We have previously shown that spermine, shortly after its ingestion, can induce the alteration of the morphology of the small intestine of suckling rats. It was proposed that this alteration is due to polyamine accumulation inside the epithelial cells. This could also be related to the fact that the intestine of the suckling rat is in an immature state. To shed light on this issue, disaccharidase and alkaline phosphatase activity assays, protein, DNA and RNA content measurements and polyamine concentration analysis were performed on the small intestine of suckling and weaned Wistar rats treated with spermine. Spermine did not induce the same intestinal alterations in weaned rats compared to suckling animals. Indeed, in sucklings, spermine administration induced a decrease of the protein, DNA, putrescine and spermidine intestinal content, suggesting a cell loss. The cell loss impaired the activity of intestinal enzymes: lactase, maltase and alkaline phosphatase. In weaned rats, the same treatment did not alter these parameters. Exogenous spermine by itself is not sufficient to induce the alterations described here and previously. The maturity degree of the small intestine could be the basis of this process.  相似文献

Severe sepsis is associated with dysfunction of the macrophage/monocyte, an important cellular effector of the innate immune system. Previous investigations suggested that probiotic components effectively enhance effector cell functions of the immune system in vivo. In this study, we produced bacteria-free, lysozyme-modified probiotic components (LzMPC) by treating the probiotic bacteria, Lactobacillus sp., with lysozyme. We showed that oral delivery of LzMPC effectively protected rats against lethality from polymicrobial sepsis induced by cecal ligation and puncture. We found that orally administrated LzMPC was engulfed by cells such as macrophages in the liver after crossing the intestinal barrier. Moreover, LzMPC-induced protection was associated with an increase in bacterial clearance in the liver. In vitro, LzMPC up-regulated the expression of cathelicidin-related antimicrobial peptide (CRAMP) in macrophages and enhanced bactericidal activity of these cells. Furthermore, we demonstrated that surgical stress or cecal ligation and puncture caused a decrease in CRAMP expression in the liver, whereas enteral administration of LzMPC restored CRAMP gene expression in these animals. Using a neutralizing Ab, we showed that protection against sepsis by LzMPC treatment required endogenous CRAMP. In addition, macrophages from LzMPC-treated rats had an enhanced capacity of cytokine production in response to LPS or LzMPC stimulation. Together, our data suggest that the protective effect of LzMPC in sepsis is related to an enhanced cathelicidin-related innate immunity in macrophages. Therefore, LzMPC, a novel probiotic product, is a potent immunomodulator for macrophages and may be beneficial for the treatment of sepsis.  相似文献

Although caloric restriction (CR) apparently has beneficial effects on the immune system, its effects on the immunological function of the intestinal mucosa are little known. The present study explored the effect of CR on the innate and adaptive intestinal immunity of mice. Balb/c mice were either fed ad libitum (control) or on alternate days fed ad libitum and fasted (caloric restriction). After 4 months, an evaluation was made of IgA levels in the ileum, the gene expression for IgA and its receptor (pIgR), as well as the expression of two antimicrobial enzymes (lysozyme and phospholipase A2) and several cytokines of the intestinal mucosa. CR increased the gene expression of lysozyme and phospholipase A2. The levels of IgA were diminished in the ileum, which apparently was a consequence of the reduced transport of IgA by pIgR. In ileum, CR increased the gene expression for most cytokines, both pro- and anti-inflammatory. Hence, CR differentially modified the expression of innate and adaptive immunity mediators in the intestine.  相似文献

溶菌酶是机体先天免疫系统中一个重要的效应分子, 参与机体多种免疫反应, 在溶菌过程中形成一个水解体系, 破坏和消除侵入体内的病原, 从而实现机体的免疫防御。从中国明对虾中克隆得到了溶菌酶基因(称为FcLyz基因), 该基因全长709 bp, 其完整的阅读框为477 bp, 编码158个氨基酸, 前18个氨基酸(-1~-18)为信号肽, 成熟肽由140个氨基酸组成(1-140aa), 其分子量为16.2 kD。经SMART分析,该基因具有1个溶菌酶1(LYZ1)结构域(19-130aa)。半定量RT-PCR分析结果表明溶菌酶虽在多种组织中有较低水平的组成性表达, 但在细菌诱导的血细胞、心脏、肝胰腺和鳃等多种组织中表达上调。将中国明对虾溶菌酶基因的成熟肽亚克隆进原核表达载体pET-30a (+)中, 转化大肠杆菌BL21(DE3), 再进行诱导表达和亲和纯化, 得到了纯化的重组溶菌酶, 并进行了抑菌活性检测。结果表明, 重组对虾溶菌酶对革兰氏阳性菌的抑菌能力较强, 最小抑菌浓度达到3.43 mmol/L, 但对革兰氏阴性菌抑制作用较小。上述结果表明, 该溶菌酶作为一种重要的免疫效应分子, 参与了对虾的免疫防御反应。  相似文献

