicles and related to the dissipation of pressure and stress at low tonicity shocks, was not seen with eel vesicles. These results indicate that the membranes from two volume transporter epithelia have different mechanisms of water permeation. Presumably the functional water channels observed in kidney vesicles are not present in eel intestine vesicles. The elastic modulus of the membrane was estimated by analysis of swelling kinetics of eel vesicles following hypotonic shock. The value obtained, 0.79 × 10−3 N cm−1, compares favorably with the corresponding value, 0.87 × 10−3 N cm−1, estimated from measurements at osmotic equilibrium. The mechanical properties of brush border membrane vesicles, BBMV, from rabbit kidney proximal tubule cells, were studied by measuring the initial and final equilibrium volumes of vesicles subjected to different osmotic shocks, using cellobiose as the impermeant solute in the preparation buffer. An elevated intracellular hydrostatic pressure was inferred from osmotic balance requirements in dilute solutions. For vesicles prepared in 18 and 85 mosm solutions, these pressures are close to 17 mosm (290 mm Hg). The corresponding membrane surface tension is 6.0 × 10−5 N cm−1 while the membrane surface area is expanded by at least 2.2%. When these vesicles are exposed to very dilute solutions the internal hydrostatic pressure rises to an estimated 84 mosm (1444 mm Hg) just prior to lysis. The corresponding maximal surface tension (pre-lysis) is 18.7 × 10−5 N cm−1, and the maximal expansion of membrane area is 6.8%. Pre-matured florets of Benibana, a cultivar of saffron thistle (Carthamus tinctorius L.) was irradiated with UV-B (280–320 nm) or UV-C (254 nm) light for 48 h at 23±1 °C and the influence of UV-light on carthamin accumulation and floret elongation was investigated. UV-C light enhances carthamin accumulation most prominently, showing a specific value of 52.3 nmol carthamin·dm−3·h−1·25μm−2 (13.9 times of control), while it restricts floret elongation by a light-suppression manner (net elongation: 0.058 mm·h−1, one ninth of control). UV-B light is also promotive for the red colour appearance (25.0 nmol carthamin·dm−3·h−1·25 μm−2, 6.7 times of control) with suppressing floret elongation (net elongation: 0.17 mm·h−1, one third of control). Heterogeneous productivity of carthamin was seen in floret tissues after continuous treating under UV-C light. Carthamin accumulation, heterogeneous carthamin productivity and decrease of floret elongation restraint under UV-lights are discussed.  相似文献

Indole-3-Acetic Acid Control on Acidic Oat Cell Wall Peroxidases   总被引:2,自引:0,他引:2  
Incubation of oat coleoptile segments with 40 μm indoleacetic acid (IAA) induced a decrease of 35–60% in peroxidase activity at the cell wall compartment. Treatment with IAA also produced a similar decrease in the oxidation of NADH and IAA at the cell wall. Isoelectric focusing of ionic, covalent, and intercellular wall peroxidase fractions showed that acidic isoforms (pI 4.0–5.5) were reduced preferentially by IAA treatment. Marked differences were found between acidic and basic wall isoperoxidases in relation to their efficacy in the oxidation of IAA. A peroxidase fraction containing acidic isoforms oxidized IAA with a V max/s0.5 value of 2.4 × 10−2 min−1· g fw−1, 4.0 times higher than that obtained for basic peroxidase isoforms (0.6 × 10−2 min−1· g fw−1). In contrast, basic isoforms were more efficient than acidic isoperoxidases in the oxidation of coniferyl alcohol or ferulic acid with H2O2 (5.6 and 2.1 times, respectively). The levels of diferulate and lignin in the walls of oat coleoptile segments were not altered by treatment with IAA. The decrease in cell wall peroxidase activity by IAA was related more to reduced oxidative degradation of the hormone than to covalent cell wall cross-linking. Hyperthermia induces transient changes in [Na+] i and [K+] i in mammalian cells. Since Cl flux is coupled with Na+ and K+ in several processes, including cell volume control, we have measured the effects of heat on [Cl] i using the chloride indicator, MQAE, with flow cytometry. The mean basal level of [Cl] i in Chinese hamster ovary cells was 12 mm. Cells heated at 42.0° or 45.0°C for 30 min had about a 2.5-fold increase in [Cl] i above unheated control values when measured immediately after heating. There was about a 3-fold decrease in [Na+] i under the same conditions, as measured by Sodium Green. The magnitude of the increase in [Cl] i depended upon time and temperature. The [Cl] i recovered in a time-dependent fashion to control values by 30 min after heating. When cells were heated at 45.0°C for 30 min in the presence of 1.5 mm furosemide, the