1. The distribution of phospholipase D (phosphatidylcholine phosphatido-hydrolase, EC 3.1.4.4) was examined in the tissues of a number of plants and seeds. 2. The highest activities were found in various swollen storage tissues of certain plants: cabbage, central stalk; cauliflower, flower; celery, swollen leaf stalk; Kohl rabi, swollen stem; carrot, root; pea and marrow, seed. 3. Appreciable activity was retained in pea seeds for at least 1 year after drying. After germination and growth in the dark the total activity present in the cotyledons and also in the whole seedling decreased. 4. In the growing pea seedling (7 days old), about 3% of the total activity was in the plumule, 9% in the root and the remainder in the cotyledons. However, the activity in the root on a dry-weight basis was higher than that in the cotyledons. In both the root and the plumule the activity on a wet- or a dry-weight basis was highest in the growing tip. 5. The activity per dry weight in the roots and aerial parts of pea plants declined to low values as growth continued, but roots struck from cuttings of mature plants showed the same high activity as found in roots from young seedlings with cotyledons attached. 6. The total phospholipids present in the cotyledons of pea seeds were depleted on germination and growth. Of the individual phospholipids, phosphatidylcholine and phosphatidylethanolamine showed the same loss in 11 days as the whole phospholipid fraction, whereas phosphatidylinositol was decreased to a greater extent and cardiolipin and phosphatidylserine were not decreased. There was no increase of phosphatidic acid, as might have been expected if the phospholipids had disappeared through phospholipase D hydrolysis. 7. It is concluded that phospholipase D in plant storage tissues and seeds may be related to the rapid growth involved in their formation rather than being necessary for the utilization of their food reserve substances.  相似文献

In this study we predicted and found evidence to support two general themes about new production systems measurement. First, there are two tiers of measures that are becoming popular to gauge performance. These two tiers are relatively independent-system-level measures like uptime, and business-level outcomes like return on investment. Second, when measures from these two categories are significantly correlated, they are likely to be for the time or flexibility measures at the system level. The rationale for this thesis is that modernization programs implement the leading edge of manufacturing strategies, and time or flexibility are replacing quality as priorities in leading-edge domestic manufacturing.  相似文献

Knowledge of the correlation between juvenileand mature-plant traits is critical in determining the opportunities for early stage selection. The effects of early stage selection on mature-plant performance have rarely been quantified. This study was conducted to identify seedling traints in red clover (Trifolium pratense L.) that correlate to mature-plant traits and to evaluate the effect of seedling selection on forage yields and other mature-plant traits. The results showed that relationships between most seedlingand mature-plant traits were weak (r ranged from 0.170 to 0.239). Nevertheless, selecting the top 10% seedlings for petiole length, days from emergence to full expansion of the 4th leaf (D4LE), or leaves per seedling, produced a mature-plant population with higher individual plant dry weight (IPDW1) and higher annual yield in 1993 (Y93). Selection for leaves per seedling increased IPDW1 by 23.2%. Selection for petiole length and the smallest D4LE increased Y93 by 15.7% and 13.8%, respectively. Furthermore, substantial expected genetic gains were obtained for IPDW1 and Y93 when selecting for some seedling traits. We conclude that plants and families with low potential yield can be eliminated at the seedling stage in red clover. This will allow breeders to increase the number of superior plants to be field tested or to conduct a more rigorous evaluation of the selected plants. Among the five selection schemes tested for direct selection of mature-plant traits, mass selection produced the largest genetic gain.  相似文献

Timing of cotyledon damage affects growth and flowering in mature plants   总被引:2,自引:0,他引:2  
Although the effects of herbivory on plant fitness are strongly linked to age, we understand little about how the timing of herbivory at the seedling stage affects growth and reproduction for plants that survive attack. In this study, we subjected six north-western European, dicotyledonous grassland species (Leontodon autumnalis, Leontodon hispidus, Plantago lanceolata, Plantago major, Trifolium pratense and Trifolium repens) to cotyledon removal at 7, 14 and 21 d old. We monitored subsequent growth and flowering (number of inflorescences recorded, and time taken for first flowers to open) over a 107 d period. Cotyledon removal reduced growth during establishment (35 d) for all species, and a further three exhibited reduced growth at maturity. Four species developed fewer inflorescences, or had delayed flowering after cotyledon removal. Although early damage (7 d old) had the greatest long-term effect on plant performance, responses varied according to the age at which the damage occurred and the species involved. Our results illustrate how growth and flowering into the mature phase is affected by cotyledon damage during different stages of seedling ontogeny, and we highlight the ways in which ontogenetic variation in seedling tolerance of tissue loss might impact upon plant fitness in mature plant communities.  相似文献

