Dynamics of Introduced Populations of Phragmidium violaceum and Implications for Biological Control of European Blackberry in Australia 下载免费PDF全文

Phragmidium violaceum causes leaf rust on the European blackberry (Rubus fruticosus L. aggregate). Multiple strains of this pathogen have been introduced into southern Australia for the biological control of at least 15 taxa of European blackberry, a nonindigenous, invasive plant. In climates conducive to leaf rust, the intensity of disease varies within and among infestations of the genetically variable host. Genetic markers developed from the selective amplification of microsatellite polymorphic loci were used to assess the population genetic structure and reproductive biology of P. violaceum within and among four geographically isolated and diseased infestations of the European blackberry in Victoria, Australia. Despite the potential for long-distance aerial dispersal of urediniospores, there was significant genetic differentiation among all populations, which was not associated with geographic separation. An assessment of multilocus linkage disequilibrium revealed temporal and geographic variation in the occurrence of random mating among the four populations. The presence of sexual spore states and the results of genetic analyses indicated that recombination, and potentially random migration and genetic drift, played an important role in maintaining genotypic variation within populations. Recombination and genetic differentiation in P. violaceum, as well as the potential for metapopulation structure, suggest the need to release additional, genetically diverse strains of the biocontrol agent at numerous sites across the distribution of the Australian blackberry infestation for maximum establishment and persistence. 相似文献

对来自新疆的104个小麦品种、高代品系及35个含有已知抗叶锈基因载体品种,在苗期接种12个中国小麦叶锈菌生理小种进行抗叶锈基因推导分析和分子检测;2007-2008年和2008-2009年连续2年度对这些材料进行成株抗叶锈性鉴定并筛选慢叶锈性品种。研究结果显示,在41个品种中共鉴定出6个已知抗叶锈基因Lr26、Lr34、Lr50、Lr3ka、Lr1和Lr14a,其中Lr26存在于21个品种中,Lr34在17个品种被发现,Lr1和Lr14a分别存在于3个品种中,还有2个品种携带Lr3ka以及1个品种携带Lr50。2年田间抗叶锈性鉴定筛选出7个慢叶锈性品种,可用于小麦抗病育种。 相似文献

Phragmidium violaceum is a rust fungus with potential for the biological control of European blackberry (Rubus fruticosus) in Australia. The collection, selection, purification and multiplication in Europe of isolates of the rust is described. Species of European blackberry naturalised in Australia showed different levels of susceptibility when inoculated with a pool of 15 isolates highly pathogenic to Rubus procerus. Selection of individual isolates on the four most widespread blackberry species showed that only two of the isolates would be required to obtain the best attack on these four species. 相似文献

Field surveys in 2006 confirmed that the exotic rust fungus Phragmidium violaceum was widespread on Rubus armeniacus and Rubus laciniatus in the Pacific Northwest of the USA. The origin and dispersal pattern of this obligate biotrophic pathogen in the USA were investigated by comparing the genetic diversity and structure of 27 isolates each from the USA and Europe, and 20 isolates from Australia where an invasion occurred in 1984. Analysis of 11 microsatellite loci revealed 74 unique genotypes, with the European population having a significantly higher level of allelic diversity and number of private alleles compared to populations from the USA and Australia. Principal coordinate analysis (PCA), analysis of molecular variance and pairwise comparisons of Φ confirmed a strong level of differentiation among continental populations, with little divergence between isolates from the USA and Europe, but a high level of differentiation between these isolates and those from Australia. These results were broadly supported by the Bayesian cluster analysis, which indicated that at K = 3 the clustering of the isolates corresponds to their geographic origin. Bayesian clustering, PCA as well as insignificant migration estimates from Europe to the USA suggest that the USA population is not a direct descendant from the European P. violaceum population. There was a weak association between genetic and geographic distance among the USA isolates, suggesting invasion was initially localized prior to dispersal or that the population may have been present for some time prior to first detection in 2005. 相似文献

