Soybean seeds [Glycine max (L.) Merr.] synthesize de novo andaccumulate several non-storage, soluble polypeptides duringnatural and precocious seed maturation. These polypeptides havepreviously been coined ‘maturation polypeptides’.The objective of this study was to determine the fate of maturationpolypeptides in naturally and precociously matured soybean seedsduring rehydration, germination, and seedling growth. Developingsoybean seeds harvested 35 d after flowering (mid-development)were precociously matured through controlled dehydration, whereasnaturally matured soybean seeds were harvested directly fromthe plant. Seeds were rehydrated with water for various timesbetween 5 and 120 h. Total soluble proteins and proteins radio-labelledin vivo were extracted from the cotyledons and embryonic axesof precociously and naturally matured and rehydrated seed tissuesand analyzed by one-dimensional PAGE and fluorography. The resultsindicated that three of the maturation polypeptides (21, 31and 128 kDa) that had accumulated in the maturing seeds (maturationpolypeptides) continued to be synthesized during early stagesof seed rehydration and germination (5–30 h after imbibition).However, the progression from seed germination into seedlinggrowth (between 30 and 72 h after imbibition) was marked bythe cessation of synthesis of the maturation polypeptides followedby the hydrolysis of storage polypeptides that had been synthesizedand accumulated during seed development. This implied a drasticredirection in seed metabolism for the precociously maturedseeds as these seeds, if not matured early, would have continuedto synthesize storage protein reserves. Protein synthesis was studied during precocious and natural soybean seed (Glycine max [L.] Merr.) maturation. Developing seeds harvested 35 days after flowering were precociously matured through controlled dehydration. Total soluble proteins and proteins labeled with [35S]methionine were extracted from control, developing seeds and from precociously and naturally matured seeds and were analyzed by one-dimensional PAGE and fluorography. The results demonstrated that several polypeptides which were designated “mature polypeptides,” were synthesized de novo during precocious and natural seed maturation. Two of these polypeptides, 31 and 128 kilodalton in mass, also stained intensely with Coomassie blue, suggesting their abundant accumulation during seed maturation. Results from in vitro translation experiments showed that the mRNAs corresponding to these “maturation polypeptides” accumulated during precocious maturation and in naturally matured seeds, but not in seeds freshly harvested 35 days after flowering (control). The role of the “maturation polypeptides” is currently unknown; however, their presence and that of their corresponding mRNAs was coincident with the ability of matured seeds to establish seedling growth. This study has demonstrated that precocious seed maturation treatments may be extremely useful for investigations of metabolic events and molecular control mechanisms affecting soybean seed maturation.  相似文献

Immature Glycine max (L.) Merrill seeds were grown and matured in liquid medium at 25°C under fluorescent light. In standard medium containing minerals, 146 millimolar sucrose and 62.5 millimolar glutamine (osmolality 0.24), precocious germination seldom occurred with a starting seed size of less than 300 milligrams fresh weight. Frequency of precocious germination increased with increased starting seed size. Sucrose concentration strongly affected precocious germination while glutamine concentration had no effect. Starting with 300 to 350 milligrams fresh weight seeds, treatments which reduced the sucrose concentration or lowered the osmolality of the culture medium stimulated precocious germination, and increased the fresh weight growth but not the dry weight growth of seeds. Increasing the osmolality to 0.38 with sucrose or mannitol prevented precocious germination without reducing dry weight accumulation in seeds. In medium with initially low osmolality, precocious germination was inhibited by addition of 1 to 100 micromolar abscisic acid to the medium without a reduction in seed growth. During growth and maturation of large soybean seeds in vitro, precocious germination and other abnormal tissue growth can be prevented by high sucrose or mannitol concentrations in the medium or by addition of abscisic acid.  相似文献

