The effect of methyl bromide (MB) was tested on active and anhydrobiotic Aphelenchus avenae. A. avenae was induced into anhydrobiosis by three different techniques. Both active and anhydrobiotic nematodes were subjected to 3,000 μ1 MB/liter air for 14 periods from 0 to 82 h. Anhydrobiotic nematodes were more resistant to fumigation than active nematodes, regardless of the technique used to induce anhydrobiosis. The percent survival decreased with increasing MB exposures (μ1 MB × h). For an LD₉₅ of 45,000-54,000 μ1/1 × h were required for active nematodes and >279,000 μ1/1 × h for anhydrobiotic nematodes.  相似文献

Living Xiphinema americanum (Xa) and X. rivesi (Xr) extracted from soil samples and stored for 1-5 days at 4 or 20 C contained aseptate fungal hyphae. The fungi directly penetrated the nematode''s cuticle from spores encysted near the head. Penetration through the stoma, vulva, or anus was rare. Catenaria anguillulae (Cat), Lagenidium caudatura (Lag), Aphanomyces sp. (Aph), and Leptolegnia sp. (Lep) were isolated into pure culture from infected nematodes. The pathogenicity of these zoosporic fungi was determined by incubating mixed freshly extracted Xa and Xr in 2% soil extract (pH = 6.7, conductivity = 48 μmhos, 20 ± 2 C) containing zoospores obtained from single-spore isolates. After 4 days, Cat, Lag, Aph, and Lep had infected 78, 18, 13, and 22%, respectively, of the nematodes. Both Xa and Xr were infected by every fungus; however, the relative susceptibility of Xa and Xr to these fungi was not determined. All noninoculated control nematodes remained uninfected and alive. In a second experiment, parasitism of Xa and Xr by Aph and Lep was increased when nematodes were incubated in 2% soil extract for 4 days before exposure to zoospores. In a third experiment, parasitism of Xa and Xr by Cat was greater in diluted saturation soil extract (conductivity = 100-400 μmhos) than in undiluted saturation extract (conductivity = 780 μmhos). Cat produced small zoospores (4-μm-d), bulbous infection hyphae, and assimilative hyphae of varying diameters in nematodes, whereas Lag, Aph, and Lep produced large zoospores (8-μm-d) and tubular, uniform infection and assimilative hyphae in nematodes.  相似文献

The nematostatic activity of oxamyl, methyl-N'',N''-dimethy]-N-hydroxy-l-thiooxamimidate (oxamyl-oxime) and N,N-dimethyl-l-cyanoformamide (DMCF) was studied by immersing 10 Meloidogyne incognita second-stage juveniles into aqueous solutions of various concentrations of each chemical. At concentrations of 500 to 8,000 μg/ml, oxamyl quickly immobilized immersed juveniles. In all other concentrations studied (down to 4 μg/ml), oxamyl stopped or reduced movement of juveniles within 24 hours. DMCF also quickly immobilized juveniles at concentrations of 4,000 and 8,000 μg/ml and reduced movement at 2,000 μg/ml. Lower concentrations had no observed effect on movement. In solutions of the oxime from 2,000 to 8,000 μg/ml, some reduction of movement was observed, but most juveniles maintained some motion over a period of 24 hours. Juveniles were transferred to water from 4,000 μg/ml solutions of oxamyl and DMCF after various intervals of time in order to determine the effect of duration of exposure to the chemicals on the ability of the immobilized juveniles to recover normal motion. Some recovery was observed even after 24 hours of exposure to DMCF, but none after exposure to oxamyl for longer than 40 minutes.  相似文献

The motility of Meloidogyne incognita second-stage juveniles (J2) and their ability to induce root galls in tomato were progressively decreased upon exposure to nicotine at concentrations of 1-100 μg/ml. EC₅₀ values ranged from 14.5 to 22.3 μg/ml, but J2 motility and root-gall induction were not eliminated at 100 μg/ml nicotine. Nicotine in both resistant NC 89 and susceptible NC 2326 tobacco roots was increased significantly 4 days after exposure to M. incognita. The increase was greater in resistant than in susceptible tobacco. Root nicotine concentrations were estimated to be 661.1-979.1 μg/g fresh weight. More M. incognita were detected in roots of susceptible than in roots of resistant tobacco. Numbers of nematodes within resistant roots decreased as duration of exposure to M. incognita was increased from 4 to 16 days. Concentrations of nicotine were apparently sufficient to affect M. incognita in both susceptible and resistant tobacco roots. Localization of nicotine at infection sites must be determined to ascertain its association with resistance.  相似文献

