The effect of gibberellic acid (GA3) on ion ratios in a dwarf maize mutant (Zea mays L. d1) exhibiting normal growth after hormone treatment has been investigated by electron microprobe analysis. Gibberellic-acid treatment increased the ion content in chloroplasts and vacuoles whereas no change of the ion content was found in the cytoplasm. The short-term kinetics of growth of the excised lettuce (Lactuca sativa L.) hypocotyl were characterized with respect to the effects of gibberellic acid (GA3), indole-3-acetic acid (IAA), KCl and pH. A Hall-device-based, miniaturized, linear displacement transducer was developed to measure the growth of 2-mm hypocotyl sections with 1-m resolution. Following treatment with GA3, a lag time of less than 10 min was typically followed by an increase in growth rate with two acceleration phases, reaching a final elevated rate within about 1 h. The kinetics of the response to GA1, a mixture of GA4 and GA7, and GA9 were similar to the response to GA3. There was no response to IAA treatment either in the presence or absence of GA3. KCl alone had no effect on the growth rate, but caused an increase in rate when added after GA3, with a lag time of usually less than 1 h. Responses to pH changes had lag times of a few minutes in all cases. A shift from H2O to pH 6 buffer inhibited growth, while a shift from H2O to pH 4 buffer resulted in a transient increase to a rate comparable to that induced by GA3. A shift from pH 6 to pH 5 caused an increase in growth rate, followed by a gradual decline to an H2O control rate after more than an hour. Viable, long-lived, gibberellic acid (GA3)-responsive protoplasts have, for the first time, been isolated from aleurone layers of mature wild oat (Avena fatua L.) grain. More than 90% of the cells of aleurone layers are recovered as protoplasts, and these respond to treatment with GA3 in essentially the same manner as the tissue from which they were derived. Protoplasts become vacuolate during incubation in vitro and, although not dependent upon GA3, vacuolation is markedly stimulated by the hormone. Amylase and ribonuclease (RNase) are produced and secreted only in the presence of GA3 and only after lag periods of 3 d and 4 d respectively. The amounts of amylase produced and secreted are proportional to GA3 concentrations as low as 1.61·10-13 M. With increasing concentrations of mannitol in the culture medium both vacuolation and the GA3-induced production and secretion of enzymes are inhibited progressively, the latter being precluded by 0.6 M to 0.7 M mannitol.Abbreviations GA3 gibberellic acid3 - RNase ribonuclease  相似文献

应用酶联免疫吸附测定法(ELISA)研究了梾木种子低温层积过程中内源激素含量的动态变化,分析了内源激素与种子休眠与发芽的关系。结果表明:(1)梾木种子中IAA含量在层积处理初期剧烈降低,持续一段时间后又显著升高,但后期下降,且IAA/ABA也出现同样的变化;种子中ABA含量在层积处理前期较高,但随着处理时间的延长趋于下降;种子内GA1/3含量以及GA1/3/ABA均随层积处理时间的延长逐渐增大;种子内ZRs和iPAs含量的变化相对较为平稳,尽管有一定的波动,但整体呈渐趋增高趋势。(2)梾木种子发芽率及发芽势在未经层积处理以及处理时间少于90d的条件下均为0,但随着层积处理时间的延长二者明显上升,层积处理的时间长短对梾木种子萌发有显著影响。(3)相关分析表明,梾木种子内GA1/3含量与种子的发芽率、发芽势均呈显著正相关关系,相关系数分别为0.688、0.662;种子内GA1/3/ABA增大有利于种子休眠解除和萌发。  相似文献

Leaf discs from etiolated bean plants were found to incorporate [3H]lysine into 80 S ribosomesynthesized proteins in the presence of chloramphenicol (100 mg l–1) when exposed to light. After a 7 min pulse of [3H]lysine, the discs were transferred to the same medium but with nonradioactive lysine, and postincubation was carried out for 24 h. The number of silver grains over the plastids, after the first period of a lag phase, indicates a large increase between 12 and 24 h of postincubation. Simultaneously, the labeling of the cytoplasm becomes reduced during that period. The results show that during inhibition of the protein formation within plastids, the synthesis of plastid-destined proteins in cytoplasm, as well as their transport into plastids, can still proceed.  相似文献

