Summary The ratio of deuterium to hydrogen (expressed as D) in hydrogen released as water during the combustion of dried plant material was examined. The D value (metabolic hydrogen) determined on plant materials grown under controlled conditions is correlated with pathways of photosynthetic carbon metabolism. C3 plants show mean D values of-132 for shoots and -117 for roots; C4 plants show mean D values of -91 for shoots and-77 for roots and CAM plants a D value of-75 for roots and shoots. The difference between the D value of shoot material from C3 and C4 plants was confirmed in species growing under a range of glasshouse conditions. This difference in D value between C3 and C4 species does not appear to be due to differences in the D value (tissue water) in the plants as a result of physical fractionation of hydrogen isotopes during transpiration. In C3 and C4 plants the hydrogen isotope discrimination is in the same direction as the carbon isotope discrimination and factors contributing to the difference in D values are discussed. In CAM plants grown in the laboratory or collected from the field D values range from-75 to +50 and are correlated with 13C values. When deprived of water, the D value (metabolic hydrogen) in both soluble and insoluble material in leaves of Kalanchoe daigremontiana Hamet et Perr., becomes less negative. These changes may reflect the deuterium enrichment of tissue water during transpiration, or in field conditions, may reflect the different D value of available water in areas of increasing aridity. Whatever the origin of the variable D value in CAM plants, this parameter may be a useful index of the water relations of these plants under natural conditions.  相似文献

Chimeric genes composed of the -phaseolin promoter, an -zein coding sequence and its modified versions containing lysine codons, and a -zein polyadenylation signal were inserted into the genome of tobacco by Agrobacterium-mediated transformation. -Zein mRNA levels in the transgenic tobacco seeds 20 days after self-pollination varied between 1.0% and 2.5% of the total mRNA population. At 25 days after pollination the 19 kDa -zein was immunologically detected with a polyclonal antiserum in protein extracts from the seeds of transgenic plants. The transgenic plant with the highest level of zein gene expression had an -zein content that was approximately 0.003% of the total seed protein. The amount of -zein in other transgenic plants varied between 1 × 10–4% and 1 × 10–5% of the total seed protein. The differences in the amounts of mRNA and protein did not correlate with the lysine substitutions introduced into the -zein protein. Polysomes translating -zein mRNA isolated from tobacco seeds contained fewer ribosomes than those from maize endosperm, but this did not appear to be the cause of the inefficient protein synthesis. In vivo labelling and immunoprecipitation indicated that newly synthesized -zein was degraded in tobacco seeds with a half-life of less than 1 hour.  相似文献

-Crystallin is the major and most abundant lens protein present in the eye lens of lower vertebrates such as amphibian and piscine species. To facilitate structural characterization of-crystallins isolated from the lens of the bullfrog (Rana catesbeiana), a cDNA mixture was synthesized from the poly(A)+mRNA isolated from fresh eye lenses. cDNA encoding-crystallin was then amplified using polymerase chain reaction (PCR) based on two primers designed according to the relatively conserved N- and C-terminal sequences of known-crystallins from teleostean fishes. PCR-amplified product corresponding to-crystallin isoforms was obtained, which was then subcloned in pUC18 vector and transformed intoEscherichia coli strain JM109. Plasmids containing amplified-crystallin cDNAs were purified and prepared for nucleotide sequencing by the dideoxynucleotide chain-termination method. Sequencing several clones containing DNA inserts of about 0.54 kb revealed the presence of two isoforms with an open reading frame of 534 base pairs, covering two-crystallins each with a deduced protein sequence of 177 amino acids including the translation-initiating methionine. These-crystallins of pI 6.364 and 6.366 contain a low-methionine content of 2.81%, in contrast to 11–16% obtained for those-crystallins with high-methionine content from most teleostean lenses. Pairwise sequence comparison of bullfrog-crystallins with those published sequences of-crystallins from carp, shark,Xenopus and anotherRana frog, bovine, and human lenses indicates that there is only 46–63% sequence similarity among these species, revealing that amphibians possess a very complex and heterogeneous group of-crystallins even from closely related species ofRana frogs. The sequence analysis and comparison of various isoforms of the frog-crystallin family provide a firm basis for identifying these lens proteins as members of a multigene family more complex than that reported for mammalian-crystallins.  相似文献

