Numeric abundance, identity, and pH preferences of methanotrophic Gammaproteobacteria (type I methanotrophs) inhabiting the northern acidic wetlands were studied. The rates of methane oxidation by peat samples from six wetlands of European Northern Russia (pH 3.9–4.7) varied from 0.04 to 0.60 μg CH4 g?1 peat h?1. The number of cells revealed by hybridization with fluorochrome labeled probes M84 + M705 specific for type I methanotrophs was 0.05–2.16 × 105 cells g?1 dry peat, i.e., 0.4–12.5% of the total number of methanotrophs and 0.004–0.39% of the total number of bacteria. Analysis of the fragments of the pmoA gene encoding particulate methane monooxygenase revealed predominance of the genus Methylocystis (92% of the clones) in the studied sample of acidic peat, while the proportion of the pmoA sequences of type I methanotrophs was insignificant (8%). PCR amplification of the 16S rRNA gene fragments of type I methanotrophs with TypeIF-Type IR primers had low specificity, since only three sequences out of 53 analyzed belonged to methanotrophs and exhibited 93–99% similarity to those of Methylovulum, Methylomonas, and Methylobacter species. Isolates of type I methanotrophs obtained from peat (strains SH10 and 83A5) were identified as members of the species Methylomonas paludis and Methylovulum miyakonense, respectively. Only Methylomonas paludis SH10 was capable of growth in acidic media (pH range for growth 3.8–7.2 with the optimum at pH 5.8–6.2), while Methylovulum miyakonense 83A5 exhibited the typical growth characteristics of neutrophilic methanotrophs (pH range for growth 5.5–8.0 with the optimum at pH 6.5–7.5).  相似文献

Three identical lab-scale biocovers were packed with an engineered soil (BC 1), tobermolite only (BC 2), and a mixture of the soil and tobermolite (BC 3), and were operated at an inlet load of 338–400 g-CH4 m?2 d?1 and a space velocity of 0.12 h?1. The methane removal capacity was 293 ± 47 g-CH4 m?2 d?1 in steady state in the BC 3, which was significantly higher than those in the BC 1 and BC 2 (106 ± 24 and 114 ± 48 g-CH4 m?2 d?1, respectively). Quantitative PCR indicated that bacterial and methanotrophic densities (6.62–6.78 × 107 16S rDNA gene copy number g-dry sample?1 and 1.37–2.23 × 107 pmoA gene copy number g-dry sample?1 in the BC 1 and BC 3, respectively) were significantly higher than those in the BC 2. Ribosomal tag pyrosequencing showed that methanotrophs comprised approximately 60 % of the bacterial community in the BC 2 and BC 3, while they only comprised 43 % in the BC 1. The engineered soil favored the growth of total bacteria including methanotrophs, while the presence of tobermolite enhanced the relative abundance of methanotrophs, resulting in an improved habitat for methanotrophs as well as greater methane mitigation performance in the mixture. Moreover, a batch experiment indicated that the soil and tobermolite mixture could display a stable methane oxidation level over wide temperature (20–40 °C, at least 38 μmol g-dry sample?1 h?1) and pH (5–8, at least 61 μmol g-dry sample?1 h?1) ranges. A robust, naturally evolving methanotrophic community in landfill cover soil (LFCS) can be the simplest way to mitigate landfill methane emission. In this study, bacterial community composition in LFCS and methane oxidation potential of enriched methanotrophic consortium, in comparison to that of axenic Methylosinus sporium, was investigated. Growth and methane oxidation of the consortium was studied in liquid phase batch experiments under varying temperature (20–40°C), pH (5–10), headspace CO2, and in presence of porous adsorbent (1.3 cm3 sponge cubes). The 16S rRNA gene analysis revealed presence of both type-I and type-II methanotrophs along with few obligate methylotroph in LFCS. Though the optimal growth condition of the consortium was at 30°C and pH 7, it was more resilient in comparison to M. sporium. With increasing availability of porous adsorbent, methane consumption by the consortium was significantly improved (p < 0.001) reaching a maximum specific methane oxidation rate of 11.4 μmol mg?1 biomass h?1. Thus, inducing naturally thriving methanotrophs in LFCS is a better alternative to axenic methanotrophic culture in methane emission management.  相似文献

