Colonisation of Pinus halepensis roots by GFP-tagged Pseudomonas fluorescens Aur6 was monitored by epifluorescence microscopy and dilution plating. Aur6-GFP was able to colonise and proliferate on P. halepensis roots. Co-inoculation with the ectomycorrhizal fungus Suillus granulatus did not affect the bacterial colonisation pattern whereas it had an effect on bacterial density. Bacterial counts increased during the first 20 days of seedling growth, irrespective of seedlings being mycorrhizal or not. After 40 days, bacterial density significantly decreased and bacteria concentrated on the upper two-thirds of the pine root. The presence of S. granulatus significantly stimulated survival of bacteria in the root elongation zone where fungal colonisation was higher. The number of mycorrhizas formed by S. granulatus was not affected by co-inoculation with Aur6-GFP. Neither Aur6-GFP nor S. granulatus stimulated P. halepensis development when inoculated alone, but a synergistic effect was observed on seedling growth when bacteria and fungus were co-inoculated.  相似文献

Ecophysiological and structural traits of seedlings of the water-saver Pinus halepensis and the water-spenders Quercus coccifera and Q. ilex were studied in response to water stress under greenhouse conditions. Water deficit reduced stomatal conductance (g s) and, as a consequence, both net CO2 assimilation (A) and transpiration rate (E) were also reduced. Water stress also emphasized midday down-regulation of the photochemical efficiency (dynamic photoinhibition) reducing quantum yield of noncyclic electron transport (ΦPSII), photochemical quenching (qP) and photochemical efficiency of the open reaction centres of PSII () and involved an increase of thermal dissipation of excess energy. However, water stress not only induced dynamic photoinhibition but also brought a reduction in F v/F m (chronic photoinhibition). Despite the water-saving strategy of P. halepensis that limited net CO2 assimilation, this species showed a higher photochemical efficiency and lower photoinhibition than Quercus species. This was not the result of a different photochemical quenching but was linked to a higher value of , indicating a less severe photo-inactivation of PSII. Water stress resulted in a loss of pigment content and in an increase of the carotenoids/chlorophyll ratio, antioxidant capacity and the biomass rate allocated to roots as opposed to that assigned to leaves. P. halepensis showed a lower photoinhibition and antioxidant activity than Quercus species due to its lower pigment content and higher proportion of carotenoids allowing P. halepensis to use, in a more effective way, the lesser excess energy absorbed.  相似文献

Planting woody vegetation is frequently a first step towards the restoration of degraded drylands. Seedling establishment on unfertile soils may be favoured by applying organic amendments such as biosolids. But the outcome of such a practice is strongly dependent on the type of amendment and the application rate used. High application rates may have deleterious effects on plant performance and compromise plantation success. Thus amendment type and dose should be carefully selected to optimise benefits and minimize risks. In this study, we evaluated the effect of two organic amendments (composted and air-dried sewage sludge) applied at 5 doses (0, 15, 30, 45 and 60 Mg ha?1) on soil properties and on the performance of 1-year-old Pinus halepensis seedlings planted in a dry Mediterranean degraded area. Soil organic matter, electrical conductivity and nutrient availability increased with the application rate, but the magnitude of this increase depended on the type of amendment and the time. Organic amendments improved N and P status and promoted seedling growth. Nevertheless, at the higher application rates they showed a negative impact on seedling survival 1 and 3 years after application. Drought effects intensified by root competition with extant vegetation and reduced water availability within the planting hole were the main causes attributed to the higher mortality. Low to moderate doses showed the best combination of seedling survival and growth and can thus be recommended to promote the establishment of P. halepensis in dry Mediterranean areas.  相似文献

Abstract

The process of primary growth in 2-year-old seedlings of 11 populations of Pinus halepensis Mill. is described. At the end of the first growing season one type of apical structure was observed: type-1, a tuft of primary needles placed close together, surrounding and protecting a meristematic apex.

At the end of the 2nd growing season, three types of apical structure were observed: type-1; type-2, a terminal winter bud; and type-3; a «bud» with characteristics of both type-1 and type-2.

Morphological observation along with an anatomical examination of the winter bud led to the conclusion that the definitive growth pattern in juvenile P. halepensis is monocyclic with a variable number of summer shoots. This growth pattern is reached by some P. halepensis populations in 3–4 years, by contrast, in other pine species two years are usually needed.

The populations studied differed both in growth potential (differences in number of cycles, ratio of first cycle to total growth, growth rates) and in the developmental stages of the apical meristem.

