Slow-folding kinetics of ribonuclease-A by volume change and circular dichroism: evidence for two independent reactions. 下载免费PDF全文
The slow refolding of guanidine-HCl-denatured ribonuclease-A was studied by volume change and by kinetic CD at 222 and 276 nm. Dilatometric measurements revealed that on refolding there is a fast volume change of +232 mL/mol of protein. This is followed by a very slow nonexponential change that takes about 25 min to reach equilibrium. By adding varying amounts of (NH4)2SO4, the slow volume change curve was resolved into 2 concurrent reactions. The faster of the 2 slow events entails a negative volume change of -64 mL/mol of protein and appears to arise from proline isomerization. The slower process, attended by a positive change of +53 mL/mol of protein, has properties consistent with the "XY" reaction of Lin and Brands (1983, Biochemistry 22:563-573). This reaction is so named because the conformational nature of neither its initial (Y) nor its final state (X) is known; the transition is characterized solely by its absorbance and fluorescence kinetics. These are the first direct physical measures attributable to the "XY" process. The early formation of a compact structure in the event responsible for the rapid +232-mL/mol volume change, however, is consistent with the sequential model of folding (Cook KH, Schmid FX, Baldwin RL, 1979, Proc Natl Acad Sci USA 76:6157-6161; Kim PS, Baldwin RL, 1980, Biochemistry 19:6124-6129). The usefulness of volume change measurements as a method of detecting structural rearrangements was confirmed by finding agreement between time constants obtained from parallel volume change and kinetic CD experiments. A method is described for frequent sequential blood volume estimation in baboons using 32P for red cell volume measurements and 125I-albumin for simultaneous plasma volume measurements. Values for red cell, plasma, and total blood volumes are reported. Close correlations of the volumes to bodyweight were demonstrated. The partial molar volume (PMV) change associated with the pressure-induced structural transition of ubiquitin is analyzed by the three-dimensional reference interaction site model (3D-RISM) theory of molecular solvation. The theory predicts that the PMV decreases upon the structural transition, which is consistent with the experimental observation. The volume decomposition analysis demonstrates that the PMV reduction is primarily caused by the decrease in the volume of structural voids in the protein, which is partially canceled by the volume expansion due to the hydration effects. It is found from further analysis that the PMV reduction is ascribed substantially to the penetration of water molecules into a specific part of the protein. Based on the thermodynamic relation, this result implies that the water penetration causes the pressure-induced structural transition. It supports the water penetration model of pressure denaturation of proteins proposed earlier. 相似文献

The structural origin of the decrease in system volume upon protein denaturation by pressure has remained a puzzle for decades. This negative volume change upon unfolding is assumed to arise globally from more intimate interactions between the polypeptide chain and water, including electrostriction of buried charges that become exposed upon unfolding, hydration of the polypeptide backbone and amino acid side chains and elimination of packing defects and internal void volumes upon unfolding of the chain. However, the relative signs and magnitudes of each of these contributing factors have not been experimentally determined. Our laboratory has probed the fundamental basis for the volume change upon unfolding of staphylococcal nuclease (Snase) using variable solution conditions and point mutants of Snase (Royer CA et al., 1993, Biochemistry 32:5222-5232; Frye KJ et al., 1996, Biochemistry 35:10234-10239). Our prior results indicate that for Snase, neither electrostriction nor polar or nonpolar hydration contributes significantly to the value of the volume change of unfolding. In the present work, we investigate the pressure induced unfolding of three point mutants of Snase in which internal cavity size is altered. The experimentally determined volume changes of unfolding for the mutants suggest that loss of internal void volume upon unfolding represents the major contributing factor to the value of the volume change of Snase unfolding. 相似文献

