为寻求新的食用油资源,发展了一种快速可靠的气相色谱-质谱联用方法,用于植物籽油中脂肪酸成分的定性鉴定和含量测定。所建立的方法成功用于葡萄籽、南瓜籽和猕猴桃籽等七种植物籽油中的棕榈酸、十八烷酸、油酸、亚油酸和α-亚麻酸的定性定量分析。结果表明,刺葡萄籽油、普通葡萄籽油、国外葡萄籽油、南瓜籽油、枸杞籽油和西番莲籽油均具有相似的脂肪酸谱,尽管其中它们所含上述五种脂肪酸含量不同,由于均存在人体所必需的饱和与不饱和脂肪酸,故可以用作替代食用油。猕猴桃籽油因为其存在高含量的α-亚麻酸成分,可能是更好的食用油和营养油资源。本文首次对枸杞籽油、西番莲籽油和猕猴桃籽油脂肪酸成分进行绝对含量分析,为新的食用油资源的开发提供了重要的依据。  相似文献

A method to determine the content and composition of total fatty acids present in microalgae is described. Fatty acids are a major constituent of microalgal biomass. These fatty acids can be present in different acyl-lipid classes. Especially the fatty acids present in triacylglycerol (TAG) are of commercial interest, because they can be used for production of transportation fuels, bulk chemicals, nutraceuticals (ω-3 fatty acids), and food commodities. To develop commercial applications, reliable analytical methods for quantification of fatty acid content and composition are needed. Microalgae are single cells surrounded by a rigid cell wall. A fatty acid analysis method should provide sufficient cell disruption to liberate all acyl lipids and the extraction procedure used should be able to extract all acyl lipid classes.With the method presented here all fatty acids present in microalgae can be accurately and reproducibly identified and quantified using small amounts of sample (5 mg) independent of their chain length, degree of unsaturation, or the lipid class they are part of.This method does not provide information about the relative abundance of different lipid classes, but can be extended to separate lipid classes from each other.The method is based on a sequence of mechanical cell disruption, solvent based lipid extraction, transesterification of fatty acids to fatty acid methyl esters (FAMEs), and quantification and identification of FAMEs using gas chromatography (GC-FID). A TAG internal standard (tripentadecanoin) is added prior to the analytical procedure to correct for losses during extraction and incomplete transesterification.  相似文献

The present study aims to evaluate the effects of feeding palm oil by-products based diets on different muscle fatty acid profiles in goats. Thirty-two Cacang × Boer goats were randomly assigned to four dietary treatments: (1) control diet (CD), (2) 80% decanter cake diet (DCD), (3) 80% palm kernel cake diet (PKCD) and (4) CD plus 5% palm oil (PO) supplemented diet (CPOD). After 100 days of feeding, four goats from each group were slaughtered and longissimus dorsi (LD), infraspinatus (IS) and biceps femoris (BF) were sampled for analysis of fatty acids. Goats fed the PKCD had higher (P<0.05) concentration of lauric acid (C12:0) than those fed the other diets in all the muscles tested. Compared to the other diets, the concentrations of palmitic acid (C16:0) and stearic acid (C18:0) were lower (P<0.05) and that of linoleic acid (C18:2 n-6) was higher (P<0.05) in the muscles from goats fed the CD. It was concluded that palm kernel cake and decanter cake can be included in the diet of goats up to 80% with more beneficial than detrimental effects on the fatty acid profile of their meat.  相似文献

大脑功能与脑内脂肪酸成分密切相关,分析研究老年时脑脂肪酸组成及含量有助于阐明不同类型脂肪酸在老年认知能力降低过程中的作用.采用Y型电迷宫测试老年组(22月龄)和青年组(3月龄)大鼠的学习记忆能力,表明老年组大鼠的学习记忆能力显著下降.用气相色谱的方法分析大鼠大脑皮质脂肪酸的组成及含量,显示老年组和青年组大脑皮质均含有16种脂肪酸.以C23:0为内标对各种脂肪酸进行了定量,老年组总脂肪酸含量比青年组降低约15%,各脂肪酸中含量下降的脂肪酸有长链饱和脂肪酸(C14:0、C16:0、C18:0)及多不饱和脂肪酸(C18:3、C20:4、C22:4、C22:6),含量升高的脂肪酸有单不饱和脂肪酸C20:1、C24:1.用峰面积归一法计算了各脂肪酸的相对含量,老年组相对含量下降的脂肪酸有C18:0、C20:4、C22:6,相对含量升高的有极长链饱和脂肪酸(C20:0、C24:0)及单不饱和脂肪酸(C16:1、C18:1、C20:1、C22:1、C24:1).相关性分析显示,大鼠学习能力与脑皮质C22:6、C22:4、C20:4水平呈正相关,与C20:1、C24:1水平呈负相关.上述结果为阐明不同脂肪酸在老年大脑认知功能障碍中的作用提供了实验依据。  相似文献

