The availability of a complete genome database for the cyanobacterium Synechocystissp. PCC6803 (glucose-tolerant strain) has raised expectations that this organism would become a reference strain for work aimed at understanding the CO2-concentrating mechanism (CCM) in cyanobacteria. However, the amount of physiological data available has been relatively limited. In this report we provide data on the relative contributions of net HCO3 uptake and CO2 uptake under steady state photosynthetic conditions. Cells were compared after growth at high CO2 (2% v/v in air) or limiting CO2 conditions (20 ppm CO2). Synechocystishas a very high dependence on net HCO3 uptake at low to medium concentrations of inorganic carbon (Ci). At high Ci concentrations net CO2 uptake became more important but did not contribute more than 40% to the rate of photosynthetic O2 evolution. The data also confirm that high Ci cells of Synechocystissp. PCC6803 possess a strong capacity for net HCO3 uptake under steady state photosynthetic conditions. Time course experiments show that induction of maximal Ci uptake capacity on a shift from high CO2 to low CO2 conditions was near completion by four hours. By contrast, relaxation of the induced state on return of cells to high CO2, takes in excess of 230 h. Experiments were conducted to determine if Synechocystissp. PCC6803 is able to exhibit a `fast induction' response under severe Ci limitation and whether glucose was capable of causing a rapid inactivation in Ci uptake capacity. Unicellular algae grow well under limiting CO2 conditions, aided by a carbon concentrating mechanism (CCM). In C. reinhardtii, this mechanism is inducible and is present only in cells grown under low CO2 conditions. We constructed a cDNA library from cells adapting to low CO2, and screened the library for cDNAs specific to low CO2-adapting cells. Six classes of low CO2-inducible clones were identified. One class of clone, reported here, represents a novel gene associated with adaptation of cells to air. A second class of clones corresponds to the air-inducible periplasmic carbonic anhydrase I (CAH1). These clones represent genes that respond to the level of CO2 in the environment.  相似文献

Reports in the 1970s from several laboratories revealed that the affinity of photosynthetic machinery for dissolved inorganic carbon (DIC) was greatly increased when unicellular green microalgae were transferred from high to low-CO2 conditions. This increase was due to the induction of carbonic anhydrase (CA) and the active transport of CO2 and/or HCO3 which increased the internal DIC concentration. The feature is referred to as the `CO2-concentrating mechanism (CCM)'. It was revealed that CA facilitates the supply of DIC from outside to inside the algal cells. It was also found that the active species of DIC absorbed by the algal cells and chloroplasts were CO2 and/or HCO3 , depending on the species. In the 1990s, gene technology started to throw light on the molecular aspects of CCM and identified the genes involved. The identification of the active HCO3 transporter, of the molecules functioning for the energization of cyanobacteria and of CAs with different cellular localizations in eukaryotes are examples of such successes. The first X-ray structural analysis of CA in a photosynthetic organism was carried out with a red alga. The results showed that the red alga possessed a homodimeric β-type of CA composed of two internally repeating structures. An increase in the CO2 concentration to several percent results in the loss of CCM and any further increase is often disadvantageous to cellular growth. It has recently been found that some microalgae and cyanobacteria can grow rapidly even under CO2 concentrations higher than 40%. Studies on the mechanism underlying the resistance to extremely high CO2 concentrations have indicated that only algae that can adopt the state transition in favor of PS I could adapt to and survive under such conditions. It was concluded that extra ATP produced by enhanced PS I cyclic electron flow is used as an energy source of H+-transport in extremely high-CO2 conditions. This same state transition has also been observed when high-CO2 cells were transferred to low CO2 conditions, indicating that ATP produced by cyclic electron transfer was necessary to accumulate DIC in low-CO2 conditions. The capability to reassimilate CO2 originating from intracellular decarboxylating processes connected with the photorespiratory glycolate pathway and-or decarboxylation of C4 acids during C4 photosynthesis has been investigated with four species of