Kinetic properties of lysozyme from the hemolymph of Crassostrea virginica   总被引:1,自引:0,他引:1  
Lysozyme activity has been demonstrated in both the supernatant and pellet fractions of whole hemolymph of the American oyster, Crassostrea virginica, subjected to centrifugation at 4000 and 10,000 × g. In each case the enzyme activity is greater in the supernatant than in the pellet.The lytic activity of the molluscan lysozyme on Micrococcus lysodeikticus, like that of egg-white lysozyme, is salt dependent, is relatively heat stable, and is very sensitive to changes in ionic concentration. The optimal pH of the molluscan enzyme, however, ranges from 5.0 to 5.5, depending on the buffer employed.When tested against a number of bacteria, the oyster lysozyme has been found to be active against not only M. lysodeikticus but also Bacillus subtilis, B. megaterium, Escherichia coli, Gaffkya tetragena, Salmonella pullorum, and Shigella sonnei, although it is less active against the last four mentioned. It is not active against Staphylococcus aureus.It is postulated that the lysozyme in the serum of C. virginica has its origin in cytoplasmic phagosomes of granulocytes and is released when these organelles become ruptured.  相似文献

The knowledge concerning the humoral immunity is scarce in amphioxus Branchiostoma belcheri. This study measured the humoral parameters including lysozyme, antimicrobial activity, microbial agglutinin and haemagglutinin in amphioxus humoral fluids before and after Vibrio alginolyticus challenge. After challenged with V. alginolyticus, the lysozyme activity, growth inhibiting activities against Escherichia coli and V. alginolyticus and microbial agglutinating activities against Micrococcus lysodeikticus, Bacillus subtilis and Staphylococcus aureus were all increased significantly and haemagglutinating activities against rabbit and human A and O erythrocytes in the humoral fluids were all increased earlier. In contrast, the agglutinating activities against Vibrio harvey and E. coli in the humoral fluids were reduced in response to V. alginolyticus challenge and the haemagglutinating activity against human B erythrocytes increased later.  相似文献

The synthesis of a new series of 8-bromo-6-alkyl-1-aryl-6H-isoxazolo[4,3-e]indole derivatives is described. All the newly synthesized compounds were screened for their antibacterial activity against Escherichia coli HB101, Staphylococcus aureus pathogens (methicillin resistant S. aureus and methicillin susceptible S. aureus), Pseudomonas aeruginosa, and Bacillus subtilis; also MIC values of these compounds were determined.  相似文献

Direct injection into the hemolymph of Heliothis zea of either an entomopathogen (Bacillus thuringiensis subsp. kurstaki) or a nonpathogen (Micrococcus lysodeikticus) is followed by a rapid phagocytosis and extensive removal of the organisms within 2 hr. The bacteria that survive this initial clearance initiate a new round of growth that is clearly evident 6–8 hr after injection. When the infecting organism is M. lysodeikticus, a second period of clearance occurs 8–12 hr after injection and nearly complete removal (many by lysis) is evident by the 12th hr. Larvae usually survive infection with this organism. When B. thuringiensis is the infecting organism, 60–80% of the phagocytized bacteria are lysed, however, the second wave of clearance seen with M. lysodeikticus does not occur; instead, the bacteria multiply extensively and death of the larvae results 12–16 hr after injection. This death does not appear to be caused either by crystalline protein or by the β-exotoxin. Analysis of hemolymph proteins using one-dimensional polyacrylamide gel electrophoresis indicated that although some quantitative changes were observed in some experiments, in the faster moving proteins when the infecting agent was B. thuringiensis, they were not consistent enough to support the idea that hemolymph proteins were either synthesized or used up during the time larvae were responding to the infectious agent. Dramatic changes were evident when the larvae were near death. No changes were ever observed when M. lysodeikticus was used as the infecting organism. A rapid response to infection using free spores of B. thuringiensis (sickness within 2–4 hr followed by death at 6–8 hr) may indicate that the spore germinating process is accompanied by release of a highly toxic material.  相似文献

