Seeds of 25 plant species from calcareous fen hay meadows were exposed to different experimental conditions and their germination was characterised. Constant temperature inhibited germination especially in Cyperaceae . Both gibberellic acid and potassium nitrate failed to terminate dormancy. Increased germination rates were found in dicot species after treatment with gibberellic acid. Temperature fluctuations increased germination of Cyperaceae as well as dicotyledons. Treatment with gibberellic acid removed the chilling requirement in some of the species. Dormancy of small seeds with thin seed coats was broken by the application of gibberellic acid or fluctuating temperature; large thick-coated seeds were unaffected by gibberellic acid. No obligatory darkness requirement was found in any species; three species germinated irrespective of light treatments. All other species achieved higher percentage germination in daylight or in red (670 nm) light. Permanent darkness and far-red light (730 nm) reduced germination drastically. The results indicate that germination characteristics of the species investigated can be related to their seed bank types.  相似文献

One-month-old fruits of Acer ginnala with winged pericarp attached gave 44% germination and this was not increased by cold treatment at 4°C for 0, 10, 20, or 30 days, gibberellic acid treatment at 0, 1, 10, 100 or 1000 mg litre-1, or ethephon treatment at 0, 2, 20, 200 or 2000 mg litre-1. After 6 months of storage at 20–25 °C, germination of untreated fruits fell to 5% but could be restored to that of 1-month-old fruits by incubation at 4 °C for 30 days. After 9 months storage, no germination occurred in untreated fruits. Cold treatment (30 days at 4 °C partially restored germination (26%). Treatment with either gibberellic acid (1000 mg litre-1) and 30 days at 4 °C (40%) or ethephon (100 mg litre-] and 30 days at 4 °C improved germination (69%). The combination of all three treatments, i.e. 100 mg litre-1 gibberellic acid, 100 mg litre-1 ethephon and 30 days at 4 °C, optimised germination (86%). Thus, dormancy of A. ginnala developed during storage but could be reversed by a combination of treatment with low temperature and growth regulators. The highest germination (86%) was achieved after low temperature and growth regulator treatment of stored fruit.  相似文献

All seeds of Aeluropus lagopoides and Urochondra setulosa germinated under non-saline conditions except for Sporobolus ioclados which showed only 40 % germination. Increase in salinity substantially inhibited germination and few seeds germinated at 400 mM NaCl. Germination at 200 mM NaCl was alleviated in U. setulosa by the application of gibberellic acid and fusicoccin, in A. lagopoides by thiourea, betaine, kinetin, fusicoccin and ethephon, and in S. ioclados by gibberellin and ethephon. High salinity (400 mM NaCl) induced germination inhibition was alleviated by proline, kinetin, fusicoccin and ethephon only in A. lagopoides.  相似文献

The allelopathic potential of the dry fruits of Washingtonia filifera (L. Linden) H. Wendl. was investigated. Leachates from fruits inhibited the germination of lettuce, wheat, red cabbage and cucumber seeds. The inhibitory effect was partly neutralized by kinetin (20 mg 1−1) and gibberellic acid (50 mg 1−1). The effect of kinetin was more pronounced at 25°C than at 20°C. Substances inhibiting germination were localized in the pericarp of the fruit and were resistant to high temperature.  相似文献

The effects of 4 chemicals on the germination promotion of stratified and unstratified seeds of loblolly pine (Pinus taeda) and baldcypress (Taxodium distichum) were studied. The chemicals used were gibberellic acid, kinetin, potassium nitrate and thiourea, each at 3 different concentrations. Stratification promoted the germination of both seed species. Certain concentrations of gibberellic acid, potassium nitrate and thiourea improved the germination of unstratified loblolly pine and baldcypress seeds while kinetin had no stimulatory effect. All 4 chemicals at specific concentrations promoted the germination of loblolly pine seeds stratified for a short period of time. Considering both speed and completeness of germination, best results were obtained when 21-day stratified seeds were treated with either gibberellic acid (100 mg/1) or kinetin (10 mg/1). In baldcypress, on the other hand, none of these chemicals had any stimulatory effect on the germination of stratified seeds. Germination of both species of seeds was either partially or completely inhibited by the highest concentration of thiourea (30,000 mg/1) used.  相似文献

