Spirodela polyrrhiza, a fast-growing duckweed with high starch and low lignin content, shows promise as a feedstock for bioenergy. Abscisic acid (ABA) is a biological hormone that controls plant growth and stress response. The effects of different ABA concentrations (0, 1.0 × 10?5, 1.0 × 10?4, 1.0 × 10?3, 1.0 × 10?2, and 1.0 × 10?1 mg/L) on duckweed biomass growth, carbon dioxide fixation, formation of photosynthetic pigments (Chlorophyll a (Chla), Chlorophyll b (Chlb), and carotenoids), the activities of soluble starch synthase (SSS) and starch branching enzyme (SBE), and the starch content of biomass were investigated in this study. ABA at concentrations lower than 1.0 × 10?3 mg/L promoted carbon dioxide fixation, whereas it inhibited carbon dioxide fixation at concentrations over 1.0 × 10?3 mg/L. ABA enhanced SSS and SBE activities at concentrations lower than 1.0 × 10?2 mg/L. ABA treatment increased the content of Chla, Chlb, and carotenoids and resulted in the enhancement of starch content. Chla content gradually increased with the increasing concentration of ABA (1.0 × 10?5 to 1.0 × 10?2 mg/L). After culturing for 10 days, starch content in 1.0 × 10?2 mg/L ABA medium reached 35.3% of dry weight (DW), which was the highest level in this study. This suggests that there is a great potential to develop a technology to increase starch accumulation in duckweed which can be used as an alternative to corn, sugarcane, or other food crops as a starch source.  相似文献

Graft copolymers (SPAN) of polyacrylonitrile (PAN) onto starch were prepared from gelatinized starch varieties with ammonium ceric nitrate as an initiator. The molecular weight of the PAN branches increased for the varieties of starches in the order high amylose maize starch < maize starch < waxy maize starch. SPAN samples were saponified with aqueous NaOH, and the aqueous solution of the resulting polymer (HSPAN) was cast into film in a forced-air oven at 35°C. The water absorbency of the HSPAN film formed from waxy maize starch was the highest (1200 g H2O (g dry sample)−1) and that from high amylose maize starch was the lowest (530 g g−1). SPAN samples from maize starch were partially hydrolyzed with dilute hydrochloric acid. The resulting polyacrylonitriles with low molecular weight starch end groups (LSPAN) were also saponified. The resulting saponified product (HLSPAN) was cast into film. The absorbencies of HLSPAN films were found to be far larger (up to 6000 g g−1) than those of the corresponding HSPAN films. The absorbency increased with increasing molecular weight of PAN in the initial SPAN up to a molecular weight of 1−1·5 × 106. The absorbency decreased significantly when HSPAN and HLSPAN films were subjected to heat treatment at 135°C or above. The crosslinks present in HSPAN and HLSPAN films prepared at 35°C and those formed during heat treatment were considered to have different structures: the former formed between carbohydrate alkoxide ions and nitrile groups at the early stages of saponification and the latter formed between carbohydrate and copoly(acrylate-acrylamide) chains and/or between copoly(acrylate-acrylamide) chains.  相似文献

Production of an amylase-degrading raw starch by Gibberella pulicaris   总被引:1,自引:0,他引:1  
An endophytic fungus, Gibberella pulicaris, produced an amylase which degraded raw starches from cereals and other crops including raw potato, sago, tapioca, corn, wheat and rice starch. In each case, glucose was the main product. Among the raw starches used, raw potato starch gave the highest enzyme activity (85 units mg–1 protein) and raw wheat starch the lowest (49 units mg–1 protein). The highest amylase production (260 units mg–1 protein) was achieved when the concentration of raw potato starch was increased to 60 g l–1. Optimum hydrolysis was at 40°C and pH 5.5.  相似文献

Carbon isotope ratios (expressed as δ13C values) were determined for various sources of starch and the starch fractions amylose and amylopectin. The δ13C values of amylose were consistently less negative, 0.4–2.3 ‰, than those of amylopectin in kernal starch from maize (Zea mays) and barley (Hordeum vulgare) and in tuber starch from potato (Solanum tuberosum). Kernel starch isolated from the maize mutants wx1 and ae1, with known genetic lesions in the starch biosynthetic pathway, also showed significant differences in δ13C values. Collectively, these results suggest that variation in carbon isotope ratios in the amylose and amylopectin components of starch may be attributed to isotopic discrimination by the enzymes involved in starch biosynthesis.  相似文献

