Summary Cytogenetic analysis of 121 sperm from a man heterozygous for a t(13;14) Robertsonian translocation was performed using the technique of in vitro penetration of hamster eggs. The frequency of sperm that were chromosomally unbalanced with respect to the translocation was 27%. The frequency of chromosomally normal (36%) and balanced (38%) complements was approximately equal, as theoretically expected. There was no evidence for an interchromosomal effect since the frequency of numerical chromosomal abnormalities (2.5%) and structural chromosomal abnormalities (10.7%) — both unrelated to the translocation — were within the normal range of control donors. The ratio of X-and Y-chromosome bearing sperm was equal, and there was no evidence for preferential segregation of the X chromosome with the translocation.  相似文献

Twenty-three purebred Alpine (n=8), Saanen (n=7) and Damascus (n=8) goat bucks raised at the Institute of Reproduction and Artificial Insemination in Thessaloniki, Greece (40 degrees 37 minutes N, 22 degrees 58 minutes E and altitude 32 m above sea level), were used to study the effect of photoperiod on semen production. Samples were collected with an artificial vagina and examined immediately after collection. In spite of the variation in nearly all semen characteristics studied among the 3 breeds of bucks, there was significant seasonal variation in both semen quantity (volume, concentration and total number of spermatozoa per ejaculate) and quality (percentage of motile spermatozoa, percentage of abnormal spermatozoa and rate of progressive motility). The best semen was produced during the breeding season (late summer and autumn). However, the magnitude of these seasonal effects was not sufficient to prevent bucks from being used for breeding throughout the year. Nevertheless, individual differences in the semen quantity and quality among bucks within a breed make individual evaluation of semen necessary to select the most fertile males for breeding.  相似文献

Karyotypes were prepared for ten Rahaji bulls using cultured peripheral blood lymphocytes. C-banded metaphases were prepared to identify the sex chromosomes and possible translocations. Out of the ten bulls, studied one bull had a 2n number of 59 chromosomes and carried a Robertsonian translocation. The possible importance of the Robertsonian translocation in Zebu cattle in Nigeria is discussed.  相似文献

The objective of this study was to investigate the effect of 2 levels of prebreeding nutrition on reproduction in yearling does artificially inseminated (AI) by the intrauterine laparoscopic method. Forty-two does (Alpine = 22 and Saanen = 20) were randomly penned in groups of 7 and were fed 1 of 2 diets. The diets contained 3.2 Mcal DE/d (MAINT) or 3.5 Meal DE/d (HIGH), which was 10 and 20% higher than the National Research Council recommendations for maintenance requirements. The does were on the 2 feed treatments for 8 wk, after which the MAINT group was switched to the HIGH group diet. A week later, they were fitted with Veramix sponges to synchronize estrus. The sponges were removed from 22, 10 and 10 randomly picked does after 17, 22 and 23 d, respectively. All the does showed estrus within 48 h of removing the sponges. Previously frozen Alpine or Saanen semen (0.5 ml) was deposited into the uterus of does exhibiting standing estrus after anesthetizing them with zylazine and ketamine. Pure breeding was practiced. All the does lost weight prior to breeding. Seventeen does (41%) conceived and kidded by AI while the rest returned to estrus about 23 d later. A significant difference (P < 0.05) in the kidding percentage was observed between the 2 breeds (Alpine = 64% and Saanen = 16%), while the kidding percentage between the 2 diets did not differ (P > 0.05). Of the does that kidded, seven (41%) had singletons, eight (47%) had twins, one had triplets and one had quadruplets. Average litter size (kids/doe kidding) by AI was 1.76. Although the does lost weight prior to breeding, this did not affect their reproduction.  相似文献

A 5-year-old female Walker hound was presented to the Washington State University Veterinary Teaching Hospital as a result of a narrowing of the vulva, which prevented natural breeding. All other physical and clinical findings were normal. Cytogenetic analysis disclosed a chromosome number of 77, with three metacentric chromosomes comprised of two X chromosomes and a Robertsonian translocation of two acrocentric autosomes, chromosomes 21 and 33. Cytogenetic analysis of two full-sister siblings with histories of absence of estrus disclosed one with the same translocation and one with a normal female chromosome constitution. The propositus was artificially inseminated with semen from a karyotypically normal male Walker hound and gave birth to nine live grossly normal pups, six females and three males. Another female pup was stillborn but was grossly normal. Cytogenetic analysis of the live pups disclosed that four (three males and one female) of the nine had the same translocation in all lymphocytes. The remaining five pups (five females) had normal female chromosome constitutions. The litter size was average for this breed. This is a previously unreported Robertsonian translocation in dogs.  相似文献

