Summary The peripolar cell is a unique cell type in the mammalian glomerulus. Peripolar cells are said to be identifiable during light microscopy by their cytoplasmic granules and by their position at the vascular pole; and during scanning electron microscopy by their distinctive surface morphology. We used both techniques to count peripolar cells in 6 normal rat kidneys. Scanning microscopy revealed that 55(±5)% of glomeruli contained at least one peripolar cell whereas light microscopy revealed granulated peripolar cells in only 4(±2)% of glomeruli. Vascular poles which contained peripolar cells previously identified by scanning were then examined by light and by transmission electron microscopy. Serial sections through these peripolar cells demonstrated the absence of cytoplasmic granules. Our observations suggest that the majority of peripolar cells in the rat contain no granules.  相似文献

A case of pulmonary carcinoma with dual (glandular and endocrine) differentiation diagnosed by fine needle aspiration cytology is reported. By light microscopy, the neoplastic cells showed mucicarmine, periodic-acid Schiff, and Grimelius silver positivity. On immunostaining, neuron specific enolase, chromogranin, calcitonin and serotonin were demonstrated. Electron microscopy revealed two different types of distinctive cytoplasmic inclusions in the same cell, namely: carcinoid type of neurosecretory granules and mucin droplets. Diagnostic criteria and possible histogenesis of this rare tumour entity are discussed.  相似文献

Pollen of ten species of Panax and six species of Aralia was examined in light microscopy and scanning and transmission electron microscopy. Grains of both genera have similar complex apertures, short columellae, and overlapping tectal sculptures, suggesting a close relationship. Most species of Panax have pollen characterized by striato-reticulate tecta, short columellae, thick foot layers, costa ectocolpi, and lalongate endoapertures. The eastern North American P. trifolius, commonly known as the dwarf ginseng, has a distinctive pollen morphology and exine structure, supporting the hypothesis of its phylogenetically isolated position. Pollen of the eastern Asian P. ginseng (ginseng) can be distinguished from the eastern North American P. quinquefolius (American ginseng) by differences in ultrastructure. The monophyly of the three medicinally important species, P. ginseng, P. notoginseng, and P. quinquefolius, suggested by triterpenoid data, is not supported by pollen data. The results of the pollen study are generally congruent with those from the sequences of nuclear ribosomal DNA.  相似文献

The development of the follicles was divided into a series of 12 distinctive stages based on various morphological criteria. Specific binding of 3H-ouabain and ruthenium red (RR) to membranes of T-tubules in crayfish muscles was used to identify the subfraction containing vesicles originating from the T-system. The microsomal fraction was prepared by differential centrifugation, and subfractions were separated in continuous or discontinuous sucrose density gradients. 3H-ouabain binding was estimated by scintillation counting; RR binding was examined by electron microscopy. The light subfraction was identified using both methods as that containing vesicles of T-tubules. Protein separation by SDS-electrophoresis revealed marked differences between the subfraction containing vesicles of T-tubules and other subfractions, the most distinctive feature being the presence of a protein of Mr 46,000 predominantly in the light subfraction.  相似文献

The most distinctive feature of the human pathogenic fungus is a polysaccharide capsule that is essential for virulence and is composed primarily of glucuronoxylomannan (GXM) and galactoxylomannan (GalXM). GXM mediates multiple deleterious effects on host immune function, yet relatively little is known about its physical properties. The average mass of Cryptococcus neoformans GXM from four antigenically different strains ranged from 1.7 to 7 x 10(6) daltons as calculated from Zimm plots of light-scattering data. GalXM was significantly smaller than GXM, with an average mass of 1 x 10(5) daltons. These molecular masses imply that GalXM is the most numerous polysaccharide in the capsule on a molar basis. The radius of gyration of the capsular polysaccharides ranged between 68 and 208 nm. Viscosity measurements suggest that neither polysaccharide altered fluid dynamics during infection since GXM behaved in solution as a polyelectrolyte and GalXM did not increase solution viscosity. Immunoblot analysis indicated heterogeneity within GXM. In agreement with this, scanning transmission electron microscopy of GXM preparations revealed a tangled network of two different types of molecules. Mass per length measurements from light scattering and scanning transmission electron microscopy were consistent and suggested GXM molecules self-associate. A mechanism for capsule growth is proposed based on the extracellular release and entanglement of GXM molecules.  相似文献

