Some metabolites present in the cell may affect its colloidal structure. If the cytoplasm is in the state of a gel, elastic
stresses may appear as a result of non-uniform concentration of some metabolites which affect mechanical properties of gels.
This paper investigates the question whether mechanical stresses, thus produced, can result in such a deformation of the cell
which may lead to a division. The result of this study indicates that this is not possible. Elastic stresses, produced by
non-uniformities of concentration of metabolites which affect the colloidal structures of the cytoplasm, may produce only

Subacute granulomatous thyroiditis (SAT), also known as de Quervain's thyroiditis or painful subacute thyroiditis, is the commonest thyroid condition responsible for neck tenderness. Other causes of pain in the thyroid gland should be taken into consideration during differential diagnosis, especially when a patient presents with misleading or equivocal signs and symptoms. We report the case of a 39 year-old woman diagnosed as having SAT whose clinical, biochemical and radiological presentation varied significantly from the common SAT manifestation. A tentative diagnosis of SAT was made based on the presented symptoms, ultrasonography and fine-needle biopsy results. However, biochemical analysis suggested neither inflammatory process nor the presence of thyrotoxicosis. Moreover, technetium scan of the thyroid revealed normal uptake of the isotope and there was neither clinical nor ultasonographic response for corticosteroids. The patient's symptoms, despite being prescribed typical treatment, gradually deteriorated and the pain became increasingly debilitating. Eventually, the patient underwent total thyroidectomy. As a result, she has become free of symptoms, but the macroscopic picture of thyroid gland, noted during the operation, gave a suspicion of neoplastic process. Nevertheless, histological study of flow samples confirmed the tentative diagnosis of de Quervain's thyroiditis, despite all previous findings that were not suggestive of it. This report confirms the likelihood that SAT can present atypically. Additionally, it indicates that surgical treatment may be considered in patients with severe, debilitating, persistent thyroid gland pain connected with SAT clinical course. 相似文献

Background
The recalcitrance of lignocellulosic cell wall biomass to deconstruction varies greatly in angiosperms, yet the source of this variation remains unclear. Here, in eight genotypes of short rotation coppice willow (Salix sp.) variability of the reaction wood (RW) response and the impact of this variation on cell wall recalcitrance to enzymatic saccharification was considered.Results
A pot trial was designed to test if the ‘RW response’ varies between willow genotypes and contributes to the differences observed in cell wall recalcitrance to enzymatic saccharification in field-grown trees. Biomass composition was measured via wet chemistry and used with glucose release yields from enzymatic saccharification to determine cell wall recalcitrance. The levels of glucose release found for pot-grown control trees showed no significant correlation with glucose release from mature field-grown trees. However, when a RW phenotype was induced in pot-grown trees, glucose release was strongly correlated with that for mature field-grown trees. Field studies revealed a 5-fold increase in glucose release from a genotype grown at a site exposed to high wind speeds (a potentially high RW inducing environment) when compared with the same genotype grown at a more sheltered site.Conclusions
Our findings provide evidence for a new concept concerning variation in the recalcitrance to enzymatic hydrolysis of the stem biomass of different, field-grown willow genotypes (and potentially other angiosperms). Specifically, that genotypic differences in the ability to produce a response to RW inducing conditions (a ‘RW response’) indicate that this RW response is a primary determinant of the variation observed in cell wall glucan accessibility. The identification of the importance of this RW response trait in willows, is likely to be valuable in selective breeding strategies in willow (and other angiosperm) biofuel crops and, with further work to dissect the nature of RW variation, could provide novel targets for genetic modification for improved biofuel feedstocks.5.

