Summary It is shown that the ouabain-resistant (OR) furosemide-sensitive K+(Rb+) transport system performs a net efflux of K+ in growing mouse 3T3 cells. This conclusion is based on the finding that under the same assay conditions the furosemidesensitive K+(Rb+) efflux was found to be two- to threefold higher than the ouabain-resistant furosemide-sensitive K+(Rb+) influx. The oubain-resistant furosemide-sensitive influxes of both22Na and86Rb appear to be Cl dependent, and the data are consistent with coupled unidirectional furosemide-sensitive influxes of Na+, K+ and Cl with a ratio of 1 1 2. However, the net efflux of K+ performed by this transport system cannot be coupled to a ouabain-resistant net efflux of Na+ since the unidirectional ouabain-resistant efflux of Na+ was found to be negligible under physiological conditions. This latter conclusion was based on the fact that practically all the Na+ efflux appears to be ouabainsensitive and sufficient to balance the Na+ influx under such steady-state conditions. Therefore, it is suggested that the ouabain-resistant furosemide-sensitive transport system in growing cells performs a facilitated diffusion of K+ and Na+, driven by their respective concentration gradients: a net K+ efflux and a net Na+ influx.  相似文献

Summary Ouabain-insensitive, furosemide-sensitive Rb+ influx (J Rb) into HeLa cells was examined as functions of the extracellular Rb+, Na+ and Cl concentrations. Rate equations and kinetic parameters, including the apparent maximumJ Rb, the apparent values ofK m for the three ions and the apparentK i for K+, were derived. Results suggested that one unit molecule of this transport system has one Na+, one K+ and two Cl sites with different affinities, one of the Cl sites related with binding of Na+, and the other with binding of K+(Rb+). A 11 stoichiometry was demonstrated between ouabain-insensitive, furosemidesensitive influxes of22Na+ and Rb+, and a 12 stoichiometry between those of Rb+ and36Cl. The influx of either one of these ions was inhibited in the absence of any one of the other two ions. Monovalent anions such as nitrate, acetate, thiocyanate and lactate as substitutes for Cl inhibited ouabain-insensitive Rb+ influx, whereas sulfamate and probably also gluconate did not inhibitJ Rb. From the present results, a general model and a specialized cotransport model were proposed: 1) In HeLa cells, one Na+ and one Cl bind concurrently to their sites and then one K+ (Rb+) and another Cl bind concurrently. 2) After completion of ion bindings Na+, K+(Rb) and Cl in a ratio of 112 show synchronous transmembrane movements.  相似文献

Summary We have found that Simon's neutral, noncyclic, Li+-selective complexone, which has imide and ether ligands, renders lipid bilayer membranes selectively permeable to certain cations and anions. The present paper characterizes the ability of this molecule to carry monovalent cations; and we show it to be most selective for Li+ among the alkali cations, the first reconstitution of Li+-selective permeation in lipid bilayer membranes. This complexone acts as an equilibrium-domain carrier for Ag+> Li+>Tl+>Na+>NH 4 + >Rb+>Cs+ over a wide range of experimental conditions. The major type of membrane-permeating species formed is a 21 carrier/cation complex dominant except at the lowest salt and carrier concentrations where a 11 carrier/cation, with a similar selectivity sequence, can be detected. Among the groupIa cations the selectivity sequence in bilayers, Li+>Na+>K+>Rb+>Cs+, is similar to that previously found for this molecule in thick solvent-polymer membrane electrodes. We find this carrier to be more selective to Ag+ than to any other monovalent cation yet studied. This high Ag+ selectivity is used, together with the dependence of the selectivity on the nature of the N-amide substitutents, to argue that the imide oxygens play a major role as ligands.  相似文献

