Plant mitochondria can differ in size, shape, number and protein content across different tissue types and over development. These differences are a result of signaling and regulatory processes that ensure mitochondrial function is tuned in a cell-specific manner to support proper plant growth and development. In the last decade, the processes involved in mitochondrial biogenesis are becoming clearer, including; how dormant seeds transition from empty promitochondria to fully functional mitochondria with extensive cristae structures and various biochemical activities, the regulation of nuclear genes encoding mitochondrial proteins via regulators of the diurnal cycle in plants, the mitochondrial stress response, the targeting of proteins to mitochondria and other organelles and connections between the respiratory chain and protein import complexes. All these findings indicate that mitochondrial function is a part of an integrated cellular network, and communication between mitochondria and other cellular processes extends beyond the known exchange or transport of metabolites. Our current knowledge now needs to be used to gain more insight into the molecular components at various levels of this hierarchical and complex regulatory and communication network, so that mitochondrial function can be predicted and modified in a rational manner. 相似文献

A considerable surge of interest in the mismatch repair (MMR) system has been brought about by the discovery of a link between Lynch syndrome, an inherited predisposition to cancer of the colon and other organs, and malfunction of this key DNA metabolic pathway. This review focuses on recent advances in our understanding of the molecular mechanisms of canonical MMR, which improves replication fidelity by removing misincorporated nucleotides from the nascent DNA strand. We also discuss the involvement of MMR proteins in two other processes: trinucleotide repeat expansion and antibody maturation, in which MMR proteins are required for mutagenesis rather than for its prevention. 相似文献

The maximum stresses to which a wide range of mammalian limb tendons could be subjected in life were estimated by considering the relative cross-sectional areas of each tendon and of the fibres of its muscle. These cross-sectional areas were derived from mass and length measurements on tendons and muscles assuming published values for the respective densities. The majority of the stresses are low. The distribution has a broad peak with maximum frequency at a stress of about 13 MPa, whereas the fracture stress for tendon in tension is about 100 MPa. Thus, the majority of tendons are far thicker than is necessary for adequate strength. Much higher stresses are found among those tendons which act as springs to store energy during locomotion. The acceptability of low safety factors in these tendons has been explained previously (Alexander, 1981). A new theory explains the thickness of the majority of tendons. The muscle with its tendon is considered as a combined system which delivers mechanical energy: the thickness of the tendon is optimized by minimizing the combined mass. A thinner tendon would stretch more. To take up this stretch, the muscle would require longer muscle fibres, which would increase the combined mass. The predicted maximum stress in a tendon of optimum thickness is about 10 MPa, which is within the main peak of the observed stress distribution. Individual variations from this value are to be expected and can be understood in terms of the functions of the various muscles. 相似文献

Flexistyly is a recently documented stylar polymorphism involving both spatial and temporal segregation of sex roles within hermaphroditic flowers. Using the experimental manipulation of stigma movement in self-compatible Alpinia mutica, we tested the hypothesis that selection for reducing interference between male and female function drives the evolution and/or maintenance of stigma movement. In experimental arrays, anaflexistylous (protogynous) flowers served as pollen donors competing for mating opportunities on cataflexistylous (protandrous) flowers. The pollen donors were either manipulated so their stigmas could not move or were left intact, and their success was determined using allozymes to assess the paternity of recipient seeds. We found that manipulated flowers sired a significantly smaller proportion of seeds, showing that stigma movement in unmanipulated plants increased male fitness. This result was strongest under conditions in which pollen competition was expected to be highest, specifically when pollinators visited multiple donor plants before visiting recipient flowers. 相似文献

