Bluegill were exposed to copper (2–0 mg/1−1 unfiltered) for 24, 48, 72 or 96 h at 24° C. This concentration approximated the 96 h LC20. Water content of liver increased 4–8% after 24 h exposure and muscle 4–6% by 48 h. These changes persisted throughout the 96 h experiments. Copper exposed fish exhibited decreases in muscle ATP, ADP, total adenylates, and energy charge by 48 h. There was a trend toward recovery of ATP with further exposure. Liver ATP of exposed fish was lower than controls at all intervals with the greatest difference evident at 48 h. No significant changes in brain adenylates were observed. Muscle and liver lactic acid was unchanged at 48 h exposure, therefore tissue hypoxia was not the cause of the adenylate changes. It was concluded that copper causes decreases in muscle and liver ATP several days before probable death of copper exposed fish and the changes seen are the result of dilution by increased tissue water, and the copper acting on certain cellular enzymes involved in detoxification and energy metabolism.  相似文献

Observations near a Swedish pulp mill at the Bothnian Sea during 1982-1992 revealed low fish abundance and a disturbed community structure. The presence of endocrine disrupting or toxic substances in effluent water was indicated by biomarker responses, impaired reproduction and stimulated growth in perch (Perca fluviatilis). In 1992 the bleaching process was altered and a secondary treatment system was installed, improving the quality and decreasing the amount of effluent. This paper presents the results of follow-up studies during 1995-1998 with the objective to analyse any recovery of the fish community due to changes in mill operations. Although abundance and diversity of the fish community did recover, an effect of enrichment, indicated by high catches of cyprinids, was still visible in the vicinity of the mill outlet. Sexual maturation was delayed, gonad development was retarded, average growth rate was faster, and the condition factor was higher in perch caught in the vicinity of the effluent outlet, than in perch captured elsewhere. It is concluded that substances inhibiting reproduction and causing growth disturbance were still present in the effluents in effective concentrations.  相似文献

A hallmark of anoxia tolerance in western painted turtles is relative constancy of tissue adenylate concentrations during periods of oxygen limitation. During anoxia heart and brain intracellular compartments become more acidic and cellular energy demands are met by anaerobic glycolysis. Because changes in adenylates and pH during anoxic stress could represent important signals triggering metabolic and ion channel down-regulation we measured PCr, ATP and intracellular pH in turtle brain sheets throughout a 3-h anoxic-re-oxygenation transition with 31P NMR. Within 30 min of anoxia, PCr levels decrease 40% and remain at this level during anoxia. A different profile is observed for ATP, with a statistically significant decrease of 23% occurring gradually during 110 min of anoxic perfusion. Intracellular pH decreases significantly with the onset of anoxia, from 7.2 to 6.6 within 50 min. Upon re-oxygenation PCr, ATP and intracellular pH recover to pre-anoxic levels within 60 min. This is the first demonstration of a sustained reversible decrease in ATP levels with anoxia in turtle brain. The observed changes in pH and adenylates, and a probable concomitant increase in adenosine, may represent important metabolic signals during anoxia.  相似文献

Citrobacter freundii was grown aerobically in a chemostat on a mineral medium witn galactose or glucose as carbon and energy sources under limitation by carbon or nitrogen source respectively. At various specific growth rates ranging from 7 to 95% μmax the culture in steady state was analysed and growth yield, specific metabolic rate of substrate utilization, intracellular concentration of pyruvate, ATP, ADP, AMP and energy charge were determined and plotted as functions of dilution rate. In all four types of experiments the physiological state of cells remained practically independent of dilution rate up toD = 0.6 μmax, and at a given specific growth rate nearly independent on μmax and type of limitation. At approximatelyD = 0.6 μmax, which is close to the maximum output dilution rateD m, the physiological state of the cells changed: growth yields decreased and intr cellular pyruvate and adenylates concentrations increased. Consequently, in a given medium two dilution rates exist at which growth rate dx/dt is the same but the physiology of the population is quite different.  相似文献