heat-induced [Cl] i increase was completely blocked. Since furosemide inhibits the Na+/K+/2Cl cotransporter, Cl channels, and even ClHCO3 exchange, these ion transporters may be involved in the heat-induced increase in [Cl] i . Changes in volume of intestinal brush border membrane vesicles of the European eel Anguilla anguilla were measured as vesicles were exposed to media with different osmotic pressures. Preparing the vesicles in media of low osmotic pressure allowed the effects of a small hydrostatic pressure to become a significant factor in the osmotic equilibration. By applying LaPlace's law to relate pressure and volume and assuming a linear relation between membrane tension and area expansion, we estimate an initial membrane tension at 4.02 × 10−5 N cm−1 and an area compressibility elastic modulus at 0.87 × 10−3 N cm−1. The elastic modulus estimate falls in the low range of values reported for membranes from other tissues in other species. This lower modulus quantitatively accounts for why eel intestinal vesicles show measurable changes in volume in hypotonic media while rabbit kidney vesicles do not. Cell-volume changes induced by terbutaline (a specific β2-agonist) were studied morphometrically in rat fetal distal lung epithelium (FDLE) cells. Cell-volume changes qualitatively differed with the concentration of terbutaline. Terbutaline of 10−10–10−8 m induced transient cell swelling. Terbutaline of 10−7 m induced transient cell swelling followed by slow cell shrinkage. Terbutaline of 10−6–10−5 m induced rapid cell shrinkage followed by slow cell shrinkage. Terbutaline of 10−3 m induced transient cell shrinkage; then cell volume oscillated during stimulation. Benzamil of 10−6 m suppressed the cell swelling induced by 10−10–10−8 m terbutaline and quinine of 10−3 m inhibited the cell shrinkage induced by 10−6–10−5 m terbutaline. These results suggest that cell swelling would be induced by NaCl influx and the cell shrinkage is by KCl efflux. Dibutyryl cyclic AMP (DBcAMP) also induced similar cell-volume changes over a wide range of concentrations (10−9–10−3 m): a low concentration induced transient cell swelling; a high concentration, rapid and slow cell shrinkage. Forskolin (10−4 m), like terbutaline (10−5 m), induced rapid cell shrinkage followed by slow cell shrinkage, and this decrease in the cell volume was enhanced by the presence of benzamil. On the other hand, cell shrinkage was induced by ionomycin (even low concentration; 3 × 10−10 m ionomycin), and after that cell volume remained at a plateau level. Removal of extracellular Ca2+ abolished the cell swelling caused by terbutaline of 10−10–10−8 m. With removal of extracellular Ca2+, the initial, rapid cell shrinkage induced by 10−5 m terbutaline became transient, but we still detected slow cell shrinkage similar to that in the presence of extracellular Ca2+. Overall, at low concentrations (10−10–10−8 m), terbutaline induced benzamil-sensitive cell swelling in FDLE cells, which was cAMP- and Ca2+-dependent; high concentrations (≥−6) induced quinine-sensitive rapid cell shrinkage, which was Ca2+-dependent; high concentrations (≥−7) induced slow cell shrinkage, which was cAMP-dependent. These findings suggest that terbutaline regulates cell volume in FDLE cells by cytosolic cAMP and Ca2+ through activation of Na+ and K+ channels. The gating cycle of CFTR (Cystic Fibrosis Transmembrane conductance Regulator) chloride channels requires ATP hydrolysis and can be interrupted by exposure to the nonhydrolyzable nucleotide AMP-PNP. To further characterize nucleotide interactions and channel gating, we have studied the effects of AMP-PNP, protein kinase C (PKC) phosphorylation, and temperature on gating kinetics. The rate of channel locking increased from 1.05 × 10−3 sec−1 to 58.7 × 10−3 sec−1 when AMP-PNP concentration was raised from 0.5 to 5 mm in the presence of 1 mm MgATP and 180 nm protein kinase A catalytic subunit (PKA). Although rapid locking precluded estimation of P o or opening rate immediately after the addition of AMP-PNP to wild-type channels, analysis of locking rates in the presence of high AMP-PNP concentrations revealed two components. The appearance of a distinct, slow component at high [AMP-PNP] is evidence for AMP-PNP interactions at a second site, where competition with ATP would reduce P o and thereby delay locking. All channels exhibited locking when they were strongly phosphorylated by PKA, but not when exposed to PKC alone. AMP-PNP increased P o at temperatures above 30°C but did not cause locking, evidence that the stabilizing interactions between domains, which have been proposed to maintain CFTR in the open burst state, are relatively weak. The temperature