Tracking techniques are vital for the understanding of the biology and ecology of organisms. While such techniques have provided important information on the movement and migration of large animals, such as mammals and birds, scientific advances in understanding the individual behaviour and interactions of small (mm-scale) organisms have been hampered by constraints, such as the sizes of existing tracking devices, in existing tracking methods. By combining biology, chemistry and physics we here present a method that allows three-dimensional (3D) tracking of individual mm-sized aquatic organisms. The method is based on in-vivo labelling of the organisms with fluorescent nanoparticles, so-called quantum dots, and tracking of the organisms in 3D via the quantum-dot fluorescence using a synchronized multiple camera system. It allows for the efficient and simultaneous study of the behaviour of one as well as multiple individuals in large volumes of observation, thus enabling the study of behavioural interactions at the community scale. The method is non-perturbing – we demonstrate that the labelling is not affecting the behavioural response of the organisms – and is applicable over a wide range of taxa, including cladocerans as well as insects, suggesting that our methodological concept opens up for new research fields on individual behaviour of small animals. Hence, this offers opportunities to focus on important biological, ecological and behavioural questions never before possible to address.  相似文献

Thirteen years after the demonstration of quantum dots as biological imaging agents, and nine years after the initial commercial introduction of bioconjugated quantum dots, the brightness and photostability of the quantum dots has enabled a range of investigations using single molecule tracking. These materials are being routinely utilized by a number of groups to track the dynamics of single molecules in reconstituted biophysical systems and on living cells, and are especially powerful for investigations of single molecules over long timescales with short exposure times and high pointing accuracy. New approaches are emerging where the quantum dots are used as 'hard-sphere' probes for intracellular compartments. Innovations in quantum dot surface modification are poised to substantially expand the utility of these materials.  相似文献

Successful drug delivery by functionalized nanocarriers largely depends on their efficient intracellular transport which has not yet been fully understood. We developed a new tracking technique by encapsulating quantum dots into the core of wheat germ agglutinin-conjugated nanoparticles (WGA-NP) to track cellular transport of functionalized nanocarriers. The resulting nanoparticles showed no changes in particle size, zeta potential or biobinding activity, and the loaded probe presented excellent photostability and tracking ability. Taking advantage of these properties, cellular transport profiles of WGA-NP in Caco-2 cells was demonstrated. The cellular uptake begins with binding of WGA to its receptor at the cell surface. The subsequent endocytosis happened in a cytoskeleton-dependent manner and by means of clathrin and caveolae-mediated mechanisms. After endosome creating, transport occurs to both trans-Golgi and lysosome. Our study provides new evidences for quantum dots as a cellular tracking probe of nanocarriers and helps understand intracellular transport profile of lectin-functionalized nanoparticles.  相似文献

Self-illuminating quantum dot conjugates for in vivo imaging   总被引:1,自引:0,他引:1  
Fluorescent semiconductor quantum dots hold great potential for molecular imaging in vivo. However, the utility of existing quantum dots for in vivo imaging is limited because they require excitation from external illumination sources to fluoresce, which results in a strong autofluorescence background and a paucity of excitation light at nonsuperficial locations. Here we present quantum dot conjugates that luminesce by bioluminescence resonance energy transfer in the absence of external excitation. The conjugates are prepared by coupling carboxylate-presenting quantum dots to a mutant of the bioluminescent protein Renilla reniformis luciferase. We show that the conjugates emit long-wavelength (from red to near-infrared) bioluminescent light in cells and in animals, even in deep tissues, and are suitable for multiplexed in vivo imaging. Compared with existing quantum dots, self-illuminating quantum dot conjugates have greatly enhanced sensitivity in small animal imaging, with an in vivo signal-to-background ratio of > 10(3) for 5 pmol of conjugate.  相似文献

Semiconductor quantum dots show promise as alternatives to organic dyes for biological labelling because of their bright and stable photoluminescence. The typical quantum dots is CdSe because colloidal synthesis for nanocrystals of this semiconductor is well established. CdSe is usually passivated with zinc sulfide. While the cytotoxicity of bulk CdSe is well documented, questions about (CdSe)ZnS potential toxicity and behaviour in vivo remain unanswered. The distribution and stability of (CdSe)ZnS quantum dots in Wistar line rats' digestive tract were investigated. Hydrophobic quantum dots were mixed with fat or sonificated in water and administered orally. The distribution and stability of quantum dots moving through the digestive system of rats was followed by fluorescence spectroscopy. In both ways prepared quantum dots were degraded in the digestive tract of animals. Quantum dots mixed with fat were more stable and degraded more slowly than quantum dots sonificated in water. The data obtained suggest possible toxicity of (CdSe)ZnS quantum dots due to the liberation of Cd(2+).  相似文献

