Simultaneous quantitative determination of the three naturally occurring juvenile hormones in insects (JH-I, JH-II and JH-III) was performed on haemolymph samples of both normally developing locusts and locusts implanted with active corpora allata, using capillary gas chromatography with electron capture detection.In fourth instar female larvae, 24–48 hr after the third ecdysis, as well as in adult females, 18 days after the imaginal ecdysis, only JH-III was detected. In fifth instar female larvae JH-III was present in very low concentrations, if at all.After implantation of four pairs of corpora allata taken from young fourth instar female larvae or one pair or corpora allata taken from adult females into fifth instar female larvae 0–24 hr after ecdysis, an elevation of the JH-III titre was observed. Neither JH-I nor JH-II could be detected. The amount of JH-III, already elevated 2 hr after implantation, remained high for several days in comparison to that of control insects. On the third day after the subsequent moult the JH-III level was comparable to that of normally developing fifth instar larvae. Factors involved in the achievement of the haemolymph JH-titre are discussed.  相似文献

Effect of sublethal concentration of B. thuringiensis on the first, third, fourth and fifth instar larvae of the American bollworm, H. armigera was investigated to study their response to food consumption, digestion, utilization, and their development till adult formation. The young larvae surviving B. thuringiensis treatment in their first instar and third instar delayed larval period by two to three days, but did not consume more food as compared to control. However, they showed higher digestibility of food as compared to control, which was compensated by their reduced ability to utilize the digested food for body substance. Contrary to the effect on first and third instar larvae, the fifth instar larvae surviving B. thuringiensis treatment in its fourth instar consumed less food, showed less absorption efficiency in digesting food, but compensated by increase in the utilization of ingested and digested food into body substance. Insects surviving B. thuringiensis HD-1 sublethal toxicity adapted to normal larval growth when fed on untreated food, depending upon insect growth prior to treatment. The moths emerging from B.thuringiensis treated larvae had sex ratio favouring females, and adult pairs laid less fertile eggs than those from the untreated ones. The response of B. thuringiensis treated larvae to their food and developmental needs is discussed.  相似文献

The plant chemical azadirachtin was administered, either in artificial diet or by oral injection, to fifth instar larvae of the tobacco budworm, Heliothis virescens (Fabr.). At a dietary concentration of 0.03125 ppm, azadirachtin significantly reduced the amount of diet consumed and the weight gained by the larvae. Higher dietary concentrations (0.25 and 0.5 ppm) were necessary to reduce the efficiency of larval conversion of digested and ingested food, respectively. However, the approximate digestibility increased at the dietary concentration of 0.25 ppm.Orally injected azadirachtin (0.25 and 0.5 μg) delayed moulting to the pupal stage, produced defective pupae or adults, and inhibited development to the adult stage. Higher doses (5.0 and 10.0 μg) reduced the pre-pupal weight loss normally associated with pupation, and completely inhibited pupation. At the critical dose of 1.0 μg (the minimal dose that disrupted development to the pupal stage), azadirachtin had less of an effect on older than on younger larvae. Larvae injected on the first day of the fifth instar failed to pupate, whereas approx 40% of those injected on subsequent days pupated.The results suggest that azadirachtin affects H. virescens in a manner similar to other tested species of insects. The significance of these results, especially regarding hormonal events in the insects, is discussed.  相似文献

In fifth instar larvae of Locusta migratoria the haemolymph lipid concentration is elevated after injection of adipokinetic hormone (AKH). This hyperlipaemic response in larvae remains substantially lower than in adults; over 75% of the mobilized lipid consists of diacylglycerol. In addition, unlike adult locusts, fifth instar larvae also exhibit a consistent, though moderate, hypertrehalosaemic response to AKH. The increases of both lipid and carbohydrate concentrations in larvae are dose-dependent, showing a significant linear regression on log dose in the range 0.2–20 pmol AKH.Glycogen phosphorylase in the fat body of fifth instar larvae as well as young adults is activated on injection of AKH, the percentage active phosphorylase increasing linearly with log dose in the range 0.04–20 pmol AKH. For a given response, a somewhat higher dose of AKH is needed in larvae than in young adults.Fat body glycogen phosphorylase is strongly activated during the period of the larval-adult ecdysis, when active phosphorylase accounts for almost half of the total enzyme, which is approximately ten times more than it is two days before, and two days after the ecdysis.The corpora cardiaca of fifth larval instar locusts already possess the potencies to elevate carbohydrate and lipid concentrations in larval haemolymph, and to activate fat body glycogen phosphorylase.  相似文献

