Explants of the Indo-Pacific sponge Pseudosuberites aff. andrewsi were fed with the microalgae Chlorella sorokiniana and Rhodomonas sp. It was microscopically observed that these algae were ingested and digested by the sponge cells, suggesting that they were consumed by the sponges. The algae were further used for two growth experiments with five explants of P. aff. andrewsi and four explants of P. andrewsi. Growth was measured as the increase in projected body area. The explants showed considerable growth (up to 730% in 54 days for P. aff. andrewsi and up to 680% in 22 days for P. andrewsi), which is much higher than previously reported growth rates for sponges. Growth started after a stationary phase of 5–20 days in which the projected body area did not increase. The growth of P. aff. andrewsi appeared to be linear and was inhibited at the end of the experiment. Two explants of P. andrewsi showed exponential growth instead of linear growth. Hence, no general statements about the growth kinetics of these sponges can be made at this time. However, the high growth rates found in this study suggest a promising future for cultivation of sponges in closed systems.  相似文献

Oxygen microprofiles were measured over the boundary layer and into the tissue of 10-day-old cultivated tissue fragments (explants of 2-4 cm3) from the choanosome of the cold-water sponge Geodia barretti with oxygen-sensitive Clark-type microelectrodes. At this time of cultivation, the surface tissue and the aquiferous system of the explants is regenerating, which makes oxygen and nutrient supply by pumping activity impossible. Oxygen profiles showed a parabolic shape, indicating oxygen flux over a diffusive boundary layer and into the tissue. Oxygen was always depleted only 1 mm below the sponge surface, leaving the major part of the explants anoxic. Diffusive oxygen flux into the explant was calculated from three oxygen profiles using Fick's first law of diffusion and revealed 9 μmol O2 cm-3 day-1, which is in the lower range of in situ oxygen consumption of whole sponges. The ability of G. barretti to handle continuous tissue anoxia enables choanosomal explants to survive the critical first weeks of cultivation without a functional aquiferous system, when oxygen is supplied to the sponge explant by molecular diffusion over its surface.  相似文献

Interactions between guanine nucleotide regulatory proteins (G proteins) and muscarinic acetylcholine receptors (mAChRs) were studied in vivo following carbachol treatment. Rat brain homogenates were separated by high speed ultracentrigation into heavy and light membrane and 300,000 g supernate franctions. The G proteins were partially purified by Sephadex-G200 and heptylamine-Sepharose and the mAChRs by (3,2′-aminobenzhydryloxy)-tropane-(ABT)-affinity chromatographies. Radioligand binding assays showed that acute carbachol induced a biphasic translocation of the mAChRs and G proteins into the light membrane fraction with an initial release at 5–10 min and a second phase at 60 min. Portions of the released mAChRs and the G proteins, were found in the 300,000 g supernates and light membranes and were eluted in the same peak fractions from a Sephadex G-200 column. This dually labelled peak dissociated in the presence of digitonin, suggesting close association between the mAChR and G protein. ABT-affinity chromatography yielded dually labelled mAChR-G protein fractions which eluated as a single radioactive peak on a second ABT column. the partially purified G proteins from these fractions were photoaffinity labelled with 8-azidoguanosine-5′-triphosphate, [γ-32P]. SDS-PAGE autoradiography revealed the presence of G and Gi which may be released simultaneously with the mAChRs from the plasma membrane. In addition, a 110,000 molecular weight polypeptide was dually labelled by [3H]-PrBCM and [γ-32P]-8-azido-GTP suggesting the presence of a “mAChR-G protein complex.” These findings provide direct evidence for the release of mAChRs and G proteins and a mAChR-G protein complex by agonist occupation of the mAChRs.  相似文献

