Flowering phenology of cleistogamous annual plants Impatiens noli-tangere and Persicaria thunbergii was studied with reference to their mortality in streamside environments in northern Japan. I. noli-tangere produced flower-buds and flowers from late June to October, whereas P. thunbergii did so from mid-August to October. Both species were often killed by rapid current and submergence after heavy rainfall,
which was unpredictable in the timing and intensity. P. thunbergii was more tolerant to rapid current and submergence than I. noli-tangere. Some individuals of I. noli-tangere were killed by herbivory from sawfly larvae in summer, but P. thunbergii individuals were seldom killed by herbivory. In consequence, the percentage of individuals that survived until the end of
September was higher in P. thunbergii than in I. noli-tangere. These situations are assumed to select a long reproductive life in I. noli-tangere and autumn flowering in P. thunbergii. 相似文献

Flowering timing is very important for the reproductive success of higher plants. However, effects of salt on plant flowering
and the underlying molecular mechanisms are largely unknown. Here, we show that salt stress delays flowering in Arabidopsis in a dose-dependent manner. Mild salt stress (≤50 mM NaCl) promoted and prolonged the vegetative growth, whereas high salinity
(≥100 mM NaCl) largely delayed or inhibited the transition from vegetative growth to reproductive development. The gibberellin
(GA)-pathway plays an important role in this phenotype, and application of exogenous GA could restore late flowering induced
by salt. In addition, the CONSTANS (CO)/FLOWERING LOCUS T (FT) module may also play a critical role in mediating the effects of salt on flowering. The mRNA abundance of CO was significantly reduced by salt stress in a dose-dependent manner. The constans (co-2) mutants did not respond to moderate salt stress, whereas over-expressing CO manifested no delay in flowering time in response to salinity. Expression of FT, SOC1 and LFY in the downstream of the pathways was also reduced by salt according to dose. Moreover, salt-sensitive mutant salt overly sensitive3 (sos3) exhibited greater sensitivity in flowering, further suggesting that ion disequilibrium mediates salt-induced late flowering.

Plants contain three classes of hemoglobin genes of which two, class 1 and class 2, have a structure similar to classical
vertebrate globins. We investigated the effect of silencing the class 1 non-symbiotic hemoglobin gene, GLB1, and the effect of overexpression of GLB1 or the class 2 non-symbiotic hemoglobin gene, GLB2, in Arabidopsis thaliana. Lines with GLB1 silencing had a significant delay of bolting and after bolting, shoots reverted to the rosette vegetative phase by formation
of aerial rosettes at lateral meristems. Lines with overexpression of GLB1 or GLB2 bolted earlier than wild type plants. By germinating the lines in a medium containing the nitric oxide (NO) donor, sodium
nitroprusside (SNP), it was demonstrated that both GLB1 and GLB2 promote bolting by antagonizing the effect of NO, suggesting that non-symbiotic plant hemoglobin controls bolting by scavenging
the floral transition signal molecule, NO. So far, NO scavenging has only been demonstrated for class 1 non-symbiotic hemoglobins.
A direct assay in Arabidopsis leaf cells shows that GLB1 as well as the class 2 non-symbiotic hemoglobin, GLB2, scavenge NO in vivo. NO has also been demonstrated to be a growth stimulating signal with an optimum at low concentrations. It was observed that
overexpression of either GLB1 or GLB2 shifts the optimum for NO growth stimulation to a higher concentration. In conclusion, we have found that expression of NO
scavenging plant hemoglobin is involved in the control of bolting in Arabidopsis. 相似文献