A solid phase combinatorial library was designed based on X-ray structures and in-silico models to explore an inducible S4+ pocket, which is formed by a simple side-chain rotation of Tyr95. This inducible S4+ pocket is unique to β-tryptase and does not exist for other trypsin-like serine proteases of interest. Therefore, inhibitors utilizing this pocket have inherent advantages for being selective against other proteases in the same family. A member of this library was found to be a potent and selective β-tryptase inhibitor with a suitable pharmacokinetic profile for further clinical evaluation. 相似文献

Leaves on transgenic tobacco plants expressing yeast-derived invertase in the apoplast develop clearly demarcated green and bleached sectors when they mature. The green areas contain low levels of soluble sugars and starch which are turned over on a daily basis, and have high rates of photosynthesis and low rates of respiration. The pale areas accumulate carbohydrate, photosynthesis is inhibited, and respiration increases. This provides a model system to investigate the sink regulation of photosynthetic metabolism by accumulating carbohydrate. The inhibition of photosynthesis is accompanied by a decrease of ribulose-1,5-bisphosphate and glycerate-3-phosphate, and an increase of triosephosphate and fructose-1,6-bisphosphate. The extracted activities of ribulose-1,5-bisphosphate carboxylase, fructose-1, 6-bisphosphatase and NADP-glyeraldehyde-3-phosphate dehydrogenase decreased. The activity of sucrose-phosphate synthase remained high or increased, an increased portion of the photosynthate was partitioned into soluble sugars rather than starch, and the pale areas showed few or no oscillations during transitions between darkness and saturating light in saturating CO2. The increased rate of respiration was accompanied by an increased level of hexose-phosphates, triose-phosphates and fructose-1,6-bisphosphate while glycerate-3-phosphate and phosphoenolpyruvate decreased and pyruvate increased. The activities of pyruvate kinase, phosphofructokinase and pyrophosphate: fructose-6-phosphate phosphotransferase increased two- to four-fold. We conclude that an increased level of carbohydrate leads to a decreased level of Calvin-cycle enzymes and, thence, to an inhibition of photosynthesis. It also leads to an increased level of glycolytic enzymes and, thence, to a stimulation of respiration. These changes of enzymes are more important in middle- or long-term adjustments to high carbohydrate levels in the leaf than fine regulation due to depletion of inorganic phosphate or high levels of phosphorylated metabolites.Abbreviations Fru 1,6bisP

This study tested several aspects of a model proposed by Williamson (1990, 1991) in which stresses in plant cell walls, detected by stress-receptive portions of inelastic cellulose microfibrils, orient microtubules via interactions with cell wall-linked transmembrane proteins. Young expanding cells of pea root tips have highly ordered transverse arrays of microtubules oriented perpendicular to the direction of cell expansion. The recovery of these ordered MT arrays after depolymerisation with oryzalin was assessed. It was shown that treating roots with disruptors of microfibril synthesis (2,6-dichlorobenzonitrile and calcofluor white) or the disruption of Arg-Gly-Asp (RGD)-mediated wall-membrane links did not affect the orientation of recovering microtubule arrays. Furthermore, cell wall stresses themselves appeared unnecessary for regeneration of transverse arrays. The relevance of these findings to Williamson's hypothesis is discussed. 相似文献

Roads and highways contribute enormously to habitat fragmentation, because they can inhibit or even block animal movement
across them, which may result in the ultimate division of the populations adjacent to the roads into smaller isolated subpopulations.
The isolation reduces gene flow and increases risk of extinction due to a decrease in the genetic diversity of the isolated
population. The aim of the present study is to determine whether highways can cause genetic subdivision of the bank vole Myodes glareolus (Schreber, 1780) and yellow-necked mouse Apodemus flavicollis (Melchior, 1834). The study was carried out at three sites in the Highway D1 (Prague-Brno) in the Czech Republic, where a
previous study demonstrated a barrier effect of the highway avoiding the interchange of individuals of both species. The genetic
structure was determined from the analysis of six DNA microsatellites loci in M. glareolus and five in A. flavicollis. We found only weak genetic differences between populations living at opposite sides of the highway in either of the species
and a low degree of subdivision, but significant positive correlation between genetic and geographical distance, which suggests

Observatories are designed to collect data for a range of uses. The Australian Acoustic Observatory (A2O) was established to collect environmental sound, including audible species calls, from 344 recorders at 86 sites around Australia. We examine the potential of the A2O to monitor near threatened, threatened, endangered and critically endangered species, based on their vocal behaviour, geographic distributions in relation to the sites of the A2O and on some knowledge of habitat use. Using IUCN and EPBC lists of threatened and endangered species, we extracted species that vocalized in the audible range, and using conservative estimates of their geographic ranges, determined whether there was a possibility of hearing them at these sites. We found that it may be possible to detect up to 171 threatened species at sites established for the A2O, and that individual sites have the potential to detect up to 40 threatened species. All 86 sites occurred in locations where threatened species could possibly be detected, and the list of detectable species included birds, amphibians, and mammals. We have incidentally detected one mammal and four bird species in the data during other work. Threatening processes to which potentially detectable species were exposed included all but two IUCN threat categories. We concluded that with applications of technology to search the audio data from the A2O, it could serve as an important tool for monitoring threatened species. 相似文献

