The effects of various growth factors on endothelial cell survival in vitro were studied. Using rat heart endothelial cells, the cell survival curves were obtained; the cells were cultured until confluent, the medium was changed to serum-free medium with or without growth factors, and the cells were counted after 3, 6, 9, and 12 days. Transforming growth factor-beta, which is known as a potent growth inhibitor for vascular endothelial cells, shortened the rat heart endothelial cell's survival period, while epidermal growth factor or transforming growth factor-alpha prolonged survival. Insulin did not affect the rat heart endothelial cell's survival. Our data indicate that growth factors play a role not only in cell proliferation but also in cell survival in vitro. In addition, elevated levels of growth inhibitors such as transforming growth factor-beta may cause tissue damage in vivo by affecting cell survival.  相似文献

In this paper we compare two alternative theoretical approaches for simulating the growth of cell aggregates in vitro: individual cell (agent)-based models and continuum models. We show by a quantitative analysis of both a biophysical agent-based and a continuum mechanical model that for densely packed aggregates the expansion of the cell population is dominated by cell proliferation controlled by mechanical stress. The biophysical agent-based model introduced earlier (Drasdo and Hoehme in Phys Biol 2:133-147, 2005) approximates each cell as an isotropic, homogeneous, elastic, spherical object parameterised by measurable biophysical and cell-biological quantities and has been shown by comparison to experimental findings to explain the growth patterns of dense monolayers and multicellular spheroids. Both models exhibit the same growth kinetics, with initial exponential growth of the population size and aggregate diameter followed by linear growth of the diameter and power-law growth of the cell population size. Very sparse monolayers can be explained by a very small or absent cell-cell adhesion and large random cell migration. In this case the expansion speed is not controlled by mechanical stress but by random cell migration and can be modelled by the Fisher-Kolmogorov-Petrovskii-Piskounov (FKPP) reaction-diffusion equation. The growth kinetics differs from that of densely packed aggregates in that the initial spread, as quantified by the radius of gyration, is diffusive. Since simulations of the lattice-free agent-based model in the case of very large random migration are too long to be practical, lattice-based cellular automaton (CA) models have to be used for a quantitative analysis of sparse monolayers. Analysis of these dense monolayers leads to the identification of a critical parameter of the CA model so that eventually a hierarchy of three model types (a detailed biophysical lattice-free model, a rule-based cellular automaton and a continuum approach) emerge which yield the same growth pattern for dense and sparse cell aggregates.  相似文献

In general, growth and differentiation are mutually exclusive, but they are cooperatively regulated during the course of development. Thus, the process of a cell's transition from growth to differentiation is of general importance for the development of organisms, and terminally differentiated cells such as nerve cells never divide. Meanwhile, the growth rate speeds up when cells turn malignant. The cellular slime mold Dictyostelium discoideum grows and multiplies as long as nutrients are supplied, and its differentiation is triggered by starvation. A critical checkpoint (growth/differentiation transition or GDT point), from which cells start differentiating in response to starvation, has been precisely specified in the cell cycle of D. discoideum Ax-2 cells. Accordingly, integration of GDT point-specific events with starvation-induced events is needed to understand the mechanism regulating GDTs. A variety of intercellular and intracellular signals are involved positively or negatively in the initiation of differentiation, making a series of cross-talks. As was expected from the presence of the GDT point, the cell's positioning in cell masses and subsequent cell-type choices occur depending on the cell's phase in the cell cycle at the onset of starvation. Since novel and multiple functions of mitochondria in various respects of development including the initiation of differentiation have been directly realized in Dictyostelium cells, they are also reviewed in this article.  相似文献

