Based on a comparison of the 2nd derivative spectra of cytochrome P-450 LM2, cytochrome c and hemoglobin with the corresponding mixtures of aromatic amino acids and the individual aromatic amino acids a significant red shift of the maxima of the absorption bands appearing as distinct minima in the 2nd derivative spectrum have been observed in the proteins. This red shift has been shown to be caused by a lowered polarity of the immediate surroundings of the chromophores. The band positions of the aromatic amino acids in the 3 investigated hemoproteins are nearly the same and therefore do not allow to qantitate conformation dependent spectral changes of the chromophores. In agreement with X-ray crystallographic data aromatic amino acids of cytochrome c and of hemoglobin are subjected to spectral changes at the transition from the oxidized to the reduced state. These redox linked conformational changes are indicated by significant changes of the amplitudes of the aromatic acids in the 2nd derivative spectrum. A quantitative evaluation of the amplitudes revealed characteristics specific for cytochrome c and hemoglobin. Surprisingly the 2nd derivative spectrum in the median UV-region of cytochrome P-450 LM2 does not show any significant change at reduction.  相似文献

The molecular origin of the observed reduction of chloride ion sensitivity in the oxygen binding characteristics of the human embryonic hemoglobin Gower II has been probed by using site-directed mutant forms of the protein. Separate mutation of the three amino acids previously identified as candidates for the suppression of chloride sensitivity in the epsilon chain allowed us to unambiguously identify the His beta 77-->epsilon Asn as the origin of the lower sensitivity towards chloride ions in hemoglobin Gower II, which allows oxygen exchange from the mother to the late embryo under physiological conditions.  相似文献

目的:接枝淀粉包裹血红蛋白制备新型人造红细胞的代替品。方法:利用油酸接枝淀粉,在超声条件下下自组装,包裹天然牛血红蛋白,并鉴定其物理化学及生物学性能。测定包封率、红外光谱分析(FTIR)、电镜观察形态学及粒径,测定P50和Hill系数。结果:人工红细胞呈圆球形,平均粒径250nm,包封率高,具有良好的携氧、释氧能力。结论:成功制备了人工纳米红细胞,为进一步临床应用提供了基础。  相似文献

The covalent binding of chemical carcinogens and mutagens to hemoglobin has been proposed as a dose monitor for environmental exposure. The binding of chloroform to hemoglobin in rats was demonstrated to result from the formation of addition adducts to amino acids in the globin. The altered amino acids were isolated with an amino acid analyzer employing ion exchange chromatography. The covalent binding of orally-administered [14C]chloroform to rat hemoglobin reached a peak within 10 h. Afterwards, the amount of chloroform bound decreased slowly with half remaining at 7 weeks. The extent of chloroform binding was linearly dependent upon dose between 0.1 and 100 μmol/kg. Above 100 μmol/kg chloroform the binding to hemoglobin increased at a reduced rate. The extent of [14C]chloroform binding resulting from either 10 daily doses of 0.1 μmol/kg or a single 1.0 μmol/kg was similar. The binding to hemoglobin in three strains of mice and rats varied from 85 ± 7 μmol/g Hb for Swiss (CFN) mice to 152 ± 14 for Fisher-344 rats. We have demonstrated that the binding of chloroform to hemoglobin appears to have the following attributes of a dose monitor: (1) easily accessible from laboratory animals (and humans); (2) dose dependent; (3) stability, so that exposure can be determined weeks after it has ceased; (4) integration of low exposures which individually would be undetectable.  相似文献

The influence of single amino acid substitutions on the stability of alpha- and beta-chains of human hemoglobin was investigated by the computer method. The method was based on characteristics of protein tertiary structure and physico-chemical properties of amino acids. Frequencies of unstable mutations in the total mutational spectra of alpha- and beta-subunits of human hemoglobin were analysed: instability was produced by 26% of mutations in the alpha-subunit and by 32% in the beta-subunit. These results support the idea that certain limitations exist for stability changes produced by amino acid substitutions.  相似文献