The seed oils of three species (Litsea cubeba (Lour.) Pers, L. auriculata Chien et Cheng, L. subcoriacea Yang et P. H. Huang) were examined and the fatty acid composition of these oils was determined by GLC. Their major fatty acid was identified as lauric acid, Its amount ranged from 34.6%–75.4%. The major acid of the fruit coat oil from L. subcoriacea Y. H. Huang was different from that of the seed oil. The former contained 50% linoleic acid. The unsaturated C10, C12, C14 acids of the seed oil from L. cubeba (Lour.) Pers were separated by distillation, column chromatography and were identified by Periodate-Perman-ganate oxidation, IR, NMR and MS. They are cis-4-decenoic, cis-4-dodecenoic and cis-4-tetradecenoic acids respectively.  相似文献

利用气相色谱法,对杜仲籽油和紫苏籽油的脂肪酸组成、α-亚麻酸含量等进行了比较研究。结果发现,两者不仅脂肪酸GC指纹图谱较为相似(脂肪酸组成、含量基本相同),而且外观、气味、折光率等质量指标也非常相近。说明了杜仲籽油具有与紫苏籽油同样的开发价值。  相似文献

One of the main challenges for the successful production and use of microalgae for biodiesel production is to obtain a satisfactory level of fatty acid methyl esters (FAME). The aims of this study are to identify the best method of lipid extraction and provide high FAME levels and to evaluate their fatty acid profiles. Six lipid extraction methodologies in three microalgae species were tested in comparison with the direct transesterification (DT) of microalgal biomass method. The choice of extraction method affected both the oily extract yield and the FAME composition of the microalgae and consequently may affect the properties of biodiesel. The efficiency of different lipid extraction methods is affected by the solvent polarity, which extracts different target compounds from lipid matrix. Dichloromethane/methanol extraction and Folch extraction produced the largest oil extract yields, but extraction with hexane/ethanol resulted in the best ester profile and levels. Performing DT reduces the volume of extractor solvent, the time and cost of FA composition analysis, as well as, presents less steps for fatty acid quantification. DT provided biomass FAME levels of 50.2, 636.4, and 258.2 mg.g?1 in Nannochlorophisis oculata, Chaetoceros muelleri, and Chlorella sp., respectively. On the basis of an analysis of the fatty acids profiles of different species, C. muelleri is a promising microalga for biodiesel production. Depending on the extraction method, Chlorella sp. and N. oculata can be considered as an alternative in obtaining arachidonic (Aa) and eicosapentaenoic (EPA) acids.  相似文献

不同甘蓝型油菜高含油量种质资源的脂肪酸成分分析   总被引:4,自引:0,他引:4  
甘蓝型油菜是我国最为重要的油料作物之一.目前我国面临着植物油严重不足的局面.提高含油量是目前甘蓝型油菜育种的主要方向之一.目前.我国育种工作者已经筛选出了大量含油量超过50%的甘蓝型油菜种质.本文对两种类型的高含油量甘蓝型油菜种质进行了脂肪酸组分分析,结果显示芥酸含量高的种质和芥酸含量低的种质的脂肪酸在菜籽油中变化范围和平均值明显不同.油酸和亚油酸在低芥酸高含油量甘蓝型油菜种质中的含量较高,约占整个油分的80%左右,其他脂肪酸组分则在10%以下;而芥酸则是高芥酸高含油量种质的最主要的成分,约占整个油分的44%左右,油酸、亚油酸等的含量则均在10%左右.相关性分析表明:芥酸和其他脂肪酸之间呈现显著负相关,因此.可以通过降低高芥酸油菜种质中的芥酸含量来提高比如油酸和亚油酸的含量.从而使这些种质的油分更适合人类的健康.  相似文献