A model flow field representative of Kolmogorov eddies in turbulence is proposed, and its two parameters are expressed in terms of the known bioreactor variables epsilon, the rate of turbulent power dissipation, and nu, the fluid kinematic viscosity. The trajectory through this flow field of a small sphere representing a cell is determined, and from that the time-varying local shear rate can be found. This allows calculation of the shear stress at any point on the sphere's surface as it rotates in and moves through the model eddy. The maximum shear stress imposed on the cell by the surrounding turbulence has a range of 0.5-5 dyn/cm(2), and can be estimated by 5.33rho(epsilonnu)(1/2). The shear stress has two major frequency components with ranges of 1-4 and 20-80 s(-1); the higher frequency component is estimated by 0.678(epsilon/nu)(1/2). The rotation of the direction of the shear stress vector at each point on the cell's surface is also discussed. Two ways in which external stresses might affect cell growth are proposed.  相似文献

BACKGROUND: When using traditional microbiological techniques to monitor cell proliferation and viability, stressed, sublethally injured, or otherwise "viable but nonculturable" cells often go undetected. Because of this, such cells often are not considered by mathematical models used to predict bioprocess performance on scale-up and inaccuracies result. Therefore, analytical techniques, decoupled from postsampling growth, are desirable to rapidly monitor individual cell physiologic states during microbial fermentations. METHODS: Microbial cells, including Escherichia coli, Rhodococus sp., and Sacharomyces cerevisiae, were taken at various stages from a range of fermentation processes and stained with one of three mixtures of fluorescent stains: rhodamine 123/propidium iodide, bis-oxonol/propidium iodide, or bis-oxonol/ethidium bromide/propidium iodide. An individual cell's physiologic state was assessed with a Coulter Epics Elite analyzer based on the differential uptakes of these fluorescent stains. RESULTS: It was possible to resolve an individual cell's physiologic state beyond culturability based on the functionality of dye extrusion pumps and the presence or absence of an intact polarized cytoplasmic membrane, enabling assessment of population heterogeneity. This approach allows the simultaneous differentiation of at least four functional subpopulations in microbial populations. CONCLUSIONS: Fluorescent staining methods used in our laboratories have led to a functional classification of the physiological state of individual microbial cells based on reproductive activity, metabolic activity, and membrane integrity. We have used these techniques extensively for monitoring the stress responses of microorganisms in such diverse areas as bioremediation, biotransformation, food processing, and microbial fermentation; microbial fermentation is discussed in this article.  相似文献

The population development of five human tumor cell lines is examined under the influence of elutriator-prepared human monocytes in a serum-free hormone- and growth factor-supplemented medium. Analysis was performed by electronic counting and sizing of tumor cell nuclei and flow cytometric detection of cell cycle phases. Tumor cell death is triggered at rather low monocyte:tumor cell ratios (1:2 to 1:4) whereas it is strongly reduced at high monocyte densities. Furthermore, it is shown that confluence of the target cell population is a necessary prerequisite for lysis. The data suggest that in monocyte/tumor cell cocultures the decision on target cell lysis is not made by the effector cell, but rather by the target cell and that the criterion for this decision is the target cell's ability or inability to respond to a monocyte challenge by arresting the cell cycle in G1. Interactions between target cells play an important role in determining the result of this decision process. A common basis is suggested for this kind of density-dependent monocyte-triggered lysis and density-dependent cell death in 3T3 cell cultures as described previously.  相似文献

Tips of young Phycomyces sporangiophores were dusted with starch grains, and growth photographically recorded. Rates of longitudinal displacement from the cell tip of individual markers were determined, also corresponding rates of change of cell diameter. From these the magnitude and spatial distribution of "relative elemental growth rates" along both longitudinal and circumferential axes of the cell were obtained. Growth rates in these two directions are functions of distance from the cell apex, and have different spatial distributions. In particular, rates of growth in cell circumference are complexly patterned. Relative elemental growth rates in length and in girth are approximately equal and maximal at the cell's apex, with a value of 2.4 mm. mm.–1 hr.–1. The characteristic shape of the tip is maintained constant in the face of its changing substance and position. This shape reflects a steady state of the cell's constituent growth patterns. At every point the growing membrane simultaneously expands in the two dimensions of its surface. The degree of polarization or directional preference of growth is measured by the ratio of longitudinal to circumferential relative elemental growth rate at any point. The ratio is not constant, but changes with position along the tip. This fact does not support the idea that membrane growth is based upon a quantal "growth event." Possible causal factors in oriented membrane growth are discussed.  相似文献

