Ion (K+, Na+, Mg2+, Ca2+ and Cl) flows and partitioning in thepetiole and lamina of leaf 6 of castor bean {Ricinus communisL.) plants growing in the presence of a mean of 71 mol m–3NaCl were described by an empirical modelling technique. Thiscombined data on changes in ion contents of petiole and lamina,ion: carbon molar ratios of phloem bleeding sap and pressure-inducedxylem exudates of the leaf with previously described informationon the economies of C and N in identical leaf material. Datawere expressed as daily exchanges of ions in xylem and phloem,or depicted as models of ion balance and transport activityof petiole and lamina during four consecutive phases of leaflife. The early import phase was characterized by high intakeof K and Mg through phloem, and of Ca mainly through xylem,but only limited intake of Na and Cl. The next phase up to fullleaf expansion showed similar relative differences in xylemintake between ions and the onset of rapid phloem export fromthe lamina of K and Mg, some export of Na and Cl but scarcelyany of Ca. The next mature phase, marked by maximal photosynthesisand transpiration by the leaf, showed high xylem intake of allions in xylem. This was more than matched by phloem export ofMg and K, but by only fractional re-export of Na and Cl andagain very limited cycling through the leaf of Ca. The finalpre-senescence phase exhibited similar behaviour, but with generallygreater contribution to phloem transport from mobilization ofion reserves of the lamina. The petiole retained particularlylarge amounts of Na and Cl in its early growth, thereby protectingthe lamina from excessive entry of salt, but these petiolarpools, together with those or other nutrient ions, were laterpartially mobilized to the lamina via the xylem stream. Datawere discussed in relation to the relatively high salt toleranceexhibited by the species. Calcium is believed to be transported with water in the xylem. Consistent with this proposal, low‐transpiring organs such as potato Solanum tuberosum tubers are known to suffer from calcium deficiency. Although roots on tubers and stolons have been shown to supply water to tubers, there is no direct evidence for the calcium transport pathway to tubers. Both a xylem and a phloem transport pathway have been suggested. We investigated in vivo calcium transport to developing potato, cv. Dark Red Norland and cv. Russet Burbank, tubers using 45Ca in a controlled environment facility. Whole plant split pot experiments allowed the placement of 45Ca either in the main (basal) root or the tuber and stolon areas of the pot. The results showed that 45Ca was transported to the shoot with the transpiration stream from both areas but was not re‐translocated to tubers or the main (basal) root system even 57 days after 45Ca application. Radioactivity could only be detected in the tuber when 45Ca was fed to the stolon and tuber area. When 45Ca was fed to specific tubers, radioactivity was detected in the aerial shoot; however, no activity was detected in other tubers or the main (basal) roots. In another set of experiments, roots on a stolon near a tuber were precisely fed 45Ca and Safranin O. The radioactive signal exactly overlapped the water transport pathway in the tuber marked with Safranin O dye, suggesting that water and calcium can be simultaneously transported from stolon roots to the tuber. No transport of 45Ca across the tuber periderm was detected 8 days after 45Ca was applied to the tuber periderm. This indicated that no significant transport of calcium occurs from the soil across the periderm. Our results provide evidence that: (1) calcium is not re‐translocated via the phloem from the aerial shoot tubers and main (basal) roots; (2) the main root system does not supply calcium to the tuber; (3) calcium is not transported across the periderm to the interior tuber tissue; (4) calcium is transported to the tuber via the xylem along with water, and the roots on the stolon associated with the tuber supply water and calcium to the developing tuber; and (5) transpirational demand is a significant determinant of calcium distribution within the plant.  相似文献