In the present report, we described the effects of indole-3-acetic acid (IAA), zeatin (ZT), gibberellin (GA3), and abscisic acid (ABA) on in vitro pollen germination and pollen tube growth in Torenia fournieri L. The results showed that IAA and GA3 stimulated in vitro pollen tube growth, ABA inhibited pollen tube growth, and ZT had no significant effect on the process. The stimulating effect of exogenous IAA was particularly distinct, and led to synchronous growth of straighter and more slender pollen tubes compared with the controls. However, no significant changes were found in the germination of the treated pollen. The auxin efflux inhibitor, 10 μM 1-N-naphthylphthalamic acid (NPA), was also found to stimulate pollen tube growth. We measured the content of hormones (free IAA, ZT, GA3, and ABA) in the stigmas and styles before and after pollination. The hormone contents of stigmas measured 0.5 h after pollination (0.5 HAP) showed that ABA content decreased, whereas the content of IAA, ZT, or GA3 did not change significantly. The hormone level in pollinated styles (4 HAP) when pollen tubes had grown into the middle part of style was characterized by an increase in free IAA and GA3 and a decrease in ABA, which was in agreement with the results that IAA and GA3 promoted but ABA inhibited pollen tube growth in vitro. Furthermore, the change of IAA level in styles was most notable, which was accordant to the fact that auxin stimulated significantly pollen tube growth in vitro. Using immunoenzyme and immunogold labeling techniques and an anti-IAA monoclonal antibody, we confirmed that free IAA was present throughout style tissues, and distributed in the nucleus and cytoplasm of style cells. All these results suggested that hormones, especially IAA, play important roles in pollen tube growth of T. fournieri. Gibberellin A4 (GA4) was identified for the first time in the garden pea (Pisum sativum) L.), by gas chromatography-mass spectrometry. However, in wild-type shoots the level of GA4 was only about 6% of the level of GA1, and it is therefore unlikely that GA4 plays a major role per se in the control of pea stem elongation. In shoots of the le mutant, GA4 was not detected, while the level of GA9 was approximately twice that found in the wild-type. The le mutation also markedly reduced the elongation response to applied GA9. It appears, therefore, that in Pisum the le mutation blocks the 3-hydroxylation of GA9 to GA4, in addition to the 3-hydroxylation of GA20 to GA1. In contrast, the le mutation did not reduce the response to applied GA5, suggesting the step GA5 to GA3 is not catalysed by the enzyme controlled by the Le gene. The step GA5 to GA3 was confirmed in peas by metabolite analysis after treatment with deuterated GA5.  相似文献

Summary Treatment of isolated barley aleurone layers with gibberellic acid (GA3) resulted in a progressive inhibition of cell-wall synthesis after a 4-h lag period. The incorporation of both [14C]arabinose and [14C]glucose into the cell wall was inhibited by GA3, but analysis of the labelled sugars in the polymerized product showed that the process most affected by the hormone treatment was pentosan biosynthesis. Labelling kinetics and pulse-chase analysis indicated that the pentosans were synthesized in the cytoplasm and subsequently transferred to the cell wall; GA3 did not significantly affect the latter step. The GA3-inhibited labelling of the cell-wall pentosans cannot be explained on the basis of an effect on uptake of radioactive cell-wall precursor, expansion of the free pentose pool, or degradation of newly-formed pentosan. GA3 inhibited the activity of a membrane-bound arabinosyl transferase present in the aleurone layers. The production of gibberellic acid (GA3) by Fusarium moniliforme M-7121 in solid-state culture was evaluated in flask cultures as well as in 3-I horizontal rotary reactors. The highest production rate of GA3 was with 80% (w/v) maize flour mixed with wheat bran. The optimum initial moisture content was inversely dependent on the ambient relative humidity. The initial water activity range for optimal growth and GA3 accumulation was about 0.98 to 0.99, which is unusually high for a filamentous fungus. A low O2 concentration resulted in a much decreased GA3 yield and the appearance of a yellow to reddish pigmentation in the mycelium. The lag phase was short and rapid growth continued for up to 2 days in the rotary reactor, with a maximum specific growth rate of 0.12 h–1. The maximum rate of GA3 production occurred during the subsequent 3 to 10 days of incubation and the final GA3 concentration reached was 18.7 mg to 19.3 mg/g dry culture. 关于中国沙棘克隆生长调节研究目前局限于外在机制,旨在探讨其克隆生长对灌水强度的响应规律及其激素调控的内在机制。结果表明:随着灌水强度的增大,分株生长、克隆繁殖、克隆扩散能力先升后降,IAA(吲哚-3-乙酸)、ZR(玉米素核苷)、GA_3(赤霉酸)含量及其与ABA(脱落酸)的比值先升后降而ABA含量先降后升。同时,分株生长、克隆繁殖、克隆扩散能力与IAA、ZR、GA_3含量以及IAA/ABA、ZR/ABA、GA_3/ABA呈极显著或显著正相关,与ABA含量呈极显著负相关。灌水强度过小或过大,IAA、ZR、GA_3含量以及IAA/ABA、ZR/ABA、GA_3/ABA低而ABA含量高,克隆生长潜力受到抑制,种群以分株小、数量少(分布稀疏)、扩散(水平根延伸和分枝)能力弱为特征,克隆生长格局倾向于"游击型"、种群早衰概率高;灌水强度适宜,IAA、ZR、GA_3含量以及IAA/ABA、ZR/ABA、GA_3/ABA高而ABA含量低,克隆生长潜力得以充分发挥,种群以分株大、数量多(分布密集)、扩散能力强为特征,克隆生长格局倾向于"聚集型"、种群稳定性高。随着灌水强度过小-适宜-过大的连续变化,中国沙棘通过改变激素状况调控克隆生长,从而形成与灌水强度相适应的克隆生长格局连续体"游击型-聚集型-游击型",种群稳定性呈"低-高-低"的连续变化过程。由此可见:灌水强度诱导内源激素发生改变,激素特征调控克隆生长格局,克隆生长格局决定种群稳定性。  相似文献