Evaluation of Food Additives and Low-toxicity Compounds for the Control of Bean Rust and Wheat Leaf Rust 总被引:1,自引:0,他引:1
The efficacy of low‐toxicity chemicals as possible alternatives to synthetic fungicides for the control of Uromyces appendiculatus and Puccinia triticina was evaluated. A preliminary selection of food additives was performed through in vitro and in vivo preliminary screenings. The ED50 and minimum inhibition concentration (MIC) values showed that most of the food additives used in this study were more toxic to U. appendiculatus than to P. triticina. Acetic acid, potassium carbonate, sodium carbonate and sodium molybdate were the food additives that were more toxic to the urediniospores of P. triticina. Selected compounds and concentrations were tested on bean and wheat plants grown in pots under controlled conditions. Acetic acid, ammonium bicarbonate, potassium acetate, potassium benzoate, potassium bicarbonate, potassium carbonate, sodium acetate and sodium citrate at 0.03, 0.09, 0.03, 0.006, 0.012, 0.012, 0.03 and 0.03 m , respectively, significantly reduced the disease severity of U. appendiculatus without causing any injury to bean leaves. Ammonium bicarbonate, potassium bicarbonate, sodium bicarbonate and sodium citrate at 0.12, 0.03, 0.12 and 0.03 m , respectively, were the most effective in reducing the disease severity caused by P. triticina without causing any injury to wheat leaves. 相似文献

The inheritance of resistance in red raspberry (Rubus idaeus) to yellow rust (Phragmidium rubi-idaei) was studied in a diallel cross among the cultivars Boyne, Meeker, Mailing Jewel, Glen Prosen and Glen Clova. The progenies and clonally propagated parents were exposed initially in a rust-infected plantation where the incidence and severity of infection was assessed at the telial stage. The following spring leaves on new canes of the same plants were inoculated with urediniospores in a glasshouse at 18 ± 2 °C. The latent period and number of uredinia per cm2 of abaxial leaf surface were determined 13 and 18 days after inoculation. Cv. Boyne developed only chlorotic flecking on inoculated leaves in the glasshouse and no sporulation was observed. It is postulated that this reaction was determined by a major gene which is designated Yr. Some of the other parents showed incomplete resistance of the slow rusting type, and segregants of Boyne selfed that were susceptible also showed low levels of rust, which suggests that Boyne carries both major- and minor-gene resistance. Diallel analyses of both the field and glasshouse data from the susceptible segregates indicated that the degree of incomplete resistance present was determined predominantly by additive gene action, though small but significant non-additive effects also occurred: cv. Meeker was the most resistant parent and contributed the most resistance to the progenies. The Meeker progenies also segregated for gene H, which determines cane hairiness and is known to be associated with resistance to other diseases. Segregants without hairs (genotypes hh) were found to be the more resistant to rust. 相似文献

Isolated and interactive effects of angular leaf spot (caused by Phaeoisariopsis griseola) and rust (caused by Uromyces appendiculatus) on leaf gas exchange and yield was studied in common bean (Phaseolus vulgaris L. cv. Carioca) plants. Gas exchange was measured on 37, 44, 51, and 58 d after planting using a portable photosynthesis system. The inoculation of plants with P. griseola (P), U. appendiculatus (U), and the combination of both pathogens (P+U) caused a significant reduction of net photosynthetic rate (P
N) and yield. The reduction of stomatal conductance (g
s), P
N, and yield was higher under P and combination of P+U than under U treatment. By effect of U, the reduction on yield was higher than the reductions on gas exchange parameters. On the treatment P+U, a reduction of 23 % in P
N and a correspondent reduction of 32 % in yield was observed. The interactive effects of the pathogens on yield could be explained in part by the decreases in g
s and in P

