The chelator-buffered nutrient solutions containing excess chelator have been used frequently in the micronutrient research, but potential toxicity of the excess chelator has not been ascertained. The present study was conducted to test effects of four concentrations of excess HEDTA [ N-(2-hydroxyethyl)ethylenedinitrilotriacetic acid] and two levels of total Zn on growth, root exudation, and nutrient uptake and transport by Triticum aestivum L. (cv. Aroona) and Triticum turgidum L. conv. durum (Desf.) MacKey (cv. Durati) genotypes differing in tolerance to Zn deficiency. Excess HEDTA at 50 μM reduced root and shoot growth and caused visual toxicity symptoms (necrotic lesions) on leaves; these effects were generally absent at lower concentrations of excess HEDTA. Root exudation of phytosiderophores increased with increasing concentrations of excess HEDTA at deficient and sufficient Zn levels, and was higher in Zn-deficiency-tolerant Aroona than in Zn-deficiency-sensitive Durati wheat. Shoot and root Zn concentrations showed a saturable response to increasing Zn2+ activities in solution. Excess HEDTA at 50 μM caused an increase in shoot concentrations of Fe and a decrease in concentrations of Mn and Cu. An average rate of Zn uptake increased with an increase in Zn2+ ionic activity in solution, with Zn-deficiency-tolerant Aroona having a higher rate of Zn uptake than Zn-deficiency-sensitive Durati in the deficiency range of Zn2+ activities. Average uptake rates of Mn and Cu decreased with an increase in concentration of excess HEDTA. Similar observations were noted for transport of Mn and Cu to shoots, while Zn transport to shoots was proportional to Zn2+ activities in solution. It was concluded that excess HEDTA at 50 μM adversely affects wheat growth and physiology, while excess of 25 μM or less does not cause measurable toxicity. The effects of increase copper concentrations in medium (10–150 μM CuSO4) on growth and viability of the roots of two-week-old soybean seedlings (Glycine max L., cv. Dorintsa) were studied. Copper excess suppressed biomass accumulation and linear plant growth; copper affected root growth much stronger than shoot growth. The presence of 10 μM CuSO4 in medium suppressed accumulation of plant biomass by 40% and the root length by 70%; in the presence of 25 μM CuSO4, these indices were equal to 80 and 90%, respectively. In the presence of 50 μM CuSO4, roots ceased to grow but biomass and shoot length still increased slightly. 150 μM CuSO4 was lethal for plants. The earliest sign of excessive copper toxicity was the accumulation of MDA, indicating activation of membrane lipid peroxidation. A significant increase in MDA content was observed at plant incubation in medium with 10 μM CuSO4 for 1 h; in this case, the content of copper in the roots increased from 36 ±1.8 (in control) to 48 ± 2.4 μg/g dry wt. The number of dead cells (permeable for the dye Evans Blue) was doubled in the presence of 200 μg/g dry wt within the root; this occurred in 72 h of growth in medium with 10 μM CuSO4, in 6 h at 25 μM CuSO4, in 3 h at 50 μM CuSO4, and 1 h at 150 μM CuSO4. Toxicity of copper excess was manifested stronger in dividing and elongation cells of the root apex (root meristem and the zone of elongation) than in more basal root regions. Copper excess resulted in the formation of breaks in the surface cell layers of the root tips and affect root morphology. When plant grew in medium with 10 μM CuSO4, a distance of lateral root formation zone from the root tip decreased markedly, and spherical swellings were formed on the tips of lateral roots. The higher copper concentrations (50 and 150 μM) suppressed completely the development of lateral roots.  相似文献

