Sheath rot disease of rice caused by Sarocladium oryzae (Sawada) (=Acrocylindrium oryzae, Sawada) has become an important production constraint in all rice-growing countries. Pathogenicity, phytotoxic metabolites, and random amplified polymorphic DNA (RAPD) markers were used to assess the level of genetic variability of S. oryzae derived from rice cultivars, CR1018, IR36, and IR50, of different locations in North East and South India. Variability in pathogenicity, phytotoxic metabolite production, and DNA polymorphisms was detected among S. oryzae isolates. Results indicated that S. oryzae isolates produced both cerulenin and helvolic acid at concentrations 0.3–0.62 and 0.9–4.8 μg mL−1 of culture filtrate, respectively. Isolates that produce higher concentration of helvolic acid induced a high percent incidence of sheath rot disease. Oligonucleotide primers, GF and MR, generated either a simple (up to 2 bands) or complex (up to 6 bands) RAPD pattern. According to their level of similarity, S. oryzae isolates from North East and South India were grouped separately into two major clusters and 13 genotypes. Molecular- and pathogenicity-based classifications were not correlated, but a high level of genetic variability within S. oryzae isolates was identified. The molecular variability of S. oryzae isolates will be an important consideration in breeding programs to develop durable resistance for sheath rot disease.  相似文献

Summary Various methods of evaluating phenotypic stability have been proposed; however, no single method can adequately describe cultivar performance. The objectives of this study were to integrate a number of methods of evaluating stability and to use this approach for cultivar selection. These objectives were considered in the context of the broad-based oilseed rape cultivar (Brassica napus spp. oleifera) evaluation system currently used in western Canada. Regression analysis was used to assess cultivar response to environments. Cluster analysis was used to assemble cultivars into groups with similar regression coefficients (b i ) and mean yield. Three parametric stability parameters, years within locations mean square (MS; Y/L), Shukla's stability variance ( i 2 ), and Francis and Kannenberg's coefficient of variability (CV i ), were compared to determine which method would be most suitable for selection of oilseed rape cultivars from within clustered groups. Yield data from three cultivars and six breeding lines that had been tested for 2 years at 26 locations in the Western Canola Cooperative Test A were used for all calculations. The cluster analysis was successful in identifying commercially acceptable breeding lines. The parameter MS i Y/L was considered to be more appropriate than either CV i or i 2 , because it measured only the unpredictable portion of the genotype x environment interaction and was independent of the other cultivars in the test. A pot experiment was conducted to investigate CH4 emissions from a sandy paddy soil as influenced by rice cultivars and atmospheric CO2 elevation. The experiment with two CO2 levels, 370 μL L−1 (ambient) and 570 μL L−1 (elevated), was performed in a climatron, located at the National Institute for Agro‐Environmental Sciences, Tsukuba, Japan. Four rice cultivars were tested in this experiment, including IR65598, IR72, Dular and Koshihikari. Tiller number, root length and grain yield were clearly larger under elevated CO2 than under ambient CO2. IR72 and Dular showed significantly higher tiller number, root length and grain yield than Koshihikari and IR65598. Average daily CH4 fluxes under elevated CO2 were significantly larger by 10.9–23.8% than those under ambient CO2, and varied with the cultivars in the sequence Dular ≧ IR72>IR65598 ≧ Koshihikari. Dissolved organic C (DOC) content in the soil was obviously higher under elevated CO2 than under ambient CO2 and differed among the cultivars, in the sequence IR72>Dular>Koshihikari>IR65598. The differences in average daily CH4 fluxes between CO2 levels and among the cultivars were related to different root exudation as DOC content, root length and tiller number. This study indicated that Koshihikari should be a potential cultivar for mitigating CH4 emission and simultaneously keeping stable grain yield, because this cultivar emitted lowest CH4 emission and produced medium grain yield.  相似文献