Alcoholic fermentation from raw corn starch using Schizosaccharomyces pombe AHU 3179 and a raw starch saccharifying enzyme (RSSE) from Corticium rolfsii AHU 9627 was investigated. The optimum ethanol production was achieved at pH 3.5, 27°C and under the yeast cell concentration of 2.7 × 109 cells/ml. Addition of RSSE 5 units (as glucoamylase)/g raw corn starch was found sufficient. Under these optimum conditions, 18.5% (v/v, at 15°C) ethanol was obtained from 30% raw corn starch (30.8% as glucose) after incubation for 48 h.  相似文献

Sporulation of Coniothyrium minitans on 20 g glucose/l or 18 g starch/l at an initial C/N ratio of 10 was the lowest for nitrate and histidine (1 × 109 vs 5–6 × 109 spores/dish). At 100 g glucose/l or 90 g starch/l lower spore numbers as compared to the lower concentrations are obtained for all nitrogen sources tested; only the spore numbers with urea and glycine were equal with either 18 or 90 g starch/l. Spore numbers at 90 g starch/l were all a factor two or more higher than at 100 g glucose/l.  相似文献

The control of photosynthetic starch/sucrose formation in leaves of soybean (Glycine max L. Merr.) cultivars was studied in relation to stage of plant development, photosynthetic photoperiod, and nitrogen source. At each sampling, leaf tissue was analyzed for starch content, activities of sucrose-metabolizing enzymes, and labeling of starch and sucrose (by 14CO2 assimilation) in isolated cells. In three of the four varieties tested, nodulated plants had lower leaf starch levels and higher activities of sucrose phosphate synthetase (SPS), and isolated mesophyll cells incorporated more carbon (percentage of total 14CO2 fixed) into sucrose and less into starch as compared to nonnodulated (nitrate-dependent) plants. The variation among cultivars and nitrogen treatments observed in the activity of SPS in leaf extracts was positively correlated with labeling of sucrose in isolated cells (r = 0.81) and negatively correlated with whole leaf starch content (r = −0.66). The results suggested that increased demand for assimilates by nodulated roots may be accommodated by greater partitioning of carbon into sucrose in the mesophyll cells. We have also confirmed the earlier report (Chatterton, Silvius 1979 Plant Physiol 64: 749-753) that photoperiod affects partitioning of fixed carbon into starch. Within two days of transfer of nodulated soybean Ransom plants from a 14-hour to a 7-hour photoperiod, leaf starch accumulation rates doubled, and this effect was associated with increased labeling of starch and decreased labeling of sucrose in isolated cells. Concurrently, activities of SPS, sucrose synthase, and uridine diphosphatase in leaves were decreased.  相似文献

The effect of starch addition on the microbial composition and the biological conversion was investigated using two upflow anaerobic sludge bracket (UASB) reactors treating methanolic wastewater: one reactor was operated with starch addition, and another reactor was operated without starch addition. Approximately 300 days of operation were performed at 30 kg COD/m3/d, and then, the organic load of the reactors was gradually increased to 120 kg COD/m3/d. Successful operation was achieved at 30 kg COD/m3/d in both reactors; however, the methanol-fed reactor did not perform well at 120 kg COD/m3/d while the methanol-starch-fed reactor did. The granule analysis revealed the granule developed further only in the methanol-starch-fed reactor. The results of the microbial community analysis revealed more Methanosaeta cells were present in the methanol-starch-fed reactor, suggesting the degradation of starch produced acetate as an intermediate, which stimulated the growth of Methanosaeta cells responsible for the extension of granules.  相似文献