Cd accumulation in durum wheat presents a potential health risk to consumers. In an effort to understand the physiological mechanisms involved with Cd accumulation, this study examined the effects of Zn on Cd root uptake and phloem translocation in a split– root system. Durum wheat seedlings were grown in chelate-buffered nutrient solution with intact root systems divided into two sections. Each root section grew in a separate 1 l pot, one of which contained 0.2 μM CdSO4. In addition, each two-pot system contained ZnSO4 in the following combinations (in μm) (for -cd root system: +cd root system): 1:1, 1:10, 10:1,10:10, 1:19, and 19:1. Harvested plant material was analyzed for Cd and Zn. In addition, rates of Cd and Zn net uptake, translocation to the shoot, and root export (translocation from one root segment to the other) between days 8 and 22 were calculated. Results show that Zn was not translocated from one root section to its connected root section. Uptake rates of Cd increased as solution Zn concentrations increased. Cd translocation from one root section to the other decreased significantly when the Zn concentration in either pot was greater than 1 μM. These results show the potential of Zn to inhibit movement of Cd via the phloem, and suggests that providing adequate Zn levels may limit phloem loading of Cd into wheat grain. Increasing the rhizosphere activity of Zn2+ in Cd-containing soils may therefore result in reduced Cd accumulation in grain even while net Cd uptake is slightly enhanced. Copper (Cu) accumulation and tolerance mechanisms in Elsholtzia haichowensis, an indicator plant of Cu mines, were investigated under hydroponics supplied with different concentrations (0.32, 50.0, 100.0 and 200.0 μM) of Cu for 8 days. Cu at 100 and 200 μM significantly decreased the root dry weight, but had no significant effect on shoot dry weight. The plants grown in the presence of 200 μM Cu accumulated 288 and 7626 μg g−1 DW total Cu in the shoots and roots, respectively. A greater proportion of accumulated Cu was water-soluble accounting for 42–93% of the total Cu content in the shoots. The concentrations of reduced glutathione (GSH) and protein thiols were significantly enhanced under excess Cu supply. However, the concentrations of these compounds, particularly protein thiols, were much higher in the leaves than that in the roots. Three UV-absorbing peaks could be eluted out through gel filtration chromatography on Sephadex G-50. A large amount of Cu was detected in the UV-absorbing peaks in 40–50 and 70–90 ml elution fractions of the root extract, and in 40–50 and 120–140 ml elution fractions of the leaf extract. The results suggested that the adaptive Cu tolerance mechanism in E. haichowensis might involve the active participation of protein thiols which had a more important role in the leaves than in the roots.  相似文献

Understanding of the mechanisms of Mn supply from the soil and uptake by the plants can be improved by using simulation models that are based on basic principles. For this, a pot culture experiment was conducted with a sandy clay loam soil to measure Mn uptake by summer wheat (Triticum aestivum L. cv. Planet), maize (Zea mays L. cv. Pirat) and sugar beet (Beta vulgaris L. cv. Orbis) and to simulate Mn dynamics in the rhizosphere by means of a mechanistic model. Seeds of three crops were sown in pots containing 2.9 kg soil in a controlled growth chamber. Root and shoot weight, Mn content of plants, root length and root radius were determined 8 (13 days in case of sugar beet) and 20 days after germination. Soil and plant parameters were determined to run nutrient uptake model calculations. Manganese content of the shoot varied from 25 mg kg-1 for sugar beet to 34 mg kg-1 for maize. Sugar beet had the lowest root length/shoot weight ratio but the highest relative shoot growth rate, resulting in the highest shoot demand on the root. This is reflected by the Mn influx which was 0.9 × 10-7, 1.7 × 10-7 and 2.5 × 10-7 nmol cm-1 s-1 for wheat, maize and sugar beet, respectively. Nutrient uptake model calculations predicted similar influx values. Initial Mn concentration of 0.2 μM in the soil solution decreased to only 0.16 μM for wheat, 0.13 μM for maize and 0.11 μM for sugar beet at the root surface. This shows that manganese transport to the root was not a limiting step. This was confirmed by the fact that an assumed 20 times increase in maximum influx (Imax) increased the calculated Mn influx by 3.7 times. Sensitivity analysis demonstrated that for controlling Mn uptake the initial soil solution concentration (C Li), the root radius (r0), Imax and the Michaelis constant (K m) were the most sensitive factors in the listed order. Many metal transporters in plants are promiscuous, accommodating multiple divalent cations including some which are toxic to humans. Previous attempts to increase the iron (Fe) and zinc (Zn) content of rice endosperm by overexpressing different metal transporters have therefore led unintentionally to the accumulation of copper (Cu), manganese (Mn) and cadmium (Cd). Unlike other metal transporters, barley Yellow Stripe 1 (HvYS1) is specific for Fe. We investigated the mechanistic basis of this preference by constitutively expressing HvYS1 in rice under the control of the maize ubiquitin1 promoter and comparing the mobilization and loading of different metals. Plants expressing HvYS1 showed modest increases in Fe uptake, root‐to‐shoot translocation, seed accumulation and endosperm loading, but without any change in the uptake and root‐to‐shoot translocation of Zn, Mn or Cu, confirming the selective transport of Fe. The concentrations of Zn and Mn in the endosperm did not differ significantly between the wild‐type and HvYS1 lines, but the transgenic endosperm contained significantly lower concentrations of Cu. Furthermore, the transgenic lines showed a significantly reduced Cd uptake, root‐to‐shoot translocation and accumulation in the seeds. The underlying mechanism of metal uptake and translocation reflects the down‐regulation of promiscuous endogenous metal transporters revealing an internal feedback mechanism that limits seed loading with Fe. This promotes the preferential mobilization and loading of Fe, therefore displacing Cu and Cd in the seed.  相似文献

