Choline acetyltransferase was purified approximately 18,000-fold from 300 g of bovine caudate nuclei to a specific activity of 21 μmol min mg protein. The overall procedure used was: extraction of the enzyme by high salt concentration, chromatography on carboxy-methyl-Sephadex, precipitation by ammonium sulphate, affinity chromatography on Blue-Sepharose and, finally absorption on hydroxylapatite. When the enzyme absorbed on hydroxylapatite was injected into mice, it provoked reproducibly a transient production of ‘inhibitory’ antibodies, followed by higher antibody titres mainly of ‘non-inhibitory’ type. These responses were elicited by injecting less than a total of 20 μg of immunogen. The highest antibody titre was obtained less than 2 months following the initial immunisation. Species cross reactivity was investigated. This procedure should prove to be of value in the production of monoclonal antibodies to choline acetyltransferase.  相似文献

Nine styrylpyridine analogs were tested as inhibitors of choline acetyltransferase which had been highly purified from rat cerebrum and bovine caudate nuclei. In general, concentrations required to achieve 50% inhibiion (I50 values) were in the micromolar range. For some analogs, I50 values were similar to those obtained previously by other investigators who used less purified enzyme preparations. With certain analogs, however, the measured values of I50 changed as the transferase became more purified, which may indicate the presence in the extract of other molecules which can interact with the enzyme. The methods used in purification of the enzyme suggest that the molecule which modifies the activity of CAT is probably a protein. The mode of inhibition by naphthylvinylpyridinium was found to be uncompetitive with respect to both choline and acetyl coenzyme A for both the rat and bovine transferases.  相似文献

Chicken brain choline acetyltransferase was purified to homogeneity using ammonium sulfate fractionation, followed by chromatography on DEAE-Sephadex (A-25), hydroxyapatite, Sephadex G-150, immunoabsorption and Sepharose-CoA columns. A purification of 3500-fold was achieved and the final preparation had a specific activity of 2:32 μmol acetylcholine formed per minute per milligram protein. The purified chicken choline acetyltransferase migrated as a single band on polyacrylamide gel electrophoresis in the presence and absence of sodium deodecyl sulfate. The native enzyme, with a molecular weight of 67,000 daltons, consists of two subunits of identical molecular weight. Chicken choline acetyltransferase has a sharp pH optimum of 7.4. It is activated by sodium chloride and potassium chloride but inhibited by cupric ion and N-ethylmaleimide.  相似文献

Choline acetyltransferase from bovine brain caudate nucleus has been purified to a specific activity of 25–30 μmol ACh formed per min and mg protein. Disc electrophoresis at pH 9.5 of the purified enzyme showed two protein bands localized close to each other. We were not able to show if ChAT was linked to one or both bands. In SDS disc electrophoresis the enzyme preparation showed one major and one minor protein band with molecular weights of 69,000 and 34,000, respectively. Heterogeneity of the enzyme preparation was also demonstrated by immunodiffusion and immunoelectrophoresis. After ammonium sulphate precipitation no aggregation of the enzyme could be detected by gelfiltration on Ultrogel AC-34 whilst a high molecular weight fraction was occasionally observed by gelfiltration on Sephadex G-200. The enzyme was, however, separated into two molecular forms (A and B) on CM-Sephadex chromatography. Both molecular forms had the same S220w but differed in heat stability and affinity for acetyl-CoA. Both forms were inactivated by an antibody preparation raised against either a purified preparation of ChAT, or A and B separately. The highly purified enzyme preparation was inactivated more than 98% by immunoprecipitation. The antibody crossreacted with ChATs from several mammalian species, but only slightly with ChAT from pigeon. The results of binding studies with affinity columns, suggest that the enzyme contains a hydrophobic lobe and a dinucleotide fold, and that a free purine rather than a free ribosyl ring of acetyl-CoA is important for the binding of the substrate to the active site. The hydrophobic lobe may be the same as the dinucleotide fold.  相似文献