Four groups could be identified: (i) Morocco and Spain, (a limited growth, few cycles, a high ratio of 1st cycle to total growth, and growth in 2nd season almost entirely due to free growth); (ii) Algeria and Greece, (moderate to low growth, a large number of cycles, a low ratio of 1st cycle to total growth, and very early formation of apical structure with preformed primordia); (iii) Israel and Central Italy (a high growth, a large number of cycles, a medium ratio of 1st cycle to total growth, and early formation of apical structure with preformed primordia); (iiii) Greece, France and Italy, which was intermediate between group (i) and the other 2 groups.  相似文献

油松菌根促生细菌——荧光假单胞菌的分离与鉴定   总被引:3,自引:2,他引:3  
为了探讨菌根促生细菌荧光假单胞菌(Pseudomonas fluorescens)与菌根真菌的互作关系,本实验从油松菌根上分离得到36株在紫外灯下产荧光的细菌菌株,以荧光假单胞菌9702作为标准菌株,对分离菌株进行显微观察、生物学鉴定和16S rDNA序列分析,结果确定HDY-8、HDY-9、HDY-20、HDY-35共 4株菌株为荧光假单胞菌,并分别命名为P.fluorescens HDY-8、P.fluorescens HDY-9、P.fluorescens HDY-20、P.fluorescens HDY-35.用这4株细菌菌株分别与外生菌根真菌(ECMF)粘盖牛肝菌(Suillus bovinus)、褐环粘盖牛肝菌(Suillus luteus)和褐黄牛肝菌(Boletus luridus)进行纯培养互作研究.结果表明,只有P.fluorescens HDY-20对3种外生菌根真菌有不同程度的促生作用,并对S.luteus促进效果最好,S.bovinus次之,B.luridus最差;P.fluorescens HDY-20促进S.bovinus、S.luteus和B.luridus菌丝生长的最佳浓度分别为2.4×109 cfu/mL、0.8×109~2.4×109 cfu/mL和0.8×109 cfu/mL,与对照相比S.bovinus和S.luteus的生物量达到极显著差异(P<0.01),B.luridus的达到显著差异(P<0.05),且分别比对照增加6.5%、9.1%和4.3%.  相似文献

Previous studies on Tuber borchii fruit bodies in early maturation stages suggested a role of bacteria in sporocarp structural modifications. In order to verify this hypothesis, in the present study we investigated by means of microbial and ultrastructural approaches, the bacterial population of T. borchii sporocarps from intermediate maturation phases to advanced decomposition stages, paying particular attention to chitinolytic and cellulolytic bacteria and to their relationships with ascii and ascospores. We found that Pseudomonas fluorescens and spore-forming Bacillaceae, both able to degrade cellulose and chitin, are present inside the sporocarps in all maturation stages investigated. Moreover, rod-shaped bacteria seem able to erode ascus walls and colonize the interior of ascii containing mature spores. These results suggest a possible role of these bacteria in the process of ascus opening. Moreover, the presence of P. fluorescens and Bacillaceae on isolated mature spores after decontamination suggests an intimate association between these bacteria and the ascospores.  相似文献

Abstract

In the present article, magnetic resonance imaging spectroscopy (MRI) was used to study fresh black truffle, with the aim of elucidating the internal structure, the effects of fungal invasion, and physical changes occurring in post-harvest. MRI is a non-destructive and non-invasive analytical technique offering the almost unique opportunity of studying foodstuff while leaving it intact, without any sample preparation. The internal morphology and modifications induced by external agents (e.g. fungal invasion, diseases, dehydration) are some of the aspects addressed by means of this innovative analytical tool.  相似文献

Pinus sylvestris seedlings, grown in a vertical petri dish system, were inoculated with five different mycorrhizal fungi. Half of the root system in the petri dish was subjected to periodic flooding, and mycorrhizal colonization was studied. Thelephora terrestris, Laccaria laccata, and Hebeloma crustuliniforme were not sensitive to flooding, whereas Suillus flavidus and S. bovinus were highly sensitive. The latter failed to colonize the root even when flooded for only 2 min per day four times a week.  相似文献