Volume changes among the unfolded (U), native (N), and molten globule (MG) conformations of horse heart ferricytochrome c have been measured. U to N (pH 2 to pH 7) was determined in the absence of added salt to be -136 +/- 5 mL/mol protein. U to MG (pH 2, no added salt to pH 2, 0.5 M KCl) yielded + 100 +/- 6 mL/mol. MG to N was broken into two steps, N to NClx at pH 7 by addition of buffered KCl to buffered protein lacking added salt (NClx = N interacting with an unknown number, X, of chloride ions), and MG to NClx by jumping MG at pH 2 in 0.5 M KCl to pH7 at the same salt concentration. The delta V of N to NClx was -30.9 +/- 1.4 mL/mol protein, whereas MG to NClx entailed a delta V of -235 +/- 6 mL/mol. Within experimental error, the results add up to zero for a complete thermodynamic cycle. We believe this to be the first volumetric cycle to have been measured for the conformational transitions of a protein. The results are discussed in terms of hydration contributions from deprotonation of the protein, other hydration effects, and the formation and/or enlargement of packing defects in the protein's tertiary structure during the steps of folding. 相似文献

The number of eggs laid into its insect-egg host by Trichogramma minutum Riley (Hymenoptera: Chalcidoidea: Trichogrammatidae) depends on the volume of its lepidopterous host. The wasps allocated fewer progeny into Manduca sexta (L.) (Sphingidae) egg hosts partially embedded in the substrate than into fully exposed hosts, both in daylight and in total darkness. Since the hosts are of identical diameter and surface chemistry, the mechanism of volume determination is neither chemical nor visual, but essentially mechanosensory, based on accessible surface area. Before oviposition, the wasp examines the host surface by walking over it, turning at the edge, and drumming with its antennae. The number of edge turns (Nt) made by wasps when they examined glass beads presented as model hosts was inversely dependent on surface area, but independent of diameter. Conversely, the examination time (Te) was directly dependent on diameter, but independent of surface area.A mechanism is proposed for host volume measurement. While the wasp examines a potential host, the relative curvature (and hence diameter) is measured, possibly by antennal angle. If this measure is within an acceptable range, the wasp initiates its drumming walk. The measure is also used to set Tc and to establish an estimate of a maximum possible volume (and hence progeny number) to the host object. With Tc fixed, the value for Nt depends on accessible surface area, the wasp turning more often on restricted surfaces. Nt allows the wasp to correct the estimate of host volume such that an appropriate number of eggs is laid in non-spherical hosts. The use of curvature to set Te increases the accuracy of the wasp's measure of Nt, while also improving its coverage of the surface. The use of curvature and surface area allows Trichogramma to optimize its progeny allocation for a variety of host sizes and shapes.
Die bestimmung des wirtsvolumen durch die parasitische wespe Trichogramma minutum: Die rolle von krümmung und oberfläche
Zusammenfassung Bei dem Insekten-Eiparasiten Trichogramma minutum (Hymenoptera: Trichogrammatidae) hängt die Anzahl der abgelegten Eier vom Volumen der Wirtseier ab. Es wurden Untersuchungen zur Bestimmung der sensorischen Grundlagen dieses Verhaltens unternommen. Die Wespe weist sowohl im Tageslicht, als auch in totaler Dunkelheit jenen Manduca sexta (L.) (Lepidoptera: Sphingidae) Eiwirten (Diameter c. 1.3 mm) weniger eigene Eier zu, die teils (30%) in Substrat eingebettet sind, als solchen, die ganz herausragen. Da die Wirte einen identischen Radius und gleichen Oberflächengeruch haben, kann der Mechanismus der Volumenbestimmung weder chemisch noch optisch sein, sondern ist dem Wesen nach mechanosensorisch, und von der zugänglichen Oberfläche abhängig. Vor de Eiablage untersucht die Wespe die Wirtsoberfläche, indem sie darüber läuft, an den Ecken umkehrt und mit ihren Antennen trommelt. In Experimenten mit Glaskugelmodellen war die Anzahl der von der Wespe gemachten Wendungen an den Ecken (Nt) umgekehrt proportional zur Oberfläche, aber unabhängig vom Radius. Im Gegensatz dazu, war die Untersuchungszeit (Te) direkt proportional zum Radius, aber unabhängig von der Oberfläche.Ein Mechanismus für die Volumenmessung wird vorgeschlagen. Gleich beim Besteigen des potentialen Wirtes wird die relative Kurve (und infolgedessen der Radius), wahrscheinlich durch den Antennenwinkel gemessen. Wenn diese Messung innerhalb einer bestimmten Spanne liegt, beginnt die Wespe ihren Trommelgang. Die Messung wird auch dazu benutzt, ein maximales Volumen (und folglich die Anzahl der Nachkommen) des Wirtes und eine maximale Untersuchungzeit (Te) zu erzielen. Wird Te konstantgehalten, hängt die Anzahl der Wendungen (Nt) von der zugänglichen Oberfläche ab, wobei die Wespe öfter bei kleineren Oberflächen wendet. Die Anzahl der Wendungen gestattet der Wespe, die ursprüngliche Einschätzung des Wirtsvolumens so zu verändern, daß eine angemessene Anzahl von Eiern in nicht kugelige Wirte gelegt wird. Die Anwendung von kurven-und oberflächenabhängigen Messungen ermöglicht eine optimale Zuweisung von Nachkommen für eine Anzahl von unterschiedlichen Wirtsgrössen und-formen. Die Oberflächenmessung gestattet eine bessere Volumenbestimmung von nicht-kugeligen Objekten. Die gezeigte Abhängigkeit von der Oberfläche ist wahrscheinlich auch eine Anpassung an Wirte, die ihre Eier in Gruppen ablegen. Unter solchen Bedingungen von grosser Wirtsdichte ist es günstiger für die Wespe, weniger Eier in jeden Wirt der Gruppe zu legen, weil diese Verteilung zu grösseren, fruchtbareren Nachkommen führt.相似文献