以1年生紫斑牡丹幼苗为试验材料,采用不同浓度(0、100、300、500 mg/L)赤霉素(GA_3)喷施叶片处理,通过透射电镜、扫描电镜、光学显微镜观察幼苗叶片解剖结构,光合仪测定幼苗光合参数并以酶联免疫吸附法测叶片内源激素含量,探究外源GA_3对紫斑牡丹幼苗叶片解剖结构、光合特性和内源激素水平的影响。结果表明:(1)低浓度GA_3处理的紫斑牡丹叶肉细胞增大,栅栏组织外层细胞中叶绿体数量增加,高浓度GA_3处理则与之相反;GA_3处理叶片的栅栏组织/海绵组织比值(P/S)、组织结构紧密度(CTR)均下降,而其组织结构疏松度(SR)增加;GA_3处理的幼苗叶片的叶肉细胞内各叶绿体大小显著大于对照,随着GA_3处理浓度增加,紫斑牡丹叶肉细胞内叶绿体的体积趋于增大,类囊体垛叠凝聚逐渐松散,叶绿体上淀粉颗粒在300 mg/L GA_3处理中较明显;叶片气孔长度、宽度、气孔器大小、气孔开度和气孔密度随着GA_3浓度升高先升高后下降,同时叶片上表皮角质层厚度随GA_3浓度的升高而增加。(2)紫斑牡丹叶片净光合速率(P_n)、气孔导度(Cond)、蒸腾速率(T_r)、水分利用率(WUE)在100和300 mg/L GA_3处理下大都显著高于对照,且300 mg/L GA_3处理显著高于其余处理,而其在500 mg/L GA_3处理下显著低于对照。(3)紫斑牡丹叶片脱落酸(ABA)和吲哚乙酸(IAA)含量均在500 mg/L GA_3下显著高于对照,而在其余浓度处理下不同程度低于对照,叶片内源玉米素核苷(ZR)和GA_3含量均在300 mg/L GA_3处理下显著高于其余处理和对照,而其余处理相比对照均无显著变化;叶片的ZR/ABA、ZR/IAA、ZR/GA_3和(IAA+GA_3+ZR)/ABA比值都在300 mg/L GA_3处理下显著高于其他处理,叶片的IAA/ABA和ABA/GA_3比值均在500 mg/L GA_3处理下显著高于其他处理。研究发现,适宜浓度外源GA_3处理,能显著提高紫斑牡丹幼苗叶片光合速率、水分利用效率及蒸腾速率,调节植物体内源激素的含量及平衡,从而使叶片能合成较多有机物,促进幼苗生长。  相似文献

Changes in gibberellins (GAs), indole-3-acetic acid (IAA), and cytokinins associated with the transition from vegetative growth to reproductive growth in Humulus lupulus L. buds and leaves harvested at fortnight intervals were studied. During vegetative growth, GA1 increased gradually and the lowest content was observed during flower development. Both GA3 and GA4 showed a dramatic increase in the samples taken from the apical part of axillary branches from plants 4–5 m high, which corresponds to the maximum vegetative development prior to macroscopically visible inflorescences. Notable increases in the cytokinins trans-zeatin (t-Z), isopentenyladenine (iP), and the riboside and ribotide forms of iP were also obtained. The auxin, indole-3-acetic acid, was the most abundant plant hormone, and its content was highest during vegetative growth. These results show for the first time a relationship between endogenous hormone profiles and both vegetative and reproductive development in hop plants, which may be relevant for future research on the control of the flowering by exogenous hormone applications.  相似文献