The intertidal wormSipunculus nudus was exposed to various temperatures for an analysis of the integrated changes in energy and acid-base status. Animals were incubated in sea water or maintained for up to 8 days at 4 and 0°C while dwelling in the sediment. Cannulation of the animals prior to experimentation allowed the analysis of blood gas parameters ( , and pH). fell to 0 torr within 8 days at 0°C. A simultaneous reduction of ventilatory activity was derived from measurements of the pattern of coelomic fluid pressure changes associated with ventilatory movements. The increase in and an onset of anaerobic metabolism, indicated by the accumulation of end products like acetate and propionate both in the coelomic fluid and the body wall musculature, led to the development of a progressive acidosis and a deviation from the alphastat regulation of intracellular pH seen in unburied animals. The drop in intracellular pH together with the depletion of the adenylates and the phosphagen, phospho-l-arginine, reflect a significant decrease in the Gibb's free-energy change of ATP hydrolysis. These changes are interpreted to indicate lethal cold injuries, because recovery was not possible when the animals were returned to 12°C after more than 2 days of exposure to 0°C. A low critical temperature indicating the onset of cold-induced anaerobiosis is concluded to exist below 4°C owing to the insufficient response of the ventilatory system to the developing hypoxia.  相似文献

The objective of the study was to examine the long-term changes in biochemical/microbial indicators of soil quality due to clear felling of mangroves for establishment of plantations. The biochemical/microbial parameters included dissolved organic-C (DOC) and -N (DON), soil microbial biomass-C (SMBC), -N (SMBN) and -P (SMBP), soil respiration (SR), metabolic quotient (qCO2), adenylates (ATP, AMP and ADP), adenylate energy charge (AEC), ergosterol and their ratios. Results revealed that the undisturbed mangroves possessed considerably greater amounts of soil organic C, DOC and DON. Consequently, SMBC, SMBN and SMBP showed marked reductions in the plantations suggesting an average loss of 66%, 49% and 75%, respectively due to changed land use. Likewise, SR decreased by 46.4% in the plantations. Enhanced qCO2 levels in the plantations indicated a microbial community under stress with a high maintenance carbon demand, while lower qCO2 levels in the mangroves indicated an efficient microbial community and a better use of available organic substrates. The levels of ATP, AMP and ADP followed a trend identical to that of SMB and SR. Greater ergosterol concentration led to greater ergosterol/SMBC ratio suggesting a shift in the microbial community structure from a primarily fungi dominated SMB in the mangroves to a fungi recessive SMB in the plantations.  相似文献

The functional response to and recovery from coal-coking waste effluent was evaluated for sediment microbial communities. Twenty estimates of microbial population density, biomass, and activity were measured five times during a 15-month period. Significant effects on microbial communities were observed in response to both wastewater contamination and diversion of the wastewater. Multivariate analysis of variance and discriminant analysis indicated that accurate differentiation between uncontaminated and contaminated sediments required a minimum of nine estimates of community response. Total viable population density, ATP, alkaline phosphatase, naphthalene, and phenanthrene mineralization rates were found to be highly weighted variables in site discrimination. Lipid and glucose mineralization, nitrogen fixation, and sediment protein also contributed significantly to explaining variation among sites. Estimates of anaerobic population densities and rates of methane production contributed little to discrimination among sites in the environment examined. In general, total viable population density, ATP, and alkaline phosphatase activity were significantly depressed in contaminated sediments. However, after removal of this contamination, the previously affected sites demonstrated greater temporal variability but a closer approximation of the mean response at the control site. Naphthalene and phenanthrene mineralization did not follow the general trend and were elevated at the contaminated sites throughout the investigation. Results of the investigation supported the hypothesis that multiple functional measures of microbial community response are required to evaluate the effect of and recovery from environmental contamination. In addition, when long-term effects are evaluated, select physiological traits, i.e., polyaromatic hydrocarbon mineralization, may not reflect population and biomass estimates of community response.  相似文献

