The availability and uptake of Cd by lettuce (Lactuca sativa L.) in two common tropical soils (before and after liming) were studied in order to derive human health-based risk soil concentration. Cadmium concentrations ranging from 1 to 12 mg kg?1 were added to samples from a clayey Oxisol and a sandy-loam Ultisol under glasshouse conditions. After incubation, a soil sample was taken from each pot, the concentration of Cd in the soil was determined, lettuce was grown during 36 d, and the edible parts were harvested and analyzed for Cd. A positive linear correlation was observed between total soil Cd and the Cd concentration in lettuce. The amount of Cd absorbed by lettuce grown in the Ultisol was about twice the amount absorbed in the Oxisol. Liming increased the soil pH and slightly reduced Cd availability and uptake. CaCl2 extraction was better than DTPA to reflect differences in binding strength of Cd between limed and unlimed soils. Risk Cd concentrations in the Ultisol were lower than in the Oxisol, reflecting the greater degree of uptake from the Ultisol. The derived risk Cd values were dependent on soil type and the exposure scenario.  相似文献

Alleviation of soil acidity in Ultisol and Oxisol for corn growth   总被引:1,自引:0,他引:1  
Malaysian Ultisols and Oxisols are characterized by low pH, high soil solution Al concentration and Ca and/or Mg deficiencies, which are limiting to corn growth. An experiment was conducted to determine the changes in solid and soil solution phase properties of a representative Ultisol and Oxisol following applications of ground magnesium limestone (GML), gypsum and their combinations, and their effects on corn growth. A plot of pAl against lime potential (pH-1/2 pCa) showed that the points were mostly positioned between the theoretical lines for kaolinite-quartz and gibbsite equilibrium, reflecting the kaolinitic-oxidic mineralogy of the Ultisol and Oxisol. Gypsum application increased Al concentration in the soil solutions of the Ultisol, but had no significant effect on that of the Oxisol. The increase in Al concentration in the Ultisol was due to an increase in ionic strength. Gypsum application increased soil solution pH of the Oxisol due to release of OH as a result of ligand exchange between SO4 and OH ions on the oxides of Fe and/or Al. Exchangeable Al in both soils was reduced by gypsum application. The reduction was associated with solid phase immobilization through alunite formation; the soil solutions of soil samples treated with 2 and 4 t gypsum ha−1 were supersaturated with respect to alunite. Application of GML at 2 t ha−1 together with 1–2t gypsum ha−1 gave high top weight of corn. Relative top weight of corn was positively correlated with a soil solution Mg and Ca/Al concentration ratio, but negatively correlated with soil solution Al concentration. Foliar Al corn was positively correlated with soil solution Al concentration. Abstract

Remediation of toxic metals by bacteria offers a relatively inexpensive and efficient way for the decontamination of soil and associated environments. The present study was carried out to investigate the surface characteristics, adsorption, and remobilization of Cd and Cu on bacteria and their composites with soil colloidal components, which are the most active constituents in soils. The bacterial strain NTG-01 (Enterobacter aerogenes), which was both Cd- and Cu-resistant, was isolated from a heavily Cu-contaminated soil of the mining area in Daye suburb of Hubei Province, China. Batch laboratory experiments with NTG-01 and soil colloids were performed to quantify adsorption of Cu and Cd. The surface area of kaolinite and the soil colloids from an Alfisol and Ultisol increased by 3.0–8.8% after the introduction of the bacteria. In the presence of bacterial cells, the negative charges of soil colloid systems increased and the positive charges decreased, shifting pH from 4.0 to 6.5. Our results demonstrate that bacteria promote the adsorption of Cd and Cu by kaolinite and soil colloid systems. However, the heavy metals bound by the bacterial composites could also be easily released by NH4NO3 and EDTA. Caution should be taken when using such bacterial strains in bioremediation of heavy metal-contaminated soils.  相似文献