—The rates of acetylation of choline and the mono-, di-, and tri-ethyl analogues of choline by choline acetyltransferase (acetyl-CoA: choline O-acetyltransferase; EC 2.3.1.6) were studied with a partially purified enzyme from bovine caudate nucleus. All the substrates were acetylated by ChAc. The rates of acetylation at low concentrations of substrate were choline >MEC >DEC >TEC, but at high concentrations MEC was acetylated more rapidly than choline. These results have been compared to those of previous workers. The mode of binding of choline and its analogues to ChAc is discussed, and it is suggested that replacement of methyl by ethyl groups results in a lower energy of binding of the substrate to the active site of the enzyme.  相似文献

—Choline acetyltransferase was extracted from Lactobacillus plantarum by relatively gentle procedures involving penicillin treatment, osmotic shock and passage through a French pressure cell. After partial purification, the extract was compared with choline acetyltransferase of calf caudate nucleus for kinetic properties and response to a class of inhibitors which consists of analogues of styrylpyridine. Both enzymes obeyed a sequential mechanism with Michaelis constants for the bacterial enzyme, Km= 8 μm vs. acetyl-CoA and 0·44 mm vs. choline; and for the caudate nucleus enzyme, Km= 15 μm vs. acetyl-CoA and 0·8 mm vs. choline. Both were stabilized by dithiothreitol and EDTA. The extracts differed in that the bacterial enzyme was more labile and apparently was susceptible to conformational changes, which modified its response to the styrylpyridinetype inhibitors. The use of intact cells of Lactobacillus plantarum as an in vivo system for studying the inhibition of choline acetyltransferase by styrylpyridines was possible only for non-quaternary analogues, which exist as an equilibrium mixture of charged and uncharged species.  相似文献

TYROSINE HYDROXYLASE IN BOVINE CAUDATE NUCLEUS   总被引:7,自引:4,他引:3  
Approximately 80 per cent of tyrosine hydroxylase activity in bovine caudate nucleus was particle-bound. The rest of the activity was found in the soluble fraction. The enzyme activity in crude tissue preparations was inhibited, probably by the presence of endogenous inhibitors. Dilution of crude tissue preparations such as the crude mitochondrial fraction caused an increase in the specific activity. The particle-bound enzyme was solubilized by incubation with trypsin. The presence of deoxycholate increased the degree of solubilization. The activity of the solubilized enzyme from the washed particles was also inhibited, but the subsequent purification by ammonium sulphate could eliminate the inhibition. The solubilized enzyme was partially purified by ammonium sulphate fractionation and Sephadex G-150 chromatography. A tetrahydropteridine and ferrous ion were required as cofactors for the partially purified enzyme. Among various divalent cations, only ferrous ion could activate the partially purified enzyme. The enzyme was inhibited by L-α-methyl-p-tyrosine and catecholamines such as dopamine. The optimum pH was found between 5.5 and 6.0. Km values toward tyrosine, 2-amino-4-hydroxy-6,7-dimethyltetrahydropteridine and Fe2+, were approximately 5 × 10?5 M, 1 × 10?4 M and 4 × 10?4 M, respectively.  相似文献

—The isoelectric point, surface charge and Km for choline of choline acetyltransferase from different species were determined. Choline acetyltransferase from mouse and monkey brain was resolved into three molecular forms with isoelectric points at 7·1, 7·5, 8·4 and 7·0, 7·35, 8·35 respectively, whereas choline acetyltransferase from the electric organ of Torpedo and from rabbit brain showed a molecular form with isoelectric point 6·6 and 6·9, respectively. With the exception of rabbit brain enzyme, there was a good correlation between the isoelectric points and surface charges of the different choline acetyltransferases. The Km's for choline were 0·66, 0·88, 0·92 and 3·5 mM for monkey, mouse, rabbit and Torpedo choline acetyltransferase respectively. The separated molecular forms of mouse and monkey enzymes did not show any significant difference in their affinity for choline.  相似文献