Shoot cultures of nickel hyperaccumulating Alyssum murale were established from epicotyl explants of seedlings aseptically germinated on hormone-free MS medium. They were further maintained on media with 0–0.92 μM kinetin. Optimal shoot multiplication was at 0.46 μM kinetin. Inoculation by shoot wounding was performed with overnight suspension of A. rhizogenes A4M70GUS which contains GUS gene cointegrated in pRiA4. After 30 days hairy roots were produced at the wounding site in 31 explant (25% out of 124). Hairy roots were excised and further propagated on hormone-free medium as separate clones. In the first passage clones 3 and 6 could be distinguished by fast growth and spontaneous shoot regeneration. In other clones (12, 23 and 25) shoot regeneration required presence of cytokinins. The five shoot culture clones regenerated from hairy roots were further cultured on media with 0.46 μM kinetin. These shoots were characterized by good elongation and lateral shoot branching, short internodes, minute slightly curled leaves and well developed plagiotropic root system spreading over the surface of media. Thus all plants regenerated from hairy root cultures manifested the characteristic Ri syndrome phenotype. They all had a strong positive GUS reaction. PCR analysis confirmed presence of uidA sequence from the gus construct. They were also tolerant to nickel accumulating up to 24,700 μg g−1 dry weight.  相似文献

Aluminum (Al) inhibits root growth in acidic soil, but the site of action of Al remains unclear. We investigated whether the rate of Al accumulation correlates to Al-indeced rapid root growth inhibition in rice seedlings (Oryza sativa L. cv. Youngnam). Growth of roots was significantly inhibited by 100 μM AICI3, as early as 1 h after the treatment. The inhibition of root growth was strongly dependent on Al concentration (l50 = 20 (μM) and Al-exposure time (l50 = 23 min at 25 μM Al) in a solution of 10 mM KCI and 1 mM CaCl2 buffered by 10 mM Mes/KOH (pH 4.5). Using ICPES, massive uptake of Al by roots was observed even at 15 min treatment of 25 μM Al. The kinetics of Al uptake by the roots closely corresponded to the inhibitory effects of Al on root growth. When the roots of seedlings were exposed to 50 (μM Al for 1 h, then sectioned and stained with hematoxylin, all cell types of the roots showed the presence of Al in the cytoplasm. These results indicate that Al was rapidly taken up into the root cells and thereby reduced root growth.  相似文献

