Specimens of moth flies (Diptera: Psychodidae: Psychodinae) were sampled in southwest Anatolia and the European part of Turkey between 2003 and 2009. Samples contained nine species new to science: Ulomyia asiaminorica sp. n., Pneumia pellucida sp. n., Berdeniella turcica sp. n., B. aktashi sp. n., Clytocerus thracicus sp. n., Jungiella afyonica sp. n., J. geniculatoides sp. n., J. barlasi sp. n., and Telmatoscopus remotus sp.n. The new species are described and illustrated. Distributional data for a total of 45 species are provided. 63 species of Psychodinae are now known to occur in Turkey.

In a psychodid, Tinearia alternata, the initial differentiation of the polytrophic ovary occurs during the early larval stages. Early in development, each ovary anlage is a solid organ subdivided into three distinct zones: the cortex houses germ cells and somatic interstitial tissue, while two other somatic regions will give rise to the oviduct calyx and anterior part of the lateral oviduct. Germ cell cluster formation precedes the development of ovarioles. Each ovariole houses only one functional egg chamber. All ovarioles within paired ovaries are developmentally synchronized. In the larval ovaries, the newly formed egg chambers and then the ovarioles are intermingeled with and surrounded by the somatic interstitial tissue of the ovary cortex. The interstitial cells give rise to all the somatic elements of the ovarioles. In the pupal ovaries, the remaining interstitial tissue degenerates; thus, the ovarioles protrude into the body cavity. The ovaries in psychodids develop relatively large and swollen oviduct calyxes that are equivalent to receptaculum seminis (spermatheca). The morphological differentiation of germ cells within the egg chambers starts during late larval/early pupal stages. Nurse cell nuclei contain prominent nucleoli and polytene chromosomes. Oocyte growth results from accumulation of yolk and then, in the final stages of oogenesis, from an inflow of cytoplasm from the nurse cells. J. Classification of Psychodinae has been a hotly contested topic in the taxonomic literature, with multiple mutually incompatible classifications proposed. Three main points of contention can be identified: (i) the validity of Maruinini Enderlein as a tribe‐level taxon and whether it forms a monophyletic group; (ii) the placement of subtribe Trichopsychodina Ježek – specifically whether it belongs with Psychoda Latreille in Psychodini or with Paramormia Enderlein and Telmatoscopus Eaton in Paramormiini Enderlein or Mormiini Enderlein; and (iii) whether Mormia Enderlein is more closely related to Brunettia Annandale or Paramormia Enderlein and Telmatoscopus Eaton. In the present paper, these questions are investigated using a molecular phylogeny of sequences compiled from all previous molecular phylogenies relevant to Psychodinae, as well as some hitherto unpublished sequences. The resulting matrix comprised 5406 base pairs from six markers for 32 taxa, and when analysed using Bayesian inference it yielded a well‐resolved tree which was unambiguous for all previously contentious points: Maruinini sensu lato (including Setomimini) was resolved as a valid taxon, Trichopsychodina is rendered paraphyletic by Psychodini, and Mormia is not closely related to Brunettia , but instead closer to Telmatoscopus and Paramormia . A revised tribal classification of Psychodinae is proposed, recognizing the tribes Psychodini, Brunettiini, Maruinini and Pericomaini as well as 17 unplaced genera.

Polytrophic ovaries of the nematocerous dipteran, Tinearia alternata Say consists of several developmentally synchronized ovarioles each housing only one functional egg chamber with 15 nurse cells and an oocyte. At the early stages of previtellogenesis the nurse cells become polyploid and synthetically active. Their nuclei contain polytene chromosomes and prominent nucleoli. With the advance of previtellogenic growth the nurse cell cytoplasm is loaded with the growing number of ribosomes and contain perinuclear nuage material, mitochondria, electron dense bodies and aggregations of endoplasmic reticulum. All these organelles are transported into the oocyte thanks to the massive and rapid flow of the nurse cell cytoplasmic contents. Nurse cell-oocyte transport is mediated by actin cytoskeleton. Prior to the rapid cytoplasm transfer, F-actin network is associated with the nurse cell membranes while tiny bundles of microfilaments form actin baskets connected with ring canals. Nurse cells in Tinearia lack an extensive scaffold of radially oriented, F-actin bundles (cables) that would tether their nuclei in place, thus preventing ring canals from plugging. The way the nuclei are anchored to their central positions within the cells remains unclear. Towards the final stages of oogenesis nurse cells are almost devoid of cytoplasm and degenerate. Although their nuclei undergo dramatic morphological transformations, typical hallmarks of apoptotic pathway could not be clearly observed. Rapid ooplasmic streaming does not occur.