The first stage of the metastatic cascade often involves motile cells emerging from a primary tumor either as single cells or as clusters. These cells enter the circulation, transit to other parts of the body and finally are responsible for growth of secondary tumors in distant organs. The mode of dissemination is believed to depend on the EMT nature (epithelial, hybrid or mesenchymal) of the cells. Here, we calculate the cluster size distribution of these migrating cells, using a mechanistic computational model, in presence of different degree of EMT-ness of the cells; EMT is treated as given rise to changes in their active motile forces (μ) and cell-medium surface tension (Γ). We find that, for (μ > μmin, Γ > 1), when the cells are hybrid in nature, the mean cluster size, N¯Γ2.0/μ2.8, where μmin increases with increase in Γ. For Γ ≤ 0, N¯=1, the cells behave as completely mesenchymal. In presence of spectrum of hybrid states with different degree of EMT-ness (motility) in primary tumor, the cells which are relatively more mesenchymal (higher μ) in nature, form larger clusters, whereas the smaller clusters are relatively more epithelial (lower μ). Moreover, the heterogeneity in μ is comparatively higher for smaller clusters with respect to that for larger clusters. We also observe that more extended cell shapes promote the formation of smaller clusters. Overall, this study establishes a framework which connects the nature and size of migrating clusters disseminating from a primary tumor with the phenotypic composition of the tumor, and can lead to the better understanding of metastasis.  相似文献

The reproductive biology of the horned viper, Cerastes cerastes gasperettii, in Riyadh region of Saudi Arabia was investigated over a period of one year. Study of reproductive cycle of male and female C. c. gasperettii revealed that the breeding season is relatively short (April and May). Thereafter females laid eggs by mid of July and hatching probably had taken place by the end of September. No activity was observed during winter, this may indicate just a single clutch per year. Relative testis weight to body weight was drastically increased (X¯ = 0.88%) during the peak of reproductive activity (May) where maximal expansion of seminiferous tubules was also attained during April and May (X¯ = 209 μm and 191 μm, respectively). Likewise, the ovarian activity was the highest during May where ovarian parameters were greater in terms of relative ovarian weight to body weight and ova diameter being 0.46% and 2.29 mm, respectively. Fat body weight was increased drastically just before the peak of reproductive activity then started to decline during June. It could be concluded that the harsh desert conditions and similar environments certainly affect reproductive activity of Saudi Arabian reptiles including snakes.  相似文献

The Stepanov equation, relating the intensity of emission, fe(v̄), at a given frequency, and that of absorption, k(v̄), at the same frequency, is applied, in its modified form (see equation 3 in text) to suspensions of Chlorella, Porphyridium, and Anacystis and to chlorophyll solutions. This application can reveal whether the yield of fluorescence, Φ(v̄), is constant, or changes with frequency. In Chlorella (green alga) a sharp drop of Φ(v̄) is indicated towards the lower frequencies (longer waves), beginning around v̄ = 1.48 × 104cm-1 (680 mμ); the Φ(v̄) function calculated from the Stepanov equation is in fair agreement with the directly determined action spectrum for the excitation of chlorophyll fluorescence in this organism. In Porphyridium (red alga) and Anacystis (blue-green alga) application of the Stepanov equation supports the conclusions, derived from direct measurements, of a much earlier “red drop” of the fluorescence excitation spectra. Direct measurements suggest that the drop in Porphyridium may begin at about 1.53 × 104cm-1 (654 mμ); in Anacystis, it may begin already above 1.57 × 104cm-1 (<637mμ). These results confirm the relation, postulated earlier by Duysens and others, between the action spectra of photosynthesis and of chlorophyll a fluorescence in algal cells. The relation of these findings to spectroscopic evidence, suggesting the existence of two main chlorophyll a components in vivo, in green as well as in red and blue-green algae, is discussed.  相似文献

A Pasteuria isolate associated with a population of the lance nematode Hoplolaimus galeatus was discovered in Peru. The infective propagules adhered to adult stages and juveniles and were found filling the bodies of males and females. The endospore and central core diameters measured 4.5 ± 0.4 pm and 1.9 ± 0.2 μm, respectively, which differed from those reported for other Pasteuria isolates found iu North America on the same host. Examinations of endospore ultrastructure with scanning electron microscopy showed the presence of a thin layer of parasporal fibers surrounding the central core, a thin reduced layer of parasporal fibers in contact with the host''s cuticle, and a putative basal core ring.  相似文献

