珍稀药用真菌——樟芝深层发酵培养条件的优化   总被引:1,自引:0,他引:1  
对樟芝深层发酵培养基进行了筛选,并在此基础上对发酵条件进行了优化。以樟芝深层发酵菌丝体三萜产量为主要目标产物,确定发酵培养条件为:40g/L葡萄糖,6g/L豆饼粉,1g/L K_2HPO_4,0.5g/L MgSO4,VB_1 100mg/L,自然pH,接种量为20%,装液量为100mL/250mL三角瓶,转速100r/min,26℃恒温培养6d,胞内三萜产量达15.25mg/100mL发酵液。  相似文献

以菌丝体生物量和多糖产量为主要指标,对桑黄(鲍氏层孔菌)的深层发酵条件进行了优化,通过单因素试验和四因素三水平正交试验筛选出了桑黄液体发酵的培养基,结果表明,最适培养基为:葡萄糖5g/L,玉米粉40g/L,豆饼粉20g/L,KH2PO41.5g/L,MgSO41g/L。进一步通过培养条件的优化,得到最适菌丝体生长的液体发酵条件为:培养温度28℃,摇床转速140r/min,pH值自然,接种量10%,装液量100mL(250mL三角瓶),发酵周期132h。  相似文献

采用响应曲面法对乳牛肝菌发酵培养基及发酵条件进行优化,以麦芽糖、蛋白胨和装液量3个因素为变量,分析乳牛肝菌液态发酵产菌丝体的最佳条件。实验结果表明,乳牛肝菌发酵的关键因子值分别为麦芽糖33.03g/L,蛋白胨6.95g/L,装液量为102.21mL/250mL时,可得到最大菌丝体生物量23.44g/L(干重)。  相似文献

对不同样品中的真菌进行了L-精氨酸高产菌株的筛选,从红曲米中分离出1株高产L-精氨酸菌株红曲霉H13,其初始发酵液中L-精氨酸产量为1.67 g/L.通过单因素实验对其液体发酵进行研究.结果表明,最佳碳源是可溶性淀粉,最佳浓度为10%,最佳氮源是蛋白胨,最佳浓度为3%,最佳pH值4.5,温度30℃,种子培养时间为18 h,摇瓶装液量50 mL/250 mL.转速为120 r/min,发酵时间7d.在此条件下进行发酵培养,所得发酵液产L-精氨酸4.08 g/L,菌丝体干重为10.32 g/L,菌丝体中L-精氨酸含量为24.17 mg/g.  相似文献

从180余份海水、海泥样品中筛选得到60株产海藻糖较高的菌株,编号为2-14的菌株海藻糖产量最高,为127.9mg/g cell。对2-14菌株进行形态特征、培养特征及生理生化试验,鉴定该菌株为红酵母属(Rhodotorula sp.)。研究摇瓶发酵条件对红酵母海藻糖产量的影响,结果为:初始pH5.5,发酵温度28℃,装液量75mL(250mL三角瓶中)。采用优化后发酵条件红酵母海藻糖产量为193.3mg/g cell,优化前对照值为132.1mg/g cell,优化后的结果是优化前的1.46倍。在5L发酵罐中培养得到最佳发酵时间为54h,发酵罐培养发酵液中海藻糖含量最高达2.5g/L,为摇瓶培养的1.6倍。  相似文献

以红酵母菌株NZ-01为试验菌株,研究其发酵工艺与中试生产。采用摇瓶发酵优化的方式,研究培养基组分与发酵工艺条件对该菌发酵的影响,并进行中试放大生产。结果显示,该菌最适生长培养基组分为葡萄糖10g/L,蔗糖10g/L,酵母膏10g/L,牛肉膏2.5g/L;色素合成最适培养基组分为葡萄糖15g/L,蔗糖10g/L,酵母膏2.5g/L,牛肉膏5g/L。最适生长起始pH值为6.0,最适接种量为8%,生长周期为44h;最适色素合成起始pH值为7.0,最适色素合成接种量为8%,色素合成周期为48h。发酵优化后的色素产量3.88μg/mL较优化前1.71μg/mL提高了127%。中试产量达3.05μg/mL。红酵母菌NZ-01优化后的发酵条件可以应用于中试生产虾青素,有规模化生产应用潜力。  相似文献

