Sperm transport and survival in the mare   总被引:1,自引:0,他引:1  
Following the deposition of semen in the mares uterus, spermatozoa must be transported to the site of fertilization, be maintained in the female tract until ovulation occurs, and be prepared to fertilize the released ovum. Sperm motility, myometrial contractions, and a spontaneous post-mating uterine inflammation are important factors for the transport and survival of spermatozoa in the mares reproductive tract. Fertilizable sperm are present in the oviduct within 4 hours after insemination. At this time, the uterus is the site of a hostile inflammatory environment. Our data suggest that spermatozoa trigger an influx of polymorphonuclear neutrophils (PMNs) into the uterine lumen via activation of complement. Furthermore, seminal plasma appears to have a modulatory effect on the post-mating inflammation through its suppressive effect on PMN chemotaxis and migration. Spermatozoa that safely have reached the oviduct can be stored in a functional state for several days, but prolonged sperm storage in the female tract is not required for capacitation and fertilization in the horse. The caudal isthmus has been proposed as a sperm reservoir in the mare. The pattern of sperm transport and survival of spermatozoa in the mares reproductive tract are different between fertile and subfertile stallions, between fertile and some infertile mares, and between fresh and frozen-thawed semen. Possible explanations for these differences include a selective phagocytosis of damaged or dead spermatozoa, impaired myometrial activity in subfertile mares, bio-physiological changes of spermatozoa during cryopreservation, and the removal of seminal plasma during cryopreservation of equine semen.  相似文献

Inflammatory conditions of the uterus, collectively known as endometritis, can be classified as acute, chronic, active, or subclinical. This condition causes substantial reductions in mare fertility. To diagnose and treat mares with endometrial inflammation, it is imperative to identify predisposing factors and challenges to the reproductive tract of the mare in a chronological order; these factors change from those of the young maiden filly to the older, multiparous broodmare. This paper addresses predisposing factors, diagnostic procedures and therapy strategies for different mare groups.  相似文献

A long-standing problem in epididymal physiology is the fate of unejaculated spermatozoa in the cauda epididymidis under conditions such as congenital absence of the vas deferens, long-term vasectomy, or castration. There is no convincing evidence for significant absorption of spermatozoa, defective or otherwise, by spermiophagy or dissolution in the epididymis of normal animals. Spermiophagy by epithelial cells or intraluminal macrophages may take place if the duct ruptures and granulomas form (e.g., after experimental ligation), although there is no quantitative information on the rate of sperm removal by this means. In one animal model (the rabbit), the epididymis is unusually resistant to granuloma formation and has provided unique insights into a phenomenon that is suggested to be present in all species. Spermatozoa retained in the rabbit cauda epididymidis by placing ligatures on the vas deferens and corpus epididymidis degenerate after several weeks but do not decrease significantly in numbers. After castration, however, they die very rapidly and >90% disappear. It is hypothesized that, in the normal androgen-maintained epididymis, degradative pathways are present in the luminal fluid that are constitutively inhibited by survival signals emanating from the epithelium. In the absence of androgen, the intraluminal mileau changes and death signals predominate that activate degradative pathways via the ubiquitin-proteasome system, DNAses, etc., to mediate dissolution of sperm organelles and nucleoprotein. It is suggested that the latter condition is the default situation and is only prevented by the stimulatory action of androgens on the epididymal epithelium.  相似文献

Transgenesis would be greatly streamlined if sperm cells could be used as transgene vectors. Attempts to 'force' sperm cells to take up transgenes may be more worthy of further study than 'autouptake' attempts. However, thorough proof of its effectiveness will be required if sperm cell-mediated gene transfer is to be accepted, given the evolutionary implications associated with the possibility of sperm cells being able to transfer exogenous DNA.  相似文献

Elucidation of the functional significance of the presence of acetylcholinesterase in spermatozoa was deferred until considerably after the initial observations that eserine increased flagellar beat frequency in Mytilus sperm (54). Discovered about forty years ago during surveys of seminal plasma constituents (2), the enzyme was later found to be associated with the particulate fraction (4). Even though characterized as a “true” acetylcholinesterase in the sperm flagellum (3), interpretations of the role of acetylcholine as a factor in motile processes remained speculative until a wide variety of cholinergic agents was found to affect sperm cell performance (32). Those agents which inhibit acetylcholine synthesis or block cholinergic receptors generally elicit a monophasic, inhibitory, dose-dependent response on sea urchin sperm swimming speed. These include hemicholinium, curare, α-bungarotoxin and decamethonium. Cholinomimetic substances and anticholinesterases (acetylcholine and nicotine; eserine, diisopropylfluorophosphate and neostigmine) act biphasically to increase the sperm swim rate at concentrations of 1 micromolar or less while higher concentrations slow or stop the forward motion of the sperm cells. Procaine (7), which competes with Ca2+ for cell surface binding sites, initially stimulates then completely inhinits sperm cell progression. Other substances which interfere with the cells′ access to Ca2+ or with the transport of Ca2+ into and within the cell adversely affect spermatozoan function. A conceptual model has been proposed, a) to relate the uptake of Ca2+ to cholinergic regulation of ionophoric channels through the sperm cell plasma membrane and b) for this Ca2+ to trigger release of sequestered calcium for coupling of excitation to flagellar wave propagation.  相似文献

