The volume of hydrated pollen grains of Petunia hybrida L. during swelling in germination medium increased three times. The volume of desiccated pollen grains increased only two times after transfer to the same medium. This difference in swelling ability is attributed to different rigidities of the pollen grain wall,ccaused by the different hydration states. The wall of pollen grains exhibits morphological variation in many features including apertures, ornamentation and thickness, but the function of these characters remains to be clarified. It has been suggested that they are involved in the accommodation of volume changes (harmomegathy). To investigate this further, we developed a protocol that induces a controlled hydration of the pollen without affecting its metabolism and we applied it to six species differing in their pollen wall morphology. The entry of water caused pollen swelling and volume increase leading to breakage of the wall and/or of the plasma membrane, such that the per cent of intact grains was negatively correlated with the level of hydration. Qualitative and quantitative differences were observed between the species. Breakage of the exine was observed only in pollen lacking apertures and with thin exine. Variation in the exine ornamentation and thickness could explain the interspecific differences observed for the rates of breakage of the plasma membrane. Our results suggest that pollen wall morphology matters for survival and maintenance of pollen integrity further to volume increase due to hydration. We propose a rationale for future studies that should allow disentangling the contribution of different pollen morphological and physiological features to harmomegathy.  相似文献

Summary The emission of proteins from the pollen wall of Linum grandiflorum stained with Coomassie blue was followed directly in moistened grains as well as in pollen prints. Within the first minute of the grain being moistened exine-borne proteins emerged from both inter-apertural and apertural sites; subsequently, proteins of a different nature were discharged from the apertures only. In a fraction of the grains the release of intine proteins was not preceded by that of exine proteins. Pin and thrum pollen did not differ in terms of mode or site of this protein emission. The presence and emergence of exine proteins from the apertures is explained by the process of infolding of the colpal wall at desiccation and its expansion at rehydration, which causes an initial trapping and subsequent re-exposure of surface materials. This explanation may also account for the occurrence of poral sporophytic proteins in the pollens of many dictoyledons.  相似文献

To investigate the role of aquaporin-mediated water transport during pollen grain germination and tube growth, Arabidopsis thaliana plasma membrane intrinsic proteins (PIPs) were expressed in pollen of Lilium longiflorum (lily). Successful expression of AtPIPs in particle-bombarded lily pollen grains was monitored by co-expression with fluorescent proteins and single-cell RT-PCR, and by measuring the water permeability coefficient (P(os)) in swelling assays using protoplasts prepared from transformed pollen grains and tubes. Expression of AtPIP1;1 and AtPIP1;2 in pollen grains resulted in P(os) values similar to those measured in nontransformed pollen grain protoplasts (6.65 +/- 2.41 microm s(-1)), whereas expression of AtPIP2 significantly increased P(os) (AtPIP2;1, 13.79 +/- 6.38; AtPIP2;2, 10.16 +/- 3.30 microm s(-1)). Transformation with combinations of AtPIP1 and AtPIP2 did not further enhance P(os). Native pollen tube protoplasts showed higher P(os) values (13.23 +/- 4.14 microm s(-1)) than pollen grain protoplasts but expression of AtPIP2;1 (18.85 +/- 7.60 microm s(-1)) did not significantly increase their P(os) values. Expression of none of the tested PIPs had any effect on pollen tube growth rates. The ectopic expression of AtPIP2s in lily pollen increased the water permeability of the plasma membrane in pollen grains, but not in pollen tubes. The measured endogenous water permeability does not limit water uptake during tube growth, but has to be regulated to prevent tube bursting.  相似文献

Summary Dynamics of F-actin organization during activation and germination ofPyrus communis (pear) pollen was examined using rhodaminephalloidin. Prior to activation, the rhodamine-phalloidin labelling pattern appeared as circular profiles in the peripheral cytoplasm of the vegetative cell and as coarse granules around the vegetative nucleus. In activated pollen, parallel arrays of cortical F-actin were aligned circumferentially, along the polar axis in non-apertural areas of the pollen grain, and at 45° to 90° to the polar axis beneath the apertures. Some pollen also showed fluorescent granules or fusiform bodies dispersed throughout the cytoplasm, but as the number of such pollen diminished with prolonged incubation, these are being considered as intermediate patterns. In later stages, the filaments became organized as interapertural bundles traversing the three apertures. However, prior to emergence of the pollen tube, labelling became confined to a single aperture. In germinated pollen grains, actin microfilaments are aligned more or less axially with respect to the axis of the developing pollen tube.The granular labelling pattern seen around the vegetative nucleus prior to pollen activation also became clearly filamentous with pollen activation; this filamentous pattern persisted until germination when it was replaced by cables that aligned longitudinally with respect to the emerging tube axis.The results demonstrate that the organization of actin undergoes considerable changes in the period preceding pollen germination and that microfilament polarization is achieved before pollen germination.  相似文献