Toxic metal contamination in soils due industrialization is nowadays a concern to the scientists worldwide. The current study deals with the evaluation of response and tolerance by isolated metallophilic bacteria in different lead concentrations (100 ppm to 1000 ppm). By taking optical densities of the isolates, the minimum inhibitory concentration (MIC) of Pb2+ were determined.16S rRNA and MALDI-TOF MS were used for the identification of the bacteria. Total of 37 isolates were observed, among them 04 (Staphylococcus equorum, Staphylococcus warneri, Bacillus safensis and Bacillus thuringiensis), isolated were detected having efficacy of Pb2+tolerance and sequestration at varying MIC. Furthermore, B. thuringiensis was observed to have highest (900 ppm) tolerance for lead and lowest (500 ppm) for Staphylococcus warneri. Moreover, the highest (65.3%) sequestration potential has been observed for B. thuringiensis and least (52.8%) for S. warneri. The tolerance and sequestration potential properties of these isolated species can be utilised to exterminate heavy metals and reduce their toxicity from the contaminated environment.  相似文献

A search for new members of the TnrA and GlnR regulons, responsible for assimilation of nitrogen in Gram-positive bacteria, was performed. Common regulatory signals with consensus sequences ATGTNAWWWWWWWTNACAT and TGTNAWWWWWWWTNACA were identified for GlnR and TnrA, respectively. The structure was described and new potential members were found in Bacillus subtilis, B. licheniformis, Geobacillus kaustophilus, and Oceanobacillus iheyensis for the TnrA/GlnR regulons; in B. halodurans for the TnrA regulon; and in Lactococcus lactis, Lactobacillus plantarum, Streptococcus pyogenes, S. pneumoniae, S. mutans, S. agalactiae, Enterococcus faecalis, Listeria monocytogenes, Staphylococcus aureus, and St. epidermidis for the GlnR regulon.  相似文献

Novel 3-elongated arylalkoxybenzamide derivatives were designed, synthesized and evaluated for their cell division inhibitory activity and antibacterial activity. Among them, the subseries of 3-alkyloxybenzamide derivatives exhibited greatly improved on-target activity against Bacillus subtilis and Staphylococcus aureus, and remarkably increased antibacterial activity against B. subtilis ATCC9372, penicillin-susceptible S. aureus ATCC25923, methicillin-resistant S. aureus ATCC29213 (MRSA) and penicillin-resistant S. aureus PR compared with 3-methoxybenzamide. In contrast, the subseries of 3-phenoxyaklyloxybenzamide, 3-heteroarylalkyloxybenzamide and 3-heteroarylthioalkyloxybenzamide derivatives only showed a significant improvement in on-target activity and antibacterial activity against B. subtilis ATCC9372.  相似文献

In the present investigation lymphocyte bacterial binding, receptors for bacteria on lymphocytes, and the relationships between bacterial binding and lymphocyte activation were studied. Of the strains used Escherichia coli, Bacillus subtilis, and Corynebacterium xerosis bound to both purified T and B cells. Staphylococcus aureus, Staphylococcus albus, and Brucella melitensis bound chiefly to B lymphocytes. Monosaccharides and treatment of lymphocytes with lectins, enzymes, or sodium metaperiodate affected bacterial adherence. Thus the lymphocyte receptors for bacteria appeared to contain carbohydrate moieties. There was no clear-cut correlation between lymphocyte binding of bacteria and bacterium-induced leukocyte-inhibitory factor (LIF) synthesis and proliferation.  相似文献