Sperm transport and motility in the mouse oviduct: observations in situ   总被引:2,自引:0,他引:2  
Sperm transport and motility were studied through the transparent walls of the mouse oviduct by direct microscopic observation and videomicrography. Observations were made on excised female tracts 1-2 h post-coitus (pc) and 1-2 h before and after the approximate time of ovulation. Motile sperm were seen at the uterine entrance to the uterotubal junction (UTJ) in all females at 1-2 h pc, but in fewer females at later times. The intramural UTJ was usually constricted and held few sperm. The extramural UTJ and adjacent lower isthmus contained many motile sperm at 1-2 h pc. Apparently, the column of sperm moved upwards because in some females, sperm were found in the upper isthmus and not in the UTJ at the later time points. Few sperm were seen in the ampulla in the periovulatory period, and none at 1-2 h pc. There appeared to be two mechanisms retaining sperm in the lower oviduct: immobilization and adherence to the epithelium. Columns of immotile sperm were seen in the lower isthmus of some females. Motile sperm usually appeared to adhere by their heads to the oviductal epithelium, only occasionally breaking free to move vigorously about the lumen.  相似文献

The objectives of this study were 1) to determine the time of embryo transport through the mare oviduct, 2) to determine whether equine embryos increase in diameter prior to the time of oviductal transport, and 3) to assess the stage of equine embryonic development at the time of oviductal transport. The time of oviductal transport (interval from ovulation to uterine entry) was estimated by collecting embryos from the mare oviduct or uterus at 2-hour intervals from 120 to 168 h postovulation. The time of oviductal transport was 130 to 142 h, since 9 9 embryos were located in the oviduct from 120 to 128 h; 7 14 embryos were in the oviduct and 7 14 embryos were in the uterus from 130 to 142 h; and 13 14 embryos were in the uterus from 144 to 168 h postovulation. Embryos collected during the period of oviductal transport (130 h to 142 h) were not significantly larger (P>0.1) in diameter than embryos collected prior to the period of oviductal transport (162.5+/-3.7 vs 156.7+/-3.1 mum, respectively). During the period of oviductal transport, embryos collected from the uterus were not significantly larger (P>0.1) in diameter than embryos collected from the oviduct (160.7+/-3.2 vs 164.3+/-7.0 mum, respectively). During this same period 12 14 embryos were compact morulae, and 2 14 embryos were blastocysts.  相似文献

Anandamide (AEA) uptake has been described over the last decade to occur by facilitated diffusion, but a protein has yet to be isolated. In some cell types, it has recently been suggested that AEA, an uncharged hydrophobic molecule, passively diffuses through the plasma membrane in a process that is not protein-mediated. Since that observation, recent kinetics studies (using varying assay conditions) have both supported and denied the presence of an AEA transporter. In this review, we analyze the current literature exploring the mechanism of AEA uptake and endeavor to explain the reasons for the divergent views. One of the main variables among laboratories is the incubation time of the cells with AEA. Initial kinetics (at time points <1 min depending upon the cell type) isolate events that occur at the plasma membrane and are most useful to study saturability of uptake and effects of purported transport inhibitors upon uptake. Results with longer incubation times reflect events not only at the plasma membrane but also interactions at intracellular sites that may include enzyme(s), other proteins, or specialized lipid-binding domains. Furthermore, at long incubation times, antagonists to AEA receptors reduce AEA uptake. Another complicating factor in AEA transport studies is the nonspecific binding to plastic culture dishes. The magnitude of this effect may exceed AEA uptake into cells. Likewise, AEA may be released from plastic culture dishes (without cells) in such a manner as to mimic efflux from cells. AEA transport protocols using BSA, similar to the method used for fatty acid uptake studies, are gaining acceptance. This may improve AEA solution stability and minimize binding to plastic, although some groups report that BSA interferes with uptake. In response to criticisms that many transport inhibitors also inhibit the fatty acid amide hydrolase (FAAH), new compounds have recently been synthesized. Following their characterization in FAAH+/+ and FAAH-/- cells and transgenic mice, several inhibitors have been shown to have physiological activity in FAAH-/- mice. Their targets are now being characterized with the possibility that a protein transporter for AEA may be characterized.  相似文献