BackgroundThe aim of the study was to Estimate and compare the radiobiological ratio α/β with the heuristic method for a cohort of Mexican patients with prostate cancer (PCa) who were treated with external radiotherapy (RT) techniques at three Hospital Institutions in Mexico City. With the Kaplan-Meier technique and the Cox proportional hazards model, the biochemical relapse-free survival (bRFS) is determined and characterized for cohorts of Mexican patients with PCa who received treatment with external RT. Using these clinical outcomes, the radiobiological parameter α/β is determined using the heuristic methodology of Pedicini et. al.Materials and methodsThe α/β is calculated from the survival curves for different treatment schemes implemented at three distinct hospitals. The Pedicini’s techniques allow to determine the parameters α/β, k and N0 when treatments are not radiobiologically equivalent, therefore, are built up of a set of curved pairs for the biologically effective dose (BED) versus the ratio α/β, where the ratio is given by the intersection for each pair of curves.ResultsSix different values of α/β were found: the first α/β = 2.46 Gy, the second α/β = 3.30 Gy, the third for α/β = 3.25 Gy, the fourth α/β = 3.24 Gy, the fifth α/β = 3.38 Gy and the last α/β = 4.08 Gy. These values can be explained as follows: a) The bRFS of the schemes presents a statistical variation; b) The absorbed doses given to the patient present uncertainties on the physical dosimetry that are not on the modeling; c) Finally, in the model for the bRFS of Eq. (3), there are parameters that have to be considered, such as: the number of clonogenic tumor cells N0, the overall treatment time (OTT), the kick-off time for tumor repopulation Tk and the repopulation doubling time. Therefore, the mean value to α/β for all schemes has an average value of 3.29 (± 0.52) Gy.ConclusionsThe value of α/β¯=3.29(±0.52)Gy is determined from cohorts of Mexican patients with PC a treated with external radiotherapy using the time-dependent LQ model, which is a higher value with respect to the “dogma” value of α/β 1.5 Gy obtained with the LQ model without temporal dependence. Therefore, there is a possibility of optimizing treatments radiobiologically and improving the results of bRFS in Mexican patients with PCa treated with external radiotherapy.  相似文献

Great Boiling Spring is a large, circumneutral, geothermal spring in the US Great Basin. Twelve samples were collected from water and four different sediment sites on four different dates. Microbial community composition and diversity were assessed by PCR amplification of a portion of the small subunit rRNA gene using a universal primer set followed by pyrosequencing of the V8 region. Analysis of 164 178 quality-filtered pyrotags clearly distinguished sediment and water microbial communities. Water communities were extremely uneven and dominated by the bacterium Thermocrinis. Sediment microbial communities grouped according to temperature and sampling location, with a strong, negative, linear relationship between temperature and richness at all taxonomic levels. Two sediment locations, Site A (87–80 °C) and Site B (79 °C), were predominantly composed of single phylotypes of the bacterial lineage GAL35 (p̂=36.1%), Aeropyrum (p̂=16.6%), the archaeal lineage pSL4 (p̂=15.9%), the archaeal lineage NAG1 (p̂=10.6%) and Thermocrinis (p̂=7.6%). The ammonia-oxidizing archaeon ‘Candidatus Nitrosocaldus'' was relatively abundant in all sediment samples <82 °C (p̂=9.51%), delineating the upper temperature limit for chemolithotrophic ammonia oxidation in this spring. This study underscores the distinctness of water and sediment communities in GBS and the importance of temperature in driving microbial diversity, composition and, ultimately, the functioning of biogeochemical cycles.  相似文献

α-Chaconine, a steroid-glycoalkaloid from Solanum tuberosum L., was increasingly more toxic to a free-living nematode, Panagrellus redivivus, with decreasing acidity from about pH 5 to 7. A study of the toxicity to adult nematodes at three concentrations of α-chaconine in buffer from pH 4 to 7.5 indicated that the free base is the nematicidal form of the compound. The median effective doses (ED₅₀) of α-chaconine to inhibit the motility of P. redivivus were estimated as 85 μg/ml at pH 6.7, 170 μg/ml at pH 6.5, and 340 μg/ml at pH 6.2.  相似文献

