Plant-pathogen interactions involve sophisticated action and counteraction strategies from both parties. Plants can recognize pathogen derived molecules, such as conserved pathogen associated molecular patterns (PAMPs) and effector proteins, and subsequently activate PAMP-triggered immunity (PTI) and effector-triggered immunity (ETI), respectively. However, pathogens can evade such recognitions and suppress host immunity with effectors, causing effector-triggered susceptibility (ETS). The differences among PTI, ETS, and ETI have not been completely understood. Toward a better understanding of PTI, ETS, and ETI, we systematically examined various defense-related phenotypes of Arabidopsis infected with different Pseudomonas syringae pv. maculicola ES4326 strains, using the virulence strain DG3 to induce ETS, the avirulence strain DG34 that expresses avrRpm1 (recognized by the resistance protein RPM1) to induce ETI, and HrcC- that lacks the type three secretion system to activate PTI. We found that plants infected with different strains displayed dynamic differences in the accumulation of the defense signaling molecule salicylic acid, expression of the defense marker gene PR1, cell death formation, and accumulation/localization of the reactive oxygen species, H2O2. The differences between PTI, ETS, and ETI are dependent on the doses of the strains used. These data support the quantitative nature of PTI, ETS, and ETI and they also reveal qualitative differences between PTI, ETS, and ETI. Interestingly, we observed the induction of large cells in the infected leaves, most obviously with HrcC- at later infection stages. The enlarged cells have increased DNA content, suggesting a possible activation of endoreplication. Consistent with strong induction of abnormal cell growth by HrcC-, we found that the PTI elicitor flg22 also activates abnormal cell growth, depending on a functional flg22-receptor FLS2. Thus, our study has revealed a comprehensive picture of dynamic changes of defense phenotypes and cell fate determination during Arabidopsis-P. syringae interactions, contributing to a better understanding of plant defense mechanisms.  相似文献

The foraging ecology of hummingbirds involves the exploitation of a high number of patchily distributed flowers. This scenario seems to have influenced capabilities related to learning and memory, which help to avoid recently visited flowers and to allocate exploitation to the most rewarding flowers, once learning has occurred. We carried out two field experiments with the green‐backed firecrown hummingbird (Sephanoides sephaniodes, Trochilidae) in order to examine the ability of birds, first, to recall a nectar location, and secondly, to remember the location of the most rewarding flower among lower quality flowers. The first experiment showed that subjects were able to recall the location of nectar among flowers of identical appearance. In the second experiment, hummingbirds were also able to recall the location of the most rewarding nectar among less rewarding flowers with the same appearance. The results of this study suggest that S. sephaniodes can remember the location of the most rewarding patch, facilitating efficient exploitation of flowers in the absence of visual cues related to nectar quality.  相似文献

Territoriality is a potentially costly endeavor, and several mechanisms for mitigating the costs of territoriality have been investigated in the wild. For example, territory owners can reduce the costs of defending territory boundaries by prioritizing defense of the most valuable areas within territories, investing less energy in low quality areas. We staged pairwise encounters between adult male lizards on natural territories in the wild, to test whether male brown anoles, Anolis sagrei, would differentially defend certain regions of their territories over others. Based on our observations that male A. sagrei spend most of their time on elevated perches on tree trunks or branches compared with sites on the ground, we predicted that territory residents would respond more aggressively to territory invasions that took place on elevated perches than to invasions on the ground. We measured significant differences in the behavior of residents following invasion on the ground vs. on the elevated perches, and results partially supported our hypothesis. Males performed more displays and approached intruders more often when territory invasion took place on the ground, but were quicker to attack intruders that entered territories on elevated perches. Our hypothesis was only partially supported, potentially indicating that elevated perches are preferred as outposts to monitor valuable sites on the ground. Our study provides evidence that territory defense varies not just among individuals, but also within individuals at different locations in a territory.  相似文献