Two axenic, in vitro liquid suspension cultures were established for Agardhiella subulata (C. Agardh) Kraft et Wynne, and their growth characteristics were compared. This study illustrated how reliable routes for the development of suspension cultures of macrophytic red algae of terete thallus morphology can be achieved for biotechnology applications. Undifferentiated filament clumps of 2–8 mm diameter were established by induction of callus-like tissue from thallus explants, and lightly branched microplantlets of 2–10 mm length were established by regeneration of filament clumps. The filament clumps were susceptible to regeneration. Adventitious shoot formation was reliably induced from 40% to 70% of the filament clumps by gentle mixing at 100 rev min?1 on an orbital shaker. The specific growth rate of the microplantlets was higher than the filament clumps in nonagitated well plate culture (4%–6% per day for microplantlets vs. 2%–3% per day for filament clumps) at 24° C and 8–36 μmol photons·m?2·s?1 irradiance (10:14 h LD cycle) when grown on ASP12 artificial seawater medium at pH 8.6–8.9 with 20%–25% per day medium replacement. Oxygen evolution rate vs. irradiance measurements showed that relative to the filament clumps, microplantlets had a higher maximum specific oxygen evolution rate (Po,max= 0.181 ± 0.035 vs. 0.130 ± 0.023 mmol O2·g?1 dry cell mass·h?1), but comparable respiration rate (Qo= 0.040 ± 0.013 vs. 0.033 ± 0.017 mmol O2·g?1 dry cell mass·h?1), compensation point (Ic= 3.8 ± 2.4 vs. 5.7 ± 1.2 μmol photons·m?2·s?1), and light intensity at 63.2% of saturation (Ik= 17.5 ± 3.9 vs. 14.9 ± 2.6 μmol photons·m?2·s?1). The microplantlet culture was more suitable for suspension culture development than the filament clump culture because it was morphologically stable and exhibited higher growth rates.  相似文献

In spring 2005, monthly sampling was carried out at a sublittoral site near Tautra Island. Microphytobenthic identification, abundance (ABU), and biomass (BIOM), were performed by microscopic analyses. Bacillariophyceae accounted for 67% of the total ABU, and phytoflagellates constituted 30%. The diatom floristic list consisted of 38 genera and 94 species. Intact light‐harvesting pigments chl a, chl c, and fucoxanthin and their derivatives were identified and quantified by HPLC. Photoprotective carotenoids were also observed (only as diadinoxanthin; no diatoxanthin was detected). Average fucoxanthin content was 4.57 ± 0.45 μg fucoxanthin · g sediment dry mass?1, while the mean chl a concentration was 2.48 ± 0.15 μg · g?1 dry mass. Both the high fucoxanthin:chl a ratio (considering nondegraded forms) and low amounts of photoprotective carotenoids indicated that the benthic microalgal community was adapted to low light. Microphytobenthic primary production was estimated in situ (MPPs, from 0.15 to 1.28 mg C · m?2 · h?1) and in the laboratory (MPPp, from 6.79 to 34.70 mg C · m?2 · h?1 under light saturation) as 14C assimilation; in April it was additionally estimated from O2‐microelectrode studies (MPPO2) along with the community respiration. MPPO2 and the community respiration equaled 22.9 ± 7.0 and 7.4 ± 1.8 mg C · m?2 · h?1, respectively. A doubling of BIOM from April to June in parallel with a decreasing photosynthetic activity per unit chl a led us to suggest that the microphytobenthic community was sustained by heterotrophic metabolism during this period.  相似文献

Abstract.Oxygen uptake and carbon dioxide release at 28°C were determined in worker castes of twenty-six species of forest termites from the Danum Valley Conservation Area, south-east Sabah, by Warburg manometry. Metabolic rate varied inversely with body weight in a suite of soil-, wood/soil- and wood-feeding species, giving a slope (in a log–log plot) of – 0.63. However, a number of large species, actively foraging forms such as Macrotermes malaccensis, M. gilvus, Havilanditermes atripennis and Hospitalitermes hospitalis, but also the wood-feeding Schedorhinotermes sarawakensis, showed an oxygen consumption greater than expected for their body weight. Rates of methane emission were above 0.100 μmol g–1 h–1 in seventeen species, with very high fluxes in two wood/soil-feeders, Termes borneensis (0.546 ± 0.163 μmol g–1 h–1) and Prohamitermes mirabilis (0.303 ± 0.123 μmol g–1 h–1). Of the fifteen remaining species, seven were soil-feeders, five were wood-feeders, two were wood/litter-feeders and a single species fed on lichen and moss. Low or negligible CH4 emissions (< 0.100 μmol g–1 h–1) were observed in three other species, all wood-feeders. An apparent respiratory quotient (RQapp) was calculated using xCO2 and xO2 (corrected for methane emission, but not hydrogen). Mean RQapp was at or above 1.00 in eleven species and between 0.95 and 1.00 in a further six species, the two sets of species together representing all trophic groups, including lichen-feeders. This is argued to be consistent with carbohydrate being the principal substrate supporting respiration.  相似文献