R. vestervicensis is known only from a small island off the Swedish east coast. It has been suggested that it is a hybrid between R. grabowskii and R. pedemontanus . We have compared these species with DNA fingerprinting and our results support the hybridization hypothesis. All bands found in the fingerprint of R. grabowskii and about half of the bands found in R. pedemontanus were present in R. vestervicensis , suggesting that the latter species was derived from fertilization of an unreduced R. grabowskii egg cell with R. pedemontanus pollen. 相似文献

To further investigate the phylogenetic relationship between Rubus coreanus and its relatives in the section Idaeobatus, we used genomic in situ hybridization (GISH) to ascertain the degree of their genomic homology. Genomic DNA from R. parvifolius and R. inopertus hybridized throughout the centromeric and sub-terminal regions on 14 and 12 chromosomes of R. coreanus, respectively. The probes from R. niveus and R. ellipticus var. obcordatus gave robust signals at the same region of eight chromosomes. R. ellipticus and R. pinfaensis generated strong signals at the centromeric and sub-terminal parts of six chromosomes. The hybridization signals from the R. tsangii and R. corchorifolius probes existed only at the telomeric parts of four chromosomes. The two signals at the sub-terminal region on chromosome 6 of R. coreanus might be 45S rDNA repeats. These results indicated that R. coreanus and R. parvifolius shared many repeat sequences. It could be deduced that the genome of R. parvifolius was most closely related to that of R. coreanus among the species tested, R. inopertus came next, while R. tsangii and R. corchorifolius showed the farthest relationship. Mature trees of eastern white, jack, Scotch, and shortleaf pines were inoculated with 25,000-34,000 pinewood nematodes, Bursaphelenchus xylophilus, isolated from infected Scotch pines in Missouri. Equal numbers of trees of each species inoculated with distilled water served as controls. Nine of fifteen Scotch pines died within 4 months of nematode infection or during the winter and early spring following infection. A single eastern white and shortleaf pine died. No jack pines died. A single Scotch pine control died, apparently the result of natural nematode infection. No other controls died. Mean oleoresin flow did not differ among nematode-inoculated jack and shortleaf pines and their respective controls. Oleoresin flow in nematode-inoculated eastern white and Scotch pines was significantly lower than in their controls. Oleoresin flow was temporarily reduced in mortality-resistant eastern white and Scotch pines following nematode infection. Thus a sublethal impact of nematode infection on mortality-resistant host trees was documented. 相似文献

Abstract The reproductive characteristics of three Rubus species ( R. palmatus Thunberg var. coptophyllus (A. Gray) O. Kuntze, R. microphyllus L.f., and R. crataegifolius Bunge) were critically investigated using materials collected from the forest floor of an 85-year-old Hinoki ( Chamaecyparis obtusa ) plantation in central Honshu, Japan. The underground branching patterns of these species were also investigated at the nursery. R. microphyllus produced the largest number of fruits in the same cane size among the three species (164 fruits per cane of 300 cm3 in D0 2H). However, the largest number of seeds produced per cane was in R. crataegifolius , where the maximum seed number attained 74,500 per cane, which was twice as many as those of the other two species. There was no apparent difference in dry matter allocation to fruits in these species. The three species produced numerous suckers by underground branching, forming large clones. In the open site at the nursery, the number of suckers produced by underground branching increased exponentially. On the other hand, in natural populations occurring in closed sites, the number of suckers produced by underground branching in the three species was by contrast much smaller. Three Rubus species proved to possess similar perennation as well as reproductive strategies, although R. palmatus var. coptophyllus shows much lower seed production and slower sucker production than the other two species. 相似文献