Effective and rapid inactivation of cellular metabolism is a prerequisite for accurate metabolome analysis. Cold methanol quenching is commonly applied to stop any metabolic activity and, at the same time remaining the cells’ integrity. However, it is reported that especially prokaryotic cells like Escherichia coli and Corynebacterium glutamicum tend to leak intracellular metabolites during cold methanol quenching. In this work leakage of adenylates is quantified for different quenching fluids. Further, a methanol/glycerol based quenching fluid is proposed, which reduces leakage drastically compared to the commonly applied methanol/water solution (16% ATP leakage compared to more than 70%).  相似文献

The stability of ecological communities depends strongly on quantitative characteristics of population interactions (type‐II vs. type‐III functional responses) and the distribution of body masses across species. Until now, these two aspects have almost exclusively been treated separately leaving a substantial gap in our general understanding of food webs. We analysed a large data set of arthropod feeding rates and found that all functional‐response parameters depend on the body masses of predator and prey. Thus, we propose generalised functional responses which predict gradual shifts from type‐II predation of small predators on equally sized prey to type‐III functional‐responses of large predators on small prey. Models including these generalised functional responses predict population dynamics and persistence only depending on predator and prey body masses, and we show that these predictions are strongly supported by empirical data on forest soil food webs. These results help unravelling systematic relationships between quantitative population interactions and large‐scale community patterns.  相似文献

Levels of ATP and ADP were studied in primary leaves of barley (Hordeum vulgare L. cv. Viner) seedlings grown under blue (BL) or red light (RL) of various irradiances. In mature leaf segments, BL stimulated a greater accumulation of adenylates than RL. Transfer of barley seedlings from RL to BL for 48 h caused about a twofold increase in the content of adenylates, probably due to de-novo synthesis of adenine nucleotides. Weak BL was found to stimulate an increase in the adenylate content and a higher irradiance enhanced the stimulatory effect. The adenylate content increased markedly from the base towards the tip of barley leaves grown under BL but not in those grown under RL. However, the adenylate content was higher in the basalmost segment of barley leaves grown under RL, indicating that the action of RL on adenylate content proceeded more rapidly than that of BL. The same conclusion could be drawn from the results of experiments with de-etiolated leaves. A linear relationship was established between the maximum rate of CO2 fixation and the ATP or ADP content in mature segments of primary barley leaves. The possible involvement of two photoreceptors, phytochrome and cryptochrome, in the long-term light regulation of the total content of adenylates in primary barley leaves is discussed.Abbreviations BL blue light - Chl chlorophyll - RL red light  相似文献

The protozoan community in eleven activated sludge wastewater treatment plants (WWTPs) in the greater Dublin area has been investigated and correlated with key physio-chemical operational and effluent quality parameters. The plants represented various designs, including conventional and biological nutrient removal (BNR) systems. The aim of the study was to identify differences in ciliate community due to key design parameters including anoxic/anaerobic stages and to identify suitable bioindicator species for performance evaluation. BNR systems supported significantly different protozoan communities compared to conventional systems. Total protozoan abundance was reduced in plants with incorporated anoxic and anaerobic stages, whereas species diversity was either unaffected or increased. Plagiocampa rouxi and Holophrya discolor were tolerant to anoxic/anaerobic conditions and associated with high denitrification. Apart from process design, influent wastewater characteristics affect protozoan community structure. Aspidisca cicada was associated with low dissolved oxygen and low nitrate concentrations, while Trochilia minuta was indicative of good nitrifying conditions and good sludge settleability. Trithigmostoma cucullulus was sensitive to ammonia and phosphate and could be useful as an indicator of high effluent quality. The association rating assessment procedure of Curds and Cockburn failed to predict final effluent biological oxygen demand (BOD5) indicating the method might not be applicable to treatment systems of different designs.  相似文献