Modifications of the pollen grains of Pyrus communis Linneaus that occur during the digestion by Osmia cornuta (Latreille) larvae were studied histochemically. We compared the features of the pollen grains found in the anthers, in the larval cell provisions and in the alimentary canal of the 5th instar larvae. Modifications were already evident in the provisions and consisted of protoplast protrusions through the apertures and a decrease in the number of starch-containing pollen grains. After pollen grains were ingested by the larvae, the protoplast appeared retracted from the pollen wall. Pollen digestion began in the anterior part of the midgut, where we observed: (1) disorganised intine at the apertures; (2) disappearance of DAPI staining of nuclear pollen DNA; (3) fewer pollen grains containing starch than in the anthers; (4) some empty pollen grains. Pollen grains in the proctodeum appeared extremely compressed and crushed. Some grains appeared to be unaffected by the digestive process. We hypothesise that the protrusion of the intine and of the protoplast from the apertures in bee provisions could be considered a kind of pre-treatment necessary to initiate the digestion process in the larval alimentary canal.  相似文献

To study microtubule organization in germinating pear (Pyrus communis L., cv., Bartlett) pollen, we removed the pollen wall by freeze-fracturing before treating the resultant pollen protoplasts by conventional immunofluorescence procedures. Results reveal that axial bundles of microtubules are present in the generative cell of both inactivated and activated pollen grains. Microtubules are not present in the vegetative cells of inactivated pollen, but they are present in the vegetative cells of activated pollen grains. Microtubule nucleation occurs in the vegetative cell cortex. Subsequently, the microtubules grow as branching arrays through most of the vegetative cell cortex except at the apertures where they form localized converging or criss-cross patterns. Eventually, in a germinated pollen grain, the microtubules form network-like arrays through most of the pollen grain and a collar of short arrays at the base of the pollen tube. It is suggested that the role of vegetative cell microtubules in pollen germination is indirect through their mediation of the conformational changes in actin organization that are essential for pollen germination.  相似文献

从发育的角度研究了中国特有单种属DichotomanthesKurz及与其系统学研究有关的外类群Prinsepiautilis的花粉形态 ,扫描电镜观察显示Dichotomanthes花粉粒自脱离四分体胼胝质膜开始至成熟二核花粉粒不同发育时期 ,花粉形态和外壁纹饰未见变化 ,仅花粉体积随成熟度增加而有所增大。而Prinsepiautilis ,其花粉粒刚脱离四分体时形状和成熟花粉明显不同 ,成熟花粉极面观为三裂圆形 ,赤道面观为圆形 ,外壁具清晰的平行条纹 ,但幼嫩花粉粒的形状很特别 ,极面观为深三裂圆形 ,赤道面观亦见花粉在两条沟之间下陷而沟部外突 ,明显为角萌发孔花粉 ,且花粉体积较成熟者小 ,而外壁纹饰同成熟者相比无根本性差异。前述两种植物花粉在不同成熟期体积有明显差异 ,而外壁纹饰在不同成熟期不存在质的变化并相对稳定 ,说明花粉外壁纹饰这一性状在蔷薇科中具有较为重要的分类学意义。DichotomanthesKurz具典型Rosaceae花粉的三孔沟结构 ,外壁具条纹 -穴状纹饰。将其孢粉学特征同Rosaceae 4个亚科有关类群的同类资料相比较 ,并结合其它形态解剖与细胞学等研究结果 ,支持将Dichotoman thes置入Maloideae下而不赞同将其另立亚科或置于Prunoideae之下。此外 ,由于Prinsepiautilis的花粉在其发育初期具角萌发孔花粉 ,与Cunoniacea  相似文献