The basic reproductive number (R₀) and the distribution of the serial interval (SI) are often used to quantify transmission during an infectious disease outbreak. In this paper, we present estimates of R₀ and SI from the 2003 SARS outbreak in Hong Kong and Singapore, and the 2009 pandemic influenza A(H1N1) outbreak in South Africa using methods that expand upon an existing Bayesian framework. This expanded framework allows for the incorporation of additional information, such as contact tracing or household data, through prior distributions. The results for the R₀ and the SI from the influenza outbreak in South Africa were similar regardless of the prior information (R^0 = 1.36–1.46, μ^ = 2.0–2.7, μ^ = mean of the SI). The estimates of R₀ and μ for the SARS outbreak ranged from 2.0–4.4 and 7.4–11.3, respectively, and were shown to vary depending on the use of contact tracing data. The impact of the contact tracing data was likely due to the small number of SARS cases relative to the size of the contact tracing sample.  相似文献

In a replicated field plot experiment, the population density of Meloidogyne incognita was monitored biweekly through the overwintering period (December through April) between soybean crops. The population survived as second-stage juveniles whose numbers remained stable through the winter months and did not decline until February. The yields of plots planted with a M. incognita susceptible cultivar were negatively correlated with the numbers of juveniles recovered at all preplanting sampling dates. In the mid-winter period (December through February), a regression equation describing the relationship predicted a yield reduction (slope) equivalent to 5.36 kg/ha for each juvenile in a 10-cm³ soil sample. In two subsequent field experiments, conducted in different sites and years, mid-winter (November) sampling gave yield reduction predictions of 4.65 and 6.69 kg/ha. Tests of the null hypothesis gave no evidence to indicate that the three slopes differed (P = 0.05). A regression analysis of combined data from the three experiments determined a mid-winter predictive yield reduction of 5.31 kg/ha for each juvenile in the 10-cm³ sample. As the sampling time approached the planting date, there were changes in the predictive yield reductions due to each juvenile in a sample. These are best described by the equation, γ̂ (yield loss) = 54.47 - 0.67X + 0.0023X², where X equals the days remaining between sampling and planting. Soil sampling should be performed during mid-winter (November through January) for the most reliable prediction of soybean yield loss.  相似文献

A method for the cryopreservation of third-stage infective juveniles (IJ) of Steinernema carpocapsae and Heterorhabiditis bacteriophora was developed. Cryoprotection was achieved by incubating the nematodes in 22% glycerol (S. carpocapsae) or 14% glycerol (H. bacteriophora) for 24 hours, followed by 70% methanol at 0 C for 10 minutes. The viability of S. carpocapsae frozen in liquid nitrogen as 20 μl volumes spread over cover slip glass was > 80%. Survival of H. bacteriophora frozen on glass varied from 10 to 60% but was improved to > 80% by replacing the glass with filter paper. Cryopreservation and storage of 1-ml aliqots of S. carpocapsae IJ resulted in > 50% survival after 8 months; pathogenicity was retained and normal in vitro development took place. Trehalose and glycerol levels increased and glycogen levels decreased during incubation of S. carpocapsae IJ in glycerol. Normal levels of trehalose, glycerol and glycogen were restored during post freezing rehydration.  相似文献

''Floratam'' and ''FX-313'' St. Augusfinegrasses (Stenotaphrum secundatum) were compared in a time-course experiment for host suitability and susceptibility to the lance nematode, Hoplolaimus galeatus. Nematode densities were determined in the soil and acid-fuchsin stained roots 42, 84, 126, 168, and 210 days after pots containing 230 cm³ of autoclaved native Margate fine sand/pot were infested with 104 ± 9 nematodes and maintained at 25 ± 2 C in the laboratory. ''FX-313'' was a more suitable host for H. galeatus. Numbers of H. galeatus reached a maximum at 210 days after inoculation, with 5,550 and 4,120 nematodes (adults plus juveniles)/pot for ''FX-313'' and ''Floratam,'' respectively. Root and shoot dry weights of both grasses were not affected by H. galeatus throughout the experiment. Three polyploid, 2n = 30 to 32 (''Floratam,'' ''FX-10,'' and ''Bitterblue'') and three diploid, 2n = 18 (''FX-313,'' ''Florida Common,'' and ''Seville'') S. secundatum genotypes were inoculated with H. galeatus (99 ± 9/pot) and compared with uninoculated controls 210 days after inoculation. St. Augustinegrass genotypes differed as hosts of H. galeatus. ''FX-313'' and ''Florida Common'' represented the high and low extremes, respectively, for nematode reproduction (9,750 and 5,490 nematodes/pot or 4,239 and 2,387 nematodes/100 cm³ of soil). However, differences in root and shoot growth were not detected 210 days after inoculation with H. galeatus.  相似文献