1. The incorporation of l-[1-14C]leucine into the proteins of liver slices and into the serum albumin and lipoproteins transported by these slices was investigated. 2. Transport rates were found to be dependent on the K+ content of the slices. 3. The effect of K+ on transport of serum albumin and of serum lipoprotein can be separated from any effect on synthesis by altering K+ concentrations after inhibition of protein synthesis by cycloheximide or puromycin. 4. The effect of low K+ concentrations is reversible. 5. There is linear relationship between the K+ content of the slices and the transport of protein. A simple method is described for maintaining various steady concentrations of K+ in the liver slices. 6. K+ may be replaced by Rb+. Cs+ is partly effective, but NH4+ and Li+ are no more effective than Na+. 7. We found evidence that K+ content rather than the flux rates of K+ or Na+ is important in this effect. 8. These results are probably important in ethionine and carbon tetrachloride poisoning in the rat, and may be significant in liver transplantation.  相似文献

Summary The bumetanide-sensitive uptake of Na+, K(Rb) and Cl has been measured at 21°C in ferrent red cells treated with (SITS+DIDS) to minimize anion flux via capnophorin (Band 3). During the time course of the influx experiments tracer uptake was a first-order rate process. At normal levels of external Na+ (150mm) the bumetanide-sensitive uptake of K+ was dependent on Cl and represented almost all of the K+ uptake, the residual flux demonstrating linear concentration dependence. The uptake of Na+ and Cl was only partially inhibited by bumetanide indicating that pathways other than (Na+K+Cl) cotransport participate in these fluxes. The diuretic-sensitive uptake of Na+ or Cl was, however, abolished by the removal of K+ or the complementary ion indicating that bumetanide-sensitive fluxes of Na+, K+ and Cl are closely coupled. At very low levels of [Na] o (<5mm) K+ influx demonstrated complex kinetics, and there was evidence of the unmasking of a bumetanide-sensitive Na+-independent K+ transport pathway. The stoichiometry of bumetanide-sensitive tracer uptake was 2Na1K3Cl both in cells suspended in a low and a high K+-containing medium. The bumetanide-sensitive flux was markedly reduced by ATP depletion. We conclude that a bumetanide-sensitive cotransport of (2Na1K3Cl) occurs as an electroneutral complex across the ferret red cell membrane.  相似文献

Summary Southern Corn Leaf Blight is caused by a toxin produced by a virulent form ofHelminthosporium maydis (Race T). The toxin has been shown to uncouple oxidative phosphorylation and dissipate Ca2+ gradients in mitochondria isolated from susceptible, but not resistant, corn. The possibility that the toxin acted by increasing the permeability of membranes to ions was tested using a planar bilayer membrane system. Addition of the toxin to the bilayer system, under voltage-clamp conditions, resulted in stepwise increases in current across the phospholipid bilayer, a response characteristic for channel formers. Single-channel conductance in 1m KCl is 27 pS which corresponds to 1.7×107 ions sec–1 channel–1 at 100 mV applied potential. The toxin channels are: (i) fairly uniform in conductance, (ii) ideally selective for K+ over Cl, and (iii) most conductive to H+. The channel showed the following selectivity for alkali metal cations: Rb+>K+>Cs+>Na+>Li+ (169731) based on the most frequently observed conductance in 1m chloride salts. The toxin showed no voltage dependence over the range of –100 to +100 mV. Channel formation was also a property of a synthetic analog (Cmpd IV) of the toxin. The ability of the native toxin to form channels may be a mode of toxin action on mitochondrial membranes from susceptible corn.  相似文献

Summary To study the physiological role of the bidirectionally operating, furosemide-sensitive Na+/K+ transport system of human erythrocytes, the effect of furosemide on red cell cation and hemoglobin content was determined in cells incubated for 24 hr with ouabain in 145mm NaCl media containing 0 to 10mm K+ or Rb+. In pure Na+ media, furosemide accelerated cell Na+ gain and retarded cellular K+ loss. External K+ (5mm) had an effect similar to furosemide and markedly reduced the action of the drug on cellular cation content. External Rb+ accelerated the Na+ gain like K+, but did not affect the K+ retention induced by furosemide. The data are interpreted to indicate that the furosemide-sensitive Na+/K+ transport system of human erythrocytes mediates an equimolar extrusion of Na+ and K+ in Na+ media (Na+/K+ cotransport), a 1:1 K+/K+ (K+/Rb+) and Na+/Na+ exchange progressively appearing upon increasing external K+ (Rb+) concentrations to 5mm. The effect of furosemide (or external K+/Rb+) on cation contents was associated with a prevention of the cell shrinkage seen in pure Na+ media, or with a cell swelling, indicating that the furosemide-sensitive Na+/K+ transport system is involved in the control of cell volume of human erythrocytes. The action of furosemide on cellular volume and cation content tended to disappear at 5mm external K+ or Rb+. Thein vivo red cell K+ content was negatively correlated to the rate of furosemide-sensitive K+ (Rb+) uptake, and a positive correlation was seen between mean cellular hemoglobin content and furosemide-sensitive transport activity. The transport system possibly functions as a K+ and waterextruding mechanism under physiological conditiosin vivo. The red cell Na+ content showed no correlation to the activity of the furosemide-sensitive transport system.  相似文献

