Sporostatic and Sporocidal Properties of Aqueous Formaldehyde   总被引:5,自引:1,他引:4  
Aqueous formaldehyde is shown to exert both sporostatic and sporocidal effects on Bacillus subtilis spores. The sporostatic effect is a result of the reversible inhibition of spore germination occasioned by aqueous formaldehyde; the sporocidal effect is due to temperature-dependent inactivation of these spores in aqueous formaldehyde. The physicochemical state of formaldehyde in solution provides a framework with which to interpret both the sporostatic and sporocidal properties of aqueous formaldehyde.  相似文献

Summary The level of inhibition of germination of spores added to cell-free culture filtrates of Fusarium oxysporum Schlecht. ex Fr. declines with time. This decline can be accounted for by the decreasing levels of volatile sporostatic factors and a stable vacuolation factor produced by F. oxysporum. After the disappearance of the volatile sporostatic factors, a stable principle is recognised in the culture filtrate by a persistent level of inhibition of spore germination which is not completely alleviated by the addition of nutrients. A stable inhibitor of spore germination has been isolated and characterised as nonanoic acid, and evidence is presented that this compound is widely produced among fungi and can inhibit the germination of spores of many fungal species. The contribution of nonanoic acid to sporostasis in cultures of F. oxysporum is assessed.  相似文献

The bacteriocin, boticin E, was produced by only a few strains of those clostridia which are nontoxigenic but otherwise identical to Clostridium botulinum type E. Boticin preparations from four different strains had identical spectra against indicator cultures. Experiments with bacterial lawns showed boticin to be sporostatic for all tested nonproteolytic C. botulinum (types B, E, and F) and nontoxigenic type E-related strains which included the producing strains as well as those different from type E in the fermentation of one to three carbohydrates. Boticin had no detectable effect on vegetative cells of boticinogenic strains but killed those of all other strains whose spores were sensitive. Cultures that were growing in an agar medium were more sensitive to the bacteriocin than those growing in broth. Vegetative cells of indicator strains adsorbed boticin, but cells of a boticin-resistant mutant did not. Boticin did not lyse suspensions of vegetative cells which had been killed previously by exposure to air but lysed actively growing protoplasts and L-forms of a strain whose normal vegetative cells are susceptible to lysis. Sporostasis resulted from inhibition of germination rather than of outgrowth. Proteolytic strains of C. botulinum (types A, B, and F) were resistant to boticin E.  相似文献

Two bacteriocins (boticins) were elaborated without induction by strain S5, a nontoxigenic variant of Clostridium botulinum type E. After separation of the two active entities by gel filtration on Sephadex G-50, a large particle with boticin activity was isolated by density gradient ultracentrifugation, and a small soluble boticin was purified by continuous curtain electrophoresis and chromatography on sulfoethyl-Sephadex. Large and small boticins were purified 200- and 3,000-fold, respectively, with yields of 50% or more. The small boticin, a basic substance with molecular weight under 30,000, was the predominant species; the large boticin, a negatively charged particle with particle weight greater than 40 x 10(6), represented less than 20% of the total activity. Both purified boticins were resistant to heat and were attacked by proteolytic enzymes, but the large boticin was less thermostable and less sensitive to proteolytic enzymes than was the smaller variety. The activity of the large boticin was not reduced by treatment with urea or deoxyribonuclease. Both boticins exhibited sporostatic and bactericidal activities for C. botulinum type E, strain 070. A suspension of type E strain 070 vegetative cells was rendered nonviable within 9 min by the small boticin. The lethal action of this bacteriocin was not reversed by trypsin.  相似文献