We present direct quasielastic neutron scattering measurements, in vivo, of macromolecular dynamics in Escherichia coli. The experiments were performed on a wide range of timescales to cover the large panel of internal and self-diffusion motions. Three major internal processes were extracted at physiological temperature: a fast picosecond process that corresponded to restricted jump diffusion motions and two slower processes that resulted from reorientational motions occurring in ∼40 ps and 90 ps, respectively. The analysis of the fast process revealed that the cellular environment leads to an appreciable increase in internal molecular flexibility and diffusive motion rates compared with those evaluated in fully hydrated powders. The result showed that the amount of cell water plays a decisive role in internal molecular dynamics. Macromolecular interactions and confinement, however, attenuate slightly the lubricating effect of water, as revealed by the decrease of the in vivo parameters compared with those measured in solution. The study demonstrated that standard sample preparations do not mimic accurately the physiological environment and suggested that intracellular complexity participates in functional dynamics necessary for biological activity. Furthermore, the method allowed the extraction of the self-diffusion of E. coli macromolecules, which presented similar parameters as those extracted for hemoglobin in red blood cells.  相似文献

Five plant species were cultivated on a soil from the Neckar alluvial fan near Heidelberg (FRG) polluted by the emissions of a cement plant. Thallium, cadmium and lead concentrations in seedlings and mature plants were determined by atomic absorption analysis. AdditionallyBrassica napus L.napus was grown on soils containing 5 different concentrations of heavy metals, achieved by mixing two similar soils, from the same area but with different metal concentrations. Thallium and cadmium were shown to be taken up by roots whilst lead which was also absorbed, was deposited mainly on the plant surface. However during cultivation in the winter months, a remarkable deposit of lead via the roots was found. Thallium in the soil from a anthrorogen source was more available to plants than thallium of geological origin. During the lifetime of a plant concentrations of thallium and cadmium were always highest in the seedling. The decrease in metal concentration with maturity depended on the plant species and the element, but was not a function of the metal concentration in the soil.  相似文献

Summary The relative rates of production by mature Digitalis purpurea plants of cholesterol, campesterol, stigmasterol and sitosterol isolated from the glycoside and lipid fractions of the plant extract, were estimated. Plants were exposed to an atmosphere of 14CO2 in a growth chamber and the radioactivity of the individual sterols assessed at intervals over 25 days on a gas-liquid radio chemical chromatography (GLRC). Incorporation of 14CO2 occurred within 12 hours into both fractions of the extract. The 5-ene sterols were produced at a similar rate over a period of 25 days but the lipid fraction was about 100% more radioactive than the glycoside fraction.  相似文献

Summary A review of past work and a comparison of the well-known disk technique with Stocker's whole leaf method for determining water deficit of some hardwood trees reveals confusion in existing terms and methods. The water relations of leaf disks cut from broadleaf trees cannot be assumed to be the same as whole leaves, since the excised disk usually requires more water per unit weight to saturate. Stocker's term wasser defizit (WD) and the whole leaf method, when modified to allow shorter equilibration schedules, remain the best way to express and measure water deficits in forest trees.  相似文献

There is growing interest in measuring the antioxidant status of plant tissues. This protocol describes the oxygen radical absorbance capacity (ORAC) assay, which measures antioxidant inhibition of peroxyl radical-induced oxidations and is a measure of total antioxidant capacity. The assay is performed in a microplate and is assessed with a 96-well multi-detection plate reader. Total antioxidant capacity of 64 experimental samples can easily be analyzed in 1 d. This assay is presented along with rapid assays for total phenolic content and total ascorbate content. Overall, these assays provide a general diagnostic tool of the antioxidant capacity in leaf tissue extracts.  相似文献

Cultures of Lavandula latifolia Medicus were established from axillary buds of mature field-grown plants. Explants were initially cultured on media with two different macronutrient combinations and benzyladenine or kinetin added either individually or with naphthaleneacetic acid. Subsequently, explants were subcultured in Murashige and Skoog medium supplemented with 20% coconut milk, 0.57 M indoleacetic acid and 8.88 M benzyladenine. Shoot proliferation from axillary buds was not affected by seasonal fluctuations in the stock plants but depended on the macronutrient composition and on the type and concentration of cytokinin tested. Best results were obtained in explants initially cultured in media with Murashige and Skoog constituents and supplemented with 5 M benzyladenine. In vitro-grown shoots were used to induce multiple shooting by transferring them to subculture medium. Shoots were rooted on Murashige and Skoog medium with macronutrients at half-strength. Plantlets were transferred to soil and grown to maturity.  相似文献