Immature embryos of soybeans (Glycine max L. Merr.) can be inducedto germinate precociously by depleting the endogenous pool ofabscisic acid (ABA). Washing the embryos or allowing the embryosto dry slowly within or out of detached pods causes a gradualdecline in ABA content. The extent of germination is correlatedwith the length of washing or drying treatments, which in turn,affects the level of endogenous ABA. Providing embryos are threeweeks of age or older, maturation and precocious germinationcan be induced by treating embryos in a way that causes a reductionin embryo ABA content. Merr.] develop, they undergoa change in seed moisture. When excised prematurely from thepod and planted, seeds do not exhibit seedling growth until63 d after flowering (DAF) when the seed moisture has fallenbelow 60%. In contrast, seed germination (radicle protrusion)can occur when seeds as young as 35 DAF (68–79% moisture)are excised, but this germination docs not lead to comparableseedling growth frequencies unless seeds are first given a moistureloss treatment to artificially reduce their moisture below 60%.A moisture loss treatment applied at 35 DAF thus enables seedto undergo the transition from germination (cell expansion)to seedling growth (cell division and expansion) to the extentthat treated immature seed have a vigour index comparable toseeds matured on the plant (100%). The pattern of protein synthesisin vivo was examined in 35 DAF seed using [35S]-methionine incorporation.When moisture loss treatment was applied for 24 h to 35 DAFseeds, seeds synthesized several new polypeptides when comparedwith untreated seeds at the same developmental stage. The sameseed samples showed 0% seedling growth in the absence of moistureloss treatment and 80% seedling growth when the treatment hadbeen applied. Moisture loss from soybean seeds appears to bea prerequisite for the synthesis of new proteins which may bepart of the metabolic process or processes that allow the soybeanseed to undergo the transition from seed germination to seedlinggrowth. During in vitro growth and maturation of soybean seeds, cessationof embryo growth and dry weight accumulation occurred in thepresence of abundant C and N nutrients. Axis followed by cotyledontissues changed from green to yellow, and post-harvest germinationpotential declined if cultured after yellowing of axis tissues.A tissue specific accumulat;on of methanol occurred during thein vitro culture of immature seeds (i.e. initially 50 to 70mg fresh weight) to maturity in liquid medium. Methanol accumulatedto 3.0 g m–3 or 50 µg seed–1 in the medium,while methanol decreased from 37 to about 3.0 µg g–1fresh weight in cotyledons. By contrast, axis tissues increased20-fold in methanol concentration to 90 µg g–1 during20 d in culture. Ethanol was present only in trace amounts inaxis tissues and medium. Addition of exogenous methanol vapourto in situ grown seeds during precocious maturation decreasedsubsequent seedling vigour and germination with increasing levelsof exposure. Methanol accumulation in axis tissues during thegermination phase was not correlated with high temperature andtissue water content treatments which simulated pre-harvestdeterioration of seeds. However, the accumulation of methanolduring in vitro seed development and maturation in liquid culturemay contribute to reduced post-harvest germination performance. Freshly harvested seeds of soybean (Glycine max L. Merr.) orseeds shelled, then dried, were non-viable. Seeds dried in intactpods, even when only 17% of normal size, matured into viableseeds and produced healthy plants. These seeds maintained activityof various enzymes but gained little soluble protein while air-dryingin intact pods. There was a qualitative change in seed proteinsassociated with maturation. Seeds matured in intact pods havea greater proportion of protein as slow-moving bands and havecompletely lost one fast-moving band compared with seeds shelledbefore drying. 采用砂培方式,研究了外源5-氨基乙酰丙酸(ALA)对盐胁迫下菘蓝种子的萌发、幼苗叶片的可溶性糖含量、丙二醛(MDA)含量及其抗氧化酶活性的影响,探讨ALA缓解菘蓝受盐胁迫伤害的响应机制。结果显示:(1)菘蓝种子萌发及幼苗生长在100 mmol·L-1 NaCl胁迫下受到明显的抑制,种子发芽率、发芽势、发芽指数、活力指数与自然含水量均显著降低,丙二醛含量、可溶性糖含量以及超氧化物歧化酶(SOD)、过氧化物酶(POD)活性显著升高。(2)盐胁迫下适宜浓度的ALA处理显著提高了种子萌发率、自然含水量及SOD、POD和CAT活性,降低了可溶性糖和丙二醛的含量,并以16.7 mg·L-1 ALA处理盐胁迫下菘蓝种子的发芽率、发芽势最大,其幼苗的SOD、POD、CAT活性最强。研究表明,盐胁迫显著抑制菘蓝种子的萌发及幼苗生长,适宜浓度的ALA能够有效缓解盐胁迫对菘蓝种子萌发及幼苗生长的伤害,提高植株的抗盐性,并以16.7 mg·L-1 ALA处理效果最佳。  相似文献