Outside-out configuration of the patch clamp technique was used to test whether an intracellular application of G protein activator (GTPS) affects ATP-activated Ca2+-permeable channels in rat macrophages without any agonist in the bath solution. With 145 mm K+ (pCa 8.0) in the pipette solution, activity of channels permeable to a variety of divalent cations and Na+ was observed and general channel characteristics were found to be identical to those of ATP-activated ones. Absence of extracellular ATP makes it possible to avoid the influence of ATP receptor desensitization and to study the channel selectivity using a number of divalent cations (105 mm) and Na+ (145 mm) as the charge carriers. Permeability sequence estimated by extrapolated reversal potential measurements was: Ca2+ Ba2+ Mn2+ Sr2+ Na+ K+ = 68 30 26 10 3.5 1. Slope conductances (in pS) for permeant ions rank as follows: Ca2+ Sr2+ Na+ Mn2+ Ba2+ = 19 18 14 12 10. Unitary Ca2+ currents display a tendency to saturate with the Ca2+ concentration increase with apparent dissociation constant (K d ) of 10 mm. No block of Na+ permeation by extracellular Ca2+ in millimolar range was found. The data obtained suggest that (i) activation of some G protein is sufficient to gate the channels without the ATP receptor being occupied, (ii) the ATP receptor activation results in the gating of a special channel with the properties that differ markedly from those of the receptoroperated or voltage-gated Ca2+-permeable channels on the other cell types.DeceasedThe authors are grateful to K. Summary Recently we proposed that cytoplasmic acidification of low K+ (LK) sheep erythrocytes may stimulate ouabain-resistant Cl-dependent K+ flux (K+Cl cotransport), also known to be activated by cell swelling, treatment with N-ethylmaleimide (NEM), or removal of cellular bivalent cations. Here we studied the dependence of K+ transport on intracellular and extracellular pH (pH i , pH o ) varied either simultaneously or independently using the Cl/HCO 3 exchange inhibitor 4,4, diisothiocyanatostilbene-3,2-disulfonic acid (DIDS). In both control and NEM-treated LK cells volumes were kept near normal by varying extracellular sucrose. Using DIDS as an effective pH clamp, both K+ efflux and influx of Rb+ used as K+ congener were strongly activated at acid pH i and alkaline pH o . A small stimulation of K+ (Rb+) flux was also seen at acid pH i in the absence of DIDS, i.e., when pH i pH o . Anti-L l serum, known to inhibit K+Cl cotransport, prevented the pH i -stimulated K+ (Rb+) fluxes. Subsequent to NEM treatment at pH 6, K+ (Rb+) fluxes were activated only by raising pH, and thus were similar to the pH activation profile of K+ (Rb+) fluxes in DIDS-treated cells with pH o varied at constant physiologic pH i . Anti-L l , which inhibited NEM-stimulated K+ (Rb+) fluxes, failed to do so in NEM-plus DIDS-treated cells. Summary Toxicity screening ofEscherichia coli NCIB 9484 andBacillus subtilis 007, NCIB 168 and NCIB 1650 has shown Cs+ to be the most toxic Group 1 metal cation. However, toxicity and accumulation of Cs+ by the bacteria was affected by two main external factors; pH and the presence of other monovalent cations, particularly K+. Over the pH range 6–9 bothE. coli andB. subtilis showed increasing sensitivity towards caesium as the pH was raised. The presence of K+ and Na+ in the laboratory media used lowered caesium toxicity and lowered acumulation of the metal. In order to assess accurately Cs+ toxicity towards the bacterial strains it was therefore necessary to define the K+:Cs+ ratio in the external medium. The minimum inhibitory K+:Cs+ concentration ratio for theBacillus strains tested was in the range 12–13 whileE. coli had a minimum inhibitory K+:Cs+ concentration ratio of 16.  相似文献

