Fall armyworm, Spodoptera frugiperda (J.E. Smith) (Lepidoptera: Noctuidae), is a major economic pest throughout the Western Hemisphere. Populations can be subdivided into two morphologically identical but genetically distinct strains (corn-strain and rice-strain) that differ in their host plant preferences. These strains can be distinguished by using polymorphisms in the mitochondrial cytochrome oxidase 1 gene. Additional sequence analysis of this locus identified two sites that were highly polymorphic in the corn-strain population and that produced four different haplotype subgroups. Comparisons of the frequency distribution of these haplotypes found no seasonal or plant host specificities, but they did demonstrate that the Brazil corn-strain population is different from corn-strain fall armyworm found in Florida. The development of a rapid means of distinguishing fall armyworm populations originating from Brazil versus Florida provides an opportunity for investigating and comparing the genetic complexity and long-range movements of this important agricultural pest.  相似文献

Fall armyworm is a major economic pest throughout the Western Hemisphere. Previous studies of populations in the southern United States, Brazil, and the Caribbean demonstrated the existence of two morphologically identical but genetically distinct host strains that can only be distinguished using genetic markers, including polymorphisms in the mitochondrial Cytochrome Oxidase I (COI) gene and in the Z-chromosome linked Triose phosphate isomerase (Tpi) gene. The strains differ in some physiological and behavioral characteristics, most notably their preference for different plant hosts, but are capable of hybridizing in the laboratory and in the field. These traits suggest that the strains are in the process of divergence, which may or may not be hemispheric in scope. The objective of this study was to determine whether the two strains are present in Argentina. It was found that the strain-diagnostic haplotypes of the COI and Tpi genes subdivided the Argentina population into two major groups. Each group displayed biases in their distribution among different host plants that were generally consistent with expected strain behavior. The overall results indicated that Argentina fall armyworm exhibit similar genetics and behavior to populations in the rest of the hemisphere. In addition, the Argentina populations had comparable haplotype frequencies to those from Brazil and Texas, consistent with possible interactions with these fall armyworm groups, but appeared to have had minimal exchanges with those from Puerto Rico or Florida.  相似文献

Several restriction sites in the cytochrome oxidase I gene of fall armyworm, Spodoptera frugiperda (J.E. Smith), were identified by sequence analysis as potentially being specific to one of the two host strains. Strain specificity was demonstrated for populations in Florida, Texas, Mississippi, Georgia, and North Carolina, with an AciI and SacI site specific to the rice (Oryjza spp.)-strain and a BsmI and HinfI site joining an already characterized MspI site as diagnostic of the corn (Zea mays L.)-strain. All four of these sites can be detected by digestion of a single 568-bp polymerase chain reaction-amplified fragment, but the use of two enzymes in separate digests was found to provide accurate and rapid determination of strain identity. The effectiveness of this method was demonstrated by the analysis of almost 200 adult and larval specimens from the Mississippi delta region. The results indicated that the corn-strain is likely to be the primary strain infesting cotton (Gossypium spp.) and that an unexpected outbreak of fall armyworm on the ornamental tree Paulownia tomentosa (Thunb.) Sieb. & Zucc. ex Steud. was due almost entirely to the rice-strain.  相似文献

Microsatellite and mitochondrial DNA (mtDNA) variability data were used to study infestations of the Mediterranean fruit fly, Ceratitis capitata (Wiedemann) in Florida in 1997 and 1998. A total of 132 flies collected in monitoring traps or as larvae removed from fruit were examined at three polymorphic mtDNA restriction sites and two microsatellite loci. All of the flies sampled in Florida in 1997 displayed the mitochondrial AAB haplotype and represent a novel introduction of Mediterranean fruit flies into the state. All flies collected in central Florida in 1998 also displayed the AAB haplotype. Microsatellite analysis of these specimens from 1998 detected only alleles that were present in 1997. These results strongly indicate that the 1998 Florida outbreaks were derived from the Florida populations from the previous year. According to our analyses, the Mediterranean region is the most likely source for the 1997 Florida infestation. Flies from a small outbreak in Miami Springs, Dade County, FL, early in 1998 had a different mtDNA haplotype, characterized by the AAC restriction pattern. Microsatellites of these specimens showed significant differences in their allelic distribution from AAB flies, indicating an origin from a separate source population. South America is the most likely source for the Miami Springs flies.  相似文献