Two sediment cores retrieved from the continental slope in the Benguela Upwelling System, GeoB 1706 (19°33.7′S 11°10.5′E) and GeoB 1711 (23°18.9′S, 12°22.6′E), reveal striking variations in planktonic foraminiferal abundances during the last 160,000 years. These fluctuations are investigated to assess changes in the intensity and position of the upwelling centres off Namibia. Four species make up over 95% of the variation within the core, and enable the record to be divided into episodes characterized by particular planktonic foraminiferal assemblages. The fossil assemblages have meaningful ecological significance when compared to those of the modern day and the relationship to their environment. The cold-water planktonic foraminifer, Neogloboquadrina pachyderma sinistral [N. pachyderma (s)], dominates the modern-day, coastal upwelling centres, and Neogloboquadrina pachyderma dextral and Globigerina bulloides characterize the fringes of the upwelling cells. Globorotalia inflata is representative of the offshore boundary between newly upwelled waters and the transitional, reduced nutrient levels of the subtropical waters. In the fossil record, episodes of high N. pachyderma (s) abundances are interpreted as evidence of increased upwelling intensity, and the associated increase in nutrients. The N. pachyderma (s) record suggests temporal shifts in the intensity of upwelling, and corresponding trophic domains, that do not follow the typical glacial-interglacial pattern. Periods of high N. pachyderma (s) abundance describe rapid, discrete events dominating isotope stages 3 and 2. The timing of these events correlates to the temporal shifts of the Angola-Benguela Front (Jansen et al., 1997) situated to the north of the Walvis Ridge. Absence of high abundances of N. pachyderma (s) from the continental slope of the southern Cape Basin indicates that Southern Ocean surface water advection has not exerted a major influence on the Benguela Current System. The coincidence of increased upwelling intensity with the movement of the Angola-Benguela Front can be interpreted mainly by changes in strength and zonality of the trade wind system.  相似文献

Photosynthetic features of Zostera marina L. and its autotrophic epiphyte community were investigated in a population inhabiting a shallow (1.3 m depth) water meadow in Great Harbor, Woods Hole, MA (U.S.A.). Photosynthesis versus irradiance (P-I) relationships were measured with respect to leaf age determined by the leaf position in the shoot bundle and by location of the tissue along the leaf axis. Therefore both age and light intensity gradients along the leaf axis were considered. The maximum photosynthesis (Pmax) per dm2 typically increased nearly two-fold along the leaf axis from leaf bases to apices. Photosynthetic rate on a chlorophyll (Chl) basis did not increase as dramatically along the leaf axis, and rates were usually lowest in tissues with the highest Chl content. The P-I relationships of leaves of different ages did not reveal photoinhibition even at light intensities > 1400 μE • m−2 • s −1. Furthermore, no photoinhibition was observed in tissues from leaf blade bases, which never experienced high light levels (> 500 μE • m −2 • s−1) in situ in Great Harbor. The initial slopes of the P-I curves and light compensation and saturation values varied along the leaf axis in relation to in situ light intensity gradients and in relation to leaf or tissue age. It appeared that leaf and/or tissue age was more important than light environment in determining P-I responses. The contribution of the autotrophic epiphyte community on Z. marina leaves to total photosynthesis per dm2 was between 27 and 50%, and between 10 and 44% per mg chlorophyll. These levels of epiphyte photosynthesis can double the primary production of Z. marina leaves. No detrimental effects of epiphyte cover were realized in leaf maximal photosynthesis or P-I relationships. Non-epiphytized leaves and leaves from which epiphytes were removed showed essentially identical photosynthetic features. Light intensity and age gradients along the leaf axis control both the photosynthetic performance of the leaves and epiphyte biomass and photosynthesis.  相似文献

Explants of the tropical sponge Pseudosuberites andrewsi were fed with the marine diatom Phaeodactylum tricornotum. The food was supplied either as intact algae or as a filtered crude extract. Growth (measured as an increase in underwater weight) was found in both experiments. The explants fed with intact algae increased to an average underwater weight of 255% of the initial weight in 45-60 days. The explants fed with crude extract increased to an average of 200% of the initial weight in 30 days. These results show that it is possible to grow a sponge using a single microorganism species as a food source. In addition, it was demonstrated that sponges are also capable of growing on non-particulate food. Therefore, this study is an important step forward towards the development of controlled, in vivo sponge cultures.  相似文献