Phlebotominae sand fly specimens were prepared for histological and physiological studies. Different fixatives were tested on sectioned and whole bodied adult females in order to obtain good fixation and provide satisfactory penetration of the embedding media. All fixed specimens were infiltrated (up to seven days under 5 C) and embedded in hydroxyethyl metacrylate. Two-three m sections were stained, mounted in Canada balsam and observed by light microscopy. Best results were achieved when whole bodied insects were double fixed in Bouin's and Carnoy's fluids (4 h/2 h) and stained in Hematoxilin/Eosin or fixed in calcium formaldehyde and stained in mercury bromophenol blue.

A study on the phlebotomine sand fly fauna in Belo Horizonte city, state of Minas Gerais, Brazil, was carried out. From April 2001 to March 2003, monthly systematic collections were performed in three houses from each of the nine regions of the city, using CDC light traps for four consecutive days. The traps were set into the houses and in peridomestic areas totaling 54 traps. A number of 3871 sand fly specimens of the genera Lutzomyia and Brumptomyia were collected. Sixty eight percent of the specimens were L. longipalpis and 16% L. whitmani, insect vectors of visceral and American cutaneous leishmaniasis, respectively. Environmental factors such as temperature, humidity, and frequency of precipitation suggest that the number of insects increases after rainy periods. During the same period mentioned above, seasonal captures were carried out in parks and green areas of Belo Horizonte, using Shannon trap. A total of 579 phlebotomine sand flies were collected from which 398 (68.7%) were females with the predominance of L. whitmani and L. monticola. Those specimens were used for natural infection examination, by polymerase chain reaction. No Leishmania DNA was present in any of the specimens tested.

Phlebotomus papatasi is a proven vector of Leishmania major which is one of the causative agents of cutaneous leishmaniasis in the Old World. Although it has a wide geographical range, its population structure is not yet well understood. In an effort to better understand the population dynamics of this vector, we developed a panel of di‐ and trinucleotide microsatellite markers, using a magnetic bead hybridization enrichment protocol. These microsatellite loci showed three to seven alleles with an expected heterozygosity range between 0.702 and 0.876. The level of polymorphisms found in this study suggests that these microsatellite loci can be used for population analysis of P. papatasi.

When females of the screw-worm fly, Chrysomya bezziana Villeneuve, were given a choice of solutions of protein, sucrose and water, their protein intake was substantial only on days 2, 3 and 4 after emergence, and during the 3 days following the first oviposition. Protein consumption by males remained consistently low for 10 days following emergence.Protein ingestion by laboratory-bred autogenous and anautogenous females, autogenous wild females of C. bezziana, and females of the anautogenous blowly species, Chrysomya megacephala (F), was compared. During the first ovarian cycle (days 1–6), protein solution accounted for 9.5% and 9.4% of the total liquid intake of laboratory-bred and wild anautogenous females of C. bezziana. Protein ingestion by C. megacephala females during the first ovarian cycle represented 23% of the total liquids. During the second ovarian cycle in C. bezziana, protein ingestion represented 21% of total liquids. The presence of dry sucrose or sucrose solution of high concentration depressed protein intake during the first ovarian cycle in C. bezziana. Most feeding by females occurred during the photophase with early morning and late evening peaks in carbohydrate ingestion and a late afternoon peak in protein ingestion. The ecological implications of the results are discussed.