ABSTRACT. A new procedure is described that utilizes Percoll gradients for purifying micronuclei (MIC) and macronuclei (MAC) from Tetrahymena thermophila. Separation of MIC from MAC during certrifugation in Percoll gradients occurs as a result of their difference in size rather than density. Three kinds of tests were used to evaluate the purity of the nuclei: visualization of the nuclei by light microscopy; examination of the nuclei by electron microscopy; and Southern blots of MIC and MAC DNA probed with the 5s rRNA genes or a fragment from the MAC extrachromosomal rDNA molecule. When examined under the light microscope, the isolated MIC and MAC have much lower nuclear cross contamination levels than previous methods have reported. MIC's contaminated with less than 1 MAC in 1000 MIC and MAC's contaminated with less than 1 MIC in 500 MAC can be routinely prepared. Quantitative analyses of electron micrographs gave higher estimates of cross contamination in our purified nuclei, which may, in part, be explained by the difficulty in identifying small MIC or MAC fragments. Southern blots of MIC and MAC DNA probed with 5s rDNA confirmed the level of MAC contamination in the MIC estimated by light microscopy during purification of the nuclei. The level of nucleolar contamination in the MIC was estimated at 10% by Southern blots of MIC and MAC DNA, derived from a heterokaryon with distinctive MIC and MAC Bam HI sites, using an rDNA probe.  相似文献

ABSTRACT. Bulk maceration of Early Devonian (Lochkovian) deposits from the Welsh Borderland has yielded eight specimens of a new type of sporangium characterized by its elongate shape and distinctive spores. The specimens have been examined using scanning electron, transmission electron and light microscopy. The elongate sporangia occur isolated and are fragmented to varying degrees. They contain trilete spores that possess a proximal surface with shallow murornate ornament and a distal surface that is laevigate. The spores belong to the dispersed spore genus Scylaspora , and this is the first report of such spores in situ . Ultrastructural studies demonstrate that the spore walls are bilayered with a lamellate inner layer overlain by an essentially homogeneous outer layer. There is little or no associated extra-exosporal material. To date this is the earliest reported example of lamellate wall ultrastructure in trilete spores. Models of spore wall development are suggested in the light of evidence provided by spore wall ultrastructure. Detailed comparisons of the characters preserved in the fossils (morphological, anatomical and ultrastructural), with those in other fossil and extant plants, currently shed little light on the phylogenetic relationships of these specimens, primarily due to the paucity of comparable data. It is proposed that the plant is probably of vascular status, but in the absence of evidence for vascular tissue, it must be classified as rhyniophytoid.  相似文献

The aim of this work is to study the characteristics of the dromedary nervous lobe and determine how the seasons condition its organization. To this end, electron microscopy was performed and examined quantitatively on animals from winter and summer periods. The results show a higher number of cells in the nervous lobe in summer than in winter. The most abundant glial elements in winter are light pituicytes engulfing neurosecretory nerve fibers making neuroglial contact, and dark pituicytes containing numerous heterogeneous light bodies. In summer, the most distinctive glial cells may be pituicytes in a phagocytic state making contact with characteristic large light bodies that could represent a degenerative process of large neuropeptide storage. Granular pituicytes were also observed in contact with glial and neuronal components. However, lipid droplets, described in pituicytes of other mammals, were not observed in our samples. Quantitative analysis of neurovascular contacts revealed that the number of nerve terminals contacting the basal lamina did not differ between summer and winter, but the mean number of glial processes increased in winter. Our data provides evidence that the storage of neuropeptides is very marked in summer and that, associated with an autophagic and phagocytic phenomenon, this suggests an adaptation to anticipate any situation that would cause dehydration of the dromedary. Thus, in its tough environment, the animal remains permanently prepared to avoid any large water loss.  相似文献

An apparently healthy Norwegian lemming (Lemmus lemmus) caught in northern Finland was observed to have a whitish body 0.5 to 1.0 mm in diameter in the external layer of the cerebral cortex. By light microscopy a highly lobulated cyst of Frenkelia sp. was observed. By transmission electron microscopy lemmus) collected in the cyst wall was seen consisting of a parasitophorous vacuolar membrane, an underlying electron-dense layer and a granular layer. The membrane was only slightly convoluted. The protrusions of the cyst wall appeared round but were often not distinctive. A very thin septum divided the interior of the cyst into compartments packed with bradyzoites and maturing zoites. The bradyzoites were elongate measuring 5-8 x 1.5-2 microm. This is the first electron microscopical study of Frenkelia sp. from L. lemmus.  相似文献