Red light and gibberellic acid were about equally effective in promoting germination of Grand Rapids lettuce (Lactuca sativa L.) seeds. With initial far red light treatment more than 80% remained dormant in subsequent dark storage. After 2 days of dark storage, red light effectively promoted germination, while gibberellic acid action was weak. With between 2 and 10 days of dark storage, gibberellic acid had little effect, while promotion by red light decreased slowly and finally disappeared. After 10 days of dark storage, both gibberellic acid and red light were required for germination. The dark storage treatment interferes with phytochrome-independent germination processes and cannot be overcome by added gibberellic acid. However, storage may also decrease the effectiveness of endogenous gibberellins. Phytochrome-dependent germination seems to require only low levels of endogenous gibberellin activity or the addition of gibberellic acid. Gibberellins and red light appear to act on germination by regulation of sequential sites of a branched-looped pathway.  相似文献

The spores of the fern Anemia phyllitidis contain abundant quantities of lipid as reserve material. Germination of these spores can be induced either by red light or, even in the dark, by gibberellic acid. The effects of both these factors on lipid degradation, lipase and isocitrate lyase activities, and on the fatty acid composition have been studied in the course of the germination process. During germination in darkness with gibberellic acid, the fatty acid composition remained similar to that in the ungerminated spore. In contrast, when spores were germinated in red light, α-linolenic acid was synthesized. Little activity of lipase and isocitrate lyase could be detected in the dry spore. Red light or gibberellic acid affected a dramatic increase of the activities of these enzymes. Lipid breakdown and lipase activity were more active in red light, however. Permanent stimuli were necessary for growth and complete lipid degradation. Induction of germination simultaneously with both factors revealed an additivity of the effects of red light and gibberellic acid.  相似文献

Seedlings of the self-fertilizing species Lactuca scariola L. grown continuously in 8 h days did not flower even one year from sowing. Seedlings grown in 16 h days uatil flower buds appeared 96 days after germination were either transferred to 8 h days or treated weekly with gibberellic acid (GA3), abscisic acid (ABA) or chlormequat (CCC) and retained, together with untreated control plants, in 16 h days. Each growth regulator caused characteristic morphological changes in the treated plants. All these plants flowered and produced seeds but the seeds showed distinct differences in weight, in their time to germination and in the seedlings which they produced. Germination and seedling characters depended on the light regime during germination as well as on the chemical applied to the parent plant and the rate of application. The parental treatment also affected the shape and size of the seedlings on a given day after germination, and certain treatments of the parent plant (transfer from long to short days and treatment with CCC in long days) advanced the flowering date of the seedlings. The gibberellin level in the seeds was raised, in increasing order, by treatment of the parent plant with 100 mg/1 GA3, transfer from long to short days, 10 mg/1 GA3, and 5000 mg/l CCC. It is suggested that the effect of day length on plant performance is mediated by the level of growth regelating substances within the plant and that the behaviour of seeds can be modified by the parental environment via the accumulation of different levels of certain growth factors in the seeds. A rise of one growth substance in the parent plant can result in the accumulation of a different one in the seeds.  相似文献

The ripe seeds of Eranthis hiemalis (L.) Salisb., the winter aconite, contain undeveloped embryos. At 20–25°C the embryos grow only little, and the seeds do not germinate. Rapid embryo development starts if the seeds, after 3 weeks of “after-ripening” at 20–25°C, are placed at low temperature, 3–4°C; germination then takes place after 2–3 months, Embryo development without germination occurs when the seeds are placed in gibberellic acid solutions at 20–25°C. Embryo development is inhibited at low temperature by the specific inhibitor of gibberellin biosynthesis, 2-chlorethyl cholin chloride, but is restored by the simultaneous addition of gibberellic acid. It is suggested that one early effect of the cold is to bring about a synthesis of gibberellin.  相似文献