—Tyramine β-hydroxylase catalyzes the biosynthesis of octopamine in the lobster nervous system. This enzyme has been characterized and a rapid microassay, based on the enzymic release of tritiated water from [1,2-(side chain) 3H] tyramine, has been developed. Lobster tyramine β-hydroxylase resembled mammalian dopamine β-hydroxylase. The most conspicuous differences were that the lobster enzyme was inhibited by anions, particularly fumarate, and had a higher affinity for substrates. Tyramine β-hydroxylase activity was present in both particulate and soluble fractions of homogenates of the lobster nervous system. Bound activity, extracted by repeated freezing and thawing, was partially purified. The enzyme had the following properties: (1) The optimum pH for the conversion of tyramine to octopamine was 7·4. (2) The apparent Michaelis constant for tyramine was 0·15 mm and for ascorbic acid was 0·2 mm at pH 6·6. (3) The purified enzyme was inhibited by salts; the degree of inhibition was sensitive to the anion and decreased in the order chloride ? fumarate > sulphate > acetate. (4) Tyramine β-hydroxylase was inhibited by metal chelating agents and by cupric sulphate at concentrations greater than 10?4m ; N-ethylmaleimide had no significant effect on activity in concentrations up to 3 mm . (5) The purified enzyme also β-hydroxylated dopamine to form norepinephrine, with an apparent Michaelis constant of 0·24 mm . This activity co-purified with tyramine β-hydroxylase, suggesting that a single enzyme catalyzed both reactions.  相似文献

WhenArthrobacter P1 is grown on choline, betaine, dimethylglycine or sarcosine, an NAD+-dependent formaldehyde dehydrogenase is induced. This formaldehyde dehydrogenase has been purified using ammonium sulphate fractionation, anion exchange- and hydrophobic interaction chromatography. The molecular mass of the native enzyme was 115 kDa±10 kDa. Gel electrophoresis in the presence of sodium dodecyl sulphate indicated that the molecular mass of the subunit was 56 kDa±3 kDa, which is consistent with a dimeric enzyme structure. After ammonium sulphate fractionation the partially purified enzyme required the addition of a reducing reagent in the assay mixture for maximum activity. The enzyme was highly specific for its substrates and the Km values were 0.10 and 0.80 mM for formaldehyde and NAD+, respectively. The enzyme was heat-stable at 50° C for at least 10 min and showed a broad pH optimum of 8.1 to 8.5. Choline acetyltransferase activity in homogenates, or in partially purified extracts of rat brain cerebra, was increased by 11–37% in the presence of ethanol when incubated at 38°C with [14C] acetyl-CoA, choline chloride and alcohol concentrations of 0.17M to 1.02M. In preincubation experiments with enzyme preparations and ethanol, inactivation of the enzyme by the alcohol, which occurs at incubation times longer than 20 minutes, could be at least partially prevented by the addition of certain components of the incubation mixture to the preincubation mixture.  相似文献

A carboxylesterase (EC 3.1.1.1) was purified from germinated finger millet by ammonium sulphate fractionation, diethylaminoethyl-cellulose chromatography and Sephadex G-200 filtration. The homogeneity of the enzyme was established by Polyacrylamide gel electrophoresis, isoelectric focussing and sodium dodecyl sulphate-polyacrylamide gel electrophoresis. The enzyme has a single polypeptide chain with a molecular weight of 70,000. The amino acid analysis of the purified enzyme revealed that it contained a greater number of neutral and acidic, compared to, basic amino acid residues. The isoelectric pH of the enzyme was found to be 5·1. Studies with different organophosphate and carbamate inhibitors showed that this enzyme was more sensitive to organophosphate inhibitors than carbamates. The rate constantsk i andl 50 for different inhibitors were calculated. The product inhibition studies with this enzyme showed linear competitive inhibition with acetate and linear noncompetitive inhibition with 1-naphthol  相似文献

