Here, we study the therapeutic effect of Acanthopanax senticosus total flavonoids (ASTFs) using a mouse intestinal inflammation model. The inflammation model used in the present study was developed through lipopolysaccharide (LPS) treatment of mice. The experimental mice were divided into a control group, model group (10 mg/kg LPS), dexamethasone group (1 mg/kg DEX) and ASTF low-, medium- and high-dosage groups (200, 400 and 800 mg/kg, respectively). The morphological and structural changes in the ileum, jejunum and duodenum were observed using HE staining. The number of intestinal goblet cells (GCs) was calculated based on PAS staining. The contents of interleukin (IL)-1β, IL-6, prostaglandin E2 (PGE2) and tumor necrosis factor α (TNF-α) were determined using enzyme-linked immunosorbent assay (ELISA) and the related mRNA expression level were measured by RT-PCR. The protein expression levels of Toll-like receptor 4 (TLR4), MyD88, p65 and p-p65 were measured using Western blotting. In addition, the 16S rRNA sequences of bacterial taxa were amplified and analyzed to assess changes in the intestinal microbes of LPS-induced mice and also in response to regulation by ASTF. Following intervention with ASTF, different therapeutic effects were shown according to the various dosages tested, all of which resulted in improved intestinal morphology and an increased number of intestinal GCs, while the contents of IL-1β, IL-6, PGE2 and TNF-α and the related mRNA expression level were significantly reduced. The TLR4, MyD88 and p-p65/p-65 protein expression levels were also significantly reduced. In addition, 16S rRNA sequencing results show that LPS disrupts the structure of mouse gut microbes, though we observed that normal microbial status can be restored through ASTF intervention. 相似文献

This study aimed at assessing the impact of four barley forms on total tract apparent digestibility of dietary fibre in horses fed a large amount of starch in the morning meal (0.27% BW). Processed barley forms had a greater pre-caecal starch digestibility than the whole form. Based on this result, we hypothesised that using barley-processing methods would limit the potential dumping of undegraded starch in the hindgut of horses and, consequently, the potential negative effect on fibre degradation in the hindgut. In a 4×4 latin square design, four mature geldings fitted with a right ventral colon-fistula were fed a meadow hay : concentrate (62 : 38; dry matter (DM) basis) diet at 1.7% BW. The concentrate was made of 80% barley distributed either as whole grain or as processed forms: 2.5 mm ground, pelleted or steam-flaked. For each period, total tract apparent digestibilities of DM, NDF and ADF were determined over 3 consecutive days by total faecal collection, whereas pH, volatile fatty acids (VFA) concentrations and cultural functional bacteria counts (total anaerobic, cellulolytic bacteria, lactic acid producers, amylolytic bacteria and lactic acid utilisers) in colonic content were evaluated on 1 day 4 h after the morning meal. Total tract apparent digestibility of DM and dietary fibre was influenced (P<0.05) by barley form. Diets including thermo-mechanically treated barley forms led to a higher (P<0.05) total tract apparent digestibility of NDF than those constituted of ground barley and also led to a greater (P<0.05) total tract apparent digestibility of ADF than those made of whole or ground barley forms. However, no significant difference was observed in colonic pH, VFA concentrations and cultural bacteria concentrations. Owing to a high starch supply in the morning meal, the concentration of the functional bacteria in the colonic content averaged 7.8 log CFU/ml, 5.9 NPM/ml, 6.9 and 7.3 CFU/ml for total anaerobic, cellulolytic, amylolytic and lactic acid-utilising bacteria, respectively. Consequently, providing horses with pelleted or steam-flaked instead of ground barley forms may limit the negative impact of starch on fibre digestibility in horses fed a high level of starch in the morning meal (0.27% BW). Animals are often exposed to or can ingest heavy metals along with their food. Therefore, we tested whether the hindgut microbiota of Mastotermes darwiniensis possesses the capability to form methyl mercury. The termite M. darwiniensis (Isoptera) was fed with saw dust containing different concentrations of inorganic mercury. Methyl mercury was determined by purge‐and‐trap capillary gas chromatography‐atom fluorescence spectrometry (CGC‐AFS) using ethyl mercury chloride as the internal standard. Total mercury concentrations were determined in the termite tissue by inductive coupled plasma‐isotope dilution mass spectrometry (ICP‐IDMS) after microwave‐assisted digestion. The obtained results showed in vivo methyl mercury production in terrestrial insects for the first time. Desulfovibrio intestinalis isolated from M. darwiniensis was identified as a biomethylating species of the intestinal microbiota. 相似文献

