This work was conducted to study the effects of fertilization with nitrogen (NaNO2, NH4C1 or CO(NH2 2) on NaCl salinized maize and safflower plants. Transpiration rate, stomatal frequency, dry matter and pigment contents were reduced under salinity. However under salinization-fertilization treatments, these parameters were generally raised. The carbohydrate contents exhibited variable results, in dependence on the plant type tested as well as on the treatment applied. The nitrogen contents of variously treated plants were raised except in the case of safflower plants salinized with only NaCl.  相似文献

The sucrose non‐fermenting‐1‐related protein kinase 2 (SnRK2) family represents a unique family of plant‐specific protein kinases implicated in cellular signalling in response to osmotic stress. In our studies, we observed that two class 1 SnRK2 kinases, SnRK2.4 and SnRK2.10, are rapidly and transiently activated in Arabidopsis roots after exposure to salt. Under saline conditions, snrk2.4 knockout mutants had a reduced primary root length, while snrk2.10 mutants exhibited a reduction in the number of lateral roots. The reduced lateral root density was found to be a combinatory effect of a decrease in the number of lateral root primordia and an increase in the number of arrested lateral root primordia. The phenotypes were in agreement with the observed expression patterns of genomic yellow fluorescent protein (YFP) fusions of SnRK2.10 and ‐2.4, under control of their native promoter sequences. SnRK2.10 was found to be expressed in the vascular tissue at the base of a developing lateral root, whereas SnRK2.4 was expressed throughout the root, with higher expression in the vascular system. Salt stress triggered a rapid re‐localization of SnRK2.4–YFP from the cytosol to punctate structures in root epidermal cells. Differential centrifugation experiments of isolated Arabidopsis root proteins confirmed recruitment of endogenous SnRK2.4/2.10 to membranes upon exposure to salt, supporting their observed binding affinity for the phospholipid phosphatidic acid. Together, our results reveal a role for SnRK2.4 and ‐2.10 in root growth and architecture in saline conditions.  相似文献

Salinity caused a consistent reduction in the growth of cowpea plants and water content in their leaves. The total as well as the pigment fractions, except carotenoids, exhibited lower values than those of control plants at almost all salinity levels. With the rise of salinization, the total nitrogen and potassium contents in the leaves were decreased but the sodium content was increased and phosphorus content was not significantly affected as compared with the controls. The application of IAA to salt-treated plants increased the water content in the leaves but it had no effect on the number of leaves and the stem length. The pigment contents in the leaves were either promoted or inhibited with the application of IAA, depending upon the level of salinization. The application of IAA to plants irrigated with water of the highest salinity level was effective in increasing the potassium content in the leaves as compared with the control, but it had no effect on the other mineral elements.  相似文献

We tested the response of Enteromorpha intestinalis to fluctuating reduced salinity regimes which may occur in coastal estuaries due to both natural and anthropogenic influences. In a fully crossed two factor experiment, we subjected E. intestinalis to 0, 5, 15 and 25 psu water enriched with nutrients for 1-, 5-, 11- and 23-day periods. Each period was followed by 24 h of exposure to 25 psu (ambient) water that was not nutrient enriched. Following 24 h in ambient salinity water, algae were returned to reduced salinity conditions for the appropriate period and the cycle continued over the 24 days for which all treatments were maintained. Exposure to 0 psu for 5 days or longer resulted in loss of pigmentation, decreased wet and dry biomass, increased wet wt:dry wt ratios, decreased removal of nitrogen (N) and phosphorus (P) from the water column and an accumulation of NH(4) in the water column. More frequent exposure to ambient salinity in the 1-day treatment mitigated these effects. Across all salinity levels tested, biomass increased as frequency of exposure to ambient salinity increased. At all durations of exposure to low salinity tested, biomass increased as salinity level increased. We conclude that growth of E. intestinalis is decreased by reduced salinity. E. intestinalis is able to withstand exposure to 0 psu but there is a temporal limit to this tolerance that is somewhere between 1 and 5 days. Populations of E. intestinalis in coastal estuaries may suffer from freshwater inputs if salinity conditions are persistently reduced.  相似文献

Aims

Heat stress is a growing concern in crop production because of global warming. In many cropping systems heat stress often occurs simultaneously with other environmental stress factors such as mineral nutrient deficiencies. This study aimed to investigate the role of adequate magnesium (Mg) nutrition in mitigating the detrimental effects of heat stress on wheat (Triticum aestivum) and maize (Zea mays).