Summary Previous studies indicate a particular sensitivity of red blood cell Na+-Li+ countertransport activity to small variations in the fatty acid composition of membrane phospholipids. To assess whether the interindividual variability of Na+-Li+ countertransport is related to differences in the species pattern of erythrocyte phosphatidylcholine (PC) and phosphatidylethanolamine (PE) in vivo, the molecular species composition of PC and PE as well as the kinetics of Na+-Li+ countertransport were analyzed in parallel in normo- and hyperlipidemic donors. Both in diacyl PC and in diacyl-PE the species 160/204 and 160/182 were, respectively, positively and negatively related to the apparent maximal velocity of Na+-Li+ countertransport. The sum of all species with 204 at sn2 of diacyl-PE exhibited a strong positive (r = 0.82, 2p < 0.001), and those containing 182 a negative correlation (r = –0.63, 2p < 0.01) to the transport activity. Essentially similar connections were observed between these species and the apparent affinity of the transport system for intracellular Na+. To evaluate whether the associations between molecular species of membrane phospholipids and Na+-Li+ countertransport activity were indicative of a causal relationship, the species 160/204-PC and 160/182-PC were selectively introduced into the erythrocyte membrane by means of the PC-specific transfer protein. Replacement of 11% of native PC by 160/182-PC inhibited the transport rate by about 25%. Exchange of 6 and 9% of PC with 160/204-PC, in contrast, accelerated the transport rate by 30 and 60%, respectively. The accordance between the in vivo relations and the results of the in vitro modification strongly suggests that elevations and reductions in the arachidonic acid and linoleic acid content of membrane PC and PE contribute to the interindividual variability of red blood cell Na+-Li+ counter-transport activity and its acceleration in hyperlipidemias.The authors wish to thank Dr. W.O. Comparison of the rate of accumulation of 86Rb+ by intact cells of Rhodobacter capsulatus during short periods of illumination with the Rb+-dependent membrane ionic current measured by electrochromism supports the view that both activities reflect the operation of a low-affinity K+ transport system. In experiments performed under similar conditions the ratio of 86Rb+ uptake to charge uptake was approx. 1.0, suggesting that the transport system operates as a uniporter. The addition of NH inf4 sup+ to a cell suspension led to an increase in membrane ionic current but failed to inhibit the accumulation of 86Rb+ during illumination. The presence of K+ and NH inf4 sup+ inhibited the increase in cellular ATP levels at the onset of illumination. This effect was prevented by Cs+. The results are considered within the context of the hypothesis (Golby et al. Eur J Biochem 187: 589–597) that NH inf4 sup+ can be transported by the K+ carrier and in the context of an alternative hypothesis that NH inf4 sup+ increases the affinity of the K+ transport system for its natural substrate and for Rb+.Abbreviations pH pH difference across the cytoplasmic membrane - electrical potential difference across the cytoplasmic membrane  相似文献