Spodoptera frugiperda (J.E. Smith) is composed of two genetically distinct strains, the so-called corn strain and the rice strain. Whether the two strains differ in their host use is unclear, because laboratory experiments have not been able to show consistent host performance or preference differences between them, and field studies showed high rates of hybridization, as well as some degree asymmetric host use. To determine the distribution of the two strains and their association with host plants, we collected fall armyworm larvae from different crops (corn, rice, alfalfa, and sorghum) and grasses in 15 different localities over 4 yr in Argentina, Brazil, and Paraguay. The strain identity was analyzed using two polymorphisms in the mitochondrial cytochrome oxidase subunit I gene. We identified the corn and rice haplotypes and three types of populations were characterized based on the frequencies of the individuals that belonged to any of these haplotypes: in 44% of populations the corn haplotype predominated, in 44% of populations the rice haplotype was the most frequent, and 11% of populations showed both haplotypes at similar proportions. In total, eight populations (47%) showed the expected pattern, two populations (12%) were polymorphic within the same field, and seven populations (41%) showed the inverse pattern. Taken together, there was no consistent pattern of host association between the two sympatric genotypes and their respective host plants. This investigation supports the need for additional studies to determine which other forces keep the genotypes separate, and what is the degree of genetic differentiation between these populations.  相似文献

草地贪夜蛾Spodoptera frugiperda J. E. Smith是一种世界性入侵害虫,为了明确入侵陕西的草地贪夜蛾的种群生物型并了解其发生及扩散规律,本研究对采集自陕西省8个地市180个草地贪夜蛾样本分别进行了基于COI和Tpi分子标记的生物型鉴定。分析发现入侵陕西的草地贪夜蛾84%是水稻型母本与玉米型父本杂交形成的杂合玉米型草地贪夜蛾。COI分子标记的结果显示,样本中水稻型占比为84.44%,玉米型为15.56%;基于Tpi基因片段的结果表明除商洛样本SL-3外,其它样本均为玉米型。值得注意的是,SL-3与非洲特异型序列同源性达100%,此非洲特异单倍型为陕西省内首次、国内第2次出现。本研究为草地贪夜蛾的迁飞扩散规律及早期预警提供了新的理论基础。  相似文献

Strips coated with conidia of Metarhizium anisopliae (Metschinkoff; Deuteromycetes: Hyphomycetes) to control the parasitic mite, Varroa destructor (Anderson and Trueman) in colonies of honey bees, Apis mellifera (Hymenoptera: Apidae) were compared against the miticide, tau-fluvalinate (Apistan) in field trials in Texas and Florida (USA). Apistan and the fungal treatments resulted in successful control of mite populations in both locations. At the end of the 42-day period of the experiment in Texas, the number of mites per bee was reduced by 69-fold in bee hives treated with Apistan and 25-fold in hives treated with the fungus; however mite infestations increased by 1.3-fold in the control bee hives. Similarly, the number of mites in sealed brood was 13-fold and 3.6-fold higher in the control bee hives than in those treated with Apistan and with the fungus, respectively. Like the miticide Apistan, the fungal treatments provided a significant reduction of mite populations at the end of the experimental period. The data from the broodless colonies treated with the fungus indicated that optimum mite control could be achieved when no brood is being produced, or when brood production is low, such as in the early spring or late fall. In established colonies in Florida, honey bee colony development did not increase under either Apistan or fungal treatments at the end of the experimental period, suggesting that other factors (queen health, food source, food availability) play some major role in the growth of bee colonies. Overall, microbial control of Varroa mites with fungal pathogens could be a useful component of an integrated pest management program for the honey bee industry.  相似文献