The occurrence of various species of Brassicaceae with indehiscent fruits in the cold deserts of NW China suggests that there are adaptive advantages of this trait. We hypothesized that the pericarp of the single-seeded silicles of Isatis violascens restricts embryo expansion and thus prevents germination for 1 or more years. Thus, our aim was to investigate the role of the pericarp in seed dormancy and germination of this species. The effects of afterripening, treatment with gibberellic acid (GA3) and cold stratification on seed dormancy-break were tested using intact silicles and isolated seeds, and germination phenology was monitored in an experimental garden. The pericarp has a role in mechanically inhibiting germination of fresh seeds and promotes germination of nondormant seeds, but it does not facilitate formation of a persistent seed bank. Seeds in silicles in watered soil began to germinate earlier in autumn and germinated to higher percentages than isolated seeds. Sixty-two percent of seeds in the buried silicles germinated by the end of the first spring, and only 3% remained nongerminated and viable. Twenty to twenty-five percent of the seeds have nondeep physiological dormancy (PD) and 75–80% intermediate PD. Seeds with nondeep PD afterripen in summer and germinate inside the silicles in autumn if the soil is moist. Afterripening during summer significantly decreased the amount of cold stratification required to break intermediate PD. The presence of both nondeep and intermediate PD in the seed cohort may be a bet-hedging strategy.  相似文献

The dormancy breaking effect of sodium azide was studied in seeds of several genetically pure lines of Avena fatua L. isolated from field populations. Sodium azide (0.8 and 1 m M ) induced germination in several dormant lines (characterized by long term dormancy) after two weeks of treatment. By about five weeks, germination was nearly complete in azide treated seeds as compared to little or no germination in controls. (2-chloroethyl) trimethylammonium chloride (an inhibitor of gibberellin biosynthesis) completely inhibited the azide effect suggesting that stimulation of germination by azide requires gibberellin biosynthesis. Azide was very effective in breaking dormancy in lines AN-51, AN-86, AN-127 and AN-265, but failed to induce germination in Montana 73. In this line there was a synergism between azide and gibberellic acid in promotion of germination. Thus, at least two metabolic blocks are involved in the stimulation of germination in this line. Salicylhydroxamic acid (an inhibitor of alternative respiration) at 3 m M completely inhibited the germination induced by 1 m M azide. At this concentration, salicylhydroxamic acid did not inhibit germination in 1) genetically nondormant seeds (line SH-430), 2) afterripened seeds of a dormant line (AN-51), and 3) gibberellic acid-treated dormant seeds. These findings suggest that salicylhydroxamic acid-sensitive process(es), presumably alternative respiration, is necessary for the stimulation of germination in the presence of azide, but not in the germination of genetically nondormant, gibberellic acid-treated dormant, or afterripened seeds.  相似文献

Morphactin-butylester (a flourene-9-carboxylic acid derivative) inhibited seed germination of two strains of lettuce. Morphactin induced inhibition of germination could be partially or wholly reversed by simultaneous addition of gibberellic acid. However, gibberellic acid played very little part in reversing the inhibitory effect of morphactin on seedling growth. It is concluded that gibberellin can not reverse all the growth effects induced by morphactin.  相似文献