The use of rosemary essential oil (RO) and its combination with nisin (RO+N) in preventing the multiplication of Alicyclobacillus acidoterrestris in orange juice was evaluated. The minimum inhibitory and bactericidal concentrations (MIC and MBC) for RO were both 125 μg ml−1 while RO+N displayed a synergistic effect. The use of RO and RO+N at concentrations of 1, 4 and 8× MIC in orange juice for 96 h was evaluated in terms of their sporicidal effectiveness. With regard to the action against A. acidoterrestris spores, RO at 8× MIC was sporostatic, whereas RO+N at 1× MIC was sporicidal. Morphological changes in the structure of the micro-organism after treatment were also observed by microscopy. Furthermore, flow cytometric analysis showed that most cells were damaged or killed after treatment. In general, the antioxidant activity after addition of RO+N decreased with time. The results demonstrate that using the combination of RO and nisin can prevent the A. acidoterrestris growth in orange juice.  相似文献

应用逐步回归法,分析了5~9月松嫩盐碱草地植物群落的生物量、丰富度、多样性和均匀度与同月土壤因子间的关系。松嫩盐碱草地植物群落特征受若干土壤因子的共同影响,不同月份影响因子的变化较复杂。5~7月土壤盐分和养分共同影响着松嫩盐碱草地植物群落生物量,但是土壤盐分因子与群落生物量的相关性更明显(5月的Ca2+质量分数,6月和7月的Mg2+质量分数)。土壤盐分因子在整个生长季内对松嫩盐碱草地植物群落生物量的直接作用都大于土壤养分因子的。6月时影响松嫩盐碱草地植物群落丰富度、Simpson指数和Shannon指数的土壤养分因子增加,其中土壤全氮质量分数的直接作用是所有土壤因子中最大的。8月植物群落生物量、Simpson指数和Shannon指数仅受土壤盐分因子影响,其中生物量与土壤pH值以及Simpson指数与土壤碱化度都是极显著负相关,Shannon指数与土壤含盐量显著负相关。6~9月的松嫩盐碱草地植物群落均匀度也同时受土壤养分和盐分因子的作用,其中6月和9月时土壤盐分因子对群落均匀度的直接作用更突出。生长末期(9月)松嫩盐碱草地植物群落生物量与土壤因子间的关系以及生长初期(5月)植物群落均匀度与土壤因子间的关系都不明显。  相似文献

为探究太湖流域丘陵区典型土地利用类型(如竹林地和茶园)土壤水分的控制因素,在不同深度土壤水分定期观测的基础上,根据前7d降雨量将研究时段划分为干旱状态和湿润状态,利用分类与回归树(CART)方法得出不同干湿状态下土壤水分分布的主控因子,并借助典范对应分析(CCA)定量分析不同土地利用类型、不同土壤深度土壤水分格局与环境因子关系。结果表明:(1)高程、土地利用类型和土层厚度对土壤水分分布的相对贡献率最大,但在不同干湿状态下其影响程度存在差异;(2)干旱状态时土壤水分主要受高程、坡度、地形湿度指数(TWI)和剖面曲率等地形因素的作用,而土层厚度和粘粒也分别为0—20 cm和20—40 cm深度土壤水分的主控因子;(3)在湿润状态下,茶园0—20 cm土壤水分的主控因素为地形因子,在20—40 cm则以土壤性质为主,竹林地两个深度的土壤水分受地形和土壤性质的作用都很强,其中20—40 cm深度土壤水分与环境因子的关系较0—20 cm深度更为复杂。  相似文献

【目的】调查宁夏银北盐碱地油葵田的土壤动物多样性及群落结构,探讨不同改良时间盐碱地土壤动物多样性及群落结构与环境因子之间的相关性。【方法】生物与环境因子之间的关系研究一直是生态学研究的热点,尤其是恢复生态学中如何设计环境因子来恢复生物群落的结构和功能最近引起了国内外学者的关注。以宁夏银北盐碱地油葵田为对象,研究了不同改良时间盐碱地中油葵田土壤动物与环境因子的相互关系,采用野外调查和室内试验相结合。【结果】共采集12个土壤动物类群,优势类群为弹尾目(60.67%)、蜱螨亚纲(23.24%)和鞘翅目(6.45%);土壤动物物种多样性、丰富度指数和均匀度在未改良盐碱地中较高。【结论】土壤pH值、碱化度和全盐质量分数是土壤动物分布的限制因素,土壤动物的数量随土壤pH值、碱化度和全盐质量分数的降低而增加。这些结果为盐碱地改良的恢复与评价提供了重要的数据支持和基础资料。  相似文献