After standing for 1h, ten subjects (7 male, 3 female) assumed a supine position for a further hour. Whole body bioelectrical impedance increased progressively during the hour spent in the supine position: after 60 min supine the increase was 13(6 to 32). Blood and plasma volumes, estimated from haematocrit and haemoglobin concentration, increased by 8.0(6.7 to 12.4)% and 16.7(12.3 to 20.8)% (median(range)) respectively after 60 min supine. Serum potassium concentration had fallen after 10 min supine (4.1(0.1)mmol 1–1; mean(SEM)) relative to the standing value (4.6(0.1)mmol 1–1) and was unchanged thereafter. Serum osmolality (P=0.991) and sodium (P=1.000) and chloride (P=0.998) concentrations remained unchanged throughout the study. The fall in serum potassium concentration in the supine position does not appear to be a simple dilutional effect consequent upon increases in blood and plasma volume as there was no effect of postural change on serum sodium or chloride concentrations. 相似文献

In many clinical situations, it is crucial to determine circulating blood volume (BV) easily and to repeat this measurement. The Dye DensitoGram Analyzer® (DDG, Nihon Kohden Corp) measures semi-automatically BV, using an injection of IndoCyanine Green (ICG, 10 mg), and avoiding intermittent blood samples. The DDG was used during a 90-day microgravity simulation by Head-Down-Tilt bed rest (HDT) to measure BV and compared with the calculation of the plasma volume (PV) variations according to Dill and Costill's formula (DC). Seventeen healthy volunteers were included: 8 control subjects (Co) and 9 subjects submitted to a resistive exercise counter-measure (CM). Measurements were performed, one day before HDT, on days 3 and 90 of HDT and on day 9 after HDT. A double measurement of the BV was performed to assess the repeatability of this method. On the last day of HDT a significant decrease (p < 0.05) in the PV was noted with the DDG (Co: − 12.3 ± 5.7%, CM: − 9.0 ± 5.3%) and DC; (Co: − 4.7 ± 1.8%, CM: − 6.8 ± 2.5%). A good repeatability of the technique was shown with a low intrasubjects coefficient of variation (4.95 ± 0.95%) and an acceptable intersubjects coefficient of variation (15.30 ± 1.13%). No correlation was noted between DDG and DC (r2 = 0.27). The DDG gives a good repeatability, not affected by the microgravity exposure. Thanks to its capacity to measure accurately the BV within 7-10 min, this device presents major advantages for clinical use and research purpose. 相似文献