Abstract The wave‐exposed nature of much of the southwestern Australian coastline considerably reduces the protective influence of seagrasses, and sediment movement appears to be relatively unaffected by their presence. Present seagrass restoration efforts focus on the deployment of large mechanically transplanted “sods” of seagrass as a means of combating the negative effects of water motion on transplant survival. The aim of this study was to investigate the combined role of wave energy and transplant spacing on sediment movement and transplant survival to provide guidance for seagrass transplantation in areas of high wave energy. One hundred sixty sods (0.25 m2) of seagrass were mechanically extracted from a mixed meadow consisting of Amphibolis griffithii (Cymodoceaceae) and Posidonia coriacea (Posidoniaceae) and planted in a high wave energy site with the treatments configured as three replicates of 16 sods placed in 4 × 4–meter squares at distances of 0.5, 1.0, and 2.0 meters apart. An additional 16 single sods were planted randomly throughout the site. Monitoring was conducted at two monthly intervals and consisted of counting the number of sods surviving and measuring the shoot density of seagrass species within each surviving sod. Sediment height was monitored using a series of sediment plates and an electronic sediment level sensor. Sod spacing had no significant effect upon transplant survival, which remained above 90% for 4 months after transplantation and then declined with the onset of winter (June to August). After 14 months individual sod survival was between 9% and 40%. Initial shoot densities were 200 to 500 shoots/m2 and declined to less than 50 shoots/m2. Sediment fluctuations up to 35 cm were noted, occasionally taking place over a matter of hours, and storms during winter caused significantly increased sediment movement. This probably curtailed rhizome extension and prevented the expansion of the transplants. This study indicates that the ability of seagrasses to influence sediment would appear to vary with the prevailing hydrodynamic regime and that a reappraisal of the notion that all seagrass communities trap sediment is necessary.  相似文献

Human activities have inflicted profound damage upon many ecosystems, and ecologists are now seeking effective means of restoring ecosystems to their natural state. Industrial ports and harbors are highly modified and often depauperate in native fauna. They are typically characterized by poor water quality and modified community composition, both of which may hinder attempts to reintroduce native species. Here, we conducted a field experiment to separate the effects of the recipient environment and community composition on the success of endemic sponge explants in Port Kembla Harbor, NSW, Australia. A reciprocal transplant was conducted between communities originating from six sites that varied in water quality and community composition, enabling us to assess the relative factors simultaneously. A colony of the endemic sponge Tedania anhelans was then inserted into the center of each community, and we quantified the survival, growth, and metal bioaccumulation of sponges over three months. Endemic sponges consistently performed better against resident assemblages when water quality was good. Sponges transplanted to cleaner sites had over double the survivorship and approximately three times the surface area of sponges transplanted to disturbed sites. These patterns were independent of community composition. Bioaccumulation of metals in sponges was correlated with survival; however, other factors such as turbidity may be required to explain sponge mortality at some sites. This study adds to evidence that remediation of the physical and chemical environments may be a prerequisite for biological remediation and demonstrates the value of experimental transplants in assessing restoration potential.  相似文献

天津芦苇(Phragmites australis)沼泽具有重要的生态功能。目前天津地区水体咸化、氮污染和水资源短缺问题严重, 显著影响了芦苇湿地的植物生理生态过程。植物稳定碳同位素组成(碳稳定同位素比值(δ13C))能够记录与植物生长过程相关联的环境变化信息, 反映植物对环境变化的生理生态适应特性。该研究调查了天津七里海、北大港和大黄堡湿地芦苇叶片的δ13C分布特征, 探讨了影响该地区叶片δ13C值变化的主要因素。研究表明: 1)天津芦苇湿地植物叶片δ13C的变化范围在-26.3‰ - -23.6 ‰之间, 平均值为-25.8‰; 2)芦苇叶片δ13C与底泥相对含水量呈显著负相关关系, 与底泥有效氮和全氮含量呈显著正相关关系, 而与底泥盐度和磷含量没有显著相关关系; 水分条件和底泥氮营养状况是影响叶片的δ13C值变化的主要因素; 3)淹水条件下, 芦苇叶片δ13C与叶片质量氮含量呈显著正相关关系, 与叶碳氮比呈显著负相关关系, 8月份七里海湿地干涸打破了此相关关系。当前环境压力下, 天津沼泽湿地干涸极大地改变了芦苇的氮、水平衡和植物对水、氮资源的利用策略, 而湿地干涸对该过程的影响要高于盐度和氮负荷增加。  相似文献