The influence of the age of the rat on the maturation of disaccharidase activities induced by spermine was studied. Three-day- and 9-day-old rats were used in the experiment. Spermine was administered orally or directly into the stomach using thin tubing daily for 3 days, and disaccharidase activities in the jejunum were measured. While spermine caused maturation of not only lactase activity but also maltase and sucrase activities in 9-day-old rats, it only caused maturation of lactase activity in 3-day-old rats. Histological studies showed no significant changes in the jejunum of 3-day-old rats treated with spermine.  相似文献

Aloe vera polysaccharides have traditionally been used in Asian cultures as medicinal plants to enhance immunity and reduce oxidative injury. The current investigation was conducted to examine the effects of A. vera polysaccharides on various in vivo parameters of innate immunity and antioxidant enzymes activities in oral ulcer animals. Forty wistar rats were randomly divided into the following 1 control group and 3 experimental groups (each group contained 10 rats). Rats in experimental groups were orally fed by A. vera polysaccharides. Rats in control group were orally fed by same volume of saline. The results showed that A. vera polysaccharides enhanced immunity activity and exerted antioxidant effects compared with vehicle controls. These results demonstrate, for the first time, that A. vera polysaccharides are effective in enhancing innate immunity and suppressing oxidative injury in oral ulcer animals.  相似文献

The aim of this work was to establish the effect of the cis9,trans11 conjugated linoleic acid (CLA) isomer on mucosal immunity during early life in rats, a period when mucosal immunoglobulin production is poorly developed, as is also the case in humans. CLA supplementation was performed during three life periods: gestation, suckling, and early infancy. The immune status of supplemented animals was evaluated at two time points: at the end of the suckling period (21-day-old rats) and 1 week after weaning (28-day-old rats). Secretory IgA was quantified in intestinal washes from 28-day-old rats by ELISA technique. IgA, TGFbeta, and PPARgamma mRNA expression was measured in small intestine and colon by real time PCR, using Taqman specific probes and primers. IgA mucosal production was enhanced in animals supplemented with CLA during suckling and early infancy: in 28-day-old rats, IgA mRNA expression was increased in small intestine and colon by approximately 6- and 4-fold, respectively, and intestinal IgA protein by approximately 2-fold. TGFbeta gene expression was independent of age and type of tissue considered, and was not modified by dietary CLA. Gene expression of PPARgamma, a possible mediator of CLA's effects was also upregulated in animals receiving CLA during early life. In conclusion, dietary supplementation with CLA during suckling and extended to early infancy enhances development of the intestinal immune response in rats.  相似文献

The role of insulin in the regulatory mechanisms governing cessation of intestinal absorption of macromolecules and sucrase development was studied in the adrenalectomized suckling rat (ADX rat). 2. Intestinal absorption of bovine immunoglobulin (IgG) infused orally was dose-dependently suppressed to 35-75% in ADX rats repeatedly injected subcutaneously with insulin. 3. When insulin was administered orally, the IgG absorption was also suppressed. 4. Intestinal sucrase activity was also induced precociously by insulin administered subcutaneously and orally. 5. These results suggest that insulin plays a role in the maturation of suppression mechanisms for macromolecular transmission and sucrase development in the small intestine of the suckling rat.  相似文献

Apostichopus japonicus (sea cucumber) is one of the economically important farmed echinoderm species in Northern China. As a crucial enzyme in innate immunity, lysozyme plays a key role in the overall defense against pathogens in A. japonicus. In the present study, a lysozyme gene from A. japonicus was cloned by PCR and expressed in Pichia pastoris using the expression vector pPIC9K. The expressed lysozyme had a molecular mass of ~14 kD, as shown by SDS-PAGE and Western-blotting. The expression condition was optimized, and the highest expression level was achieved by induction with 1% methanol at pH 5.0 for 120 h. The recombinant lysozyme was purified by affinity chromatography using a Ni-NTA column. The specific activity of the purified lysozyme was 34,000 U/mg using Micrococcus lysodeikticus as substrates. It exhibited antimicrobial activity toward M.lysodeikticus, as detected by growth inhibition on agar plate and turbidity assay, suggesting a potential application of A. japonicus lysozyme as an antimicrobial agent in A. japonicus aquaculture.  相似文献

Chitotriosidase is a chitinolytic enzyme expressed by maturing macrophages and preformed in neutrophil granules, suggesting a role in antimicrobial defence. Although available evidence supports a role in anti-fungal immunity, there is a lack of an obvious phenotype in humans homozygous for a mutation which renders chitotriosidase inactive. This may be explained by compensatory effects of enzymes co-expressed with chitotriosidase, such as lysozyme. We have found that chitinase is highly expressed in mouse and human eye, particularly in lacrimal glands. Chitotriosidase is the only member of the chitinase/chilectin gene cluster expressed in the murine eye. As lacrimal glands also produce lysozyme, we have asked whether chitotriosidase, in addition to its documented anti-fungal effects, has synergistic anti-bacterial properties with lysozyme. The effect of recombinant chitotriosidase on the growth of five Gram-positive (Bacillus cereus, Bacillus subtilis, Listeria innocua, Staphylococcus aureus and Staphylococcus aureus OatA(+/-)) and two Gram-negative strains (Escherichia coli and Pseudomonas aeruginosa), were tested in a luminometric assay. Recombinant chitotriosidase did not inhibit bacterial growth and did not synergize with lysozyme. Though the expression of chitotriosidase in the eye supports a role in innate immunity, the antimicrobial spectrum appears to be complementary to lysozyme and may indeed be limited to fungi.  相似文献