In Drosophila melanogaster, gametes formed by oocytes in which Robertsonian translocations were induced in an immature stage usually show chromosomal imbalance. It is estimated that fewer than 20% of the gametes bearing newly induced Robertsonian translocations “fusing” X and fourth chromosomes are of balanced constitution. In contrast, when the two acrocentric pairs, X and fourth chromosomes, are replaced by an X-4 Robertsonian translocation, treatment of immature oocytes of homozygotes produces some 5–6-fold fewer sex-chromosome trisomics than do females of normal karyotype. In the place of such trisomics (having separate sex chromosomes), there is a much smaller number of compound-X chromosomes formed and a number of compound-fourth chromosomes as well. However, the production of “XO” males is not appreciably smaller in the translocation homozygotes. A number of possible mechanisms to account for this are suggested. The findings are consistent with the expectations of the hypothesis that radiation-induced nondisjunction results from improper conjunctions of heterologues, brought about by chromatid interchange7–12, 16.  相似文献

We report a patient with mosaicism for two different Robertsonian translocations, both involving chromosome 21. She carries an unbalanced cell line with an i(21q) and a balanced cell line with a rob(21q22q). She is phenotypically normal but has two children who inherited the i(21q) and have Down syndrome. We demonstrate that both abnormal chromosomes are dicentric and that the proband’s 21/21 rearrangement is an isochromosome formed from a maternally derived chromosome 21. We propose a model in which the i(21q) is the progenitor rearrangement in the proband, which subsequently participated in a nonreciprocal rearrangement characteristic of a jumping translocation. A new Robertsonian translocation was found in several animals of the Blonde d'Aquitaine breed. The fused chromosome analyzed by G- and R-banding results from the fusion of chromosomes 21 and 27. C-banding suggests that the fused chromosome is dicentric.  相似文献

A Robertsonian translocation results in a metacentric chromosome produced by the fusion of two acrocentric chromosomes. Rb heterozygous mice frequently generate aneuploid gametes and embryos, providing a good model for studying meiotic nondisjunction. We intercrossed mice heterozygous for a (7.18) Robertsonian translocation and performed molecular genotyping of 1812 embryos from 364 litters with known parental origin, strain, and age. Nondisjunction events were scored and factors influencing the frequency of nondisjunction involving chromosomes 7 and 18 were examined. We concluded the following: 1. The frequency of nondisjunction among 1784 embryos (3568 meioses) was 15.9%. 2. Nondisjunction events were distributed nonrandomly among progeny. This was inferred from the distribution of the frequency of trisomics and uniparental disomics (UPDs) among all litters. 3. There was no evidence to show an effect of maternal or paternal age on the frequency of nondisjunction. 4. Strain background did not play an appreciable role in nondisjunction frequency. 5. The frequency of nondisjunction for chromosome 18 was significantly higher than that for chromosome 7 in males. 6. The frequency of nondisjunction for chromosome 7 was significantly higher in females than in males. These results show that molecular genotyping provides a valuable tool for understanding factors influencing meiotic nondisjunction in mammals.  相似文献

A new dicentric Robertsonian translocation is described in a Holstein cow. The translocation appears to have arisen spontaneously from the centric fusion of autosomal acro centrics 14 and 28 which resulted in a diploid chromosome number of 59. Cytogenetic analysis of 58 boars at an artificial insemination (AI) centre revealed the presence of a reciprocal chromosome translocation, rcp(1;11)(q−;p+), in two Duroc boars. Pedigree analysis of these two boars suggested familial transmission of the chromosome rearrangement. The reproductive consequences of this translocation were determined in a herd of sows that had received semen doses from these and other boars. All sows underwent multiple AI, with different groups established retrospectively depending on the percentage of semen doses provided by the carrier boars ([number of carrier boar doses/total number doses provided] x 100): 0%, 25%, 50%, 75%, 100%. The fertility rates (percentage of successful multiple AIs/total multiple AIs) recorded for multiple AI including semen doses from the carrier boars were not significantly different from those recorded when all semen doses were supplied by normal-karyotype boars. A reduction in litter size of 29.38% was observed, however, in litters sired by one of the carrier boars when its participation in multiple AI was 100%. The number of live-born piglets per litter gradually decreased (P < 0.05) as the percentage participation in multiple AI (25, 50, or 75%) of the carrier boar increased. In addition, both carrier boars sired some piglets with signs of cleft palate and complex malformations of the front legs; these died soon after birth. In conclusion, the boars carrying the translocation rcp(1;11)(q−;p+) showed reduced reproductive performance.  相似文献

The low number of piglets per litter was the reason why a Finnish Yorkshire breed boar used for artificial insemination (AI) was taken for cytogenetic study. The boar had a reciprocal translocation between autosomes 8 and 10, resulting in a reduction in its litter size of about 19%. In addition the litter sizes were counted from 16 female offspring of this 2n = 38,XY; rcp(8;10)(p1.1;q1.3) boar.  相似文献