A method was developed for the inoculation of flowers of cut roses with conidia of Botrytis cinerea. Flower buds were inoculated by spraying of conidial supensions that were ultrasonicated for 10 s. The differences in susceptibility between 8 rose cvs to infections of isolate Bc-33 and the differences in pathogenicity between 14 5. cinerea isolates to cv. ‘Sonia’ were evaluated. Isolates obtained from rose flowers caused higher infection rates than those obtamed from various other hosts. The CVS ‘Madelon’, ‘Melody’ and ‘Sonia’ were found to be highly susceptible, whereas the cvs ‘Caramboie’, ‘Gabriella’, ‘Pasadena’ and ‘Rubinette’ were only slightly susceptible. The lower disease severity in the less susceptible cvs was based on a retardation of the growth of mfection hyphae in the petals, which may have been due to partial resistance. The formation of symptoms is effectuated already by the initial stages in the infection process. Thus, the apparent existing partial resistance to infections of B. cinerea cannot prevent the loss of ornamental value. 相似文献

Leymus mollis (Trin.) Pilger (NsNsXmXm, 2n = 28), a wild relative of common wheat, possesses many potentially valuable traits that could be transferred to common wheat during breeding programs. In this study, the karyotypic constitution of a wheat - L. mollis 3D(3Ns#1) disomic substitution line isolated from the F5 progeny of octoploid Tritileymus M842-16 x Triticum durum cv. D4286, which was designated as 10DM57, was determined using genomic in situ hybridization (GISH), fluorescent in situ hybridization (FISH), SSR markers, and EST- STS markers. Screening of mitosis and meiosis showed that 10DM57 had a chromosome karyotype of 2n = 42 =21Ⅱ. GISH indicated that 10DM57 was a line with 40 chromosomes from wheat and two of the Ns chromosomes from L. mollis, which formed a ring bivalent in pollen mother cells at metaphase I. FISH analysis showed that the chromosome 3D may be replaced by 3Ns#1 in 10DM57. DNA markers, including SSR and EST-STS primers, showed that the pair of wheat chromosome 3D in 10DM57 was substituted by the pair of chromosome 3Ns#t from L. mollis. Evaluation of the agronomic traits showed that, compared with its common wheat relative 7182, 10DM57 was resistant to leaf rust while the spike length and number of spikes per plant were improved significantly, which correlated with a higher wheat yield. The new germplasm, 10DM57, could be exploited as an intermediate material in wheat genetic and breeding programs. 相似文献

以高产小豆品种(系)‘2000-75’、‘冀红9218’和低产品种(系)‘红宝1号’、‘湾选1号’为材料,测定小豆开花至成熟期,根系与始花节位叶片生长指标、保护酶活性、可溶性蛋白含量等变化,分析其根系与始花叶协同衰老的相关性,以揭示小豆根系活力与地上部叶片衰老之间的关系。结果表明:(1)各品种(系)开花后,植株根系伤流强度随着花后生育进程的推进呈单峰增长趋势,但根系活力的衰老起始期晚于叶片功能的衰退开始期。(2)从开花至成熟期,小豆根系活力与叶片中超氧化物歧化酶、过氧化氢酶、叶绿素含量、可溶性蛋白、叶片净光合速率都存在正相关关系,与丙二醛含量呈负相关关系。(3)与低产品种相比,高产小豆品种(系)‘2000-75’和‘冀红9218’的根系活力强,叶片功能期持续时间长,叶绿素含量下降速度慢,保护性酶含量高,使花后小豆的功能叶捕获光能的能力增强,从而形成较高光合能力的小豆群体,最终获得高产。 相似文献