Methods

Wheat and maize plants were grown in solution culture with low or adequate Mg supply at 25/22 °C (light/dark). Half of the plants were, then, exposed to heat stress at 35/28 °C (light/dark). Development of leaf chlorosis and changes in root and shoot growth, chlorophyll and Mg concentrations as well as the activities of major antioxidative enzymes were quantified in the experimental plants. Additionally, maize plants were analyzed for the specific weights (e.g., dry or fresh weight per a given leaf surface area) and soluble carbohydrate concentrations of sink and source leaves.

Results

Visual leaf symptoms of Mg deficiency were aggravated in wheat and maize when exposed to heat stress. In both species, root growth was more sensitive to Mg deficiency than shoot growth, and the shoot-to-root ratios peaked when heat stress was combined with Mg deficiency. Magnesium deficiency markedly reduced soluble carbohydrate concentrations in young leaf; but resulted in substantial increase in source leaves. Magnesium deficiency also increased activities of antioxidative enzymes, especially when combined with heat stress. The highest activities of superoxide dismutase (up to 80 % above the control), glutathione reductase (up to 250 % above the control) and ascorbate peroxidase (up to 300 % above the control) were measured when Mg-deficient plants were subjected to heat, indicating stimulated formation of reactive oxygen species (ROS) in Mg deficient leaves under heat stress.

Conclusions

Magnesium deficiency increases susceptibility of wheat and maize plants to heat stress, probably by increasing oxidative cellular damage caused by ROS. Ensuring a sufficiently high Mg supply for crop plants through Mg fertilization is a critical factor for minimizing heat-related losses in crop production.  相似文献

Development of antifouling strategies requires knowledge of how fouling organisms would respond to climate change associated environmental stressors. Here, a calcareous tube built by the tubeworm, Hydroides elegans, was used as an example to evaluate the individual and interactive effects of ocean acidification (OA), warming and reduced salinity on the mechanical properties of a tube. Tubeworms produce a mechanically weaker tube with less resistance to simulated predator attack under OA (pH 7.8). Warming (29°C) increased tube volume, tube mineral density and the tube’s resistance to a simulated predatory attack. A weakening effect by OA did not make the removal of tubeworms easier except for the earliest stage, in which warming had the least effect. Reduced salinity (27 psu) did not affect tubes. This study showed that both mechanical analysis and computational modeling can be integrated with biofouling research to provide insights into how fouling communities might develop in future ocean conditions.  相似文献

This review summarizes our current state of knowledge on the structural organization and functional pattern of photosynthetic water splitting in the multimeric Photosystem II (PS II) complex, which acts as a light-driven water: plastoquinone-oxidoreductase. The overall process comprises three types of reaction sequences: (1) photon absorption and excited singlet state trapping by charge separation leading to the ion radical pair [Formula: see text] formation, (2) oxidative water splitting into four protons and molecular dioxygen at the water oxidizing complex (WOC) with P680+* as driving force and tyrosine Y(Z) as intermediary redox carrier, and (3) reduction of plastoquinone to plastoquinol at the special Q(B) binding site with Q(A)-* acting as reductant. Based on recent progress in structure analysis and using new theoretical approaches the mechanism of reaction sequence (1) is discussed with special emphasis on the excited energy transfer pathways and the sequence of charge transfer steps: [Formula: see text] where (1)(RC-PC)* denotes the excited singlet state (1)P680* of the reaction centre pigment complex. The structure of the catalytic Mn(4)O(X)Ca cluster of the WOC and the four step reaction sequence leading to oxidative water splitting are described and problems arising for the electronic configuration, in particular for the nature of redox state S(3), are discussed. The unravelling of the mode of O-O bond formation is of key relevance for understanding the mechanism of the process. This problem is not yet solved. A multistate model is proposed for S(3) and the functional role of proton shifts and hydrogen bond network(s) is emphasized. Analogously, the structure of the Q(B) site for PQ reduction to PQH(2) and the energetic and kinetics of the two step redox reaction sequence are described. Furthermore, the relevance of the protein dynamics and the role of water molecules for its flexibility are briefly outlined. We end this review by presenting future perspectives on the water oxidation process.  相似文献