We determined the complete amino acid sequences of the Erabu sea snake (Laticaudia semifasciata) hemoglobin by analyzing the intact globin chains, enzymatically digested fragments, and chemical cleavage fragments to clarify the molecular evolution and phylogenetic classification of the sea snake. The Erabu sea snake has two types of hemoglobin components, Hb-I and Hb-II, which contain different α- and β-chains. This is the second report of the complete primary structure for hemoglobin of snakes. The sequences were compared with those of other reptilian hemoglobins. Amino acids at positions critical for the structure and physiological functions of hemoglobin were loosely conserved. The requirements for binding of ATP and of diphosphoglycerate as allosteric effectors of β-globins seemed to be fulfilled.  相似文献

Carboxypeptidase activity (hydrolysis of N-carbobenzoxy-l-phenylalanyl-l-alanine) is high in a number of temperate zone cereals, originating in Asia Minor (wheat, barley, oats, wild oats, rye, triticale) compared to other cereals originating in central America or Asia (maize, sorghum, rice). However, endopeptidase activity (hydrolysis of azocasein or hemoglobin) is relatively much higher in the latter group. Comparison of trichloroacetic acid (TCA)-soluble products derived from the hydrolysis of hemoglobin showed that carboxyterminal amino acids (histidine, arginine, and tyrosine), are released when extracts from wheat and barley endosperms are used. With extracts from corn endosperms, much more TCA-soluble ultraviolet- absorbing material is released, but very little is released as free amino acids within the first 2 hours and the expected C-terminal amino acids of hemoglobin are not detected in significant amounts. These results suggest that the method of hydrolysis of the storage proteins may be significantly different in these two classes of cereals.  相似文献

A procedure to separate the α and β globin chains of rabbit hemoglobin, denatured with sodium dodecyl sulfate in the presence of mercaptoethanol, on a column of polyacrylamide gel was developed. The identity of the two separated chains was verified by (a) differences in distribution of radioactivity between the chains when the hemoglobin samples were labeled uniformly with various 3H- or 14C-labeled amino acids; (b) the analysis of the chain distribution of radioactivity in purified hemoglobin isolated from rabbit reticulocytes, pulse-labeled with [3H] leucine; and (c) the separation pattern of a mixture of authentic [α-3H]- and [β-14C]-labeled globin chains. The globin chains of human hemoglobin A also could be separated in a similar manner. This procedure is particularly useful when only microgram quantities of hemoglobin are available for study.  相似文献

Soybean lipoxygenase-1 was irreversibly inactivated by various peroxy acids containing a cis,cis-1,4-pentadiene group. Among these compounds, 15(S)-hydroperoxyeicosatetraenoic acid (15(S)-HPETE)2 was found to be the most effective in the inactivation of lipoxygenase. Although the prior exposure of 15(S)-HPETE to hemoglobin abolished the inhibitory effect of 15(S)-HPETE, the simultaneous inclusion of hemoglobin potentiated the inactivation of lipoxygenase by 15(S)-HPETE alone. Interestingly, the potentiating effect of hemoglobin was observed only in the incubations with peroxy acids possessing the cis,cis-1,4-pentadiene. In either the presence or the absence of hemoglobin, it was commonly observed that the enzyme inactivation, which was maximal at pH 10, was significantly protected by tocopherol, but neither by mannitol nor ethanol, and that the inclusion of arachidonic acid or linoleic acid prevented the enzyme inactivation. Based on these results, it is suggested that the selective inactivation of lipoxygenase by these peroxy acids may be due to unstable intermediates produced from hydroperoxy acids bound to the active site of lipoxygenase.  相似文献