In two experiments with growing-finishing pigs six different dietary fats were added to a conventional diet (control - C) to study the effects of dietary monounsaturated (MUFA) and polyunsaturated fatty acids (PUFA) on the fatty acid composition of backfat and kidney fat at similar amounts of double bonds in feed (Exp. 1:7% pork fat - PF, 4.95% olive oil - OO, 3.17% soybean oil - SO) or a constant amount of 5% of processed fats (Exp. 2: partially hydrogenated fat - SAT, fractionated pork fats: olein - OLE, stearin - STE). Compared with the control, PUFA were only slightly increased in backfat of pigs fed PF, OLE, STE or OO, although dietary PUFA intake was up to 70% higher. With SO PUFA were significantly increased in adipose tissues, predominantly at the expense of MUFA. Consequently, a non-linear relationship was found between PUFA intake and proportion in backfat. MUFA were incorporated at the expense of SFA, therefore, adipose tissues of OO fed animals were lowest in SFA. Despite comparable amounts of double bonds in feed (Exp. 1), the degree of unsaturation measured as fat score (sum of double bonds) was in the order SO > OO > PF > C. In contrast, the proportion of SFA was C > PF = SO > OO. Regarding the decisive role of SFA for fat consistency it may be concluded that MUFA should also be considered in feeding recommendations for pigs. Furthermore, in case of a high dietary supply of MUFA, a simple index of double bonds might not be sufficiently conclusive to judge pig fat quality.  相似文献

Fatty acid composition is an important phenotypic trait in pigs as it affects nutritional, technical and sensory quality of pork. Here, we reported a genome-wide association study (GWAS) for fatty acid composition in the longissimus muscle and abdominal fat tissues of 591 White Duroc×Erhualian F2 animals and in muscle samples of 282 Chinese Sutai pigs. A total of 46 loci surpassing the suggestive significance level were identified on 15 pig chromosomes (SSC) for 12 fatty acids, revealing the complex genetic architecture of fatty acid composition in pigs. Of the 46 loci, 15 on SSC5, 7, 14 and 16 reached the genome-wide significance level. The two most significant SNPs were ss131535508 (P = 2.48×10−25) at 41.39 Mb on SSC16 for C20∶0 in abdominal fat and ss478935891 (P = 3.29×10−13) at 121.31 Mb on SSC14 for muscle C18∶0. A meta-analysis of GWAS identified 4 novel loci and enhanced the association strength at 6 loci compared to those evidenced in a single population, suggesting the presence of common underlying variants. The longissimus muscle and abdominal fat showed consistent association profiles at most of the identified loci and distinct association signals at several loci. All loci have specific effects on fatty acid composition, except for two loci on SSC4 and SSC7 affecting multiple fatness traits. Several promising candidate genes were found in the neighboring regions of the lead SNPs at the genome-wide significant loci, such as SCD for C18∶0 and C16∶1 on SSC14 and ELOVL7 for C20∶0 on SSC16. The findings provide insights into the molecular basis of fatty acid composition in pigs, and would benefit the final identification of the underlying mutations.  相似文献

通过气相色谱-质谱法分析了水酶法提取的石榴籽油的脂肪酸组成。结果表明:石榴籽油含有丰富的不饱和脂肪酸,总不饱和脂肪酸含量达94%以上。采用Schall烘箱法,以过氧化值(POV)为参考指标,研究了光线、温度及抗氧化剂对石榴籽油氧化稳定性能的影响。结果表明,温度、光线对石榴籽油的氧化过程都有影响,温度的影响更为显著;叔丁基对苯二酚(TBHQ)对石榴籽油具有良好的抗氧化效果,并与抗坏血酸(VC)具有较好的协同增效作用;生育酚(VE)和没食子酸丙酯(PG)对石榴籽油无显著抗氧化作用。  相似文献

Lipid metabolism in the rumen is responsible for the complex fatty acid profile of rumen outflow compared with the dietary fatty acid composition, contributing to the lipid profile of ruminant products. A method for the detailed dimethylacetal and fatty acid analysis of rumen contents was developed and applied to rumen content collected from lambs fed lucerne or concentrate based diets supplemented with soybean oil. The methodological approach developed consisted on a basic/acid direct transesterification followed by thin-layer chromatography to isolate fatty acid methyl esters from dimethylacetal, oxo- fatty acid and fatty acid dimethylesters. The dimethylacetal composition was quite similar to the fatty acid composition, presenting even-, odd- and branched-chain structures. Total and individual odd- and branched-chain dimethylacetals were mostly affected by basal diet. The presence of 18∶1 dimethylacetals indicates that biohydrogenation intermediates might be incorporated in structural microbial lipids. Moreover, medium-chain fatty acid dimethylesters were identified for the first time in the rumen content despite their concentration being relatively low. The fatty acids containing 18 carbon-chain lengths comprise the majority of the fatty acids present in the rumen content, most of them being biohydrogenation intermediates of 18∶2n−6 and 18∶3n−3. Additionally, three oxo- fatty acids were identified in rumen samples, and 16-O-18∶0 might be produced during biohydrogenation of the 18∶3n−3.  相似文献