Larvae of Diacrisia casignetum Kollar were reared in groups of 100, 50 and 10 on fresh sunflower leaves at 27 ± 1°C temperature, 75 ± 5% r.h. and 12 h light each day. Rates of feeding, excretion and weight gain increased with age of the larva and were greatest in the sixth instar. Differences in these rates attributable to density were statistically heterogeneous (P < 0.001) whereas, of those attributable to instar, the weight gain was significant (P < 0.10). The second-instar larvae had highest levels of protein and carbohydrates in the body tissues; and the sixth-instar larvae had, the least. The pattern of protein assimilation in various instars was similar to that of glycogen except that proportional differences in the latter were pronounced in fifth and sixth instars. Consumption index, growth rate and approximate digestibility were high in the early instars whereas efficiencies of conversion of ingested and digested food were high in advanced instars. Food utilization, as reflected by efficiency of conversion of ingested and digested values in advanced instars, indicated the amount needed for maintenance decreased and more energy was left for growth and reproduction.  相似文献

Application of methoprene to fourth (penultimate) instar larvae of the silkworm Bombyx mori induced the appearance of the feeding dauer larvae at the fifth (last) instar and prevented pupal metamorphosis. Methoprene also increased the protein concentrations of hemolymph last instar larvae by preventing sequestration of storage proteins by the fat body. Usually, the female-specific storage protein 1 (SP1)* disappears from the male hemolymph at the time of the last larval instar. However, exposure of male larvae to methoprene at the penultimate instar enhanced the accumulation of SP1 in the hemolymph. The SP1 accumulated in males did not differ in molecular weight and immunoreactivity from the SP1 produced in female larvae. Both sexes of fourth instar larvae allatectomized on day 1 instantly accumulated SP1 in the hemolymph, and methoprene application after allatectomy suppressed the hemolymph accumulation of the SP1. In contrast, if allatectomy was carried out at a later stage of the fourth larval instar, SP1 concentration in hemolymph of fifth instar larvae did not increase, suggesting the different juvenile hormone action for regulation of SP1 synthesis in the penultimate instar larvae of silkworms.  相似文献

为研究马铃薯甲虫Leptinotarsa decemlineata(Say)幼虫取食新烟碱杀虫剂噻虫嗪和吡虫啉处理(有效成分18 g/100 kg种薯拌种)播种90 d后马铃薯叶片的消化吸收和生长发育情况,在室内采用重量法测定了幼虫取食量、体重增加量、食物利用率,近似消化率以及蛹重等营养指标。 结果表明:70%噻虫嗪种子处理可分散粉剂拌种后,马铃薯甲虫幼虫的取食量、体重增加量、食物利用率、食物转化率、近似消化率和相对生长率以及蛹重显著低于10%吡虫啉可湿性粉剂拌种和空白对照(P<0.05),发育历期显著长于10%吡虫啉可湿性粉剂拌种和空白对照(P<0.05);而10%吡虫啉可湿性粉剂拌种后,马铃薯甲虫幼虫的取食量和3~4龄期的体重增加量与空白对照差异显著(P<0.05),蛹重、1~2龄幼虫的体重增长量、食物利用率、食物转化率、近似消化率、相对生长率和生长发育历期与空白对照差异不显著(P>0.05)。 说明70%噻虫嗪种子处理可分散粉剂拌种对马铃薯甲虫幼虫的取食有显著抑制作用,不能很好地满足其生长发育营养物质的需要,幼虫取食其叶片后,近似消化率和相对生长速率显著降低,生长发育历期显著延长。 这有利于减少当季世代数,降低危害。  相似文献

Summary Although newly-emerged Colias philodice readily accept Medicago sativa, Melilotus alba, and Coronilla varia, fifth instar larvae reared on any single plant species display a highly significant induced feeding preference for their rearing host. Forced host-switching reveals that fifth instar relative growth rate (RGR) on M. sativa and M. alba is significantly reduced by prior feeding on either alternative host. Moreover, regardless of rearing diet, switching to a novel host during the fifth instar results in reduced RGR, relative consumption rate (RCR), efficiency of conversion of digested food (ECD), and pupal weight. These results support the hypothesis that induction of feeding preference is an adaptive response that predisposes larvae to feed on the plant species they are most capable of utilizing for growth.  相似文献