The phlebotomine sand fly Lutzomyia serrana (Damasceno & Arouck) was mass-reared tinder conditions of varying densities in an effort to improve colony production efficiency. To do this, the experimental carrying capacity of a standard rearing chamber was determined, i.e., the optimum population size in relation to density (individuals per unit of space). Rearing chambers of 100 cm3 were populated with 1-50 L. serrana engorged females and an equal number of males. Laboratory conditions were maintained at 23-26 degrees C and 85-95% RH. The following parameters were recorded for each experimental chamber (three replicates): (1) female mortality without oviposition, (2) number of eggs oviposited and (3) number of adults emerging from the egg cohort. Female mortality began to increase substantially in the 26-female chamber, from 5.7% to 15% and finally reaching 60.2% in the 46-50 female chambers. In the chambers containing 1-20 females, egg number and realized adult progeny increased linearly to reach an asymptote. In the 20-50 female chambers, the number of eggs ranged from 420 to 699, and adult production from 306 to 432. The optimum carrying capacity for the 100-cm3 chambers was 22 +/- 2 females. Beyond this number, auto-regulation was initiated, i.e., female mortality without oviposition increased as the number of females per chamber increased. Total number of eggs and adult production was similar in all chambers containing 20-50 females. In conclusion, for optimizing production of mass reared sand flies, determination of the carrying capacity is essential to optimize use of insectary resources, to avoid loss of valuable potentially ovipositing females, and to increase overall production efficiency.

A new species of parthenogenetic, autogenic and apparently extremely endemic phlebotomine is described from a sandstone cave located in primary terra firme forest to the North of the city of Manaus. Specimens were collected in the aphotic zone of the Refúgio do Maruaga cave by light trap and reared from bat guano. The adult morphology suggests a closer relationship to some Old World Phlebotominae than to species of Lutzomyia Fran?a encountered in the surrounding rainforest, but it shares characteristics with the recently proposed Neotropical genera Edentomyia Galati, Deanemyia Galati and Oligodontomyia Galati.

We measured reproductive and population parameters of adult sand flies, Phlebotomus papatasi (Scopoli, 1786) (Diptera: Psychodidae), in environmental chambers maintained at temperatures of 15, 18, 20, 25, 28, and 32 degrees C. Based on cohorts of adults at each temperature regime, horizontal life tables were constructed using established laboratory colonies initiated from specimens collected in Sanliurfa Province, southeastern Anatolia, Turkey. The fecundity and longevity of the insects were both highly variable, depending on the temperature. At 15 degrees C, all of the cohort females died before laying eggs, so the construction of a life table for this temperature regime was not possible. Within a range of 18 to 32 degrees C, the longevity of adult P. papatasi increased as the temperature decreased; at 15 degrees C, the mean survival times of females and males were 19.04 +/- 6.94 days (9-35) and 17.84 +/- 7.11 days (9-33), respectively. While the highest number of eggs was found in the cohort at 28 degrees C (44.08 +/- 7.79), this was only 3.60 +/- 1.55 in the cohort at 32 degrees C and 2.8 +/- 0.9 in the cohort at 18 degrees C. This result showed that extreme temperatures negatively affect the fecundity of this species. The cohort reared at 28 degrees C exhibited the highest intrinsic rates of population increase (r(m)) for P. papatasi. The r(m) ranged from 0.098 at 28 degrees C to 0.007 at 18 degrees C. The cohort placed at 28 degrees C was found to be significantly different (P < 0.01) from the other cohorts producing the fewest progeny in terms of net reproductive rate, R(0), (15.87). The values for mean generation time (T) were estimated to vary from 36 days to 271 days depending on temperature. Principal Component Analysis (PCA) confirmed results from the previous studies that the cohort at 28 degrees C orientated and clustered as a distinct group along the first two PCs.

Espíndola, A., Buerki, S., Jacquier, A., Je?ek, J. & Alvarez, N. (2012). Phylogenetic relationships in the subfamily Psychodinae (Diptera, Psychodidae). —Zoologica Scripta, 41, 489–498. Thanks to recent advances in molecular systematics, our knowledge of phylogenetic relationships within the order Diptera has dramatically improved. However, relationships at lower taxonomic levels remain poorly investigated in several neglected groups, such as the highly diversified moth‐fly subfamily Psychodinae (Lower Diptera), which occurs in numerous terrestrial ecosystems. In this study, we aimed to understand the phylogenetic relationships among 52 Palearctic taxa from all currently known Palearctic tribes and subtribes of this subfamily, based on mitochondrial DNA. Our results demonstrate that in light of the classical systematics of Psychodinae, none of the tribes sensu Je?ek or sensu Vaillant is monophyletic, whereas at least five of the 12 sampled genera were not monophyletic. The results presented in this study provide a valuable backbone for future work aiming at identifying morphological synapomorphies to propose a new tribal classification.