A complex system of muddy fluid-discharging and methane (CH4)-releasing seeps was discovered in a valley of the river Mukhrinskaya, one of the small rivers of the Irtysh Basin, West Siberia. CH4 flux from most (90%) of these gas ebullition sites did not exceed 1.45 g CH4 h−1, while some seeps emitted up to 5.54 g CH4 h−1. The δ13C value of methane released from these seeps varied between −71.1 and −71.3‰, suggesting its biogenic origin. Although the seeps were characterized by low in situ temperatures (3.5 to 5°C), relatively high rates of methane oxidation (15.5 to 15.9 nmol CH4 ml−1 day−1) were measured in mud samples. Fluorescence in situ hybridization detected 107 methanotrophic bacteria (MB) per g of mud (dry weight), which accounted for up to 20.5% of total bacterial cell counts. Most (95.8 to 99.3%) methanotroph cells were type I (gammaproteobacterial) MB. The diversity of methanotrophs in this habitat was further assessed by pyrosequencing of pmoA genes, encoding particulate methane monooxygenase. A total of 53,828 pmoA gene sequences of seep-inhabiting methanotrophs were retrieved and analyzed. Nearly all of these sequences affiliated with type I MB, including the Methylobacter-Methylovulum-Methylosoma group, lake cluster 2, and several as-yet-uncharacterized methanotroph clades. Apparently, microbial communities attenuating methane fluxes from these local but strong CH4 sources in floodplains of high-latitude rivers have a large proportion of potentially novel, psychrotolerant methanotrophs, thereby providing a challenge for future isolation studies.  相似文献

Telemetered heart rate (fH) was examined as an indicator of activity and oxygen consumption rate (VO2) in adult, cultivated, Atlantic salmon, Salmo salar L. Heart rate was measured during sustained swimming in a flume for six fish at 10° C [mean weight, 1114 g; mean fork length (f. l.), 50·6 cm] and seven fish at 15° C (mean weight, 1119 g; mean f. l., 50·7 cm) at speeds of up to 2·2 body lengths/s. Semi–logarithmic relationships between heart rate and swimming speed were obtained at both temperatures. Spontaneously swimming fish in still water exhibited characteristic heart rate increases associated with activity. Heart rate and Vo2 were monitored simultaneously in a 575–1 circular respirometer for six fish (three male, three female) at 4° C (mean weight, 1804 g; mean F. L., 62· cm) and six fish (three male, three female) at 10° C (mean weight, 2045 g; mean f. l., 63·2 cm) during spontaneous but unquantified activity. Linear regressions were obtained by transforming data for both fH and Vo2 to log values. At each temperature, slopes of the regressions between fH and Vo2 for individual fishes were not significantly different, but in some cases elevations were. All differences in elevation were between male and female fish. There were no significant differences in regression slope or elevation for fish of the same sex at the two temperatures and so regressions were calculated for the sexes, pooling data from 4 and 10° C. There was no significant difference in the mean ± S. D. Vo2 between the sexes at 4° C (male, 66·0 ± 59·6 mgO2 kg?1 h?1; female, 88·0 ± 60·1 mgO2 kg?1 h?1) or 10° C (male, 166·2 ± 115·4 mgO2 kg?1 h?1; female, 169·2 ± 111–1 mgO2 kg?1h?1). Resting Vo2 (x?± s. d.) at 4°C was 36·7 ± 8.4 mgO2 kg?1 h?1, and 10° C was 72·8 ± 11·9 mgO2 kg?1 h?1. Maximum Vo2 (x?± S. D.) at 4° C was 250·6 ± 40·2 mgO2 kg?1 h?1, and at 10° C was 423·6 ± 25·2 mgO2 kg?1 h?1. Heart rate appears to be a useful indicator of metabolic rate over the temperature range examined, for the cultivated fish studied, but it is possible that the relationship for wild fish may differ.  相似文献

