Many boreal and polar lichens occupy bipolar distributional ranges that frequently extend into high mountains at lower latitudes. Although such disjunctions are more common among lichens than in other groups of organisms, the geographic origin of bipolar lichen taxa, and the way and time frame in which they colonized their ranges have not been studied in detail. We used the predominantly vegetative, widespread lichen Cetraria aculeata as a model species. We surveyed the origin and history of its bipolar pattern using population genetics, phylogenetic and genealogical reconstruction methods. Cetraria aculeata originated in the Northern Hemisphere and dispersed southwards during the Pleistocene. The genetic signal suggests a Pleistocene dispersive burst in which a population size expansion concurred with the acquisition of a South‐American range that culminated in the colonization of the Antarctic.  相似文献

Lichens are symbioses between fungi (mycobionts) and photoautotrophic green algae or cyanobacteria (photobionts). Many lichens occupy large distributional ranges covering several climatic zones. So far, little is known about the large‐scale phylogeography of lichen photobionts and their role in shaping the distributional ranges of lichens. We studied south polar, temperate and north polar populations of the widely distributed fruticose lichen Cetraria aculeata. Based on the DNA sequences from three loci for each symbiont, we compared the genetic structure of mycobionts and photobionts. Phylogenetic reconstructions and Bayesian clustering methods divided the mycobiont and photobiont data sets into three groups. An amova shows that the genetic variance of the photobiont is best explained by differentiation between temperate and polar regions and that of the mycobiont by an interaction of climatic and geographical factors. By partialling out the relative contribution of climate, geography and codispersal, we found that the most relevant factors shaping the genetic structure of the photobiont are climate and a history of codispersal. Mycobionts in the temperate region are consistently associated with a specific photobiont lineage. We therefore conclude that a photobiont switch in the past enabled C. aculeata to colonize temperate as well as polar habitats. Rare photobiont switches may increase the geographical range and ecological niche of lichen mycobionts by associating them with locally adapted photobionts in climatically different regions and, together with isolation by distance, may lead to genetic isolation between populations and thus drive the evolution of lichens.  相似文献

The aim of the research is to explore the overall in vitro antioxidant activity, total phenol content, reduction power and antimicrobial activity of the methanol extracts of the lichens Cetraria pinastri, Cladonia digitata, Cladonia fimbriata, Fulgensia fulgens, Ochrolechia parella and Parmelia crinita. The methanol extract of the Cetraria pinastri showed a strong antioxidant activity, whereas the extracts of the species Fulgenesi fulgens, Cladonia fimbriata and Parmelia crinita showed the moderate one and the extract of the species Ochrolechia parella and Cladonia digitata the weak one. The methanol extract of the lichen Cetraria pinastri had the biggest total phenol content (32.9 mg/g of the dry extract). A certain correlation was established between the antioxidant activity and the total phenol content for the researched lichen extracts. The work also explores the antimicrobial activity of the methanol extracts of the mentioned species of lichens against six bacterial and eleven fungi species by the disc-diffusion method and by establishing the minimum inhibitory concentration (MIC). The methanol extracts of the lichens Cetraria pinastri and Parmelia crinita showed the strongest both antibacterial and antifungal activity against most of the tested microorganisms. These researches suggest that the lichens Cetraria prunastri can be used as new sources of the natural antioxidants and the substances with antimicrobial features.  相似文献

Phylogenetic relationships within the family Parmeliaceae (lichenized ascomycetes) with emphasis on the heterogeneous group of cetrarioid lichens are reconstructed. The results are based on cladistic analyses of DNA-sequences, morphological and chemical data. Almost all currently recognized cetrarioid genera were included in the analyses together with parmelioid and alectorioid members of the presumably monophyletic family Parmeliaceae. We tried to sample taxonomic diversity of the family as widely as possible. The ITS1-5.8S-ITS2 region of the rDNA and a partial β-tubulin gene from 126 samples representing 82 species were analysed. Cetrarioid lichens were identified as a monophyletic group, supported by both ITS and β-tubulin characters. This group was reanalysed using 47 morphological, anatomical and secondary chemistry characters combined with the DNA data matrix. ITS and β-tubulin sequences provide congruent information, and a clear correlation between DNA-data and conidial shape is observed. The current taxonomy of the cetrarioid lichens is discussed and compared with the phylogenetic trees obtained here. A comprehensive study of the phylogeography of some bipolar or subcosmopolitic species with representatives from both hemispheres was performed. Cetraria aculeata is the only taxon where correlation between DNA-data and geographic origin is observed.  相似文献

Background and Aims

Phenotypic variability is a successful strategy in lichens for colonizing different habitats. Vagrancy has been reported as a specific adaptation for lichens living in steppe habitats around the world. Among the facultatively vagrant species, the cosmopolitan Cetraria aculeata apparently forms extremely modified vagrant thalli in steppe habitats of Central Spain. The aim of this study was to investigate whether these changes are phenotypic plasticity (a single genotype producing different phenotypes), by characterizing the anatomical and ultrastructural changes observed in vagrant morphs, and measuring differences in ecophysiological performance.