Negative-ion fast atom bombardment tandem mass spectrometry has been used in the characterization of non-, mono-, di- and trisulfated disaccharides from heparin and heparan sulfate. The positional isomers of the sulfate group of monosulfated disaccharides were distinguished from each other by negative-ion fast atom bombardment tandem mass spectra, which provide an easy way of identifying the positional isomers. Embryonic, larval, and juvenile development of an Indian cyprinid fish, Chela dadiburjori, is described from laboratory-reared specimens. The eggs, measuring 0.7–0.9mm in diameter, were demersal, almost spherical in shape, transparent and unpigmented, with a pale yellow yolk and no oil globule. Hatching occurred 50–61h after fertilization at ca. 27°C. The newly hatched larvae, measuring 2.4–2.6mm in body length (BL), had melanophores on the body with 14–16+14–17=29–31 myomeres. Two dark transverse bands on the ventral body surface and one melanophore on the lower margin of the eye in newly hatched larvae were diagnostic. Additionally, a cement organ for adhering to objects was present on the forehead of yolk sac larvae <3.1mm BL. The yolk was completely absorbed at 3.5mm BL. Notochord flexion was initiated at 5.0mm BL and finished at 6.0mm BL. Aggregate numbers of all fin rays were completed at 9.2mm BL. Squamation was initiated on the caudal peduncle at 8.0mm BL and completed at 10mm BL. The eggs of C. dadiburjori resembled those of the closely related species Devario malabaricus and Danio rerio. The larvae and juveniles of C. dadiburjori were also similar to those of the latter species in general morphology, especially the presence of body melanophores in newly hatched individuals and a distinctive lateral streak on the head during the period from yolk sac to postflexion larvae. However, early yolk sac larvae of C. dadiburjori were more similar to those of Devario malabaricus than Danio rerio in having a cement organ on the forehead. Larvae and juveniles of C. dadiburjori differed from those of the latter two species in pigmentation on the ventral body surface at hatching and around the mouth during the period from preflexion to early postflexion larvae and in having a dark lateral streak or band on the body in postflexion larvae and juveniles.  相似文献

Zusammenfassung Der Verlauf des Nahrungsflusses im Darmtrakt der Kleinzikade Euscelidius variegatus wird nach Verfütterung von farbstoffhaltiger Nährlösung ermittelt. Es wird der Beweis erbracht, daß die aufgenommene Nahrungsmenge in der Filterkammer geteilt wird und die beiden Anteile den Darmtrakt auf zwei verschiedenen Wegen in Richtung Rektalblase passieren. Ein Anteil der aufgenommenen Nährlösung wird über einen Kurzschlußweg in der Filterkammer sowohl über den Filterkammerdarm als auch über die Kryptonephridien direkt in den Enddarm gepumpt, während die in der Magentasche der Filterkammer verbleibenden Nahrungsanteile über einen langen Verdauungsweg zum After gelangen. Hierbei wird der Magentascheninhalt in den Magen gedrückt. Von dort aus passiert er den Mitteldarm und erreicht über den Enddarm den After. Der Kurzschlußweg und der Verdauungsweg können gleichzeitig benutzt werden. Der Kurzschlußweg wird von der Nahrung jedoch in viel kürzerer Zeit durchströmt als der längere Verdauungsweg.

Summary The leafhopper Euscelidius variegatus is fed with synthetic food, coloured with 1% Azorubin-S. Its flow in the alimentary tract has been studied. It has been found that the sucked-in food is divided into two parts in the filter chamber, each taking different way in the alimentary tract for its flow. One part of the food is pumped into the hindgut via the short circuit way going through the filter chamber once over the Filterkammerdarm and also over the kryptonephries. That part of the food, which remains in the pocket of the filter chamber takes the long digestion way to the anus over stomach, midgut and hindgut. Both the ways could be used at the same time. But the food takes much shorter time for its passage through the short circuit way as compared to the time needed for the long digestion way.