Cellular metabolite analyses by 13C-NMR showed that C. reinhardtii cells assimilate acetate at a faster rate in heterotrophy than in mixotrophy. While heterotrophic cells produced bicarbonate and CO2 aq, mixotrophy cells produced bicarbonate alone as predominant metabolite. Experiments with singly 13C-labelled acetate (13CH3-COOH or CH3-13COOH) supported that both the 13C nuclei give rise to bicarbonate and CO2 aq. The observed metabolite(s) upon further incubation led to the production of starch and triacylglycerol (TAG) in mixotrophy, whereas in heterotrophy the TAG production was minimal with substantial accumulation of glycerol and starch. Prolonged incubation up to eight days, without the addition of fresh acetate, led to an increased TAG production at the expense of bicarbonate, akin to that of nitrogen-starvation. However, such TAG production was substantially high in mixotrophy as compared to that in heterotrophy. Addition of mitochondrial un-coupler blocked the formation of bicarbonate and CO2 aq in heterotrophic cells, even though acetate uptake ensued. Addition of PSII-inhibitor to mixotrophic cells resulted in partial conversion of bicarbonate into CO2 aq, which were found to be in equilibrium. In an independent experiment, we have monitored assimilation of bicarbonate via photoautotrophy and found that the cells indeed produce starch and TAG at a much faster rate as compared to that in mixotrophy and heterotrophy. Further, we noticed that the accumulation of starch is relatively more as compared to TAG. Based on these observations, we suggest that acetate assimilation in C. reinhardtii does not directly lead to TAG formation but via bicarbonate/CO2 aq pathways. Photoautotrophic mode is found to be the best growth condition for the production of starch and TAG and starch in C. reinhardtii.  相似文献

Energy crises and environmental pollution have caused considerable concerns; duckweed is considered to be a promising new energy plant that may relieve such problems. Lemna aequinoctialis strain 6000, which has a fast growth rate and the ability to accumulate high levels of starch was grown in both Schenk & Hildebrandt medium (SH) and in sewage water (SW). The maximum growth rates reached 10.0 g DW m−2 day−1 and 4.3 g DW m−2 day−1, respectively, for the SH and SW cultures, while the starch content reached 39% (w/w) and 34% (w/w). The nitrogen and phosphorus removal rate reached 80% (SH) and 90% (SW) during cultivation, and heavy metal ions assimilation was observed. About 95% (w/w) of glucose was released from duckweed biomass hydrolysates, and then fermented by Angel yeast with ethanol yield of 0.19 g g−1 (SH) and 0.17 g g−1 (SW). The amylose/amylopectin ratios of the cultures changed as starch content increased, from 0.252 to 0.155 (SH) and from 0.252 to 0.174 (SW). Lemna aequinoctialis strain 6000 could be considered as valuable feedstock for bioethanol production and water resources purification.  相似文献

Present study aims to optimize the production of starch and total carbohydrates from Arthrospira platensis. Growing concerns toward unprecedented environmental issues associated with plastic pollution has created a tremendous impetus to develop new biomaterials for the production of bioplastic. Starch-based biopolymers from algae serve as sustainable feedstock for thermoplastic starch production due to their abundant availability and low cost. A. platensis was cultivated in Zarrouk's medium at 32 ± 1°C and exposed to red light with a photoperiod of 12:12 hr light/dark. Growth kinetics studies showed that the maximum specific growth rate (μmax) obtained was 0.059 day−1 with the doubling time (td) of 11.748 days. Subsequently, Zarrouk's medium with different concentrations of sulfur, phosphorus and nitrogen was prepared to establish the nutrient-limiting conditions to enhance the accumulation of starch and total carbohydrates. In this study, the highest starch accumulated was 6.406 ± 0.622 mg L−1 under optimized phosphorus limitation (0.025 g L−1) conditions. Nitrogen limitation (0.250 g L−1) results demonstrated significant influenced (p < 0.05) on total carbohydrates (67.573 ± 2.893 mg L−1) accumulation in A. platensis. The starch accumulation in A. platensis was significantly affected (p < 0.05) by phosphorus limitation (0.0025 g L−1). Subsequently, the optimized phosphorus concentration was coupled with mixotrophic cultivation to further enhance the starch accumulation. The results obtained indicated that, the starch (11.426 ± 0.314 mg L−1) and carbohydrates (43.053 ± 2.986 mg L−1) concentration obtained was significantly high (p < 0.05) under mixotrophic cultivation. Therefore, it shown that nutrient limitation and mixotrophic cultivation are viable strategies to enhance the accumulation of starch and total carbohydrates in A. platensis.  相似文献

Summary Production of lipid from starch in a two-component mixed culture in a fed batch process was studied. Saccharomycopsis fibuliger was used as the amylolytic microorganism and Rhodosporidium toruloides, which does not grow on starch, as the lipid producer. The concentration of nitrogen can be used as an external regulator of growth and competition in this process. The total biomass concentration decreased and the relative amount of Rh. toruloides increased with decreasing initial concentration of nitrogen in the medium. The highest lipid concentration (9.7 g·l-1), highest overall lipid production rate (0.15 g·l-1·h-1) and highest concentration of lipid in biomass (36.5%) were obtained in cultures with an initial nitrogen concentration of 0.5 g·l-1. Compared to monocultures of Lipomyces starkeyi and Aspergillus oryzae on starch, the mixed culture showed slightly lower conversion of starch to lipid but was superior in the final lipid concentration and the overall lipid production rate.  相似文献