<Emphasis Type="Italic">Agrobacterium</Emphasis>-mediated genetic transformation of <Emphasis Type="Italic">Perilla frutescens</Emphasis> 总被引:3,自引:0,他引:3
A reproducible plant regeneration and an Agrobacterium tumefaciens-mediated genetic transformation protocol were developed for Perilla frutescens (perilla). The largest number of adventitious shoots were induced directly without an intervening callus phase from hypocotyl explants on MS medium supplemented with 3.0 mg/l 6-benzylaminopurine (BA). The effects of preculture and extent of cocultivation were examined by assaying -glucuronidase (GUS) activity in explants infected with A. tumefaciens strain EHA105 harboring the plasmid pIG121-Hm. The highest number of GUS-positive explants were obtained from hypocotyl explants cocultured for 3 days with Agrobacterium without precultivation. Transgenic perilla plants were regenerated and selected on MS basal medium supplemented with 3.0 mg/l BA, 125 mg/l kanamycin, and 500 mg/l carbenicillin. The transformants were confirmed by PCR of the neomycin phosphotransferase II gene and genomic Southern hybridization analysis of the hygromycin phosphotransferase gene. The frequency of transformation from hypocotyls was about 1.4%, and the transformants showed normal growth and sexual compatibility by producing progenies. 相似文献

Bigtooth maple (Acer grandidentatum) is a promising ornamental tree that is not widely used in managed landscapes. Tissue culture has not been used successfully
to propagate this taxon. We cultured single- and double-node explants from greenhouse-grown, 2-y old seedlings of bigtooth
maples, which are indigenous to New Mexico, Texas, and Utah, on Murashige–Skoog (MS), Linsmaier–Skoog (LS), Driver–Kuniyuki
Walnut (DKW), and Woody Plant (WPM) tissue culture media. Media affected shoot proliferation (P = 0.0242) but the zone of explant origin (P = 0.7594) did not. After four 30-d subcultures, explants on DKW media and WPM media produced 3.6 and 3.5 shoots per explant,
respectively. Sprouting rates were highest on DKW, making DKW the best overall media for shoot proliferation. Double-node
microshoots were rooted in vitro on DKW containing indole acetic acid (IAA). Microshoots represented six genotypes from three locations within Texas and New
Mexico. Rooting percentage increased up to 15% as IAA concentration increased (P = 0.0040). There was 100% survival of rooted microshoots in vented Phytatrays containing one perlite: one peat moss (v/v). We conclude that DKW can be used to proliferate microshoots, and IAA induces rooting in microshoots of bigtooth maple. 相似文献

Two experiments were performed to determine how application of the cytokinin benzyladenine (BA) influenced flowering in Doritaenopsis and Phalaenopsis orchid clones. In the first experiment, two vegetative orchid clones growing in 15-cm pots were transferred from a 28°C greenhouse
that inhibited flowering to a 23°C greenhouse for flower induction (day 0). A foliar spray (0.2 L m−2) containing BA at 100, 200, or 400 mg L−1 or 25, 50, or 100 mg L−1 each of BA and gibberellins A4 + A7 (BA+GA) was applied on days 0, 7, and 14. Plants treated with BA alone at 200 or 400 mg L−1 had a visible inflorescence 3–9 days earlier and had a mean of 0.7–3.5 more inflorescences and 3–8 more flowers per plant
than nontreated plants. The application of BA+GA had no effect on inflorescence number and total flower number at the rates
tested. In the second experiment, three orchid clones received a single foliar spray of BA at 200 mg L−1 at six time points relative to time of transfer from 29°C to 23°C (−1, 0, +1, +2, +4, or +6 weeks). A separate group of plants
received a BA application at week 0 but was maintained at 29°C. Inflorescence number was greatest in all three orchid clones
when plants were treated with BA 1 week after the temperature transfer. Plants that were sprayed with BA and maintained at
29°C did not initiate inflorescences. The promotion of flowering by the application of BA suggests that cytokinins at least
partially regulate inflorescence initiation of Doritaenopsis and Phalaenopsis, but its promotion is conditional and BA application cannot completely substitute for an inductive low temperature. 相似文献