Apis mellifera foragers perform waggle dances to communicate the presence of highly desirable nectar sources to their forager-mates. Each waggle dance consists of several waggle-runs (straight movements of the dancer closely aligned on the comb surface) that carry spatial information that the dance followers can use to locate the food source being advertised. To address how this complex motor display responds to unpredictable fluctuations in its main triggering stimulus, i.e., sucrose stimulation, we analyzed the effects of an increase in reward on the direction of consecutive waggle-runs as well as other components of the waggle dance. Results show that a sudden increase in reward may increase the directional scatter among consecutive waggle-runs, especially those performed at the beginning of the dance. However, a simultaneous and rapid increase in the duration of the signal—together with a more regular alignment of the later waggle-runs within the signal— seems to compensate the initial increase in directional scatter so that the transfer of directional information remains effective. These results point out that the regulation of dance maneuvers depends on the dancers motivation to forage. 相似文献

The authors review the anticipated benefits from tourism for protected areas in Madagascar, the mechanisms that have been adopted, and how these are working at national and local levels. The contribution of tourism to the maintenance of Protected Areas in Madagascar is assessed. The anticipated benefits of tourism envisaged by the 1991 National Environmental Action Plan are considered to be over-optimistic. The growth rate in tourist arrivals is found to be smaller than previously anticipated as a result of infrastructural constraints at national and local scales. It is suggested that revenues from tourism are inadequate in meeting park management costs although the local transfer of entrance fee revenues to development projects is found to be more beneficial. It is also argued that the benefits of revenue sharing can be effective only for a limited number of protected areas until unfrastructure is improved and protected areas become more accessible to tourists. 相似文献

γ-Glutamyltransferase activity was detected in the plasma membrane of the highly differentiated hepatoma cell line Fao, (0.93 mU/mg cell protein). Dexamethasone (1 μM) provoked a 2–3-fold increase in the activity of the enzyme in the presence of fetal calf serum. Maximal induction occurred 48–72 h after addition of the glucocorticoid to the cell culture medium. The hormonal specificity was demonstrated by the relative potencies of several glucocorticoids and sex steroids: hydrocortisone and corticosterone increased γ-glutamyltransferase activity while tetrahydrocorticosterone and all sex steroids tested were ineffective. The effect of dexamethasone on γ-glutamyltransferase activity was specific since the activities of several other plasma membrane enzymes were not modified. The mechanism of the dexamethasone-induced increase in γ-glutamyltransferase activity was neither by modification of the affinity of the enzyme for its substrates nor by alteration of the subcellular distribution of the enzyme. This increase was prevented by cycloheximide and actinomycin D. The data presented are consistent with a specific glucocorticoid receptor-mediated induction of γ-glutamyltransferase activity in Fao cells. The kinetic parameters of the induction process by glucocorticoids are very similar to those found in adult rat liver. These results suggest that the Fao cell line is a very convenient system for the study of the molecular mechanisms of glucocorticoid effects on differentiated cells. 相似文献

In two experiments, the effect has been investigated of a mildand a more prolonged drought on the spatial distribution ofgrowth, epidermal cell lengths and cell wall peroxidase activitiesin the leaf elongation zone of the grass species Lolium temulentumL. In both experiments drought reduced the size of the elongationzone and local rates of elongation within it. Abrupt increasesin cell wall-associated peroxidase activity occurred at or closeto the position where elongation ceased in the leaf elongationzones of well-watered and mildly drought-stressed plants. Moreprolonged drought caused a 200300% increase in the cellwallassociated peroxidase activity in the elongation zone only.The significant increase in the elongation zone cell wall peroxidaseactivity and its spatial variation provides evidence of a potentiallycausal role for cell wall-associated peroxidase in restrictingcell expansion during drought.