The effects of liming and inoculation with the arbuscular mycorrhizal fungus, Glomus intraradices Schenck and Smith on the uptake of phosphate (P) by maize (Zea mays L.) and soybean (Glycine max [L.] Merr.) and on depletion of inorganic phosphate fractions in rhizosphere soil (Al-P, Fe-P, and Ca-P) were studied in flat plastic containers using two acid soils, an Oxisol and an Ultisol, from Indonesia. The bulk soil pH was adjusted in both soils to 4.7, 5.6, and 6.4 by liming with different amounts of CaCO3.In both soils, liming increased shoot dry weight, total root length, and mycorrhizal colonization of roots in the two plant species. Mycorrhizal inoculation significantly increased root dry weight in some cases, but much more markedly increased shoot dry weight and P concentration in shoot and roots, and also the calculated P uptake per unit root length. In the rhizosphere soil of mycorrhizal and non-mycorrhizal plants, the depletion of Al-P, Fe-P, and Ca-P depended in some cases on the soil pH. At all pH levels, the extent of P depletion in the rhizosphere soil was greater in mycorrhizal than in non-mycorrhizal plants. Despite these quantitative differences in exploitation of soil P, mycorrhizal roots used the same inorganic P sources as non-mycorrhizal roots. These results do not suggest that mycorrhizal roots have specific properties for P solubilization. Rather, the efficient P uptake from soil solution by the roots determines the effectiveness of the use of the different soil P sources. The results indicate also that both liming and mycorrhizal colonization are important for enhancing P uptake and plant growth in tropical acid soils.  相似文献

We have investigated the effects of low molecular weight organic acid ligands on the adsorption of the insecticidal toxin from Bacillus thuringiensis (Bt) by the colloidal (<2 μm particle-size) fraction of some soils. The desorption of the bound toxin by NaCl and phosphate buffer has also been measured. The soils used were a red soil (Ultisol), a latosol (Oxisol), a yellow brown soil (Alfisol) and a yellow cinnamon soil (Alfisol) from central and southern China. The adsorption isotherms were all of the L-type, and the data fitted the Langmuir equation (R2 > 0.97). When present at low concentrations, organic acids (acetate, oxalate, citrate) had an inhibitory effect on toxin adsorption. Uptake, however, was promoted when the organic acid concentration exceeded 10 mM. The toxin was very strongly bound by the soils but the soil-toxin interaction weakened in the presence of organic acids. A small portion of the toxin was adsorbed by electrostatic and ligand exchange interactions. The addition of organic acids appeared to enhance these interactions. Responsible Editor: Thomas B. Saliva is known to modulate the adhesion of bacteria in the oral cavity. The present work was performed to assess the effect of salivary components on the adhesion of Escherichia coli to a model oral surface. Several genetically engineered E. coli strains were used to examine the role of type 1 fimbriation in the interaction of these strains with salivary components in solution or adsorbed to hydroxyapatite. High (MG1) and low (MG2) molecular weight salivary mucins, and secretory immunoglobulin A (sIgA), were found to interact with the surface of E. coli, and these interactions were independent of the expression of fimbriae or capsule. In contrast, fimbriated strains of E. coli adhered to a greater extent to saliva-coated synthetic hydroxyapatite (HAP) than did nonfimbriated strains. Testing of salivary components separated by gel filtration chromatography revealed that only high-molecular-weight components promoted adhesion of E. coli to HAP. Additional studies found that purified MG2 and sIgA promoted the adhesion of E. coli to HAP. Expression of type 1 fimbriae enhanced adhesion, while mannose inhibited adhesion of fimbriated strains, to saliva-coated HAP and to HAP coated with MG2 and sIgA. We conclude that salivary MG2 and sIgA may provide receptors for the adhesion of type 1 fimbriated E. coli to oral surfaces. Received: 10 February 1996 / Accepted: 11 March 1996  相似文献

The effect of a series of xerogel coatings modified with aminoalkyl/fluorocarbon/hydrocarbon groups on the adhesion of a new test species, the filamentous brown alga Ectocarpus crouaniorum, has been explored, and compared with the green alga Ulva linza. The results showed that E. crouaniorum adhered weakly to the less polar, low wettability coatings in the series, but stronger adhesion was shown on polar, higher surface energy coatings containing aminoalkyl groups. The results from a separate series of coatings tuned to have similar surface energies and polarities after immersion in artificial seawater (ASW), but widely different surface charges, demonstrated that surface charge was more important than surface energy and polarity in determining the adhesion strength of both E. crouaniorum and U. linza on xerogel coatings. No correlation was found between adhesion and contact angle hysteresis. X-ray photoelectron spectroscopy analysis of samples after immersion in ASW confirmed the presence of charged ammonium groups on the surface of the aminoalkylated coatings.  相似文献