Frequent assessment of pathogen diversity is one of the most important criteria in designing disease management programmes. A study on diversity of field isolates of Rhizoctonia solani from sheath blight-infected rice fields of south India has been carried out. A total of 236 R. solani isolates were obtained from 45 locations in the surveyed area. Sclerotial features such as colour, size and shape and distribution pattern were varying among isolates. However, no other morphological features found to differ among isolates. Majority of the R. solani isolates were fast growers as they attained complete mycelial growth within 2 days in a 90-mm Petri plate and the emergence of sclerotial structures was seen even in 4 days of incubation. Selected 10 R. solani isolates exhibited considerable variations in pathogenicity on three different rice cultivars. Vellai ponni was found to be the most susceptible rice cultivar to all the field isolates of R. solani.  相似文献

In almond, gametophytic self-incompatibility is controlled by a single multiallelic locus (S-locus). In styles, the products of S-alleles are ribonucleases, the S-RNases. Cultivated almond in California have four predominant S-alleles (S a, S b, S c, S d). We previously reported the cDNA cloning of three of these alleles, namely S b, S c and S d. In this paper we report the cloning and DNA sequence analysis of the S a allele. The Sa-RNase displays approximately 55% similarity at the amino-acid level with other almond S-RNases (Sb, Sc, and Sd) and this similarity was lower than that observed among the Sb, Sc and Sd-RNases. Using the cDNA sequence, a PCR-based identification system using genomic DNA was developed for each of the S-RNase alleles. Five almond cultivars with known self-incompatibility (SI) geno-types were analyzed. Common sequences among four S-alleles were used to create four primers, which, when used as sets, amplify DNA bands of unique size that corresponded to each of the four almond S-alleles; S a (602 bp), S b (1083 bp), S c (221 bp) and S d (343 bp). All PCR products obtained from genomic DNA isolated from the five almond cultivars were cloned and their DNA sequence obtained. The nucleotide sequence of these genomic DNA fragments matched the corresponding S-allele cDNA sequence in every case. The amplified products obtained for the S a- and S b-alleles were both longer than that expected for the coding region, revealing the presence of an intron of 84 bp in the S a-allele and 556 bp in the S b-allele. Both introns are present within the site of the hypervariable region common in S-RNases from the Rosaceae family and which may be important for S specificity. The exon portions of the genomic DNA sequences were completely consistent with the cDNA sequence of the corresponding S-allele. A useful application of these primers would be to identify the S-genotype of progeny in a breeding program, new varieties in an almond nursery, or new grower selections at the seedling stage. In studies on the infection of potato cultivars with different field isolates of Phoma exigua var. foveata, cultivar × isolate interactions, although sufficiently large to influence the relative pathogenicities of isolates on different cultivars, were not substantial enough to affect cultivar ranking order. Cultivar rank was markedly influenced by pathogen isolate only when both field and culture collection isolates were compared. This suggests that the complications of cultivar × isolate interactions can be avoided in cultivar screening tests by the use of recent field isolates of high pathogenicity. It was found that cultivar was considerably more important than isolate in determining gangrene lesion size. The need to consider both lesion diameter and depth when estimating rot size following point inoculation of different cultivars was confirmed.  相似文献

The interaction of an excess of the title ligands L with the cis-Pt(phos)2 moieties gives compounds a-bcis-[Pt(L-O)2(phos)2] (a, phos = P(Ph)3; b, phos = 1/2 dppe), in which O- is preferred to S-coordination. Such preference is confirmed by the fact that the same products are obtained by reaction of excess of L with the previously reported a-d complexes [Pt(L-O,S)(phos)2]+, (c, phos = PPh3, d, phos = 1/2 dppe), for which chelate ring opening occurs with rupture of Pt-S rather than Pt-O bonds. Compound a can be obtained also by oxidative addition of HL to [Pt(PPh3)3]. The Pt-O bonds in compounds a-d are stable towards substitution by Me2SO, pyridine and tetramethylthiourea. Substitution of L’s occurs with N,N′-diethyldithiocarbamate, which forms a very stable chelate with Pt(II). Thiourea and N,N′-dimethylthiourea also react, because they give rise to cyclometallated products [Pt(phos)2(NRC(S)NHR)]+ (R = H, CH3), with one ionised thioamido group, as revealed by an X-ray investigation of [Pt(PPh3)2(NHC(S)NH2)]+. The preference of O versus S coordination, as well as the stability of the Pt-O bonds, are discussed in terms of antisymbiosis.  相似文献