为构建完整的声学标记追踪体系并评估其在浅水草型湖泊的应用效果, 于2019年7月—2019年12月在河北省白洋淀生态修复示范区鲥鯸淀内,使用Vemco定位系统进行鱼类声学标记追踪实验。实验以MS-222为麻醉剂, 采用手术缝合法将声学标记植入鱼体腹腔内。对比乌鳢和鲫的术后结果, 发现: (1)鲫的死亡率和感染率均为75%, 乌鳢的死亡率和感染率为41.67%和13.33%; (2)乌鳢腹腔狭小, 在植入标记后需进行舒缓按压, 以保证探头不会压迫肠道而对生理活动造成影响。声学标记信号的接收量分析表明: (1)日间接收机接收量显著高于夜间(P<0.05); (2)方差分析结果表明晴天和雨天的接收量存在显著差异(P<0.05); (3)秩相关检验显示接收量与温度之间相关性不显著(P>0.05); (4)路径追踪结果显示乌鳢喜好活动于水草覆盖度较高的边缘区域。通过构建适宜浅水草型湖泊的声学标记追踪方法, 可促进野外环境下鱼类分布和行为生态学的定量研究, 为湖泊生态修复的效果评估提供技术手段。  相似文献

The spectroscopic properties of quantum dots can be strongly influenced by the conditions of their synthesis. In this work, we have characterized several spectroscopic properties of commercial, streptavidin functionalized quantum dots (QD525, lot 1005-0045, and QD585, lot 0905-0031, from Invitrogen). This is the first step in the development of calibration beads to be used in a generalizable quantification scheme of multiple fluorescent tags in flow cytometry or microscopy applications. We used light absorption, photoexcitation, and emission spectra, together with excited state lifetime measurements, to characterize their spectroscopic behavior, concentrating on the 400- to 500-nm wavelength ranges that are important in biological applications. Our data show an anomalous dependence of emission spectrum, lifetimes, and quantum yield (QY) on excitation wavelength that is particularly pronounced in the QD525. For QD525, QY values ranged from 0.2 at 480 nm excitation up to 0.4 at 450 nm and down again to 0.15 at 350 nm. For QD585, QY values were constant at 0.2 between 500 and 400 nm, but they dropped to 0.1 at 350 nm. We attribute the wavelength dependencies to heterogeneity in size and surface defects in the QD525, consistent with characteristics described previously in the chemistry literature. The results are discussed in the context of bridging the gap between what is currently known in the physical chemistry literature of quantum dots and the quantitative needs of assay development in biological applications.  相似文献

Quantifying and understanding movement is critical for a wide range of questions in basic and applied ecology. Movement ecology is also fostered by technological advances that allow automated tracking for a wide range of animal species. However, for aquatic macroinvertebrates, such detailed methods do not yet exist. We developed a video tracking method for two different species of benthic macroinvertebrates, the crawling isopod Asellus aquaticus and the swimming fresh water amphipod Gammarus pulex. We tested the effects of different light sources and marking techniques on their movement behavior to establish the possibilities and limitations of the experimental protocol and to ensure that the basic handling of test specimens would not bias conclusions drawn from movement path analyses. To demonstrate the versatility of our method, we studied the influence of varying population densities on different movement parameters related to resting behavior, directionality, and step lengths. We found that our method allows studying species with different modes of dispersal and under different conditions. For example, we found that gammarids spend more time moving at higher population densities, while asellids rest more under similar conditions. At the same time, in response to higher densities, gammarids mostly decreased average step lengths, whereas asellids did not. Gammarids, however, were also more sensitive to general handling and marking than asellids. Our protocol for marking and video tracking can be easily adopted for other species of aquatic macroinvertebrates or testing conditions, for example, presence or absence of food sources, shelter, or predator cues. Nevertheless, limitations with regard to the marking protocol, material, and a species’ physical build need to be considered and tested before a wider application, particularly for swimming species. Data obtained with this approach can deepen the understanding of population dynamics on larger spatial scales and of the effects of different management strategies on a species’ dispersal potential.  相似文献