The sequence analysis of llama (Lama glama, Camelidae) hemoglobin is described. The chains were separated, cleaved by trypsin as previously described, quantitatively characterized and sequenced in the sequenator. The llama hemoglobin differs from the human hemoglobin in that it has 25 different amino acids in the alpha chain and 24 different amino acids in the beta chain. The interaction between protein and phosphate is discussed. The earlier finding that the O2 affinity of the llama hemoglobin is dependent on its content of 2, 3-bisphosphoglycerate is interpreted here as a mutation of the 2, 3-bisphosphoglycerate contact position beta2 His in human hemoglobin to beta2 Asn in llama hemoglobin, whereby one of the four 2, 3-bisphosphoglycerate contact points is interrupted. This interruption gives rise to a diminished reduction of intrinsic oxygen affinity in the hemoglobin molecule and explains, on a molecular basis, the increased oxygen affinity of the llama hemoglobin, and consequently, the high-altitude respiration of the llama. By analogy, the increased O2 affinity of human fetal hemoglobin is interpreted according to previous physiological investigations on blood and fetal hemoglobin by the inactivation of the phosphoglycerate contact point beta143 His in the adult hemoglobin by mutation to gamma 143 Ser in the fetal hemoglobin. With respect to respiration in horses (2, 3-bisphosphoglycerate contact beta2 Gln), measurement of atomic parameters show that the amido group of the glutamine is situated close enough to the 2, 3-bisphosphoglycerate oxygen to build a hydrogen bond with the phosphate. Consequently, the explanation of the low-altitude respiration of the horse lies in the fact that glutamine and histidine fulfill sterochemically an identical function.  相似文献

A strong interaction between cytochrome b5 and hemoglobin has been demonstrated by titration curves in isoelectric focusing — electrophoresis. The pH of maximum interaction is in the pH range 8.0–8.3, which suggests a predominant role of Lys of met hemoglobin in the binding to acidic amino acids of cytochrome b5. The stoichiometry of the complex appears to be 1:1 (cytochrome b5: hemoglobin subunit) with similar binding affinities for α and β chains.  相似文献

Globin prepared from hemoglobin of adult tupai (Tupaia glis) was separated into alpha and beta polypeptide chains by CM-cellulose column chromatography. The S-aminoethylated alpha polypeptide chain and S-carboxymethylated beta polypeptide chain were each digested with trypsin, and the sequences of all the peptides thus obtained were established. The ordering of these tryptic peptides in the alpha and beta polypeptide chains was deduced from the homology of their primary structures with that of human adult hemoglobin. In this way the primary structures of the alpha and beta polypeptide chains of tupai hemoglobin were established; 27 amino acids in the alpha polypeptide chain and 26 in the beta chain differ from those in human adult hemoglobin.  相似文献

The effects of short chain (C1-C5) aldehydes, ketones, acids, alcohols and ethers on murine erythroleukemia (MEL) cells were examined to determine which particular chemical moieties and some of their combinations stimulated hemoglobin synthesis in these cells. The C4 series of compounds was active at lower concentrations than homologs of shorter chain lengths. Within an homologous series the potency and efficacy of the alcohol was always less than that of the acid and aldehyde compounds. Though heptanoic acid was found to be an inducer of hemoglobin synthesis in MEL cells, the 4,6-dioxoheptanoic acid analog is a potent inhibitor of hemoglobin synthesis. Analysis of porphyrin content of MEL cells incubated with the inducers 2-butanone, 2-methoxyethanol, acetone and methanol, showed that increased hemoglobin synthesis was always accompanied by the accumulation of porphyrins, most of which was protoporphyrin. These studies suggest that low molecular weight ketones, aldehydes, acids, ethers and alcohols can correct the defect in erythroid differentiation exhibited by MEL cells and they further suggest that the physiological trigger for inducing hemoglobin synthesis in these cells is less discriminating than previously recognized.  相似文献

The effects of free fatty acids on hemoglobin conversion and lipid peroxidation were studied in hemoglobin-containing liposomes (hemosomes) formed from an equimolar mixture of phosphatidylcholine (PC) and phosphatidylethanolamine (PE). It was shown that in hemosomes oxyhemoglobin is converted into hemichrome by the interaction of saturated fatty acids (arachidic, stearic, palmitic, myristic and lauric). This is accompanied by accumulation of primary and secondary products of lipid peroxidation. All fatty acids, except for lauric acid, have a stabilizing effect on lipid peroxidation in liposomes prepared from an equimolar mixture of PC and PE. The formation of lipid peroxidation products is inhibited by superoxide dismutase, D-alpha-tocopherol, D-mannitol and thiourea. The relationships between hemoglobin conversion and lipid peroxidation in hemosomes under effects of fatty acids were studied. The mechanisms of these reactions are discussed.  相似文献