At present it is unclear how Ag dose-dependent T cell functions, such as cytokine production, reflect TCR affinity and how the signal strength afforded by the Ag dose affects the kinetics of cytokine production by the individual T cell. We used a computer-assisted ELISPOT approach to address these issues. IFN-gamma release by a clonal population of CD4 T cells was monitored on a clonal population of APC while titrating the nominal peptide. The frequency of cytokine-producing cells, the net per-cell output of cytokine, and the onset of cytokine production were each found to be functions of the signal strength. Sigmoidal dose-response curves were seen at the clonal population level, but the activation thresholds for the individual T cells followed a Gaussian distribution. Moreover, the overall dose-response curve of the T cell clone revealed cyclic changes, becoming increasingly shifted toward lower Ag concentrations with the duration of time that elapsed since the last restimulation with Ag. Therefore, responsiveness to Ag ("functional avidity") is not a constant parameter of a T cell clone but a function of the T cell's history of last Ag encounter. The implications of such shifting activation thresholds are discussed for autoimmune disease.  相似文献

A general model is proposed for describing the growth behavior of mammalian cell populations, which features:(a) a cell cycle time distribution function with properties such that mean and variance increase with increasing population size; (b) maturation age and maturation rate functions which constrain the maturational pathways of individual cells; and (c) a death rate function, where cell death is construed as irreparable damage to a cell's reproductive apparatus. The biological implications of the model are discussed, and methods for relating the model to real cell systems by means of commonly used experimental techniques are described. This study addresses the mechanism of wall stress relaxation in growing plant cells. The current viscoelastic model of cell wall relaxation, which dates from the work of Preston, Cleland, Lockhart, and others in the 1960s, has serious shortcomings. It has been shown however that the theory of loss of stability (LOS) can be applied to materials in tension, leading to the conclusion that the relaxation of stresses in the walls of any pressure vessel is rigorously modeled using LOS. We propose that LOS also provides a more appropriate and versatile model of stress relaxation in growing plant cells. We argue that when treated as a manifestation of LOS, the regulation of cell turgor has a rigorous and demonstrable basis in the geometrical and physical properties of the cell wall and the cell's ability to import water. Thus plant cell growth can be regarded as an inherently self-limiting process, tunable by biochemical or structural means. Lastly, despite the current limitations of our model, we apply direct measurement of elastic modulus, wall thickness and cell radius obtained from cylindrical Chara corallina cells to generate an initial calculation of critical pressures in a hypothetical spherical cell with the same material properties.  相似文献

Cultures of the CRL-1606 hybridoma (ATCC) have been reported to undergo continuous proliferation with simultaneous death during nutrient limited fed-batch fermentations. The bcl-2 proto-oncogene has been shown to prevent cell death under a variety of otherwise death inducing conditions. We were interested in elucidating the nature of the massive death observed in cultures of CRL-1606, specifically with respect to the possible environmental causes, and the ability of overexpressed human bcl-2 (hbcl-2) to mitigate cell death. Abortive proliferation, or continuous proliferation in the presence of continuous death, could be induced in serum free cultures of CRL-1606 through the withdrawal of insulin provided the culture was competent for cell proliferation. Culture competency for proliferation was found to be solely determined by the presence of cell culture nutrients. Abortive proliferation was defective in cultures transfected with hbcl-2 and the enhanced viability observed resulted from an increased viable cell population and at the expense of the nonviable cell population normally found in untransfected cultures. Abortive proliferation was also observed in serum containing cultures upon serum shiftdowns. Like the insulin-supplemented serum free culture system, hbcl-2 transfected cultures exhibited defects in the abortive proliferation process. These results suggest that the massive death observed during nutrient-limited fed-batch fermentation originate, in part, from growth or survival factor limitations. Hence, approaches to design cell culture media that account for the cell's proliferation requirements without accounting for the cell's survival requirements may represent a cell death sentence. Given the transformed nature of the hybridomas, we conclude that the abortive proliferation of CRL-1606 is a consequence of inappropriate cell cycle entry in a survival factor limited environment.  相似文献

