Background/Objective

The CDKAL1 gene is among the best-replicated susceptibility loci for type 2 diabetes, originally identified by genome-wide association studies in humans. To clarify a physiological importance of CDKAL1, we examined effects of a global Cdkal1-null mutation in mice and also evaluated the influence of a CDKAL1 risk allele on body mass index (BMI) in Japanese subjects.

Methods

In Cdkal1-deficient (Cdkal1 −/−) mice, we performed oral glucose tolerance test, insulin tolerance test, and perfusion experiments with and without high-fat feeding. Based on the findings in mice, we tested genetic association of CDKAL1 variants with BMI, as a measure of adiposity, and type 2 diabetes in Japanese.

Principal Findings

On a standard diet, Cdkal1 −/− mice were modestly lighter in weight than wild-type littermates without major alterations in glucose metabolism. On a high fat diet, Cdkal1 −/− mice showed significant reduction in fat accumulation (17% reduction in %intraabdominal fat, P = 0.023 vs. wild-type littermates) with less impaired insulin sensitivity at an early stage. High fat feeding did not potentiate insulin secretion in Cdkal1 −/− mice (1.0-fold), contrary to the results in wild-type littermates (1.6-fold, P<0.01). Inversely, at a later stage, Cdkal1 −/− mice showed more prominent impairment of insulin sensitivity and glucose tolerance. mRNA expression analysis indicated that Scd1 might function as a critical mediator of the altered metabolism in Cdkal1 −/− mice. In accordance with the findings in mice, a nominally significant (P<0.05) association between CDKAL1 rs4712523 and BMI was replicated in 2 Japanese general populations comprising 5,695 and 12,569 samples; the risk allele for type 2 diabetes was also associated with decreased BMI.

Conclusions

Cdkal1 gene deletion is accompanied by modestly impaired insulin secretion and longitudinal fluctuations in insulin sensitivity during high-fat feeding in mice. CDKAL1 may affect such compensatory mechanisms regulating glucose homeostasis through interaction with diet.  相似文献

Cirsium setidens is a perennial medicinal herb that is rich in flavonoids. We investigated in this study the effect of a C. setidens ethanol extract (CSE) on the development of nonalcoholic fatty liver in mice fed a high-fat diet (HF). C57BL/6J mice were fed either a control diet (CON) or HF for 8 weeks, and then fed CON, HF, or HF with 100 mg/kg of BW CSE (HF+CSE) for an additional 7 weeks. The final body weight and adipose tissue weight of the mice in the HF+CSE group were significantly lower than those in the HF group. CSE also markedly diminished both the lipid droplets in the liver tissues and decreased the hepatic and serum triglycerides (TG) concentrations. CSE strongly increased the hepatic mRNA levels of carnitine palmitoyltransferase (CPT1) and medium-chain acyl-CoA dehydrogenase (MCAD), the fatty acid β-oxidation enzymes. The hepatic levels of phosphorylated-AMP-activated protein kinase (AMPK) were significantly higher in the HF+CSF group than in the HF group. These results suggest that CSE inhibited hepatic fat accumulation by up-regulating the expression of the fatty acid β-oxidation genes.  相似文献

Though widely employed for clinical intervention in obesity, metabolic syndrome, seizure disorders and other neurodegenerative diseases, the mechanisms through which low carbohydrate ketogenic diets exert their ameliorative effects still remain to be elucidated. Rodent models have been used to identify the metabolic and physiologic alterations provoked by ketogenic diets. A commonly used rodent ketogenic diet (Bio-Serv F3666) that is very high in fat (~94% kcal), very low in carbohydrate (~1% kcal), low in protein (~5% kcal), and choline restricted (~300 mg/kg) provokes robust ketosis and weight loss in mice, but through unknown mechanisms, also causes significant hepatic steatosis, inflammation, and cellular injury. To understand the independent and synergistic roles of protein restriction and choline deficiency on the pleiotropic effects of rodent ketogenic diets, we studied four custom diets that differ only in protein (5% kcal vs. 10% kcal) and choline contents (300 mg/kg vs. 5 g/kg). C57BL/6J mice maintained on the two 5% kcal protein diets induced the most significant ketoses, which was only partially diminished by choline replacement. Choline restriction in the setting of 10% kcal protein also caused moderate ketosis and hepatic fat accumulation, which were again attenuated when choline was replete. Key effects of the 5% kcal protein diet – weight loss, hepatic fat accumulation, and mitochondrial ultrastructural disarray and bioenergetic dysfunction – were mitigated by choline repletion. These studies indicate that synergistic effects of protein restriction and choline deficiency influence integrated metabolism and hepatic pathology in mice when nutritional fat content is very high, and support the consideration of dietary choline content in ketogenic diet studies in rodents to limit hepatic mitochondrial dysfunction and fat accumulation.  相似文献