An experiment was carried out in open-top chambers located in eastern Spain. One-yr-old Pinus halepensis Mill. seedlings were exposed during three consecutive summers to the following ozone (O3) treatments: charcoal-filtered air (CFA), non-filtered air (NFA) or non-filtered air plus 40 nl l−1 O3, 9 h d−1, 5 d wk−1 (NFA+40). Seasonal variations in Aleppo pine performance were observed since reductions in chlorophyll and cellular peroxidase levels associated with increases in superoxide dismutase activity, were recorded during the summer. Similarly, a reduction in epoxidation state was found at midday during the summer, derived from an activation of the xanthophyll cycle associated to an increment in radiation and temperature levels.
The first O3-induced effects were recorded in previous-year needles (1991) during the first summer exposure as an increase in extracellular and total peroxidase activities and in zeaxanthin levels in the NFA+40 treatment along with a trend to a higher SOD activity in this treatment. A carry-over effect was detected since a lower winter recovery of chlorophyll levels was found in the NFA+40 seedlings along with a reduction of xanthophyll levels. A reduction in chlorophyll levels was observed in the previous-year needles (1992) from the NFA+40 treatment at the end of the second fumigation period. Realistic ozone exposures induced alterations in plant antioxidative systems and plant pigments as shown in this paper. These observations together with the reductions in stomatal conductance and net photosynthesis recorded in the same experiment, indicate that Aleppo pine is a species sensitive to ozone.  相似文献

Net photosynthesis, transpiration, dark respiration rates and stomatal and mesophyll resistances were studied in young potted seedlings of Pinus halepensis Mill. under gradually decreasing soil and leaf water potentials. Stomatal resistance under non-limiting xylem water potentials was 6–7 times higher than mesophyll resistance. Stomata started to close at threshold xylem water potentials of −0.8 MPa, whereas mesophyll resistance started to increase at about −1.4 MPa. Decreasing xylem water potentials increased the CO2 compensation point and decreased the water use efficiency (expressed by the photosynthesis to transpiration ratio) and dark respiration rate. It is concluded that at least part of the drought resistance characteristics of P. halepensis are associated with a sensitive stomatal mechanism which enables an efficient control of water loss.  相似文献

A series of substituted kynurenines (3-bromo-dl, 3-chloro-dl, 3-fluoro-dl, 3-methyl-dl, 5-bromo-l, 5-chloro-l, 3,5-dibromo-l and 5-bromo-3-chloro-dl) have been synthesized and tested for their substrate activity with human and Pseudomonas fluorescens kynureninase. All of the substituted kynurenines examined have substrate activity with both human as well as P. fluorescens kynureninase. For the human enzyme, 3- and 5-substituted kynurenines have kcat and kcat/Km values higher than l-kynurenine, but less than that of the physiological substrate, 3-hydroxykynurenine. However, 3,5-dibromo- and 5-bromo-3-chlorokynurenine have kcat and kcat/Km values close to that of 3-hydroxykynurenine with human kynureninase. The effects of the 3-halo substituents on the reactivity with human kynureninase may be due to electronic effects and/or halogen bonding. In contrast, for the bacterial enzyme, 3-methyl, 3-halo and 3,5-dihalokynurenines are much poorer substrates, while 3-fluoro, 5-bromo, and 5-chlorokynurenine have kcat and kcat/Km values comparable to that of its physiological substrate, l-kynurenine. Thus, 5-bromo and 5-chloro-l-kynurenine are good substrates for both human as well as bacterial enzyme, indicating that both enzymes have space for substituents in the active site near C-5. The increased activity of the 5-halokynurenines may be due to van der Waals contacts or hydrophobic effects. These results may be useful in the design of potent and/or selective inhibitors of human and bacterial kynureninase.  相似文献

Truffles are hypogeous ectomycorrhizal (EM) fungi belonging to the genus Tuber. Although outplanting of truffle-inoculated host plants has enabled the realization of productive orchards, truffle cultivation is not yet standardized. Therefore, monitoring the distribution of fungal species in different truffle fields may help us to elucidate the factors that shape microbial communities and influence the propagation and fruiting of Tuber spp. In this study, we compared the fungal biodiversity in cultivated and natural Tuber melanosporum truffle fields located in Central Italy. To this end, ectomycorrhizas (ECM) and soil samples were molecularly analyzed, and an inventory of the fungi associated with Quercus pubescens plants colonized by T. melanosporum, Tuber aestivum or Tuber brumale was compiled. T. melanosporum and T. aestivum were dominant on the cultivated plants, and the number of EM species was markedly lower in the cultivated sites than in the natural sites. However, in the same site, EM biodiversity was higher in T. brumale-colonized plants than in T. melanosporum-colonized plants. These results suggest that different Tuber spp. may have different competitive effects on the other mycobionts. Additionally, in keeping with our previous findings, we found that the number of T. melanosporum genotypes recovered from the soil samples was higher than that of the underlying ECM.  相似文献