Aerobic methane oxidation has been mostly studied in environments with moderate to low temperatures. However, the process also occurs in terrestrial thermal springs, where little research on the subject has been done to date. The potential activity of methane oxidation and diversity of aerobic methanotrophic bacteria were studied in sediments of thermal springs with various chemical and physical properties, sampled across the Kunashir Island, the Kuriles archipelago. Activity was measured by means of the radioisotope tracer technique utilizing 14C-labeled methane. Biodiversity assessments were based on the particulate methane monooxygenase (pmoA) gene, which is found in all known thermophilic and thermotolerant methanotrophs. We demonstrated the possibility of methane oxidation in springs with temperature exceeding 74 °C, and the most intensive methane uptake was shown in springs with temperatures about 46 °C. PmoA was detected in 19 out of 30 springs investigated and the number of pmoA gene copies varied between 104 and 106 copies per ml of sediment. Phylogenetic analysis of PmoA sequences revealed the presence of methanotrophs from both the Alpha- and Gammaproteobacteria. Our results suggest that methanotrophs inhabiting thermal springs with temperature exceeding 50 °C may represent novel thermophilic and thermotolerant species of the genera Methylocystis and Methylothermus, as well as previously undescribed Gammaproteobacteria.  相似文献

Nitrite-dependent anaerobic methane oxidation (n-damo) process, mediated by Candidatus Methylomirabilis oxyfera of the candidate phylum NC10, was discovered recently which plays an important role in coupling the global nitrogen and carbon cycles. However, the distribution and diversity of this new anaerobic methane-oxidizing microorganism have not been investigated in desert lakes yet. The present study successfully retrieved n-damo bacterial 16S rRNA and pmoA gene sequences using PCR technique from lakes in Badain Jaran Desert of China. Phylogenetic analyses showed that n-damo bacteria widely occurred in brine and freshwater lakes on the desert with high diversity, including both sediment and water samples. The results of quantitative PCR indicated that the abundance of the 16S rRNA gene in lake sediments varied from 1.12?±?0.68?×?105 to 1.64?±?0.70?×?105 copies g?1 (dry weight), while that in water samples per milliliter was generally one order of magnitude lower than sediments. Correlation analyses suggested that n-damo bacterial abundance and diversity strongly depended on salinity. In lake sediments, the distribution, abundance, and diversity of n-damo bacteria were significantly associated with depth due to the concentration gradient of the NOx- and ammonium. This study provided new insights into both the n-damo community patterns and its interaction with ambient environmental factors in the desert lake ecosystem.  相似文献

Biological treatment using methane-oxidizing bacteria (MOB) immobilized on six porous carrier materials have been used to mitigate methane emission. Experiments were performed with different MOB inoculated in building materials at high (~20 % (v/v)) and low (~100 ppmv) methane mixing ratios. Methylocystis parvus in autoclaved aerated concrete (AAC) exhibited the highest methane removal rate at high (28.5?±?3.8 μg CH4 g?1 building material h?1) and low (1.7?±?0.4 μg CH4 g?1 building material h?1) methane mixing ratio. Due to the higher volume of pores with diameter >5 μm compared to other materials tested, AAC was able to adsorb more bacteria which might explain for the higher methane removal observed. The total methane and carbon dioxide-carbon in the headspace was decreased for 65.2?±?10.9 % when M. parvus in Ytong was incubated for 100 h. This study showed that immobilized MOB on building materials could be used to remove methane from the air and also act as carbon sink.  相似文献

Measurements of bimodal oxygen uptake have been made in a freshwater air-breathing fish,Notopterus chitala at 29.0±1(S.D.)°C. xhe mean oxygen uptake from continuously flowing water without any access to air, was found to be 3.58±0.37 (S.E.) ml O2 · h?1 and 56.84+4.29 (S.E.) ml O2 · kg?1 · h?1 for a fish weighing 66.92 + 11.27 (S.E.) g body weight. In still water with access to air, the mean oxygen uptake through the gills were recorded to be 2.49 ± 0.31 (S.E.) ml O2 · h?1 and 38.78 ± 1.92 (S.E.) ml O2 · kg?1 · h?1 and through the accessory respiratory organs (swim-bladder) 6.04±0.87 (S.E.) ml O2 · h?1 and 92.32±2.91 (S.E.) ml O2 · kg?1 · h?1 for a fish averaging 66.92±11.27 (S.E.) g. Out of the total oxygen uptake (131.10 ml O2 · kg?1 · h?1), about 70% was obtained through the aerial route and the remainder 30% through the gills.  相似文献