Foregut fermentation occurs in mammalian ruminants and in one bird, the South American folivorous hoatzin. This bird has an enlarged crop with a function analogous to the rumen, where foregut microbes degrade the otherwise indigestible plant matter, providing energy to the host from foregut fermentation, in addition to the fermentation that occurs in their hindguts (cecum/colon). As foregut fermentation represents an evolutionary convergence between hoatzins and ruminants, our aim was to compare the community structure of foregut and hindgut bacterial communities in the cow and hoatzin to evaluate the influences of host phylogeny and organ function in shaping the gut microbiome. The approach used was to hybridize amplified bacterial ribosomal RNA genes onto a high-density microarray (PhyloChip). The results show that the microbial communities cluster primarily by functional environment (foreguts cluster separately from hindguts) and then by host. Bacterial community diversity was higher in the cow than in the hoatzin. Overall, compared with hindguts, foreguts have higher proportions of Bacteroidetes and Spirochaetes, and lower proportions of Firmicutes and Proteobacteria. The main host differences in gut bacterial composition include a higher representation of Spirochaetes, Synergistetes and Verrucomicrobia in the cow. Despite the significant differences in host phylogeny, body size, physiology and diet, the function seems to shape the microbial communities involved in fermentation. Regardless of the independent origin of foregut fermentation in birds and mammals, organ function has led to convergence of the microbial community structure in phylogenetically distant hosts. 相似文献

Feeding a diet with a small amount of hay and a high proportion of concentrate given in large meals entails a risk of colic to horses as this can impact the hindgut microbial ecosystem. To counteract this potential negative effect, one feeding strategy is to modify the distribution sequence of concentrate and hay. The purpose of the present study was to assess the impact of feeding the concentrate meal before or with the hay meal on fermentative parameters postprandial variations in caecum and right ventral colon contents and faeces. We focused on the comparison of pH value and volatile fatty acid (VFA) concentrations between the three compartments. Four horses fitted with two cannulas in the caecum and right ventral colon were fed a 100% hay diet (H) for a 3-week period, and then a 40% concentrate–60% hay diet for two consecutive periods of 3 weeks each. Concentrate and hay were offered simultaneously in two equal meals at 0800 and 1700 h (HC1) during one period, or separately in two equal meals at 0800 and 1600 h for the concentrate and at 1000 and 1730 h for hay (HC2) during the second one. During the 2nd week of each diet period (H, HC1, HC2), caecal, right ventral colonic and faecal samples were collected before (0) and at 2, 4, 6 and 8 h after the morning meal. pH values and VFA concentrations were measured. Adding concentrate to the hay was associated with a decrease of pH value in the caecum, right ventral colon and faeces and an increase of faecal VFA concentrations (P<0.01). An impact of the concentrate feeding sequence was observed on caecal pH and faecal propionate concentration (P<0.001) but none on the other parameters investigated. In the caecum and right ventral colon, pH values decreased and VFA concentrations increased within the range of 4 to 8 h after the morning meal. Our data suggested that the variation of faecal pH could be an indicator of those of the caecal and right ventral colonic pH with a delay of 2 h. The present study showed that hay and commercial pelleted could be safely distributed simultaneously, thus making easier every day horse feeding. 相似文献