目的是通过测定不同条件下类胡萝卜素的产量找出粘红酵母发酵生产类胡萝卜素的最优条件。探讨了不同碳源、氮源对粘红酵母菌体生长和色素形成的影响,并通过正交实验确定了最佳条件组合。实验结果表明,最适发酵培养条件为:蔗糖40g/L、酵母粉20g/L、转速150r/min、装液量30mL/500mL、发酵时间84h。在此条件下,粘红酵母摇瓶发酵的生物量、类胡萝卜素含量及产量分别达15.17g/L、718.6μg/g、10.9mg/L,依次比初始发酵提高了1倍、7.4倍和15.8倍。发酵过程动态分析表明,84h色素产量达最高峰。  相似文献

蝙蝠蛾拟青霉液体发酵工艺优化及菌丝体腺苷含量的检测   总被引:2,自引:0,他引:2  
通过摇瓶液体培养的方法,以菌丝体生物量为主要考察指标,筛选蝙蝠蛾拟青霉PH-2菌株液体发酵的培养基成分和条件。进一步采用正交试验,以菌丝体生物量和腺苷含量为联合指标,确定PH-2菌株液体发酵高产菌丝体和腺苷的培养基配方。通过试验获得的最佳配方为:玉米淀粉40g/L,葡萄糖5g/L,玉米浆25g/L,硫酸镁·7H2O 1g/L,磷酸二氢钾1g/L,硫酸锌1.5g/L,维生素B1 25mg/L;培养条件为:初始pH 6.0,装液量150mL/500mL,接种量8%(V/V),温度26℃,转速120r/min,培养7d。在该工艺下,菌丝体生物量达到25.2g/L。应用高效液相色谱(HPLC)法,检测到菌丝体中腺苷含量为2.76mg/g。该方法简便、快速、准确,可作为蝙蝠蛾拟青霉菌丝体或其相关产品中腺苷的检测手段。本工艺的优化试验获得了较高的菌丝体收率,菌丝体质量良好。  相似文献

The fermentation of Grifola frondosa was investigated in the shake flasks and a 5-L jar fermenter in batch and fed-batch modes. In the shake-flask experiments, the preferable mycelial growth and exopolysaccharide (EPS) production was observed at relatively low pH; maltose and glucose were preferred carbon sources for high mycelial production. The EPS was doubled after 13 d of cultivation when glucose was increased from 2% to 4%. Yeast extract (YE) (0.4%) in combination with corn steep powder (CSP) (0.6%) and YE (0.8%) in combination with CSP (1.2%) were preferred nitrogen sources for high mycelial production and EPS production, respectively. All plant oils tested significantly stimulate cell growth of G. frondosa but they failed to enhance EPS production. The EPS products usually consisted of two fractions of different molecular sizes varied by the plant oils used. The fed-batch fermentation by glucose feeding was performed when the glucose concentration in the medium was lower than 0.5% (5g/L), which greatly enhanced the accumulation of mycelial biomass and EPS; the mycelial biomass and EPS were 3.97g/L and 1.04g/L before glucose feeding, which reached 8.23g/L and 3.88g/L at 13 d of cultivation. In contrast, the mycelial biomass and EPS in the batch fermentation were 6.7g/L and 3.3g/L at 13 d of cultivation.  相似文献

The optimal fermentation medium and conditions for mycelial growth and water-soluble exo-polysaccharides production by Isaria farinosa B05 were investigated. The medium components and fermentation conditions were optimized according to the one at a time method, while the concentration of medium components was determined by the orthogonal matrix method. The results showed that the optimal fermentation medium was as follows: sucrose 3.5% (w/v), peptone 0.5%, yeast extract 0.2%, K(2)HPO(4) 0.1%, and MgSO(4) 0.05%. The suitable fermentation conditions were as follows: initial pH 7.0, temperature 25 degrees C, medium volume 75 mL/250 mL, inoculum volume 5% (v/v), time 5d. In such optimal nutrition and environmental conditions, the maximal mycelial yield was 2.124 g/100 mL after 4 day's fermentation, while maximal water-soluble exo-polysaccharides production reached 2.144 g/L after 5 day's fermentation.  相似文献

