Welfare Quality® (WQ) assessment protocols place the emphasis on animal-based measures as an indicator for animal welfare. Stakeholders, however, emphasize that a reduction in the time taken to complete the protocol is essential to improve practical applicability. We studied the potential for reduction in time to complete the WQ broiler assessment protocol and present some modifications to the protocol correcting a few errors in the original calculations. Data was used from 180 flocks assessed on-farm and 150 flocks assessed at the slaughter plant. Correlations between variables were calculated, and where correlation was moderate, meaningful and promising (in terms of time reduction), simplification was considered using one variable predicted from another variable. Correlation analysis revealed a promising correlation between severe hock burn and gait scores on-farm. Therefore, prediction of gait scores using hock burn scores was studied further as a possible simplification strategy (strategy 1). Measurements of footpad dermatitis, hock burn, cleanliness and gait score on-farm correlated moderately to highly with slaughter plant measurements of footpad dermatitis and/or hock burn, supporting substitution of on-farm measurements with slaughter plant data. A simplification analysis was performed using footpad dermatitis, hock burn, cleanliness and gait scores measured on-farm predicted from slaughter plant measurements of footpad dermatitis and hock burn (strategy 2). Simplification strategies were compared with the full assessment protocol. Close agreement was found between the full protocol and both simplification strategies although large confidence intervals were found for specificity of the simplified models. It is concluded that the proposed simplification strategies are encouraging; strategy 1 can reduce the time to complete the on-farm assessment by ~1 h (25% to 33% reduction) and strategy 2 can reduce on-farm assessment time by ~2 h (50% to 67% reduction). Both simplification strategies should, however, be validated further, and tested on farms with a wide distribution across the different welfare categories of WQ.  相似文献

The Welfare Quality® protocols provide a multidimensional assessment of welfare, which is lengthy, and hence limited in terms of practicality. The aim of this study was to investigate potential ‘iceberg indicators’ which could reliably predict the overall classification as a means of reducing the length of time for an assessment and so increase the feasibility of the Welfare Quality® protocol as a multidimensional assessment of welfare. Full Welfare Quality® assessments were carried out on 92 dairy farms in England and Wales. The farms were all classified as Acceptable or Enhanced. Logistic regression models with cross validation were used to compare model fit for the overall classification on farms. ‘Absence of prolonged thirst’, on its own, was found to correctly classify farms 88% of the time. More generally, the inclusion of more measures in the models was not associated with greater predictive ability for the overall classification. Absence of prolonged thirst could thus, in theory, be considered to be an iceberg indicator for the Welfare Quality® protocol, and could reduce the length of time for a farm assessment to 15 min. Previous work has shown that the parameters within the Welfare Quality® protocol are important and relevant for welfare assessment. However, it is argued that the credibility of the published aggregation system is compromised by the finding that one resource measure (Absence of prolonged thirst) is a major driver for the overall classification. It is therefore suggested that the prominence of Absence of prolonged thirst in this role may be better understood as an unintended consequence of the published measure aggregation system rather than as reflecting a realistic iceberg indicator.  相似文献

Welfare concerns for intensive beef production have often been raised, but on-farm welfare assessment studies are rare. The aim of this study was to apply the Welfare Quality® (WQ) welfare assessment system for fattening cattle on beef bull farms to evaluate the state of welfare at the level of WQ measures and of aggregated scores, as well as overall classification. In addition, the purpose was to evaluate two ways of providing feedback information to the farmers with regard to possible welfare improvements on the farms. The study was conducted in Austria, Germany and Italy on a total of 63 beef bull farms with deep litter or cubicle-housing systems. Assessments were carried out 3 times (1 month and 7 months apart from the initial visit). In every country, farmers were assigned to two treatment groups (feedback from initial visit as written report, F, written feedback plus oral advice, FA) and a control group (C), which did not receive any feedback. At the criterion level, the highest average welfare scores were obtained from ‘Absence of prolonged hunger’ (94/100 points) followed by ‘Absence of pain induced by management procedures’ (88/100) and ‘Comfort around resting’ (77/100). Most welfare concerns related to the criteria ‘Absence of disease’ (40/100), ‘Expression of social behaviour’ (44/100) and ‘Positive emotional state’ (48/100), thus indicating room for improvements. Two-thirds of the farms achieved the ‘Enhanced’ level, about one-third was judged ‘Acceptable’ and only one farm ‘Excellent’. After 6 months of monitoring period, there was no significant welfare improvement in both the treatment groups as compared with the control group. Reasons for the lack of effect may mainly be seen in the short monitoring period and a lack of external incentives. In conclusion, the WQ assessment system revealed areas for improvement, but longer term studies and investigations on alternative ways of transferring outcomes from on-farm welfare assessments to farmers should be carried out in future.  相似文献