Male chimpanzees, Pan troglodytes, cooperate to defend a community range within which resident females range in smaller core areas. There has been debate over exactly what males are defending, whether mates, territory or both. One hypothesis holds that males are defending mates, and that an increase in community range size will lead directly to the acquisition of more females. However, males frequently attack females as well as males at the edge of the community range. We examined 18 years of observational data on the Gombe chimpanzees to determine the behaviour of males during extragroup encounters, and the consequences of changes in community range size on the number of adult females and indirect measures of food availability. Males were always aggressive to males from other communities, and often attacked adult females, especially those that were not sexually receptive, were older, and/or had more than one offspring. The number of females did not increase with range size, but several measures suggested an increase in food availability with range size. These measures include more time spent in large foraging parties, higher encounter rates with resident females, more encounters with sexually receptive females and higher female reproductive rates. These findings suggest that males defend a feeding territory for their resident females and protect them from sexual harassment. Although a large range may eventually attract more females, this is not an immediate consequence of range expansion. Male number was not correlated with community range size. 相似文献

Animal hybridization is increasingly recognized as common and evolutionarily important, but the role of behavior in promoting hybridization events is not well understood. Understanding the behavioral causes of hybridization requires understanding the ecological, demographic, and phenotypic influences on mate choice in hybridizing taxa. Here, I review how these influences on mate choice can contribute to hybridization by (1) circumventing (female) choice, (2) bringing formerly isolated species into sympatry, (3) masking cues important for mate recognition, (4) altering the traits or preferences involved in mate recognition, or (5) altering the costs and benefits of mate choice. In particular, hybridization in response to either high direct costs of mate choice or high direct benefits of heterospecific mating may be widespread, a possibility that awaits further testing. Adopting a mate choice perspective to the study of hybridization challenges the assumption of hybrids as reproductive “mistakes” and allows for functional explanations regarding the occurrence and maintenance of hybridization. As evidence of the prevalence and evolutionary importance of animal hybridization accumulates, investigations into the role of behavior will be increasingly important for our understanding of diversification and for conservation applications. 相似文献

Despite seminal papers that stress the significance of silicon (Si) in plant biology and ecology, most studies focus on manipulations of Si supply and mitigation of stresses. The ecological significance of Si varies with different levels of biological organization, and remains hard to capture. We show that the costs of Si accumulation are greater than is currently acknowledged, and discuss potential links between Si and fitness components (growth, survival, reproduction), environment, and ecosystem functioning. We suggest that Si is more important in trait-based ecology than is currently recognized. Si potentially plays a significant role in many aspects of plant ecology, but knowledge gaps prevent us from understanding its possible contribution to the success of some clades and the expansion of specific biomes. 相似文献

Mitochondrial genome integrity is fundamental to mammalian cell viability. Since mitochondrial DNA is constantly under attack from oxygen radicals released during ATP production, DNA repair is vital in removing oxidatively generated lesions in mitochondrial DNA, but the presence of a strong base excision repair system has not been demonstrated. Here, we addressed the presence of such a system in mammalian mitochondria involving the primary base lesion repair enzyme DNA polymerase (pol) β. Pol β was localized to mammalian mitochondria by electron microscopic-immunogold staining, immunofluorescence co-localization and biochemical experiments. Extracts from purified mitochondria exhibited base excision repair activity that was dependent on pol β. Mitochondria from pol β-deficient mouse fibroblasts had compromised DNA repair and showed elevated levels of superoxide radicals after hydrogen peroxide treatment. Mitochondria in pol β-deficient fibroblasts displayed altered morphology by electron microscopy. These results indicate that mammalian mitochondria contain an efficient base lesion repair system mediated in part by pol β and thus pol β plays a role in preserving mitochondrial genome stability. 相似文献

The levels of proteins required for methyl-directed mismatch repair appear to decline in stationary-phase and nutritionally-deprived cells of Escherichia coli. It has been hypothesized that error-correction by the system also declines, and this decline is responsible for adaptive or stationary-phase mutations. However, evidence in support of this hypothesis is lacking. The mismatch repair system is no less effective in correcting errors during prolonged selection than it is during growth. Furthermore, mismatch repair proteins supplied in excess reduce both growth-dependent and adaptive mutation. 相似文献