The gut microbiomes of birds and other animals are increasingly being studied in ecological and evolutionary contexts. Numerous studies on birds and reptiles have made inferences about gut microbiota using cloacal sampling; however, it is not known whether the bacterial community of the cloaca provides an accurate representation of the gut microbiome. We examined the accuracy with which cloacal swabs and faecal samples measure the microbiota in three different parts of the gastrointestinal tract (ileum, caecum, and colon) using a case study on juvenile ostriches, Struthio camelus, and high‐throughput 16S rRNA sequencing. We found that faeces were significantly better than cloacal swabs in representing the bacterial community of the colon. Cloacal samples had a higher abundance of Gammaproteobacteria and fewer Clostridia relative to the gut and faecal samples. However, both faecal and cloacal samples were poor representatives of the microbial communities in the caecum and ileum. Furthermore, the accuracy of each sampling method in measuring the abundance of different bacterial taxa was highly variable: Bacteroidetes was the most highly correlated phylum between all three gut sections and both methods, whereas Actinobacteria, for example, was only strongly correlated between faecal and colon samples. Based on our results, we recommend sampling faeces, whenever possible, as this sample type provides the most accurate assessment of the colon microbiome. The fact that neither sampling technique accurately portrayed the bacterial community of the ileum nor the caecum illustrates the difficulty in noninvasively monitoring gut bacteria located further up in the gastrointestinal tract. These results have important implications for the interpretation of avian gut microbiome studies. 相似文献

A Pt and calomel electrode combination were used to determine the redox potentials of the gut contents in two termites, Zootermopsis nevadensis and Cubitermes severus. Strongly reducing conditions occurred in the paunch of Z. nevadensis (mean Eh = ?160 mV), consistent with many evidences that anaerobic fermentation of wood polymers occurs at this site. In C. severus, a soil-feeder, equivalent regions of the hindgut were more midly reducing (P1: mean Eh = ?104 mV; P3: mean Eh = ?47 mV) while the colon appeared microaerobic or aerobic. It is argued that these conditions are more appropriate to the digestion of humic materials. Potentials consistent with aerobic conditions were found throughout the guts of Periplaneta americana, Locusta migratoria and Glomeris marginata, although the cockroach hindgut was more reducing than the equivalent structures in the other non-termite species. 相似文献

Abstract In forested stream ecosystems of the north and eastern United States, larvae of the aquatic crane fly Tipula abdominalis are important shredders of leaf litter detritus. T. abdominalis larvae harbor a dense and diverse microbial community in their hindgut that may aide in the degradation of lignocellulose. In this study, the activities of cellulolytic and hemicellulolytic enzymes were demonstrated from hindgut extracts and from bacterial isolates using model sugar substrates. One of the bacterial isolates was further characterized as a member of the family Microbacteriaceae. Taxonomic position of the isolate within this family was determined by a polyphasic approach, as is commonly employed for the separation of genera within the family Microbacteriaceae. The bacterial isolate is Gram-type positive, motile, non-sporulating, and rod-shaped. The G + C content of the DNA is 64.9 mol%. The cell wall contains B2γ type peptidoglycan, D- and L-diaminobutyric acid as the diamino acid, and rhamnose as the predominant sugar. The predominant fatty acids are 12-methyltetradecanoic acid (ai-C15:0) and 14-methylhexadecanoic acid (ai-C17:0). The isolate forms a distinct lineage within the family Microbacteriaceae, as determined by 16S rRNA sequence analysis. We propose the name Crocebacterium ilecola gen. nov., sp. nov., to accommodate this bacterial isolate. The type species is T202T (ATCC BAA-1359; GenBank Accession DQ826511). 相似文献

In the cockroach, Leucophaea maderae, the hindgut is innervated by the proctodeal nerves, branches of nerves XI of the terminal abdominal ganglion.Phasic afferent activity was recorded from the 11th-proctodeal nerve in response to distention of the hindgut by saline injection. With methylene blue staining, eight multipolar sensory receptors of type II (Dethier, 1963, The Physiology of Insect Senses. Methuen, London) were identified on the ventral and lateral surfaces of the rectum, four on each side. The hindgut of Drosophila larvae consists of nine domains that have been distinguished by specific gene expression patterns. In the present study, we examined the ultrastructure of the hindgut of Drosophila larvae, with special reference to the domains, in order to determine whether or not the domains are morphologically distinct functional units. Each domain showed specific ultrastructural features that suggested specific corresponding functions. According to the morphological features, terms are proposed for each domain: the imaginal ring; the \"pylorus,\" which has a thick cuticular layer and well-developed sphincter muscles; the \"large intestine,\" which occupies a major middle portion of the hindgut and has a unique dorsal and ventral subdivision; \"border cells,\" which delineate the anterior and posterior borders of the large intestine and the border between the dorsal and ventral domains of the large intestine; and the \"rectum,\" which is situated at the posterior end of the hindgut and has a thick cuticular layer and sphincter muscles. The morphological features indicate that the large intestine has active absorptive activities. The domains, which have been distinguished by gene expressions, were demonstrated to be functional tissue units of the gut. 相似文献