the genus Flaveria (Asteraceae). The C3-C4 intermediate species F. pubescens and F. anomala reassimilated CO2 much more efficiently than the C3 species F. cronquistii and, with respect to this feature, behaved similarly to the C4 species F. trinervia. Therefore, under atmospheric conditions the intermediate species photorespired with rates only between 10–20% of that measured with F. cronquistii. At low oxygen concentrations (1,5%) the reassimilation potential of F. anomala approached that of F. trinervia and was distinct from that found with F. pubescens. The data are discussed with respect to a possible sequence of events during evolution of C4 photosynthesis. The increasing CO2 concentration in Earths atmosphere is expected to cause a greater decline in the nutritional quality of C3 than C4 plants. As a compensatory response, herbivorous insects may increase their feeding disproportionately on C3 plants. These hypotheses were tested by growing the grasses Lolium multiflorum C3) and Bouteloua curtipendula C4) at ambient (370 ppm) and elevated (740 ppm) CO2 levels in open top chambers in the field, and comparing the growth and digestive efficiencies of the generalist grasshopper Melanoplus sanguinipes on each of the four plant × CO2 treatment combinations. As expected, the nutritional quality of the C3 grass declined to a greater extent than did that of the C4 grass at elevated CO2; protein levels declined in the C3 grass, while levels of carbohydrates (sugar, fructan and starch) increased. However, M. sanguinipes did not significantly increase its consumption rate to compensate for the lower nutritional quality of the C3 grass grown under elevated CO2. Instead, these grasshoppers appear to use post-ingestive mechanisms to maintain their growth rates on the C3 grass under elevated CO2. Consumption rates of the C3 and C4 grasses were also similar, demonstrating a lack of compensatory feeding on the C4 grass. We also examined the relative efficiencies of nutrient utilization from a C3 and C4 grass by M. sanguinipes to test the basis for the C4 plant avoidance hypothesis. Contrary to this hypothesis, neither protein nor sugar was digested with a lower efficiency from the C4 grass than from the C3 grass. A novel finding of this study is that fructan, a potentially large carbohydrate source in C3 grasses, is utilized by grasshoppers. Based on the higher nutrient levels in the C3 grass and the better growth performance of M. sanguinipes on this grass at both CO2 levels, we conclude that C3 grasses are likely to remain better host plants than C4 grasses in future CO2 conditions.  相似文献

Aphanocapsa 6308 metabolizes both NaHCO3 and Na2CO3. The short term incorporation (5-s) metabolic pattern and the patterns of incorporation of bicarbonate for exponential versus stationary phase cultures differ, however. Cells were equilibrated for 10 min in air and distilled water prior to injection of either NaH14CO3 at pH 8.0, or Na2 14CO3 at pH 11.0. Hot ethanol extracts were analyzed via paper chromatography and autoradiography for products of CO2 fixation. At 5 s, malate (51.5%) predominates slightly as a primary bicarbonate fixation product over 3-phosphoglycerate (40.3%); 3-phosphoglycerate is the primary product of carbonate fixation. At 60 s, the carbonate and bicarbonate labelling patterns are similar. Cells in stationary phase fix in 5 s a greater proportion of bicarbonate into malate (36% vs. 14% for 3-phosphoglycerate) than do cells in exponential growth. Likewise, 60 s incorporations show a large amount of bicarbonate fixed into aspartate (30.9%) in stationary phase cells over that of exponential phase (11.6%). These data suggest an operative C4 pathway for purposes not related to carbohydrate synthesis but rather as compensation for the incomplete tricarboxylic acid cycle in cyanobacteria. The role of carbon dioxide in glucose metabolism of Bacteroides fragilis   总被引:2,自引:0,他引:2  