All 17 examined Polish isolates of Fusarium moniliforme Sheldon [syn. F. verticillioides (Sacc.) Nirenberg], originating from wheat, maize and barley kernels, exhibited pathogenicity to wheat seedlings of the cultivar ‘Almari’ (5 isolates-very strong, 3 strong, 3 medium, 5 weak and only one very weak). The same isolates were able to produce the mycotoxins fumonisins on rice under laboratory conditions. Fumonisin B1 (FB1) was detected in concentrations ranging from 1.3 to 2240mg/kg and fumonisin B2(FB2) from 0.7 to 600 mg/kg. The yield of the fumonisins produced in vitro was not related to the pathogenicity of individual isolates.  相似文献

This study was carried out in Desert Research Center and Faculty of Agriculture, Zagazig University, Egypt, under North Sinai conditions during three growing seasons, i.e., summer 2018, fall 2018/2019 and summer 2019 to assess the effect of radiation mutants on leaf histological features and genetic stability of the productivity of some potato cultivars under drought stress conditions. Results reveal that the genotypes can be statistically classified based on regression coefficient (bi), deviation from regression (S2di) to 4 groups (with low in S2di are considered in all groups) as: (i) Genotype with elevated average, bi = 1, it is considered as stable genotype where Cara cultivar (both generations) was included. (ii) Genotype with elevated average, bi > 1 as genotype with average stability where spunta cultivar was involved. (iii) Genotype with low mean, bi < 1 as genotype with low stability where hermes (both primary (M1) and secondary (M2) radiated generations) and Caruso (2nd generation) cultivars were involved. (iv) Genotypes including a few bi values in one generation, as genotype including low stability but are not recommended for use in this generation where Caruso cv in M1 was included. The results indicated that 20 Gy irradiation exposure revealed that Spunta cultivar produced markedly high mean combined over yield during M1 (11.771 ton/fed) and M2 (10.97 ton/fed) generations than other genotypes and ranked first over all environments. It proves that Hermes could be employed as anti-stress genotypes under stress conditions (negative conditions or poor yielding). However, spunta followed by cara cv. represented a good performance in M2 production yield (10.97 and 8.51 ton/fed, respectively), slight drift from the regression line and coefficient close to 1, therefore, both cultivars were excellent between genotypes in shape of yield stability and is recommended for different conditions. According to anatomical studies, 80 % from field capacity (FC) decreased the thickness of medvein and lamina of potato cv. spunta, also, dimensions of medvein bundle and mean diameter of vessels. In conclusion, plants treated with gamma ray at level 20 and grown under 80 % FC induced prominent increase in all previous characters.  相似文献

The effect of N form and Si nutrition on rice (Oryza sativa L.) susceptibility to blast disease (caused by Pyricularia oryzae Cav.) was assessed in the greenhouse with nutrient solution culture. The N form supplied to the susceptible cultivar IR50 affected the relative infection efficiency (RIE) of P. oryzae measured as lesions/cm2 leaf. Plants given NO3 - were more susceptible than plants receiving NH4 +-N. This result may partially explain why plants grown in nonflooded soil, where NO3 - is the main source of inorganic N, are more susceptible to blast than plants grown in flooded soils, where NH4 + is the main inorganic N source. Nitrate-N and Mn concentration were higher in leaf blades of plants grown with NO3 -. Total-N, Si, and Fe concentration were not affected by N form. The addition of Si significantly increased IR50 resistance to blast. With 2.2 mol m-3 Si in solution, RIE values were lower by more than 90% than the control with no Si added in solution. The effect of Si accumulation in leaves at various positions was further studied in cultivars having differing levels of resistance (IR50, IR36, and IAC165). Silicon addition significantly reduced RIE in the three cultivars. Silicon concentration in the topmost leaves (the only leaves showing typical blast lesions) was not significantly different among the three cultivars when 2.2 mol m-3 Si was used. Silicon was an important component in the mechanism of resistance to blast and it was effective regardless of the original level of resistance of the cultivar used. Abstract