Effects of exogenous gibberellic acid (GA; 10 and 100 μM) application on growth, protein and nitrogen contents, ammonium (NH4 +) content, enzymes of nitrogen assimilation and antioxidant system in pea seedlings were investigated under chromium (VI) phytotoxicity (Cr VI; 50, 100 and 250 μM). Exposure of pea seedlings to Cr and 100 μM GA resulted in decreased seed germination, fresh and dry weight and length of root and shoot, and protein and nitrogen contents compared to control. Compared to control, Cr and 100 μM GA led to the significant alteration in nitrogen assimilation in pea. These treatments decreased root and shoot nitrate reductase (NR), glutamine synthetase (GS) and glutamine 2-oxoglutarate aminotransferase (GOGAT) activities (except 50 μM Cr alone for GOGAT) while glutamate dehydrogenase (GDH) activity and NH4 + content increased. Compared to control, the root and shoot activities of superoxide dismutase (SOD) and ascorbate peroxidase (APX) increased (except APX activity at 250 μM Cr + 100 μM GA) while catalase (CAT), glutathione reductase (GR) and dehydroascorbate reductase (DHAR) activities were decreased (except GR at 100 μM GA alone) following exposure of Cr and 100 μM GA. Total ascorbate and total glutathione in root and shoot decreased by the treatments of Cr and 100 μM GA while their levels were increased by the application of 10 μM GA compared to Cr treatments alone. It has been reported that application of 10 μM GA together with Cr alleviated inhibited levels of growth, nitrogen assimilation and antioxidant system compared to Cr treatments alone. This study showed that application of 10 μM GA counteracts some of the adverse effects of Cr phytotoxicity with the increased levels of antioxidants and sustained activities of enzymes of nitrogen assimilation; however, 100 μM GA showed apparently reverse effect under Cr phytotoxicity.  相似文献

Rice seedlings were grown in hydroponic culture to determine the effects of external Zn and P supply on plant uptake of Cd in the presence or absence of iron plaque on the root surfaces. Iron plaque was induced by supplying 50 mg l−1 Fe2+ in the nutrient solution for 2 day. Then 43-day-old seedlings were exposed to 10 μmol l−1 Cd together with 10 μmol l−1 Zn or without Zn (Zn–Cd experiment), or to 10 μmol l−1 Cd with 1.0 mmol l−1 P or without P (P–Cd experiment) for another 2 day. The seedlings were then harvested and the concentrations of Fe, Zn, P and Cd in dithionite–citrate–bicarbonate (DCB) extracts and in roots and shoots were determined. The dry weights of roots and shoots of seedlings treated with 50 mg l−1 Fe were significantly lower than when no Fe was supplied. Adsorption of Cd, Zn and P on the iron plaque increased when Fe was supplied but Cd concentrations in DCB extracts were unaffected by external Zn or P supply levels. Cd concentrations in shoots and roots were lower when Fe was supplied. Zn additions decreased Cd concentrations in roots but increased Cd concentrations in shoots, whereas P additions significantly increased shoot and root Cd concentrations and this effect diminished when Fe was supplied. The percentage of Cd in DCB extracts was significantly lower than in roots or shoots, accounting for up to 1.8–3.8% of the plant total Cd, while root and shoot Cd were within the ranges 57–76% and 21–40% respectively in the two experiments. Thus, the main barrier to Cd uptake seemed to be the root tissue and the contribution of iron plaque on root surfaces to plant Cd uptake was minor. The changes in plant Cd uptake were not due to Zn or P additions altering Cd adsorption on iron plaque, but more likely because Zn or P interfered with Cd uptake by the roots and translocation to the shoots.  相似文献

The effects of copper on the growth, tolerance indices, mineral composition (N, P, K, Fe, Zn and Mn) and metal uptake of reed (Phragmites australis [Cav. Trin. ex Steudel]) and maize (Zea mays L.) were investigated in hydroponic experiments at copper concentrations ranging from 0.5 to 157 M Cu. A reduction in root length was shown to be a good indicator of copper toxicity, concentrations of 15.7 and 78.7 M Cu inhibiting root growth in maize and reed, respectively. The reed was significantly more tolerant of copper than maize and at 7.85 M Cu (external concentration), reed can be described as a Cu tolerant plant, and maize as a Cu non-tolerant species. As a result of Cu toxicity, the concentrations of macronutrients N, P and K decreased in both shoot and root of maize, while the concentrations were hardly affected in reed tissues. Fe concentration increased in shoots and roots of maize and in roots of reed with increasing Cu treatments, leading to highly significant (p<0.01) linear relationships between tissue Fe and Cu concentrations. The bioconcentration factor (BCF) of Cu was higher in roots than in shoots of both plant species, ranging from 612 to 1592 in reed for the Cu treatments tested. In the roots of maize, BCF of Cu increased from 349 to 1931 when increasing Cu in nutrient solution from 7.85 M to 78.5 M. Therefore, reed could be useful in wastewater treatments for the removal of Cu. However, the use of reed in phytoextraction of Cu from contaminated soils is limited by the low accumulation rate in shoots and although reed can be more efficient than maize for Cu phytoextraction, harvesting the full biomass, including roots, may be required.  相似文献