Fluorescent nanocrystals, specifically quantum dots, have been a useful tool for many biomedical applications. For successful use in biological systems, quantum dots should be highly fluorescent and small/monodisperse in size. While commonly used cadmium-based quantum dots possess these qualities, they are potentially toxic due to the possible release of Cd2+ ions through nanoparticle degradation. Indium-based quantum dots, specifically InP/ZnS, have recently been explored as a viable alternative to cadmium-based quantum dots due to their relatively similar fluorescence characteristics and size. The synthesis presented here uses standard hot-injection techniques for effective nanoparticle growth; however, nanoparticle properties such as size, emission wavelength, and emission intensity can drastically change due to small changes in the reaction conditions. Therefore, reaction conditions such temperature, reaction duration, and precursor concentration should be maintained precisely to yield reproducible products. Because quantum dots are not inherently soluble in aqueous solutions, they must also undergo surface modification to impart solubility in water. In this protocol, an amphiphilic polymer is used to interact with both hydrophobic ligands on the quantum dot surface and bulk solvent water molecules. Here, a detailed protocol is provided for the synthesis of highly fluorescent InP/ZnS quantum dots that are suitable for use in biomedical applications.  相似文献

An automated technique for the identification, tracking and analysis of biological cells is presented. It is based on the use of nanoparticles, enclosed within intra-cellular vesicles, to produce clusters of discrete, point-like fluorescent, light sources within the cells. Computational analysis of these light ensembles in successive time frames of a movie sequence, using k-means clustering and particle tracking algorithms, provides robust and automated discrimination of live cells and their motion and a quantitative measure of their proliferation. This approach is a cytometric version of the moving light display technique which is widely used for analyzing the biological motion of humans and animals. We use the endocytosis of CdTe/ZnS, core-shell quantum dots to produce the light displays within an A549, epithelial, lung cancer cell line, using time-lapse imaging with frame acquisition every 5 minutes over a 40 hour time period. The nanoparticle moving light displays provide simultaneous collection of cell motility data, resolution of mitotic traversal dynamics and identification of familial relationships allowing construction of multi-parameter lineage trees.  相似文献

The Photoluminescence of quantum dots have been found to be a useful tool for the detection of small to medium sized analyte molecules in a host-guest environment. By the incorporation of quantum dots into molecularly imprinted polymers, which can offer shape and selectivity, the former can respond by quenching the photoluminescence emission upon template binding. In this work host polymers were synthesized and cased into thin films using functional monomers such as methacrylic acid (MAA), CdSe/ZnS core-shell derivatized with 4-vinyl pyridine and ethylene glycol dimethacrylic acid (EGDMA) as a cross-linker. Near-infrared fluorescent type II quantum dots for sentinel lymph node mapping   总被引:20,自引:0,他引:20  
The use of near-infrared or infrared photons is a promising approach for biomedical imaging in living tissue. This technology often requires exogenous contrast agents with combinations of hydrodynamic diameter, absorption, quantum yield and stability that are not possible with conventional organic fluorophores. Here we show that the fluorescence emission of type II quantum dots can be tuned into the near infrared while preserving absorption cross-section, and that a polydentate phosphine coating renders them soluble, disperse and stable in serum. We then demonstrate that these quantum dots allow a major cancer surgery, sentinel lymph node mapping, to be performed in large animals under complete image guidance. Injection of only 400 pmol of near-infrared quantum dots permits sentinel lymph nodes 1 cm deep to be imaged easily in real time using excitation fluence rates of only 5 mW/cm(2). Taken together, the chemical, optical and in vivo data presented in this study demonstrate the potential of near-infrared quantum dots for biomedical imaging.  相似文献