Aims: To evaluate the suitability of commercially available Petrifilm? EC plates for enumeration of Escherichia coli from soil. Methods and Results: A confirmed E. coli strain isolated from liquid swine manure was inoculated into sterilized sandy clay loam and loam soils at the concentrations of 102, 103, 105 CFU g?1 of soil. The efficiency of recovery on Petrifilm? EC plates for soils spiked with E. coli was compared with standard membrane filtration techniques on m‐FC basal medium supplemented with 3‐bromo‐4‐chloro‐5‐indoyl‐β‐d ‐glucopyranoside (BCIG) and most probable numbers (MPN) techniques in E. coli medium with 4‐methylumbelliferyl‐β‐d ‐glucuronide (EC‐MUG) broth. Petrifilm? EC and m‐FC (BCIG) methods were then assessed for the ability to recover E. coli from field soils applied with swine manure. No significant differences (P > 0·05) were observed between Petrifilm? EC, m‐FC (BCIG) and MPN methods for the recovery of E. coli from spiked samples, irrespective of soil type. However, recovery of E. coli from manure‐applied field soil samples showed a significant difference (P < 0·05) between the Petrifilm? EC method and the m‐FC method in enumerating E. coli possibly as a result of false positives on m‐FC. Conclusion: The Petrifilm? EC method is suitable for the enumeration of E. coli from soil with a detection limit of 10 CFU g?1 soil. Significance and Impact of the Study: The commercially available Petrifilm? EC method is comparatively low cost, easy to use method for the enumeration of E. coli from soil without the need for further confirmation tests.  相似文献

In our previous study [Hong Y, Brown DG (2009) Appl Environ Microbiol 75(8):2346–2353], the adenosine triphosphate (ATP) level of adhered bacteria was observed to be 2–5 times higher than that of planktonic bacteria. Consequently, the proton motive force (Δp) of adhered bacteria was approximately 15% greater than that of planktonic bacteria. It was hypothesized that the cell surface pH changes upon adhesion due to the charge‐regulated nature of the bacterial cell surface and that this change in surface pH can propagate to the cytoplasmic membrane and alter Δp. In the current study, we developed and applied a charge regulation model to bacterial adhesion and demonstrated that the charge nature of the adhering surface can have a significant effect on the cell surface pH and ultimately the affect the ATP levels of adhered bacteria. The results indicated that the negatively charged glass surface can result in a two‐unit drop in cell surface pH, whereas adhesion to a positively charged amine surface can result in a two‐unit rise in pH. The working hypothesis indicates that the negatively charged surface should enhance Δp and increase cellular ATP, while the positively charged surface should decrease Δp and decrease ATP, and these results of the hypothesis are directly supported by prior experimental results with both negatively and positively charged surfaces. Overall, these results suggest that the nature of charge on the solid surface can have an impact on the proton motive force and cellular ATP levels. Biotechnol. Persistent urinary tract infections (UTI) are often caused by E. coli adhered to urothelium. This type of cells is generally recognized as very tolerant to antibiotics which renders difficult the treatment of chronic UTI. This study investigates the use of lytic bacteriophages as alternative antimicrobial agents, particularly the interaction of phages with E. coli adhered to urothelium and specifically determines their efficiency against this type of cells. The bacterial adhesion to urothelium was performed varying the bacterial cell concentrations and the period and conditions (static, shaken) of adhesion. Three collection bacteriophages (T1, T4, and phiX174 like phages) were tested against clinical E. coli isolates and only one was selected for further infection experiments. Based on the lytic spectrum against clinical isolates and its ability to infect the highest number of antibiotic resistant strains, the T1-like bacteriophage was selected. This bacteriophage caused nearly a 45% reduction of the bacterial population after 2 h of treatment. This study provides evidence that bacteriophages are effective in controlling suspended and adhered cells and therefore can be a viable alternative to antibiotics to control urothelium- adhered bacteria.  相似文献