针对目前关于草地螟Loxostege sticticalis L.雄成虫生殖系统结构和功能的研究相对缺乏的状况,本文利用光镜和扫描电镜系统研究了草地螟雄蛾的内外生殖器官及相关结构。草地螟雄蛾内外生殖器官集中于腹部第510腹节;内生殖器官位于510腹节;内生殖器官位于58腹节内腔中,由睾丸(testes)、贮精囊(seminal vesicle)、输精管(vas deferens)、附腺(accessory gland)和射精管(ejaculatory ducts)组成;外生殖器官为腹部第9、10腹节的特化结构,包括抱握器(harpes)、背兜(tegumen)、基腹弧(vinculum)、钩形突(uncus)、颚形突(gnathos)、阳茎囊(aedeagal caecum)和阳茎(phallus)。外生殖器中抱握器的端刺(furcella)方向为雄蛾区别于其它种类的一个重要形态学特征。该研究首次明确了草地螟雄蛾的生殖系统结构,并为锥额野螟蛾属中种间分类提供更多的科学依据。  相似文献

The dorsal surface of the tongue of the little tern, Sterna albifrons, has a distinctive anterior region for five-sixths of its length and a terminal posterior region. The anterior region observed by scanning electron microscopy is distinguished along its forward half by a median line from which median papillae protrude. The hind half of the anterior region has a median sulcus without papillae. The deciduous epithelium on both sides of the median line and sulcus bears scattered epithelial protrusions. The posterior lingual region has neither median papillae nor deciduous epithelium. So-called giant conical papillae are located in a transverse row between anterior and posterior regions. Delicate microridges adorn the surfaces of all outer epithelial cells in both regions. Examination of the dorsal lingual epithelium by light and electron microscopy provides histologic and cytologic criteria for distinguishing anterior and posterior regions. Basal cells are nearly alike throughout the dorsal epithelium. Intermediate layer cells of the anterior region contain numerous tonofibrils in electron-dense bundles composed of 10 nm tonofilaments. The outer layer is composed of electron-dense, well-keratinized cells, and electron-lucent epithelial protrusions are present on the exposed surface of the outermost cells. Median papillae are composed of typical keratinized cells, which are nearly filled with keratin filaments. Intermediate layer cells in the posterior region of the tongue are nearly filled with unbundled tonofilaments. There is only a very thin outer keratinized layer in this region.  相似文献

Three Cocconeis Ehrenberg species, epiphytic on the leaves of Posidonia oceanica (L.) Delile, were studied by light and electron microscopy. A new species, C. pseudonotata, is described which is closely related to C. notata Petit and C. diruptoides Hustedt. Gross morphology and the presence of a sigmoid raphe and sternum are the characters shared by the three species, whereas the central areas of sternum and raphe-sternum valves, and the stria and areola densities are the main distinctive characters for the species when observed by light microscopy. Scanning and transmission electron microscopic observations revealed that the fine structure of areolae must be also considered in order to identify the species. Comparison among the three species has been carried out by observations on material from our samples, as well as on the type material of C. diruptoides and cleaned samples from the Hustedt collection. The study also provides new information on the geographical distribution of these species.  相似文献

Psychodiella sergenti is a recently described specific pathogen of the sand fly Phlebotomus sergenti, the main vector of Leishmania tropica. The aim of this study was to examine the life cycle of Ps. sergenti in various developmental stages of the sand fly host. The microscopical methods used include scanning electron microscopy, transmission electron microscopy and light microscopy of native preparations and histological sections stained with periodic acid-Schiff reaction. Psychodiella sergenti oocysts were observed on the chorion of sand fly eggs. In 1st instar larvae, sporozoites were located in the ectoperitrophic space of the intestine. No intracellular stages were found. In 4th instar larvae, Ps. sergenti was mostly located in the ectoperitrophic space of the intestine of the larvae before defecation and in the intestinal lumen of the larvae after defecation. In adults, the parasite was recorded in the body cavity, where the sexual development was triggered by a bloodmeal intake. Psychodiella sergenti has several unique features. It develops sexually exclusively in sand fly females that took a bloodmeal, and its sporozoites bear a distinctive conoid (about 700 nm long), which is more than 4 times longer than conoids of the mosquito gregarines.  相似文献