As the salt-affected areas are expected to increase substantially in subsequent years, the impact of salinity on plant growth and yield is likely to increase. One of the first consequences of plant exposure to high saline concentrations is the formation of reactive oxygen species (ROS). In order to allow adjustment of the cellular redox state, plant antioxidative system has to be activated. This system involves several enzymes and compounds, as the sulphur-containing metabolite glutathione (GSH). Therefore, our aim was to determine whether adequate sulphur nutrition might alleviate the adverse effects of salt stress on barley plants grown in the presence of different sulphate application rate and exposed to 100 mM NaCl, by studying differences in growth parameters, lipid peroxidation, sulphate and thiol accumulation and sulphur assimilation pathway. In salt-treated plants, an adequate sulphur supply allows adequate GSH synthesis (high-thiol concentration) thus avoiding the effects of ROS on photosynthetic functions (no effect on both chlorophyll and protein content), whereas in S-deficient plants, salt stress leads to excess ROS production that induces stress and plants showed reduction of photosynthetic efficiency (loss of chlorophyll and protein contents). As thiol levels are more abundant in S-sufficient plants than in those S-deficient, one might expect that S-sufficient plants are more able to remove the harmful effects of high salinity. The comparison of malondialdehyde levels between +S and ?S salt-treated plants strongly supports this idea. In conclusion, we found that plant sulphur nutritional status plays a key role in the metabolic modifications necessary to cope with salt stress.  相似文献

Use of an in-season measure of crop N sufficiency to guide fertilizer management is one approach to match the supply of N to the crop N demand. This study examined use of gene expression in leaf tissue of field-grown potatoes for use in assessment of potato N sufficiency. Potato cultivar ‘Shepody’ was grown with six fertilizer N rates (0–250 kg N ha–1). Leaf disks were collected weekly for quantification of the expression of N uptake/transport, N assimilation, and amino acid metabolism genes in leaf tissue by nCounter. Many of the genes evaluated were responsive to crop N supply, but the response varied widely among sampling dates. The exception was an ammonium transporter gene (AT1) which was highly expressed, was relatively consistent across sampling dates, was closely related to root zone soil nitrate concentration across N rates and sampling dates, and was highly negatively correlated with total tuber yield. The level of expression of AT1 in leaf tissue was as good as or better than conventional chemical or optical measures of potato N sufficiency in the current study.  相似文献

Green photosynthetic bacteria harvest light and perform photosynthesis in low-light environments, and contain specialized antenna complexes to adapt to this condition. We performed small-angle neutron scattering (SANS) studies to obtain structural information about the photosynthetic apparatus, including the peripheral light-harvesting chlorosome complex, the integral membrane light-harvesting B808-866 complex, and the reaction center (RC) in the thermophilic green phototrophic bacterium Chloroflexus aurantiacus. Using contrast variation in SANS measurements, we found that the B808-866 complex is wrapped around the RC in Cfx. aurantiacus, and the overall size and conformation of the B808-866 complex of Cfx. aurantiacus is roughly comparable to the LH1 antenna complex of the purple bacteria. A similar size of the isolated B808-866 complex was suggested by dynamic light scattering measurements, and a smaller size of the RC of Cfx. aurantiacus compared to the RC of the purple bacteria was observed. Further, our SANS measurements indicate that the chlorosome is a lipid body with a rod-like shape, and that the self-assembly of bacteriochlorophylls, the major component of the chlorosome, is lipid-like. Finally, two populations of chlorosome particles are suggested in our SANS measurements.  相似文献