NADH specific glutamate dehydrogenase (GDH) activity was examined in roots and shoots of maize seedlings grown in half-strength Hoagland’s solution containing NH4NO3 as sole nitrogen source under irradiance of 60 W m−2 and temperature of 25±2°C. When 5,5′-dithio-bis (2-nitrobenzoic acid) (DTNB) was supplied to the assay mixture, it inhibited NADH-GDH activity in both roots and shoots, irrespective of whether the enzymes were extracted from light- or dark-treated roots and shoots. In each case the inhibition increased with the increase in DTNB concentration. At the maximum concentration of DTNB used (20 μM) the inhibition of shoot NADH-GDH was more pronounced than inhibition of root enzyme. This indicated differences in shoot and root NADH-GDH.  相似文献

Summary In vitro regeneration of plants from root culture of Melia azedarach seedlings was obtained. The origin and mode of development of the regenerated shoot buds were studied by means of histological analysis and scanning electron microscopy (SEM). Maximum shoot bud regeneration was achieved when root segments were cultured on Murashige and Skoog (MS) medium at quarter strength with 3% sucrose and 0.44 μM benzyladenine (BA) and kept under light (116 μmol m−2 s−1). Shoot bud elongation was achieved on MS with 0.44 μM BA, 0.46 μM kinetin (KIN), and 3.26 μM adenine sulphate (AD). Regenerated shoots were rooted on MS with 12.26 μM indole-3-butyric acid (IBA) for 4 d and subsequently in MS lacking plant growth regulators for 26 d. Plants were established in a potting substrate. Histological analysis of roots from intact seedlings (without treatment) demonstrated that during the early life of the roots, M. azedarach lacks preformed buds. In contrast, when the roots were excised and cultured in vitro, the histology and SEM observations revealed that buds originated from meristematic groups of cells, which had been formed from the pericycle and several layers beneath. These meristematic groups of cells grew towards the periphery of the cortex by crushing the outer layer of cortical cells. Further develoment led to the differentiation of leaf primordia and a shoot apical meristem.  相似文献

A simple, high-frequency and reproducible protocol for induction of adventitious shoot buds and plant regeneration from leaf-disc cultures of Jatropha curcas L. has been developed. Adventitious shoot buds were induced from very young leaf explants of in vitro germinated seedlings as well as mature field-grown plants cultured on Murashige and Skoog’s (MS) medium supplemented with thidiazuron (TDZ) (2.27 μM), 6-benzylaminopurine (BA) (2.22 μM) and indole-3-butyric acid (IBA) (0.49 μM). The presence of TDZ in the induction medium has greater influence on the induction of adventitious shoot buds, whereas BA in the absence of TDZ promoted callus induction rather than shoot buds. Induced shoot buds were multiplied and elongated into shoots following transfer to the MS medium supplemented with BA (4.44 μM), kinetin (Kn) (2.33 μM), indole-3-acetic acid (IAA) (1.43 μM), and gibberellic acid (GA3) (0.72 μM). Well-developed shoots were rooted on MS medium supplemented with IBA (0.5 μM) after 30 days. Regenerated plants after 2 months of acclimatization were successfully transferred to the field without visible morphological variation. This protocol might find use in mass production of true-to-type plants and in production of transgenic plants through Agrobacterium/biolistic-mediated transformation.  相似文献

We investigated the effect of salt stress on enzymatic activity of superoxide dismutase (SOD) isozymes in shoot and root tissues of salt tolerant and sensitive wheat (Triticum aestivum L. and Triticum durum Defs.) cultivars. Ten day old seedlings were subjected to 0.7 M NaCl stress for 3 and 5 days. Seedlings treated in the same manner without salt stress served as controls. Activity of SOD isozymes in root and shoot extracts was determined by activity staining of native polyacrylamide gels. In both shoot and root extracts of examined cultivars two isozymes of SOD, namely MnSOD and Cu/ZnSOD were identified. Cu/ZnSOD activity comprised 90 % of total SOD activity in both root and shoot tissues. Salt stress caused 1–1.5 fold increase in MnSOD activity of shoots in tolerant cultivars when compared with non-stressed controls. Under stress conditions, compared to controls all cultivars exhibited reduced MnSOD activity in root tissues. Cu/ZnSOD activity, on the other hand, was remarkably enhanced (3–4 fold) in root extracts of the tolerant cultivars, whereas it was reduced in the sensitive ones.  相似文献