The growth inhibitory activity of seven rice (Oryza sativa L.) cultivars and the secretion level of momilactone B from these rice cultivars were determined to understand chemical basis of the interaction of rice with other plant species. All rice cultivars inhibited the growth of hypocotyls and roots of lettuce (Lactuca sativa L.) seedlings when the lettuce was grown together with the rice, and showed different range of the inhibitory activity. These results suggest that all rice cultivars may possess allelopathic activity and the activity may be cultivar dependent. Momilactone B, which is a potent growth inhibitor, was found in root exudates of all rice cultivars, and the momilactone B concentration was also cultivar-dependent. The allelopathic activity of each rice cultivar was closely correlated with momilactone B concentration in the root exudates. The present results suggest that rice cultivars possess various allelopathic activities and their allelopathic activity may depend on the secretion level of momilactone B. Therefore, momilactone B may play an important role in rice allelopathy and in the chemical interactions of rice with other plant species.  相似文献

Ammonia-oxidizing bacteria (AOB) populations were studied on the root surface of different rice cultivars by PCR coupled with denaturing gradient gel electrophoresis (DGGE) and fluorescence in situ hybridization (FISH). PCR-DGGE of the ammonium monooxygenase gene (amoA) showed a generally greater diversity on root samples compared to rhizosphere and unplanted soil. Sequences affiliated with Nitrosomonas spp. tended to be associated with modern rice hybrid lines. Root-associated AOB observed by FISH were found within a discrete biofilm coating the root surface. Although the total abundance of AOB on root biofilms of different rice cultivars did not differ significantly, there were marked contrasts in their population structure, indicating selection of Nitrosomonas spp. on roots of a hybrid cultivar. Observations by FISH on the total bacterial community also suggested that different rice cultivars support different bacterial populations even under identical environmental conditions. The presence of active AOB in the root environment predicts that a significant proportion of the N taken up by certain rice cultivars is in the form of NO3 -N produced by the AOB. Measurement of plant growth of hydroponically grown plants showed a stronger response of hybrid cultivars to the co-provision of NH4 + and NO3 . In soil-grown plants, N use efficiency in the hybrid was improved during ammonium fertilization compared to nitrate fertilization. Since ammonium-fertilized plants actually receive a mixture of NH4 + and NO3 with ratios depending on root-associated nitrification activity, these results support the advantage of co-provision of ammonium and nitrate for the hybrid cultivar.  相似文献

Rice blast, caused by the fungus Magnaporthe grisea, is a globally important disease of rice that causes annual yield losses. The segregation of genes controlling the virulence of M. grisea on rice was studied to establish the genetic basis of cultivar specificity in the interaction of rice and M. grisea. The segregation of avirulence and virulence was studied in 87 M. grisea F1 progeny isolates from a cross of two isolates, Guy11 and JS153, using resistance-gene-differential rice cultivars. The segregation ratio indicated that avirulence and virulence in the rice cultivars Aichi–asahi and K59, respectively, are controlled by single major genes. Genetic analyses of backcrosses and full-sib crosses in these populations were also performed. The χ2 test of goodness-of-fitness for a 1:1 ratio indicated that one dominant gene controls avirulence in Aichi-asahi and K59 in this population. Based on the resistance reactions of rice differential lines harboring known resistance genes to the parental isolates, two genetically independent avirulence genes, AVR–Pit and AVR–Pia, were identified. Genetic linkage analysis showed that the SSR marker m355–356 is closely linked to AVR–Pit, on the telomere of chromosome 1 at a distance of approximately 2.3 cM. The RAPD marker S487, which was converted to a sequence-characterized amplified region (SCAR) marker, was found to be closely linked to AVR–Pia, on the chromosome 7 telomere at a distance of 3.5 cM. These molecular markers will facilitate the positional cloning of the two AVR genes, and can be applied to molecular-marker-assisted studies of M. grisea populations.  相似文献