The effect of CO2 concentration on growth and glucose fermentation of Bacteroides fragilis was studied in a defined mineral medium. Batch culture experiments were done in closed tubes containing CO2 concentrations ranging from 10% to 100% (with appropriate amounts of bicarbonate added to maintain the pH at 6.7). These experiments revealed that CO2 had no influence on growth rate or cell yield when the CO2 concentration was above 30% CO2 (minimum available CO2–HCO 3 - , 25.5 mM), whereas a slight decrease in these parameters was observed at 20% and 10% CO2 (available CO2–HCO 3 - , 17 and 8.5 mM, respectively). If CO2–HCO 3 - concentrations were below 10 mM, the lag phase lengthened and a decrease in maximal growth rate and cell yield were observed. The amount of acetate made decreased, while d-lactate concentration increased. A net production of CO2 allowed growth under conditions of extremely low concentrations of added CO2.When B. fragilis was grown in continuous culture with 100% CO2 or 100% N2, the dilution rate influenced the concentrations of acetate, succinate, propionate, d-lactate, l-malate and formate formed. Decreasing the dilution rate favored propionate and acetate production under both conditions. When the organism was grown with 100% N2, the amount of propionate formed was greater than the amount of succinate formed at all dilution rates. Except at slow dilution rates the reverse was true when 100% CO2 was used. B. fragilis was unable to grow at dilution rates faster than 0.154 h-1 when grown with 100% N2; the Y glc max was 67.9 g DW cells/mol glucose and m s was 0.064 mmol glucose/g DW·h. If the gas atmosphere was 100% CO2 the organism was washed out of the culture when the dilution rate exceeded 0.38 h-1; the Y glc max was 59.4 g DW cells/mol glucose and m s was 0.094 mmol glucose/g DW·h.Measurement of the phosphoenolpyruvate (PEP) carboxykinase (E.C. 4.1.1.49) with whole, permeabilized cells of B. fragilis showed an increase of specific enzyme activity with decreasing CO2 concentrations. The growth response of Dutch salt marsh species (C3 and C4) to atmospheric CO2 enrichment was investigated. Tillers of the C3 speciesElymus athericus were grown in combinations of 380 and 720 11-1 CO2 and low (O) and high (300 mM NaCl) soil salinity. CO2 enrichment increased dry matter production and leaf area development while both parameters were reduced at high salinity. The relative growth response to CO2 enrichment was higher under saline conditions. Growth increase at elevated CO2 was higher after 34 than 71 days. A lower response to CO2 enrichment after 71 days was associated with a decreased specific leaf area (SLA). In two other experiments the effect of CO2 (380 and 720 11-1) on growth of the C4 speciesSpartina anglica was studied. In the first experiment total plant dry weight was reduced by 20% at elevated CO2. SLA also decreased at high CO2. The effect of elevated CO2 was also studied in combination with soil salinity (50 and 400 mM NaCl) and flooding. Again plant weight was reduced (10%) at elevated CO2, except under the combined treatment high salinity/non-flooded. But these effects were not significant. High salinity reduced total plant weight while flooding had no effect. Causes of the salinity-dependent effect of CO2 enrichment on growth and consequences of elevated CO2 for competition between C3 and C4 species are discussed.  相似文献

The long-term effects of exogenous sucrose (3 percnt;) on growth, photosynthesis and carbon metabolism ofin vitro tomato plantlets were investigated under two sets of growth conditions that respectively favor source- or sink-limitations of photosynthesis: 1) low photosynthetic photon flux (PPF) (50 μmol m−2 · s−1) and low CO2 concentration (400 μmol mol−1) and 2) high PPF (500 μmol m−2 · s−1 and high CO2 concentration (4000 μmol mol−1). The supply of sucrose under source-limitation conditions increased the growth, the maximal photosynthetic rate, the chl content, the maximal quantum yield of Photosystem II estimated by the Fv/Fm chl fluorescence ratio as well as the soluble sugars (hexoses, sucrose) and starch contents in roots, young and mature leaves when compared to those of photo-autotrophic plantlets. Also, sucrose feeding under these conditions strongly increased the activity of sucrose synthase (SS) (EC 2.4.1.13) in roots and young leaves whereas the activities of sucrose phosphate synthase (SPS) (EC 2.4.1.14), acid invertase (INV) (EC 3.2.1.26) and ADP-glucose pyrophosphorylase (ADPGppase) (EC 2.7.7.27) were highly stimulated in roots and mature leaves. Contrary to these observations, the supply of sucrose to plantlets developed under high PPF and CO2 concentration decreased growth and led to a somewhat lower maximal photosynthetic rate relative to photo-autotrophic plantlets. These negative responses to exogenous sucrose were accompanied by stronger accumulations of hexose and starch, larger stimulation of INV in mature leaves developed under conditions of sink limitation than those from source limitation conditions. Moreover, under high PPF and high CO2 concentration, exogenous sucrose led to a marked repression of the SPS activity and caused much lower stimulations of ADPGppase in mature leaves than those observed at low PPF and low CO2 concentration. We therefore conclude that under our experimental conditions, the interactive effects of exogenous sucrose and environmental conditions on growth and photosynthesis could be rationalized by the source-sink equilibrium of thein vitro tomato plantlets.  相似文献