Experiments were conducted to test whether the facultatively phytophagous predator Orius insidiosus (Say) (Heteroptera: Anthocoridae) ingested phloem, xylem or mesophyll contents from soybean plants (Glycine max L.). Potential uptake of phloem sap was examined by radiolabeling photosynthate with 14CO2 and then measuring the accumulation of radiolabeled metabolites in feeding animals. Most O. insidiosus feeding on radiolabeled plants ingested no or very low levels of label; only 3% ingested small amounts of label, indicating the experimental insects fed very little, if at all, on the phloem. In contrast, well known phloem feeding insects used as positive controls accumulated substantial levels of labeled metabolites after feeding on known host plants. O. insidiosus did feed on xylem contents, as shown by ingestion of safranin-labeled xylem fluid. A few of the insects showed signs of feeding on the mesophyll, as indicated by the presence of chloroplasts in the gut. However, the small diameter of the food canal may cause limited passage of chloroplasts, which would contribute to an underestimation of the frequency of mesophyll feeding. Some radiolabeled metabolites remain in the mesophyll so those insects that ingested low levels of radiolabel probably ingested label from the mesophyll, which supports the notion that some level of mesophyll feeding occurred. Feeding site determines the nutrients ingested during phytophagy. These insects obtain water from the xylem, and may ingest small amounts of starches, sugars, and amino acids from the mesophyll. The results suggest that facultative phytophagy by this heteropteran predator primarily provides the insect with water, but also may provide some nutrients that supplement a prey diet and help the predator survive periods when prey are scarce.  相似文献

Indoleacetic acid (IAA)-oxidase from both secondary phloem and xylem was dependent on 2,4-dichlorophenol for activity, and was enhanced by addition of Mn2+. The pH optimum was 6.0 from both tissues. IAA-oxidase and its inhibitors were distributed differently in the secondary phloem and secondary xylem of carrot root. In the phloem a high IAA-oxidase activity was distributed uniformly along the radius but in the xylem a somewhat lower concentration decreased from the cambium. IAA-oxidase inhibitor in the phloem increased exponentially from a very low concentration near the cambium, whereas in the xylem an appreciable concentration was present near the cambium, decreasing linearly with distance from the cambium. Longitudinal gradients in the xylem parallel studies by other workers with the greatest IAA-destroying capacity present in older tissues. In the xylem inhibitor decreased and IAA-oxidase increased from the root apex. In the phloem IAA-oxidase was uniform, whereas the inhibitor increased in older tissue.

The IAA-oxidase inhibitors in phloem and xylem may be different. In the xylem the IAA-oxidase inhibitor may be a lignin precursor present in young cells which disappears as lignification proceeds. In the phloem IAA-oxidase reacting with endogenous IAA appears to form a physiologically active product.

Summary Homogenates of differentiating xylem and phloem tissue have higher cellulase activities than cambial samples; the highest activity is always found in phloem. Callus tissue, in which no vascular differentiation occurs, contains only low cellulase activity. The results suggest that cellulase is involved in vascular differentiation. Different pH optima of cellulase activity were found: in cambium, xylem and phloem tissue, cellulase activity with an optimum at about pH 5.9 is predominantly membrane-bound; it is sedimentable at 100,000 g and releasable by Triton X-100. The same may be true of activity with an optimum at pH 5.3. Phloem tissue also contains a soluble, cytoplasmic cellulase of high activity at pH 7.1, and xylem tissue contains cytoplasmic cellulase with an optimum at pH 6.5. Low cellulase activity with a pH optimum similar to that of xylem homogenates was found in xylem sap. Cellulase activity in abscission zones increases greatly just before leaf abscission. Abscission zone cellulase has two pH optima, et 5.3 and 5.9; both activities are increased by Triton treatment of homogenates. The possible existence of several different cellulases forming part of a cellulase complex, and the rôle of the enzymes in hydrolysing wall material during cell differentiation are discussed.  相似文献