Basil (Ocimum basilicum L.) seedlings were cultured on liquid medium in controlled conditions. Two varieties differing in leaf size were compared. When plants were 30?days old, the medium was supplemented with 50?mM NaCl. After 15?days of treatment, root, stem and leaf biomass, leaf number, and leaf surface area were measured. Ion accumulation was determined in roots, stems, and leaves. Photosynthetic parameters (CO2 fixation rate, internal CO2 concentration, stomatal conductance) as well as transpiration rate were determined on separate leaves. Electrolyte leakage and malondialdehyde content were used to estimate damage to membranes and lipid peroxidation, respectively. Several antioxidant enzymatic activities were used as proxies of oxidative stress. High Na+ concentration was reached in leaf tissues. Salt restricted whole plant biomass deposition rate by diminishing leaf number and leaf expansion, as well as photosynthetic activity were estimated from whole plant biomass production per unit leaf surface area. Diminished stomatal conductance restricted CO2 fixation rate, and decrease in chlorophyll content presumably limited photosynthetic activity. Lipid peroxidation revealed damages to membranes. The magnitude of these responses differed between the two varieties, indicating that an intraspecific variability in salt response exists in basil.  相似文献

Absorption of excess light energy by the photosynthetic machinery results in the generation of reactive oxygen species (ROS), such as H2O2. We investigated the effects in vivo of ROS to clarify the nature of the damage caused by such excess light energy to the photosynthetic machinery in the cyanobacterium Synechocystis sp. PCC 6803. Treatments of cyanobacterial cells that supposedly increased intracellular concentrations of ROS apparently stimulated the photodamage to photosystem II by inhibiting the repair of the damage to photosystem II and not by accelerating the photodamage directly. This conclusion was confirmed by the effects of the mutation of genes for H2O2-scavenging enzymes on the recovery of photosystem II. Pulse labeling experiments revealed that ROS inhibited the synthesis of proteins de novo. In particular, ROS inhibited synthesis of the D1 protein, a component of the reaction center of photosystem II. Northern and western blot analyses suggested that ROS might influence the outcome of photodamage primarily via inhibition of translation of the psbA gene, which encodes the precursor to D1 protein.  相似文献

We assessed the role of plant growth-promoting rhizobacteria (PGPR) strains viz. Burkholdera cepacia SE4, Promicromonospora sp. SE188 and Acinetobacter calcoaceticus SE370 in counteracting salinity and drought stress to cucumber plants. The control plants had stunted growth, while PGPR-treated plants had significantly higher biomass and chlorophyll contents under salinity and drought stress. The ameliorative effects of PGPR-application were also evidenced by the increased water potential and decreased electrolytic leakage. The PGPR-applied plants had reduced sodium ion concentration, while the potassium and phosphorus were abundantly present as compared to control under stress. Oxidative stress was mitigated by PGPR through reduced activities of catalase, peroxidase, polyphenol oxidase, and total polyphenol as compared to control. The control plants showed up-regulation of stress-responsive abscisic acid as compared to PGPR application, while salicylic acid and gibberellin 4 were significantly higher in PGPR. In conclusion, the PGPR application might be used in marginalized agricultural lands to increase crop productivity.  相似文献

Halorhodospira halochloris is an anaerobic, halophilic, purple photosynthetic bacterium belonging to γ-Proteobacteria. H. halochloris is also characteristic as a thermophilic phototrophic isolate producing bacteriochlorophyll (BChl) b. Here, we report the complete genome sequence of H. halochloris DSM 1059. The genetic arrangement for this bacterium’s photosynthetic apparatus is of particular interest; its genome contains two sets of puf operons encoding the reaction center and core light-harvesting 1 (LH1) complexes having almost identical nucleotide sequences (e.g., 98.8–99.9% of nucleotide identities between two sets of pufLM genes, but 100% of deduced amino acid sequence identities). This duplication of photosynthetic genes may provide a glimpse at natural selection in action. The β-polypeptides of the LH1 complex in purple bacteria usually contain two histidine residues to bind BChl a; however, those of H. halochloris were revealed to have four histidine residues, indicating unusual pigment organization in the LH1 complex of this species. Like in other BChl b-producing phototrophs, the genome of H. halochloris lacks the divinyl reductase genes bciA and bciB. The phylogeny of chlorophyllide a oxidoreductase, which catalyzes committed steps in the synthesis of BChl a and BChl b, indicates that evolution toward BChl b production is convergent. Geranylgeranyl reductase (BchP) of H. halochloris has an insertion region in its primary structure, which could be important for its unusual sequential reduction reactions.