The effect of gibberellin A1 (GA1) on production of ethylene by cowpea (Vigna sinensis cv Blackeye pea no. 5) epicotyl explants and its relationship to epicotyl elongation was investigated. The explants were placed upright in water and incubated in sealed culture tubes or in large jars. GA, and IAA in ethanol solution were injected into the subapical tissues of the decapitated epicotyls. Cowpea epicotyl explants elongated after GA but not after IAA treatment, and they were very sensitive to exogenous ethylene. As little as 0.14 1/1 ethylene reduced significantly GA1-induced epicotyl elongation.Treatment with GA1 induced the production of ethylene which began 10 h after GA application, showed a peak at about 22 h and then declined. The yield of ethylene was proportional to the amount of GA, injected. The inhibition of epicotyl elongation in closed tubes was avoided by absorbing ethylene released with Hg(Cl04)2 , or by adding AVG to the incubation solution to inhibit ethylene production. Treatment with IAA elicited a rapid production of ethylene which ceased about 10 h after application. The roles of gibberellic acid (GA3) and fusicoccin (FC) in the elongation growth and acidification of the medium by excised hypocotyl sections of lettuce (Lactuca sativa L.) were investigated. Hypocotyl sections incubated in buffer without GA3 elongate optimally at pH 4.0–4.25 while sections incubated with GA3 show the same growth between pH 4.25 and 6.0. Preincubation of sections at pH 6.0 for 6 h does not affect the subsequent elongation response to acidic medium (pH 4.25); however, the sections become refractory to further acid treatment after their initial burst of growth in response to pH 4.25. Sections made refractory to acid are responsive to GA3 application, however, and the rate of growth in response to GA3 of sections pretreated for 6 h at pH 4.25 is 85% of that of sections pretreated at pH 6.0. Although preincubation of sections for 48 h in medium at pH 6.0 abolishes the GA3 response, it does not affect the response to buffer at pH 4.25. FC stimulates elongation growth in letuce hypocotyls at an optimal concentration of 1 M, and pretreatment of sections at pH 4.25 does not affect this elongation response. Although both GA3 and FC increase elongation of the section, neither causes appreciable acidification of the medium. Addition of KCl or NaCl to FC-treated sections causes rapid medium acidification but addition of salts to GA3-treated tissue does not cause acidification. Abrasion of the hypocotyl to remove the cuticle does not enhance acidification of the medium by the sections nor deos it affect elongation of the sections in response to GA3 or FC. Medium acidification by the sections is not a passive process since it is abolished both by low temperature (2° C) and metabolic inhibitors (carbonyl cyanide-m-chlorophenyl-hydrazone, azide). The acidification of the medium by barley (Hordeum vulgare L.) roots in response to FC is also dependent on the presence of KCl. Several of the 16,17-dihydro gibberellins (GAs) inhibit elongation in a variety of species. In a study of their mechanism of action we have investigated the effect of exo-16,17-dihydro-Ga5 (diHGA5) on the metabolism of GA20 in dwarf rice (Oryza sativa cv. Tan-ginbozu). A mixture of [3H]- and [3H]-GA20 (100 ng per plant) was applied in microdrops to 4 d old seedlings which were harvested 72 h later. Concurrent treatment with diHGA5 at 100 ng or 333 ng per plant reduced GA20-induced elongation of the second leaf sheath by 41–66%. There was a concomitant reduction in the amount of [2H2]GA1 present at harvest, measured by gas chromatography-mass spectrometry-selected ion monitoring. The [2H2]GA29 content was also reduced. There was no clear effect of diHGA5 on the total radioactivity recovered, or on conversion of the [3H]GA20 to putative [3H]GA conjugates, or on the amount of [2H2]GA20 found. No free [2H2]GA8 was detected. In other experiments there was little effect of diHGA5 on elongation induced by treatment with GA1. Seed maturation of Pisum sativum cv. Progress No. 9 proceeds more slowly in winter than in summer even when the parent plants are grown in greenhouse conditions with light-and heat-supplementation. For parent plants grown under summer and winter conditions the metabolism of [3H]GA9 in cultured seeds is qualitatively different in seeds of equivalent age and qualitatively the same in seeds of equivalent weight. 13-Hydroxylation of [3H]GA9[3H]GA20 is restricted to early stages of seed development. 2-Hydroxylation of [3H]GA92-OH-[3H]GA9 has only been observed at a stage of development after endogenous GA9 has accumulated. 2-OH-GA9 has been shown to be endogenous to pea and is named GA51. H2-GA31 and its conjugate have not been shown to be present in pea and may be induced metabolites of [3H]GA9. The metabolism of GA20GA29 is used to illustrate a technique of feeding [2H][3H]GAs in order to distinguish a metabolite from the same endogenous compound. The in vitro conversion of [3H]GA20[3H]GA29, and the virtual non-metabolism of [3H]GA29 have been confirmed for seeds in intact fruits. The cell wall loosening enzymes viz. glycosidases, polygalacturonase and xylanase were analyzed in cytoplasmic and wall bound fractions extracted from control and hormone (GA3 NAA, PAA) treated internodes, as they are known to play a key role in cell wall metabolism. Among the glycosidases, wall bound β-glucosidase and α-galactosidase activities were significantly correlated with age of control internodes. Cytoplasmic α-galactosidase showed significant correlation in hormone treated internodes. Maximum correlation was observed in GA3, followed by PAA and NAA. Wall bound xylanase activity was well correlated with length only in NAA treated internodes and less after GA3 treatment while cytoplasmic xylanase showed correlation with intrnode length only in control and after NAA treatment. Cytoplasmic polygalacturonase showed correlation with internode length only after GA3 treatment while wall bound polygalacturonase showed correlation with internode length after NAA treatment. The possible role of these enzymes in internode development is discussed.  相似文献