Genetic data were used to compare the structure of native house fly populations collected in North Dakota, Texas, and Florida. Recombination studies with mutant markers on chromosomes 3 and 4 indicated a lack of inversion polymorphism among the three populations in those areas of the genetic map studied. Significant differences were observed among flies from the three regions with regard to the frequency of 1) females that produced only male progeny, and 2) male-determining 3rd chromosomes (IIIm chromosomes). However, the North Dakota and Texas flies were more similar to each other than to the Florida flies since populations from the two former areas possessed a low frequency of both male-producing females and IIIm chromosomes; in contrast, the Florida population was void of females that produced males only and a high percentage if not all Florida males appeared to possess the IIIm male-determining mechanism. Tests for recessive lethal 3rd chromosomes showed that there was no significant difference in the frequency of lethal factors recovered from the North Dakota and Texas flies; the presence of IIIm chromosomes in Florida males precluded the recovery of lethal factors from this population by the method employed. The data suggest that house fly strains to be employed in genetic control programs should 1) originate from target control areas to avoid possible behavioral differences existing among flies from different locales, 2) be initiated with as many flies as possible to provide a background for the maintenance of variability, and 3) be renewed periodically with field-collected material since the genotype may be capable of rapid reorganization in response to laboratory selection pressures.  相似文献

Field specimens ofEuseius mesembrinus (Dean) from Florida and Texas were found to be morphologically similar regardless of differences in the lengths of dorsal shield setae or the St IV macroseta. Populations from both states were reproductively compatible. Taxonomic characters ofE. mesembrinus were illustrated using both light and scanning electron microscopy. Lengths of dorsal shield setae and the macroseta on the tarsus of leg IV of field-collected specimens from Florida and Texas, laboratory cultures, and their crosses (F1) showed inconsistent variance. Multivariate analysis of variance and canonical discriminate analysis revealed that the source of variance can be attributed to ecological factors. The fall armyworm Spodoptera frugiperda, an invasive insect pest native to the Americas, has established populations throughout eastern China. The North China Plain—a key corn-producing area in East China with a unique topography—was invaded by fall armyworm in 2019 and is seriously threatened by this migratory pest. However, the spatiotemporal extent of the migratory movements of fall armyworm from the North China Plain remains poorly understood. Using an air transport-based trajectory modeling approach that incorporates flight behavior, we simulated the potential nocturnal migration trajectories of fall armyworm from the North China Plain based on historical meteorological data from June to October of 2015–2019, and examined the night-time atmospheric conditions associated with their possible flights. The emigration patterns showed monthly variation in the main landing area and common migration direction. The displacement of newly emerged moths from the North China Plain was concentrated in the Northeast China Plain (including Liaoning, Jilin and Heilongjiang provinces) before late summer, after which they were most likely to undertake return flights to the south (especially into Hubei, Anhui and Hunan provinces). This southwest-northeast aerial migration corridor follows the topography of East China and is affected by the East Asian monsoon. These topographic-atmospheric conditions have resulted in the North China Plain becoming a key stopover for fall armyworm populations engaging in multigenerational long-distance migration across East China. These findings contribute to our knowledge of fall armyworm migration and will aid in the implementation of management and control strategies against this highly migratory agricultural pest.  相似文献

Diaprepes abbreviatus (L.) (Coleoptera: Curculionidae) is a root weevil introduced into the United States from the Caribbean in 1964. It is associated with >300 plants, including citrus, sugarcane, and potatoes. D. abbreviatus is widespread in Florida, and it has recently been detected in limited areas of California and Texas. The purpose of this research is to evaluate the utility of 16S ribosomal (16S rRNA) and cytochrome oxidase I (COI) mitochondrial markers for the delineation of genetic populations of D. abbreviatus in Florida and for the characterization of patterns of dispersion among these populations. We also assessed these markers as genetic tools for the clarification of taxonomic uncertainties in specimens from Dominica (Lesser Antilles). We analyzed 111 weevils from six Florida populations and six specimens from Dominica. In Florida, we found three haplotypes with only one haplotype in each population. Florida haplotypes differed by one to three nucleotide substitutions, possibly the result of a recent divergence from one source population or three different introductions from closely related populations from the Caribbean. In contrast, specimens from Dominica showed a high genetic variability with three 16S haplotypes and six unique COI haplotypes, delineating two mitochondrial clades. We show that these mitochondrial markers are useful for phylogeographic studies of D. abbreviatus.  相似文献