地表和地下土壤动物群落空间格局及其与环境因子的空间作用关系,是揭示地表-地下生态系统格局与过程及生物多样性维持机制的重要基础。于2011年在三江平原农田生态系统,在50m×50m的空间尺度内,基于地统计空间分析方法,揭示地表和地下土壤螨群落及不同螨类物种丰富度的空间格局,并分析这种空间格局与土壤含水量、土壤p H值及大豆株高空间格局的空间关联性。半方差函数和普通克里格插值表明,8月份地表、地下和10月份地下螨群落及这些群落内大部分螨类物种在特定空间尺度内形成集群,表现为空间异质性特征,且这种空间分异多由结构性因素或结构性因素和随机性因素共同调控。交叉方差函数表明,土壤螨群落和不同螨类物种的空间格局与环境因子的空间格局在多种尺度上表现出复杂的空间关联性(正的或负的)。但简单Mantel检验仅发现8月份地表中气门亚目未定种1(Mesostigmata unidentified sp.1)和大豆株高存在明显的正的空间关联性。研究结果表明地下螨群落和生长季节的地表螨群落具有明显的空间异质性结构,地表和地下螨群落及大多数螨类物种丰富度与土壤含水量、土壤p H值及大豆株高的空间关联性并不显著。促进地表-地下生态系统土壤动物群落空间格局研究,为地表-地下格局与过程研究奠定基础。  相似文献

土壤动物与土壤生态系统的环境要素密切相关,探明土壤动物与环境要素之间的相互关系对深刻认识土壤生态系统的运行机制、评价土壤生态系统功能具有重要意义.影响土壤动物的环境要素可以分为土壤性质和土壤外部环境两方面,前者主要指土壤基本理化性质、土壤水分和土壤污染状况,后者包括植被、土地利用、地貌和气候等环境要素.本文从以上角度对我国土壤动物与土壤环境要素关系研究进行总结和归纳,认为当前我国相关研究中主要存在以下若干问题:与环境指示作用整合性研究较少,研究方法有待改进,对环境要素的综合性及其时空变异性认识不足.并提出了相应的建议:与实际研究需要相结合,借鉴国外先进方法经验,进行多要素综合研究以及开展大区域长期监测等.  相似文献

通过野外选择样地,设置样方,调查了温带地区松嫩草原区农牧林复合系统中小型土壤动物群落的生态特征.结果表明,中小型土壤动物的优势类群是球角跳虫科(Hypogastruridae)、辐螨(Actinedida)、革螨(Gamasida)和甲螨(Oribatida),常见类群是节跳虫科(Isotomidae)和叶蝉科(Jassidae),从玉米田、草地和杨树林3个子系统的对比分析看,草地物种最丰富,多样性最高,生境条件最优越,中小型土壤动物的个体数和类群数随土层深度的增加而递减,表聚性明显;但3个子系统的垂直分布特征不完全相同,运用主成分分析方法,分析了主要土壤环境要素和气侯要素与中小型土壤节肢动物群落的相关关系,发现土壤环境要素是影响中小型土壤动物群落的主导因子。  相似文献

农田土壤固碳速率是评价土壤固碳效应和潜力的重要指标,精确估算区域农田土壤固碳速率对土壤地力及环境效应均具有重要意义.本研究选取黄淮海平原典型潮土区河南省封丘县为研究区域,按照土壤利用-土壤类型联合单元布点法,于2011年采集了70个耕层土样,测定了土壤有机碳含量、机械组成、容重、pH,并与全国第二次土壤普查(1981年)数据进行对比分析,结合地统计方法和GIS技术研究了该地区近30年农田土壤固碳速率的空间变异特征,利用显著性检验、回归分析、方差分析等方法定量分析了该区域农田土壤固碳速率的影响因素.结果表明: 近30年封丘县域土壤固碳速率平均值为0.33 t C·hm-2·a-1,变异系数为74%,属于中等变异性;土壤固碳速率的变化在东西方向上表现为西高东低、中部高南北低,呈片状分布,区域结构性因素是引起农田土壤固碳速率空间分布差异的主导因素,如土壤类型、机械组成、容重、pH,可解释空间变异的59.5%,其次是随机性因素,如秸秆还田量、施肥量,可解释空间变异的40.5%.  相似文献