Spin-State-Selective Excitation (S3E), which forexample selectively excites amide proton resonances corresponding toexclusively either the or the spin state of the covalentlybound 15N atom is employed for E.COSY-type extraction ofheteronuclear J coupling constants. Instead of having one spectrum with twopeaks (corresponding to the or spin state of15N), S3E generates two spectra, each with onlyone peak for each 15N nucleus. These two spectra are generatedfrom the same data set, so that there is no reduction in sensitivitycompared to conventional 1JNH-resolved methods.Another interesting feature in comparison with conventional methods is that1JNH can be suppressed during the evolutionperiod, meaning that no heteronuclear multiplet structure is visible in the1 frequency dimension. Summary The impact of extraneously furnished D(+)-glucose and D(–)-fructose, on the synthesis of new oligosaccharides from sucrose solution byC. lycopersici andF. semitectum, is presented in the present paper. The results are significant, in that they show, that the behaviour of the organisms is determined by the shell-wrapped molecular memory enshrined in the nucleus of the cell; and the signal of optical rotatory power is interpreted by the operator-structural genes determining the configuration of the enzyme-fructosidase.  相似文献

Type E botulinum neurotoxin is produced byClostridium botulinum along with a neurotoxin binding protein which helps protect the neurotoxin from adversepH, temperature, and proteolytic conditions. The neurotoxin binding protein has been purified as a 118-kDa protein. Secondary structure content of the neurotoxin binding protein as revealed by far-UV circular dichroism spectroscopy was 19% -helix, 50%-sheets, 28% random coils, and 3%-turns. This compared to 22% -helix, 44%-sheets, 34% random coils, and no-turns of the type E botulinum neurotoxin. The complex of the two proteins revealed 25%-helix, 45%-sheets, 27% random coils, and 3%-turns, suggesting a significant alteration at least in the-helical folding of the two proteins upon their interaction. Tyrosine topography is altered considerably (28%) when the neurotoxin and its binding protein are separated, indicating strong interaction between the two proteins. Gel filtration results suggested that type E neurotoxin binding protein clearly complexes with type E neurotoxin. The interaction is favored at lowpH as indicated by an initial binding rate of 8.4 min–1 atpH 5.7 compared to 4.0 min–1 atpH 7.5 as determined using a fiber optic-based biosensor. The neurotoxin and its binding protein apparently are of equivalent antigenicity, as both reacted equally on enzyme-linked immunosorbent assay to polyclonal antibodies raised against the toxoid of their complex.  相似文献

The conformational properties of the oligosaccharide chain of GM1 ganglioside containingN-glycolyl-neuraminic acid, -Gal-(1-3)--GalNAc-(1-4)-[-Neu5Gc-(2-3)]--Gal-(1-4)--Glc-(1-1)-Cer, were studied through NMR nuclear Overhauser effect investigations on the monomeric ganglioside in dimethylsulfoxide, and on mixed micelles of ganglioside and dodecylphosphocholine in water. Several interresidual contacts for the trisaccharide core--GalNAc-(1-4)-[-Neu5Gc-(2-3)]--Gal-were found to fix the relative orientitation of the three saccharides, while the glycosidic linkage of the terminal -Gal-was found to be quite mobile as the -Gal-(1-3)--GalNAc-disaccharide exists in different conformations. The impacts of two species of ants (Lasius niger and Pristomyrmex pungens) attending the cotton aphid (Aphis gossypii) on the number of emerging adults of the aphids primary parasitoid (Lysiphlebus japonicus) and hyperparasitoids were assessed experimentally at a no-pesticide mandarin grove. L.japonicus females were observed foraging frequently in aphid colonies attended by either ant species, with more females in P.pungens-attended than in L.niger-attended colonies, but rarely in aphid colonies where ants were artificially excluded. Females were less often attacked by P.pungens workers than by L.niger workers, so that they stayed longer and oviposited into more aphids in P.pungens-attended than in L.niger-attended colonies. Attendance by each ant species reduced predator numbers in aphid colonies, compared to colonies where ants were absent, although P.pungens was slightly less effective in repelling predators. Therefore, both ant species incidentally protected parasitized aphids from predators. Consequently, L.japonicus-mummies were abundant in colonies attended by either ant species, with more mummies in P.pungens-attended colonies, and were scarce in ant-excluded colonies. The hyperparasitism and predation on L.japonicus larvae within mummies occurred more frequently in P.pungens-attended than in L.niger-attended colonies, but mummy predation rate was only 20% in the former. As a result, the number of emerging L.japonicus adults did not differ significantly between aphid colonies attended by the two ant species, but significantly more hyperparasitoids emerged in colonies attended by the relatively less aggressive P.pungens than in colonies attended by L.niger.  相似文献