A reliable and reproducible method for plant regeneration in vitro of two important temperate eucalypts, Eucalyptus nitens and E. globulus, has been developed which utilises seedling explants. Highly regenerative callus was obtained from individual cotyledon and hypocotyledon explants of both species following cultivation on Murashige and Skoog’s (MS) basal nutrient medium supplemented with 30 g l−1 sucrose, 5–10% (v/v) coconut water, 0.8% agar, 1 mg l−1 -naphthalene-acetic acid (NAA) and 0.5 mg l−1 N6 benzylaminopurine (BAP). Shoot differentiation was observed 7–8 weeks after transfer of callus onto regeneration medium containing 0.5 mg l−1 NAA and 1 mg l−1 BAP. In a few instances, direct shoot regeneration occurred without an intervening callus phase in both species. The frequency of plant regeneration was higher for callus derived from hypocotyl segments (30–35%) compared to cotyledonary explants (20–25%) though the average number of shoots per cotyledonary explant was generally higher than for hypocotyl explants. Somatic embryos were observed occasionally in E. nitens, arising from the surface of organogenic callus. Organised structures closely resembling somatic embryos were also observed in E. globulus. Regenerated shoots (30–40%) of both species could be rooted in modified MS media containing indole-3-butyric acid (IBA) and plantlets were successfully transferred to soil.  相似文献

Five species of amphibious stichaeoid fishes (four stichaeids, one pholidid) with distinct but overlapping distribution patterns in the rocky intertidal zone of central California were studied in the laboratory with respect to body size and shape, survival out of water, evaporative water loss and water resorption upon re-immersion. Members of all species survived periods of emersion of at least 6 h, but maximum survival times varied with species and body size. Surface area to weight ratios in relation to body length were highest in Xiphister atropurpureus (Kittlitz) and X. mucosus (Girard), lowest in Cebidichthys violaceus (Girard) and Anoplarchus purpurescens Gill and most variable in Xererpes fucorum (Jordan & Gilbert) (the pholidid). Weight-specific water loss (mg · g−1 · h−1) was highest in the two species of Xiphister followed in order by Xererpes fucorum, Cebidichthys violaceus and Anoplarchus purpurescens.

In general, adaptive relationships were found between body size, surface area, tolerance of water loss and intertidal distribution patterns. Among juvenile stichaeoids, for which problems of emersion are most critical, those of Cebidichthys violaceus possess the combination of attributes, including small surface area to weight ratio, high water content and long tolerance of emersion, that should give them an adaptive edge in occupying the upper levels of the shore. In accordance with these characteristics, C. violaceus has the highest, most exposed vertical distribution of the five study species.  相似文献

生存在不同基底颜色环境下的爬行动物种群通常表现出丰富的体色地理变异, 其体色变化的潜在机制具有多样性。变色沙蜥(Phrynocephalus versicolor)和草原沙蜥(P. frontalis)具有较近的遗传关系, 曾被认为与荒漠沙蜥(P. przewalskii)组成同一系统发育种组。本文应用光纤光谱仪(AvaSpec-2048), 通过记录沙蜥背部体表12个部位的皮肤光反射率, 定量比较在黑化环境下的深色变色沙蜥与非黑化环境下的浅色草原沙蜥自然体色变异, 研究其种群体色变异是否具有时间可逆性, 并探讨基底颜色对沙蜥体色的影响机制。研究结果表明, 黑化生境下的变色沙蜥体色显著深于非黑化枯黄色生境下的草原沙蜥。此外, 对黑化与非黑化样本开展的生境互换移植围栏实验, 即把枯黄色生境中非黑化的草原沙蜥移植于黑色的基底环境中饲养, 把黑化生境中黑化的变色沙蜥移植于枯黄色生境中饲养。结果表明, 饲养1周后黑化群体背部6个检测部位的光反射率显著变大, 其他部位均无显著变化; 而非黑化群体只有左后肢和背部右上方2个部位的皮肤光反射率发生显著变化, 其他部位反射率无显著变化。结果表明, 变色沙蜥体色变异能力比草原沙蜥强, 体色表型可能已经在两个近缘沙蜥物种中稳定遗传, 基底生境颜色的短期变化在统计学上能引起肉眼难以识别的轻微的体色变异, 个体发育相关的一些遗传因素可能对体色变异起控制 作用。  相似文献