A fundamental challenge common to studies of animal movement, behavior, and ecology is the collection of high-quality datasets on spatial positions of animals as they change through space and time. Recent innovations in tracking technology have allowed researchers to collect large and highly accurate datasets on animal spatiotemporal position while vastly decreasing the time and cost of collecting such data. One technique that is of particular relevance to the study of behavioral ecology involves tracking visual tags that can be uniquely identified in separate images or movie frames. These tags can be located within images that are visually complex, making them particularly well suited for longitudinal studies of animal behavior and movement in naturalistic environments. While several software packages have been developed that use computer vision to identify visual tags, these software packages are either (a) not optimized for identification of single tags, which is generally of the most interest for biologists, or (b) suffer from licensing issues, and therefore their use in the study of animal behavior has been limited. Here, we present BEEtag, an open-source, image-based tracking system in Matlab that allows for unique identification of individual animals or anatomical markers. The primary advantages of this system are that it (a) independently identifies animals or marked points in each frame of a video, limiting error propagation, (b) performs well in images with complex backgrounds, and (c) is low-cost. To validate the use of this tracking system in animal behavior, we mark and track individual bumblebees (Bombus impatiens) and recover individual patterns of space use and activity within the nest. Finally, we discuss the advantages and limitations of this software package and its application to the study of animal movement, behavior, and ecology.  相似文献

  1. Animal movement studies are conducted to monitor ecosystem health, understand ecological dynamics, and address management and conservation questions. In marine environments, traditional sampling and monitoring methods to measure animal movement are invasive, labor intensive, costly, and limited in the number of individuals that can be feasibly tracked. Automated detection and tracking of small‐scale movements of many animals through cameras are possible but are largely untested in field conditions, hampering applications to ecological questions.
  2. Here, we aimed to test the ability of an automated object detection and object tracking pipeline to track small‐scale movement of many individuals in videos. We applied the pipeline to track fish movement in the field and characterize movement behavior. We automated the detection of a common fisheries species (yellowfin bream, Acanthopagrus australis) along a known movement passageway from underwater videos. We then tracked fish movement with three types of tracking algorithms (MOSSE, Seq‐NMS, and SiamMask) and evaluated their accuracy at characterizing movement.
  3. We successfully detected yellowfin bream in a multispecies assemblage (F1 score =91%). At least 120 of the 169 individual bream present in videos were correctly identified and tracked. The accuracies among the three tracking architectures varied, with MOSSE and SiamMask achieving an accuracy of 78% and Seq‐NMS 84%.
  4. By employing this integrated object detection and tracking pipeline, we demonstrated a noninvasive and reliable approach to studying fish behavior by tracking their movement under field conditions. These cost‐effective technologies provide a means for future studies to scale‐up the analysis of movement across many visual monitoring systems.

The Late Cretaceous track assemblage of southeastern Poland includes intriguing ichnotaxa with Asian or North American affinities. New finds of the ceratopsian-like ichnite and small hadrosauroid track refer directly to this problem, whereas the re-evaluation of foraminifer taxa estimates the stratigraphic positions of the track-bearing horizon and allowed to understand paleogeographic context of the studied and adjacent areas. The results demonstrate that during the Late Cretaceous times, migration of dinosaurs was possible from Asia through the East European Land to Kukeritz Island and the islands at peri-Tethys area.  相似文献

The migrations and winter distributions of most seabirds, particularly small pelagic species, remain poorly understood despite their potential as indicators of marine ecosystem health. Here we report the use of miniature archival light loggers (geolocators) to track the annual migration of Sabine’s Gull Larus sabini, a small (c. 200 g) Arctic‐breeding larid. We describe their migratory routes and identify previously unknown staging sites in the Atlantic Ocean, as well as their main Atlantic wintering area in the southern hemisphere. Sabine’s Gulls breeding in northeast Greenland displayed an average annual migration of almost 32 000 km (n = 6), with the longest return journey spanning close to 39 000 km (not including local movements at staging sites or within the wintering area). On their southern migration, they spent an average of 45 days in the Bay of Biscay and Iberian Sea, off the coasts of France, Spain and Portugal. They all wintered in close association with the cold waters of the Benguela Upwelling, spending an average of 152 days in that area. On their return north, Sabine’s Gulls staged off the west African coast (Morocco, Mauritania, Senegal), spending on average 19 days at this site. This leg of migration was particularly rapid, birds travelling an average of 813 km/day, assisted by the prevailing winds. Sabine’s Gulls generally followed a similar path on their outbound and return migrations, and did not exhibit the broad figure‐of‐eight pattern (anti clockwise in the southern hemisphere and clockwise in the northern hemisphere) seen in other trans‐equatorial seabirds in the Atlantic and Pacific oceans.  相似文献