dependence of normal CFTR gating by ATP was strongly asymmetric, with the opening rate being much more temperature sensitive (Q 10= 9.6) than the closing rate (Q 10= 3.6). Stem elongation in Fuchsia × hybrida was influenced by cultivation at different day and night temperatures or in different light qualities. Internode elongation of plants grown at a day (25°C) to night (15°C) temperature difference (DIF+10) in white light was almost twofold that of plants grown at the opposite temperature regime (DIF−10). Orange light resulted in a threefold stimulation of internode elongation compared with white light DIF−10. Surprisingly, internode elongation in orange light was similar for plants grown at DIF−10 and DIF+10. Flower development was accelerated at DIF−10 compared with DIF+10 in both white and orange light. To examine whether the effects of DIF and light quality on shoot elongation were related to changes in gibberellin metabolism or plant sensitivity to gibberellins (GAs), the stem elongation responses of paclobutrazol-treated plants to applied gibberellins were determined. In the absence of applied gibberellins paclobutrazol (>0.32 μmol plant−1) strongly retarded shoot elongation. This inhibition was nullified by the application of about 10–32 nmol of GA1, GA4, GA9, GA15, GA19, GA20, GA24, or GA44. The results are discussed in relation to possible effects of DIF and light quality on endogenous gibberellin levels and gibberellin sensitivity of fuchsia and their effects on stem elongation. Permeability of Boric Acid Across Lipid Bilayers and Factors Affecting It   总被引:13,自引:0,他引:13  
Boron enters plant roots as undissociated boric acid (H3BO3). Significant differences in B uptake are frequently observed even when plants are grown under identical conditions. It has been theorized that these differences reflect species differences in permeability coefficient of H3BO3 across plasma membrane. The permeability coefficient of boric acid however, has not been experimentally determined across any artificial or plant membrane. In the experiments described here the permeability coefficient of boric acid in liposomes made of phosphatidylcholine was 4.9 × 10−6 cm sec−1, which is in good agreement with the theoretical value. The permeability coefficient varied from 7 × 10−6 to 9.5 × 10−9 cm sec−1 with changes in sterols (cholesterol), the type of phospholipid head group, the length of the fatty acyl chain, and the pH of the medium. In this study we also used Arabidopsis thaliana mutants which differ in lipid composition to study the effect of lipid composition on B uptake. The chs1-1 mutant which has lower proportion of sterols shows 30% higher B uptake compared with the wild type, while the act1-1 mutant which has an increased percentage of longer fatty acids, exhibited 35% lower uptake than the wild type. Lipid composition changes in each of the remaining mutants influenced B uptake to various extents. These data suggest that lipid composition of the plasma membrane can affect total B uptake. Developing high-yielding rice genotypes is decisive to ensure global food security with current population growth and the threat of environmental pressures. Cytoplasmic male sterility (CMS) system provides a valuable approach for commercial exploitation of heterosis and producing high-yielding and quality hybrid rice. Three CMS lines and ten diverse restorers were crossed using line × tester mating design. The obtained thirty F1 hybrids and their thirteen parents were evaluated. Yield traits as well as certain floral traits characters that influence the efficiency of crossing and hybrid seed production as the duration of floret opening (min), stigma exsertion (mm), stigma length (mm), opening floret angle, and anther length (mm) were assessed. Highly significant variations were detected among parents, crosses, and parents vs. crosses for all the studied traits. The CMS line L2 and the restorer T5 were determined as good combiners for stigma exsertion, stigma length, opining floret angle, and duration of floret opening. Besides, the hybrids L1 × T1, L1 × T3, L2 × T2, L2 × T5, L3 × T4, L3 × T5, and L3 × T9 exhibited positive SCA effects for most floral traits. Moreover, the CMS lines L1 and L3 as well as the restorers T1, T2, T3, T6, and T9 were identified as good general combiners for grain yield and certain related traits. The hybrids L1 × T1, L1 × T5, L1 × T7, L2 × T3, L2 × T4, L2 × T5, L2 × T10, L3 × T1, L3 × T2, and L3 × T6 displayed positive SCA effects for grain yield and one or more of its attributes. Both additive and non-additive gene effects were involved in the governing inheritance of all evaluated traits. The biochemical variations among the certain evaluated genotypes were further studied. The esterase and peroxidase isozymes were applied for verifying the genetic diversity at the protein level among the used CMS lines, restorers, and their crosses. All the applied isozymes displayed polymorphism for the parents and their crosses. The banding pattern and intensity differences provided accurate results on the reliable variability among the tested genotypes.  相似文献