Sample sizes of welfare assessment protocols must warrant to reflect prevalences on-farm properly – regardless of farm size. Still, solely a fixed sample size was specified for the Welfare Quality® protocol for sows and piglets. The present study investigated whether animals may be assessed from only one body side as applied in the protocol and whether the pre-set sample size of 30 animals mirrors the prevalences of the animal-based indicators on-farm in the gestation unit considering different farm sizes. All indicators were assessed for both sides of an animal’s body by one observer on 13 farms in Germany, which were visited five times within 10 months. The farm visits were treated as independent since different animals were housed in the gestation units. The number of sows in the gestation units varied between 18 and 549 animals. The comparison of sides was carried out calculating exact agreement between animals’ sides and a Wilcoxon signed-rank test (W). The results signified that it is sufficient to assess the animal from one side (exact agreement: 88.3% to 99.5%, except for bursitis (70.0%); W: P-values 0.14 to 0.92). However, if side preferences existed in the indicator bursitis a potential bias must be considered. In the following, the sample size was evaluated by comparing samples’ prevalences against true prevalence, that is, the prevalence of all observed animals in the gestation unit in each farm visit. Therefore, subsets of data were generated by applying simple random sampling without replacement. The samples randomly included the animals’ right or left sides. Linear regression was rated as appropriate provided: coefficient of determination R2 ≥ 0.90, slope = 1 and intercept = 0 signifying exact agreement. The results revealed that the sample size required by the protocol and the application of calculation formulas are solely appropriate to mirror the prevalences of frequent indicators in the gestation unit, for example, bursitis (mean prevalence 34.4%). Using a proportion of animals, for example, a sample of 30% of all observed animals in a farm visit, pointed out that proportions must increase with indicators’ underlying prevalence narrowing 0.00%. Local infections (mean prevalence 13.3%) needed samples including 60% of all observed animals in each farm visit, whereas vulva lesions (mean prevalence 7.28%) only reached accuracy with the inclusion of 70% of the animals. Indicators with a mean prevalence of <1% were not analysed but can most likely only be ascertained by the assessment of all animals.  相似文献

Animal welfare in livestock production is of great interest to consumers. The organic farming approach strives to ensure animal welfare based on preventive measures, but there are very few scientific studies that compare the actual differences in animal welfare between organic and conventional farms. Those studies that have been carried out frequently focus on specific aspects of animal welfare, mostly health issues. The aim of the present study, therefore, was to investigate the effects of the farming system on the welfare of dairy cows in a more holistic way. Although this study was carried out in just two federal states of Germany, the results could serve as a suitable model for the whole country. We used the Welfare Quality assessment protocol to measure welfare for dairy cattle (Welfare Quality®, 2009) and the results showed significant differences (P < 0.05) between organic and conventional farms, but there was also considerable variance between individual farms of the same farming system. Organic farms scored higher in all four Welfare Quality® principles: “Good Feeding”, “Good Housing”, “Good Health” and “Appropriate Behavior” compared to conventional farms. In particular, organic farms obtained higher scores with respect to Welfare Quality® measures of resting comfort, which contributes to a lower percentage of lameness; organic farms also implemented less painful methods for disbudding, or indeed carried out no disbudding, and provided access to pasture and outdoor exercise. However, organic farms still have room for improvement, especially with respect to animal health. Therefore, outcome‐based specifications should be included in the current (purely action-oriented) European regulation of organic production (EC, 2008; EU, 2018) to safeguard the health‐related aspects of animal welfare.  相似文献