Aims Grazing activities degrade soil aggregates, reduce vegetation coverage and affect the amount of deposited material, and make the land more vulnerable to wind erosion. Although livestock increase was considered as the main issue leading to the degradation, only very few studies have quantitatively investigated the relationship between grazing and soil erosion. The relationship between different stocking rates and sediment flux, and sediment soil particle was studied to reveal the mechanism of different grazing intensities on soil erosion process, to provide basic parameters for grazing optimization in the Stipa breviflora desert steppe. Methods In the Stipa breviflora desert steppe research area, BSNE collecting sand boxes were set in the randomly distributed paddock experiment sites for 11 year with different grazing intensities (0.15、0.30、0.45、0 sheep·hm-2·month-1, corresponding to light grazing LG, moderate grazing MG, heavy grazing HG and control CK, respectively). The quantitative relationship between grazing intensity and sediment flux, and the characteristics of sediment soil particle were conducted in four sampling periods through 2 years (April 2013 to April 2015).Important findings (1) Grazing intensity had a significant effect on the sediment flux (p< 0.05), and the sediment flux increased with the increase of grazing intensity. The response of sediment flux to grazing intensity was variable with season. The daily average sediment flux (13.12 g·m-1·d-1) during the period of April to October was smaller than that from October to April (18.74 g·m-1·d-1). The sediment flux difference of different grazing intensities was greater from April to October, with the 5 times daily average sand flux in the heavy grazing paddock that in the control. The average sediment flux difference of different grazing intensities was small from October to April. (2) The relationship between the natural logarithm of sediment flux at different height and the vertical height had a better binomial fitting from April to October, and there was no obvious regular pattern about flux vertical distribution from October to April, and the vertical flux difference of grazing intensities was mainly expressed in 0-50 cm layer. (3) Sand sediment particle ≤250 μm accounted for more than 85% of the total sediment, the sand sediment particle of ≤50 μm) size was significantly enriched, and the enrichment ratio increased with the increase of vertical height. The enrichment ratio of 125-250 μm particle and 50-125 μm particle decreased with the increase of vertical height, and the enrichment ratio of 125-250 μm particle was smaller than that of 50-125 μm particle (p< 0.05). Therefore grazing intensity had different influence on the sand flux in Stipa breviflora desert steppe, the greater the grazing intensity, the heavier the wind erosion was, and the effect of grazing intensity on grassland was enhanced by wind erosion.  相似文献

以内蒙古短花针茅(Stipa breviflora)草原为研究对象, 在放牧11年的样地布设BSNE集沙仪, 通过2013年4月到2015年4月4个采样期对短花针茅草原放牧强度与风沙通量的定量关系及风蚀物粒度特征进行了研究。结果表明: (1)放牧强度对风沙通量有显著影响(p < 0.05)。随着放牧强度增强, 风沙通量逐渐增加; 风沙通量对放牧强度的响应存在季节差异, 生长季的日平均风沙通量小于非生长季, 且生长季不同放牧强度间风沙通量差异较大, 其中重度放牧区风沙通量是对照区的5倍, 而非生长季不同放牧强度间风沙通量差异较小, 重度放牧区是对照区的1.7倍; (2)生长季不同高度风沙通量的自然对数和垂直高度之间存在较好的二项式拟合结果, 而非生长季风沙通量的垂直分布没有明显规律; 放牧强度间通量的垂直分布差异主要表现在0-50 cm高度; 在同样高度, 放牧强度大, 垂直通量也大; (3)风蚀沉积物中粒径≤250 μm颗粒富集度随垂直高度变化而变化; 风沙沉积物中, ≤250 μm的颗粒占沉积物总量的85%以上, 沉积物中粒径≤50 μm的颗粒有明显的富集作用, 且随垂直高度增加富集比逐渐增加; 粒径为125-250 μm和50-125 μm的颗粒表现为随垂直高度增加富集比降低的趋势, 且125-250 μm粒径的富集比显著小于50-125 μm颗粒的富集比; 荒漠草原放牧强度对风沙通量有不同程度的影响, 放牧强度越大, 风蚀越重, 风蚀作用强化了重度放牧对草地退化的影响作用。  相似文献