Root-synthesized cytokinins are transported to the shoot and regulate the growth, development, and stress responses of aerial tissues. Previous studies have demonstrated that Arabidopsis (Arabidopsis thaliana) ATP binding cassette (ABC) transporter G family member 14 (AtABCG14) participates in xylem loading of root-synthesized cytokinins. However, the mechanism by which these root-derived cytokinins are distributed in the shoot remains unclear. Here, we revealed that AtABCG14-mediated phloem unloading through the apoplastic pathway is required for the appropriate shoot distribution of root-synthesized cytokinins in Arabidopsis. Wild-type rootstocks grafted to atabcg14 scions successfully restored trans-zeatin xylem loading. However, only low levels of root-synthesized cytokinins and induced shoot signaling were rescued. Reciprocal grafting and tissue-specific genetic complementation demonstrated that AtABCG14 disruption in the shoot considerably increased the retention of root-synthesized cytokinins in the phloem and substantially impaired their distribution in the leaf apoplast. The translocation of root-synthesized cytokinins from the xylem to the phloem and the subsequent unloading from the phloem is required for the shoot distribution and long-distance shootward transport of root-synthesized cytokinins. This study revealed a mechanism by which the phloem regulates systemic signaling of xylem-mediated transport of root-synthesized cytokinins from the root to the shoot.

Phloem unloading via the apoplastic pathway is essential for shoot distribution and long-distance translocation of root-synthesized cytokinins from the root to the shoot through the xylem.  相似文献

When indole-3-acetic acid (IAA) is applied to woody shoots cambialdivision is stimulated and the cambial derivatives differentiateto produce xylem tissue. When gibberellic acid (GA) is applied,cambial division occurs but the resultant derivatives on thexylem side of the cambium remain undifferentiated. The relativelevels of applied IAA and GA are important in determining whethermainly xylem or phloem tissue is produced. High IAA/low GA concentrationsfavour xylem formation, whereas low IAA/high GA concentrationsfavour phloem production. The new phloem tissue produced asa result of hormone treatment is fully differentiated, containingsieve elements and sieve plates. IAA is important in promotingthe elongation of the cambial derivatives to produce xylem vesseland fibre elements, though in the case of xylem fibres appliedGA causes further elongation. Water and solute flows in the coupled system of xylem and phloem were modeled together with predictions for xylem and whole stem diameter changes. With the model we could produce water circulation between xylem and phloem as presented by the Münch hypothesis. Viscosity was modeled as an explicit function of solute concentration and this was found to vary the resistance of the phloem sap flow by many orders of magnitude in the possible physiological range of sap concentrations. Also, the sensitivity of the predicted phloem translocation to changes in the boundary conditions and parameters such as sugar loading, transpiration, and hydraulic conductivity were studied. The system was found to be quite sensitive to the sugar-loading rate, as too high sugar concentration, (approximately 7 MPa) would cause phloem translocation to be irreversibly hindered and soon totally blocked due to accumulation of sugar at the top of the phloem and the consequent rise in the viscosity of the phloem sap. Too low sugar loading rate, on the other hand, would not induce a sufficient axial water pressure gradient. The model also revealed the existence of Münch “counter flow”, i.e., xylem water flow in the absence of transpiration resulting from water circulation between the xylem and phloem. Modeled diameter changes of the stem were found to be compatible with actual stem diameter measurements from earlier studies. The diurnal diameter variation of the whole stem was approximately 0.1 mm of which the xylem constituted approximately one-third.  相似文献

Summary A sequence of histoautoradiographs suggests that absorbed foliarapplied calcium-45 moves in the oat leaf upwards to the tip through the xylem vessels and when present at high concentration, downwards through the phloem elements. 45Ca accumulates in the outer bundle-sheath cells, in the phloem elements of the smaller, less differentiated bundles and in the mesophyll and epidermal cells. Tetraena mongolica Maxim is a narrowly monotypic genus of Zygophyllaceae found in a very limited area in the western part of Inner Mongolia, China. The plant is called "oil firewood" and its stems and branches are used as fuelwood. As triacylglycerol (TAG) is the main component of the plant oil, the TAG content was analyzed, as were the distribution of oleosomes in different tissues of the stem. This was in order to ascertain whether the term "oil firewood" referred to this storage lipid. Stems of T. mongolica indeed contained high levels of TAG (approximately 46 mg/g of dry matter or DM). The concentration of TAG in phloem (90 mg/g of DM) was much higher than that in xylem (20mg/g of DM), and semi-thin sections stained by Sudan Black B showed that almost all cells in the phloem contained oleosomes whereas in the xylem, oleosomes were found only in parenchymatous cells. These results suggest that T. mongolica has a high capacity to accumulate TAG in its stem cells.  相似文献