The biomass yield of freshwater filamentous sulfur bacteria of the genus Beggiatoa, when grown lithoheterotrophically or mixotrophically, has been shown to increase 2 to 2.5 times under microaerobic conditions (0.12 mg/l oxygen) as compared to aerobic conditions (9 mg/l oxygen). The activity of the glyoxylate cycle key enzymes have been found to increase two to three times under microaerobic conditions (at an O2 concentration of 2 mg/l), and the activities of the sulfur metabolism enzymes increased three to five times (at an O2 concentration of 0.1–0.5 mg/l). It has also been found that, under microaerobic conditions, thiosulfate was almost completely oxidized to sulfate by the bacteria, without accumulation of intermediate metabolites. At the same time, a 2- to 15-fold decrease in the activities of the tricarboxylic acid cycle enzymes involved in the reduction of NAD and FAD was observed. Reorganization of the respiratory chain after changes in aeration and type of nutrition was also observed. It has been found that, in cells grown heterotrophically, the terminal part of the respiratory chain contained an aa 3-type oxidase, whereas, during mixotrophic, lithoheterotrophic, and autotrophic growth, aa 3-type oxidase synthesis was inhibited, and the synthesis of a cbb 3-type oxidase, which is induced under microaerobic conditions, was activated. Abutilon theophrasti (C3) and Amaranthus retroflexus (C4), were grown from seed at four partial pressures of CO2: 15 Pa (below Pleistocene minimum), 27 Pa (pre-industrial), 35 Pa (current), and 70 Pa (future) in the Duke Phytotron under high light, high nutrient, and wellwatered conditions to evaluate their photosynthetic response to historic and future levels of CO2. Net photosynthesis at growth CO2 partial pressures increased with increasing CO2 for C3 plants, but not C4 plants. Net photosynthesis of Abutilon at 15 Pa CO2 was 70% less than that of plants grown at 35 Pa CO2, due to greater stomatal and biochemical limitations at 15 Pa CO2. Relative stomatal limitation (RSL) of Abutilon at 15 Pa CO2 was nearly 3 times greater than at 35 Pa CO2. A photosynthesis model was used to estimate ribulose-1,5-bisphosphate carboxylase (rubisco) activity (Vcmax), electron transport mediated RuBP regeneration capacity (J max), and phosphate regeneration capacity (PiRC) in Abutilon from net photosynthesis versus intercellular CO2 (AC i) curves. All three component processes decreased by approximately 25% in Abutilon grown at 15 Pa compared with 35 Pa CO2. Abutilon grown at 15 Pa CO2 had significant reductions in total rubisco activity (25%), rubisco content (30%), activation state (29%), chlorophyll content (39%), N content (32%), and starch content (68%) compared with plants grown at 35 Pa CO2. Greater allocation to rubisco relative to light reaction components and concomitant decreases in J max and PiRC suggest co-regulation of biochemical processes occurred in Abutilon grown at 15 Pa CO2. There were no significant differences in photosynthesis or leaf properties in Abutilon grown at 27 Pa CO2 compared with 35 Pa CO2, suggesting that the rise in CO2 since the beginning of the industrial age has had little effect on the photosynthetic performance of Abutilon. For Amaranthus, limitations of photosynthesis were balanced between stomatal and biochemical factors such that net photosynthesis was similar in all CO2 treatments. Differences in photosynthetic response to growth over a wide range of CO2 partial pressures suggest changes in the relative performance of C3 and C4 annuals as atmospheric CO2 has fluctuated over geologic time.  相似文献

Summary The proliferation and survival of avocado nodal cultures of juvenile origin were affected by the form and concentration of nitrogen. Optimum growth was achieved on modified Murashige and Skoog medium containing 67% KNO3 and 33% NH4NO3 with total N of 40 mM supplemented with 100 mg l−1 myo-inositol, 1 mg l−1 thiamine HCl, 30 g l−1 sucrose, and 4.44 μM BA with a 16-h photoperiod (120–150 μmol m−2 s−1). Proliferating shoots and plantlets were photosynthetically active. Better shoot growth and accumulation of higher biomass occurred in a CO2-enriched environment than under ambient CO2 conditions. CO2 assimilation efficiency, however, was higher under the latter conditions than in a CO2-enhanced environment, e.g., 31±7 and 17±2 μmol CO2 m−2 s−1, respectively. The net CO2 assimilation rates of in vitro grown plantlets were comparable to those of seedlings ex vitro.  相似文献