In the current study, we explored the ameliorative capacity of Leifsonia xyli SE134, a plant growth-promoting rhizobacteria (PGPR), against copper (Cu) stress on tomato grown under elevated Cu levels of 50 and 100?mM. Initially, L. xyli SE134 modulated innate gibberellins (GAs) and indole-3-acetic acid (IAA) metabolism in response to elevated Cu toxicity. The IAA contents increased, whereas that of bioactive GAs decreased in relation to Cu concentration gradient in the broth media. Furthermore, exposure to elevated Cu caused detrimental effects on the physiological attributes as revealed by attenuated shoot length, root length, stem diameter, shoot dry weight, root dry weight, and chlorophyll content in non-inoculated tomatoes as compared to L. xyli SE134 inoculated plants. The growth rescuing effect of L. xyli SE134 may be attributed to the modulation of endogenous amino acids contents in plants, such as glutamic acid, threonine, phenylalanine, glycine, proline, and arginine. Moreover, L. xyli SE134 inoculation stimulated total polyphenol and flavonoid content, reduced super oxide dismutase activity, strongly inhibited Cu, and increased phosphorus and iron content in plants grown under elevated Cu stress. In the absence of Cu toxicity, L. xyli SE134 significantly enhanced amino acid content, improved total flavonoids, and increased phosphorus content, thus resulting in higher plant growth.  相似文献

The Raf-like protein kinase abscisic acid (ABA) and abiotic stress-responsive Raf-like kinase (ARK) previously identified in the moss Physcomitrium (Physcomitrella) patens acts as an upstream regulator of subgroup III SNF1-related protein kinase2 (SnRK2), the key regulator of ABA and abiotic stress responses. However, the mechanisms underlying activation of ARK by ABA and abiotic stress for the regulation of SnRK2, including the role of ABA receptor-associated group A PP2C (PP2C-A), are not understood. We identified Ser1029 as the phosphorylation site in the activation loop of ARK, which provided a possible mechanism for regulation of its activity. Analysis of transgenic P. patens ark lines expressing ARK-GFP with Ser1029-to-Ala mutation indicated that this replacement causes reductions in ABA-induced gene expression, stress tolerance, and SnRK2 activity. Immunoblot analysis using an anti-phosphopeptide antibody indicated that ABA treatments rapidly stimulate Ser1029 phosphorylation in the wild type (WT). The phosphorylation profile of Ser1029 in ABA-hypersensitive ppabi1 lacking protein phosphatase 2C-A (PP2C-A) was similar to that in the WT, whereas little Ser1029 phosphorylation was observed in ABA-insensitive ark missense mutant lines. Furthermore, newly isolated ppabi1 ark lines showed ABA-insensitive phenotypes similar to those of ark lines. Therefore, ARK is a primary activator of SnRK2, preceding negative regulation by PP2C-A in bryophytes, which provides a prototype mechanism for ABA and abiotic stress responses in plants.

The effect of acclimation salinity and salinity changes on the concentration of high-energy phosphate metabolites and arginine kinase (AK) flux was examined in vivo in juvenile blue crabs using 31P-nuclear magnetic resonance (NMR). Crabs were acclimated for 7 days to a salinity of 5 or 35 per thousand and then placed in a flow apparatus that could sustain the animals while NMR spectra were acquired. Crabs were subjected to either hyperosmotic salinity changes, where an animal acclimated to 5 per thousand was exposed to a salinity of 35 per thousand, or hyposmotic changes, which involved the reciprocal exchange. Neither acclimation salinity nor salinity change had a significant effect on the concentrations of arginine phosphate, inorganic phosphate or ATP. 31P-NMR saturation transfer experiments were used to determine the effect of salinity on the forward and reverse flux of the AK reaction. There was no significant effect of acclimation salinity or salinity change on the flux rate through this reaction. This is in contrast to previous results, which showed that AK flux in isolated muscle was sensitive to prevailing osmotic conditions (Holt and Kinsey, J. Exp. Biol. 205 (2002) 1775-1785). The present study indicates that the integrated osmoregulatory capacity of the intact animal is sufficient to preserve cellular energy status and enzyme function during acute salinity changes.  相似文献