Summary. Potassium pyroantimonate precipitation was used to locate loosely bound calcium in rice (Oryza sativa L.) lodicules before and after anthesis, and flowering of panicles was accelerated by treatment with methyl jasmonate. From 1 day to 4 h before anthesis, the number of calcium precipitates in the cell walls and vacuole membranes decreased gradually, whereas they increased remarkably in the cytoplasm and nucleolus. At the beginning of anthesis, the number of calcium granules in lodicules reduced sharply, but there was a large accumulation of flocculent precipitates in the vacuoles. After anthesis, the flocculent precipitates decreased in number until they disappeared, whereas the granular precipitates started to accumulate once again. The rice florets treated with 2 mM methyl jasmonate were induced to open within 10–30 min and they then closed 0.5–1 h later. The nucleolus, cytoplasm, and vacuole membrane of the lodicule cells contained many calcium granules during flowering, although the cell walls lacked calcium. At 1 h after treatment, the number of calcium granules had decreased, while flocculent precipitates were regularly observed in the nondegenerated cells. At 6 h after treatment, calcium grains started to reappear in the cell walls. These changes in calcium precipitates before and after anthesis indicate that the opening and closing of florets correlates with the calcium level in lodicule cells. In addition, excised panicles, with florets judged to be nearing anthesis, were soaked in 2–200 mM EGTA solution for 2 min after treatment with 2 mM methyl jasmonate. The results indicate that EGTA had an antagonistic effect on the methyl jasmonate-induced floret opening in rice. We have used current/voltage (I/V) analysis to investigate the role played by extracellular mucilage in the cellular response to osmotic shock in Lamprothamnium papulosum. Cells lacking extracellular mucilage originated in a brackish environment (1/3 seawater). These were compared, first with cells coated with thick (∼50 μm) extracellular mucilage, collected from a marine lake, and second, with equivalent mucilaginous marine cells, treated with heparinase enzyme to disrupt the mucilage layer. Histochemical stains Toluidine Blue and Alcian Blue at low pH identified the major component of the extracellular mucilage as sulfated polysaccharides. Treating mucilage with heparinase removed the capacity for staining with cationic dyes at low pH, although the mucilage was not removed, and remained as a substantial unstirred layer. Cells lacking mucilage responded to hypotonic shock with depolarization (by ∼95 mV), cessation of cyclosis, due to transient opening of Ca2+ channels, and opening of Ca2+-activated Cl channels and K+ channels. Cell conductance transiently increased tenfold, but after 60 min was restored to the conductance prior to hypotonic shock. Mucilaginous cells depolarized by a small amount (∼18 mV), but Ca2+ channels failed to open in large enough numbers for cyclosis to cease. Likewise most Ca2+-activated Cl channels failed to open and conductance increased only ∼1.2-fold above the prehypotonic level. After 60 min conductance was less than the conductance prior to hypotonic shock. Heparinased mucilaginous cells recovered several aspects of the hypotonic response in cells lacking mucilage. These cells depolarized (by ∼103 mV); cyclosis ceased, indicating that Ca2+ channels had opened, and conductance increased to ∼4 times the value prior to hypotonic shock, indicating that Ca2+-activated Cl channels opened. However, after 60 min, these cells had neither restored membrane potential (and remained at positive values), nor decreased their conductance. It was not possible to determine whether K+ channels had opened. The heparinased cells recovered the normal hypotonic response of mucilaginous cells when heparinase was washed out. Apical seawater cells, which lacked mucilage, were unaffected by heparinase treatment. The results demonstrate that the presence of extracellular sulfated polysaccharide mucilage impacts upon the electrophysiology of the response to osmotic shock in Lamprothamnium cells. The effect of cyanide (CN) on voltage-activated or cAMP-induced passive chloride conductance (G Cl ) was analyzed in isolated toad skin. Comparatively low