During growth of Acetobacterium woodii on fructose, glucose or lactate in a medium containing less than 0.04% bicarbonate, molecular hydrogen was evolved up to 0.1 mol per mol of substrate. Under an H2-atmosphere growth of A. woodii with organic substrates was completely inhibited whereas under an H2/CO2-atmosphere rapid growth occurred. Under these conditions H2+CO2 and the organic substrate were utilized simultaneously indicating that A. woodii was able to grow mixotrophically. Clostridium aceticum differed from A. woodii in that H2 was only evolved in the stationary phase, that the inhibition by H2 was observed at pH 8.5 but not at pH 7.5, anf that in the presence of fructose and H2+CO2 only fructose was utilized.The hydrogenase activity of fructose-grown cells of C. aceticum amounted to only 12% of that of H2+CO2-grown cells. With A. woodii a corresponding decrease of the activity of this enzyme was not observed.  相似文献

Seedlings of loblolly pine (Pinus taeda L.) were grown under varying conditions of soil nitrogen and atmospheric carbon dioxide availability to investigate the interactive effects of these resources on the energetic requirements for leaf growth. Increasing the ambient CO2 partial pressure from 35 to 65 Pa increased seedling growth only when soil nitrogen was high. Biomass increased by 55% and photosynthesis increased by 13% after 100 days of CO2 enrichment. Leaves from seedlings grown in high soil nitrogen were 7.0% more expensive on a g glucose g–1 dry mass basis to produce than those grown in low nitrogen, while elevated CO2 decreased leaf cost by 3.5%. Nitrogen and CO2 availability had an interactive effect on leaf construction cost expressed on an area basis, reflecting source-sink interactions. When both resources were abundant, leaf construction cost on an area basis was relatively high (81.8±3.0 g glucose m–2) compared to leaves from high nitrogen, low CO2 seedlings (56.3±3.0 g glucose m–2) and low nitrogen, low CO2 seedlings (67.1±2.7 g glucose m–2). Leaf construction cost appears to respond to alterations in the utilization of photoassimilates mediated by resource availability.  相似文献

A marine filamentous cyanobacterium capable of rapid growth under N2-fixing conditions has been isolated from the Texas Gulf Coast. This organism appears to be an Anabaena sp. and has been given the strain designation CA. Cultures grown on mineral salts medium bubbled with 1% CO2-enriched air at 42°C show a growth rate of 5.6±0.1 generations per day with molecular nitrogen as the sole nitrogen source. This growth rate is higher than any other reported in the literature to date for heterocystous cyanobacteria growing on N2. Under similar growth conditions, 7.5 mM NH4Cl yields a growth rate of 6.6±0.1 generations per day while 7.5 mM KNO3 allows for a growth rate of 5.8±0.4 generations-day. Nitrogen-fixation rates, as measured by acetylene reduction, show maximum activity values in the range of 50–100 nmoles ethylene produced/minxmg protein. These values compare favorably with those obtained from heterotrophic bacteria and are much higher than values reported for other cyanobacteria. Growth experiments indicate that the organism requires relatively high levels of sodium and grows maximally at 42°C. Because of its high growth rate on N2, this newly isolated organism appears ideal for studying nitrogen metabolism and heterocyst development among the cyanobacteria.  相似文献

In order to better elucidate fixed-C partitioning, nutrient acquisition and water relations of prairie grasses under elevated [CO2], we grew the C4 grass Bouteloua gracilis (H.B.K.) lag ex Steud. from seed in soil-packed, column-lysimeters in two growth chambers maintained at current ambient [CO2] (350 μL L−1) and twice enriched [CO2] (700 μL L−1). Once established, plants were deficit irrigated; growth chamber conditions were maintained at day/night temperatures of 25/16°C, relative humidities of 35%/90% and a 14-hour photoperiod to simulate summer conditions on the shortgrass steppe in eastern Colorado. After 11 weeks of growth, plants grown under CO2 enrichment had produced 35% and 65% greater total and root biomass, respectively, and had twice the level of vesicular-arbuscular mycorrhizal (VAM) infection (19.8% versus 10.8%) as plants grown under current ambient [CO2]. The CO2-enriched plants also exhibited greater leaf water potentials and higher plant water use efficiencies. Plant N uptake was reduced