Diets low in methionine extend lifespan of rodents, though through unknown mechanisms. Glycine can mitigate methionine toxicity, and a small prior study has suggested that supplemental glycine could extend lifespan of Fischer 344 rats. We therefore evaluated the effects of an 8% glycine diet on lifespan and pathology of genetically heterogeneous mice in the context of the Interventions Testing Program. Elevated glycine led to a small (4%–6%) but statistically significant lifespan increase, as well as an increase in maximum lifespan, in both males (p = 0.002) and females (p < 0.001). Pooling across sex, glycine increased lifespan at each of the three independent sites, with significance at p = 0.01, 0.053, and 0.03, respectively. Glycine‐supplemented females were lighter than controls, but there was no effect on weight in males. End‐of‐life necropsies suggested that glycine‐treated mice were less likely than controls to die of pulmonary adenocarcinoma (p = 0.03). Of the 40 varieties of incidental pathology evaluated in these mice, none were increased to a significant degree by the glycine‐supplemented diet. In parallel analyses of the same cohort, we found no benefits from TM5441 (an inhibitor of PAI‐1, the primary inhibitor of tissue and urokinase plasminogen activators), inulin (a source of soluble fiber), or aspirin at either of two doses. Our glycine results strengthen the idea that modulation of dietary amino acid levels can increase healthy lifespan in mice, and provide a foundation for further investigation of dietary effects on aging and late‐life diseases.  相似文献

Methods: Elders who had immigrated to Melbourne and Vancouver within the previous 15 years were recruited from local community centres and assigned to focus groups of 5–7 participants in Vancouver (4 groups) or Melbourne (5 groups). Results: Following an iterative process of thematic analysis, the discussions revealed that immigrants care about the comfort and appearance of their teeth, and they value Western dentistry as a supplement to traditional remedies, but they have difficulty getting culturally sensitive information about oral health care. Accessing dentistry, they explained, is distressing because of language problems and financial costs that impose on their children. Consequently, many immigrants obtain dental treatment in China when they return for occasional visits. They felt that separation of dentistry from national health care programmes in Canada and Australia disregards natural links between oral health and general health. Conclusions: The similarity of concerns in both cities suggests that dissemination of information and availability of services are the important themes influencing oral health, and that, beliefs developed over a lifetime play an important role in interpreting oral health in the host country.  相似文献

炎症小体是细胞内组装形成的大分子蛋白复合体,可将白介素-1β(IL-1β) 和IL-18 加工成熟,并诱导细胞焦亡性死亡,在协调对抗病原体感染和生理紊乱的过程中发挥重要作用。Nod 样受体蛋白3(Nod-like receptor protein 3, NLRP3) 炎症小体是迄今为止结构和功能研究得最为明确的炎症小体,其活化后参与免疫性疾病、心血管疾病、神经系统疾病等多种疾病发生及发展过程。研究显示,许多中药有效成分可以调节相关疾病靶细胞中NLRP3 炎症小体的活化。从中药有效成分调节相关疾病靶细胞(如神经细胞、肝肾细胞、内皮细胞、肿瘤细胞等)中NLRP3 炎症小体活化的机制出发,综述近4 年国内外对中药有效成分调节NLRP3 炎症小体活化的研究进展,以期阐释相关中药有效成分的作用特点,并为相关疾病的防治提供一定参考。  相似文献

Mature differentiated macrophages can self‐maintain by local proliferation in tissues and can be extensively expanded in culture under specific conditions, but the mechanisms of this phenomenon remain only partially defined. Here, we show that SIRT1, an evolutionary conserved regulator of life span, positively affects macrophage self‐renewal ability in vitro and in vivo. Overexpression of SIRT1 during bone marrow‐derived macrophage differentiation increased their proliferative capacity. Conversely, decrease of SIRT1 expression by shRNA inactivation, CRISPR/Cas9 mediated deletion and pharmacological inhibition restricted macrophage self‐renewal in culture. Furthermore, pharmacological SIRT1 inhibition in vivo reduced steady state and cytokine‐induced proliferation of alveolar and peritoneal macrophages. Mechanistically, SIRT1 inhibition negatively regulated G1/S transition, cell cycle progression and a network of self‐renewal genes. This included inhibition of E2F1 and Myc and concomitant activation of FoxO1, SIRT1 targets mediating cell cycle progression and stress response, respectively. Our findings indicate that SIRT1 is a key regulator of macrophage self‐renewal that integrates cell cycle and longevity pathways. This suggests that macrophage self‐renewal might be a relevant parameter of ageing.  相似文献