Recent research in cancer progression and treatment indicates that many forms of cancer arise from the development of a small subpopulation of abnormal cancer stem cells (CSCs) that promote cancer growth and spread. Many potential treatments preferentially interact with cells at certain stages of the cell cycle by either selective killing or halting the cell cycle, such as intense, nanosecond-duration pulsed electric fields (nsPEFs). Simple mathematical models of unfed cancer cell populations at the plateau of their growth characteristics may estimate the long-term consequences of these treatments on proliferating and quiescent cell populations. Applying such a model with no transition from the quiescent to proliferating state shows that it is possible for the proliferating cell population to fall below 1 if the quiescent cell population obtains a sufficient competitive advantage with respect to nutrient consumption and/or survival rate. Introducing small, realistic transition rates did not appreciably alter short-term or long-term population behaviour, indicating that the predicted small cell population behaviour (< 1 cell) is not an artefact of the simpler model. Experimental observations of nsPEF-induced effects on the cell cycle suggest that such a model may serve as a first step in assessing the viability of a given cancer treatment in vitro prior to clinical application.  相似文献

Plant cell morphogenesis depends critically on two processes: the deposition of new wall material at the cell surface and the mechanical deformation of this material by the stresses resulting from the cell's turgor pressure. We developed a model of plant cell morphogenesis that is a first attempt at integrating these two processes. The model is based on the theories of thin shells and anisotropic viscoplasticity. It includes three sets of equations that give the connection between wall stresses, wall strains and cell geometry. We present an algorithm to solve these equations numerically. Application of this simulation approach to the morphogenesis of tip-growing cells illustrates how the viscoplastic properties of the cell wall affect the shape of the cell at steady state. The same simulation approach was also used to reproduce morphogenetic transients such as the initiation of tip growth and other non-steady changes in cell shape. Finally, we show that the mechanical anisotropy built into the model is required to account for observed patterns of wall expansion in plant cells.  相似文献

Summary Experimental tumors developed in white Pekin ducks after intramuscular implantation of 20-methylcholanthrene. Cells derived from the primary tumor were adapted successfully to grow in vitro and have growth characteristics similar to that of established cell lines of mammalian origin. The cell density rises rapidly and the doubling time is approximately 19 hr. The duck cells have been cultured succesfully for at least 80 passages in vitro. The continuously cultured cells have the characteristic chromosome pattern of duck, and the DNA of the duck cell line hybridized with duck liver DNA. We believe we have established a continuous cell line of avian origin. Electron-microscopic examinations of the tumor cells and RNA-directed DNA polymerase of the cell-free supernate show no evidence of endogenous virus production. In a previous paper, we proposed a model in which the volume growth rate and probability of division of a cell were assumed to be determined by the cell's age and volume. Some further mathematical implications of the model are here explored. In particular we seek properties of the growth and division functions which are required for the balanced exponential growth of a cell population. Integral equations are derived which relate the distribution of birth volumes in successive generations and in which the existence of balanced exponential growth can be treated as an eigenvalue problem. The special case in which all cells divide at the same age is treated in some detail and conditions are derived for the existence of a balanced exponential solution and for its stability or instability. The special case of growth rate proportional to cell volume is seen to have neutral stability. More generally when the division probability depends on age only and growth rate is proportional to cell volume, there is no possibility of balanced exponential growth. Some comparisons are made with experimental results. It is noted that the model permits the appearance of differentiated cells. A generalization of the model is formulated in which cells may be described by many state variables instead of just age and volume.  相似文献