Differences in starch metabolism during seed development and germination of two soybean [ Glycine max (L.) Merrill] genotypes with normal seed β-amylase activity ['Williams' ( Sp 1b and 'Altona' ( Sp 1b)] and two soybean genotypes with undetectable seed β-amylase activity ['Chestnut' ( Sp 1au) and 'Altona' ( Sp 1)] were investigated. Starch and soluble sugar profiles were essentially the same during seed development and germination. Total amylase activity of Williams and Altona ( Sp 1b) peaked just prior to seed maturity and then dropped off slowly; whereas, the total amylase activity of Chestnut and Altona ( sp 1) was very low throughout seed development and germination. The differences in amylase activity between Altona ( Sp 1 b) and Altona ( sp 1) was also seen in leaves. α-Amylase activity was similar in the four genotypes when β-amylase was inhibited with Hg2+ but was higher in the two genotypes with normal β-amylase activity when β-amylase was inhibited with heat plus Ca2+. Low levels of starch phosphorylase activity were detected throughout seed development and germination, and the activity was similar in three of the genotypes and higher in Altona ( sp 1).
The protein, oil and oligosaccharide contents of mature seeds of the four genotypes were similar. Altona ( sp 1 b) and ( sp 1), which appear to be near isogenic lines, were not different in any morphological character or yield.
Altona ( Sp 1 b) showed greater hydrolysis of soybean seed starch than Altona ( sp 1), but the evidence indicates that the mutation resulting in greatly reduced or missing β-amylase activity has no effect on starch metabolism of developing and germinating soybean seeds.  相似文献

模拟华山新麦草野生土壤生境特点,分别采用聚乙二醇(PEG-6000)、钙(CaCl2)以及二者混合液3种方式浸种,研究PEG-6000和Ca2+浸种对华山新麦草种子萌发及幼苗生长的影响.结果表明:3种处理下华山新麦草的种子发芽率、发芽势和发芽指数均无显著变化;但PEG-6000浸种后,华山新麦草幼苗可溶性糖和可溶性蛋白含量增加,蛋白酶、SOD和POD活性增强;CaCl2浸种后,华山新麦草幼苗可溶性糖含量增加,蛋白酶、SOD和CAT活性增强,淀粉酶活性降低;而PEG-6000和CaCl2混合液浸种后,华山新麦草幼苗可溶性糖含量下降,SOD活性降低,华山新麦草生长受阻.据此推测华山干旱和钙离子含量高的生境特点可能是华山新麦草种群濒危的原因之一.  相似文献

以哥伦比亚生态型(Columbia-0)拟南芥(Arabidopsis thaliana)为实验材料, 人工模拟UV-B辐照处理拟南芥种子, 统计其发芽势和发芽率, 测定根长、株高、叶绿素、可溶性糖、可溶性蛋白以及丙二醛(MDA)含量, 研究UV-B辐照处理对拟南芥种子萌发和幼苗生长的影响。研究结果表明, 低剂量的UV-B辐照可以促进拟南芥种子萌发和幼苗生长, 并且最佳辐照剂量为1.0 kJ·m–2(P<0.05), 此时的发芽势、发芽率、根长、株高、叶绿素、可溶性糖及可溶性蛋白的含量均达到最大, 而丙二醛的含量变化则不明显(P>0.05)。当辐照剂量大于1.0 kJ·m–2时, 促进作用逐渐变小, 并且随着辐照剂量的增加, 表现出了抑制作用。实验结果表明, 适当剂量的UV-B辐照在一定程度上可以促进拟南芥种子萌发和幼苗生长, 而过高剂量的辐照则对其产生抑制或损伤作用。  相似文献

以不同浓度的壳聚糖对油菜种子进行包衣处理,考察其对油菜种子萌发及幼苗耐盐性的影响,并在不同盐浓度胁迫条件下对种子萌发时的发芽势、发芽率、生物量(鲜重、干重、根长、芽长)等指标进行测定,同时分析油菜幼苗叶绿素含量、可溶性蛋白及可溶性糖含量的变化。结果表明,一定浓度的壳聚糖包衣处理可提高油菜种子发芽率、发芽势、生物量、幼苗的耐盐指数、叶绿素含量、可溶性蛋白及可溶性糖的含量,其中浓度为0.25 g·L-1壳聚糖包衣处理对油菜种子萌发的促进效果较好,而浓度为0.50 g·L-1壳聚糖包衣处理对提高油菜幼苗耐盐性具有较好的促进作用。  相似文献