The effects of pressure on the sol-gel transition of κ- and ι-carrageenans were studied in KCl solutions under high pressures up to 3000 kg/cm2. The carrageenan gels were destabilized by pressure: the pressure depression of melting temperature, (dT/dP)m, was ?5.7 × 10?3 and ?4.0 × 10?3 K cm2/kg independent of KCl concentration for κ- and ι-carrageenans, respectively. The enthalpy, entropy and volume changes accompanying the gel formation were calculated from the Eldridge-Ferry's plots and the Clausius-Clapeyron equation. The volume change per unit cross-link (two disaccharide residues) was estimated to be (2.5 ~ 4.9) and (1.7 ~ 3.4) ml/mol for κ- and ι-carrageenans, respectively. The compressibility of both carrageenan molecules appeared to be larger by (1.6 ~ 2.6) × 10?12 (κ-form) and by (0.8 ~ 1.3) × 10?12cm2/dyn (i-form) in gel state as compared with in sol state These increases in volume and compressibility on gelation were attributed to a reduction of water of hydration from the carrageenan molecules, which is mainly due to a replacement of the polymer-water hydrogen bond by the polymer-polymer hydrogen bond. These results seemed not inconsistent with the idea that a double helix structure of carrageenan gels may persist in solution as well as in the solid state. 相似文献

Background
Reduced gas transfer in patients with pulmonary arterial hypertension (PAH) is traditionally attributed to remodeling and progressive loss of pulmonary arterial vasculature that results in decreased capillary blood volume available for gas exchange.Methods
We tested this hypothesis by determination of lung diffusing capacity (DL) and its components, the alveolar capillary membrane diffusing capacity (Dm) and lung capillary blood volume (Vc) in 28 individuals with PAH in comparison to 41 healthy individuals, and in 19 PAH patients over time. Using single breath simultaneous measure of diffusion of carbon monoxide (DLCO) and nitric oxide (DLNO), DL and Dm were respectively determined, and Vc calculated. Dm and Vc were evaluated over time in relation to standard clinical indicators of disease severity, including brain natriuretic peptide (BNP), 6-minute walk distance (6MWD) and right ventricular systolic pressure (RVSP) by echocardiography.Results
Both DLCO and DLNO were reduced in PAH as compared to controls and the lower DL in PAH was due to loss of both Dm and Vc (all p < 0.01). While DLCO of PAH patients did not change over time, DLNO decreased by 24 ml/min/mmHg/year (p = 0.01). Consequently, Dm decreased and Vc tended to increase over time, which led to deterioration of the Dm/Vc ratio, a measure of alveolar-capillary membrane functional efficiency without changes in clinical markers.Conclusions
The findings indicate that lower than normal gas transfer in PAH is due to loss of both Dm and Vc, but that deterioration of Dm/Vc over time is related to worsening membrane diffusion. 相似文献13.

Summary The pathway for movement of chloride ions across frog skin is not well understood. Mitochondria-rich (MR) cells have been proposed as the route for chloride across the skin. To test this hypothesis we studied the MR cells of the skin of the frog,Rana pipiens, by quantitative light microscopic determination of cell volume. MR cell volume was influenced by changes in the chloride concentration or osmolality of the outside bathing solution. MR cells shrank about 23% when all chloride was removed from the outside (mucosal) bathing solution. MR cells were also shown to be responsive to changes in the osmolality of either the mucosal or serosal bath. Osmotically-induced swelling caused by dilution of the serosal bath resulted in volume regulatory decrease. These results are consistent with the hypothesis that MR cells constitute the pathway for chloride movement across frog skin. 相似文献