草地贪夜蛾 Spodoptera frugiperda (J. E. Smith)是联合国粮农组织全球预警的超级害虫,2019年1月入侵我国云南,并随季风远距离迁飞扩散,4月23日,广东省广州市增城区首次发现并确认该虫幼虫发生为害,对当地玉米等粮食作物构成严重威胁。本研究运用基于WRF模式的昆虫三维轨迹分析程序和GrADS气象图形软件,模拟了入侵广东的草地贪夜蛾的迁飞路径及天气背景场。结果表明:按照幼虫发育历期推算,首批草地贪夜蛾成虫迁入广东的时间可能为4月8日~13日;在此期间,越南北部及华南地区的西南低空急流可为草地贪夜蛾的迁飞提供运载气流,而风切变、降雨和下沉气流为草地贪夜蛾的迫降提供便利条件;4月9-10日存在草地贪夜蛾的有效迁飞路径,入侵广东省增城区草地贪夜蛾种群的有效虫源地分布在越南及老挝北部。本研究为华南地区草地贪夜蛾春季迁入种群的监测预警及防控提供了科学依据。  相似文献

Performance of experimental Bacillus thuringiensis (Bt) MON events alone and pyramided with MON810 were evaluated over 3 yr in Georgia and Alabama. Ability of events to prevent whorl defoliation by the fall armyworm, Spodoptera frugiperda (J. E. Smith), and natural ear feeding damage by the corn earworm, Helicoverpa zea (Boddie) was assessed. In each year, near-isogenic hybrids with novel single transformation events and crosses pyramided with the MON810 event were compared with the standard single MON810 event and nontransformed susceptible control. Events were tested for resistance to whorl damage by manual infestations of fall armyworm and ear damage by natural infestations of corn earworm. All Bt events tested reduced fall armyworm whorl damage ratings per plant compared with the susceptible hybrid. All Bt treatments also had considerably less ear infestation and damage by corn earworm compared with the nontransgenic isoline. The MON841, MION849, and MON851 events reduced ear damage by H. zea but were not as effective as other novel events and were not advanced for further testing after the 1999 season. Pyramiding events compared with single events did not improve control of fall armyworm whorl damage, but they generally did prevent more ear damage by corn earworm. The MON84006 event singly and pyramided with MON810 had superior control of whorl-stage damage by S. frugiperda and ear damage by H. zea compared with MON810. Deployment of new events and genes could provide additional tools for managing the potential for insect resistance to Bt toxins. Furthermore, improved control of whorl and ear infestations by H. zea and S. frugiperda would increase the flexibility of planting corn, Zea mays L., and permit double cropping of corn in areas where these pests perennially reach damaging levels.  相似文献

Long-distance migration of adult corn earworm moths (Helicoverpazea), and several other noctuid moth species, facilitates seasonalexpansion of pest populations and consequent increased infestationsof agricultural crops on a continental scale in North America.Long-term field studies of population dynamics and migratoryflights of H. zea and fall armyworm (Spodoptera frugiperda)in the United States were evaluated using X-band radar observationsand profiles of atmospheric conditions. These studies identifiedcharacteristic patterns of migratory flight that are largelyassociated with vertical profiles of temperature and wind speed.Collective patterns of moth migrations were generally highlycorrelated with wind headings, but often at a significant angulardeviation. Preliminary analyses are presented between moth distributionsin the aerosphere estimated from discrete moth counts usingX-band radar and bulk reflectivity data from NEXRAD Dopplerradar. Identification of associations between atmospheric factorsand noctuid population dynamics and migratory flights will improvethe ability to predict infestations by pest species throughouttheir broad seasonal range expansion.  相似文献

Laboratory-germinated seedlings from Connecticut, New Jersey, Illinois, Tennessee, Texas, and Florida were compared under four photoperiod temperature programs. Under each controlled condition, seedlings of northernmost provenances demonstrated least stem elongation and earliest dormancy. Greatest sensitivity to cool temperatures and short photoperiods was demonstrated by northernmost provenances. At warm temperatures, dormant bud formation was controlled by photoperiod in all populations tested except those from Texas and Florida. The reported differences among populations reflect the effects of natural selection upon different Liquidambar populations, insuring survival of the species in various habitats.  相似文献