Allatectomy of 1-day-old female desert locusts resulted in an accumulation of lipid in the fat body. This accumulation of lipid was due to the continuation of lipid deposition in the fat body after the period of somatic growth. Somatic growth and feeding activity were unaffected by allatectomy, and so could not be indirect causes of fat body lipid accumulation. Lipid accumulation in allatectomized locusts is more likely to be related directly to a lack of juvenile hormone. Implantation of active corpora allata into 1-day-old adult female locusts resulted in a premature development of oöcytes and a decrease in fat body lipid accumulation; somatic growth was not inhibited. Implantation of active corpora allata into old allatectomized locusts resulted in a decrease in the fat body lipid content and the onset of oöcyte development. The lipid synthetic activity of the fat body, measured by the incorporation of 14C-actate into total fat body lipid, was greatly increased in allatectomized locusts after the period of somatic growth. The protein synthetic activity of the fat body, measured by the incorporation of 3H-leucine into total fat body protein, remained low after the period of somatic growth in allatectomized insects. Juvenile hormone might thus have a dual effect on fat body metabolism, that is suppressing lipid synthesis and stimulating vitellogenic protein synthesis. Increased synthesis of lipid by the fat body would then account for the accumulation of lipid in the fat body after allatectomy. Inhibition of release of lipid from the fat body is unlikely to play a part in the accumulation as allatectomy had no effect on haemolymph lipid concentrations.  相似文献

A factor that affects synthesis of sericin mRNAs of Bombyx mori was analyzed by organ transplantation and allatectomy. When silk glands of the third instar larvae were transplanted into the abdomen of fifth instar larvae, substantial amounts of sericin mRNAs were induced in the transplant. The induced sericin gene activity was suppressed upon re-transplantation into the abdomen of fourth instar larvae and induced again when the second hosts grew up to fifth instar larvae. An allatectomy performed on fourth instar larvae promoted production of these mRNAs, suggesting that the synthesis of sericin mRNA is regulated by the titer of juvenile hormone.  相似文献

为了选育出在生产上具有推广价值的美洲黑杨Populus deltoides优良抗虫品系,本研究以分月扇舟蛾Clostera anastomosis 3龄幼虫为材料,以I-72杨为对照评价和分析了18个美洲黑杨无性系在实验室条件(28±1℃,RH 70%,16L∶8D)下的抗虫性,通过选择性和非选择性取食实验筛选出了11个具有较强抗性的无性系; 测定了美洲黑杨主要营养物质和次生代谢物质含量,研究了不同无性系对分月扇舟蛾体内保护酶和消化酶活性及对该虫生长发育和食物利用的影响。结果表明:美洲黑杨不同无性系抗性的大小与它们叶片中总酚含量呈明显正相关趋势; 抗性水平不同的无性系对分月扇舟蛾幼虫的生长发育和食物利用有不同的影响,表现在幼虫体重、每日体重增加量、相对生长率、近似消化率、食物利用率和食物转化率等主要生长发育指标的变化上,同时对分月扇舟蛾幼虫取食量和排粪量亦有不同的影响。结果说明,分月扇舟蛾对次生代谢物质的反应更为敏感,抗性无性系对分月扇舟蛾幼虫体内SOD和CAT酶活性有明显的激活作用。  相似文献

Two non-cross-hybridizing cDNA clones were isolated from a lambda gt11 cDNA library prepared from Day 2 fifth instar female fat body of Manduca sexta and shown by hybrid selection to code respectively for the two storage proteins arylphorin and female-specific protein (FSP). Analysis of the developmental expression of arylphorin showed its presence during the feeding phases of the penultimate (fourth) and final (fifth) larval instars and its absence during the molt. Abdominal ligation of larvae followed by infusion of Grace's medium showed that this amino acid-rich medium was able to maintain arylphorin expression in fourth instar larvae, but not continued high expression in fifth instar larvae. This nutrient medium however was sufficient to allow initiation of expression in newly ecdysed fifth larval abdomens. Infusion of 5 micrograms 20-hydroxyecdysone (20HE) caused a significant reduction of arylphorin RNA in ligated fourth larval abdomens, whereas 50 micrograms was required in Day 2 fifth larval abdomens to suppress this RNA. Thus, both the lack of incoming nutrients and the rising titer of ecdysteroid contribute to the loss of arylphorin mRNA at the molts and at wandering. By contrast, FSP mRNA was first detected in females on Day 2 of the fifth instar, but not in males until wandering, and then was present throughout the prepupal period. In females allatectomy caused the precocious appearance of FSP mRNA which was prevented by application of 10 micrograms methoprene, a juvenile hormone analog. Expression of FSP mRNA in males however appeared to be independent of hormonal milieu.  相似文献