(1) Dopamine, β-hydroxylase (EC 1.14.2.1) was purified from bovine adrenal medullae according to the method of Foldes , Jeffrey , Preston and Austin (1972). (2) The kinetics, pH optimum and the effect of Cu2+ ions on the purified enzyme were found to resemble those of the enzyme isolated by more involved procedures. (3) The sedimentation coefficient (s20) of the homogeneous enzyme in 10 mM-phosphate buffer, pH 7·2, containing 0·1 M-NaCI was found to be 10·24 ± 0·12 (S.E.M. of 10 determinations). (4) The effect of pH on the mol. wt. of the enzyme was investigated and no large deviation was found from the native mol. wt. of 290,000 in the pH range 3·9 to 11·1. (5) The amino acid analysis of dopamine β-hydroxylase is presented, and is contrasted to that of chromogranin A purified from the same chromaffin granule lysate. (6) Treatment with either 8 M-urea or 0·1% (w/v) sodium dodecyl sulphate was found to dissociate the enzyme into three similar, non-active subunits, each of mol. wt. of the order of 100,000.  相似文献

  • 1 Choline acetyltransferase was purified from ox brain striate nuclei by an extraction step at pH 5, cation-exchange chromatography, fractional precipitation with ammonium sulphate, and chromatography on Sephadex G-200. The enzyme was obtained free of deacylases and cholinesterases, at specific activities of 01-0-3 μmol acetylcholine formed per min per mg protein.
  • 2 The enzyme was found to be a stable and relatively basic protein, with a molecular weight of 65,000.
  • 3 In the catalysed reactions, , k1, was about four times k2, and the equilibrium constant was approximately 40. For the forward reaction, the Michaelis constant for each substrate was independent of the concentration of the other (choline = 0-75 mM; acetyl-CoA = 10 μM), whereas in the back reaction one substrate increased the affinity for the other (acetylcholine = 0-75-5 MM; CoA = 25-150 μM).
  • 4 CoA inhibited acetylcholine synthesis by competing with acetyl-CoA (K1, = 16 μM). Acetylcholine slightly inhibited the forward reaction (e.g. 45 per cent in 200 mM) without competing with choline or acetyl-CoA. These data indicate an ordered reaction mechanism; acetyl-CoA probably always binds before choline.

The lactate dehydrogenase (LDH) from Nocardia asteroides was purified to homogeneity by ammonium sulphate precipitation, gel filtration on Sephadex G-150 and DEAE-Sepharose column chromatography. The purified enzyme showed a single band in native condition which indicated its homogeneity. SDS-PAGE of the purified enzyme showed the presence of three bands which correspond to molecular weights of 60, 66 and 74 kDa. The pH and temperature optima of the purified enzyme were 9.5 and 50 degrees C, respectively. The metal ions Mn++, Fe++, Co++, Mg++ and Ca++, increased the purified LDH activity. On the other hand, enzyme activity was completely inhibited by CuCl2. Potassium chloride, ammonium sulphate and sodium chloride did not alter the enzyme activity. The purified enzyme exhibited a Km value of 1.6 x 10(-5) M for pyruvate.  相似文献

对牛小脑膜区肌醇磷脂激酶进行了11 500倍纯化,过程包括:TritonX-100抽提,硫酸铵沉淀,阳离子交换层析(phosphocellulose),亲和层析(Heparin Sepharose CL-6B)和阴离子交换层析(DEAE10,FPLC)等.纯化程度可达95%以上,对SDS-PAGE电泳结果进行扫描分析测其分子质量为56 ku.纯化的肌醇磷脂激酶的特异活性为450 nmol/mg·min, 动力学性质表现为ATP的表观Km值为7.9×10-7 mol/L,PI的表观Km值为6.6×10-7 mol/L. 腺嘌呤核苷是该酶的有效抑制剂,3.5×10-7 mol/L腺嘌呤核苷可使该酶活力降低约50%,而TritonX-100对该酶活力具有刺激作用,0.5% TritonX-100可使该酶表现为最高活力.  相似文献