Because of the increasing emphasis placed upon land application as a means of wastewater disposal, it is important to evaluate the influences of different factors upon virus survival in soil. The objective of this study was to measure the effects of various environmental variables on virus persistence. Test samples of soil were placed in vials, and the soil was wetted with suspensions of virus in either distilled water, unchlorinated secondary sewage effluent, or mixtures of effluent and water. The viruses used were coxsackieviruses A9 and B3, echovirus 1, poliovirus 2, rotavirus SA11, and bacteriophages T2 and MS2. The rate of virus inactivation was evaluated statistically with regard to conditions under which the vials were incubated and to the soil characteristics. The factors that were found to influence virus survival were temperature, soil moisture content, presence of aerobic microorganisms, degree of virus adsorption to the soil, soil levels of resin-extractable phosphorus, exchangeable aluminium, and soil pH. Overall, temperature and virus adsorption to soil appeared to be the most important factors affecting virus survival.  相似文献

荒漠植物红砂叶绿素和脯氨酸累积与环境因子的相关分析   总被引:13,自引:0,他引:13  
通过测定中国境内荒漠植物红砂(Reaumuria soongorica)主要分布区内21个自然种群407株叶片的叶绿素和脯氨酸含量,以及不同种群内土壤含水量、可溶性盐分含量、有机质、全氮、全磷含量等土壤理化性状指标,分析了不同自然种群红砂叶绿素和脯氨酸含量的变异特征以及影响其变化的主要环境因子.研究结果表明:红砂种群间叶绿素含量差异显著.土壤因子对叶绿素合成的影响较气象因子大,而土壤含水量和土壤全磷含量是导致不同红砂种群叶绿素含量出现明显差异的主要原因.红砂脯氨酸含量平均值显著高于盐爪爪、骆驼刺、芨芨草等其它荒漠物种,并且与叶片含水量和土壤含水量呈显著负相关,与土壤可溶性盐分含量呈显著正相关.红砂体内脯氨酸的累积是对干旱盐渍环境的适应性反应,与抗旱性和抗盐性之间存在着一定的关系.  相似文献

面对全球变化,准确掌握陆地森林生态系统生物地球化学循环过程及其影响因素,有助于维持生态系统多功能性与稳定性,实现增汇并延长碳汇服务,持续增进并改善人民福祉。为了探究贺兰山山地森林生态系统优势建群种青海云杉(Picea crassifolia)林土壤生态化学计量特征对海拔的生态响应及其影响因素,从土壤、林分、地形影响因素的角度出发,采用冗余分析(Redundancy analysis, RDA)和方差分解分析(Variance partitioning analysis, VPA)阐明影响其非根和根围土壤生态化学计量特征空间变异的关键因素。结果表明:(1)贺兰山青海云杉林非根和根围土壤C、N养分含量均维持在较高水平,高海拔区域青海云杉生长受土壤P限制高;相较非根土壤,根围土壤虽养分含量较低,但具有较高的养分周转能力,如有机质分解与矿化、固持P的潜力等。(2)随海拔变化,非根和根围土壤养分及其生态化学计量比变化趋势不同,非根土除了全氮(TN)含量外其他养分含量及化学计量比在不同海拔间均差异显著(P<0.05),根围土全磷(TP)、碳磷比(C/P)具有显著性差异(P>0.05)。...  相似文献