Bicarbonate has been regarded as a major factor for inducing Zn deficiency in lowland rice, but the mechanisms responsible for this effect are not yet fully understood. The objective of the present study was to test whether early effects of bicarbonate (HCO3 )are inhibition of root growth due to the accumulation of organic acids induced by HCO3 . Solution culture experiments were conducted using two rice cultivars differing in susceptibility to Zn deficiency, and four bicarbonate concentrations (0, 5, 10, 20 mM). Bicarbonate (5–20 mM) strongly inhibited root growth of the Zn-inefficient cultivar within 4 days of treatments. In contrast, root growth of the Zn-efficient cultivar was slightly stimulated with bicarbonate at 5–10 mM and not affected at 20 mM. The inhibitory effect of bicarbonate on root growth in the Zn-inefficient cultivar was mainly that of impairment of new root initiation rather than suppression of elongation of individual roots. Bicarbonate (5–20 mM) increased the concentrations of malate, succinate and citrate in the roots of both cultivars, but to a greater extent for the Zn-inefficient than for the Zn-efficient cultivars. The results suggest that the impairment of root growth was likely to be the initial action of bicarbonate in inducing Zn deficiency in lowland rice, and the inhibitory effect of bicarbonate on root growth of the Zn-inefficient cultivar might result from high accumulation and an insufficient compartmentation of organic acids in the root cells.  相似文献

To develop a rice cultivar that would be suitable for direct-seedingcultivation in cooler temperate regions, we generated transgenic rice plants inwhich a rice encoding a -class glutathioneS-transferase (GST) under the control of a maize ubiquitinpromoter. GSTs have been suggested to be responsible for tolerance to variousstresses such as cold, salt and drought by detoxification of xenobioticcompounds and reactive oxygen species. A total of 87 R0 transgenicrice plants harboring a chimeric GST gene were generatedusing Agrobacterium mediated transformation. ThreeR2 lines homozygous for the transgene were assayed for GST activityand had higher GST and glutathione peroxidase activities thannon-transformants.Seedlings of the transgenic lines demonstrated greatly enhanced germination andgrowth rates at low temperature grown under submergence. The GST transgeniclines should be useful for breeding rice cultivars suitable for direct-seedingcultivation in cooler temperate regions.  相似文献

The impact of root exudates, collected from five rice cultivars, on methane (CH4) production was studied in a paddy soil under anaerobic conditions. Root exudates of the cultivars Dular, IR72 and IR65598 collected at four growth stages and of B40 and IR65600 collected at two growth stages showed that (a) CH4 production was commenced rapidly within 2 h upon exudate addition and reached a maximum within a day of addition, and (b) 7-d incubation periods were sufficient to study exudate-induced CH4 production potentials. Among different cultivars, high C releases from roots, increased the methanogenic source strength of the soil, which finally controlled CH4 production. The relationship of the amount of CH4 produced was stronger for the amount of total organic C (r = 0.920) than for the amount of organic acids (r = 0.868) added through exudates. Apparently, CH4 production and CH4 emission are more closely related to the release pattern of root exudate-C than to its individual components. The proportion of exudate-C converted to CH4 ranged between 61 and 83% and remained unaffected by cultivars and growth stages suggesting that the majority of exudate-C served as a methanogenic substrate in the anoxic rice soils. These observations indicate that the use of high-yielding cultivars with lowest excretion (for example IR65598, IR65600) would result in lowest exudate-induced CH4 production. Therefore, cultivar choice could greatly influence regional and global CH4 emissions and screening/selection of exiting rice cultivars, and/or breeding new cultivars with low exudation rates could offer an important methane mitigation option as long as yields are not compromised.  相似文献

Phytotoxicity of AAL-toxin and fumonisin B1 to six cultivars of tomato was compared with the pathogenicity of their fungal sources, Alternaria alternata and Fusarium moniliforme, respectively. These include two AAL-toxin susceptible cultivars with genotypes(asc/asc), three resistant cultivars (Asc/Asc), and a heterozygous cultivar (Asc/asc.) A. alternata spores were pathogenic to the susceptible but not to the resistant cultivars F. moniliforme was not pathogenic to any of the tomatoes. Filtrates of both fungi grown on rice containing their respective toxins caused necrosis within 48 h and eventually mortality on susceptible cultivars but not on the resistant lines. The heterozygous cultivar Asc/asc showed minimal damage and no mortality after 14 days exposure to both filtrates and both toxins. The spores of both fungi had no effect on heterozygous intact plants. Tomato leaf disc bioassays with AAL-toxin and fumonisin B1 at 1μM caused cellular leakage and reduced chlorophyll content in susceptible cultivars and minimal effects on the heterozygous and resistant varieties.  相似文献