The reproductive behavior of horned rainbow scarab beetles,Phanaeus difformis, was studied to determine the influence of morphological traits on intersexual and intrasexual interactions.Phanaeus difformis is a sexually dimorphic dung beetle in which males possess much larger horns than females, and males can be grouped into major and minor male morph categories based on horn size. Male-female pairs cooperated in nest construction and provisioning. In the laboratory, males of both morphs assisted females and were equally successful at copulating. However, in the field larger individuals had a pairing advantage due to greater success in intrasexual competition. Some males used an alternative mating tactic which involved sneaking copulations with paired females. In most cases the sneak male was smaller than the paired male.  相似文献

Regulation of β-xylosidase formation by xylose in Trichoderma reesei   总被引:1,自引:0,他引:1  
The soft-rot fungus Trichoderma reesei forms -xylosidase (EC 3.2.1.37) activity during cultivation on xylan and xylose, but not on glucose. When mycelia precultivated on glycerol were washed and transferred to fresh medium without a carbon and nitrogen source, -xylosidase formation was induced by xylan, xylobiose and xylose. A supply of 4 mm xylose and a pH of 2.5 provided optimal conditions for induction. -Xylosidase accounted for the major portion of total extracellular protein under these conditions, and could be purified to physical homogeneity by a single anion exchange chromatography step. A recombinant strain of T. reesei that carries multiple copies of the homologous xylanase II-encoding gene has a six-fold increased xylanase activity, but forms comparable -xylosidase activities. This shows that the rate of xylan hydrolysis has no effect on the induction of -xylosidase. Methyl--d-xyloside inhibited -xylosidase competitively and was a weak -xylosidase inducer. The induction by xylobiose and xylan was strongly enhanced by the simultaneous addition of methyl--d-xylosidese and xylan or xylobiose. The results suggest that a slow supply of xylose is a trigger for -xylosidase induction.  相似文献

The seeds of 19 sunflower species were compared on the basis of their protein contents and the relative proportions of their protein fractions. The globulin content varied from 50% to about 70% and the albumin content from 18% to 35% according to the species. The level of intermediateMr polypeptides showed a great variability (9.6 to 24.3%). Comparative studies onMr polymorphism were carried out by means of sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE) of non reduced and/or reduced samples using both mono- and bidimensional procedures. Polypeptide constituents of helianthinin were compared including both number and molecular size (cultivatedH. annuus was used as a standard). Studies focused on differences observed between the major two (Mr 38 000), (Mr 32 000) and (Mr 25 500), (21 000) polypeptides families constituting the main A, B, and C subunits. and polypeptides analyses permit to discriminate easilyH. petiolaris from the other species. Charge polymorphism was studied using isoelectric focusing (IEF) and IEF-PAGE in mono and bidimensional procedures in the presence or absence of 2-mercaptoethanol (2-ME). Only a specific 4 polypeptide enables an easy discrimination betweenH. petiolaris and all the other species. Detailed nomenclature of the , and , polypeptides constituting the different helianthinin globulin subunits is given via the results of pI andMr analyses. Monodimensional IEF patterns of the more basic albumins (pI > 8.0) appear to provide a more valuable approach to identifying specific protein markers.  相似文献