Methods

Specimens of vagrant and attached populations of C. aculeata were collected on the steppes of Central Spain. The fungal internal transcribed spacer (ITS), glyceraldehyde-3-phosphate dehydrogenase (GPD) and the large sub-unit of the mitochondrial ribosomal DNA (mtLSUm), and the algal ITS and actin were studied within a population genetics framework. Semi-thin and ultrathin sections were analysed by means of optical, scanning electron and transmission electron microscopy. Gas exchange and chlorophyll fluorescence were used to compare the physiological performance of both morphs.

Key Results and Conclusions

Vagrant and attached morphs share multilocus haplotypes which may indicate that they belong to the same species in spite of their completely different anatomy. However, differentiation tests suggested that vagrant specimens do not represent a random sub-set of the surrounding population. The morphological differences were related to anatomical and ultrastructural differences. Large intercalary growth rates of thalli after the loss of the basal–apical thallus polarity may be the cause of the increased growth shown by vagrant specimens. The anatomical and morphological changes lead to greater duration of ecophysiological activity in vagrant specimens. Although the anatomical and physiological changes could be chance effects, the genetic differentiation between vagrant and attached sub-populations and the higher biomass of the former show fitness effects and adaptation to dry environmental conditions in steppe habitats.  相似文献

Eight lichen species, Cetraria aculeata, Cladonia furcata, Pseudephebe pubescens, Sphaerophorus globosus, Stereocaulon alpinum, Umbilicaria antarctica, Usnea antarctica and Usnea aurantiacoatra, were collected from King George Island, maritime Antarctica, for the evaluation of antioxidant activities. Anti-linoleic acid peroxidation activity, free radical scavenging activity, reducing power and superoxide anion scavenging activity were assessed of methanol and acetone extract of the lichens in vitro. Extract of Umbilicaria antarctica, Cladonia furcata, Sphaerophorus globosus and Usnea antarctica were found to have strong in vitro antioxidant properties. In general, acetone extract exhibited stronger activities than methanol extract. The activity-guided bioautographic TLC and HPLC analysis demonstrated that lecanoric acid was the main antioxidant compound in the acetone extract of Umbilicaria antarctica, the most potent antioxidant lichen species among the test species. The results suggested that several Antarctic lichens and their substances can be used as novel bioresources of natural antioxidants.  相似文献

The number and range of taxa belonging to the Central European section Phaestoglochin (Carex L.) differ significantly among systematic classifications. Our study revealed that all of the quantitative morphological features of perigynia of Carex spicata Huds., Carex muricata subsp. muricata L., Carex muricata subsp. lamprocarpa ?elak., Carex divulsa subsp. divulsa Stokes, and Carex divulsa subsp. leersii Koch., had statistically significant differences among taxa. However, this differentiation did not reflect the known taxonomic divisions of the group and therefore, it did not represent a determining criterion for distinguishing the particular taxa. It also did not confirm the close and significant connections between subspecies within the species studied. Based on morphological features of perigynia, we showed that C. divulsa is a well-distinguished taxon, while the other taxa form a complex group. Analysis of the differentiation of perigynium features among the taxa studied against the background of the plant communities where the plants were grown indicated a distinct effect of the habitat conditions on the morphological features of perigynia.  相似文献

In Europe, the genus Dactylorhiza comprises a bewildering variety of forms that are difficult to sort into discrete species. Most Dactylorhiza species are diploid or tetraploid and contrasting hypotheses have been proposed to explain the complex variation within this group. Using PCR-RFLP analysis in eight putative species, we could identify four highly differentiated chloroplast DNA lineages. The first lineage (clade A) included the unique haplotype found in D. sambucina. Clade B grouped four haplotypes belonging mostly to D. incarnata. Clades C and D included 27 haplotypes detected in diploid D. fuchsii and in all tetraploid species investigated. Eighty percent of the chloroplast variation were consistent with currently accepted species boundaries. The imperfect agreement between chloroplast variation and species boundaries may be ascribed to incomplete lineage sorting and/or reticulation. Our cpDNA results provide strong evidence that the allotetrapolyploids have been formed through asymmetric hybridization with a member of the D. fuchsii / maculata group as the maternal parent.  相似文献