Many seeds are dormant when shed from the mother plant. This unique characteristic of plants poses challenges in conservation and many different treatments have traditionally been used to break dormancy. When germination only occurs under certain circumstances, recruitment may be insufficient and the viability of plant populations may be threatened. A marked dormancy was previously identified in the Iberian critically endangered species Pseudomisopates rivas-martinezii. The present study aimed to determine the magnitude of dormancy by estimating seed viability under different germination treatments: heat, ash, stratification, gibberellin addition, and darkness. Our results indicate that there were significant differences in viability across plant populations and treatments. Maximum seed germination was obtained under ash addition, although cold stratification alone gave a considerable enhancement. However, gibberellic acid did not improve germination and darkness had a diminishing effect. In conclusion, a cold period is required for the species to germinate, coupled with the effects of fire: (1) ash, (2) vegetation clearings, and (3) resprouting, which are major factors triggering seed germination. Although these conditions are widely found in the field, we hypothesize that a more dramatic situation may have tackled this species before the increase of human-mediated fires in the past millennia.  相似文献

Luzula spicata L. seeds are completely dormant at maturity. A germination inhibitor is present at the micropylar end. Normally, the only effective means of eliciting germination is a precise scarification of the micropylar end which inactivates the inhibitor. Exogenous application of gibberellic acid, kinetin, KNO3, and thiourea have no affect on the dormancy of unscarified seeds. Scarification of the hilar end of the seed does not elicit germination, but when gibberellic acid is applied to the hilar scarified seeds moderate germination results. Presumably, these seeds are dormant due to a deficit of endogenous gibberellin; a condition which can be overcome by the application of gibberellic acid to seeds scarified at a site in itself ineffective in producing germination. Apparently the gibberellic acid serves to initiate amylase activity in the endosperm, overcoming the inhibitor block. Luzula spicata seed dormancy is apparently unique in that a germination inhibitor is operative in conjunction with the commonly recognized gibberellin-amylase mechanism.  相似文献

Zeyheria montana Mart. has become endangered, primarily because of deforestation of its habitat, its use as a medicinal plant extract, and the strong endemism of the species. In this study, an efficient protocol was established for the micropropagation and conservation of Z. montana germplasm using isolated mature zygotic embryos as explants. Embryos germinated in vitro 4 d after isolation and inoculation on modified Murashige and Skoog (MS) medium containing 2.0 mg/l gibberellic acid (GA3). The addition of GA3 also improved the germination index and accelerated the process of germination. Nodal segments from seedlings were placed on modified ¼-strength MS medium containing 0.1 mg/l 6-benzyladenine and 0.5 mg/l GA3. Nodal segments produced 7.3 shoots per explant within 60 d. Following transfer of shoots to a medium containing 1.5 mg/l indole-3-butyric acid, roots formed. All plantlets obtained were successfully acclimatized under greenhouse conditions, and approximately 68.5 acclimatized plants could be obtained per seed each year. This protocol provides a method to preserve this rare and endangered medicinal plant.  相似文献

Seed germination behavior in two types of dimorphic fruits inSalsola komarovii Iljin (Chenopodiaceae) was studied. The germination percentage was much higher in the long-winged type than in the short-winged type when the fruits were tested for germinability. If lignified perianths were removed from the fruits, seeds of both types germinated at higher percentages and the difference in the germination percentage between two types decreased. The extract of the perianths inhibited the seed germination. Abscisic acid was detected in the extract of the perianths by enzyme-linked immunosorbent assay and its level was higher in the short-winged type than in the long-winged type. Germinability in both types was lost one year after harvest when the fruits were stored at room temperature and the germination was tested with fruits. When the seeds were tested, however, the short-winged type germinated even two years after the harvest; the germinability of the short-winged type lasted one year longer than that of the long-winged type. Light irradiation was not required for the seed germination in either type regardless of presence or absence of the perianths. No clear difference between the two types was observed either in optimal temperature for the germination or in strength of the resistance to salinity.  相似文献