Summary An electroporation method using a Bio-Rad Gene Pulser has been optimized for introducing heterologous DNA into Kluyveromyces lactis yeasts. The plasmid pCR1, derived from a native Kluyveromyces plasmid, was used to transform K. lactis. This plasmid produces a wheat -amylase and contains both the biosynthetic marker URAA and G418 resistance genes. Transformation was optimal at 4500 V/cm, 25 F, and with 0.2 g plasmid DNA. Transformation efficiencies in the range 104–105 transformants/107 cells/g DNA were obtained.  相似文献

Summary The -galactosidase gene from Bacillus subtilis KL88 was cloned into Escherichia coli and the gene product characterized for its potential use in the dairy industry. The two recombinant plasmids that we obtanied encoded a -galactosidase with the same catalytic and thermal characteristics as the native -galactosidase from B. subtilis. The recombinant -galactosidases exhibited high activity at low temperature (10°C), with maximum activity at 50°C and an optimum pH of 6.0. Its molecular weight was estimated to be 90 Kd. The restriction maps of the recombinant plasmids were constructed. The -galactosidase gene was located in a 2.3 Kb fragment.  相似文献

Summary The pathogenFusarium solani sensuSnyder &Hansen, as identified byW. L. Gordon was recorded in the present work as a new species causing cotton wilt in Egypt. Cotton dust in varying concentrations does not significally alter the normal infection of both Menoufi and Giza 26 cotton varieties towardsFusarium. Similarly the potency ofFusarium filtrate to induce wilting did not appreciably change with previous treatment of cotton plants with cotton dust. On the other hand leaves treated with 2,4-D showed the maximal water loss. Percentage infected Giza 26 cotton seedlings were found to be comparatively less in soil infected with 2,4-D treatedFusarium mycelium than in that infected with untreated mycelium.  相似文献

Summary In this paper, the results of the preceding electrophysiological study of sodium-alanine cotransport in pancreatic acinar cells are compared with kinetic models. Two different types of transport mechanisms are considered. In the simultaneous mechanism the cotransporterC forms a ternary complexNCS with Na+ and the substrateS; coupled transport of Na+ andS involves a conformational transition between statesNCS andNCS with inward- and outward-facing binding sites. In the consecutive (or ping-pong) mechanism, formation of a ternary complex is not required; coupled transport occurs by an alternating sequence of association-dissociation steps and conformational transitions. It is shown that the experimentally observed alanine- and sodium-concentration dependence of transport rates is consistent with the predictions of the simultaneous model, but incompatible with the consecutive mechanism. Assuming that the association-dissociation reactions are not rate-limiting, a number of kinetic parameters of the simultaneous model can be estimated from the experimental results. The equilibrium dissociation constants of Na+ and alanine at the extracellular side are determined to beK N <-64mm andK S <-18mm. Furthermore, the ratioK N /K N S of the dissociation constants of Na+ from the binary (NC) and the ternary complex (NCS) at the extracellular side is estimated to be <-6. This indicates that the binding sequence of Na+ andS to the transporter is not ordered. The current-voltage behavior of the transporter is analyzed in terms of charge translocations associated with the single-reaction steps. The observed voltage-dependence of the half-saturation concentration of sodium is consistent with the assumption that a Na+ ion that migrates from the extracellular medium to the binding site has to traverse part of the transmembrane voltage.  相似文献

Summary During high salinity stress, -alanine accumulates to high levels in the sea anemone,Bunodosoma cavernata. Following a salinity increase from 26 to 40 -alanine increased 28-fold from 1.5 to 41.9 moles/g dry weight. Both whole animal studies and experiments with cell free homogenates indicate that under high salinity conditions an increase in the rate of -alanine synthesis from aspartic acid as well as a decrease in the rate of -alanine oxidation are responsible for the observed accumulation of -alanine. The rate of aspartic acid decarboxylation to -alanine is about 3 times greater in anemones acclimated to 40 than for those in normal salinity water (26). -alanine oxidation to CO2 and acetyl-CoA proceeds 2.5 to 3 times slower in high salinity adaptedB. cavernata than in those acclimated to normal salinity.