Measurements and observations of pollen grains of varying structure before and after dehiscence of the anther sac illustrate large magnitudes of harmomegathic changes in volume and shape. Such changes are compared for colpate, porate, and colporate grains, and it is suggested that the nature of the harmomegathal action may serve to distinguish colpi or furrows from pores. Consideration of the requirement to allow harmomegathy while preventing wall collapse may provide at least partial explanation for evolution of the internal structure and external sculpturing of pollen grain walls.  相似文献

Quantitative studies of the adhesion of pollen grains to the stigma in Brassica oleracea revealed that self-pollen is initially less firmly bound than cross-pollen. The pollen grain tryphine, believed to be important in the adhesion process, has been shown to differ in mobility following self- and cross-pollination when observed using fluorescent probes. The hydration of the pollen grains has been investigated in vitro by measuring the changes in shape, volume and fresh weight of the imbibing grains. Whilst little change in volume could be detected there was a considerable increase in fresh weight together with a change of shape. The structure of the massulae composing the pollinium ofLoroglossum hircinum was studied before pollination and 12 and 24 hours afterwards. The grains are grouped in tetrads closely packed in massulae. The exine is only present on the outside of the massulae. The intine consists of two layers: a compact layer surrounding the pollen grain and a looser layer surrounding the pollen grain and a looser layer surrounding the tetrad. Twelve hours after pollination, pollen volume and the space between the tetrads increase due to vacuolization. Twenty-four hours after pollination, pollen volume and tetrad spacing are higher due to vacuolization and some grains have emitted pollen tubes. Pollen growth due to vacuole formation, and the absence of common walls between adjacent tetrads lead to crumbling of the massulae. The mature pollen grain does not have apertures: the site of pollen tube emission is determined after pollination. The first grains to germinate are those in the centre of the massula. The vegetative cell nucleus is the first to enter the pollen tube; the generative cell elongates and undergoes the second haploid mitosis shortly after entering the pollen tube.  相似文献

Aperture morphology of tetrad pollen of Epilobium luteum (Onagraceae: Epilobieae) from three Alaskan collections is highly variable. The first collection appears to lack apertures altogether and is presumed to consist of immature pollen gains in a genus known to achieve mature size before the apertures become distinctly protruding. A second collection has tetrads with 3- and 4-apertured grains, the apertures in the latter are often irregularly spaced and not in apposition with the apertures of neighboring members. The third collection consists of the more typical 3-apertured members that characterize the majority of Epilobium pollen grains. In all of these collections individual pollen grains (monads) are interspersed among the tetrads. The variations in the number of apertures emphasize the importance of having a comprehensive understanding of the stage of development of the pollen (taxon) examined when describing pollen collections. In the first collection this would mean the recognition that in Onagraceae apertures occur in the later stages of microspore ontogeny. In the latter two collections a thorough background of the literature of the pollen morphology on this largest Onagraceae taxon is useful for the understanding of the significance of a range of aperture numbers on Epilobium pollen grains.  相似文献

The trees group showed the highest quantity of pollen as well as taxa diversity, although its peak period was in the dry season. The weeds and grasses emitted a larger quantity of pollen in the rainy season. The dominant taxa wereAlnus, Casuarina, Compositae and Gramineae. As for their relation with meteorological parameters, we found that the increase of pollen concentration was related to high temperatures, low relative humidity and high wind speed, the latter causing an increase of airborne pollen with no dilution at all. In Japan, the problems of Sugi (Cryptomeria japonica)-pollinosis have been much discussed in recent papers and journals. The author made an investigation on the airborne pollen grains from a scientific standpoint in connection with the incidence of pollinosis. By using the Cascade Impactor the author collected 600 liters of air sample a day, at the roof of the Pharmaceutical Science Building, Toho University, Funabashi, Chiba, Japan. Each air sampling for 2 hrs was repeated three times a week since 1969. The airborne pollen grains in each sample were counted under the microscope to identify the pollen types. The pollen grains were classified into six types according to the classification of pollen grains in Japan established by lkuse. The accumulated number of pollen of each species was analysed statistically by circular plot, Weibull plot, Edwards plot and semi-logarithmic plot. In this report mainly 3B type of pollen grains (Cryptomeria, Chamaecyparis), collected in the last three years are described. The total number of pollen grains in 1988 (2820 grains) was doubled in quantity as compared with that in 1987 (1177 grains) and in 1989 (1121 grains). The 3B type of pollen grains in 1988 (1450 grains) was 3.4 times as many as in 1987 (397 grains), and 12.9 times as in 1989 (112 grains). The 3B type of pollen grains represented the major portion of total pollen, and influenced the annual amount and the dispersal period.  相似文献