Background

Residual Kidney Function (RKF) is associated with survival benefits in haemodialysis (HD) but is difficult to measure without urine collection. Middle molecules such as Cystatin C and β2-microglobulin accumulate in renal disease and plasma levels have been used to estimate kidney function early in this condition. We investigated their use to estimate RKF in patients on HD.

Design

Cystatin C, β2-microglobulin, urea and creatinine levels were studied in patients on incremental high-flux HD or hemodiafiltration(HDF). Over sequential HD sessions, blood was sampled pre- and post-session 1 and pre-session 2, for estimation of these parameters. Urine was collected during the whole interdialytic interval, for estimation of residual GFR (GFRResidual = mean of urea and creatinine clearance). The relationships of plasma Cystatin C and β2-microglobulin levels to GFRResidual and urea clearance were determined.

Results

Of the 341 patients studied, 64% had urine output>100ml/day, 32.6% were on high-flux HD and 67.4% on HDF. Parameters most closely correlated with GFRResidual were 1/β2-micoglobulin (r2 0.67) and 1/Cystatin C (r2 0.50). Both these relationships were weaker at low GFRResidual. The best regression model for GFRResidual, explaining 67% of the variation, was: GFRResidual=160.3(1β2m)4.2 Where β2m is the pre-dialysis β2 microglobulin concentration (mg/L). This model was validated in a separate cohort of 50 patients using Bland-Altman analysis. Areas under the curve in Receiver Operating Characteristic analysis aimed at identifying subjects with urea clearance≥2ml/min/1.73m2 was 0.91 for β2-microglobulin and 0.86 for Cystatin C. A plasma β2-microglobulin cut-off of ≤19.2mg/L allowed identification of patients with urea clearance ≥2ml/min/1.73m2 with 90% specificity and 65% sensitivity.

Conclusion

Plasma pre-dialysis β2-microglobulin levels can provide estimates of RKF which may have clinical utility and appear superior to cystatin C. Use of cut-off levels to identify patients with RKF may provide a simple way to individualise dialysis dose based on RKF.  相似文献

Laboratory bioassays were conducted to determine the effects of two carbamates, carbofuran (an acetylcholinesterase inhibitor) and fenoxycarb (a juvenile hormone analog), on survival and infectivity of the infective juveniles (IJ) of Steinernema feltiae Umeå strain and Steinernema carpocapsae All strain. Both insecticides caused mortality of IJ in a dose-related fashion. The two nematode species were equally sensitive to fenoxycarb (LD₅₀ ca. 0.03mg/ml). Whereas IJ of S. feltiae were several orders of magnitude more sensitive to carbofuran (LD₅₀ ≤ 0.2 μg/ml) than to fenoxycarb, S. carpocapsae IJ displayed approximately the same degree of sensitivity to carbofuran (LD₅₀ 0.01-0.03 mg/ml) as they did toward fenoxycarb. Toxicity of the carbamates was the same at all exposure periods from 24 to 168 hours'' duration. Determinations of infective doses of nematodes required to cause 50% mortality of Galleria mellonella larvae showed that the infectivity of IJ that survived exposure to either of the two carbamates was not compromised by treatment.  相似文献

Paralongidorus bullatus n. sp. from groundnut soils in Sador''é Niger is described and illustrated. It is distinguishable from most species of the genus by its body and odontostyle lengths, knob-like head, and digitate ending of the protoplasmic region of the tail. Length of the female body is 4.4-5.5 mm, of the odontostyle 132-156 μm, and of the tail 32-44 μm. Tail terminus is conoid to broadly rounded. Uteri are well developed, without sperms. Males are not found. Xiphinema parasetariae Luc 1958, a species inquirenda, is validated and, based on measurements and tail structures, X. attorodorum Luc 1961 is proposed as a synonym of X. parasetariae. Additional measurements are given for its females and juveniles.  相似文献