裸脚菇0612-9次级代谢产物具有强烈抗青绿霉活性,可作为微生物源防腐剂用于柑橘保藏,但是其发酵周期长,产出能耗大效率低。用摇瓶对裸脚菇0612-9的液体菌种培养基、培养条件进行优化并对优化后液体菌种接种种龄、接种量进行探索,最后用5L发酵罐进行放大发酵验证。取样计数测定菌丝球数量、过滤称重测定菌丝干重、HPLC监测活性物质Ⅱ的积累、牛津杯法评价抗青绿霉活性。经研究最佳碳源为玉米粉和麦芽糖,最佳氮源为蛋白胨,最佳液体菌种培养基组成为:玉米粉30g/L、麦芽糖10g/L、蛋白胨15g/L、KH2PO4 2g/L、MgSO4·7H2O 1g/L;最佳培养条件:起始pH 5、接种3×Ф7mm菌块、装液量100mL/250mL三角瓶、温度28℃、转速160r/min;优化前菌丝球数46个/10mL,菌丝干重0.28g/100mL,优化后菌球数达985个/10mL,菌丝干重达0.69g/100mL,分别为优化前的21.4倍、2.43倍;后续发酵使用种龄9d的液体菌种、接种量7.5%。优化后液体菌种在发酵罐中后续发酵周期从10d缩短至5d,缩短50%,产量比优化前提高8.28%。  相似文献

A species of mushroom, Termitomyces albuminosus, was cultured in liquid medium for production of chromium-enriched mycelium. The influence of chromium (Ⅲ ) on mycelial growth of T. albuminosus was investigated. An optimum medium composed of 5.6g/L yeast extract, 51.6g/L hydrolyzed rice, 2g/L KH2PO4, and 20mg/L chromium(Ⅲ ) with initial pH of 4.5 was obtained by using method of central composite design (CCD). After incubation of 84h, the maximal biomass of chromium-enriched mycelia reached 24.23g DMW(dried mycelial weight)/L with 272μg/g DMW chromium content in 500mL flasks containing 100mL medium with an inoculum of 8% on a shaker of 100r/min under an optimized cultivation condition at 28℃.  相似文献

采用均匀设计和单因子试验法,系统考察了圆红冬孢酵母菌(Rhodosporidiumtoruloides)在不同碳氮比条件下产油发酵情况以及添加无机盐对产油发酵的影响,通过均匀设计软件对二次多项回归方程求解及单因素分析得知在培养基组成分别为葡萄糖70g/L,硫酸铵0.1g/L,酵母粉0.75g/L,磷酸二氢钾0.4g/L,七水硫酸镁1.5g/L,初始pH6.0,在灭菌(121℃15min)后添加ZnSO41.91×10-6mmol/L、CaCl21.50mmol/L、MnCl21.22×10-4mmol/L、CuSO41.00×10-4mmol/L。发酵摇瓶装液量为250mL三角瓶装培养基50mL,接种量为10%(种龄28h)。在上述条件下,30℃振荡(200r/min)培养120h,所得菌体油脂含量高达76.1%,脂肪得率系数可达22.7。  相似文献

采用单因素和正交试验对筛选出的一株有潜力的生物防治菌——拟康氏木霉Trichoderma pseudokoningii的液态发酵条件进行优化,并进行100L发酵罐中试放大试验的研究。优化后的拟康氏木霉发酵控制参数为:培养基配方为麸皮40g/L,马铃薯50g/L,蔗糖20g/L,KH2PO4 0.5 g/L,MgSO4·7H2O 0.5g/L,CaCl2 0.25g/L;摇瓶发酵培养时间为6d,培养基的初始pH值为6.0- 7.0,发酵温度为28±1℃。通过对拟康氏木霉生长曲线的测定,确定其在发酵罐中培养的时间60h为宜,此时所获得的菌丝体干重为1.2694g/100ml发酵液。本研究结果为高效率、低成本、工业化生产具有生防作用的拟康氏木霉菌丝制剂提供了科学依据。  相似文献
