Seven polymorphic microsatellite loci have been characterized for investigating population structure in the patchily distributed herb Begonia sutherlandii. Two loci (BSU3 and BSU4) exhibited population specific null alleles; primer redesign and allele sequencing for one of these loci showed two transition mutations in the original primer site. Two loci exhibited imperfect repeat polymorphisms due to single base pair indels in the flanking region (locus BSU6) and in the microsatellite region itself (BSU7). Transversion mutations were also found in the microsatellite region of locus BSU7. The remaining three loci amplified in all individuals tested and appeared to conform to a simple stepwise mutation pattern.  相似文献

由于丽格海棠通过扦插获得的量有限,进口种子的价格又很昂贵,所以以它的叶、茎、花茎、叶柄等营养器官作为外植体,进行组织培养来快速获得丽格海棠。通过诱导芽分化和诱导愈伤两种途径,研究不同植物生长调节剂及浓度,对外植体萌发和生根的影响。研究表明基本培养基为MS,最佳分化培养基为6-BA1.00mg/L NAA0.10mg/L;最佳生根培养基为1/2MS NAA0.mg/L;花土:沙子=1:1,是丽格海棠最佳的驯化移栽基质,在该基质中生根苗移栽成活率达90%。  相似文献

The chromosome numbers of four Chinese Begonia species were counted for the first time as follows: 2 n  = 22 in B. rubropunctata , 2 n  = 18 in B. purpureofolia , 2 n  = 44 in B. pedatifida , and 2 n  = 22 in B. villifolia . The first two species had relatively large chromosomes with early condensed chromatin in the distal and interstitial or proximal regions, whereas the last two had small chromosomes with early condensed chromatin in the proximal regions. Although they have been reduced in the literature to synonyms of B. pedatifida and B. villifolia , respectively, B. rubropunctata and B. purpureofolia should be treated as distinct species because of the differences in chromosome numbers and other chromosome features.  © 2007 The Linnean Society of London, Botanical Journal of the Linnean Society , 2007, 155 , 513–517.  相似文献

Begonias are hyper-diverse and important horticultural plants. Six polymorphic microsatellite markers were developed from CT- and GT-enriched libraries of Begonia maxwelliana. The number of alleles per locus ranged from 2 to 12 and the observed heterozygosity ranged from 0.036 to 0.813. Null alleles were detected in one locus (Bma161) after Bonferroni correction. All the six markers were amplifiable in 23 selected Begonia species with the success rates of 17–100%. On average, species of the same section as B. maxwelliana (i.e. sect. Platycentrum) yielded higher transferability (91%). These markers will be useful for population genetic studies of the genus Begonia.  相似文献

Cytokinin activity has been obtained in ethanol extracts of Begonia and Bryophyllum plants. Begonia x cheimantha Everett yielded 30 to 300 μg kinetin equivalents per kg of fresh leaves in the tobacco callus bioassay. Short day conditions appeared to increase the extractable cytokinin content in the tissue. Purification by fractionation on Dowex 50 H+ columns followed by organic solvent extraction, silver precipitation, and repeated paper chromatography yielded an apparently homogeneous product which accounted for most of the activity in the Begonia extracts. It was indistinguishable from zeatin in chromatograms developed with six solvent systems. Other cytokinin active fractions were also obtained from both Begonia and Bryophyllum. Crystalline picrate preparations of active products were insufficient for identification by mass spectrometry.  相似文献

In vitro regeneration of four Begonia genotypes, B. semperflorens, B. rex, B.×elatior, and hybrid of Begonia with unknown parents Tiger was carried out starting from leaf and petiole segments as explants. Five Murashige and Skoog's derived media were tested, three of them supplemented with -naphthaleneacetic acid (NAA) and 6-benzyladenine (BA), and the other two with NAA and kinetin (KIN) in different concentrations. Shoot regeneration was preferentially induced on the BA containing media, quantitative differences being observed among explants and genotypes.  相似文献

利用播种、扦插和组织培养 3种方式繁殖变色秋海棠均获得成功。播种繁殖的发芽率约 60 % ;在 3种不同的叶插繁殖中 ,以锥形叶插成活率最高 ;组织培养以叶片为外植体、MS+BA1 +NAA0 .1固体发芽培养基较好 ,从外植体直接分化出芽原基和新芽 ,属于器官型再生方式。利用密闭容器栽培法或类似此种方法栽培变色秋海棠 ,取得较好效果。  相似文献

Two species of Begonia L. (Begoniaceae), viz. Begonia demissa Craib and Begonia subperfoliata Parish ex Kurz are reported as new records in China, and the two species assigned to Begonia sect. Parvibegonia A. DC. and Begonia sect. Diploclinium (Wight) A. DC. respectively. The type locality of Begonia demissa is in Thailand and of Begonia subperfoliata in Myanmar. The distribution of Begonia demissa is now extended to Mengyuan, Mengla County of Xishuangbanna Dai Autonomous Prefecture and that of Begonia subperfoliata to Liushun Community, Pu’er City, both in Yunnan province, China.  相似文献

The primary accumulation of oxalate in the early seedling stagesaccompanies the synthesis of both protein and reserve carbohydrate.A further accumulation accompanies the normal growth of theyoung expanding leaf. Free oxalic acid accounts for most ofthe total oxalate content at the various stages. The form in which nitrogen is supplied to the young seedling(ammonium nitrogen or nitrate nitrogen) markedly affects bothgrowth and the amount of oxalate formed. More oxalate and poorergrowth is found in plants grown on ammonium as a sole sourceof nitrogen. In the mature leaf the oxalate content remains constant undera wide variety of different conditions. No enzyme systems could be detected which might be active inthe formation of oxalate. It is concluded that oxalic acid is only formed during rapidgrowth in Begonia, and that once formed it takes little furtherpart in metabolism.  相似文献