There is a large demand for holistic welfare assessment systems that result in a singular balanced summary of welfare. The Welfare Quality® (WQ) broiler protocol summarizes 18 welfare measures into four principles (‘good feeding’, ‘good housing’, ‘good health’ and ‘appropriate behaviour’), which are then integrated into one overall category (‘excellent’, ‘enhanced’, ‘acceptable’ or ‘not classified’). But the protocol is time consuming which hampers implementation. Furthermore, WQ’s aim to assess animal welfare in a wide range of husbandry systems may decrease its ability to discriminate between flocks from the same system. We applied the protocol in the context of intensive indoor rearing to assess whether it discriminated sufficiently between flocks, could be shortened without losing essential information, and provided a balanced summary of welfare. The vast majority of the flocks (88%) received the same overall classification (acceptable) whilst all other flocks received an adjacent classification (enhanced), suggesting poor discriminative capacity. For 95% of the flocks overall classification was explained by two measures only (‘drinker space’ and ‘stocking density’). A system based on these two measures would reduce assessment time from 3.5 h to a few minutes. However, both measures’ validity can be questioned as they are risk factors for poor welfare rather than animal-based outcome measures and they suffer from methodological weaknesses. Furthermore, the possibility for such an extreme simplification raises doubts on whether the overall classification reflects a balanced summary of different welfare aspects. In line with this, overall classification was not affected by replacing single measures within the ‘good health’ and ‘appropriate behaviour’ principles with realistically attainable minima or maxima for intensively reared flocks. Even replacing either of these two principles entirely with their realistically obtainable minimum or maximum did not affect classification. Such insensitivity to change may discourage attempts to improve the welfare of intensively reared flocks when assessments are made based on the overall classification. This calls for an adjustment of the classification system, which is currently being developed by the Welfare Quality Network.  相似文献

Background

Minimally invasive breast biopsy technology is now considered a standard of care for the diagnostic evaluation of suspicious breast lesions. The aim of the current study was to present a comprehensive evaluation of the 8-gauge vacuum-assisted Mammotome® system for ultrasound-guided diagnostic biopsy and selective excision of breast lesions.

Methods

A retrospective analysis was conducted of a series of 304 consecutive 8-gauge Mammotome® procedures that were performed under ultrasound guidance by a single surgeon from March 2004 to December 2006. Multiple variables, including patient demographics, characteristics of the breast lesion (based on ultrasound and mammography), procedural and histopathology variables, and interval follow-up variables (based on ultrasound and mammography), were evaluated.

Results

Among 304 procedures, 235 (77%) were performed with the presumption of complete excision of the ultrasound lesion during Mammotome® core acquisition, while 69 (23%) were performed with only partial excision of the ultrasound lesion during Mammotome® core acquisition (diagnostic tissue sampling only). 100% of all ultrasound lesions were accurately diagnosed, demonstrating no apparent false-negative results among the 256 patients that were compliant with follow-up at a median interval follow-up duration of 11 months (range 1 to 37). Likewise, 89% of all appropriately selected ultrasound lesions were completely excised, as demonstrated on interval follow-up ultrasound at a median time of 6 months (range, 3 to 16). There were no independent predictors of successful complete excision of any given appropriately selected ultrasound lesion by the ultrasound-guided 8-gauge Mammotome® biopsy technique.

Conclusion

The 8-gauge vacuum-assisted Mammotome® system is highly accurate for ultrasound-guided diagnostic biopsy of suspicious breast lesions and is highly successful for complete excision of appropriately selected presumed benign breast lesions. This particular technology should be routinely offered to all appropriately selected patients that are evaluated by physicians involved in breast-specific health care.  相似文献

Selective screening media for the detection and identification of Aeromonas strains are needed to guide primary isolation procedures in the clinical laboratory. This study compared the selective CromoCen? AGN chromogenic agar medium for the detection and identification of Aeromonas strains that were isolated from various samples against the conventional selective agar media that are commonly used for the isolation of this organism in food, environmental and clinical samples. The Miles and Misra and ecometric methods were used to evaluate the microbiological performance of CromoCen? AGN chromogenic agar medium, which was shown to be satisfactory. A total of 14 reference Aeromonas strains, 44 wild strains and 106 clinical stool specimens were examined using both non-chromogenic selective agars that are commonly used for Aeromonas isolation and CromoCen? AGN agar. The latter exhibited 94.73% sensitivity and 100% specificity for the various samples. On CromoCen? AGN agar medium, Aeromonas formed colonies with light green, greenish and salmon pigments with or without a surrounding wide transparent zone (halo) of 2-3mm in diameter around the entire border. This medium is recommended for the isolation and potential identification of the Aeromonas genus.  相似文献

Background

The availability of both mouse and human draft genomes has marked the beginning of a new era of comparative mammalian genomics. The two available mouse genome assemblies, from the public mouse genome sequencing consortium and Celera Genomics, were obtained using different clone libraries and different assembly methods.