At 21°C the first egg generation takes 6 days to develop. During this period the oöcyte volume increases by a factor of ×5000, and compared with an oögonium the growth factor amounts to ×100,000. The growth rate of the oöcyte increases with each stage of oögenesis, until at stage 5 it reaches 2.5 × 106 μm3/hr. About 35% of the substance stored in the oöcyte originates from the nurse chamber. 30% of the volume is formed in the stage where the oöcyte synthesizes almost exclusively glycogen. Mechanical compression of cartilage is associated with a rise in the interstitial osmotic pressure, which can alter cell volume and activate volume recovery pathways. One of the early events implicated in regulatory volume changes and mechanotransduction is an increase of intracellular calcium ion ([Ca2+]i). In this study, we tested the hypothesis that osmotic stress initiates intracellular Ca2+ signaling in chondrocytes. Using laser scanning microscopy and digital image processing, [Ca2+]i and cell volume were monitored in chondrocytes exposed to hyper-osmotic solutions. Control experiments showed that exposure to hyper-osmotic solution caused significant decreases in cell volume as well as transient increases in [Ca2+]i. The initial peak in [Ca2+]i was generally followed by decaying oscillations. Pretreatment with gadolinium, a non-specific blocker of mechanosensitive ion channels, inhibited this [Ca2+]i increase. Calcium-free media eliminated [Ca2+]i increases in all cases. Pretreatment with U73122, thapsigargin, or heparin (blockers of the inositol phosphate pathway), or pertussis toxin (a blocker of G-proteins) significantly decreased the percentage of cells responding to osmotic stress and nearly abolished all oscillations. Cell volume decreased with hyper-osmotic stress and recovered towards baseline levels throughout the duration of the control experiments. The peak volume change with 550 mOsm osmotic stress, as well as the percent recovery of cell volume, was dependent on [Ca2+]i. These findings indicate that osmotic stress causes significant volume change in chondrocytes and may activate an intracellular second messenger signal by inducing transient increases in [Ca2+]i. 相似文献

To assess the relationship between blood volume (BV) and the reduction in plasma volume (PV) during exercise in individual variations, we measured BV and changes in PV in thirteen male volunteers during treadmill exercise until exhaustion. The lactate threshold (LT), as a predictor of aerobic exercise capacity, was calculated from the exercise intensity at the point of plasma lactate concentration buildup to 4 mmol. The relationship of peak VO2 with BV indicated a significant positive correlation. The strong positive relation between the shifts in PV and total PV, and resulted in a maintenance of the circulating BV. 相似文献

Origins of blood volume change due to glutamatergic synaptic activity at astrocytes abutting on arteriolar smooth muscle cells 总被引:1,自引:0,他引:1
The cellular mechanisms that couple activity of glutamatergic synapses with changes in blood flow, measured by a variety of techniques including the BOLD signal, have not previously been modelled. Here we provide such a model, that successfully accounts for the main observed changes in blood flow in both visual cortex and somatosensory cortex following their stimulation by high-contrast drifting grating or by single whisker stimulation, respectively. Coupling from glutamatergic synapses to smooth muscle cells of arterioles is effected by astrocytes releasing epoxyeicosatrienoic acids (EETs) onto them, following glutamate stimulation of the astrocyte. Coupling of EETs to the smooth muscle of arterioles is by means of potassium channels in their membranes, leading to hyperpolarization, relaxation and hence an increase in blood flow. This model predicts a linear increase in blood flow with increasing numbers of activated astrocytes, but a non-linear increase with increasing glutamate release. 相似文献

基于RS与GIS技术,以遥感影像数据、土地利用数据、森林资源二类调查数据为主要数据源,采用逐步回归法建立森林蓄积量定量估测模型。根据"蓄积量-生物量-碳储量"推算方法,对福州市森林植被碳储量和碳密度进行估算。建立福州市土地利用转移矩阵,分析2000—2010年土地利用变化影响下的福州市森林碳储量变化特征。结果表明:(1)根据不同的森林类型,即常绿阔叶林、常绿针叶林、针阔混交林分别建立的多元线性回归模型修正决定系数分别为0.599、0.679、0.694,通过模型适用性检验和精度验证。(2)2000年、2010年福州市森林植被碳储量总量分别为12.499Tg、12.642Tg,植被碳密度分别为18.694、18.708 t/hm~2,森林植被碳储量增加了1.430×10~5t。(3)福州市闽清县、永泰县、闽侯县的森林植被碳密度常年保持较高水平,并呈现出增长趋势;罗源县、长乐市、连江县森林植被碳密度较低,并呈现下降趋势。(4)2000—2010年,灌木和耕地是主要土地利用类型转出者,森林和建设用地是主要土地利用类型转入者。森林主要由灌木和耕地转化,主要向建设用地、耕地进行转化。由于土地利用变化,10年间福州市总碳储量减少了1.711×10~4t,其中土壤碳储量减少2.230×10~3t,植被碳储量减少1.489×10~4t。 相似文献