Legionella pneumophila cell surface hydrophobicity and charge are important determinants of their mobility and persistence in engineered water systems (EWS). These surface properties may differ depending on the growth phase of L. pneumophila resulting in variable adhesion and persistence within EWS. We describe the growth-dependent variations in L. pneumophila cell surface hydrophobicity and surface charge using the microbial adhesion to hydrocarbon assay and microelectrophoresis, respectively, and their role in cell adhesion to stainless steel using a quartz crystal microbalance with dissipation (QCM-D) monitoring instrument. We observed a steady increase in L. pneumophila hydrophobicity during their lifecycle in culture media. Cell surfaces of stationary phase L. pneumophila were significantly more hydrophobic than their lag and midexponential counterparts. No significant changes in L. pneumophila cell surface charge were noted. Morphology of L. pneumophila remained relatively constant throughout their lifecycle. In the QCM-D study, lag and exponential phase L. pneumophila weakly adhered to stainless steel surfaces resulting in viscoelastic layers. In contrast, stationary phase bacteria were tightly and irreversibly bound to the surfaces, forming rigid layers. Our results suggest that the stationary phase of L. pneumophila would highly favour their adhesion to plumbing surfaces and persistence in EWS.  相似文献

Soil amendment with organic materials (crop residues animal manure, and green manure) reportedly has positive effects on soil properties, from acidity to plant-nutrient availability. To examine that hypothesis, an incubation study was conducted to assess the changes in some chemical properties of three different tropical soils (Andisol, Ultisol, and Oxisol) amended with chicken manure and green manure (Leucaena leucocephala) at the rate of 10tha(-1). The results showed that organic amendments raised soil pH and EC, regardless of the type of manure used. Manuring lowered the concentrations of Mehlich-3 extractable Ca, P, Mn and Si in all soils and decreased the concentration of Mg in the Ultisol and Oxisol. However, manure amendment led to increases in the concentrations of Mg and K in the Andisol. Organic amendments caused a decrease in KCl extractable Al. Initial soluble C levels were highest in the Oxisol (60mumolg(-1)) and lowest in the Andisol (20mumolg(-1)). The concentration of soluble C decreased exponentially with duration of incubation. Three low molecular weight organic molecules (acetic acid, catechol and oxalic acid) out of the eight tested were found in all manure-amended soils. This study quantified the release of some Al chelating organic acids, the reduction of exchangeable Al, and the changes in major plant-nutrients when organic materials were added to nutrient poor, tropical acid soils.  相似文献

The transport of bacteria has been investigated extensively using iron (Fe) (hydr)oxide-coated quartz. However, few studies have investigated the effects of aluminum (Al) (hydr)oxide on the transport of bacteria. In this study, column experiments were conducted to investigate the effects of Fe/Al hydroxides on the transport of Escherichia coli (E. coli) in saturated quartz sand at different pH levels, ionic strengths (IS), and ionic compositions. Fe/Al hydroxide coatings increased the positive charge of quartz, reduced the negative charge, shifted zeta potential in a positive direction, and thus enhanced the retention of E. coli on quartz. The retention of E. coli decreased with increasing pH and increased with increasing IS. These findings were consistent with the theoretical prediction of the Derjaguin, Landau, Verwey and Overbeek (DLVO) interaction energy. Calcium ions improved the retention of E. coli in the column. Since Al-hydroxide-coated quartz had a more positive charge, the retention of E. coli was higher in Al-hydroxide-coated quartz than in Fe-hydroxide-coated quartz. When compared with quartz alone, Fe/Al hydroxide coatings significantly reduced the transport of E. coli, and the inhibitory effect of Al hydroxide was greater than that of Fe hydroxide.  相似文献