Results

We present here a critical comparison of the two latest mouse genome assemblies. The utility of the combined genomes is further demonstrated by comparing them with the human 'golden path' and through a subsequent analysis of a resulting conserved sequence element (CSE) database, which allows us to identify over 6,000 potential novel genes and to derive independent estimates of the number of human protein-coding genes.

Conclusion

The Celera and public mouse assemblies differ in about 10% of the mouse genome. Each assembly has advantages over the other: Celera has higher accuracy in base-pairs and overall higher coverage of the genome; the public assembly, however, has higher sequence quality in some newly finished bacterial artifical chromosome clone (BAC) regions and the data are freely accessible. Perhaps most important, by combining both assemblies, we can get a better annotation of the human genome; in particular, we can obtain the most complete set of CSEs, one third of which are related to known genes and some others are related to other functional genomic regions. More than half the CSEs are of unknown function. From the CSEs, we estimate the total number of human protein-coding genes to be about 40,000. This searchable publicly available online CSEdb will expedite new discoveries through comparative genomics.  相似文献

We evaluated a new protocol for measurement of cyclosporine A (CsA) 2 H after dose (C2) on the V-Twin? analyzer. Imprecision, recovery, and linearity were determined using CsA-spiked blood pools. Accuracy was evaluated using specimens from renal, cardiac, and liver transplant patients, and results were compared with those from liquid chromatography-tandem mass spectrometry (LC-MS/MS) and the Abbott TDx?/TDxFLx? assay. Cross-reactivity and interferences were assessed in the presence of 800 ng/mL CsA. Imprecision coefficients of variation were 3.3%-4.8% (within run) and 5.9%-8.7% (total). Recovery was within 10% of the expected values. Linearity was 350-2,000 ng/mL. Calibration was stable for ≥ 2 weeks. Method comparison showed regression statistics: V-Twin? = 1.01 × LC tandem MS + 36.1, r = 0.971; V-Twin? = 1.13 × Abbott - 92.4, r = 0.969. Metabolite cross-reactivity and interference (endogenous substances and drugs) were within ±10%. The C2 protocol on the V-Twin? analyzer provides acceptable assay performance and accurate determination of whole blood CsA drawn at 2 H after dose.  相似文献

Behavior and average daily gain were evaluated after mixing pigs amongst litters or maintaining them as littermate groups, after weaning and initially limit or full fed. Limit fed mixed groups fought most and gained least of the treatment groups. Limit fed littermate groups fought least of the groups. Salt marsh vegetation is one important biological element for establishing the ecological quality status (EQS) while evaluating transitional waters (TW). This intertidal plant community is sensitive to the most important pressures present in estuaries. In this study, an ecological index based multi-metric was established taking into account the species composition and ecological relations in Portuguese salt marsh habitats. During the AQuA-Index elaboration it was found that the parameters that respond better to the variability of ecological conditions present nowadays are the Shannon Diversity Index, the Shannon Maximum Diversity Index, the number of halophytic species, the Margalef Diversity Index and the Pielou Equitability Index. The application of the AQuA-Index index to eight TW systems varying in morphology and pressures, demonstrated that the metric responds to the ecological quality of each system. Also, it showed some advantages in comparison with other approaches, in zones with a relevant proportion of middle and upper marsh and where historical data were not available. This way the AQuA-Index proved to be an efficient Index for ecological quality assessment, using salt marsh vegetation as indicator.  相似文献

The objective of this study was to investigate if female growing pigs spend similar amounts of time exploring a wooden stick as they do exploring enrichment material regardless of its proximity to the feeder. Forty-eight pigs aged 18 to 26 weeks allocated to 16 pens with three pigs per pen were studied. Fifty percent of the pens had a wooden stick beside the feeder, and the rest had a similar stick opposite to the feeder. Two observers assessed the pigs by means of scan and focal sampling. The pigs spent more time (p < .0001) exploring the wood during the first week than during the rest of the study (10.9% vs 3.6%). The pigs with the wood close to the feeder spent less (p = .0001) time resting (29.9%) and more (p < .0001) time exploring (6.3%) the wood than did pigs with the wood opposite to the feeder (32.4% and 2.5%, respectively). In conclusion, a wooden stick placed close to the feeder was associated with more exploratory behaviorcompared with a similar stick placed opposite to the feeder.  相似文献