by CO2 enrichment, while P uptake appeared little influenced by CO2 regime. Under the conditions of the experiment, CO2 enrichment increased root biomass and VAM infection via stimulated growth and adjustments in C partitioning below-ground. Summary Elodea canadensis grows over a wide range of inorganic carbon, nutrient, and light conditions in lakes and streams. Affinity for HCO 3 - use during photosynthesis ranged from strong to weak in Elodea collected from seven localities with different HCO 3 - and CO2 concentrations. The response to HCO 3 - was also very plastic in plants grown in the laboratory at high HCO 3 - concentrations and CO2 concentrations varying from 14.8 to 2,200 M. Bicarbonate affinity was markedly reduced with increasing CO2 concentrations in the growth medium so that ultimately HCO 3 - use was not detectable. High CO2 concentrations also decreased CO2 affinity and induced high CO2 compensation points (360M CO2) and tenfold higher half-saturation values (800 M CO2).The variable HCO 3 - affinity is probably environmentally based. Elodea is a recently introduced species in Denmark, where it reproduces only vegetatively, leaving little opportunity for genetic variation. More important, local populations in the same water system had different HCO 3 - affinities, and a similar variation was created by exposing one plant collection to different laboratory conditions.Bicarbonate use enabled Elodea to photosynthesize rapidly in waters of high alkalinity and enhanced the carbon-extracting capacity by maintaining photosynthesis above pH 10. On the other hand, use of HCO 3 - represents an investment in transport apparatus and energy which is probably not profitable when CO2 is high and HCO 3 - is low. The future capacity of forest ecosystems to sequester atmospheric carbon is likely to be influenced by CO2-mediated shifts in nutrient cycling through changes in litter chemistry, and by interactions with pollutants like O3. We evaluated the independent and interactive effects of elevated CO2 (560 μl l−1) and O3 (55 nl l l−1) on leaf litter decomposition in trembling aspen (Populus tremuloides) and paper birch (Betula papyrifera) at the Aspen free air CO2 enrichment (FACE) site (Wisconsin, USA). Fumigation treatments consisted of replicated ambient, +CO2, +O3, and +CO2 + O3 FACE rings. We followed mass loss and litter chemistry over 23 months, using reciprocally transplanted litterbags to separate substrate quality from environment effects. Aspen decayed more slowly than birch across all treatment conditions, and changes in decomposition dynamics of both species were driven by shifts in substrate quality rather than by fumigation environment. Aspen litter produced under elevated CO2 decayed more slowly than litter produced under ambient CO2, and this effect was exacerbated by elevated O3. Similarly, birch litter produced under elevated CO2 also decayed more slowly than litter produced under ambient CO2. In contrast to results for aspen, however, elevated O3 accelerated birch decay under ambient CO2, but decelerated decay under enriched CO2. Changes in decomposition rates (k-values) were due to CO2- and O3-mediated shifts in litter quality, particularly levels of carbohydrates, nitrogen, and tannins. These results suggest that in early-successional forests of the future, elevated concentrations of CO2 will likely reduce leaf litter decomposition, although the magnitude of effect will vary among species and in response to interactions with tropospheric O3.  相似文献

Tobacco leaves (Nicotiana tabacum var. Wisconsin 38) submitted to anaerobic conditions behave in a manner similar to that of maize, sugarcane, or sorghum leaves (C4-plants); more precisely, a lag time in O2 release is exhibited when the leaves are exposed to light after treatment in the dark under pure nitrogen. Although the conditions for the appearance of this phenomenon in tobacco are somewhat different, the main features are identical to those observed with maize: abolition of the lag time upon immediate exposure to light, release of CO2 under light (illumination burst of CO2), photochemical nature of the reactions involved in the abolition of the lag time, activation of oxygen release by far-red light, and the antagonistic effect of red and far-red light on the lag time. The high CO2 compensation point of tobacco leaves permits the classification of this plant among the C3 group. A comparison of these experimental results with others from the literature suggests than the distinguishing features between C3- and C4-plants are not as sharp as generally thought.  相似文献