钙是植物必需的营养元素。酸性砂质土壤中含钙较少,导致在其土壤上生长的作物容易缺钙。另外由于果树果实、果菜类和包心叶菜类的蒸腾作用弱,导致果树和蔬菜普遍生理缺钙。根系维管束组织可能通过共质体和质外体两种途径进行钙素吸收,而果实则可通过非维管束组织直接吸收钙素。Ca2 通过Ca2 通道内流进入胞质,并通过Ca2 -ATPase和Ca2 /H 反向转运蛋白外流以保持胞质内低Ca2 浓度。为了应对植物发育和环境胁迫信号,Ca2 由质膜、液泡膜和内质网膜的Ca2 通道内流进入胞质,导致胞质Ca2 浓度迅速增加,产生钙瞬变和钙振荡,传递到钙信号靶蛋白,如钙调素、钙依赖型蛋白激酶及钙调磷酸酶B类蛋白,引起特异的生理生化反应。本文综述了植物钙素吸收、转运以及代谢研究的最新进展,包括植物对钙的需求和作物缺钙的原因,根系维管束组织及果实钙素吸收机理,Ca2 跨膜运输特性,钙的信使作用以及钙信号靶蛋白等方面内容。  相似文献

Phytochelatins (PCs) are glutathione-derived peptides that function in heavy metal detoxification in plants and certain fungi. Recent research in Arabidopsis has shown that PCs undergo long-distance transport between roots and shoots. However, it remains unknown which tissues or vascular systems, xylem or phloem, mediate PC translocation and whether PC transport contributes to physiologically relevant long-distance transport of cadmium (Cd) between shoots and roots. To address these questions, xylem and phloem sap were obtained from Brassica napus to quantitatively analyze which thiol species are present in response to Cd exposure. High levels of PCs were identified in the phloem sap within 24 h of Cd exposure using combined mass spectrometry and fluorescence HPLC analyses. Unexpectedly, the concentration of Cd was more than four-fold higher in phloem sap compared to xylem sap. Cadmium exposure dramatically decreased iron levels in xylem and phloem sap whereas other essential heavy metals such as zinc and manganese remained unchanged. Data suggest that Cd inhibits vascular loading of iron but not nicotianamine. The high ratios [PCs]/[Cd] and [glutathione]/[Cd] in the phloem sap suggest that PCs and glutathione (GSH) can function as long-distance carriers of Cd. In contrast, only traces of PCs were detected in xylem sap. Our results suggest that, in addition to directional xylem Cd transport, the phloem is a major vascular system for long-distance source to sink transport of Cd as PC–Cd and glutathione–Cd complexes.  相似文献

Recent work has demonstrated that minerals in plants are circulated between root and shoot. This occurs during the whole life time and renders possible response to changing environmental conditions. This mineral circulation occurs through intensive solute exchange between xylem and phloem in roots, stems, and leaves. The transport form of heavy metals such as iron, manganes, zinc and copper in the phloem, whether ionic or chelated, is unclear in most cases.
The unusual amino acid nicotianamine (NA) is ubiquitous throughout the plant kingdom. It is a chelator of several divalent transition metals. Its physiological role was investigated with the tomato mutant chloronerva, the only known NA-free multicellular plant. The mutant also exhibits disturbances of its iron metabolism and that of other heavy metals. This leads, among others, to a typical intercostal chlorosis and progressive iron accumulation in the leaves. From the heavy metal chelating properties of NA and from the phenotype of the mutant chloronerva it is concluded that NA is needed for normal distribution of heavy metals in young growing tissues fed via the phloem. This function could be fulfilled by mediating phloem loading or unloading of heavy metals as well as by preventing their precipitation in the alkaline phloem sap. An attempt is made to explain the chloronerva phenotype in the light of the phloem transport hypothesis of chelated iron.  相似文献