Abstract— Choline acetyltransferase from bovine brain has been extensively purified to a specific activity of 2.5 μmol ACh/min mg protein. Attempts to isolate an acetyl enzyme intermediate after incubation of the enzyme with [1-14C]acetyl-CoA were unsuccessful. Such an intermediate could only be isolated using a 30-fold less purified enzyme preparation. The protein, binding 14C in this preparation, did not correspond to choline acetyltransferase as shown by disc-electrophoresis. The highly purified enzyme could, however, be labelled when choline acetyltransferase was immobilized on a mercuribenzoate sepharose gel and incubated with [1-14C]acetyl-CoA. Subsequently, the immobilized labelled enzyme or the labelled enzyme which had been released by cysteine from the gel. formed ACh after incubation with choline. The labelling and the following formation of [14C]ACh was pH dependent.
Masking htstidine residues of the enzyme with diethylpyrocarbonate almost abolished the labelling of the immobilized enzyme and completely abolished the formation of [14C]ACh. Enzyme inhibited with 5.5'-dithiobis(2-nitrobenzoate) was partially reactivated when the thionitrobenzoatederivative was cleaved by KCN treatment to a thiocyanatederivalive. A reaction mechanism for ChAT is proposed based on the present data.  相似文献

1. Choline acetyltransferase may be isolated in either a bound or soluble form after hypo-osmotic treatment of a crude synaptosome fraction, depending on the conditions. 2. In the bound form, the enzyme appears to be associated with the larger membrane fragments rather than with synaptic vesicles. 3. The bound form is predominant at slightly acid pH values and low ionic strength, the soluble form under more physiological conditions of pH and ionic strength. 4. Sodium chloride, potassium chloride, magnesium chloride and calcium chloride at similar ionic strengths solubilize the enzyme. 5. Choline acetyltransferase was found to be soluble under these conditions after release from synaptosomes from rat and pigeon cerebra, guinea-pig cortex and rabbit cortex, caudate nuclei, diencephalon and midbrain. 6. Certain isoenzymes of lactate dehydrogenase behaved similarly.  相似文献

Acyl-CoA oxidase (EC 1.3.3.6), an enzyme that catalyses the first and rate limiting step of the β-oxidation spiral in plant glyoxysomes was purified to homogeneity from loblolly pine (Pinus taeda L.) megagametophytes isolated 9 to 11 d after imbibition at 30 °C. Homogeneity of the enzyme was confirmed by silver staining SDS-PAGE gels of peak enzymatic activity fractions from a molecular sieving column that was the last step of purification. The purification procedure included acetone extraction, heat treatment, ammonium sulphate precipitation and chromatography on three columns (phenyl-Sepharose, hydroxyapatite, and molecular sieving). The homogenous enzyme was purified 1 250-fold to a specific activity of 417.5 nkat·mg–1 protein. The enzyme was a homodimer with a native molecular mass of 150 kDa and a subunit molecular mass of 71 kDa. Polyclonal antibodies raised in rabbits were confirmed to be mono-specific for acyl-CoA oxidase by immunotitration and western blotting experiments. A western blot analysis of cell free extracts indicated that acyl-CoA oxidase was predominantly megagametophytic.  相似文献

Choline kinase (EC 2.7.1.32; ATP: choline phosphotransferase) was purified 200-fold from an extract of acetone powder of rabbit brain by a combination of acid precipitation, ammonium sulphate precipitation, DEAE cellulose chromatography, and ultrafiltration. Maximal activity of 243 nmol of phosphorylcholine synthesized. min?1 mg?l of protein occurred at pH 9.5–10.0 in the presence of 10 mm MgS04, 10 mm choline and 0.005% (w/v) bovine serum albumin. 2-Aminoethanol, 2-methylaminoethanol, and 2-dimethylaminoethanol were also phosphorlyated by the enzyme preparation. The enzyme quantitatively converted low concentrations of choline (2.5–50 μm ) to phosphorylcholine [32P] in the presence of ATP [y32P], and may, therefore, be used to measure small amounts of choline acetylcholine. There were two Km values for choline at pH 9.5; 32 μm and 0.31 mm . At pH 7.4, the higher Km was not observed and enzyme activity was maximal with 0.1 mm choline. The Km for ATP was 1.1 mm . Enzyme activity was inhibited by ATP (20 mm ), AMP, ADP, cytidine diphosphocholine (1 or 10 mm ), and activated by choline esters (1.0 mm ), NaCl or KCl(200 mm ).  相似文献