concentrations of CN inhibited G Cl almost completely and fully reversibly, regardless of whether it was applied from the mucosal or serosal side. The IC50 was 180 ± 12 μm for voltage-activated G Cl and 305 ± 30 μm for the cAMP-inducted conductance. At [CN] <100 μm, the initial inhibition frequently declined partly in the continuous presence of CN. Inhibition was independent of the presence of Ca2+. Inhibition was stronger at more alkaline pH, which suggests that dissociated CN is the effective inhibitor. The onset of the inhibition of voltage-activated or cAMP-induced G Cl by CN occurred with half-times of 34 ± 10 sec, whereas reversibility upon washout was twice as fast (18 ± 7 sec). If [CN] <200 μm was applied under inactivating conditions (serosa −30 mV), the reduction of G Cl was stronger upon subsequent voltage-activation than under steady-state activated conditions. This effect was essentially complete less than 30 sec after apical addition of CN, but G t recovered thereafter partially in the continuous presence of CN. Dinitrophenol inhibited G Cl similarly, while omission of oxygen did not affect it. These observations, as well as the time course of inhibition and the full reversibility, suggest that interference of CN with oxidative phosphorylation and subsequent metabolic depletion is not the reason for the inhibition of G Cl . We propose that the inhibition is directly on G Cl , presumably by competition with Cl at a rate-limiting site in the pathway. Location and molecular nature of this site remain to be identified. Rate and equilibrium measurements of ryanodine binding to terminal cysternae fractions of heavy sarcoplasmic reticulum vesicles demonstrate that its activation by high concentrations of monovalent salts is based on neither elevated osmolarity nor ionic strength. The effect of the ions specifically depends on their chemical nature following the Hofmeister ion series for cations (Li+ < NH+ 4 < K∼ Cs+≤ Na+) and anions (gluconate < Cl < NO3 ∼ ClO4 ∼ SCN) respectively, indicating that both are involved in the formation of the salt-protein complex that can react with ryanodine. Activation by rising salt concentrations exhibits saturation kinetics with different dissociation constants (25–11 m) and different degrees of cooperativity (n= 1.5–4.0) for the respective salts. Maximal second order binding rates between 40,000 and 80,000 (m −1· sec−1) were obtained for chlorides and nitrates of 1a group alkali ions with the exception of lithium supporting only rates of maximally 10,000 (M−1· sec−1). The nitrogen bases, NH+ 4 and Tris+, in combination with chloride or nitrate, behave divergently. High maximal binding rates were achieved only with NH4NO3. The dissociation constants for the ryanodine–protein complexes obtained by measurements at equilibrium proved to depend differently on salt concentration, yet, converging to 1–3 nm for the applied salts at saturating concentrations. The salts do not affect dissociation of the ryanodine protein complex proving that the effect of salts on the protein's affinity for ryanodine is determined by their effect on the on-rate of ryanodine binding. ATP and its analogues modify salt action resulting in elevated maximal binding rates and reduction or abolition of binding cooperativity. Linear relations have been obtained by comparing the rates of ryanodine binding at different salt concentrations with the rates or the initial amplitudes (15 sec) of salt induced calcium release from actively loaded heavy vesicles indicating that the various salts promote specifically and concentration dependently channel opening and its reaction with ryanodine. In vivo studies with leaf cells of aquatic plant species such as Elodea nuttallii revealed the proton permeability and conductance of the plasma membrane to be strongly pH dependent. The question was posed if similar pH dependent permeability changes also occur in isolated plasma membrane vesicles. Here we report the use of acridine orange to quantify passive proton fluxes. Right-side out vesicles were exposed to pH jumps. From the decay of the applied ΔpH the proton fluxes and proton permeability coefficients (PH+) were calculated. As in the intact Elodea plasma membrane, the proton permeability of the vesicle membrane is pH sensitive, an effect of internal pH as well as external pH on PH+ was observed. Under near symmetric conditions, i.e., zero electrical potential and zero ΔpH, PH+ increased from 65 × 10−8 at pH 8.5 to 10−1 m/sec at pH 11 and the conductance from 13 × 10−6 to 30 × 10−4 S/m2. At a constant pH i of 8 and a pH o going from 8.5 to 11, PH+ increased more than tenfold from 2 to 26 × 10−6 m/sec. The calculated values of PH+ were several orders