Vinpocetine is a safe nootropic agent used for neurological and cerebrovascular diseases. The anti-inflammatory activity of vinpocetine has been shown in cell based assays and animal models, leading to suggestions as to its utility in analgesia. However, the mechanisms regarding its efficacy in inflammatory pain treatment are still not completely understood. Herein, the analgesic effect of vinpocetine and its anti-inflammatory and antioxidant mechanisms were addressed in murine inflammatory pain models. Firstly, we investigated the protective effects of vinpocetine in overt pain-like behavior induced by acetic acid, phenyl-p-benzoquinone (PBQ) and formalin. The intraplantar injection of carrageenan was then used to induce inflammatory hyperalgesia. Mechanical and thermal hyperalgesia were evaluated using the electronic von Frey and the hot plate tests, respectively, with neutrophil recruitment to the paw assessed by a myeloperoxidase activity assay. A number of factors were assessed, both peripherally and in the spinal cord, including: antioxidant capacity, reduced glutathione (GSH) levels, superoxide anion, tumor necrosis factor alpha (TNF-α) and interleukin 1 beta (IL-1β) levels, as well as nuclear factor kappa B (NF-κB) activation. Vinpocetine inhibited the overt pain-like behavior induced by acetic acid, PBQ and formalin (at both phases), as well as the carrageenan-induced mechanical and thermal hyperalgesia and associated neutrophil recruitment. Both peripherally and in the spinal cord, vinpocetine also inhibited: antioxidant capacity and GSH depletion; increased superoxide anion; IL-1β and TNF-α levels; and NF-κB activation. As such, vinpocetine significantly reduces inflammatory pain by targeting oxidative stress, cytokine production and NF-κB activation at both peripheral and spinal cord levels.  相似文献

This paper analyzes the interaction between science, philosophy and politics (including ideology) in the early work of J. B. S. Haldane (from 1922 to 1937). This period is particularly important, not only because it is the period of Haldane's most significant biological work (both in biochemistry and genetics), but also because it is during this period that his philosophical and political views underwent their most significant transformation. His philosophical stance first changed from a radical organicism to a position far more compatible with mechanical materialism. The primary intellectual influence that was responsible for this shift was that of F. G. Hopkins. Later, Haldane came to accept Marxism and its official metaphysics, dialectical materialism, a move that let him accept the materialist conception of the world while still maintaining a resolute distance from mechanism. Throughout all these changes, what is most obvious is the influence of science on Haldane's philosophical views. An influence in the opposite direction is far less apparent.Parts of this paper are extracted from a longer work which concerns the interactions between philosophy and science throughout Haldane's scientific career (Sarkar forthcoming). The general conclusions reached here, from a consideration of Haldane's work only from 1922 to 1937 (see Section 6), remain the same for the rest of his life, as is detailed in the longer work. Simvastatin (SIM) is anti-inflammatory. We used low density lipoprotein receptor knockout (LDLR-/-) mice and human aortic smooth muscle cells (HASMCs) as model systems to study the effect of SIM on arterial calcification and to explore the potential mechanisms contributing to this protective effect. High-fat diet (HFD) caused the LRLR -/- to develop dyslipidemia, diabetics, atherosclerosis and aortic smooth muscle calcification. SIM, N-acetyl cysteine (NAC, a ROS scavenger) and apocynin (APO, a NADPH oxidase inhibitor) did not significantly retard the development of dyslipidemia or diabetic. However, those treatments were still effective in attenuating the HFD-induced atherosclerosis and aortic smooth muscle calcification. These findings suggest that the protective effect of SIM against aortic calcification is not contributed by the cholesterol lowering effect. SIM, NAC and APO were found to attenuate the HFD induced elevation of serum TNF-α, soluble TNFR1 (sTNFR1), 3-nitro-tyrosine. We hypothesized that the pro-inflammatory cytokine, oxidative stress and TNFR1 played a role in inducing aortic calcification. We used HASMC to investigate the role of TNF-α, oxidative stress and TNFR1 in inducing aortic calcification and to elucidate the mechanism contributes the protective effect of SIM against aortic calcification. We demonstrated that treating HASMC with TNF-α induced cell Ca deposit and result in an increase in ALP, NADPH oxidase activity, NF-kB subunit p65, BMP2, MSX2, and RUNX2 expression. SIM suppressed the TNF-α induced activation of NADPH oxidase subunit p47, the above-mentioned bone markers and TNFR1 expression. Furthermore, p65, p47 and TNFR1 siRNAs inhibited the TNF-α-mediated stimulation of BMP-2, MSX2, RUNX2 expression. SIM, APO, and NAC either partially inhibit or completely block the TNF-α induced H2O2 or superoxide production. These results suggest that SIM may, independent of its cholesterol-lowering effect, suppresses the progression of vascular diseases through the inhibition of the inflammation mediators TNF-α and TNFR1.  相似文献