Tympanate hearing has evolved in at least 6 different orders of insects, but had not been reported until recently in the Diptera. This study presents a newly discovered tympanal hearing organ, in the parasitoid tachinid fly, Ormia ochracea. The hearing organ is described in terms of external and internal morphology, cellular organization of the sensory organ and preliminary neuroanatomy of the primary auditory afferents. The ear is located on the frontal face of the prothorax, directly behind the head capsule. Conspicuously visible are a pair of thin cuticular membranes specialized for audition, the prosternal tympanal membranes. Directly attached to these membranes, within the enlarged prosternal chamber, are a pair of auditory sensory organs, the bulbae acusticae. These sensory organs are unique among all auditory organs known so far because both are contained within an unpartitioned acoustic chamber. The prosternal chamber is connected to the outside by a pair of tracheae. The cellular anatomy of the fly's scolopophorous organ was investigated by light and electron microscopy. The bulba acustica is a typical chordotonal organ and it contains approximately 70 receptor cells. It is similar to other insect sensory organs associated with tympanal ears. The similarity of the cellular organization and tympanal morphology of the ormiine ear to the ears of other tympanate insects suggests that there are potent constraints in the design features of tympanal hearing organs, which must function to detect high frequency auditory signals over long distances. Each sensory organ is innervated by a branch of the frontal nerve of the fused thoracic ganglia. The primary auditory afferents project to each of the pro-, meso-, and metathoracic neuropils. The fly's hearing organ is sexually dimorphic, whereby the tympanal membranes are larger in females and the spiracles larger in males. The dimorphism presumably reflects differences in the acoustic behavior in the two sexes.

Visceral leishmaniasis is a zoonosis whose primary vector in Brazil is the sandfly Lutzomyia longipalpis Lutz & Neiva. Presently, efforts to control the vector have not been effective in reducing the prevalence of disease. A possible alternative to current strategies is the biological control of the vector using entomopathogenic fungi. This study evaluates the effects of the fungus, Beauveria bassiana (Bals.) Vuilleman, in different developmental stages of L. longipalpis. Five concentrations of the fungus were utilized ranging from 104 to 108 conidia/ml, with appropriate controls. The unhatched eggs, larvae and dead adults exposed to B. bassiana were sown to reisolate the fungus. The fungus was subsequently identified by polymerase chain reaction (PCR) and DNA sequencing. Exposure to B. bassiana reduced the number of eggs that hatched by 59% (P < 0.01). The longevity of infected adults was 5 days, significantly lower than that of the negative control which was 7 days (P < 0.001). The longevity of the adult sandfly exposed to the positive chemical (pyrethroid, cypermetherin) control was less than 1 day. The effects of fungal infection on the hatching of eggs laid by infected females were also significant and dose-dependent (P < 0.05). With respect to fungal post-infection growth parameters, only germination and sporulation were significantly higher than the fungi before infection (P < 0.001). The identity of the reisolated fungus was confirmed by automated DNA sequencing post-passage in all insect stages. These data show that B. bassiana has good pathogenic potential, primarily on L. longipalpis larvae and adults. Consequently, the use of this fungus in sandfly control programs has potential in reducing the use of chemical insecticides, resulting in benefits to humans and the environment.

Field tests on attraction of Caribbean fruit flies to 15×20 cm colored sticky traps releaved a clear preference for orange, followed by yellow and yellow-green. Lower capture rates were obtained for dark green, blue, red, black, and white. Reflectance spectra of painted surfaces were determined and it was noted that fly capture rates were directly related to the proportion of light reflected in the 580–590 nm region. Attraction appeared to depend primarily upon the hue rather than the intensity of emission. Responses to orange were considered to indicate fruit-seeking rather than foliage-seeking behavior because a disproportionate number of females was captured (3:1 : ratio), because foliage normally reflects maximally in the yellow-green rather than the orange region, and because a variety of host fruit reflects strongly in the orange region of the spectrum.