The stimulatory effects of gibberellic acid (GA3) and the inhibitory effects of polyethylene glycol 4000 (PEG) on hypocotyl elongation and cell cycle kinetics in subapical pith cells of dwarf watermelon seedlings (Citrullus lanatus [Thunb.] Matsu and Nakai) were investigated. Mitotic indices (MI) were determined from direct counts of pith cells stained by a modified Feulgen technique. Labeling indices (LI) were determined from direct counts of labeled pith cells sampled 1.5 h after apical applications of3H-thymidine. Root application of 0.32 mM GA3 at 96, 120, or 144 h after sowing resulted in significant increases in both mitotic and labeling indices within 4.5 to 7.5 h following treatment. A single mitotic peak at 13.5 h occurred in all three treatment periods. Labeling peaks were often less defined than mitotic peaks; however, a relatively high proportion of labeled nuclei were usually observed between 7.5 and 9 h after GA3 treatment and at 16.5 h, the latter period coinciding with progression of cells into S phase from the peak period of mitosis. The results suggest that GA3 increases the proportion of rapidly dividing cells in the subapical meristem by increasing the probability that slowly cycling or nonproliferative cells in both 2C and 4C DNA states will enter the proliferative pool. The addition of PEG (200 g/l, = 1.5 mPA) to the rooting medium of dwarf watermelon seedlings inhibited hypocotyl elongation and reduced both mitotic and labeling indices simultaneously within 4.5 h after treatment. Within 24–28 h after PEG treatment, mitotic and labeling indices approached 0. Seedlings transferred from PEG to either water or GA3 exhibited rapid recovery of cell division and hypocotyl elongation. Mitotic and labeling indices increased within 4.5–7.5 h into the recovery period in either water or GA3 and reached control values within 10.5 h. GA3 hastened the recovery from PEG-induced stress. It is concluded that water stress imposed by PEG 4000 causes arrest of cell division in meristematic cells of watermelon seedlings in both G1 and G2 periods. PEG and GA treatments resulted in only a partial and transitory synchronization of the cell cycle.  相似文献

It is well known that post-bloom applications ofgibberellic acid (GA3) increase seedless grapeberry size by enhancing cell division, or cellenlargement, or both. As a consequence, total waterand sugar per berry are increased. Soluble invertaseis considered to be one of the key enzymes in theaccumulation of sugar in grape berries. To study apossible role of invertase in the GA3berry-sizing effect, different rates of post-bloomGA3 were applied to seedless grape cv. Sultanaand hexose concentration and invertase activity weremeasured. GA3 stimulated both parameters as earlyas 24 and 32 h after applications, respectively.Moreover, the increment in sugar content and enzymeactivity remained throughout the growing of the berries period and, at ripening, increases in hexosescontent (102%) and invertase activity (60%) weredetected when GA3 was applied at a rate of 45 ppm.At the same GA3 rate the pericarp cellsdoubled in size.