Sodium (Na) toxicity is one of the most formidable challenges for crop production world-wide. Nevertheless, despite decades of intensive research, the pathways of Na(+) entry into the roots of plants under high salinity are still not definitively known. Here, we review critically the current paradigms in this field. In particular, we explore the evidence supporting the role of nonselective cation channels, potassium transporters, and transporters from the HKT family in primary sodium influx into plant roots, and their possible roles elsewhere. We furthermore discuss the evidence for the roles of transporters from the NHX and SOS families in intracellular Na(+) partitioning and removal from the cytosol of root cells. We also review the literature on the physiology of Na(+) fluxes and cytosolic Na(+) concentrations in roots and invite critical interpretation of seminal published data in these areas. The main focus of the review is Na(+) transport in glycophytes, but reference is made to literature on halophytes where it is essential to the analysis.  相似文献

甾醇是一类广泛存在于生物体内的环戊烷骈多氢菲衍生物,其不仅是细胞膜的重要组成成分,还具有重要的生理和药理活性。随着合成生物学和代谢工程技术的发展,近些年来应用酵母细胞异源合成甾醇的研究不断深入。但由于甾醇是疏水性大分子,倾向于积累在酵母的膜结构中而引发细胞毒性,一定程度上限制了甾醇产量的进一步提升。因此,揭示酵母中甾醇转运机制,特别是与甾醇转运相关的转运蛋白的工作原理,有助于设计新的策略,解除酵母细胞工厂中的甾醇积累毒性、实现甾醇增产。酵母中甾醇转运主要通过蛋白质介导的非囊泡运输机制来完成,本文归纳了酵母中已报道的5类甾醇转运相关蛋白,即OSBP/ORPs家族蛋白、LAM家族蛋白、NPC样甾醇转运蛋白、ABC转运家族蛋白和CAP超家族蛋白,汇总了这些蛋白对细胞内甾醇梯度分布和稳态维持所起的重要作用。此外,本文还综述了甾醇转运蛋白在酵母细胞工厂中的应用现状。  相似文献

One of the most important waterway components is vegetation, which play a pivotal role in the flow and sediment transport. Vegetation environment and characteristics, including vegetation porosity or density, shape, flexibility and vegetation height, are significantly affected in vegetated channels. Various vegetation positions and arrangements such as patches significantly affect the sediment deposition rate and flow turbulence. This paper reviews recent works conducted on vegetated open channels, which include the effect of different vegetation arrangements and vegetation characteristics on mass transport and turbulence structure. Studies based on laboratory, field works, and modeling, have been reviewed based on previous methods used by different researchers. Methods used in vegetation porosity evaluation, rate of flow and sediment transport properties are presented. The purposes of this experiment were 1) to test the hypothesis that placing rabbit embryos into the mare's uterus would hasten oviduct transport and 2) to determine if placing fluid into the uterus of bred mares on Day 4 and/or Day 5 would subsequently disrupt the mare's pregnancy. The hypothesis that placing rabbit embryos into the mare's uterus would hasten oviduct transport was not supported, since the uterine recovery rate of equine embryos on Day 5 was not significantly higher (P>0.05) for mares receiving rabbit embryos on Day 4 than for mares receiving no uterine infusion on Day 4 (1 10 vs 0 10 , respectively). However, placing fluid into the mare's uterus on Day 4 was apparently responsible for hastened oviduct transport, since mares with media infused into the uterus on Day 4 had a significantly higher (P<0.05) recovery rate of equine embryos on Day 5 than did mares receiving either rabbit embryos or no uterine infusion on Day 4 post ovulation (5 10 vs 1 10 or 0 10 , respectively). The Day-14 pregnancy rate was significantly higher (P<0.05) for mares receiving no uterine infusion on Day 4 or Day 5 than for mares receiving uterine infusion on Day 5 or uterine infusion on both Days 4 and 5 (9 10 vs 4 10 , 2 10 and 0 10 , respectively).  相似文献

Sperm motility is a key factor in allowing us to determine semen quality and fertilizing capacity. Motility in semen is mainly controlled by K+ in salmonids, and probably also in sturgeons, and by osmotic pressure in other freshwater and seawater fish species, but other factors, such as concentration of surrounding metabolites and ions (Ca2+, Mg2+, etc.), pH and temperature also influence motility characteristics. In the present study, we have mainly reviewed and summarized the effects of temperature and pH on the motility of spermatozoa in three fish species: salmonids, cyprinids and sturgeons. Data in the literature show that motility, fertilizing ability and velocity of spermatozoa, as well as the duration of the motility period, depend on the temperature of the assay medium and also of that of the brood fish holding tank. In contrast, the pH of the swimming medium, and thus the intracellular pH of spermatozoa, has less influence on sperm motility parameters in cyprinids, salmonids and sturgeons.  相似文献