Secondary growth in the stem of Dolichos lablab is achieved by the formation of eccentric successive rings of vascular bundles. The stem is composed of parenchymatous ground tissue and xylem and phloem confined to portions of small cambial segments. However, development of new cambial segments can be observed from the obliterating ray parenchyma, the outermost phloem parenchyma and the secondary cortical parenchyma. Initially cambium develops as small segments, which latter become joined to form a complete cylinder of vascular cambium. Each cambial ring is functionally divided into two distinct regions. The one segment of cambium produces thick-walled lignified xylem derivatives in centripetal direction and phloem elements centrifugally. The other segment produces only thin-walled parenchyma on both xylem and phloem side. In mature stems, some of the axial parenchyma embedded deep inside the xylem acquires meristematic activity and leads to the formation of thick-walled xylem derivatives centrifugally and phloem elements centripetally. The secondary xylem comprises vessel elements, tracheids, fibres and axial parenchyma. Rays are uni-multiseriate in the region of cambium that produces xylem and phloem derivatives, while in some of the regions of cambium large multiseriate, compound, aggregate and polycentric rays can be noticed.  相似文献

We carried out a theoretical analysis of phloem transport based on Münch hypothesis by developing a coupled xylem-phloem transport model. Results showed that the maximum sugar transport rate of the phloem was limited by solution viscosity and that transport requirements were strongly affected by prevailing xylem water potential. The minimum number of xylem and phloem conduits required to sustain transpiration and assimilation, respectively, were calculated. At its maximum sugar transport rate, the phloem functioned with a high turgor pressure difference between the sugar sources and sinks but the turgor pressure difference was reduced if additional parallel conduits were added or solute relays were introduced. Solute relays were shown to decrease the number of parallel sieve tubes needed for phloem transport, leading to a more uniform turgor pressure and allowing faster information transmission within the phloem. Because xylem water potential affected both xylem and phloem transport, the conductance of the two systems was found to be coupled such that large structural investments in the xylem reduced the need for investment in the phloem and vice versa.  相似文献

Modification of external morphology and internal structure of plants is a key feature of their successful survival in extreme habitats. They adapt to arid habitats not only by modifying their leaves, but also show several modifications in their conducting system. Therefore, the present study is aimed to investigate the pattern of secondary growth in Leptadenia pyrotechnica (Forssk.) Decne., (Asclepiadaceae), one such species growing in Kachchh district, an arid region of Gujarat State. A single ring of vascular cambium, responsible for radial growth, divided bidirectionally and formed the secondary xylem centripetally and the phloem centrifugally. After a short period of secondary xylem differentiation, small arcs of cambium began to form secondary phloem centripetally instead of secondary xylem. After a short duration of such secondary phloem formation, these segments of cambium resumed their normal function to produce secondary xylem internally. Thus, the phloem strands became embedded within the secondary xylem and formed interxylary phloem islands. Such a recurrent behavior of the vascular cambium resulted in the formation of several patches of interxylary phloem islands. In thick stems the earlier formed non-conducting interxylary phloem showed heavy accumulation of callose on the sieve plates followed by their crushing in response to the addition of new sieve elements. Development of intraxylary phloem is also observed from the cells situated on the pith margin. As secondary growth progresses further, small arcs of internal cambium get initiated between the protoxylem and intraxylary phloem. In the secondary xylem, some of the vessels are exceptionally thick-walled, which may be associated with dry habitats in order to protect the vessel from collapsing during the dryer part of the year. The inter- and intraxylary phloem may also be an adaptive feature to prevent the sieve elements to become non-conducting during summer when the temperature is much higher.  相似文献