Leaf-feeding damage by first generation larvae of fall armyworm, Spodopter frugiperda (J. E. Smith) (Lepidoptera: Noctuidae), and southwestern corn borer, Diatraea grandiosella Dyar (Lepidoptera: Crambidae), cause major economic losses each year in maize, Zea mays L. A previous study identified quantitative trait loci (QTL) contributing to reduced leaf-feeding damage by these insects in the maize line Mp704. This study was initiated to identify QTL and their interactions associated with first generation leaf-feeding damage by fall armyworm and southwestern corn borer. QTL associated with fall armyworm and southwestern corn borer resistance in resistant line Mp708 were identified and compared with Mp704. Multiple trait analysis (MTA) of both data sets was then used to identify the most important genetic regions affecting resistance to fall armyworm and southwestern corn borer leaf-feeding damage. Genetic models containing four and seven QTL explained southwestern corn borer and fall armyworm resistance, respectively, in Mp708. Key genomic regions on chromosomes 1, 5, 7, and 9 were identified by MTA in Mp704 and Mp708 that confer resistance to both fall armyworm and southwestern corn borer. QTL regions on chromosomes 1, 5, 7, and 9 contained resistance to both insects and were present in both resistant lines. These regions correspond with previously identified QTL related to resistance to other lepidopteran insects, suggesting that broad-spectrum resistance to leaf feeding is primarily controlled by only a few genetic regions in this germplasm.  相似文献

Aim We investigated the spatio‐temporal patterns of genetic diversity in West Indian and mainland populations of a widespread parthenogenic ant (Platythyrea punctata F. Smith) to infer source populations and subsequent colonizations across its geographic range. Location Central America, Texas and the West Indies (Florida, the Bahamas, Greater and Lesser Antilles). Methods We employed phylogeographic reconstruction based on 1451 bp of mitochondrial DNA (cytochrome c oxidase subunits I and II) sequenced from 91 individuals of P. punctata. We employed standard population genetic analyses, Bayesian phylogenetic analyses, haplotype networks and molecular dating methods as performed by beast . We also employed phylogenetic analysis using two nuclear markers (970 bp) to understand the placement of P. punctata in the globally distributed genus Platythyrea. Results Based on highly reduced haplotypic variation and temporal estimates, rapid expansion and dispersal from Central America best explains the observed distribution of haplotypes. Platythyrea punctata successfully invaded the West Indies very few times. One haplotype occurred on every island surveyed from the Bahamas and Florida in the north to Barbados at the southern edge of its range. Haplotype diversity in the West Indies is quite low, despite a larger sample size relative to the mainland. Most mainland colonies collected each possessed a unique haplotype, whereas only Florida and the larger islands (the Dominican Republic, Puerto Rico and Guadeloupe) contained more than one haplotype. Island haplotypes were most similar to haplotypes collected in northern Mexico and southern Texas, but genetic distances were nevertheless high. The putative sister species of P. punctata appears to be an endemic of Hispaniola (P. strenua Wheeler & Mann), even though older, mainland populations of P. punctata are sympatric with at least two other congenerics. Main conclusions Dispersal seems very limited on the mainland, with well‐defined clades corresponding to geographical regions. Colonization of the islands from the mainland was extremely rare, but once successful there were very few barriers to expansion to nearly every island in the West Indies. We hypothesize that this invasion occurred during the late Pleistocene as the climate became warmer and less arid.  相似文献

Cytauxzoon felis is a long-recognized hemoparasite of free-ranging Florida panthers (Puma concolor coryi), but its prevalence and effect on the population has not been assessed. Red blood cell indices and white blood cell counts were compared between infected and noninfected Florida panthers and Texas cougars (Puma concolor stanleyana) from 1983-1997 in Florida (USA). The prevalence of cytauxzoonosis for both populations was 39% (11/28) for Texas cougars, 35% for Florida panthers (22/63) and 36% overall. Thirteen hematologic parameters were compared between C. felis positive and negative panthers and cougars. Florida panthers had significantly lower mean cell hemoglobin count (MCHC) and higher white blood cell (WBC), neutrophil, monocyte and eosinophil counts (P < or = 0.05) than Texas cougars. Infected Florida panthers had significantly lower mean cell hemoglobin (MCH) and monocyte counts and higher neutrophil and eosinophil counts than infected Texas cougars. Although statistically significant differences were measured for hematologic parameters in C. felis positive panthers and cougars, biologically significant differences were not likely because values were generally within expected reference ranges for healthy animals. Cytauxzoonosis does not appear to have a negative effect on the hematologic parameters of chronically infected panthers and cougars. Potential transient changes during initial infection were not evaluated.  相似文献