Summary The effect of extracellular and intracellular Na+ (Na o + , Na i + ) on ouabain-resistant, furosemide-sensitive (FS) Rb+ transport was studied in human erythrocytes under varying experimental conditions. The results obtained are consistent with the view that a (1 Na++1 K++2 Cl) cotransport system operates in two different modes: modei) promoting bidirectional 11 (Na+–K+) cotransport, and modeii) a Na o + -independent 11 K o + /K i + exchange requiring Na i + which, however, is not extruded. The activities of the two modes of operation vary strictly in parallel to each other among erythrocytes of different donors and in cell fractions of individual donors separated according to density. Rb+ uptake through Rb o + /K i + exchange contributes about 25% to total Rb+ uptake in 145mm NaCl media containing 5mm RbCl at normal Na i + (pH 7.4). Na+–K+ cotransport into the cells occurs largely additive to K+/K+ exchange. Inward Na+–Rb+ cotransport exhibits a substrate inhibition at high Rb o + . With increasing pH, the maximum rate of cotransport is accelerated at the expense of K+/K+ exchange (apparent pK close to pH 7.4). The apparentK m Rb o + of Na+–K+ cotransport is low (2mm) and almost independent of pH, and high for K+/K+ exchange (10 to 15mm), the affinity increasing with pH. The two modes are discussed in terms of a partial reaction scheme of (1 Na++1 K++2 Cl) cotransport with ordered binding and debinding, exhibiting a glide symmetry (first on outside = first off inside) as proposed by McManus for duck erythrocytes (McManus, T.J., 1987,Fed. Proc., in press). N-ethylmaleimide (NEM) chemically induces a Cl-dependent K+ transport pathway that is independent of both Na o + and Na i + . This pathway differs in many properties from the basal, Na o + -independent K+/K+ exchange active in untreated human erythrocytes at normal cell volume.     Summary K+, Cl, and Ca2+ channels in the vacuolar membrane of tobacco cell suspension cultures have been investigated using the patch-clamp technique. In symmetrical 100mM K+, K+ channels opened at positive vacuolar membrane potentials (cytoplasmic side as reference) had different conductances of 57 pS and 24 pS. K+ channel opened at negative vacuolar membrane potentials had a conductance of 43 pS. The K+ channels showed a significant discrimination against Na+ and Cl. The Cl channel opened at positive vacuolar membrane potentials for cytoplasmic Cl influx had a high conductance of 110pS in symmetrical 100mM Cl. When K+ and Cl channels were excluded from opening, no traces were found of Ca2+ channel activity for vacuolar Ca2+ release induced by inositol 1,4,5-trisphosphate or other events. However, we found a 19pS Ca2+ channel which allowed influx of cytoplasmic Ca2+ into the vacuole when the Ca2+ concentration on the cytoplasmic side was high. When Ca2+ was substituted by Ba2+, the conductance of the 19 pS channel became 30 pS and the channel showed a selectivity sequence of Ba2+Sr2+Ca2+Mg2+=10.60.60.21. The reversal potentials of the channel shifted with the change in Ca2+ concentration on the vacuolar side. The channel could be efficiently blocked from the cytoplasmic side by Cd2+, but was insensitive to La3+, Gd3+, Ni2+, verapamil, and nifedipine. The related ion channels in freshly isolated vacuoles from red beet root cells were also recorded. The coexistence of the K+, Cl, and Ca2+ channels in the vacuolar membrane of tobacco cells might imply a precise classification and cooperation of the channels in the physiological process of plant cells.  相似文献

    Summary The Ehrlich tumor cell possesses and anion-cation cotransport system which operates as a bidirectional exchanger during the physiological steady state. This cotransport system, like that associated with the volume regulatory mechanism (i.e. coupled net uptake of Cl+Na+ and/or K+) is Cl-selective and furosemide-sensitive, suggesting the same mechanism operating in two different modes. Since Na+ has an important function in the volume regulatory response, its role in steady-state cotransport was investigated. In the absence of Na+, ouabain-insensitive K+ and DIDS-insensitive Cl transport (KCl cotransport) are low and equivalent to that found in 150mm Na+ medium containing furosemide. Increasing the [Na+] results in parallel increases in K+ and Cl transport. The maximum rate of each (18 to 20 meq/(kg dry wt)·min) is reached at about 20mm Na+ and is maintained up to 55mm. Thus, over the range 1 to 55mm Na+ the stoichiometry of KCl cotransport is 11. In contrast to K+ and Cl, furosemide-sensitive Na+ transport is undetectable until the [Na+] exceeds 50mm. From 50 to 150mm Na+, it progressively rises to 7 meq/(kg dry wt)·min, while K+ and Cl transport decrease to 9 and 16 meq/(kg dry wt)·min, respectively. Thus, at 150mm Na+ the stoichiometric relationship between Cl, Na+ and K+ is 211. These results are consistent with the proposal that the Cl-dependent cation cotransport system when operating during the steady state mediates the exchange of KCl for KCl or NaCl for NaCl; the relative proportion of each determined by the extracellular [Na+].  相似文献