Regions of the gut of near-term embryos and new-born larvae of the viviparous rockfish Sebastes taczanowskii were examined histologically and immunohistochemically in order to ascertain whether they possessed the potential to absorb intact macromolecules. Compared with the hindgut of larvae 24 h after parturition, that of near-term embryos (stage 31) was found to be markedly hypertrophied and to react positively when tested for the presence of eosinophilic materials and with antibodies raised against extracts of the rockfish yolk (ab.a-E). In addition, near-term embryos incubated in Ringer solution containing 1% bovine serum albumin appeared to accumulate the protein within the epithelium of the hindgut. Furthermore, as confirmed by immunodiffusion analysis, the ovarian fluid contained materials which reacted positively to ab.a-E. These results suggest that ingestion of intact macromolecules occurs during the late embryonic stages and that the yolk-like substances detected within the hindgut epithelium of such embryos may be derived from the bathing ovarian fluid. 相似文献

To access the fermentative response of equine caecal microbial population to nitrogen availability, an in vitro study was conducted using caecal contents provided with adequate energy sources and nitrogen as limiting nutrient. Two nitrogen (N) sources were provided, protein (casein) and non-protein (urea). Caecal fluid, taken from three cannulated horses receiving a hay–concentrate diet, was mixed with a N-free buffer–mineral solution. The influence of four N levels (3.7, 6.3, 12.5 or 25 mg of N in casein or urea) was studied using the gas production technique. Total volatile fatty acids (VFA), NH3-N and gas production were measured after a 24-h incubation period. Microbial biomass was estimated using adenine and guanine bases as internal markers, and ATP production was estimated stoichiometrically. Microbial growth efficiency (YATP) and gas efficiency (Egas) were estimated. Fermentation with casein as the sole N source was generally characterized by lower total VFA, NH3-N, total gas production and higher acetate : propionate (A : P) ratio and YATP than with urea. Results herein presented indicate that, under these in vitro conditions, caecal microbial population does in fact use urea N, but less efficiently than casein in terms of microbial growth. 相似文献

苔藓植物由于含有较高浓度的不饱和脂肪酸尤其是花生四烯酸可以提高动物的御寒能力,因此驯鹿和其他许多生活在寒冷地区的食草动物以及鸟类将苔藓作为主要的食物来源。为了进一步了解苔藓被采食的情况,本实验对生活在内蒙古大兴安岭满归敖鲁古雅民族乡的驯鹿3个月份的粪便(1999年采)进行了显微观察,发现其中苔藓植物在4月份占5.63%、6月份2.2%、9月份12.92%,3个月份的粪便中均以赤茎藓(Pleurozium schreberi)为主,占苔藓总量的70%以上,曲尾藓(Dicranum spp.)、毛叶苔(Ptilidium ciliare)和沼泽皱蒴藓(Aulacomnium palustre)也有少量食用。对驯鹿生活区域内4种林型下苔藓植物的盖度和生物量的测定结果表明,驯鹿对苔藓植物的选择食用与苔藓植物的物种和丰富度相关。 相似文献

The loss of part of the lipopolysaccharides (LPS) of the outer membrane of T. ferrooxidans negatively influenced the attachment of the bacteria to minerals and the bioleaching process. LPS previously extracted from T. ferrooxidans and which had come into contact with pyrite inhibited the attachment of cells to minerals and also negatively affected the bioleaching. These results suggest that LPS play an important role in the attachment of the microorganisms and therefore, its presence or absence could affect the bioleaching process. 相似文献