Our previous studies had reported that morin, a bioflavanoid exhibited potent anti-inflammatory effect against adjuvant-induced arthritic rats. In this current study, we investigated the anti-inflammatory mechanism of morin against monosodium urate crystal (MSU)-induced inflammation in RAW 264.7 macrophage cells, an in vitro model for acute gouty arthritis. For comparison purpose, colchicine was used as a reference drug. We have observed that morin (100–300 μM) treatment significantly suppressed the levels of inflammatory cytokines (TNF-α, IL-1β, IL-6, MCP-1 and VEGF), inflammatory mediators (NO and PEG2), and lysosomal enzymes (acid phosphatase, β-galactosidase, N-acetyl glucosamindase and cathepsin D) in MSU-crystals stimulated macrophage cells. The mRNA expression of pro-inflammatory cytokines (TNF-α, IL-1β, IL-6, and MCP-1), inflammatory enzymes (iNOS and COX-2), and NF-κBp65 was found downregulated in MSU crystal stimulated macrophage cells by morin treatment, however, the mRNA expression of hypoxanthine phospho ribosyl transferse (HPRT) was found to be increased. The flow cytometry analysis revealed that morin treatment decreased intracellular reactive oxygen species levels in MSU crystal stimulated macrophage cells. The western blot analysis clearly showed that morin mainly exerts its anti-inflammatory effects by inhibiting the MSU crystal-induced COX-2 and TNF-α protein expression through the inactivation of NF-κB signaling pathway in RAW 264.7 macrophage cells similar to that of BAY 11–7082 (IκB kinase inhibitor). Our results collectively suggest that morin can be a potential therapeutic agent for inflammatory disorders like acute gouty arthritis.  相似文献

The consumption of high-fat and high-sugar diets, in the form of junk food, and binge eating are now common. Increasing evidence suggests that a high-fat diet (HFD) can induce neuroinflammation and alter behavior. I aimed to study the effects of diets of differing fat content on neuroinflammation and spatial memory using an object–place (OP) task. Thirty-two adult male rats were allocated to four groups and fed a regular diet (Regular diet), a control diet (Control diet), an HFD (60% of calories from lard), or a high-coconut oil diet (HCOD; 60% of calories from coconut oil) for 3 days. Their water intake, food consumption, body mass, and metabolic variables were measured. HFD-fed rats showed significantly poorer performance on the OP task, as assessed using the discrimination index (??0.208?±?0.094), than the Regular (0.462?±?0.078; P?<?0.0001) and Control (0.379?±?0.081; P?=?0.0003) groups. However, no significant difference was observed in spatial memory between the HCOD and Regular groups. The concentrations of neuroinflammatory markers (interleukin [IL]-1β, IL-6, tumor necrosis factor α, and nuclear factor κB) were also measured in the hippocampus and prefrontal cortex. HFD-fed rats showed significantly higher levels of neuroinflammatory markers than the Regular and Control diet-fed groups. HCOD feeding did not induce neuroinflammation in the hippocampus and prefrontal cortex compared with the Regular and Control groups.