目的:观察外源性精胺对缺氧所致的乳鼠心肌细胞凋亡的影响,并探讨其机制。方法:复制原代培养乳鼠心肌细胞缺氧损伤模型(使用pH=6.8的Hank's平衡盐溶液作为细胞培养基,排出氧气,然后在缺氧箱中培养24 h),细胞随机分为正常对照(Control)组、缺氧(Hypoxia)组和精胺干预(Hypoxia+Sp)组。Western blot检测心肌细胞多胺代谢关键酶(ODC、SSAT)蛋白质表达;CCK-8,Hoechst 33342染色观察细胞凋亡情况;光吸收法检测细胞(或培养液)内T-SOD和Caspase-3/-9活性,MDA、GSH含量;DCFH-DA染色观察细胞内活性氧(ROS)生成。结果:与正常组相比,Hypoxia组SSAT蛋白质表达、细胞凋亡率、MDA含量以及细胞内ROS生成增加,而ODC蛋白质表达、SOD活性、GSH含量降低;与Hypoxia组比较,Sp处理可减轻上述指标的变化。结论:外源性精胺可减轻缺氧引起的乳鼠心肌细胞损伤和凋亡,其机制与恢复多胺稳态和清除活性氧有关。  相似文献

Lysozyme, an antibacterial protein, has been implicated in innate immunity in invertebrates, but its activity in shrimp remained to be determined. We cloned the white shrimp lysozyme cDNA using a PCR strategy and detected its activity in haemocytes using a lytic-zone assay against Micrococcus luteus. The cloning was based on a reported EST (dbEST BE18831). The deduced amino acid sequence resulted in 150 amino with 46% identity to hen egg white lysozyme. RT-PCR was used to detect lysozyme mRNA in haemocytes. Analysis of the amino acid sequence of the shrimp lysozyme showed that it belongs to the C-type family of lysozymes. Furthermore, the lysozyme amino acid sequence contained extra residues at its C-terminus, which are characteristic of marine invertebrates. This information will be useful in future studies on the molecular mechanisms of immunity in marine invertebrates.  相似文献

原核重组表达的凡纳滨对虾(Litopenaeus vannamei)溶菌酶蛋白主要以包涵体形式存在, 经变性和复性处理后活性仍较差。研究将凡纳滨对虾溶菌酶基因(Lvlyz基因)克隆至毕赤酵母分泌型表达载体pPIC9K中, 电击转化毕赤酵母GS115细胞, 经组氨酸营养缺陷培养基筛选和PCR检测获得转化子。对其进行连续甲醇诱导表达, 利用SDS-PAGE和C端携带的6×His标签,对发酵液上清进行Western blot检测, 结果表明19.3 kD左右的条带即是重组表达的溶菌酶蛋白。用溶壁微球菌平板抑菌法鉴定表达产物具有较强的抑菌能力。研究首次利用毕赤酵母真核表达系统实现对虾溶菌酶基因的可溶性表达, 并且表达产物的活性良好。    相似文献

Acute stress can compromise acquired, and potentiate innate, immunity. Recent evidence suggests that the impact of stress on measures of immunity can be modulated by the physical activity status of the organism and that extracellular heat shock protein 72 (eHSP72) contributes to the activation of innate immunity produced by stress. Therefore, this study investigated whether physical activity status would impact the immunologically enhancing effects of stressor exposure [inescapable tail-shock stress (IS)] on innate immunity and whether changes in eHSP72 responses could play a role. Adult, male Fischer 344 rats lived with mobile (physically active) or immobile (sedentary) running wheels. After 6 wk, rats were exposed to IS or to no stress. Immediately after IS, all rats were injected subcutaneously with live Escherichia coli. Inflammation was assessed daily, and plasma eHSP72 was measured at various time points. Rats exposed to IS resolved their inflammation faster than nonstressed rats, but the beneficial impact of stress on recovery was greater in physically active rats. All rats had equal increases in circulating eHSP72 after IS. Splenocytes harvested from a separate cohort of nonstressed rats were cultured with eHSP72, and nitric oxide and cytokines were measured. Physically active rats responded to eHSP72 stimulation in vitro with a greater nitric oxide and cytokine response than sedentary rats. Thus physically active rats both recover faster than sedentary rats after bacterial challenge + IS exposure and demonstrate potentiated cellular responses to eHSP72 activation that could be important for bacterial recovery.  相似文献