The effects of ethylene (C2H4) on tetrasporogenesis of the red seaweed Pterocladiella capillacea (S. G. Gmelin) Bornet were investigated. Ethylene is a gaseous hormone that is involved in a variety of physiological processes (e.g., flowering, fruit abscission) in higher plants. To study the effects of ethylene on the reproduction of the red seaweed P. capillacea, immature tetrasporophytic thalli were exposed to a flow of ethylene for different time periods. Maximum maturation of tetrasporangia was observed at 7 d in thalli exposed to ethylene for 15 min. This maturation was accompanied by a significant increase in the free fraction of putrescine (Put) and a 5‐fold increase in the level of total RNA. These changes were specifically due to ethylene since they were blocked by the presence of the ethylene perception inhibitor silver thiosulphate (STS). Moreover, P. capillacea was determined to produce ethylene at a rate of 1.12 ± 0.06 nmol ethylene · h?1· g?1 fresh weight (fwt) with specific activities for 1‐aminocyclopropane‐1‐acrylic acid (ACC) synthase of 11.21 ± 1.19 nmol ethylene · h?1· mg?1 protein and for ACC oxidase (ACO) of 7.12 ± 0.11 nmol ethylene · h?1· mg?1 protein. We conclude that ethylene may indeed be a physiological regulator of tetrasporogenesis in this red seaweed.  相似文献

To study the effects of elevated ozone concentration on methane dynamics and a sedge species, Eriophorum vaginatum, we exposed peatland microcosms, isolated by coring from an oligotrophic pine fen, to double ambient ozone concentration in an open‐air ozone exposure field for four growing seasons. The field consists of eight circular plots of which four were fumigated with elevated ozone concentration and four were ambient controls. At the latter part of the first growing season (week 33, 2003), the methane emission was 159±14 mg CH4 m?2 day?1 (mean±SE) in the ozone treatment and 214±8 mg CH4 m?2 day?1 under the ambient control. However, towards the end of the experiment the ozone treatment slightly, but consistently, enhanced the methane emission. At the end of the third growing season (2005), microbial biomass (estimated by phospholipid fatty acid biomarkers) was higher in peat exposed to ozone (1975±108 nmol g?1 dw) than in peat of the control microcosms (1589±115 nmol g?1 dw). The concentrations of organic acids in peat pore water showed a similar trend. Elevated ozone did not affect the shoot length or the structure of the sedge E. vaginatum leaves but it slightly increased the total number of sedge leaves towards the end of the experiment. Our results indicate that elevated ozone concentration enhances the general growth conditions of microbes in peat by increasing their substrate availability. However, the methane production did not reflect the increase in the concentration of organic acids, probably because hydrogenotrophic methane production dominated in the peat studied. Although, we used isolated peatland microcosms with limited size as study material, we did not find experimental factors that could have hampered the basic conclusions on the effects of ozone.  相似文献

To evaluate the effect of cultivation, nitrogen fertilizer, and set aside on CH4 uptake after drained marshland was converted into agricultural fields, CH4 fluxes and CH4 concentrations in soil gas were in situ measured in a drained marsh soil, a set‐aside cultivated soil, and cultivated soils in Sanjiang Plain of Northeast China in August 2001. Over the measuring period, the highest CH4 uptake rate was 120.7±6.2 μg CH4 m?2 h?1 in the drained marsh soil and the lowest was 29.5±4.9 μg CH4 m?2 h?1 in the set‐aside cultivated soil, showing that there was no significant recovery of CH4 uptake ability 5 years after cultivation activity was stopped. CH4 uptake rates were significantly less in the cultivated soils than in the drained marsh soil by 30.1–74.6%, which resulted mainly from cultivation and partly from nitrogen addition. A significantly negative correlation between CH4 flux and bulk density in the cultivated soils tilled by machine suggests that cultivation reduced CH4 uptake through compaction, because of the enhanced diffusion resistance for CH4 and O2. Nitrogen fertilization slowly reduced but persistently affected CH4 uptake even after long‐term application of nitrogen.  相似文献