An account is given of early liberations and the spread of hedgehogs throughout New Zealand between 1869 and 1973. Evidence, gathered largely from questionnaires and personal observations, is summarised in tables and maps to show the present range and abundance of the species. Hedgehogs dispersed from many points of liberation; their numbers increased dramatically in the South Island between 1910 and 1940, and in the North Island between 1920 and the 1950s. By the 1940s they had colonised most of the lowland areas, and their range had extended to the foot of the bushed mountainous regions; a few were found at altitudes of 2000 m. Since 1948 they have extended their range a little to include parts of the central volcanic plateau of the North Island, areas of inland Nelson, and northern Westland. Their numbers appear to have stabilised over the past 25 years. Today, hedgehogs are most abundant in intensively farmed lowland districts, towns, and suburbs. They become less numerous with increasing altitude, and few are encountered above 800 m or in wet, bushed areas. They are absent from areas where rainfall exceeds 250 cm/year or where more than 250 frosts occur annually. Factors limiting the abundance of hedgehogs in New Zealand are discussed.  相似文献

Two novel delphinidin 3-(tri or di)-glycoside-7-glycosides were isolated from the violet–blue flowers of Vinca minor L. and V. major L. (Family: Apocynaceae), and determined to be delphinidin 3-O-[2-O-(β-xylopyranosyl)-6-O-(α-rhamnopyranosyl)-β-galactopyranoside]-7-O-(α-rhamnopyranoside) [= delpphinidin 3-(2G-xylosylrobinobioside)-7-rhamnoside] as major floral anthocyanin of V. minor and delphinidin 3-O-[6-O-(α-rhamnopyranosyl)-β-galactopyranoside]-7-O-(α-rhamnopyranoside) [= delpphinidin 3-robinobioside-7-rhamnoside] as major floral anthocyanin of V. major by chemical and spectroscopic methods. In addition, chlorogenic acid and kaempferol 3-O-[6-O-(α-rhamnopyranosyl)-β-galactopyranoside]-7-O-(α-rhamnopyranoside) [= kaempferol 3-robinobioside-7-rhamnoside (robinin)] were identified in these flowers. In this paper, the relation between the structure of floral anthocyanins and classification of Vinca species was discussed.  相似文献

In this article we test the hypothesis that the secular increase in heights in the course of the second half of the nineteenth century was associated with the rise of the breadwinner-homemaker household. In these ‘modern’ households, women raised the living standards (quality and quantity of food, hygiene and care) for all members, especially the children. We model the assumed contributions to the family budget by age and gender of household members, and find that a strong imbalance between consumers and producers in the household put severe strains on effective resource allocation, leading to lower net nutrition and lower young adult heights. We suggest a carefully calibrated consumer/producer ratio as an indicator to capture these effects. The ratio is not meant to replace others, and we show that sibling rank order as well as gender preferences also played a role in intra-household resource allocation. For our research, we have used a database with reconstructed life histories (including co-residence) of 3003 Dutch army recruits. Our results indicate that the consumer/producer ratio as experienced by recruits in their early life indeed had a strong impact (-1,8 cm) on their heights. However, this effect differed by social class, which can be explained by differences in acceptance of the income pooling model.  相似文献

We investigated the influence of drone size and potential reproductive quality on caste interactions by adding large drones reared in drone cells (DC drones; considered to be of higher quality) and small drones reared in worker cells (WC drones; of lower quality) to two observation colonies and monitoring worker–drone interactions and acceptance by workers. When initially introduced into the colonies more DC drones received trophallaxis, whereas more WC drones received aggression and eviction attempts from workers. Nevertheless, WC and DC drones were equally likely to be accepted by workers. For both drone types accepted individuals had slightly, but significantly greater weights than rejected males. Thus, workers discriminated between drones of different sizes and potential quality upon initial encounter, although these discriminations were not strongly associated with acceptance decisions. After drones were accepted, workers either showed no preference for interacting with WC or DC drones, or if a preference was shown it tended to favor WC drones. Compared to accepted DC drones, significantly more WC drones received grooming for longer periods of time and also spent more time engaged in all interactions with workers combined. DC and WC drones did not differ in the likelihood of receiving trophallaxis or the vibration signal, although for both interactions slightly more WC drones were recipients. Thus, workers may bias some interactions with accepted drones to favor smaller individuals with potential developmental deficiencies, in a manner that could contribute to the production of a greater total number of competitive males and increased colony reproductive output.  相似文献