The population dynamics of Archaea after flooding of an Italian rice field soil were studied over 17 days. Anoxically incubated rice field soil slurries exhibited a typical sequence of reduction processes characterized by reduction of nitrate, Fe3+, and sulfate prior to the initiation of methane production. Archaeal population dynamics were followed using a dual approach involving molecular sequence retrieval and fingerprinting of small-subunit (SSU) rRNA genes. We retrieved archaeal sequences from four clone libraries (30 each) constructed for different time points (days 0, 1, 8, and 17) after flooding of the soil. The clones could be assigned to known methanogens (i.e., Methanosarcinaceae, Methanosaetaceae, Methanomicrobiaceae, and Methanobacteriaceae) and to novel euryarchaeotal (rice clusters I, II, and III) and crenarchaeotal (rice clusters IV and VI) lineages previously detected in anoxic rice field soil and on rice roots (R. Grosskopf, S. Stubner, and W. Liesack, Appl. Environ. Microbiol. 64:4983–4989, 1998). During the initiation of methanogenesis (days 0 to 17), we detected significant changes in the frequency of individual clones, especially of those affiliated with the Methanosaetaceae and Methanobacteriaceae. However, these findings could not be confirmed by terminal restriction fragment length polymorphism (T-RFLP) analysis of SSU rDNA amplicons. Most likely, the fluctuations in sequence composition of clone libraries resulted from cloning bias. Clonal SSU rRNA gene sequences were used to define operational taxonomic units (OTUs) for T-RFLP analysis, which were distinguished by group-specific TaqI restriction sites. Sequence analysis showed a high degree of conservation of TaqI restriction sites within the different archaeal lineages present in Italian rice field soil. Direct T-RFLP analysis of archaeal populations in rice field soil slurries revealed the presence of all archaeal lineages detected by cloning with a predominance of terminal restriction fragments characteristic of rice cluster I (389 bp), Methanosaetaceae (280 bp), and Methanosarcinaceae/rice cluster VI (182 bp). In general, the relative gene frequency of most detected OTUs remained rather constant over time during the first 17 days after flooding of the soil. Most minor OTUs (e.g., Methanomicrobiaceae and rice cluster III) and Methanosaetaceae did not change in relative frequency. Rice cluster I (37 to 30%) and to a lesser extent rice cluster IV as well as Methanobacteriaceae decreased over time. Only the relative abundance of Methanosarcinaceae (182 bp) increased, roughly doubling from 15 to 29% of total archaeal gene frequency within the first 11 days, which was positively correlated to the dynamics of acetate and formate concentrations. Our results indicate that a functionally dynamic ecosystem, a rice field soil after flooding, was linked to a relatively stable archaeal community structure.  相似文献

Woody encroachment, a spatially explicit process of land-cover change, is known to affect the biophysical and biogeochemical properties of ecosystems. However, little information is available on the impacts of woody encroachment on N oxide emissions from savanna regions. We combined hyperspectral remote sensing and field measurements to quantify spatial patterns and estimate regional fluxes of soil N oxide emissions as they covary with vegetation cover and soil type across a semiarid rangeland in north Texas. Soil nitric oxide (NO) emissions were highly correlated with Prosopis canopy cover, allowing the extrapolation of NO fluxes from hyperspectral observations of woody cover. NO emissions were highly variable, ranging from 0 to 550 kg NO-N km–2 y–1 across the region, with the lowest emissions from shallow clay soils and highest from deeper upland clay loams. An estimate of annual NO emissions based on remotely derived Prosopis cover, temperature, and precipitation was 160 kg NO-N km–2 y–1, almost twice that of the value derived from traditional averaging of field measurements. We conclude that relationships between NO emissions and remotely sensed structure and composition are advantageous for quantifying NO emissions at the regional scale. This study also provides new insight into the role of woody encroachment on biogeochemical processes that are highly variable and otherwise difficult to measure at the regional scale.  相似文献

Two potential strategies for acquiring territories are (1) fighting and taking over a territory from its owner, or (2) waiting until a territory owner dies and then taking its place. In this paper we explore territory acquisition using these two strategies, using a population dynamical model. Factors expected to affect the predominance of each strategy are injury rate, rate of successful territory takeover, birth and death rates on the territory, and birth and death rates while non-territorial. We explore the effects of these parameters on numbers of territorial and nonterritorial fighters and waiters. Waiters predominate when injury rate, birth rate of nonterritorial individuals, and death rate of territory owners are high. Fighters predominate when rate of successful territory takeover, death rate of nonterritorial individuals, and birth rate of territory owners are high. Summary We present results of experiments designed to identify floral characteristics that influence patterns of pollen carryover by hummingbirds visiting Ipomopsis aggregata flowers. We used fluorescent dye powders as pollen analogues. For all four experimental treatments considered, amounts of dye deposited on recipient stigmas declined linearly as a function of flower position in a visitation sequence. The decline was significantly steeper when recipient flowers had pollen-carrying anthers than when they did not; whereas degree of stigma clogging and presence or absence of empty anthers did not influence carryover. From this we conclude that presence of pollen on recipient flowers significantly reduces the average number of subsequent flowers reached by donor pollen. We discuss mechanisms for this effect and its significance for the evolution of floral structure.  相似文献

Infectious diseases have been paramount among the threats to health and survival throughout human evolutionary history. Natural selection is therefore expected to act strongly on host defense genes, particularly on innate immunity genes whose products mediate the direct interaction between the host and the microbial environment. In insects and mammals, the Toll-like receptors (TLRs) appear to play a major role in initiating innate immune responses against microbes. In humans, however, it has been speculated that the set of TLRs could be redundant for protective immunity. We investigated how natural selection has acted upon human TLRs, as an approach to assess their level of biological redundancy. We sequenced the ten human TLRs in a panel of 158 individuals from various populations worldwide and found that the intracellular TLRs—activated by nucleic acids and particularly specialized in viral recognition—have evolved under strong purifying selection, indicating their essential non-redundant role in host survival. Conversely, the selective constraints on the TLRs expressed on the cell surface—activated by compounds other than nucleic acids—have been much more relaxed, with higher rates of damaging nonsynonymous and stop mutations tolerated, suggesting their higher redundancy. Finally, we tested whether TLRs have experienced spatially-varying selection in human populations and found that the region encompassing TLR10-TLR1-TLR6 has been the target of recent positive selection among non-Africans. Our findings indicate that the different TLRs differ in their immunological redundancy, reflecting their distinct contributions to host defense. The insights gained in this study foster new hypotheses to be tested in clinical and epidemiological genetics of infectious disease.  相似文献