In this experiment, bread wheat flour and isolated wheat starch were treated with ozone gas (1,500 mg/kg at 2.5 L/min) for 45 min and 30 min, respectively. Starch was isolated from treated flour. Ozone treated starch and starch isolated from ozone treated flour had similar chemical and physical properties. Chemical analysis of starch isolates indicated depolymerization of high molecular weight amylopectins; with a subsequent increase in low molecular weight starch polymers as a result of starch hydrolysis. Ozone treatment resulted in elevated levels of carboxylic groups and decreased total carbohydrate content in amylopectin fractions. 1H NMR results indicated formation of a keto group [(1→4)-3 keto] at the H-2 terminal (proton at C-2 position) and β-glucuronic acid at the H-1 terminal (proton at C-1 position). DSC transition temperatures and change in enthalpy were not affected by ozone treatment. Increased swelling power and RVA breakdown were observed in starch from ozone treated samples.  相似文献

A commercial preparation of -amylase, Biotempase, obtained from Biocon India Pvt. Ltd., and crude glucoamylase produced from Aspergillus sp. NA21 were used to hydrolyse tapioca powder, a non-conventional starchy substrate. Among various concentrations of starch (15–35%, dry weight/volume) tried for maximum liquefaction; slurry made with 25% substrate concentration proved optimal. An economical process of liquefaction was carried out using steam under pressure (0.2–0.3 bar, 104–105 °C) to liquefy a 25% slurry in just 45 min, contrary to a slower process carried out at 95 °C in a water bath. For liquefaction of starch a pH of 5.0 proved to be optimum. The dose of Biotempase as prescribed by the supplier could be reduced by 33% achieving the same degree of liquefaction, by addition of CaCl2 to the starch slurry at the concentration of 120 mg/l. The conditions for the saccharification of liquefied starch were optimized to be 60 °C and pH 5.0, producing 90% saccharification in 24 h. Supplementation of divalent ions Ca2+, Mg2+ and Zn2+ in the process of saccharification showed no effect. Finally glucose was found to be the main hydrolysis product in the saccharification of tapioca starch.  相似文献

The regulation of starch synthesis and exocellular polysaccharide synthesis by GA3 was studied with cells of sweet potato grown as suspension in glycerol medium. In the presence of GA3, and under normal cell growth, starch formation was inhibited. The incorporation activity (starch synthesis) from ADP-[14C] glucose or UDP-[14C] glucose with GA3 treated cells was reduced. On the other hand, the synthesis of exocellular polysaccharides composed of glucose, galactose, mannose and arabinose etc., was stimulated and a clear increase of the Man/Ara ratio was observed in the presence of GA3. These results may indicate that GA3 affects the regulation of starch synthesis and exocellular polysaccharide synthesis.  相似文献

Carboxymethyl starch (CMS) was obtained as a product of the reaction of starch and monochloroacetic acid (MCA) in the presence of sodium hydroxide. The influence of the molar ratio of NaOH/AGU, the molar ratio of MCA/AGU, the reaction time, reaction temperature, and the water content on the degree of substitution (DS) was studied. The optimal molar ratio of NaOH/AGU and MCA/AGU is 2.4 and 1.0, respectively. Increase of the ratio of NaOH/AGU or MCA/AGU leads to an increase in DS, but only to certain extent. The highest values of the DS were obtained when the carboxymethylation was performed at 60 °C for 2.5 h. The water content in the reaction media ethanol was optimal at 20% (v/v). The scanning electron micrographs (SEMs) revealed that the carboxymethylation affected the structural arrangement of the starch and caused granular disintegration. The particle size distribution (PSD) also displayed that the average particle diameter increased greatly after modification from 37.37 μm to 72.88 μm. Wide angle X-ray diffractometry (XRD) revealed that starch crystallinity was obviously reduced after carboxymethylation. The new bands at 1600 cm−1 and 1426 cm−1 in FT-IR indicated that the starch granules were substituted.  相似文献