The female reproductive system of the tsetse fly Glossina morsitans morsitans is analysed by scanning electron microscopy (SEM). The study focuses in particular on the choriothete, a peculiar uterine structure involved in the viviparous mode of reproduction of Glossina morsitans morsitans.Under light microscopy, the choriothete appears formed by numerous tongue-like folds projecting towards the uterine lumen and lined by a thin cuticle. SEM analysis highlights for the first time a distinctive new feature that is not visible by traditional histological methods. That is a cuticular covering of the choriothete, which shows numerous thorns in the form of crest-like structures arranged in nearly parallel lines. The role of the choriothete in pregnancy and in larval nourishment is discussed.  相似文献

Forty two surgical specimens containing myofibroblasts were studied to clarify the criteria for identifying the fibronexus, an ultrastructural feature regarded as a marker for myofibroblastic differentiation. Granulation tissue, tumour stroma, fibro-proliferative lesions (nodular fasciitis, myofibromatosis, inflammatory myofibroblastic tumour) and malignancies (myofibrosarcoma and fibrosarcoma) were studied. Comparable results were found throughout these specimens, although fibronexus junctions were better developed in reactive compared with tumoral myofibroblasts. By electron microscopy, myofibroblasts were identified by abundant rough endoplasmic reticulum, peripheral smooth-muscle myofilaments with focal densities, and fibronexus junctions. The latter were recognised as the points of convergence on the myofibroblast surfaces of intracellular myofilaments and extracellular fibronectin fibrils. The fibronectin fibrils were often co-linear with myofilaments. Also, fibronectin fibrils were dark-staining, straight and rigid-looking, and had a longitudinal filamentous substructure. A striking feature was the tendency of fibronectin fibrils to project into the surrounding extracellular space, away from the myofibroblast surface: in these respects, they differed significantly from lamina ("basement membrane"). The presence of fibronectin fibrils correlated positively with fibronectin immunostaining by light and electron microscopy. Laminin and collagen IV showed variable and weak staining in the intercellular spaces in a minority of cases and never strongly stained myofibroblast surfaces. The data emphasise that the fibronexus has a number of distinctive features permitting identification, and constitute a reference-point for pathologists wishing to use electron microscopy to refine light microscopy diagnoses of putative myofibroblastic lesions. The role of the fibronexus in the definition of the myofibroblast is discussed.  相似文献

Coloninae is a tiny and distinctive subfamily of the highly diverse family Leiodidae (round fungus and small carrion beetles). Although only one leiodid fossil is known from the Mesozoic, its taxonomic position and classification are doubtful. We describe the first definitive Coloninae fossil, Colon burmiticum Yamamoto sp. nov., from Upper Cretaceous amber of Myanmar. Our finding sheds light on the origin and early evolutionary history of this peculiar genus and its subfamily.  相似文献

Cysts of Myxobolus pendula from the gill arch of creek chub (Semotilus atromaculatus) were examined at various stages of development using light and electron microscopy. The subepithelial host connective tissue underwent dramatic changes, including degradation and remodelling of collagen and vascularisation, in response to the infection. Inflammatory cells lay in a fluid-filled space beneath the host's connective tissue and surrounded a distinctive parasite-derived matrix, composed of collagen fibril bundles embedded in cellular processes of the underlying secretory cells. This collagen matrix was resistant to degradation and invasion by leukocytes. Secretion of a matrix by M. pendula as a structural support, and a protective barrier against the host inflammatory cells is a novel observation for cyst-forming Myxosporea.  相似文献

Despite its clinical importance, the fatigue behaviour of cortical bone has not been examined as widely as its static behaviour. In the present study, specimens from the tibiae of horses have been subjected to load-controlled single step tests. The cyclic deformation behaviour was described by the development of stress-strain hysteresis parameters over the lifetime. The fatigue behaviour of bone is characterised by cyclic softening which is most distinctive towards the end of the lifetime. The microstructural damage accumulated during cyclic loading results in a loss of stiffness, asymmetrical deformation of the bone in tension and compression in cyclic creep. As shown by light and scanning electron microscopy, microcrack formation and growth is the main damage mechanism. The crack growth behaviour is strongly influenced by the microstructure, the stress components and the absolute value of the local stresses. Lower local stresses and/or compressive mean stresses lead to a dominant influence of the shear stress components with shear failure at inner interfaces. With increasing crack length, that is, higher local stress amplitudes, or tensile mean stresses, the microstructure is more and more ignored and failure occurs primarily under the influence of the normal stress components. This can be clearly seen on the fracture and specimen surfaces.  相似文献