Immature Glycine max (L.) Merr. seeds initially at 50–70mg fresh weight were successfully grown and matured in vitroin detached pods. Surface sterilized pods were floated in aliquid medium containing 5 per cent sucrose, minerals, and glutaminein 125 ml Erlenmeyer flasks and incubated at 25 °C under350–400 µE m–1 s–1 white light. Seedswhich were matured in vitro increased tenfold in dry weight,were visually similar to commercial seeds of the same size,were tolerant to desiccation and germinated with normal seedlinggrowth. 人工老化处理对结球甘蓝种子生理生化特性的影响   总被引:3,自引:0,他引:3  
以结球甘蓝品种‘冬升’种子为材料,研究高温(40℃)高湿(相对湿度100%)人工老化处理过程中种子的萌发特性、种子浸出液相对电导率和丙二醛、可溶性蛋白、可溶性糖含量以及抗氧化酶活性的变化,以揭示种子劣变的机理。结果表明:(1)人工老化处理甘蓝种子的含水量和不正常苗率均随着老化时间的延长逐渐增加,而种子发芽势、发芽率、发芽指数和活力指数的增加均逐渐降低。(2)随着处理天数的延长,老化处理甘蓝种子的浸出液电导率显著增大,浸出液可溶性糖含量逐渐升高,可溶性蛋白含量表现出显著下降趋势,而种子MDA含量呈先升高后逐渐下降的趋势。(3)在结球甘蓝种子老化进程中,其种子中SOD、POD、CAT活性变化的趋势相似,均随老化程度的加深而逐渐降低;而APX活性在老化处理的最初2d显著增加,第6天显著降低。研究发现,在结球甘蓝种子老化进程中,种子活力和萌发率显著降低,其种子浸出液电导率、可溶性蛋白含量、可溶性糖含量、保护酶活性变化与种子老化及劣变程度密切相关,膜脂过氧化作用可能是引起或加剧种子老化劣变的重要原因之一。  相似文献

The sucrose non-fermenting-related kinase complex (SnRK1) is a heterotrimeric complex that plays a central role in metabolic adaptation to nutritional or environmental stresses. Here we investigate the role of a regulatory gamma-subunit of the complex, MtSNF4b, in Medicago truncatula seeds. Western blot indicated that MtSNF4b accumulated during seed filling, whereas it disappeared during imbibition of mature seeds. Gel filtration chromatography suggested that MtSNF4b assembled into a complex (450-600 kDa) at the onset of maturation drying, and dissociated during subsequent imbibition. Drying of desiccation-tolerant radicles led to a reassembly of the complex, in contrast to sensitive tissues. Silencing of MtSNF4b using a RNA interference (RNAi) approach resulted in a phenotype with reduced seed longevity, evident from the reduction in both germination percentage and seedling vigour in aged RNAi MtSNF4b seeds compared with the wild-type seeds. In parallel to the assembly of the complex, seeds of the RNAi MtSNF4b lines showed impaired accumulation of raffinose family oligosaccharides compared with control seeds. In mature seeds, the amount of stachyose was reduced by 50-80%, whereas the sucrose content was 60% higher. During imbibition, the differences in non-reducing sugar compared with the control disappeared in parallel to the disassembly of the complex. No difference was observed in dry weight or reserve accumulation such as proteins, lipids and starch. These data suggest that the regulatory gamma-subunit MtSNF4b confers a specific and temporal function to SnRK1 complexes in seeds, improving seed longevity and affecting the non-reducing sugar content at later stages of seed maturation.  相似文献