Red algae (Rhodophyceae) are photosynthetic eukaryotes that accumulate starch granules in the cytosol. Starch synthase activity in crude extracts of Gracilaria tenuistipitata Chang et Xia was almost 9-fold higher with UDP[U-14C]glucose than with ADP[U-14C]glucose. The activity with UDP[U-14C]glucose was sensitive to proteolytic and oxidative inhibition during extraction whilst the activity with ADP[U-14C]glucose appeared unaffected. This indicates the presence of separate starch synthases with different substrate specificities in G. tenuistipitata. The UDPglucose: starch synthase was purified and characterised. The enzyme appears to be a homotetramer with a native Mr of 580 kDa and displays kinetic properties similar to other α-glucan synthases such as stimulation by citrate, product (UDP) inhibition and broad primer specificity. We propose that this enzyme is involved in cytosolic starch synthesis in red algae and thus is the first starch synthase described that utilises UDPglucose in vivo. The biochemical implications of the different compartmentalisation of starch synthesis in red algae and green algae/plants are also discussed. Starch synthesis and CO2 evolution were determined after incubating intact and lysed wheat (Triticum aestivum L. cv. Axona) endosperm amyloplasts with 14C-labelled hexose-phosphates. Amyloplasts converted [U-14C]glucose 1-phosphate (Glc1P) but not [U-14C]glucose 6-phosphate (Glc6P) into starch in the presence of ATP. When the oxidative pentose-phosphate pathway (OPPP) was stimulated, both [U-14C]Glc1P and [U-14C]Glc6P were metabolized to CO2, but Glc6P was the better precursor for the OPPP, and Glc1P-mediated starch synthesis was reduced by 75%. In order to understand the basis for the partitioning of carbon between the two potentially competing metabolic pathways, metabolite pools were measured in purified amyloplasts under conditions which promote both starch synthesis and carbohydrate oxidation via the OPPP. Amyloplasts incubated with Glc1P or Glc6P alone showed little or no interconversion of these hexose-phosphates inside the organelle. When amyloplasts were synthesizing starch, the stromal concentrations of Glc1P and ADP-glucose were high. By contrast, when flux through the OPPP was highest, Glc1P and ADP-glucose inside the organelle were undetectable, and there was an increase in metabolites involved in carbohydrate oxidation. Measurements of the plastidial hexose-monophosphate pool during starch synthesis and carbohydrate oxidation indicate that the phosphoglucose isomerase reaction is at equilibrium whereas the reaction catalysed by phosphoglucomutase is significantly displaced from equilibrium. Optimum conditions for glucose syrups production from white sorghum were studied through sequential liquefaction and saccharification processes. In the liquefaction process, a maximum dextrose equivalent (DE) of 10.98 % was achieved using 30 % (w/v) of starch and Termamyl ɑ-amylase from Bacillus licheniformis. Saccharification was performed by free and immobilized amyloglucosidase from Rhizopus mold at 1 % (w/v). DE values of 88.32 % and 79.95 % were obtained from 30 % (w/v) of starch with, respectively, free and immobilized enzyme. The immobilized Amyloglucosidase in calcium alginate beads showed reusable capacity for up to 6 cycles with 46 % of the original activity retained. The kinetic behaviour of immobilized and free enzyme gives Km value of 22.13 and 16.55 mg mL−1 and Vmax of 0.69 and 1.61 mg mL−1 min−1, respectively. The hydrolysis yield using immobilized amyloglucosidase were lower than that of the free one. However, it is relevant to reuse enzyme without losing activity in order to trim down the overall costs of enzymatic bioprocesses as starch transformation into required products in industrial manufacturing. Hydrolysis of sorghum starch using immobilized amyloglucosidase represents a promising alternative towards the development of the glucose syrups production process and its utilization in various industries.  相似文献

In the alkaliviscogram of starch of 26 nonwaxy rices grown in the tropics, gelatinization normality correlated positively with final gelatinization temperature (BEPT) of starch (r=0.969**) and negatively with alkali spreading value of milled rice (r= ?0.931**). Peak viscosity was not linearly related to amylose content. Among samples of rice starch having a high amylose (>28%) content, peak viscosity was correlated with the gel consistency of starch (r=?0.690**) and of milled rice (r=?0.644**) (n = 18). These high-amylose starches showed the widest variation in peak viscosity. Amylose content, and gel consistency were inherited from the same parent in all nine varieties and lines studied, whereas peak viscosity, gelatinization normality and the final BEPT were inherited from either parent. The starch of five waxy rices showed higher peak viscosities even at a concentration of 1.8% as compared with a 2.0% nonwaxy rice starch.  相似文献