of magnitude lower than those obtained from studies on intact leaves. Apparently, in plasma membrane purified vesicles the transport system responsible for the observed high proton permeability in vivo is either (partly) inactive or lost during the procedure of vesicle preparation. The residue proton permeability is in agreement with values found for liposome or planar lipid bilayer membranes, suggesting that it reflects an intrinsic permeability of the phospholipid bilayer to protons. In the giant-celled marine algae Valonia utricularis the turgor-sensing mechanism of the plasmalemma and the role of the tonoplast in turgor regulation is unknown because of the lack of solid data about the individual electrical properties of the plasmalemma and the vacuolar membrane. For this reason, a vacuolar perfusion technique was developed that allowed controlled manipulation of the vacuolar sap under turgescent conditions (up to about 0.3 MPa). Charge-pulse relaxation studies on vacuolarly perfused cells at different turgor pressure values showed that the area-specific resistance of the total membrane barrier (tonoplast and plasmalemma) exhibited a similar dependence on turgor pressure as reported in the literature for nonperfused cells: the resistance assumed a minimum value at the physiological turgor pressure of about 0.1 MPa. The agreement of the data suggested that the perfusion process did not alter the transport properties of the membrane barrier. Addition of 16 μm of the H+-carrier FCCP (carbonylcyanide p-trifluoromethoxyphenyhydrazone) to the perfusion solution resulted in a drop of the total membrane potential from +4 mV to −22 mV and in an increase of the area-specific membrane resistance from 6.8 × 10−2 to 40.6 × 10−2Ωm2. The time constants of the two exponentials of the charge pulse relaxation spectrum increased significantly. These results are inconsistent with the assumption of a high-conductance state of the tonoplast (R. Lainson and C.P. Field, J. Membrane Biol. 29:81–94, 1976). Depending on the site of addition, the pore-forming antibiotics nystatin and amphotericin B affected either the time constant of the fast or of the slow relaxation (provided that the composition of the perfusion solution and the artificial sea water were replaced by a cytoplasma-analogous medium). When 50 μm of the antibiotics were added externally, the fast relaxation process disappeared. Contrastingly, the slow relaxation process disappeared upon vacuolar addition. The antibiotics cannot penetrate biomembranes rapidly, and therefore, the findings suggested that the fast and slow relaxations originated exclusively from the electrical properties of the plasmalemma and the tonoplast respectively. This interpretation implies that the area-specific resistance of the tonoplast is significantly larger than that of the plasmalemma (consistent with the FCCP data) and that the area-specific capacitance of the tonoplast is unusually high (6.21 × 10−2 Fm−2 compared to 0.77 × 10−2 Fm−2 of the plasmalemma). Thus, we have to assume that the vacuolar membrane of V. utricularis is highly folded (by a factor of about 9 in relation to the geometric area) and/or contains a fairly high concentration of mobile charges of an unknown electrogenic ion carrier system. The effects of jasmonic acid (JA) on elongation growth of coleoptile segments from etiolated maize (Zea mays L.) were investigated in the presence and absence of auxin. When supplied alone, at physiological concentrations (10−9, 10−8, and 10−5 m), JA (or methyl-JA) inhibited growth. JA at a similar range of concentrations also inhibited auxin-induced elongation growth. To determine whether this effect on growth depended on endogenous abscisic acid (ABA), we grew maize coleoptiles in the presence of norflurazon (an inhibitor of carotenoid biosynthesis) that results in reduced endogenous ABA levels. Growth of etiolated coleoptile segments from these plants was inhibited by JA (or methyl-JA) in both the absence and presence of auxin. Previously, we have observed a correlation between elongation growth and cytosolic pH (pHi), in which auxin lowers pHi, and growth inhibitors such as ABA raise pHi. We examined the effect of low concentrations of methyl-JA on pHi with dual emission dye, carboxy seminaphthorhodafluor-1, and confocal microscopy. To confirm these studies, we also used in vivo 31P NMR spectrometry to ascertain the changes in pHi after addition of jasmonate to maize coleoptiles. Coleoptiles grown in either the absence or presence of norflurazon responded to methyl-JA or JA by increases in pHi of approximately 0.2 pH unit. This response occurs over a period of 15–20 min and appears to be independent of endogenous ABA.