To determine whether phospholipid is required for the activity of mammalian signal peptidase, the enzyme was partially purified from porcine pancreas and then extensively freed of phospholipid by SP-Sephadex C-50 chromatography. gamma-Secretase cleavage of the amyloid precursor protein already subjected to a prior beta-secretase cleavage generates beta-amyloid (Abeta) peptide fragments, which are major constituents of the amyloid plagues found in Alzheimer's disease brain tissues. gamma-Secretase activity and components of the gamma-secretase complex are found in the endoplasmic reticulum-Golgi intermediate compartment, the Golgi, the trans-Golgi network, the plasma membrane, the endosomal-lysosomal system and recently, the mitochondria. Abeta fragments have been shown to be neurotoxic, leading to mitochondrial dysfunction and enhanced apoptotic cell death. However, if Abeta fragments are indeed detrimental to neurons, the widespread presence of enzymatic activity that would result in their generation in the cell appears to make little sense. The presence of a gamma-secretase complex in the mitochondrion, an organelle that is particularly susceptible to Abeta toxicity, is even more puzzling. Emerging evidence suggests that both secreted and intracellular Abeta fragments have endogenous functions. Also, while the fibrillogenic Abeta1-42 is clearly neurotoxic, the more abundant and soluble Abeta1-40 is an antioxidant and could potentially be neuroprotective in several ways. A "physiological" amount of Abeta1-40 production by cellular gamma-secretase activity may be part of the neuron's natural counter against oxidative damage, in addition to endogenous roles in neuronal survival and modulation of synaptic transmission. In any case, whether Abeta is produced locally in the mitochondria and the function for mitochondrial Abeta, if produced, is yet unclear. 相似文献

Constitutional mismatch repair deficiency syndrome is a rare autosomal recessive syndrome caused by homozygous mutations in mismatch repair genes. This is characterized by the childhood onset of brain tumors, colorectal cancers, cutaneous manifestations of neurofibromatosis-1 like café au lait spots, hematological malignancies, and occasionally other rare malignancies. Here, we would like to present a family in which the sibling had glioblastoma, and the present case had acute lymphoblastic lymphoma and colorectal cancer. Mammals generally ejaculate many more spermatozoa than seem to be needed for fertilization. This apparent profligacy has not been explained, but observations made in marsupials may shed light on it. The Virginia opossum, Didelphis virginiana, inseminates only about three million spermatozoa, a very low number. As a corollary, relatively few (ca. 13 X 10(6] are stored in each cauda epididymidis. However, some 5% of the spermatozoa that the opossum ejaculates populate the oviduct about 12 h later when ovulation can be anticipated--a success rate in the female orders of magnitude greater than in eutherian mammals. It is not certain what determines the unusually efficient transport to and the high survival rate of spermatozoa in the oviduct of Didelphis, but two unusual features suggest themselves as possible contributors. Didelphis (and all other American marsupial) spermatozoa undergo a head-to-head pairing in the epididymis by the acrosomal face; this serves to isolate the acrosome of ejaculated spermatozoa from the female milieu until the pairs separate in the oviduct. Secondly, spermatozoa are housed in special crypts in the isthmus of the oviduct. Australian marsupials, which usually lack such features, store spermatozoa in the epididymis in numbers more close to those in comparably sized eutheriam mammals. Exceptions which store very low sperm numbers there can be seen in one Australian Family, the Dasyuridae . The spermatozoa of dasyurids are not paired, but the species examined possess distinctive sperm storage crypts in the oviducal isthmus similar to those in the opossum. The present findings suggest that where mechanisms exist that could protect the acrosome and, or, the whole spermatozoon in the female tract, a much lower level of sperm production can be maintained without compromising fertility. While the number ejaculated typically by any one species is probably determined ultimately by several interacting factors, it therefore seems likely that a most important one in this respect relates to conditions spermatozoa face in the female tract. 相似文献