Fluorescent quantum dots have great potential in cellular labeling and tracking. Here, PEG encapsulated CdSe/ZnS quantum dots have been conjugated with Tat peptide, and introduced into living mesenchymal stem cells. The Tat peptide conjugated quantum dots in mesenchymal stem cells were assessed by fluorescent microscopy, laser confocal microscope and. flow cytometry. The result shows that Tat peptide conjugated quantum dots could enter mesenchymal stem cells efficiently. The Tat-quantum dots labeled stem cells were further injected into the tail veins of NOD/SCID beta2 M null mice, and the tissue distribution of these labeled cells in nude mice were examined with fluorescence microscope. The result shows that characteristic fluorescence of quantum dots was observed primarily in the liver, the lung and the spleen, with little or no quantum dots accumulation in the brain, the heart, or the kidney.  相似文献

Through new tracking techniques, data on timing and routes of migration in long‐distance migrant birds are accumulating. However, studies of the consistency of migration of the same individuals between years are still rare in small‐sized passerine birds. This type of information is important to understand decisions and migration abilities at the individual level, but also for life history theory, for understanding carry over effects between different annual cycle stages and for conservation. We analysed individual repeatability of migration between years in great reed warblers Acrocephalus arundinaceus; a medium‐sized European songbird migrating to sub‐Saharan Africa. In seven males, with geolocator data from 2–4 yr per bird, we found low to moderate (non significant) repeatability in timing of migration parameters (R ≤ 0.41), but high (and significant) repeatability for most spatial parameters, i.e. autumn route (R = 0.64) and stopover sites (R = 0.59–0.87) in Europe, and wintering sites (R = 0.77–0.99) in sub‐Saharan Africa. This pattern of high spatial but low temporal within‐individual repeatability of migration between years contrasts other tracking studies of migrating birds that generally have found consistency in timing but flexibility in routes. High spatial consistency of migration in the great reed warbler may be due to it being a specialist in wetlands, an unevenly distributed habitat, favouring a strategy of recurrence at previously visited sites. Low temporal repeatability may be caused by large between‐year variation in carry‐over effects from the breeding season, high flexibility in decision rules during migration or high sensitivity to environmental factors (weather, wind) during migration.  相似文献

  1. Individual space and resource use are central issues in ecology and conservation. Recent technological advances such as automated tracking techniques are boosting ecological research in this field. However, the development of a robust method to track space and resource use is still challenging for at least one important ecosystem component: motile aquatic macroinvertebrates. The challenges are mostly related to the small body size and rapid movement of many macroinvertebrate species and to light scattering and wave signal interference in aquatic habitats.
  2. We developed a video tracking method designed to reliably assess space use behavior among individual aquatic macroinvertebrates under laboratory (microcosm) conditions. The approach involves the use of experimental apparatus integrating a near infrared backlight source, a Plexiglas multi‐patch maze, multiple infrared cameras, and automated video analysis. It allows detection of the position of fast‐moving (~ 3 cm/s) and translucent individuals of small size (~ 5 mm in length, ~1 mg in dry weight) on simulated resource patches distributed over an experimental microcosm (0.08 m2).
  3. To illustrate the adequacy of the proposed method, we present a case study regarding the size dependency of space use behavior in the model organism Gammarus insensibilis, focusing on individual patch selection, giving‐up times, and cumulative space used.
  4. In the case study, primary data were collected on individual body size and individual locomotory behavior, for example, mean speed, acceleration, and step length. Individual entrance and departure times were recorded for each simulated resource patch in the experimental maze. Individual giving‐up times were found to be characterized by negative size dependency, with patch departure occurring sooner in larger individuals than smaller ones, and individual cumulative space used (treated as the overall surface area of resource patches that individuals visited) was found to scale positively with body size.
  5. This approach to studying space use behavior can deepen our understanding of species coexistence, yielding insights into mechanistic models on larger spatial scales, for example, home range, with implications for ecological and evolutionary processes, as well as for the management and conservation of populations and ecosystems. Despite being specifically developed for aquatic macroinvertebrates, this method can also be applied to other small aquatic organisms such as juvenile fish and amphibians.