Endemic Hawaiian species in the genus Plantago show considerable morphological and ecological diversity. Despite their variation, a recent phylogenetic analysis based on DNA sequence data showed that the group is monophyletic and that sequence variation among species and morphotypes is low. This lack of sequence polymorphisms resulted in an inability to resolve species and population affinities within the most recently derived clade of this lineage. To assess species boundaries, population genetic structure and interpopulation connectivity among the morphologically and ecologically distinct populations within this clade, genetic variation was examined using eight microsatellite loci. Within‐population genetic diversity was found to be lowest in the Maunaiu, Hawai'i population of the endangered P. hawaiensis, and highest in the large P. pachyphylla population from 'Eke, West Maui. Isolation by distance across the range of populations was detected and indicated restricted dispersal. This result is likely to be attributable to few interisland dispersal events in the evolutionary history of this lineage. Genetic differentiation within islands tended to be higher among populations occurring in contrasting bog and woodland habitats, suggesting ecological barriers to gene flow and the potential role of ecological divergence in population diversification. Overall, these results are consistent with findings from phylogenetic analysis of the entire lineage. Our data bring new insights regarding patterns of dispersal and population genetic structure to this endemic and endangered group of island taxa. As island environments become increasingly fragmented, information of this type has important implications for the successful management of these fragile populations and habitats.  相似文献

Aim To test whether marine biogeographical patterns observed at the community level are also important within species. It is postulated that historical hydrogeographic barriers have driven in situ diversification. Location The intertidal and shallow subtidal zones of southern Australia, New Zealand and nearby islands. Australia's temperate marine communities are characterized by a high degree of endemism and show strong biogeographical structure along an east–west axis. Methods Phylogeographical analysis of the widespread asteriid sea‐star Coscinasterias muricata Verrill across southern Australia and New Zealand. Forty‐two samples from 27 locations were included in phylogenetic analyses of mitochondrial (CO1; control region) and nuclear (ITS2) DNA sequences. Results Analysis of mtDNA revealed a deep phylogenetic split within Australian C. muricata, strongly correlated with latitude. ‘Northern’ haplotypes (latitude ≤ 37.6° S, nine sites, 15 samples) were 7.3–9.4% divergent from ‘southern’ haplotypes (latitude ≥ 37.6° S, 19 sites, 27 samples), consistent with late Pliocene separation. Eastern and western representatives of the ‘northern’ clade were 0.5–1.0% divergent, probably reflecting Pleistocene isolation. The ‘southern’ clade of Australia is also represented in New Zealand, indicating Pleistocene oceanic dispersal. Nuclear DNA (ITS2) sequences yielded relatively little phylogenetic resolution, but were generally congruent with mtDNA‐based groupings. Main conclusions The phylogeographical pattern detected within Australian C. muricata closely resembles marine biogeographical groupings proposed on the basis of community and species distributions. Recurring evolutionary patterns may have been driven by the hydrographic history of southern Australia. Specifically, we suggest that Plio‐Pleistocene temperature change and the repeated opening and closure of Bass Strait promoted allopatric divergence and perhaps cryptic speciation in C. muricata.  相似文献

The paraphyletic diatom genus Nitzschia comprises over 1000 morphologically distinct pennate taxa, known from the benthos and plankton of freshwater, brackish, and marine environments. The principal diagnostic characters for delimitation of Nitzschia species include valve shape, the position and structure of the raphe, presence/absence and shape of the proximal raphe endings and terminal raphe fissures, areola structure, and specific morphometric features such as cell size, and stria and fibula density. In this study, we isolated 12 diatom strains into culture from samples collected at the surface or greater depths of the southeastern Adriatic Sea. Morphological analyses included LM, SEM, and TEM observations, which, along with specific morphometric features, allowed us to distinguish three new Nitzschia species. These findings were congruent with the results of phylogenetic analyses performed on nuclear‐encoded SSU (18S) rDNA and chloroplast‐encoded rbcL and psbC genes. One of the new species (Nitzschia dalmatica sp. nov.) formed a lineage within a clade of Bacillariaceae containing members of the Nitzschia sect. Dubiae, which was sister to Psammodictyon. A second lineage was part of a novel clade that is significantly distinct from other Nitzschia species sequenced so far and includes Nitzschia adhaerens sp. nov. and N. cf. adhaerens. A further new species was found, Nitzschia inordinata sp. nov., which appeared as the sister group to the N. adhaerens clade and the conopeoid Nitzschia species in our phylogenetic trees. Our findings contribute to the overall diversity of genus Nitzschia, especially in identifying some deep branches within the Bacillariaceae, and highlight under‐scoring of this genus in marine plankton.  相似文献