The balloon flower (Platycodon grandiflorum) is a popular traditional medicinal plant used in Korea to treat conditions such as bronchitis, asthma, tuberculosis, diabetes,
and inflammatory diseases. Recently, immunopharmacological research identified triterpenoid and saponin as important active
compounds in P. grandiflorum. To study and extract these compounds and other metabolites from P. grandiflorum, a technique was developed for producing hairy root cultures, which are a reliable source of plant compounds. To achieve this,
the activity of Agrobacterium rhizogenes was exploited, which can transfer DNA segments into plant genomes after infecting them. In this study, the A. rhizogenes strain R1000 was determined that had the highest infection frequency (87.5%) and induced the most hairy roots per plant,
and the concentration of antibiotics (75 mg/l kanamycin) was elucidated for selection after transformation. Wild-type and
transgenic hairy roots contained various phenolic compounds, although both of them had similar concentrations of phenolic
compounds. In the future, the protocols described here should be useful for studying and extracting valuable metabolites such
as phenolic compounds from P. grandiflorum hairy root cultures. 相似文献

To ensure that the initiation of flowering occurs at the correct time of year, plants need to integrate a diverse range of
external and internal signals. In Arabidopsis, the photoperiodic flowering pathway is controlled by a set of regulators that include CONSTANS (CO). In addition, Arabidopsis plants also have a family of genes with homologies to CO known as CO-LIKE (COL) about which relatively little is known. In this paper, we describe the regulation and interactions of a novel member of
the family, COL5. The expression of COL5 is under circadian and diurnal regulation, but COL5 itself does not appear to affect circadian rhythms. COL5, like CO, is regulated by GIGANTEA. Furthermore, COL5 is expressed in the vascular tissue. Using COL5 over-expressing lines we show that, under short days, constitutive expression of COL5 affects flowering time and the expression of the floral integrator genes, FLOWERING LOCUS T and SUPPRESSOR OF OVEREXPRESSION OF CO 1. Constitutive expression of COL5 partially suppresses the late flowering phenotype of the co-mutant plants. However, plants with loss of COL5 function do not show altered flowering. Taken together, our results suggest that COL5 has COL activity, but may either not
have a role in regulating flowering in wild-type plants or may act redundantly with other flowering regulators.

Two repeated DNA sequences isolated from a partial genomic DNA library of Helianthus annuus, p HaS13 and p HaS211, were shown to represent portions of the int gene of a Ty3 /gypsy retroelement and of the RNase-Hgene of a Ty1 /copia retroelement, respectively. Southern blotting patterns obtained by hybridizing the two probes to BglII- or DraI-digested genomic DNA from different Helianthus species showed p HaS13 and p HaS211 were parts of dispersed repeats at least 8 and 7 kb in length, respectively, that were conserved in all species studied. Comparable hybridization patterns were obtained in all species with p HaS13. By contrast, the patterns obtained by hybridizing p HaS211 clearly differentiated annual species from perennials. The frequencies of p HaS13- and p HaS211-related sequences in different species were 4.3x10(4)-1.3x10(5) copies and 9.9x10(2)-8.1x10(3) copies per picogram of DNA, respectively. The frequency of p HaS13-related sequences varied widely within annual species, while no significant difference was observed among perennial species. Conversely, the frequency variation of p HaS211-related sequences was as large within annual species as within perennials. Sequences of both families were found to be dispersed along the length of all chromosomes in all species studied. However, Ty3 /gypsy-like sequences were localized preferentially at the centromeric regions, whereas Ty1/ copia-like sequences were less represented or absent around the centromeres and plentiful at the chromosome ends. These findings suggest that the two sequence families played a role in Helianthusgenome evolution and species divergence, evolved independently in the same genomic backgrounds and in annual or perennial species, and acquired different possible functions in the host genomes. 相似文献