High frequency plantlet regeneration was achieved in cotyledonary nodes of Aegle marmelos. Cotyledonary nodes from 1 mo. old in vitro grown seedlings of A. marmelos were cultured on Murashige and Skoog (MS) medium supplemented with benzyl adenine (BA) (0–8.8 μM), kinetin (KIN) (0–9.4 μM), and indole-3-acetic acid (IAA) (0–1.14 μM) either alone or in combinations. The highest regenerative response was observed on medium containing 6.6 μM BA + 1.14 μM IAA where approximately 86.6% of the cultures responded with an average shoot numbers of 487.5 per explant in 7-wk time. Cultures maintained on KIN-supplemented medium showed very poor response. In vitro responded shoots were transferred to root induction medium consisting of half-strength MS supplemented with auxins IAA, indole-3-butyric acid (IBA), or α-naphthalene acetic acid (NAA). Rooting was best in medium supplemented with 14.7 μM IBA. Rooted plantlets were acclimatized and transferred to the field with 80% survival rate.  相似文献

The brassinosteroid, 24-epibrassinolide, caused observable inhibition of the growth of aseptically-cultured excised tomato ( Lycopersicon esculentum Mill. cv. Best of All) roots at a concentration of 0.01 μ M . Root formation and outgrowth in tomato shoot cuttings and root biomass in intact tomato seedlings were also depressed by epibrassinolide, but only at a higher concentration (0.1μ M ). Concentrations of the steroidal estrogens, estrone and estradiol (as sulphate derivatives), in excess of 1.0 μ M , reduced cultured root growth as well as root number and total (but not mean) root length in shoot cuttings. Seedling root growth was only slightly inhibited by estradiol but not by estrone. Both epibrassinolide and estrogens caused morphological abnormalities, such as epinasty and leaf-inrolling in cuttings and seedlings. Epibrassinolide stimulated extension growth of the hypocotyl and epicotyl in cuttings and of the hypocotyl in seedlings, but estrogens exerted no effects on shoot growth.  相似文献

We investigated whether low-pH-induced manganese (Mn) deficiency causes low-pH-induced root hair formation in lettuce seedlings. Both the number and length of root hairs increased in 0 μM Mn (Mn-free) at pH 6 and decreased in 3 mM Mn (excess Mn) at pH 4 compared with the values in 10 μM Mn (normal Mn). These results indicate an inhibitory effect of Mn on both root hair initiation and elongation. The time dependency of root hair induction caused by Mn deficiency corresponded to that caused by low pH. Within 1 h after the pH of the culture medium was reduced from pH 6 to pH 4, the Mn uptake by roots decreased to 43% of that at pH 6. These results suggest that low-pH-induced Mn deficiency promotes root hair formation. At low pH, the rate of Mn uptake was reduced in areas >2 mm from the root tip. Roots with low-pH-induced root hairs still showed low Mn uptake during 3 h of incubation at pH 6. Multiple shoots were induced by culturing nodal explants excised from 1-month-old aseptic seedlings of red pepper (Capsicum annuum L. cv. Pusa Jwala) on Murashige and Skoog (MS) medium supplemented with (0.1–10 μM) thidiazuron (TDZ). The rate of multiple shoot induction per explant was maximum (14.4 ± 0.06) on MS medium supplemented with 1.0 μM TDZ. Regenerated shoots were elongated well on growth regulator free MS medium. Adventitious roots were induced two weeks after transfer of elongated shoots to MS medium supplemented with auxins (IAA, IBA or NAA) in different concentrations. Optimum root formation frequency was obtained in medium containing 1.0 μM IBA. Ex-vitro rooting was also achieved by pulse treatment with 300 μM IBA for 10 min. Rooted shoots were transplanted in plastic pots containing garden soil (with 90 % survival rate), where they grew well and attained maturity. Regenerated plants were phenotypically and cytologically normal.  相似文献