Food consumption, utilization and ecological efficiency of Zygogramma bicolorata were studied in larvae and adults up to 45 days old. The study revealed that sex had a significant influence on dry food consumption in adults but not in larvae. The other parameters of ecological efficiencies, viz. efficiency of conversion of ingested food (E.C.I.), tissue growth and dry biomass, were highest in the fourth instar and in adult females. The Consumption Index (C.I.) was higher in males than females in both larval and adult stages. Developmental and growth rates were relatively higher in the first and second instars. The fourth instar and adult females were the most efficient consumers and converters of food. Food intake by females was an important determinant of the number of eggs laid.  相似文献

为探明植食性昆虫对受重金属胁迫的寄主植物的生理生态响应机制,本研究用Cd胁迫下银中杨的叶片饲养舞毒蛾幼虫,分析舞毒蛾幼虫对食物的利用情况以及其对Cd的排毒代谢机制.结果表明: 取食Cd胁迫下银中杨的叶片后,舞毒蛾3~6龄幼虫体内的Cd浓度和Cd含量均显著高于对照,但随着幼虫龄期增长,其体内Cd浓度显著降低,而Cd含量有不同程度的提高;舞毒蛾幼虫粪便和虫蜕中的Cd浓度均显著高于对照;舞毒蛾3~5龄幼虫的食物消耗率显著高于对照,而转化率显著低于对照;3~4龄幼虫的食物利用率均与对照差异不显著,但在5龄时显著低于对照.说明在Cd胁迫下,舞毒蛾幼虫能通过有效的排毒代谢途径将体内富集的部分Cd排出体外,且高龄幼虫的排毒代谢能力强于低龄幼虫;舞毒蛾幼虫体质量的增加会对体内的Cd浓度形成一种稀释效应;舞毒蛾幼虫能通过调整食物消耗率和转化率之间的比例,来维持其正常生长发育所需的食物利用率,但超过一定限度后仍会造成食物利用率降低.  相似文献

The objective of this work was to assess, through consumption and utilization of natural food measurements, whether mulberry cultivars, Morus spp., could be recommended to rear the silkworm, Bombyx mori L., in a commercial scale. The mulberry cultivars Miura (standard), Korin and Tailandesa and the hybrids FM 3/3, FM 86, SK 1 and SK 4 were tested. Seventy five fifth-instar commercial hybrid larvae were individualized in gerbox? unities and maintained in a rearing room (25 ± 3oC and 80 ± 10% RH).The mulberry leaves of each of the five cultivars used to feed the silkworm larvae were submitted to bromatological analysis. The dry weight of larvae at the beginning and at the end of the fifth instar, the food consumed and the feces eliminated were recorded to determine the following indexes: relative consumption rate (RCR), relative metabolic rate (RMR), relative growth rate (RGR), approximate digestibility (AD), efficiency of conversion of ingested food (ECI), efficiency of conversion of digested food (ECD), metabolic cost (MC). The bromatological composition analysis of mulberry leaf cultivars revealed that the hybrid SK 4 presented superior nutritional quality compared to the standard cultivar Miura, due to its higher content in crude protein and ethereal extract, and lower detergent fiber content. The hybrid SK 4 was the most adequate food to the silkworm larvae because it was ingested in small amounts, thus providing good digestibility to the larvae, low metabolic cost, good growth rate and one of the highest efficiencies in the conversion of the ingested food and ingested and digested in biomass.  相似文献