Given sufficient time and limited gene flow, evolutionary lineages tend to transform into separate species. Mechanisms preventing assimilation during repeated gene‐flow events include divergent adaptations and the development of pre‐ or postzygotic isolation. We analysed the morphological and genetic boundaries of three species of the rock lizard clade Darevskiarudis’ (Darevskia rudis, Darevskia valentini, and Darevskia portschinskii) in relation to the environment, and tried to reconstruct evolutionary pathways underlying the observed separation among the species. We studied the geographic distribution of the scalation traits, microsatellite genotypes, and mitochondrial haplotypes. Our analyses showed consistent morphological and genetic patterns at the centre of the ranges for each species, but asymmetric distribution of alleles and scalation characters within the current contact zones among the species. The genetic and morphological diversification of the clade has been shaped during glacial isolation in an area of Southern Caucasus, away from the Black Sea Coast. The ancestral lineage of D. portschinskii separated from the common D. rudis–D. valentini lineage in the middle Pleistocene, and the two latter lineages separated in relatively recent geological time. Neither of the lineages attained complete lineage sorting; moreover, isolation and migration modelling have helped to detect recombinant gene flow from D. rudis to D. portschinskii (but not to D. valentini). This is most likely linked with climatically more similar suitable habitats between D. rudis and D. portschinskii than between D. valentini and the other two species. In itself, the isolation period was insufficient for the development of intrinsic isolation mechanisms in the system studied. It has been proposed that high morphological similarity between closely related species of small-eared shrews resulted from a recent divergence and intermittent population connectivity, presumably due to Pleistocene climatic fluctuations and associated changes in forest habitat distribution. Here we examined the morphological variation of two sister species of small-eared shrews inhabiting cloud forests from Mexico, Cryptotis obscurus and C. mexicanus. We then used ecological niche modelling to provide compelling evidence for current environmental barriers for population connectivity, and for detecting divergent ecological niches between candidate species. Our results indicated that the species boundaries in this clade should be subject to change. High morphological similarity suggested that populations of C. obscurus and C. mexicanus located west of the Isthmus of Tehuantepec, a major geographic barrier for montane species, are conspecific. Niche divergence between these two putative species was not supported indicating niche conservatism across the evolutionary history of these small-eared shrews. In addition, several barriers seem to play a main role for current lineage divergence between populations within this clade. The population located east of the Isthmus, previously referred to C. mexicanus, might prove to represent a new species based on morphological distinction and current geographic isolation. We have highlighted that estimating species’ potential distributions provides insights to evaluating the effect of geographic barriers on lineage divergence and making stronger inferences when delimiting species.  相似文献

Mat-forming lichens dominating high-latitudinal habitats vary in color and geometry. Widespread species are light greenish yellow (usnic acid) and reflect solar radiation, whereas melanic species absorbing most solar wavelengths are spatially more restricted. Color thereby influences lichens’ energy budget and thus their hydration and photosynthetically active periods. By using well-defined cushions from early successional stages on glacier forelands – three melanic(m) and three usnic(u) mat-forming lichens with hair-like branches (Alectoria ochroleuca(u), Gowardia nigricans(m)), hollow terete branches (Cladonia uncialis(u), Cetraria muricata(m)), and flat branches (Flavocetraria nivalis(u), Cetraria islandica(m)) – we quantified hydration traits and analyzed how color and cushion size affect water loss rate (WLR) and duration of active periods. Main findings: 1) WLR declined with cushion size and was highest in melanic lichens. 2) Active periods were longer for usnic than for melanic lichens and increased with size in all groups. 3) Size, color, and taxon nested in color significantly influenced WLR and duration of active periods in linear mixed models. 4) Hair lichen cushions had shorter active periods than growth forms with terete or flat branches due to their more open canopy architecture and lower water holding capacity (WHC). 5) WHC measured for isolated branches highly underestimated WHC for intact cushions.  相似文献