High species richness, resource availability and disturbance are community characteristics associated with forest invasibility. We categorized commonly measured community variables, including species composition, topography, and landscape features, within both mature and 15-year-old clearcuts in West Virginia, USA. We evaluated the importance of each variable for predicting the degree of forest invasion by early-establishing exotic invasive plants. Biotic variables, including overall richness (excluding exotic invasive species) and mutually exclusive native and exotic non-invasive species richness, were the strongest indicators of invasibility. Sites that were located on northeast-facing slopes, more mesic conditions, or in clearcuts were more likely to be invaded by exotic invasive plants. Invasion of clearcut sites was more dependent on available microsites (e.g., lower solar radiation, northeast-facing slopes, and lower elevations) within each site than on the condition of the surrounding landscape, whereas invasion into the mature forests was dependent more on the surrounding landscape (e.g., proximity to paved roads). Our results indicate that exotic invasive plant species in our study area respond similarly as other plant species to resource availability and that competitive interactions are relatively unimportant. Current invasion into this landscape is more likely to be a passive reaction to site conditions instead of a driver of change.  相似文献

Many plants are adapted to flower at particular times of year, to ensure optimal pollination and seed maturation. In these plants flowering is controlled by environmental signals that reflect the changing seasons, particularly daylength and temperature. The response to daylength varies, so that plants isolated at higher latitudes tend to flower in response to long daylengths of spring and summer, while plants from lower latitudes avoid the extreme heat of summer by responding to short days. Such responses require a mechanism for measuring time, and the circadian clock that regulates daily rhythms in behaviour also acts as the timer in the measurement of daylength. Plants from high latitudes often also show an extreme response to temperature called vernalisation in which flowering is repressed until the plant is exposed to winter temperatures for an extended time. Genetic approaches in Arabidopsis have identified a number of genes that control vernalisation and daylength responses. We describe an extension of the method of Myers et al. (1982) to measure with high precision the uniformity of contractile motions that occur between sarcomeres in the isolated cardiac muscle cell (guinea pig and rat). The image of the striations, observed with modulation contrast microscopy, was detected by a linear array of photodiodes. Sarcomere length was measured greater than 500/s from the frequency of the array's video signal at two selectable regions of the cell. A precision test grating demonstrated that method resolves known differences in the spacing between two contiguous striations to +/- 0.01 micron and that the effects of image translation and microscopic resolution are minor. The distribution of striation spacing appears to be discrete in isolated segments of the cell, and patches of fairly uniform length can be identified that are laterally contiguous. When electrically triggered, contraction is synchronous and the sarcomeres shorten and relengthen smoothly. The contrast between the striations is transiently enhanced during relengthening, an indication that the contracting cell can not be treated as a simple grating. Pauses that occur during late in relengthening (and transient contractile alternans) are characterized by very synchronized activity. These forms of irregular contractile behavior are not explained by desynchronization of a mechanism of release of intracellular calcium. Background and aims

As plants approach maturity and start to senesce, the primary sink for phosphorus (P) is the seed but it is unclear how plant P status affects the resulting P concentration and speciation in the seed and remaining plant parts of the residues. This study was established to measure how P speciation in different parts of wheat and canola is affected by plant P status.

Methods

Wheat and canola grown in the glasshouse were supplied three different P rates (5, 30 and 60 kg P ha?1 equivalent). At physiological maturity, plants were harvested and P speciation was determined for all plant parts (root, stem, leaf, chaff/pod and seed) and rates of P application, using solution 31P nuclear magnetic resonance (NMR) spectroscopy.

Results

Phytate was the dominant form of P in seed whereas orthophosphate was the dominant form of P in other plant parts. The distribution of P species varied with P status for canola but not for wheat. The phytate content of wheat chaff increased from 10 to 45 % of total P as the P rate increased. Canola pods did not show a similar trend, with most P present as orthophosphate.

Conclusions

Although minor differences were observed in P speciation across the three P application rates and plant parts, the effect of this on P cycling from residues into soil is likely to be relatively minor in comparison to the overall contribution of these residues to soil P pools. This glasshouse experiment shows the dominant P form in crop residues that is returned to soil after harvest is orthophosphate, regardless of plant P status.  相似文献

Somaclones regenerated from three wheat (Triticum aestivum L.) cultivars, Glennson, Pavon and PAK-16171 were evaluated for variation in agronomic and morphological characters. Calli were initiated from germinating seeds on Linsmaier and Skoog (LS) medium plus 2 mg/l 2,4-dichlorophenoxyacetic acid, 2% sucrose and 1% agar. Calli were isolated and regenerated into whole plants on LS medium containing 0.1 mg/l indole - 3-acetic acid and 0.5 mg/l benzyladenine. Comparisons among the somaclones and their parents were made for plant height, spike length, number of grains per spike, and 100 grain weight. Significant variation was observed in these characters between the somaclones and parents. Genotypic differences were observed among the somaclones for many of these agronomic and morphological characters.  相似文献