By using sites in the Great Marshes at Barnstable (Massachusetts, USA) this study examines the effects of a set of environmental parameters on the foraminiferal distribution. The studied parameters are: elevation above mean high water; salinity of the porewater; various sediment characteristics; vegetation; and food source. Relations between the environmental parameters and foraminiferal properties (frequencies, densities and diversities) are quantified with correlation coefficients. For the first time Siphotrochammina lobata and Balticammina pseudomacrescens are documented in the New England region.

The following species show a significant correlation with one or more of the studied parameters and are designated as key-species: Haplophragmoides manilaensis, Jadammina macrescens, Balticammina pseudomacrescens, Miliammina fusca and Tiphotrocha comprimata. Based on cluster analysis and the presence, absence or dominance of the key-species characteristic associations are distinguished. The distribution of three associations is indicative of specific marsh environments: the marsh fringe, the middle marsh and the marsh edge. These three marsh units are separated by their own salinity regime, flooding and sediment characteristics.

The marsh fringe is typified by the H. manilaensis Association and experiences freshwater input (seepage, surface runoff and rainwater) and only slight marine influence, resulting in low salinity values (2.5–20‰). The width of the marsh fringe is variable, dependent on the amount of seepage which in turn is controlled by the permeability of the basement and the peat. The J. macrescens Association characterizes the middle marsh where salinities are controlled by infiltration of sea- and rainwater and by evaporation. Salinity values are higher than 20‰, while temporarily salinity can reach extreme high values during periods without flooding and high evaporation rates (e.g., 44‰). The fully marine M. fusca Association occupies the daily flooded marsh edge where the salinities have the same values as Cape Cod bay water (ca. 28‰).

Unlike many other salt marshes the distribution of foraminiferal assemblages in the Great Marshes does not show a vertical zonation with respect to mean high water. This shows that a worldwide applicable model for paleoenvironmental studies in salt marshes based on foraminifera is not feasible. Each salt marsh has its own characteristics. Regional factors such as climate play an important role in the salinity regime, while the local upland characteristics determine if seepage takes place. Thus each marsh has its own foraminiferal fingerprint showing the opportunistic behaviour of the salt marsh agglutinants. A surface study is an indispensable first step in assessing the value of foraminifera as paleo-ecological indicators.  相似文献

调水调沙工程的实施为黄河三角洲带来大量淡水恢复退化湿地的同时,也改变了湿地的水沙状况并带来大量的外源物质。本研究采用3因素4水平的正交试验,以黄河口滨岸潮滩湿地典型的先锋物种碱蓬为研究对象,探讨了其对淹水深度、泥沙埋深及外源氮输入的适应机制。结果表明: 泥沙埋深对碱蓬叶片蛋白含量及超氧化物歧化酶(SOD)活性的影响较大;氮输入对过氧化物酶(POD)活性的影响较大,而3种处理对过氧化氢酶(CAT)活性的影响无显著差异。淹水深度对叶、茎及总干重具有显著影响,且随淹水深度的增加呈现减小趋势,最大值(25.70、40.86、69.73 g)均出现在2 cm淹水处理;而氮输入及泥沙埋深对碱蓬干重的影响不显著。极差分析表明,淹水深度对碱蓬叶、茎、根及总干重的影响最为显著,其次为氮输入和泥沙埋深,2 cm淹水深度+12 cm埋深深度+9 g·m-2氮输入量最有利于碱蓬的生长。可见,淹水深度的变化对碱蓬的生长起到决定性的作用,在调水调沙等工程的实施过程中应注重淹水深度的把控。  相似文献

Mussels, Mytilus edulis, were exposed for periods of 6 weeks at various locations in Dutch coastal waters during 1989 and 1990. “Survival in air” showed to be a sensitive response parameter for indicating pollution induced environmental stress in transplanted mussels sampled from eight field sites. Increased tissue contaminant levels, especially PCBs and PAHs, correlated with a reduced survival time during aerial exposure. Three weeks exposure of mussels in the laboratory to 1 μg · 1−1 PCBs affected the aerial survival time negatively. Laboratory experiments did not indicate that lowered salinity influences the “Survival in air” response after sufficient acclimation (15 days), facilitating the use of this response parameter in both marine and estuarine waters.  相似文献