To assess local elasticity in the red cell's spectrin-actin network, nano-particles were tethered to actin nodes and their constrained thermal motions were tracked. Cells were both immobilized and controllably deformed by aspiration into a micropipette. Since the network is well-appreciated as soft, thermal fluctuations even in an unstressed portion of network were expected to be many tens of nanometers based on simple equipartition ideas. Real-time particle tracking indeed reveals such root-mean-squared motions for 40-nm fluorescent beads either tethered to actin directly within a cell ghost or connected to actin from outside a cell via glycophorin. Moreover, the elastically constrained displacements are significant on the scale of the network's internodal distance of approximately 60-80 nm. Surprisingly, along the aspirated projection-where the network is axially extended by as much as twofold or more-fluctuations in the axial direction are increased by almost twofold relative to motions in the unstressed network. The molecular basis for such strain softening is discussed broadly in terms of force-driven transitions. Specific considerations are given to 1) protein dissociations that reduce network connectivity, and 2) unfolding kinetics of a localized few of the red cell's approximately 10(7) spectrin repeats.  相似文献

We have developed a stochastic model that describes the orientation response of bipolar cells grown on a cyclically deformed substrate. The model was based on the following hypotheses regarding the behavior of individual cells: (a) the mechanical signal responsible for cell reorientation is the peak to peak surface strain along the cell's major axis (p-p axial strain); (b) each cell has an axial strain threshold and the threshold is normally distributed in the cell population; (c) the cell will avoid any direction where the p-p axial strain is above its threshold; and (d) the cell will randomly orient within the range of directions where the p-p axial strains are less than the cell's threshold. These hypotheses were tested by comparing model predictions with experimental observations from stretch experiments conducted with human melanocytes. The cells were grown on elastic rectangular culture dishes subjected to unidirectional cyclic (1 Hz) stretching with amplitudes of 0, 4, 8, and 12%. After 24 h of stimulation, the distribution of cell orientations was determined by measuring the orientations of 300–400 randomly selected cells. The 12% stretch experiment was used to determine the mean, 3.5%, and the standard deviation, 1.0%, of the strain threshold for the cell population. The Kolmogorov-Smirnov test was then used to determine if the orientation distributions predicted by the model were different from experimentally measured distributions for the 4 and 8% stretches. No significant differences were found between the predicted and experimental distributions (4%: p = 0.70; and 8%: p = 0.71). These results support the hypothesis that cells randomly orient, but avoid directions where the p-p axial strains are above their thresholds.  相似文献

Presently, worldwide attempts are being made to apply stem cells and stem cell-derived products to a wide range of clinical applications and for the development of cell-based therapies. In order to harness stem cells and manipulate them for therapeutic application, it is very important to understand the basic biology of stem cells and identify the factors that govern the dynamics of these cells in the body. Several signaling pathways have emerged as key regulators of stem cells. Some of these signaling pathways regulate the stem cell's proliferative capacity and therefore act as direct regulators of the stem cell, whereas others are involved in shaping and maintaining the stem cell niche and therefore act as indirect regulators of the stem cell. It is difficult to identify which signaling pathways critically affect the stem cell's behavior and which are important for maintaining the quiescent population. A stem cell receives different extrinsic signals compared with the bulk population and responds to them differently. In order to manipulate these adult cells for therapeutic approaches it is crucial to identify how signaling pathways regulate stem cells either directly by regulating proliferative status or indirectly by influencing the niche. The main challenge is to identify whether different factors provide diverse extrinsic signals to the stem cell and its daughter cell population, or whether there are intrinsic differences in stem cell and daughter cell populations that is reflected in their behavior. In this study, we will focus on the various aspects of stem cell biology and differentiation, as well as exploring the potential strategies to intervene the differentiation process in order to obtain the desired yield of cells applicable in regenerative medicine.  相似文献