Extracts from the plant Lycopus europaeus L. are traditionally used in mild forms of hyperthyroidism. High doses caused a reduction of TSH or thyroid hormone levels in animal experiments, whereas in hyperthyroid patients treated with low doses of Lycopus an improvement of cardiac symptoms was reported without major changes in TSH or thyroid hormone concentrations. Lycopus extract was tested in thyroxine treated hyperthyroid rats (0.7 mg/kg BW i.p.). Co-treatment with an hydroethanolic extract from L. europaeus L. started one week later than T4-application and lasted 5.5 weeks. As reference substance atenolol was used. The raised body temperature was reduced very effectively even by the low dose of the plant extract, whereas the reduced gain of body weight and the increased food intake remained unaffected by any treatment. No significant changes of thyroid hormone concentrations or TSH levels were observed. Lycopus extract and atenolol reduced the increased heart rate and blood pressure. The cardiac hypertrophy was alleviated significantly by both treatment regimes. beta-Adrenoceptor density in heart tissue was significantly reduced by the Lycopus extract or the beta-blocking agent showing an almost equal efficacy. Although the mode of action remains unclear, these organo-specific anti-T4-effects seem to be of practical interest, for example in patients with latent hyperthyroidism.  相似文献

Dispersal limitation is often involved when the species composition of a dry abandoned grassland shows a slow response to resumed regular mowing. A seed-addition experiment, using 32 species which do not belong to the local species pool, was performed on Monte San Giorgio (southern Switzerland) to test whether the low recruitment success was due to dispersal limitation or due to unfavourable microsite conditions. In October 1997, 20 species were individually sown in six 3 × 4 m blocks of a 2 × 2 factorial “partial” split-plot design with treatments of abandonment vs. mowing and undisturbed vs. root-removed soil, this last being applied in small naturally-degradable pots. Moreover, 12 species were sown only in the treatments on undisturbed soil. Seedlings of sown and spontaneously germinating seeds were observed on 16 occasions over one 12-month period. Seeds of 31 out of the 32 species germinated. Twenty-four species showed germination rates higher than 5% and different seasonal germination patterns. Established vegetation, especially the tussocks ofMolinia arundinacea, reduced the quality of microsites for germination. Whereas a few species germinated better under the litter ofMolinia arundinacea, many more germinated better under the more variable microsite conditions of a mown grassland.  相似文献

Summary One of the foremost technical issues addressed in reintroduction and restoration projects is the feasibility of establishing living plants. To advance the recovery process, the germination requirements of 201 threatened Western Australian seed‐bearing taxa representing a range of life forms, families and ecophysiological characteristics were studied. Procedures used to stimulate germination in otherwise dormant seed involved pretreatment using thermal shock, scarification, seed coat removal, soaking in an aqueous smoke solution and/or additions of the growth hormone gibberellic acid (GA3). Sixty‐one taxa germinated under the basic trial conditions of light (12‐ h photoperiod), temperature (constant 15°C) and moisture, without additional pretreatments. These taxa were generally those with canopy‐stored seeds in the families Proteaceae and Casuarinaceae, and small‐seeded taxa in Myrtaceae. Taxa with soil‐stored seeds required single or multiple cues to stimulate germination. Seeds in the families Fabaceae and Mimosaceae were dependent on cracking of the seed coat, mechanically through nicking of the testa or through thermal shock, as were several non‐leguminous species of the Sterculiaceae and Liliaceae. Complete or partial removal of seed coats, in conjunction with GA3 enhanced germination percentage in some taxa of the Myoporaceae, Lamiaceae and Myrtaceae. Application of GA3 also enhanced germination percentage in members of the Epacridaceae. Several taxa previously stimulated by aqueous smoke solutions were equally responsive to additions of GA3 after complete seed coat removal. In general, species with seed weights greater than 10 mg germinated better under a range of conditions than those with lighter seeds. There was no difference in germinability between resprouter and seeder species, and no obvious relationship between seed weight and germination rate. In the light of previous studies these results indicate that the relationship between germination requirements and ecophysiological characteristics is similar for both threatened and common species. These data will enable better prediction of likely dormancy breaking cues for other related species and will greatly assist the process of recovery and restoration work for mining operations and community bushland regeneration as well as single species reintroductions.  相似文献