The effect of live bacteria (Micrococcus lysodeikticus and Vibrio anguillarum), and PAMPs (poly I:C, zymosan, LPS, LTA and CpG) on the production of intermediate toxic radicals (respiratory burst activity and production of nitric oxide) and mytilin B, myticin C and lysozyme gene expression was studied in vivo and in vitro. In vitro, bacteria were able to modulate the haemocytes' respiratory burst activity, being significantly increased after 6 h of incubation. The effect of pathogen-associated molecular patterns (PAMPs) was also studied. Zymosan produced an increase of the PMA-mediated response but an inhibition of the zymosan-mediated response. A significant increase of nitric oxide production was found at all the sampled time points (1, 3 and 6 h) in comparison with controls on both, the Gram-positive and Gram-negative bacteria. The in vivo responses measured on haemocytes after M. lysodeikticus injection were faster than those induced by V. anguillarum. However, V. anguillarum induced stronger in vitro effects. Mytilin B, myticin C and lysozyme in vitro gene expression, occurred at short times after infection. The maximum in vitro expression was detected 3 h post-infection. The differences between M. lysodeikticus and V. anguillarum in different measured parameters may suggest that different signalling pathways might be involved. Moreover, among all assayed PAMPs, LPS elicited the highest response.  相似文献

The Bacillus subtilis 168 autolytic glycosidase degrades Micrococcus lysodeikticus cells or cell walls, whereas the B. subtilis autolytic amidase does not. The criteria used to establish this fact included: the determination of chemical bonds broken, heat-inactivated kinetics, pH dependence curves, and the physical separation of glycosidase from amidase. The physical separation involved LiCl elution from two different ion-exchange materials, walls from B. subtilis 168 strain βAO, and walls from mutant strain βA173 derived from strain βAO. No evidence was obtained for B. subtilis vegetative bacteria making any more autolysins than one autolytic amidase and one autolytic glycosidase.  相似文献

Plasmids isolated from Bacillus subtilis and Staphylococcus epidermidis were transformed into Staphylococcus aureus. Heterologous transformation was susceptible to restriction in S. aureus but could be performed in restriction-negative mutants or in heat-treated host bacteria. Three plasmids isolated from S. epidermidis were transformed into S. aureus with this technique and characterized. Two of them, pTE109 and pCE109, appear to be similar to two tet and cml plasmids previously isolated from S. aureus. The third, pPE109, carries penicillin and cadmium resistance and shows a restriction enzyme pattern which differs from known penicillinase plasmids in S. aureus.  相似文献

The shuttle plasmid pGK12, as well as several Staphylococcus aureus plasmids, was introduced into Streptococcus lactis by intergeneric protoplast fusion with Bacillus subtilis. Riboflavin(RF)-productivity of reseoflavin(RoF)-resistant strains of Staphylococcus aureus, Bacillus pumilus and Bacillus subtilis was studied. Two of three types of resistant strains of S. aureus were shown to produce more RF than the parent strains. Resistant strains of B. pumilus were also RF-producing, and one type (fp) of the resistant strains from B. subtilis HW produced a large amount of RF in the culture medium. The hereditary stability of RF-producing properties of the fp type was also shown.  相似文献

BackgroundBacillus anthracis is the causative agent of anthrax, a disease associated with a very high mortality rate in its invasive forms.MethodsWe studied a number of ebselen analogs as inhibitors of B. anthracis thioredoxin reductase and their antibacterial activity on Bacillus subtilis, Staphylococcus aureus, Bacillus cereus and Mycobacterium tuberculosis.ResultsThe most potent compounds in the series gave IC50 values down to 70 nM for the pure enzyme and minimal inhibitory concentrations (MICs) down to 0.4 μM (0.12 μg/ml) for B. subtilis, 1.5 μM (0.64 μg/ml) for S. aureus, 2 μM (0.86 μg/ml) for B. cereus and 10 μg/ml for M. tuberculosis. Minimal bactericidal concentrations (MBCs) were found at 1–1.5 times the MIC, indicating a general, class-dependent, bactericidal mode of action. The combined bacteriological and enzymological data were used to construct a preliminary structure–activity-relationship for the benzoisoselenazol class of compounds. When S. aureus and B. subtilis were exposed to ebselen, we were unable to isolate resistant mutants on both solid and in liquid medium suggesting a high resistance barrier.ConclusionsThese results suggest that ebselen and analogs thereof could be developed into a novel antibiotic class, useful for the treatment of infections caused by B. anthracis, S. aureus, M. tuberculosis and other clinically important bacteria. Furthermore, the high barrier against resistance development is encouraging for further drug development.General significanceWe have characterized the thioredoxin system from B. anthracis as a novel drug target and ebselen and analogs thereof as a potential new class of antibiotics targeting several important human pathogens.  相似文献