Juvenile hormone (JH)-dependent vitellogenin (Vg) synthesis in the fat body of Locusta migratoria is normally limited to sexually mature adult females. As a step toward examining the basis of this limitation, we have tested female and male locusts in a series of stages after the third larval molt for inducibility of Vg synthesis by the synthetic JH analog, methoprene. We find that in the fourth and fifth larval instars fat body of both sexes can be induced to produce Vg, but in the adult stage females respond strongly while no more than trace amounts can be induced in males. Quantitative assays show relative responsiveness in the order: adult female > fifth instar female > fifth instar male ? adult male. During the fifth instar of both sexes, maximal vitellogenic response was obtained in midinstar. After the larval-adult ecdysis, female fat body was unresponsive during the first 4 days, then responsiveness increased and by Day 8 after ecdysis fat bodies were fully as competent to produce Vg as at Day 14, the usual maximum of the first vitellogenic cycle due to endogenous JH. Larval and adult female fat bodies implanted into male larvae are competent for Vg synthesis after metamorphosis, so that the differences between adult male and female cannot be imposed by the male milieu intérieur during the larval-adult molt. In male and female precocious adults, produced by treatment of fourth instars with precocene, fat body responded to methoprene as in normal adults. We conclude that factors intrinsic to the fat body cells, determined early in development, are responsible for differential gene programing in males and females, which is partially expressed by the fifth instar but fully manifest only after a molt in the absence of JH.  相似文献

ABSTRACT. Removal of the antennae from Schistocerca gregaria early in the third or fourth instar resulted in fifth instar nymphs with a green haemolymph and cuticle. Antennectomized adults had a lower somatic dry weight and a smaller fat body than operated control insects. In addition, their rate of sexual development increased and some of their morphometric parameters were altered. Implantation of corpora allata into fourth instar nymphs induced a green coloration of the haemolymph after a few days. It is suggested that antennectomy of Schistocerca nymphs resulted in an increased effectiveness of the corpus allatum hormone with its concomitant effects on nymphal pigmentation, growth and development, and adult morphometries. It is further suggested that antennectomy of crowded locusts may mimic, for them, the uncrowded condition, by reducing sensory input.  相似文献

The uzi fly, Blepharipa zebina, is a well-known larval endoparasitoid of the tropical tasar silkworm, Antheraea mylitta. The present study dealt with the effect of the number of maggots developing per host on host nutritional parameters, parasitoid development and reproduction. Nutritional indices for ingestion, digestion, approximate digestibility, relative consumption rate, relative growth rate, and gain in body weight declined significantly with the increase in parasitoid burden, but the efficiency of conversion of digested food recorded a significant increase. The efficiency of conversion of ingested food remained little affected. The developmental period was significantly extended in larvae parasitized with 5 and 10 maggots per larva (mpl). Cocoon shell weight decreased by 27-63.5% in parasitized groups (1, 2, and 5 mpl) while larvae parasitized with 10 mpl could not spin cocoons. The maggot development period, recovery percentage, and fecundity of the uzi fly declined significantly with the increase in number of maggots developing per host.  相似文献

Fourth and 5th instar larvae ofHeliothis armigera (Hübner) parasitised byEucelatoria bryani Sabrosky consumed significantly less chickpea flour diet than unparasitised larvae of the same age in the laboratory. Significant reduction in diet consumption, larval weight gain and frass produced at 24 and 48 h following parasitisation was observed in 5th instar host larvae. Parasitised larvae retained a greater percentage of food ingested (AD) than did the unparasitised ones. The effect of diets prepared from whorl tissue of resistant and susceptible corn genotypes, Zea mays L., on the larval growth, development, and physiology of fall armyworm, Spodoptera frugiperda (J. E. Smith), was analyzed. Larvae reared on an optimized artificial diet had a higher growth rate and developed faster than those reared on lyophilized whorl tissue from resistant and susceptible genotypes. Larvae reared on the resistant material were smaller and had a longer developmental period. Larvae reared on yellow-green and green whorl sections from resistant plants were significantly smaller than those reared on the same sections of susceptible plants. There was no significant difference in weight when larvae were reared on the yellow whorl regions from either resistant or susceptible lines. Physiological indices were determined for larvae fed resistant and susceptible lyophilized and fresh whorl material. Larvae fed resistant lyophilized material had significantly lower growth rate (GW) and efficiency of conversion of ingested food to body substance (ECI) than those reared on artificial diet or susceptible material. However, there were no significant differences in consumption index (CI), approximate digestibility (AD) and efficiency of conversion of digested food to body substance (ECD) between larvae reared on lyophilized tissue from resistant and susceptible genotypes. Larvae reared on fresh yellow-green whorl sections from resistant plants had significantly lower GW, ECI, and ECD than those reared on susceptible material. In contrast, no significant differences in any of the estimated food consumption and utilization indices were observed between larvae reared on fresh yellow whorl sections from resistant or susceptible plants. These results suggest that some components of whorls from resistant plants, especially the yellow-green region, inhibit food utilization in fall armyworm larvae.  相似文献