    Summary The specific activity of the Na+/K+/Cl cotransporter was assayed by measuring the initial rates of furosemide-inhibitable86Rb+ influx and efflux. The presence of all three ions in the external medium was essential for cotransport activity. In cultured smooth muscle cells furosemide and bumetanide inhibited influx by 50% at 5 and 0.2 m, respectively. The dependence of furosemide-inhibitable86Rb+ influx on external Na+ and K+ was hyperbolic with apparentK m values of 46 and 4mm, respectively. The dependence on Cl was sigmoidal. Assuming a stoichiometry of 112 for Na+/K+/Cl, aK m of 78mm was obtained for Cl. In quiescent smooth muscle cells cotransport activity was approximately equal to Na+ pump activity with each pathway accounting for 30% of total86Rb+ influx. Growing muscle cells had approximately 3 times higher cotransport activity than quiescent ones. Na+ pump activity was not significantly different in the gorwing and quiescent cultures. Angiotensin II (ANG) stimulated cotransport activity as did two calcium-transporting ionophores, A23187 and ionomycin. The removal of external Ca2+ prevented A23187, but not ANG, from stimulating the cotransporter. Calmodulin antagonists selectively inhibited86Rb+ influx via the cotransporter. Beta-adrenoreceptor stimulation with isoproterenol, like other treatments which increase cAMP, inhibited cotransport activity. Cultured porcine endothelial cells had 3 times higher cotransport activity than growing muscle cells. Calmodulin antagonists inhibited cotransport activity, but agents which increase cAMP or calcium had no effect on cotransport activity in the endothelial cells.  相似文献

    Summary In this short survey differences between species and varieties in the four major mechanisms that affect selective uptake of potassium and sodium to the plant within the root are considered. These include influx selectivity, K+/Na+ exchange at the plasmalemma, and selectivity at the tonoplast as well as at the symplasm-xylem boundary. The affinity of various plants for potassium influx in system 1 is rather uniform although varietal differences in barley have been observed. Differences are much more pronounced for sodium influx, for which Helianthus showed rather high and Fagopyrum rather low affinity. There is substantial variation between species in the efficiency of K+/Na+ exchange at the plasmalemma of cortical root cells; the three cereals Hordeum, Triticum, and Secale were highly efficient while K+/Na+ exchange in Atriplex, Helianthus and Allium was poor, even if the cytoplasmic sodium content was accounted for. Apparently there was no direct relation between salt tolerance and K+/Na+ exchange. The observed differences in the efficiency of K+-dependent sodium extrusion or K+/Na+ exchange were not due to the use of excised roots, they were observed also when roots of whole seedlings were investigated. At the tonoplast a 11 exchange of vacuolar potassium for sodium has been observed in roots of Hordeum. By this exchange sodium ions are removed from the symplasm and potassium ions are recovered from vacuoles and thus made available for transport to the shoot. Indications for specific differences in this exchange have been observed; the exchange appears to be more efficient in Helianthus than in Hordeum roots. More comparative studies are needed here. At the boundary between symplasm and xylem vessels selectivity can be set up during xylem release of cations and there are reports that suggest a preference for sodium (Lycopersicum cheesemanii, Solanum pennellii, and Suaeda) and for varietal differences amongst tomatoes. Selectivity at this boundary, the plasmalemma of the xylem parenchyma cells was described in this paper by the selectivity ratio of transport that relates the rates of xylem transport to the cytoplasmic sodium and potassium concentrations. Based on this ratioAtriplex hortensis was shown to discriminate for sodium during xylem release while there was little selectivity in Hordeum and possibly some discrimination in favour of K+ in Allium roots.     Influx of the K+ analogue Rb+ was measured through the ouabain-sensitive Na+/K+ pump and the ouabain-insensitive “leak” pathways in Cl? or NO in mature red cells from adult pigs and in reticulocytes naturally occurring in 7-day-old piglets. In reticulocytes, Rb+ influxes by the two pathways were of about equal magnitude in Cl? (13 and 10 mmoles/liter cells × hr) and at least 25-fold larger than in mature red cells (0.5 and 0.4 mmoles/liter cells × hr). In Na + media, a portion of the ouabain-insensitive “leak” flux of Rb+ was Cl? dependent (Rb+Cl? transport) as NO replacement reduced Rb+ influx by 90% in reticulocytes and by 40% in mature red cells. The sulfhydryl reagent N-ethylmaleimide (NEM) stimulated Rb+Cl? transport about twofold in reticulocytes and up to 13-fold in mature red cells. When reticulocytes matured to erythrocytes during in vitro incubation, about 90% of both ouabain-sensitive Rb+ pump and ouabain-insensitive Rb+Cl? influx were lost. In contrast, the NEM-stimulated Rb+Cl? transport changed much less throughout this period, suggesting an entity operationally but not necessarily structrually distinct from the basal Rb+Cl? transport. Although the experimental variability precluded a full assessment of significant changes in the small Na+/K+(Rb+) pump and Rb+Cl? fluxes in mature pig red cells kept for the same time period in vitro, Rb+ flux changes in reticulocytes appear to be maturational in nature, reflecting parallel activity transitions of Na+/K+ pump and Cl?-dependent K+ fluxes in vivo.  相似文献