The pollen grain morphology of 30 species of 27 genera from the four subfamilies of the Cyperaceae have been studied with LM and SEM. Several methods were tested in order to find the optimal treatment for the delicate Cyperaceae pollen grains. For LM, treatment with wetting agent Agepon and KOH yielded the best results, while critical point drying (CPD) after fixation with alcohol proved to be the best method for studying pollen grains of Cyperaceae with SEM. Our results show that the Mapanioideae have a pollen grain type (only one distal ulcus) clearly different from the other Cyperaceae. Representatives of the examined Sclerioideae and Caricoideae show a similar pollen grain type (mostly one distal ulcus and three lateral pores) while in the Cyperoideae different pollen grain types are found.  相似文献

Aerobiological study of Fagaceae pollen in the middle-west of Spain   总被引:1,自引:0,他引:1  
The concentration of airborne Fagaceae pollen in Salamanca and the correlations with some meteorological parameters have been examined. Castanea and Quercus pollen grains were collected from 1998 to 2004 using a Burkard spore trap. No pollen grains of Fagus were found. The main pollen season took place in April and May for Quercus and in June and July for Castanea. Yearly variations on these dates could be related to the influence of meteorological factors such as rainfall, temperature, or dominant winds. The highest values appeared in the year 2004 for both taxa. The Fagaceae airborne content was mainly due to Quercus pollen, Castanea having a scarce pollen content in the city of Salamanca. The highest counts of Fagaceae pollen grains were found from mid May to early June due to the pollen behavior of oaks. The cumulative counts varied over the years, with a mean value of 2,384 pollen grains, a highest total of 6,036 in 2004 and a lowest total of 954 in 2001. No cyclic variations were observed. Daily pollen concentrations presented positive correlation with temperature, negative with relative humidity and slightly negative with rainfall using Spearman's correlation coefficients, only in the case of Castanea, because the particular hourly distribution of rainfall during the spring might affect Quercus airborne pollen.  相似文献

The phenology of pollen release and pollen capture by Pistacia vera was studied in the field and laboratory respectively. Inflorescences of Pistacia vera were examined in a wind tunnel to determine whether the behavior of airborne conspecific pollen around receptive flowers differed as a result of changes in the shape and size of the inflorescence. In addition, the behavior of unclumped (single) and clumped pollen grains was studied to determine differences in the probability of their capture. Wind speeds within a commercial orchard during pollen shedding averaged 0.9–2.2 m/sec and atmospheric pollen concentrations were highest between 0900–1100 hr MST. Each of three stages in inflorescence development (defined on the basis of the number of exserted stigmas) was examined under identical ambient airflow conditions with equal concentrations of airborne pollen (1,000 grains/m3). The general pattern of pollen grain motion involves direct inertial collision by windward surfaces and by sedimentation of pollen onto leeward surfaces; clumped pollen rarely sedimented onto leeward surfaces. Small changes in ambient wind speed (0.5 m/sec to 1.0 m/sec) produced significant changes in the pattern of pollen motion around inflorescences and altered the number of pollen grains captured by leeward surfaces. Thus, wind pollination in P. vera is affected both by windspeed and by the shape or size of flower clusters. Differences in the behavior of clumped and unclumped pollen result from their inertial properties and responsiveness to local changes in the direction and speed of airflow. Unclumped pollen has a higher probability of being captured by leeward surfaces. The apparent insensitivity of pollen motion to differences in inflorescence size may ensure equitable pollination during the acropetal development of flowers.  相似文献

Pollen aperture polymorphism is studied in Viola diversifolia, where all plants produce three- and four-apertured pollen grains. We tested whether there are genetic differences among plants for the proportions of the different pollen morphs, and whether the morphs differ in gametophytic performance. Results show that the more apertures a pollen grain has, the more quickly it germinates but that few-apertured pollen grains have faster growing pollen tubes and longer life expectancies. The proportions of the different pollen morphs, together with pollen tube growth rates, may be inherited traits based on differences among maternal families. These results suggest that the different pollen morphs are favored in different pollination ecology situations. The production of several pollen morphs by the same individual could therefore be evolutionarily advantageous.  相似文献