Gut cell losses contribute to overall feed efficiency due to the energy requirement for cell replenishment. Intestinal epithelial cells are sloughed into the intestinal lumen as digesta passes through the gastrointestinal tract, where cells are degraded by endonucleases. This leads to fragmented DNA being present in faeces, which may be an indicator of gut cell loss. Therefore, measuring host faecal DNA content could have potential as a non-invasive marker of gut cell loss and result in a novel technique for the assessment of how different feed ingredients impact upon gut health. Faecal calprotectin (CALP) is a marker of intestinal inflammation. This was a pilot study designed to test a methodology for extracting and quantifying DNA from pig faeces, and to assess whether any differences in host faecal DNA and CALP could be detected. An additional aim was to determine whether any differences in the above measures were related to the pig performance response to dietary yeast-enriched protein concentrate (YPC). Newly weaned (∼26.5 days of age) Large White × Landrace × Pietrain piglets (8.37 kg ±1.10, n = 180) were assigned to one of four treatment groups (nine replicates of five pigs), differing in dietary YPC content: 0% (control), 2.5%, 5% and 7.5% (w/w). Pooled faecal samples were collected on days 14 and 28 of the 36-day trial. Deoxyribonucleic acid was extracted and quantitative PCR was used to assess DNA composition. Pig genomic DNA was detected using primers specific for the pig cytochrome b (CYTB) gene, and bacterial DNA was detected using universal 16S primers. A pig CALP ELISA was used to assess gut inflammation. Dietary YPC significantly reduced feed conversion ratio (FCR) from weaning to day 14 (P<0.001), but not from day 14 to day 28 (P = 0.220). Pig faecal CYTB DNA content was significantly (P = 0.008) reduced in YPC-treated pigs, with no effect of time, whereas total faecal bacterial DNA content was unaffected by diet or time (P>0.05). Faecal CALP levels were significantly higher at day 14 compared with day 28, but there was no effect of YPC inclusion and no relationship with FCR. In conclusion, YPC reduced faecal CYTB DNA content and this correlated positively with FCR, but was unrelated to gut inflammation, suggesting that it could be a non-invasive marker of gut cell loss. However, further validation experiments by an independent method are required to verify the origin of pig faecal CYTB DNA as being from sloughed intestinal epithelial cells. 相似文献

BACKGROUND AND AIMS: Roridula plants capture insects but have no digestive enzymes. It has been hypothesized that Roridula leaves absorb nitrogen from the faeces of obligately associated, carnivorous hemipterans. But rapid movement across the leaf surfaces of most plant leaves is prevented by the presence of an impermeable cuticle. However, in carnivorous plants, cuticular gaps or pores in digestive/absorptive cells allow rapid movement across the leaf surface. Recently, it was suggested that the hemipteran-plant interaction constituted a new pathway for plant carnivory. Here, a further adaptation to this pathway is described by demonstrating how Roridula plants probably absorb hemipteran faeces rapidly through their leaf cuticles. METHODS: The dye neutral red was used to document the rapidity of foliar absorption and TEM to determine the nature of cuticular discontinuities in the leaf of Roridula. KEY RESULTS: Aqueous compounds diffuse rapidly across the cuticle of Roridula's leaves but not across the cuticles of co-occurring, non-carnivorous plant leaves. Furthermore, immature Roridula leaves were unable to absorb neutral red whereas mature leaves could. Using TEM, cuticular gaps and pores similar to those in other carnivorous plants were found in the epidermal cells of mature Roridula leaves. CONCLUSIONS: The leaf cuticle of Roridula is very thin (0-120 nm) and cell wall elements project close to the leaf surface, possibly enhancing foliar absorption. In addition to these, cuticular gaps were frequently seen and probably perform a function similar to those found in other carnivorous plants: namely the absorption of aqueous compounds. The cuticular gaps of Roridula are probably an adaptation to plant carnivory, supporting the newly described pathway. 相似文献

ABSTRACT. Behavioural studies show that fullness of the hindgut influences the size of meals taken by fifth-instar Locusta migratoria L. nymphs, with the relevant information coming from the ileum and passing via the terminal abdominal ganglion. Morphological and electrophysiological studies suggest that stretch receptors on the posterior ileum, associated with branches of the rectal nerves, register extension of the ileal fold and probably also an increase in ileal volume. A system in which the volumetric feedbacks from both the crop and hindgut interact in the regulation of meal size is postulated. 相似文献