Coding plays a universal and pervasive role in biological organization, in forms such as genetic coding (DNA to protein translation), RNA processing, gene regulation, protein modification, cell signalling, immune responses, epigenetic development and natural language. Nevertheless, the ways and means by which organic codes are formed and used are still poorly understood. A formal model is presented in this paper to investigate the emergence of conventional codes among code users. The relationship between the formation and the usage of codes is discussed, and a biological mechanism involving coding is identified in the context of the immune system.  相似文献

Summary Observations were made on the nectar-feeding of hummingbirds in three areas of natural forest in the coastal mountains of southeastern Brazil. Five of the seven hummingbird species observed are endemic to the region. The observations are summarised and discussed, and limited comparison is made with the feeding ecology of Andean hummingbirds. The data suggest that two plant families of prime importance in temperate forest in the Andes, the Rubiaceae and Ericaceae, are unimportant in southeastern Brazilian montane forest. The close fit between the bill ofPhaethornis eurynome and the corollas of three species of plants suggests coevolution. The data presented are preliminary and incomplete, and indicate the need for more thorough research into the relationships between hummingbirds and plants in this region.
Kolibris und die von ihnen besuchten Blüten im Küstengebirge Südostbrasiliens
Zusammenfassung Die Beobachtungen über blütenbesuchende Kolibris stammen aus drei Naturwaldgebieten im Küstengebirge Südostbrasiliens. 5 der 7 beobachteten Kolibriarten sind in dieser Region endemisch. Die Zusammenfassung der Beobachtungen gestattet unter Einschränkung gewisse Vergleiche mit der Nahrungsökologie andiner Kolibris. Die Daten machen wahrscheinlich, daß Rubiaceae und Ericaceae, zwei Pflanzenfamilien von vorrangiger Bedeutung in den Wäldern der gemäßigten Zone der Anden, im südostbrasilianischen Bergwald unwichtig sind. Die enge Anpassung des Schnabels vonPhaethornis eurynome an die Gestalt der Blütenkrone dreier Pflanzenarten läßt Koevolution vermuten. Die Beobachtungen sind jedoch unvollständig und forden weitere intensive Untersuchung der Beziehungen zwischen Kolibris und Pflanzen in den bearbeiteten Gebieten.

The interplay between pathogen effectors, their host targets, and cognate recognition proteins provides various opportunities for antagonistic cycles of selection acting on plant and pathogen to achieve or abrogate resistance, respectively. Selection has previously been shown to maintain diversity in plant proteins involved in pathogen recognition and some of their cognate pathogen effectors. We analyzed the signatures of selection on 10 Arabidopsis thaliana genes encoding defense signal transduction proteins in plants, which are potential targets of pathogen effectors. There was insufficient evidence to reject neutral evolution for 6 genes encoding signaling components consistent with these proteins not being targets of effectors and/or indicative of constraints on their ability to coevolve with pathogen effectors. Functional constraints on effector targets may have provided the driving selective force for the evolution of guard proteins. PBS1, a known target of an effector, showed little variation but is known to be monitored by a variable guard protein. Evidence of selection maintaining diversity was present at NPR1, PAD4, and EDS1. Differences in the signatures of selection observed may reflect the numbers of effectors that target a particular protein, the presence or absence of a cognate guard protein, as well as functional constraints imposed by biochemical activities or interactions with plant proteins.  相似文献

1998年在河北南皮棉区转Bt基因棉棉田和常规棉综防田的生物群落变化的研究表明,与常规棉综防田相比,转Bt基因棉棉田的节肢动物种类增加30.7%,功能团多样性增加66.9%,天敌类和中性节肢动物类的丰富度分别增加97.6%和158.0%,害虫类的丰富度减少45.7%。就功能团的种群数量来看,主要害虫棉铃虫、蚜虫和蓟马种群数量分别减少98.9%、69.0%和72.6%;而次要害虫害蝽、粉虱、叶蝉类昆虫的种群数量分别增加了2.2、3.2和14.8倍,棉花生长中期,转Bt基因棉棉田中的基位物种丰富度大于中位和顶位物种;常规棉田中的差异则更大,出现天敌控制“空缺”。综合分析表明,转Bt基因棉棉田生态系统比常规棉综防田稳定;转Bt基因棉棉田的中性节肢动物和次要害虫在害虫-天敌的营养链中起到了重要的调控作用。  相似文献