Cold hardened seedlings of winter wheat (Triticum aestivum L. em Thell) show an hypoxic hardening response: an exposure to low temperature flooding increases the tolerance of plants to a subsequent ice encasement exposure. Seedlings of winter barley (Hordeum vulgare L.) do not show such a response in similar experimental conditions. During ice encasement, there are general declines in adenylate energy charge (AEC), total adenylates and ATP:ADP ratios in the crown tissues of two winter wheat cultivars, and a winter barley, but rates of decline are faster in the barley. When the ice period is preceded by low temperature flooding of the whole plant, levels of the adenylate components are raised significantly in the wheats, and to a lesser extent in the barley. The survival of plants in ice preceded by flooding is related to the increased initial level of adenylates at the onset of the ice encasement stress, and the maintenance of higher levels of adenylates and ATP in the early stages of ice encasement as a result of accelerated rates of glycolysis. Higher survival of both winter wheat and barley plants during ice encasement in the light is also associated with significantly higher levels of AEC and adenylates in the early stages of ice encasement.  相似文献

应用PFU法收集微型生物群落,监测鸭儿湖的农药污染。原生动物对五个氧化塘内农药废水的毒性反应,可用群集过程来表示,无论室内或是野外试验,结果基本一致。其中微型生物群落的结构参数有原生动物种类组成和种数、叶绿素a含量、水细菌数量;功能参数有反映原生动物群集过程中的S_(eq)、G、t_(90%)和微型生物的呼吸速度。功能和结构的参数均能较准确地反映出氧化塘系统的净化过程。PFU法可应用于预报污染物的生物效应浓度,初认为是一种经济、快速、正确的生物学监测方法。  相似文献

Here, we analyze how the set of nucleotides in the cell is equilibrated and how this generates simple rules that help the cell to organize itself via maintenance of a stable non‐equilibrium state. A major mechanism operating to achieve this state is thermodynamic buffering via high activities of equilibrating enzymes such as adenylate kinase. Under stable non‐equilibrium, the ratios of free and Mg‐bound adenylates, Mg2+ and membrane potentials are interdependent and can be computed. The adenylate status is balanced with the levels of reduced and oxidized pyridine nucleotides through regulated uncoupling of the pyridine nucleotide pool from ATP production in mitochondria, and through oxidation of substrates non‐coupled to NAD+ reduction in peroxisomes. The set of adenylates and pyridine nucleotides constitutes a generalized cell energy status and determines rates of major metabolic fluxes. As the result, fluxes of energy and information become organized spatially and temporally, providing conditions for self‐maintenance of metabolism.  相似文献

It is still unclear if performance recovery in postischemic hearts is related to their tissue level of high-energy phosphates before reflow. To test the existence of this link, we monitored performance, metabolism and histological damage in isolated, crystalloid-perfused rat hearts during 20 min of low-flow ischemia (90% coronary flow reduction) and reflow. To prevent interference from different ischemia times and perfusing media compositions, the ischemic ATP level was varied by changing energy demand (electrical pacing at 330 min–1). Under full coronary flow conditions, work output, as well as ATP and phosphocreatine contents were the same in control, spontaneously contracting (n = 23) and paced (n = 21) hearts. During low-flow ischemia, the higher work output (p < 0.0001) in paced hearts decreased their tissue content of ATP, phosphocreatine and total adenylates and purines (p < 0.05), as opposed to maintained values in control hearts. During reflow, the recovery of mechanical performance and O2 uptake was 94 ± 5% and 110 ± 9% (p = NS vs. baseline) in controls, vs. 71 ± 5% and 74 ± 6% in paced hearts (p < 0.004 vs. baseline). The levels of ATP and total adenylates and purines remained constant in control, but were markedly depressed (p < 0.05 vs. baseline) in paced hearts. Phosphocreatine+creatine was the same in both groups. These data, together with the observed lack of creatine kinase leakage and of structural damage, indicate that myocardial recovery during reflow reflects the tissue level of ATP, phosphocreatine and total adenylates and purines during ischemia, regardless of physical cell damage.  相似文献