The distribution of the enzyme nitrate reductase (NR) within the thallus of the brown alga Laminaria digitata (Huds.) Lamour is described for plants sampled from the east coast of Scotland in May and June when growth rates are at a maximum. Highest NR activities (≈ 0.2 μmol NO3? reduced·g?1 wet wt·h?1) occurred in the mature blade. NR activities declined towards the basal meristematic region of the blade. Activities in the stipe and holdfast were also low, being between 0.05 and 0.07 μmol NO3? reduced·g?1 wet wt·h?1. The activities of the enzyme glutamine synthetase (GS), which is important in the assimilation of NH4+, showed a similar distribution within the blade to those of NR.The transverse profile of NR activity in the stipe exhibited a decline from the outer to the inner tissues. Maximum activities (0.13 μmol NO3? reduced·g?1 wet wt·h?1) occurred in the meristoderm, while those of the cortex and medulla were 0.04 and 0.01 μmol NO3? reduced·g?1 wet wt·h?1 respectively.These data indicate that most of the NO3? assimilation occurs in the mature blade rather than in the meristematic tissue where there is a high nitrogen demand for growth. The data are consistent with the maintenance of meristematic growth by the internal transport of organic nitrogen from the mature blade.  相似文献

The experiment was organized in a 3×2 factorial arrangement with three dietary fat blends and a basal (20 mg kg?1 diet) or supplemented (220 mg kg?1) level of α-tocopheryl acetate. Dietary vitamin E and monounsaturated to polyunsaturated fatty acid ratio (dietary MUFA/PUFA) affected muscle α-tocopherol concentration (α-tocopherol [log μg g?1]=0.18 (±0.105)+0.0034 (±0.0003)·dietary α-tocopherol [mg kg?1 diet] (P<0.0001)+0.39 (±0.122)·dietary MUFA/PUFA (P<0.0036)). An interaction between dietary α-tocopherol and dietary MUFA/PUFA exists for microsome α-tocopherol concentration (α-tocopherol [log μg g?1]=1.14 (±0.169) (P<0.0001)+0.0056 (±0.00099)·dietary α-tocopherol [mg kg?1 diet] (P<0.0001)+0.54 (±0.206)·dietary MUFA/PUFA (P<0.0131)?0.0033 (±0.0011)·dietary α-tocopherol [mg kg?1)]×dietary MUFA/PUFA (P<0.0067)), and hexanal concentration in meat (hexanal [ng·g?1]=14807.9 (±1489.8)?28.8 (±10.6) dietary α-tocopherol [mg·kg?1] (P<0.01)?8436.6 (±1701.6)·dietary MUFA/PUFA (P<0.001)+24.0 (±11.22)·dietary α-tocopherol·dietary MUFA/PUFA (P<0.0416)). It is concluded that partial substitution of dietary PUFA with MUFA lead to an increase in the concentration of α-tocopherol in muscle and microsome extracts. An interaction between dietary α-tocopherol and fatty acids exists, in which at low level of dietary vitamin E inclusion, a low MUFA/PUFA ratio leads to a reduction in the concentration of α-tocopherol in microsome extracts and a concentration of hexanal in meat above the expected values.  相似文献

Microbial oxidation is the only biological sink for atmospheric methane. We assessed seasonal changes in atmospheric methane oxidation and the underlying methanotrophic communities in grassland near Giessen (Germany), along a soil moisture gradient. Soil samples were taken from the surface layer (0–10 cm) of three sites in August 2007, November 2007, February 2008 and May 2008. The sites showed seasonal differences in hydrological parameters. Net uptake rates varied seasonally between 0 and 70 μg CH4 m−2 h−1. Greatest uptake rates coincided with lowest soil moisture in spring and summer. Over all sites and seasons, the methanotrophic communities were dominated by uncultivated methanotrophs. These formed a monophyletic cluster defined by the RA14, MHP and JR1 clades, referred to as upland soil cluster alphaproteobacteria (USCα)-like group. The copy numbers of pmoA genes ranged between 3.8 × 105–1.9 × 106 copies g−1 of soil. Temperature was positively correlated with CH4 uptake rates (P<0.001), but had no effect on methanotrophic population dynamics. The soil moisture was negatively correlated with CH4 uptake rates (P<0.001), but showed a positive correlation with changes in USCα-like diversity (P<0.001) and pmoA gene abundance (P<0.05). These were greatest at low net CH4 uptake rates during winter times and coincided with an overall increase in bacterial 16S rRNA gene abundances (P<0.05). Taken together, soil moisture had a significant but opposed effect on CH4 uptake rates and methanotrophic population dynamics, the latter being increasingly stimulated by soil moisture contents >50 vol% and primarily related to members of the MHP clade.  相似文献