Bremia lactucae Regel (Chromista, Peronosporaceae) is an economically destructive pathogen, which causes downy mildew disease on lettuce
(Lactuca sativa L.) worldwide. The ribosomal internal transcribed spacer (ITS) of Bremia lactucae isolates was analyzed for the first time. The ITS region of lettuce downy mildew was observed to have a size of 2458 bp;
thereby, having one of the longest ITS sizes recorded to date. The majority of the extremely large sized ITS2 length of 2086
was attributed to the additional presences of nine repetitive elements with lengths of 179–194 bp, which between them shared
the low homology of 48–69%. Comparison of the ITS2 sequences with the B. lactucae isolates from other host plants showed that isolates present on Lactuca sativa were distinct from those on L. indica var. laciniata, as well as Hemistepta and Youngia. We suggest the high degree of sequence heterogeneity exhibited in the ITS2 region of B. lactucae may warrant the specific detection and diagnosis of this destructive pathogen or its division into several distinct species. 相似文献

The warty cabbage Bunias orientalis is an invasive pest in much of central Europe, including much of Germany since the 1980s, whereas in other countries, such
as The Netherlands, it is a less common exotic species. Here, healthy larvae of Mamestra brassicae, which has been found feeding on B. orientalis plants in Germany, and larvae parasitized by one of its major larval endoparasitoids Microplitis mediator, were reared on both herbivore-induced and noninduced leaves of B. orientalis originating from single large populations growing in The Netherlands and central Germany. Herbivore performance was less
negatively affected than parasitoid performance by differences in plant quality. Development times in both M. brassicae and Mi. mediator were shorter on Dutch than German plants and also shorter on noninduced than induced plants. Moreover, survival and body
size of the parasitoid was more strongly affected by plant population and induction than survival of healthy M. brassicae. Chemical analyses of defensive secondary metabolites [glucosinolates (GS)] revealed that concentrations of the major GS sinalbin
were constitutively expressed in German plants whereas they were induced in Dutch plants. However, in separate feeding bioassays
in which preference for induced and noninduced leaves was compared separately, L3 instars of M. brassicae preferred noninduced German plants over Dutch plants but induced Dutch plants over German plants, revealing that changes
in primary metabolites or an unidentified non-GS compound mediates population-related differences in plant quality. The results
reveal asymmetric effects of plant quality in exotic plants on organisms in the second and third trophic level. 相似文献

Polygonum viviparum is one of the first ectomycorrhizal (EM) plant species colonising primary successional sites at the Rotmoos glacier forefront
(Tyrolean Alps, Austria). On a site with soil development of about 150 years (2,400 m above sea level), mycobionts of P. viviparum were identified by morphotyping and fungal ribosomal deoxyribonucleic acid internal transcribed spacer sequencing. For studying
seasonal dynamics and spatial heterogeneity, ectomycorrhizae were sampled on five plots during all seasons. P. viviparum root tips were always EM. In total, 18 mycobiont taxa of the following genera were identified: Cenococcum (1), Cortinarius (2), Helvella (1), Inocybe (3), Russula (1), Sebacina (2), Thelephora (2) and Tomentella (6). All were non-specific EM partners of EM plants. As early as 2 weeks after spring snow melt, EM were well developed,
vital and showed high mycobiont diversity. The relative abundance of senescent root tips was lowest in spring and increased
throughout the year, with a maximum in winter (frozen soil). Thus, mycobiont growth and physiological activity obviously start
when soil is still under snow cover: We speculate that water availability is one important initiation factor for mycorrhizal
development under snow cover, when temperatures still range around the freezing point. Irrespectively of the season, most
abundant mycobionts at this primary successional site belonged to the genera Tomentella, Sebacina and Cenococcum, also in frozen soil. Spatial heterogeneity was high when considering species composition and diversity indices. Overall
mycobionts species richness was restricted at this site, probably because of the limited availability of fungal partners.
We regard the presence/absence of fungal partner and limiting abiotic impacts of the environment as key factors for the symbiotic
status of P. viviparum.