Sponges are the dominant organisms on many coral reefs and through feeding they may greatly reduce the concentration of suspended food particles. Retention efficiencies of the tubular sponges Aplysina lacunosa, Callyspongia vaginalis and Niphates digitalis were examined on a coral reef located in the Florida Keys. Replicate ambient and exhalant water samples were collected in situ from individuals of each species and analysed using two methods. Retention efficiencies of suspended particles (0.75-18 µm) examined using Coulter counter analysis were similar among the three sponge species, averaging 86%. For all sponges, particle retention decreased as particle size increased from 0.7 to 18 µm. Water samples plated on to Marine Agar produced 54 microbial types. Retention efficiencies of culturable microbes were similar among the three species, averaging 82%. This study suggests that the coral reef sponges Aplysina lacunosa, Callyspongia vaginalis and Niphates digitalis play an important role in the transfer of energy between the pelagic and benthic environments.  相似文献

The aim of this study was the production of high numbers of M. anisopliae blastospores with enhanced germination efficiency under conditions of water-stress (ecological fitness). Different nitrogen sources and concentrations were screened for their ability to induce blastospore production while keeping pH and water activity (aw) at fixed levels (6.8, and 0.98 aw, respectively). After optimum nitrogen status was determined (cornsteep solid (CS) + yeast extract (YE); or cottonseed flour (CF) + YE), the effect of interaction of nitrogen source, pH (3.5, 5, 6.8, 8, 9 and 10) and solutes for aw adjustment (KCl, NaCl, PEG 200) on blastospore production, endogenous polyol content (glycerol, erythritol, arabitol and mannitol) and total protein, were determined. For both ionic (NaCl, KCl) and non-ionic solutes (PEG 200), optimum blastospore production (between 4×107 and 2×108 blastospores ml-1) and growth occurred in the pH range 6.8-8, with the CF + YE nitrogen profile giving higher yields than CS + YE. Optimum conditions for high erythritol and total protein endogenous concentrations (40.37-73.44 and 14.33-18.90 mg g-1 fresh weight, respectively) occurred between pH 6.8 and 8 by ionic aw modification (KCl, NaCl) and with the CF + YE nitrogen profile. Germination of blastospores produced under these cultural conditions was between 62 and 89% under conditions of water-stress (0.96 aw). On the other hand, blastospores with lower amounts of erythritol and total protein content had decreased germination (8-67%). These results could have significant implications for developing a liquid fermentation medium for the production of high numbers of fungal propagules with enhanced efficacy under non-optimum environmental conditions.  相似文献

Monoerucin is a monoacylglycerol incorporating a cis monounsaturated fatty acid, 22 carbon atoms long, with the double bond at carbon number 13. The original temperature-composition phase diagram for the monoerucin/water system, constructed on the basis of ‘consistency’ and light microscopy, includes unexpectedly only the lamellar and fluid isotropic phases [Lutton, E.S., 1965. J. Am. Oil Chem. Soc. 42:1068–1070]. In separate studies using X-ray diffraction, the cubic phase [Larsson et al., 1978. J. Sci. Food Agric. 29:909–914] and the inverted hexagonal (HII) phase [Caffrey, M., 1989. Biophys. J. 95:11–21] were shown to be the dominant phases. We have resorted to the traditional isoplethal diffraction method for constructing the equilibrium phase diagram of monoerucin in water with a view to resolving the controversy regarding the thermotropic and lyotropic properties of the system. The study confirms the presence of the pure HII phase that extends from ca. 10 to 22% (w/w) water and from ca. 35 to 125°C, and its coexistence with excess water above ca. 24%(w/w) water in the same temperature range. The low temperature region of the diagram is dominated by the lamellar crystal (Lc) phase, with the existence of the lamellar liquid crystal (L) and cubic-Ia3d phases at intermediate temperatures and hydration levels. Differential scanning calorimetry and polarizing light microscopy were used to confirm the reported equilibrium phase behavior.