以盆栽2年生富士苹果树(砧木为平邑甜茶M.hupehensisReld)为试材,研究了盐胁迫对苹果矿质营养平衡的影响.结果表明,在盐胁迫下,苹果各器官不同时期的单位干样中Ca、Mg、Fe和Zn含量的平均值没有明显变化,但各元素与Na的比值明显下降,特别是在高盐(3‰NaCl)胁迫下下降更为明显,从而破坏了树体内元素平衡.在无盐和盐胁迫下,苹果各器官中Ca含量的顺序为主干韧皮部>叶片、新梢>根>主干木质部;Mg含量为新梢、根>主干木质部、主干韧皮部、叶片;Fe含量为根>叶片>主干韧皮部、新梢>主干木质部;Zn含量为新梢>叶片>根、主干韧皮部>主干木质部.与对照相比,器官中各元素含量在胁迫期间表现出不同程度的波动性.  相似文献

Understanding the regulation of calcium uptake, xylem transport and its impacts on growth and leaf gas exchange is a subject that has received insufficient recent attention. Calcium (Ca) is unique within the group of key elements required for plant growth in that it also has a role in cellular signalling via regulation of changes in its cytoplasmic concentration. Its mobility, within the plant, is however somewhat constricted by its chemistry and cellular signalling role, and its adsorptive capacity within the aopoplast and the xylem. Supply and demand for Ca is achieved by a homeostatic balance which if perturbed can cause a number of distinctive physiological conditions, often related to Ca deficiency. In this issue Rothwell and Dodd present experiments with bean (Phaseolus vulgaris) and pea (Pisum sativum) plants grown in a field soil exposed to the processes of soil liming (application of Ca carbonate (CaCO3). Given that there is evidence of free Ca in the xylem sap altering stomatal conductance it is reasonable to ask the question does liming elevate Ca in the transpiration stream which may explain the observed reduced growth which they hypothesise is due to Ca-induced stomatal closure. They show that liming doubled soil exchangeable Ca, reduced stomatal conductance and shoot biomass in both species compared with unlimed controls. However, xylem sap Ca concentration increased only in bean. Interestingly, the same was not true for the pea where the root xylem sap concentration remained unchanged despite an increase in soil available Ca. Given that stomatal conductance decreased in both species, but in response to a lime-induced increase in xylem sap Ca in only one; this questions the role of Ca in inducing stomatal closure. They propose that their data suggest that as yet unidentified antitranspirant causes stomatal closure in both species not the increase in xylem sap Ca per se.  相似文献

Growth and mineral composition of fruits in potted trees were studied at two temperatures (19 and 24°C) in two experiments with Golden Delicious and one with Cox's Orange Pippin under a controlled environment. In Golden Delicious the high temperature promoted fruit growth markedly, but in Cox's Orange Pippin the stimulation was much less pronounced. The levels of K, N, Mg, and P (expressed as amounts per fruit) were increased at the higher temperature. This increase was almost completely the result of differences in fruit growth. With respect to Ca the high temperature reduced Ca influx in one Golden Delicious experiment and had hardly any effect in the other, but favoured influx in Cox's Orange Pippin. In one Golden Delicious experiment the temperature was raised 7 weeks after blossoming, which resulted in a temporary drop of Ca. When the same temperature change was applied 4 weeks earlier, Ca continued to increase. It is assumed that mineral influx occurs via the xylem during the first few weeks after fruit set and via the phloem throughout. The results are discussed in terms of differences in mobility in the phloem for the various minerals and of a shift in the ratio between xylem and phloem movement induced by differences in the growth rate of the fruits.  相似文献