Enterohemorrhagic E. coli O157: H7 (EHEC) is a major foodborne pathogen capable of causing diarrhea and vomiting, with further complications such as hemolytic‐uremic syndrome (HUS). The aim of this study was to use the real‐time PCR method to quantify the survival of Escherichia coli O157:H7/pGFP in phyllosphere (leaf surface), rhizosphere (volume of soil tightly held by plant roots), and non‐rhizosphere soils (sand and clay) irrigated with contaminated water and compare the results obtained between real‐time PCR method and conventional plate counts. The real‐time PCR probe was designed to hybridize with the (eae) gene of E. coli O157:H7. The probe was incorporated into real‐time PCR containing DNA extracted from the phyllosphere, rhizosphere, and non‐rhizosphere soils irrigated with water artificially contaminated with E. coli O157:H7. The detection limit for E. coli O157:H7 quantification by real‐time PCR was 2.3 × 103 in the rhizosphere and phyllosphere samples. E. coli O157:H7 survived longer in rhizosphere soil than the non‐rhizosphere soil. The concentration of E. coli O157:H7/pGFP in rhizosphere soils was ≥ 104 CFU/g in both soils at day 12 based on both plate count and real time PCR, with the clay soil significantly (P = 0.05) higher than the sandy soil. This data showed that E. coli O157H:7 can persist in the environment for more than 50 d, and this may pose some risk for both animal and human infection and provides a very significant pathway for pathogen recontamination in the environment.  相似文献

Summary The relations between the Schoenus phytocoena and their site conditions are elucidated by stand and species ordinations (ORDINA, RA) and by comparisons of ordinations of environmental variables alone and ordination of the combined environmental and species variables (RA). Correlations between the environmental variables show that they may be gathered into two contrasting groups, the carbon and the carbonate groupings, respectively. This first direction of variation is related to hydrological conditions causing the differences in thickness of organic soils, content of organic carbon and dry weight of intact soil per unit volume. The second direction of variation is associated with a nutrient factor complex, in this study represented by available phosphate.The carbon grouping and a constantly high level of mainly topogeneous soil water is typical of sites of the Oxycoccus-Schoenus association. The Sesleria group of the Primula-Schoenus association is related to sites with a periodically low table of topogeneous or soligeneous soil water and mineral or mucky soils with high values of carbonate content, pH and CEC and a fairly low nutrient state, while the sites of the Bartsia-Ophrys and Valeriana groups have a fairly high level of mainly soligeneous soil water and mucky soils with variable carbonate content, intermediate pH and CEC and somewhat higher nutrient state. Periodically flooded sites with topogeneous soil water and mineral soils are typical of the Cladium-Schoenus nigricans, Schoenus intermedius-Schoenus ferrugineus and Glaux phytocoena.Nomenclature is the same as in Tyler (1980).This study is part of my doctor's thesis. Organic and inorganic carbon (C) fluxes transported by water were evaluated for dominant hydrologic flowpaths on two adjacent headwater catchments in the Brazilian Amazon with distinct soils and hydrologic responses from September 2003 through April 2005. The Ultisol-dominated catchment produced 30% greater volume of storm-related quickflow (overland flow and shallow subsurface flow) compared to the Oxisol-dominated catchment. Quickflow fluxes were equivalent to 3.2 ± 0.2% of event precipitation for the Ultisol catchment, compared to 2.5 ± 0.3% for the Oxisol-dominated watershed (mean response ±1 SE, n = 27 storms for each watershed). Hydrologic responses were also faster on the Ultisol watershed, with time to peak flow occurring 10 min earlier on average as compared to the runoff response on the Oxisol watershed. These different hydrologic responses are attributed primarily to large differences in saturated hydraulic conductivity (K s). Overland flow was found to be an important feature on both watersheds. This was evidenced by the response rates of overland flow detectors (OFDs) during the rainy season, with overland flow intercepted by 54 ± 0.5% and 65 ± 0.5% of OFDs for the Oxisol and Ultisol watersheds respectively during biweekly periods. Small volumes of quickflow correspond to large fluxes of dissolved organic C (DOC); DOC concentrations of the hydrologic flowpaths that comprise quickflow are an order of magnitude higher than groundwater flowpaths fueling base flow (19.6 ± 1.7 mg l−1 DOC for overland flow and 8.8 ± 0.7 mg l−1 DOC for shallow subsurface flow versus 0.50 ± 0.04,mg l−1 DOC in emergent groundwater). Concentrations of dissolved inorganic C (DIC, as dissolved CO2–C plus HCO3–C) in groundwater were found to be an order of magnitude greater than quickflow DIC concentrations (21.5 mg l−1 DIC in emergent groundwater versus 1.1 mg l−1 DIC in overland flow). The importance of deeper flowpaths in the transport of inorganic C to streams is indicated by the 40:1 ratio of DIC:DOC for emergent groundwater. Dissolved CO2–C represented 92% of DIC in emergent groundwater. Results from this study illustrate a highly dynamic and tightly coupled linkage between the C cycle and the hydrologic cycle for both Ultisol and Oxisol landscapes: organic C fluxes strongly tied to flowpaths associated with quickflow, and inorganic C (particularly dissolved CO2) transported via deeper flowpaths.  相似文献