应用火焰原子吸收光度法(FAAS)对苦参、黄柏及黄柏胶囊中微量元素铜、铬的含量进行了测定,并对FAAS若干条件进行了优化选择。结果表明,该法的平均回收率在95.2%~104.8%之间,相对标准偏差在1.76%以下,结果可靠,方法简便,可用于实际样品的测定。  相似文献

再生障碍性贫血是由多种原因引起的骨髓造血功能衰竭,以全血细胞减少为特征的一种综合病症。目前普遍认为其发病机理与免疫因素介导的造血干细胞受损导致造血微环境异常相关。近年来,中医药以其独特的理论体系和治疗方法,在再生障碍性贫血的病因病机及防治防等研究方面取得了较大进展。中医药防治再生障碍性贫血一般从肾精亏虚、毒损髓络、生血乏源入手,以补肾填精、生血解毒为治疗法则,采用中药方剂或/和联合西药,可提高治疗有效率,降低西药的毒副作用,并可以调控T淋巴细胞的分化及相关细胞因子的表达。中药治疗可以改善骨髓造血微环境、不同程度的恢复骨髓的造血功能,提高患者的生存质量。现就中医药对此病的相关研究作一综述。  相似文献

Syndrome of Chinese medicine is an understanding of the regularity of disease occurrence and development and its performance of symptoms. Syndrome is the key to recognize diseases and the foundation to treat them. However, because of the complexity of the concept and the limitation of present investigations, the research of syndrome is hard to go further. Proteomics has been received extensive attention in the area of medical diagnosis and drug development. In the holistic and systemic context, proteomics have a convergence with traditional Chinese medicine (TCM) syndrome, which could overcome the one-sidedness and singleness of TCM and avoid the complication and tedious processes. Chinese medicine has a wealth of experience and proteomics has a substantial research potential, the integration of the two aspects will bring a great enhancement of our knowledge of disease.  相似文献

微生物发酵中药材属于传统中药炮制中一种获得新药的重要技术方法。目前;现代生物技术的发展逐渐对微生物发酵中药的机制和工艺进行了解析;然而;难以找到合适的发酵菌种、微生物发酵中药的机制不明晰、中药发酵的终点不易判定、缺乏质控指标、发酵工艺规范不统一等问题依然凸显。因此;从微生物发酵中药的种类与功能、微生物发酵中药材机制及微生物发酵中药工艺三个方面对现有的研究进展进行了综述;总结了微生物在中药发酵过程中的具体发酵机制;并对微生物发酵中药的发展远景进行展望;为中药发酵菌种的选择、发酵终点的判定、质量标准的制定、工艺的进一步规范及中药产业链新药物的研发和升级提供理论依据和技术参考。  相似文献

用耐甲氧西林的金葡菌(MRSA)及金葡菌标准株为试验菌,测定了14种中药的乙醇提取物与4种抗菌药物的联合抗菌作用。狭叶十大功劳、马齿苋、厚朴、桔梗、益母草、夏枯草与4种抗菌药物对两株金葡菌存在联合作用;泽漆、马鞭草、乌头、车前草、白曼陀罗、杜仲与4种抗菌药物无联合作用;芍药与青霉素、头孢曲松产生很好的联合效应;鸡骨常山能提高环丙沙星的抗菌作用。筛选与抗菌素有联合作用的中草药对抑制金黄色葡萄球菌是可行的。  相似文献

本文讨论了中医现代化和中西医结合的有关问题,指出中医现代化工作可分为三个方面:第一方面,应当培养大批临床上过硬的中医人才,在临床上有所突破,以此为基础进行理论研究;第二方面,将中医理论转换成现代语言;第三方面,中医与包括现代生物医学在内的多学科结合。阐述了现代生物医学科技前沿与中西医结合交叉而产生的新学科一光子中医学。指出光子中医学将为中西医结合提供理论和技术平台,成为多学科研究中医的代表性学科,并将成为中医现代化的一个重要方向,为人类卫生事业作出重大贡献。  相似文献