Weedy rice represents an important resource for rice improvement. The F1 hybrid between the japonica wide compatibility rice cultivar 02428 and a weedy rice accession from Yunnan province (SW China) suffered from pollen sterility. Pollen abortion in the hybrid occurred at the early bicellular pollen stage, as a result of mitotic failure in the microspore, although the tapetum developed normally. Genetic mapping in a BC1F1 population (02428//Yunnan weedy rice (YWR)/02428) showed that a major QTL for hybrid pollen sterility (qPS-1) was present on chromosome 1. qPS-1 was fine-mapped to a 110 kb region known to contain the hybrid pollen sterility gene Sa, making it likely that qPS-1 is either identical to, or allelic with Sa. Interestingly, F1 hybrid indicated that Dular and IR36 were assumed to carry the sterility-neutral allele, Sa n . Re-sequencing SaM and SaF, the two component genes present at Sa, suggested that variation for IR36 and Dular may be responsible for the loss of male sterility, and the qPS-1 sequence might be derived from wild rice or indica cultivars. A phylogenetic analysis based on microsatellite genotyping suggested that the YWR accession is more closely related to wild rice and indica type cultivars than to japonica types. Thus it is probable that the YWR accession evolved from a spontaneous hybrid between wild rice and an ancient cultivated strain of domesticated rice.  相似文献

The study aims at assessment of morphological, molecular and pathogenic variability of Alternaria helianthi, incitant of leaf blight of sunflower. Morphological characteristics determined for 26 isolates of A. helianthi from India revealed variations in shape of culture, pigmentation, conidial measurements, number of septa and colony growth. The conidia of isolate Ah‐7 were long, while conidia of isolate Ah‐15 were short. Based on cultural characters, isolates were classified into four groups. Genetic variability of the isolates was assessed by random amplified polymorphic DNA analysis. Good polymorphism was observed and cluster analysis indicated presence of six genetically distinct groups among the isolates. The isolates Ah‐1, Ah‐7 and Ah‐14 were genetically distinct. Resistant sources are not available in cultivated sunflower, while wild Helianthus species possess resistance to multiple stresses. We evaluated reaction of wild Helianthus species to isolates of A. helianthi. Among wild Helianthus species, H. tuberosus followed by H. occidentalis showed moderately to highly resistant reaction to all the isolates and recorded less disease incidence. The species H. argophyllus followed by H. laevigatus showed more disease incidence. The cultivated sunflower recorded susceptible reaction to most of the isolates and recorded high disease incidence. The isolates differed significantly for pathogenic reaction and were grouped into three pathogenicity groups; low, medium and high. Six isolates induced <20% disease incidence and were included in the low pathogenicity group. Majority of isolates were in the medium pathogenicity group. Six isolates i.e. Ah‐9, Ah‐10, Ah‐18, Ah‐20, Ah‐24 and Ah‐26 induced more than 50% disease incidence and were considered high pathogenicity group. Our results demonstrate the existence of considerable variation in resistance of Helianthus species to A. helianthi and also in morphological and genetic characters of A. helianthi isolates prevalent in India.  相似文献

Verticillium wilt of lettuce caused by Verticillium dahliae can cause severe economic damage to lettuce producers. Complete resistance to race 1 isolates is available in Lactuca sativa cultivar (cv.) La Brillante and understanding the genetic basis of this resistance will aid development of new resistant cultivars. F1 and F2 families from crosses between La Brillante and three iceberg cultivars as well as a recombinant inbred line population derived from L. sativa cv. Salinas 88 × La Brillante were evaluated for disease incidence and disease severity in replicated greenhouse and field experiments. One hundred and six molecular markers were used to generate a genetic map from Salinas 88 × La Brillante and for detection of quantitative trait loci. Segregation was consistent with a single dominant gene of major effect which we are naming Verticillium resistance 1 (Vr1). The gene described large portions of the phenotypic variance (R 2 = 0.49–0.68) and was mapped to linkage group 9 coincident with an expressed sequence tag marker (QGD8I16.yg.ab1) that has sequence similarity with the Ve gene that confers resistance to V. dahliae race 1 in tomato. The simple inheritance of resistance indicates that breeding procedures designed for single genes will be applicable for developing resistant cultivars. QGD8I16.yg.ab1 is a good candidate for functional analysis and development of markers suitable for marker-assisted selection.  相似文献