Fifty-six strains of rapidly growing mycobacteria (RGM) and 14 strains of aerobic actinomycetes as quality controls (QC) were tested in the API (RAPID) Coryne system version 2. Both groups yielded codes with low identification scores, considerable overlaps, and similar diagnoses. No species-specific codes were observed. For centuries, entire male pigs have been castrated to reduce the risk of boar taint. However, physical castration of pig is increasingly being questioned with regard to animal welfare considerations. Immunization against gonadotrophin releasing hormone (GnRH) provides an alternative to physical castration. Using the currently available commercial product (Improvac®; Pfizer Animal Health), a two-dose regimen of a GnRH vaccine is administered. After the second vaccination, a substantial increase in feed consumption has been reported, which may be associated with increased body fatness and decreased feed efficiency when compared with unvaccinated entire male pigs. The aim of the present study was to investigate the effect of a feed restriction on these traits and on the behaviour of 120 group-housed entire males (five pigs/pen) vaccinated against GnRH. The first vaccination was performed at 62 days of age and the second (V2) at 130 days of age. Pigs were slaughtered in two batches 4 to 5 weeks after V2. They were either offered feed ad libitum over the 22 to 114 kg BW range (AL treatment) or ad libitum up to a maximum of 2.50 (R2.50 treatment) or 2.75 kg/day per pig (R2.75 treatment). Behavioural observations and skin lesion scoring were conducted 1 week before V2, and 1 and 3 weeks after V2. At slaughter, the volumetric lean meat content was measured using an X-ray computed tomography scanner. Between V2 and slaughter, the average feed intakes for the R2.75 and R2.50 treatments were 15% and 22% lower than the average AL feed intake (3.20 kg/day), respectively. Feed restriction was associated with a reduced average daily gain after V2 (846, 932 and 1061 g/day in the R2.50, R2.75 and AL groups, P < 0.01) but had no effect on the feed conversion ratio (3.00 kg feed/kg BW gain on average, P = 0.62). No difference was observed in the lean meat content (71.8%, 70.7% and 70.4% in the R2.50, R2.75 and AL groups, P = 0.14), despite a reduced backfat thickness measured in restrictively fed pigs (12.0, 13.0 and 13.6 mm in the R2.50, R2.75 and AL groups, P < 0.01). Higher skin lesion scores were observed 3 weeks after V2 in R2.50 and R2.75 pigs than in the AL ones (scores 33.4, 27.7 and 25.5, respectively, P = 0.04). These results, combined with an unimproved feed efficiency and no marked change in carcass characteristics, suggest that immunologically castrated pigs should not be restrictively fed during the late finishing period.  相似文献

Invasive aspergillosis is a serious problem for immunocompromised patients, especially if neutropenic. The diagnosis of this infection is complicated, since clinical symptoms are often similar to those of other fungal diseases. The chance of detecting the presence of a specific antigen in the serum could confirm the suspected clinical diagnosis and. perhaps, be useful for the follow-up of the patient. The Medical Mycology Committee of the Associazione Microbiologi Clinici Italiani (AMCLI) decided to evaluate in a multicenter prospective study (from I November 1998 to 28 February 1999) the performance of the Platelia Aspergillus Kit (Bio-Rad) for the detection of Aspergillus galactomannan in human serum. The enrolled patients included various groups of immunosuppressed patients (mostly neutropenic). Blood samples were drawn at the time of enrollment. This decision was based upon a clinical diagnosis of probable aspergillosis (antibiotic non-responsive fever for at least 96 hours, cough, hemophthosis and positive chest X-ray). Additional blood samples were drawn on days 3, 6, 9, 12, 15 and 21. Culture and histopathologic examinations were performed according to the individual laboratory workflow. For each patient the laboratory filled a form with all the available clinical information, to create a database on which to evaluate the results of the test. During the study, 187 patients with various kinds of immunosuppression were enrolled. A total of 256 sera were tested: for 117 patients (62.6%) only the basal sample was tested, whereas for the 70 symptomatic patients (37.4%) multiple specimens (range: 1-6) were tested. The results allowed the laboratories to exclude (68.6%) or confirm (31.5%: confirmed and/or probable) the clinical diagnosis of invasive aspergillosis; 4 cases remained undetermined. Based on the results of this study, it seems that the use of this test should be limited to those patients with clinical symptoms of aspergillosis.  相似文献