In agricultural landscapes, linear habitats, such as hedgerows at field margins increase structural connectivity among forest
patches, potentially providing dispersal corridors for forest herbs. The spatial structure of linear habitats, however, also
results in edge effects and perturbations that can influence the individual and population performance of forest plants. This
study compares the stage structure and components of growth and reproduction of 14 Trillium grandiflorum populations in hedgerows and forests. Hedgerow Trillium tended to grow faster and, when mature, produced more flowers and more ovules per flowers than forest Trillium, a pattern possibly associated to differences in nutrients and light availability between the two habitats. Seed production
and germination rate, however, did not differ between hedgerows and forests. At the population level, seedlings and juveniles
were proportionally less abundant in hedgerows than in forests. Although well-established plants can thrive in hedgerows,
reduced recruitment may eventually limit the capacity to establish new populations and therefore hamper migration along hedgerow-corridors.
Considering the strategies by which plants persist in linear habitats becomes particularly relevant at a time when species
are expected to be much in need of dispersal corridors because of climatic stress. 相似文献

The seeds of Theobroma cacao (cacao) are the source of cocoa, the raw material for the multi-billion dollar chocolate industry. Cacao’s two most important
traits are its unique seed storage triglyceride (cocoa butter) and the flavor of its fermented beans (chocolate). The genome
of T. cacao is being sequenced, and to expand the utility of the genome sequence to the improvement of cacao, we are evaluating Theobroma grandiflorum, the closest economically important species of Theobroma for its potential use in a comparative genomic study. T. grandiflorum differs from cacao in important agronomic traits such as flavor of the fermented beans, disease resistance to witches’ broom
and abscission of mature fruits. By comparing genomic sequences and analyzing viable inter-specific hybrids, we hope to identify
the key genes that regulate cacao’s most important traits. We have investigated the utility in T. grandiflorum of three types of markers (microsatellite markers, single-strand conformational polymorphism markers and single nucleotide
polymorphism (SNP) markers) developed in cacao. Through sequencing of amplicons of 12 diverse individuals of both cacao and
T. grandiflorum, we have identified new intra- and inter-specific SNPs. Two markers which had no overlap of alleles between the species were
used to genotype putative inter-specific hybrid seedlings. Sequence conservation was significant and species-specific differences
numerous enough to suggest that comparative genomics of T. grandiflorum and T. cacao will be useful in elucidating the genetic differences that lead to a variety of important agronomic trait differences. 相似文献

An effective protocol was developed for in vitro regeneration of the Melothria maderaspatana via indirect organogenesis in liquid and solid culture systems. Organogenesis was achieved from liquid culture calluses derived from leaf and petiole explants of mature plants. Organogenic calluses (98.2?±?0.36 and 94.8?±?0.71%) were induced from both leaf and petiole explants on Murashige and Skoog (MS) liquid medium containing 6.0 µM 2,4-dichlorophenoxyacetic acid (2,4-D) and 0.5 µM thidiazuron (TDZ); and 6.0 µM 2,4-D and 1.0 µM benzyladenine (BA) combinations, respectively. Adventitious shoot regeneration (68.2?±?0.06 shoots per explant) was achieved on MS medium supplemented with 2.0 µM BA, 4.0 µM TDZ, 10% v/v coconut water and 0.06 mM glutamine from leaf-derived calluses. Petiole-derived calluses produced adventitious shoots (45.4?±?0.09 shoots per explant) on MS medium fortified with 2.0 µM BA, 4.0 µM TDZ, 10% v/v coconut water, and 0.08 mM glutamine. Elongation of shoots occurred in MS medium with 2.0 µM gibberellic acid (GA3). Regenerated shoots (2–3 cm in length) rooted (74.2?±?0.38%) and hardened (85?±?1.24%) when they were transferred to 1/2-MS medium supplemented with 3.0 µM indole-3-butyric acid (IBA) followed by garden soil, vermiculate, and sand (2:1:1 ratio) mixture. The elongated shoots (4–5 cm in length) were exposed simultaneously for rooting as well as hardening (100%) in moistened [(1/8-MS basal salt solution with 5 µM IBA and 100 mg l?1 Bavistin® (BVN)] garden soil, vermiculate, and sand (2:1:1 ratio) mixture.