    Macroscopic instantaneous and time-dependent currents have been measured in the vacuolar membrane of Beta vulgaris using a patch clamp configuration analogous to whole cell mode. At low cytosolic Ca2+ and in the absence of Mg2+, only an instantaneous current was observed. This current is carried predominantly by cations (PKPCl 71, pnapcl 41 and arginine is also conducted). The instantaneous current can be activated by ATP4– (e.g., ATP-activated mean K+ current density was –20 mA.m–2 at a membrane voltage of –20 mV) and by increasing cytosolic pH and Mg2+ (raising Mg2+ from 0 to 0.4 mm induced a mean current density increase of –7 mA.m–2 at –20 mV). Such current can be activated by simultaneous addition of putative in vivo concentrations of ATP4–/MgATP/Mg free 2+ (in the presence of bafilomycin to inhibit the vacuolar ATPase) and further modulated by cytosolic pH. With vacuolar K+ concentration greater than that of the cytosol, activation of the instantaneous current would mediate vacuolar K+ release over the range of physiological membrane voltage. It is argued that the ATP4–-activated current, in addition to acting as a K+ mobilization pathway, could provide a counter-ion (shunt) conductance, allowing the two electrogenic H+ pumps which reside in the vacuolar membrane to acidify the vacuolar lumen.A separate time-dependent current, which was not observed at low Ca2+ concentrations (less than 500 nm) could also be elicited by addition of Mg2+ at the cytoplasmic membrane face.     Summary The volume regulatory response of the Ehrlich ascites tumor was studied in KCl-depleted, Na+-enriched cells. Subsequent incubation in K+-containing NaCl medium results in the reaccumulation of K+, Cl, water and the extrusion of Na+. The establishment of the physiological steady state is due primarily to the activity of 2 transport systems. One is the Na/K pump (K M for K 0 + =3.5mm;J max=30.1 mEq/kg dry min), which in these experiments was coupled 1K+/1 Na+. The second is the Cl-dependent (Na++K+) cotransport system (K M for K 0 + =6.8mm;J max=20.8 mEq/kg dry min) which mediates, in addition to net ion uptake in the ratio of 1K+1Na+2Cl, the exchange of K i + for K 0 + . The net passive driving force on the cotransport system is initially inwardly directed but does not decrease to zero at the steady state. This raises the possibility of the involvement of an additional source of energy. Although cell volume increases concomitant with net ion uptake, this change does not appear to be a major factor regulating the activity of the cotransport system.  相似文献