Pereskia aculeata Miller (Cactaceae) is an invasive alien plant from Central and South America that has become a problematic environmental weed in South Africa. A potential biological control agent, the stem-wilter, Catorhintha schaffneri Brailovsky & Garcia (Coreidae), was collected in southern Brazil and imported into quarantine in South Africa. Field host range data suggested that C. schaffneri has a host range restricted to P. aculeata. No-choice nymph survival tests were then conducted on 27 test plant species in 9 families. Survival to the adult stage was only recorded on P. aculeata and the closely related Pereskia grandifolia Haw. (Cactaceae). Mortality was significantly higher on P. grandifolia with only 3% of the nymphs reaching the adult stage compared with 74% on P. aculeata indicating that P. aculeata is the primary host plant. P. grandifolia is native in South America and is of no agricultural importance in South Africa so any feeding on P. grandifolia in South Africa would have no negative environmental or economic consequences. In other tests, adult survival on P. aculeata [25.8 days (SE ± 3.74)] was significantly longer than on other test plant species [4.3 days (SE ± 0.36)] further confirming the host specificity of the species. Impact studies conducted in quarantine indicated that C. schaffneri is damaging to P. aculeata, significantly reducing the number of leaves and the shoot lengths of plants, even at relatively low insect densities. C. schaffneri is safe for release in South Africa and is likely to be a damaging and effective agent.  相似文献

The Neotropical catfish genus Kronichthys contains three species distributed along coastal rivers of southern and southeastern Brazil. Although phylogenetic hypotheses are available, the molecular and morphological diversity and species boundaries within the genus remain unexplored. In this study, the authors generated mitochondrial data for 90 specimens combined with morphometric and meristic data to investigate species diversity, species boundaries and putative morphological signatures in Kronichthys. Phylogenetic and species delimitation results clearly show the presence of four genetic lineages, three within Kronichthys heylandi along the coast from Rio de Janeiro to southern São Paulo and a single lineage encompassing both the nominal species Kronichthys lacerta and Kronichthys subteres from the Ribeira de Iguape basin to Santa Catarina in southern Brazil. Nonetheless, morphological data show overlapped ranges in morphometrics and a definition of only two morphotypes, with clear phenotypic differences in the teeth number: K. heylandi differs from K. subteres + K. lacerta by the higher number of premaxillary teeth (30–52 vs. 19–28) and higher number of dentary teeth (28–54 vs. 17–28). Headwater captures and connections of paleodrainages because of sea-level fluctuations represent the two major biogeographic processes promoting species diversification and lineage dispersal of Kronichthys in the Atlantic coastal range of Brazil.  相似文献

Systematists and taxonomists have benefited greatly from the emergence of molecular methods. Species identification has become straightforward through DNA barcoding and the rapid build‐up of massive DNA barcode reference libraries. In animals, mitonuclear discordance can significantly complicate the process of species identification and delimitation. The causes of mitonuclear discordance are either biological (e.g., introgression, incomplete lineage sorting, horizontal gene transfer androgenesis) or induced by operational factors (e.g., human error with specimen misidentification or incorrect species delimitation). Moreover, endosymbionts may play an important role in promoting fixation of mitochondrial genomes. Here, we study the mitonuclear discordance of wolf spiders species (Lycosidae) (independent cases from Alopecosa aculeata and Pardosa pullata groups) that share identical COI DNA barcodes. We approached the case utilizing double‐digest restriction site‐associated DNA sequencing (ddRADseq) to obtain and analyse genomic‐scale data. Our results suggest that the observed cases of mitonuclear discordance are not due to operational reasons but result from biological processes. Further analysis indicated introgression and that incomplete lineage sorting is unlikely to have been responsible for the observed discrepancy. Additional survey of endosymbionts provided ideas on further research and their role in shaping mitochondrial DNA distribution patterns. Thus, ddRADseq grants an efficient way to study the taxonomy of problematic groups with insight into underlying evolutionary processes.  相似文献

Butterflies of the genus Polyura form a widespread tropical group distributed from Pakistan to Fiji. The rare endemic Polyura epigenes Godman & Salvin, 1888 from the Solomon Islands archipelago represents a case of marked island polymorphism. We sequenced museum specimens of this species across its geographic range to study the phylogeography and genetic differentiation of populations in the archipelago. We used the Bayesian Poisson tree processes and multispecies coalescent models, to study species boundaries. We also estimated divergence times to investigate the biogeographic history of populations. Our molecular species delimitation and nuclear DNA network analyses unambiguously indicate that Malaita populations form an independent metapopulation lineage, as defined in the generalized lineage concept. This lineage, previously ranked as a subspecies, is raised to species rank under the name Polyura bicolor Turlin & Sato, 1995  stat. nov. Divergence time estimates suggest that this lineage split from its sister taxon in the late Pleistocene. At this time, the bathymetric isolation of Malaita from the rest of the archipelago probably prevented gene flow during periods of lower sea level, thereby fostering allopatric speciation. The combination of molecular species delimitation methods, morphological comparisons, and divergence time estimation is useful to study lineage diversification across intricate geographic regions.  相似文献