通过对土壤肥力与板栗枝干病害关系的研究表明,在土壤物理因素中,土壤类型以粘土、石砾土的栗园发病较重,壤土、沙壤土、黄棕壤的栗园发病较轻;土壤容重与感病指数呈正相关;土层厚度与感病指数呈负相关。在土壤化学因素中,有机质、速效磷与病情呈负相关;pH值、全氮和钾含量与病情关系不显著,并通过对土壤肥力因素与病情关系的逐步回归分析,筛选出土层厚度、土壤容重、粘性土、有机质和速效磷5个因素对病情贡献较大。以此建立了病情预测的回归模型,根据这一模型,可以预测研究范围内的任一给定条件下病害可能发生的水平,该研究将为今后的板栗生产和管理提供指导。  相似文献

为了揭示不同产地土壤因子对关黄柏酚酸类、生物碱类和柠檬苦素类化学成分含量的影响,本研究对31个样点的15个土壤因子及所产药材的10种化学成分含量进行检测。通过冗余分析考察土壤因子与化学成分间的相关性,用t-value双序图解读各土壤因子对各化学成分含量的影响,并通过聚类分析考察样点土壤特征对药材化学特征的影响。结果显示:土壤中的盐分、速效磷、阳离子交换量、速效氮、速效钾、有机质、有效锰、硫和有效铜对关黄柏化学成分含量有显著影响;3-O-阿魏酰奎尼酸、4-O-阿魏酰奎尼酸、5种生物碱、黄柏内酯含量随上述土壤因子含量的上升呈下降趋势;紫丁香酚苷和黄柏酮主要呈上升趋势。研究结果表明环境土壤因子的差异会导致关黄柏多种化学成分的含量发生显著性改变。  相似文献

土壤呼吸作用作为陆地生态系统碳循环的重要组成部分,是当前碳循环研究中的热点问题.对于土壤呼吸作用主要组成部分土壤微生物呼吸作用和根系呼吸作用影响因子的研究,有助于准确地评估全球碳收支.本文从气候、土壤、植被及地表覆被物、大气CO2浓度、人为干扰等方面综述了土壤微生物呼吸作用和根系呼吸作用的主导影响因子,指出这些影响因子不仅直接或间接地影响土壤微生物呼吸作用和根系呼吸作用,而且它们之间相互作用、相互影响,且各影响因子的地位和作用会随时空尺度变化发生相应改变.在此基础上,论文提出了未来土壤呼吸作用的研究重点.  相似文献

This study was performed to evaluate the effects of Korean propolis against foodborne pathogens and spores of Bacillus cereus and to investigate the antimicrobial activity against B. cereus structure by transmission electron microscopy (TEM). The antimicrobial effects of the Korean propolis were tested against foodborne pathogens including Gram-positive (B. cereus, Listeria monocytogenes and Staphylococcus aureus) and Gram-negative (Salmonella typhimurium, Escherichia coli and Pseudomonas fluorescence) bacteria by agar diffusion assay. Gram-positive bacteria were more sensitive than were Gram-negative bacteria. The vegetative cells of B. cereus were the most sensitive among the pathogens tested with minimum inhibitory concentration (MIC) of 0.036 mg/μl of propolis on agar medium. Based on MIC, sensitivity of vegetative cells of B. cereus and its spores was tested in a nutrient broth with different concentrations of propolis at 37°C. In liquid broth, treatment with 1.8 mg/ml propolis showed bactericidal effect against B. cereus. B. cereus vegetative cells exposed to 7.2mg/ml of propolis lost their viability within 20 min. Against spores of B. cereus, propolis inhibited germination of spores up to 30 hours, compared to control at higher concentration than vegetative cells yet acted sporostatically. The bactericidal and sporostatic action of propolis were dependent on the concentration of propolis used and treatment time. Electron microscopic investigation of propolis-treated B. cereus revealed substantial structural damage at the cellular level and irreversible cell membrane rupture at a number of locations with the apparent leakage of intracellular contents. The antimicrobial effect of propolis in this study suggests potential use of propolis in foods.  相似文献