Microbial communities play a vital role in maintaining soil health. A multiphasic approach to assess the effect of pulp and paper mill effluent on both the structure and function of microbial soil communities is taken. Bacterial communities from agricultural soils irrigated with pulp and paper mill effluent were compared to communities form soils irrigated with well water. Samples were taken from fields in the state of Uttarakhand, India, where pulp and paper mill effluent has been used for irrigation for over 25 years. Comparisons of bacterial community structure were conducted using sequencing of the 16S rRNA gene from both isolates and clone libraries attained from the soil. Community-level physiological profiling was used to characterize the functional diversity and catabolic profile of the bacterial communities. The multiphasic approach using both physiological and molecular techniques proved to be a powerful tool in evaluating the soil bacterial community population and population differences therein. A significant and consistent difference in the population structure and function was found for the bacterial communities from soil irrigated with effluent in comparison to fields irrigated with well water. The diversity index parameters indicated that the microbial community in pulp and paper mill effluent irrigated fields were more diverse in both structure and function. This suggests that the pulp and paper mill effluent is not having a negative effect on the soil microbial community, but in fact may have a positive influence. In terms of soil health, this finding supports the continued use of pulp and paper mill effluent for irrigation. This is however only one aspect of soil health which was evaluated. Further studies on soil resistance and robustness could be undertaken to holistically evaluate soil health in this situation.  相似文献

The cryoprotective additives glycerol and dimethylsulphoxide were found to be toxic to Chlorella cells at concentrations greater then 2.5% w/v. Polyvinylpyrrolidone, was not damaging up to a concentration of 15% w/v. Chlorella 211/7a had a recovery rate greater than 95% at all rates of cooling studied. With Chlorella 211/8h the survival was lower than 0.1% at all rates examined. The addition of dimethylsulphoxide (5% w/v) to Chlorella 211/8h increased the recovery, particularly at the faster rates of cooling; with polyvinylpyrrolidone (10% w/v) there was an optimum range of cooling rate.Cells of Chlorella 211/7a from the exponential phase of growth were found to be damaged both by a temperature reduction from 25°C to 0°C (thermal shock) and by freezing and thawing. The effect of a low phosphate concentration on intracellular adenine nucleotide content, oxygen consumption and poly--hydroxybutyrate deposition was investigated with N-free and NH 4 + batch cultures of Azotobacter vinelandii. When the microorganisms were cultured under low-phosphate concentrations the cells contained much larger amounts of poly--hydroxybutyrate, but displayed lower oxygen consumption activities and energy charge values than did control cells. Also, the ratio ATP to ADP was much higher in control cells and the intracellular levels of ATP were lower in low-phosphate cells.  相似文献

Abstract Predicting variations in microbial growth rates (μ) in Nature requires a method that will detect departures from optimal μ, determine the time scale at sub-optimal μ, and measure the restoration or reversibility toward optimal μ. Isotopic flux measurements are equivocal because of uncertainties in their relationship to μ, particularly in non-steady state condictions. After perturbing a natural microbial community and a clone of the phytoplankter Selenastrum capricornutum with two different toxicants, we examined the kinetics of the adenylate energy charge, and followed changes in biomass over time with the total adenylate and chlorophyll a concentrations. We determined that exposure to cadmium resulted in a reduction in μ; reversible within a short time, whereas exposure to the industrial effluent caused an exponential loss of adenylates and complete inhibition of growth. Upon perturbing a microbiol community, the study of adenylate energy charge kinetics detects departures from optimal μ, measures their duration and reversibility.  相似文献

Aspergillus niger Yang No. 2 and its mutant strain SL1 were grown in solid state fermentation. Samples were taken after 2, 4 and 6 days of incubation and the mycelia were analysed for their intracellular concentrations of some organic acids and adenylates and the activities of selected enzymes. Strain Yang No. 2 contained high concentrations of citrate with very little oxalate, while strain SL1 contained lower concentrations of citrate but considerably higher concentrations of oxalate. As the fermentation proceeded, strain Yang No. 2 showed a much higher ratio of ATP:AMP than did strain SL1. In addition, the enzyme ATP:citrate lyase became undetectable during citrate accumulation in strain Yang No. 2, while its activity remained high during oxalate accumulation in strain SL1. It is proposed that citrate accumulation by strain Yang No. 2 during solid state fermentation is due to blockage of its metabolism in the mitochondrion via inhibition of isocitrate dehydrogenase by the high ATP:AMP ratio, and in the cytosol by repression of ATP:citrate lyase activity.  相似文献