In this study the effects of seed size variation on germinationand seedling vigour have been investigated within and betweenploidy levels of diploid and related autotetraploid Dactylixglomerata. Rates of seed germination and seedling growth werecompared in two contrasting environments using diploid and tetraploidseeds of equal and also different biomass. Within each ploidylevel, seed biomass had no effect on either the overall percentagenor the rate of germination. In contrast, the comparison ofseeds of equal biomass but differing in ploidy level showedthat seeds from tetraploid plants germinated faster and to ahigher percentage than those from diploid plants. With respectto seedling growth, heavier seeds from the tetraploid genotypesgave seedlings of significantly higher biomass than those fromlighter tetraploid and diploid seeds throughout the 2 monthsof study. Interestingly, seeds of equal biomass but from differentploidy levels produced seedlings more similar than those fromthe extreme seed weight categories. These differences were maintainedin two different environmental conditions. The influence of abscisic acid (ABA) on the precocious germinationand storage protein production of pea seeds has been examinedusing embryo and pod culture. The precocious germination ofembryos in culture could not be inhibited fully by ABA on apermissive medium (2% sucrose) even at 0.1 mol m–3. However,increasing the sucrose concentration to 5% caused near completeinhibition when ABA was added to the medium. Embryos of differentweights cultured on a high osmoticum (mannitol-containing medium),equivalent to 10% sucrose, did not show any consistent differencein ABA content. When fluridone was added to a non-permissiveculture medium, no decrease in ABA content of the embryos couldbe observed and no precocious germination was induced. In contrast,fluridone was able to prevent the accumulation of ABA in seedspresent in pods cultured in its presence from an early stageof development. These seeds, however, grew normally and reachedmaturity, did not germinate precociously in vivo, were desiccationtolerant and still produced storage protein message whetheror not ABA was included in the culture medium. Immature Glycine max (L.) Merrill seeds, initially between 50and 450 mg f. wt, were grown and matured successfully in vitro.Excised seeds were floated in a liquid medium containing 5 percent sucrose, minerals and glutamine in flasks incubated at25 °C under 300 to 350 µE m–2 s–1 fluorescentlight. During 16 to 21 d in culture, seeds grew to a matured. wt of 100 to 600 mg per seed at an average rate of 5 to 25mg d. wt per seed d–1 depending on initial size. Growthrates were maximal during the first 8 to 10 d in vitro but declinedwith loss of green colour in the cotyledons. Seed coats rupturedwith rapid cotyledon expansion during the first 2 d in culture.Embryos were tolerant to desiccation and 80 to 90 per cent germinatedif removed from culture before complete loss of green colour.The growth of excised seeds in vitro exceeded the growth ofseeds in detached pods, but when windows were cut in pods topermit direct exposure of seeds to the medium, seed growth wascomparable. Compared to seeds, somatic embryos accumulated relatively low levels and different types of storage carbohydrates. The regulation of starch accumulation was studied to determine its effects on desiccation tolerance and vigor of dry somatic embryos. Somatic embryos of Medicago sativa are routinely matured through three phases: 7 days of development; 10 days of phase I maturation, a rapid growth phase; and 10 days of phase II maturation, a phase leading to the acquisition of desiccation tolerance. The control of starch deposition was investigated in alfalfa somatic embryos by manipulating the composition of the phase I maturation medium with different levels of sucrose, abscisic acid, glutamine and different types of carbohydrates and amino acids. After phase II maturation, mature somatic embryos were collected for desiccation and subsequent conversion, or for biochemical analyses. Starch deposition occurred primarily during phase I maturation, and variations in the composition of this medium influenced embryo quality, storage protein and starch accumulation. A factorial experiment with two levels of glutamine × three levels of sucrose showed that increasing the sucrose concentration from 30 to 80 g/l increased embryo size and starch content, but had minimal effect on accumulation of storage proteins; glutamine also increased embryo size, but decreased starch content and increased accumulation of the high salt soluble S-2 (medicagin) storage proteins. ABA did not influence any of the parameters tested when included in phase I maturation at concentration up to 10 μM. Replicating sucrose with maltose, glucose, or glucose and fructose did not alter embryo size or starch accumulation (mg/g fresh weight), but replacement with fructose alone reduced embryo size, and replacement with glucose alone reduced germination. Suplementation with the amino acids, asparagine, aspartic acid and glutamine increased seedling vigor, but decreased the starch content of embryos. The data indicate that starch accumulation in somatic embryos is regulated by the relative availability of carbon versus nitrogen nutrients in the maturation medium. The quality of mature somatic embryos, determined by the rate of seedling development (conversion and vigor), correlated with embryo size, storage protein and free amino acid but not with starch. Therefore, further improvements in the quality of somatic embryo may be achieved through manipulation of the maturation medium in order to increase storage protein, but not starch deposition.  相似文献