One day after application, urea-N remaining in the floodwater and determined as water-soluble N (urea-N + NH4 +-N) was used to calculate the potential N loss from lowland rice soils. Actual N loss calculated from 15N balance measurements using forced air exchange (airflow rate: 20 L min-1) in greenhouse pots. Conditions for variable potential N loss were created by manipulating the method of urea application and duration of presubmergence or by selecting soils with diverse cation exchange capacities (CEC). Potential N loss tended to be lower than actual N loss; the differences were, however, nonsignificant. The method of urea application that led to the lowest potential N loss from a Guthrie silty clay loam (Typic Fragiaquult) also led to the least 15N loss and vice-versa (r=0.99**). Duration of presubmergence did not alter the relationship between potential and actual N loss although it influenced the rate of urea hydrolysis in floodwater. The primary depencence of actual N loss on water-soluble N was maintained in soils differing in CEC (r=0.83**). The association between potential and actual N loss was closer for high-CEC soils ( 20 cmol [+] kg-1 soil, r=0.91**) than for low-CEC soils (<20 cmol [+] kg-1 soil, r=0.85**). Ammonia volatilization could be more closely predicted by potential N loss than could apparent denitrification.The results of this study suggest that potential N loss calculated from one-time determination of water-soluble N in floodwater can be a good index of actual N loss from flooded, puddled rice soils. Notable exceptions are to be expected for soils in which water-soluble N gets lost from floodwater either before (soils with fast urea hydrolysis in floodwater) or after (soils with steady leaching) determination of potential N loss.  相似文献

Changes in surface charge density of liposomes induced by E. coli endotoxin were studied by microelectrophoresis. Endotoxin altered the surface charge of phosphatidylcholine liposomes from neutral to negative. The negative charge of the endotoxin-phosphatidylcholine complex was neutralized electrostatically by binding with Ca2+ (2 mM). Phosphatidylcholine liposomes were made positive by addition of the positively charged detergent, hexadecyltrimethylammonium chloride. Endotoxin made the positively charged liposomes less charged. On the other hand, phosphatidylserine liposomes which were negatively charged became less charged in the presence of high concentration of endotoxin (8 mg/ml). The endotoxin effect on phosphatidylserine liposomes was abolished by EDTA (1 mM) but potentiated by CaCl2 (0.1–2 mM). The effect of phosphate on the surface charge and cadmium (Cd) adsorption was examined in seven soils that varied in their variable-charge components. The effect of phosphate on immobilization and phytoavailability of Cd from one of the soils, treated with various levels of Cd (0–10 mg Cd kg–1 soil), was further evaluated using mustard (Brassica juncea L.) plants. Cadmium immobilization in soil was evaluated by a chemical fractionation scheme. Addition of phosphate, as KH2PO4, increased the pH, negative charge and Cd adsorption by the soils. Of the seven soils examined, the three allophanic soils (i.e., Egmont, Patua and Ramiha) exhibited greater increases in phosphate-induced pH, negative charge and Cd2+ adsorption over the other four non-allophanic soils (i.e., Ballantrae, Foxton, Manawatu ad Tokomaru). Increasing addition of Cd enhanced Cd concentration in plants, resulting in decreased plant growth (i.e., phytotoxicity). Addition of phosphate effectively reduced the phytotoxicity of Cd. There was a significant inverse relationship between dry matter yield and Cd concentration in soil solution. Addition of phosphate decreased the concentration of the soluble + exchangeable Cd fraction but increased the concentration of inorganic-bound Cd fraction in soil. The phosphate-induced alleviation of Cd phytotoxicity can be attributed primarily to Cd immobilization due to increases in pH and surface charge.  相似文献