The interaction of two cytolytic toxins, Cyt1Ab from Bacillus thuringiensis subsp. medellin and Cyt2Ba from Bacillus thuringiensis subsp. israelensis, with Bacillus sphaericus was evaluated against susceptible and resistant Culex quinquefasciatus and the nonsensitive species Aedes aegypti. Mixtures of B. sphaericus with either cytolytic toxin were synergistic, and B. sphaericus resistance in C. quinquefasciatus was suppressed from >17,000- to 2-fold with a 3:1 mixture of B. sphaericus and Cyt1Ab. This trait may prove useful for combating insecticide resistance and for improving the activity of microbial insecticides.  相似文献

Active 50 S ribosomal subunits from Bacillus licheniformis and Bacillus subtilis can be reconstituted in vitro from dissociated RNA and proteins. The reconstituted 50 S sub-units are indistinguishable from native 50 S subunits in sedimentation on sucrose gradients and in protein composition. The procedure used is similar to that developed for reconstitution of Bacillus stearothermophilus 50 S subunits, though the optimal conditions are somewhat different. Hybrid ribosomes can be reconstituted with 23 S RNA and proteins from different sources (B. stearothermophilus and B. licheniformis or B. subtilis). The thermal stability of these ribosomes depends on the source of the proteins, and not on the source of 23 S RNA.  相似文献

Bacteria can either exist in the planktonic (free floating) state or in the biofilm (encased within an organic framework) state. Bacteria biofilms cause industrial concerns and medical complications and there has been a great deal of interest in the discovery of small molecule agents that can inhibit the formation of biofilms or disperse existing structures. Herein we show that, contrary to previously published reports, d-amino acids do not inhibit biofilm formation of Bacillus subtilis (B. subtilis), Staphylococcus aureus (S. aureus), and Staphylococcus epidermis (S. epidermis) at millimolar concentrations. We evaluated a diverse set of natural and unnatural d-amino acids and observed no activity from these compounds in inhibiting biofilm formation.  相似文献

Transposon mutagenesis with the Enterococcus faecalis transposon Tn917 is a genetic approach frequently used to identify genes related with specific phenotypes in gram-positive bacteria. We established an arbitrary PCR for the rapid and easy identification of Tn917 insertion sites in Staphylococcus epidermidis with six independent, well-characterized biofilm-negative Tn917 transposon mutants, which were clustered in the icaADBC gene locus or harbor Tn917 in the regulatory gene rsbU. For all six of these mutants, short chromosomal DNA fragments flanking both transposon ends could be amplified. All fragments were sufficient to correctly identify the Tn917 insertion sites in the published S. epidermidis genomes. By using this technique, the Tn917 insertion sites of three not-yet-characterized biofilm-negative or nonmucoid mutants were identified. In the biofilm-negative and nonmucoid mutant M12, Tn917 is inserted into a gene homologous to the regulatory gene purR of Bacillus subtilis and Staphylococcus aureus. The Tn917 insertions of the nonmucoid but biofilm-positive mutants M16 and M20 are located in genes homologous to components of the phosphoenolpyruvate-sugar phosphotransferase system (PTS) of B. subtilis, S. aureus, and Staphylococcus carnosus, indicating an influence of the PTS on the mucoid phenotype in S. epidermidis.  相似文献

Staphylococcus xylosus, Staphylococcus equorum, and Staphylococcus epidermidis strains were isolated from Bryndza cheese and identified using PCR method. The antimicrobial susceptibility of these strains was assessed using disc diffusion method and broth microdilution method. The highest percentage of resistance was detected for ampicillin and oxacillin, and in contrary, isolates were susceptible or intermediate resistant to ciprofloxacin and chloramphenicol. Fourteen of the S. xylosus isolates (45 %) and eleven of the S. equorum isolates (41 %) exhibited multidrug resistance. None of the S. epidermidis isolate was multiresistant. The phenotypic resistance to oxacillin was verified by PCR amplification of the gene mecA.  相似文献