Leaf anatomy of 14 species in the subgenus lndocarex from China was studied. The anatomical characters are proved to be systematically valuable. (1) Characters of lamina transverse
section: All leaves of these 14 species are dorsiventral. The outline mostly V-shaped, occasionally flat or nearly flat, with adaxial lateral rib in each half of lamina and some of them
flanged. The cells of adaxial surface larger than those of abaxial surface, and the epidermal
cells over veins usually smaller than others. Air-cavities between vascular bundles are well developed, and bulliform cells also well developed in most taxa. The vascular bundles are collaterai, bundle sheaths double-layered, and the outer sheath parenchymatous and the inner
sheath fibrous. (2) Characters of lamina epidermis: The shape of the cell on both surfaces is
generally rectangular, and the anticlinal wall of epidermal cell sinuous; stomata is paracytic,
elliptic to oblong, rarely sub-circular; prickles occur on adaxial surfaces of certain species;
papillae are only obvious on abaxial surface of C. moupinensis Franch. The characters of
transverse section and epidermis of leaf blades of these 14 species differ from each other to
certain degree, and closely related species are similar in anatomical characters. The anatomical characters of lamina are of value for classification at specific and sectional level of the subgenus Indocarex. Despite of the variation of these characters among species, a certain num
ber of characters appears to be shared by the members of the subgenus, and some of the common characters are primitive. In addition, some gross morphological characters are common and primitive also. Therefore, the subgenus Indocarex may be primitive in the genus Carex. The anatomical and morphological characters of C. scaposa C. B. Clakre and C. densifimbriata Tang et Wang ex S. Y. Liang are distinct. The two species and their allies should be treated as section instead of subsection. Glasshouse and field trials were conducted to determine the efficacy of a talc-based powder formulation of Pseudomonas fluorescens strain Pf1 in controlling groundnut leaf spot ( Cercosporidium personatum ) and rust ( Puccinia arachidis ). Seed treatment with the talc-based powder formulation of the bacterium alone effectively reduced the severity of leaf spot and rust. When the treated seeds were sown in soil, the antagonist moved to the rhizosphere and multiplied well in it. Foliar application with the powder formulation effectively controlled the groundnut leaf spot and rust. P. fluorescens multiplied well in the phyllosphere after foliar application of powder formulation. Combined application of the P. fluorescens formulation to seed and foliage effectively controlled leaf spot and rust, and increased the pod yield in greenhouse and field tests. 相似文献

Fusarium oxysporum f. sp. albedinis, the bayoud disease agent of date palm, grows on a mineral medium containing the cell walls of date palm roots as a sole carbohydrate source. The growth and development of pathogen under these conditions was related to the production of extracellular cell wall-degrading enzymes (CWDE): cellulases, polygalacturonases, polygalacturonate transeliminases, and pectinmethylesterases. The mycelial growth and the sporulation of pathogen were higher in the presence of cell walls of susceptible cultivars (BFG, JHL, BSK) than in the presence of those of resistant cultivars (IKL, SLY, BSTN). After 8 d of fungal culture, the activity of CWDE was equal whatever is the origin of the cell walls (resistant or susceptible cultivars). After 16 d of culture, the activity of these CWDE was higher when the parasite was cultivated on the cell walls of the susceptible cultivars than on those of the resistant cultivars. A positive correlation was observed between CWDE activities and the growth and the sporulation of F. oxysporum after 16 d of culture. These results clearly show a relation between the susceptibility/resistance of the cell walls of the roots of the date palm to the parasitic CWDE and the susceptibility/resistance of the cultivars. 相似文献

Chromobacterium violaceum DSM 30191 accumulated a homopolyester of 3-hydroxyvaleric acid (3HV) up to 65% of the cellular dry matter during cultivation in fed-batch cultures with valeric acid as sole carbon source and during cell starvation of the nitrogen source. From fructose, gluconate, propionate or hexanoate a homopolyester of 3-hydroxybutyrate (3HB) was accumulated. Poly(3HV) homopolyster was also accumulated by two different strains of C. violaceum, whereas two other strains of C. violaceum and three strains of Janthinobacterium lividum accumulated poly(3HB-co-3HV) copolyesters from valerate. The composition of the biosynthetic poly(3HV) was confirmed by various nuclear magnetic resonance spectroscopic methods. Differential scanning calorimetry analysis of four poly(3HV) samples that were isolated from different batches of cells revealed glass transition temperatures between –10 and –12°C and melting points between 107 and 112°C. Viscosity measurements gave intrinsic viscosities between 62.5 and 124.8 × 10–2 dl/g for these samples, indicating approximate relative molecular masses between 60 000 and 145 000 of the biosynthetic poly(3HV).