Globin prepared from hemoglobin of the brown lemur (Lemur fulvus fulvus) was separated into alpha and beta chains by chromatography on a CM 52 column. The S-aminoethylated alpha and beta chains were each digested with trypsin and resulting peptides were isolated. The amino acid sequences of the tryptic peptides were established. The ordering of these peptides in the alpha and beta chains was deduced from the homology of their amino acid sequences with that of human adult hemoglobin. The primary structure of brown lemur hemoglobin thus obtained differs from that of human hemoglobin in 15 amino acids in the alpha chain and 26 in the beta chain.  相似文献

本文采用丹酰氯聚酰胺薄层分析法研究了65名3—7岁健康儿童的血浆游离氨基酸和血红蛋白浓度。结果表明:(1)赖氨酸促进蛋白质合成不仅供机体生长需要,同样也供合成血红蛋白;(2)贫血前期已有血浆游离氨基酸的变化,继而出现贫血,提示儿童时期生长旺盛,应注重补充某些氨基酸消耗和补充维生素、叶酸、铁元素等造血原料。  相似文献

The effect of chemical modification of hemoglobin with six derivatives of benzene isothiocyanate has been studied. The negatively charged reagents (isothiocyanates of benzoic and benzenesulfonic acids) markedly inhibit the interaction of hemoglobin with allosteric effectors such as H+, Cl- and organic phosphates; the affinity for heme ligands in the absence of effectors is reduced but cooperativity is maintained, making these modified hemoglobins suitable models for a possible 'blood substitute'. The only uncharged reagent tested (isothiocyanate of benzenesulfonamide) increases the oxygen affinity of hemoglobin and affects only slightly the interaction with heterotropic ligands; its potential use as an antisickling drug is under study.  相似文献

目的:研究复方α-酮酸对维持性血液透析(MHD)患者营养及脂质代谢的影响。方法:将40例MHD患者随机分为两组:(1)实验组(n=20),予常规蛋白饮食,并服用复方α-酮酸12片/d;(2)对照组(n=20),子常规蛋白饮食,不服用复方α-酮酸。分别于治疗前、治疗后3个月后测定各项血生化指标及人体测量学指标。结果:经3个月治疗后,实验组血白蛋白、前白蛋白较前明显升高(P〈0.01),总胆固醇和甘油三酯明显下降(P〈0.01),血红蛋白无明显变化(P〉0.05);对照组血红蛋白、血白蛋白、前白蛋白及血脂无明显变化;实验组及对照组治疗前后体重指数、肱三头肌皮褶厚度、上臂肌围无明显变化(P〉0.05)。结论:复方α-酮酸可有效改善维持性血液透析患者营养状况以及脂质代谢,但长期使用该疗法的疗效和安全性有待于进一步的研究。  相似文献

The burrowing brittle star Hemipholis elongata (Say) possesses hemoglobin-containing coelomocytes (RBCs) in its water vascular system. The RBCs, which circulate between the arms and body, are thought to play a role in oxygen transport. The hemoglobin of adult animals has a moderate affinity for oxygen (P(50) = 11.4 mm Hg at pH 8, 20 degrees C, measured in cellulo) and exhibits cooperativity (Hill coefficient > 1.7). The hemoglobin of juveniles has a higher affinity (P(50) = 2.3 mmHg at pH 8.0, 20 degrees C) and also exhibits cooperativity. The oxygen-binding properties of the hemoglobin are relatively insensitive to pH, temperature, and hydrogen sulfide. Adult hemoglobin is a heterogeneous mixture composed of three major fractions. The combined results of electrospray mass spectrometry and oxygen-binding experiments performed on purified fractions indicate that the native hemoglobin is in the form of homopolymers. A partial amino acid sequence (about 40 amino acids) of adult hemoglobin reveals little homology with holothurian hemoglobins.  相似文献
